2. Acetic acid and hydrogen peroxide improved defense-related biochemical responses of onion bulbs to black mold rot caused by Aspergillus niger L.

Abstract

During field conditions and storage, onion bulbs are often severely affected by Aspergillus niger L., which leads to substantial yield and quality loss due to black mold. In the El-Minia region of Egypt, we isolated several strains of A. niger from different onion bulbs. To combat the black mold rot disease caused by A. niger, we tested the individual and combined effects of acetic acid and hydrogen peroxide against the most virulent strain under both in vitro and storage (180 days) conditions. Additionally, we measured the levels of antioxidant compounds such as phenolic acids, polyphenols, and ascorbic acid. Our pathogenicity tests helped us identify the most virulent isolate. We found that the environmentally friendly compound peroxyacetic acid, derived from the combination of hydrogen peroxide and acetic acid, reduced black mold infection in both, in vitro and storage conditions. Treating postharvest onion bulbs with 0.2 g L−1 acetic acid and 0.4 g L−1 hydrogen peroxide promised to mitigate the development of black mold rot for 180 days of storage. Furthermore, our results indicated that these treatments increased bioactive antioxidant compounds such as ascorbic acid, phenolic compounds, and polyphenols. These compounds, as secondary metabolites, served as the first line of defense against onion rot disease during storage. In conclusion, applying peroxyacetic acid proved a practical approach to controlling black mold rot disease development in onion bulbs during storage. [ABSTRACT FROM AUTHOR]

Abstract

Bu çalışma kapsamında, Kuzey Agripark Firmasına ait seralarda yetiştirilen ve ıslah materyali olarak kullanılan 17 genotipe ait 319 kavun bitkisi kullanılmıştır. Bu genotiplerin 13 ayrı morfolojik karakter açısından gözlemi yapılmıştır. Morfolojik çalışmalar sonucu elde edilen verilerin istatistiksel analizleri, Temel Bileşenler Analizi (TBA) ve Kümeleme (Cluster) Analiz Yöntemleri kullanılarak gerçekleştirilmiştir. Analizlerde SAS v 9.3 ve Minitab 17.0 bilgisayar paket programları kullanılmıştır. Genotiplerin TBA ve kümeleme analizi ile sınıflandırılması sonucunda donör bitkilere ait 4 farklı grup elde edilmiştir. On yedi genotipe ait bitkilerden elde edilen 76 tane double haploid bitkinin ise 6 morfolojik karakter için gözlemi yapılmış ve elde edilen verilerin istatistiksel analizleri, Temel Bileşenler Analizi (TBA) ve Kümeleme (Cluster) Analiz Yöntemleri kullanılarak gerçekleştirilmiştir. Bu analizlerin sonucunda double haploid bitkilere ait 4 farklı grup belirlenmiştir. Moleküler karakterizasyon çalışmalarında 47 adet SSR primeri tüm genotiplerde taranmış ve polimorfik olan 7 adet SSR primeri çalışmada kullanılmıştır. SSR analizleri sonucu elde edilen veriler için binom veri matriksi oluşturulmuştur. SAS v 9.3 ve Minitab 17.0 bilgisayar paket programları kullanılarak Çok Değişkenli Kümeleme Analizi ve Temel Bileşenler Analizi verilerin analizi için kullanılmıştır. Bu analizlerin sonucunda 17 genotipe ait bitkilerin moleküler olarak gruplandırılmasında 5 farklı grup, 76 tane double haploid bitkinin moleküler gruplandırılmasında ise yine 5 farklı grup oluşmuştur. Ayrıca 76 adet double haploid kavun bitkisinin önemli bir fungal hastalık olan Fusarium oxysporum f. sp. melonis'in 0,1 ve 2 ırkları için moleküler testlemesi SCAR ve CAPS markırları kullanılarak yapılmıştır. 33 tane bitkinin her üç ırka da dayanıklı olduğu bulunmuştur.

In this study, 319 melon lines belonging to 17 genotypes and grown in greenhouse of Kuzey Agripark Company were used. 13 different morphological characters of these genotypes were observed and recorded. Statistical analysis of obtained data was performed by using cluster and principal component analysis (PCA). SAS 9.3 and Minitab 17.0 statistical packaged software were used for data analysis. After classification of genotypes with PCA and cluster analysis, 4 distinct groups (heterotic) belonging to parental lines were obtained.Six morphological characters of 76 double haploid plants belonging to 17 different types of genotype were also observed and statistical analysis of the obtained data were done by using PCA and Cluster analysis as stated above. As a result of these analyses, 4 different groups (heterotic) belonging to double haploid plants were determined. In the molecular characterization, 47 different SSR primers were screened for both 319 melon lines and 76 double haploid plants and 7 polymorphic SSR primers were selected and used in the study. As a result of SSR analysis, binomial data matrix was formed. SAS 9.3 and Minitab 17.0 statistical packaged software were used to perform multivariate cluster analysis and PCA on obtained data. As a result of these analyses, 5 different groups, in molecular classification of plants belonging to 17 genotype and 76 double haploid plants were determined.Beside, the 76 double haploid melon were screened for most serious fungal diseases, Fusarium oxysporum f. sp. melonis races 0,1 and 2, by using SCAR and CAPS molecular markers. 33 of these melonis plants were found to be resistance to 3 races of Fusarium oxysporum f. sp. 84

8. Short-term outcome of suture rectopexy in children with rectal prolapse: laparoscopic versus posterior sagittal approach.

Abstract

Context Innumerable surgical options addressing persistent rectal prolapse are available. This study compared the short-term outcome of laparoscopic suture rectopexy (LSR) with posterior sagittal rectopexy (PSR). Patients and methods A prospective randomized study was carried out on patients requiring rectal prolapse surgery. Patients were randomly allocated into LSR and PSR groups. Patients with neurological/musculoskeletal deficits, lower gastrointestinal tract anomalies and those with previous pelvic or perineal surgeries were excluded. Results A total of 66 patients, who had suture rectopexy done, were followed up for a minimum of 6 months following surgery. There were 33 LSR and 33 PSR. The mean duration of symptoms was 19 months (range: 6 months to 7.5 years). The mean age at operation was 5.9 years (range: 2.5–12 years), with a slight female predominance (54.5%). The mean operative time was 87.2 and 51.3 min for LSR and PSR, respectively. The mean postoperative hospital stay was 41.18 and 31.87 h for PSR and LSR, respectively. LSR had better Manchester Scar Scale scores compared with PSR (mean: 6.45 and 10.09, respectively). LSR patients resumed unrestricted activities earlier than those of PSR (mean: 9.84 and 15.15 days, respectively). Both groups showed comparable improvements in bowel functions and quality of life. Complications were a transient partial recurrence in one LSR patient (3.1%) and two wound infections in PSR group (6.2%). There was one conversion to laparotomy in LSR group (3.1%). Conclusion Both techniques seemed equally effective in eliminating rectal prolapse. Without longer operative times and conversion to laparotomy, LSR would have been absolutely superior to PSR.
