Abstract

Biodegradation test of aromatic compounds by pure cultureof bacterial isolate I4 from bacterial tablet using mineral mediumas growth medium was done. Aromatic compounds tested ascarbon sources used were phenol, p-chlorphenol, p-nitrophenol,2,4 dichlorphenol and benzene. The oval colony forming and highmotility isolate is an aerobic and gram negatif bacteria. Theisolate was able to use citric acid as carbon source, producedacid .from methyl red test and gave negative result of indol test.The isolate formed a slightly blue-green colour colony on agarslant, and it did not use ethanol as carbon source. Thecharacteristic of the isolate I4 indicated that it belonged to thegenus Pseudomonas. Test using selective media confirmed thatthe isolate I4 is a Pseudomonas fluorescence. The capability ofthe bacteria to oxidize of 2,4 dichlorphenol was relatively fast.Experimental results showed that the bacteri grew on benzene atconcentration of 10% (v/v), and degraded 2,4 dichlophenol withrelatively high rate.Keywords: Bacterial tablet, aromatic compounds, phenol. pchlorphenol,2,4 dichlorphenol, p-nitrophenol, benzene, pure culture,Pseudomonas fluorescence, isolate I4

Abstract

Carwash wastewater (CW) contains potentially harmful compounds. Due to water scarcity, its reuse is indispensable. Phytoremediation is one of methods to eliminate pollutants by using plants, such as vetiver grass. This study observed the growth characteristic of vetiver in CW and its capacity in pollutant removal. Treatment with two factors and three replicates was done: Media for vetiver growth was tap water (TW) without carwash wastewater (M0), mixture of 50% tap water + 50% carwash wastewater (M1), and 100% of carwash wastewater without tap water (M2). Media grown without and with vetiver was of V0 and V1 respectively. Individual stem of vetiver was planted hydroponically in a chamber. Plant growth was observed once a week. At the harvesting (day 70), root, stem and leaf of the vetiver were collected separately Results showed that vetiver has a capacity to adapt, survive and growing in CW media. Plant generation achieved 70.1-81.8%, 60.6-75.8%, and 71.7-78.5% for stem, leaf and root respectively. Within 70 days, the pollutant that covered of 78.5 and 57.9% N, 83.5 and 69.0% P, 76.0 and 65.3% COD, 68.6 and 64.8% BOD, 81.3 and 59.5% detergent, 98.6 and 95.8% phenol, 73.3 and 61.5% Pb, and 88.5 and 82.8% Zn could be removed by vetiver, i.e. in growth media M1 and M2, respectively. Abstract

Biohydrogen production research was carried out using raw materials of agricultural organic waste that was obtained from markets around the Bandung city. The organic part, which consisted of agricultural waste material, mainly fruit and vegetable waste, was crushed and milled using blender. The sludge that produced from milling process was then used as a substrate for mixed culture microorganism as a raw material to produce biohydrogen. As much as 1.2 kg.day-1 of sludge (4% of total solid) was fed into bioreactor that had a capacity of 30L. Experiment was done under anaerobic fermentation using bacteria mixture culture that maintained at pH in the range of 5.6-6.5 and temperature of 25-30oC on semi-continuous mode. Parameters of analysis include pH, temperature, total solid (TS), organic total solid (OTS), total gas production, and hydrogen gas production. The results showed that from 4% of substrate resulted 897.86 L of total gas, which contained 660.74 L (73.59%) of hydrogen gas. The rate of hydrogen production in this study was 11,063 mol.L-1.h-1.
