Summary: The role of maternal tissue in embryogenesis remains enigmatic in many complex organisms. Here, we investigate the contribution of maternal tissue to apical–basal patterning in the kelp embryo.Focussing on Undaria pinnatifida, we studied the effects of detachment from the maternal tissue using microsurgery, staining of cell wall modifications, morphometric measurements, flow cytometry, genotyping and a modified kelp fertilisation protocol synchronising kelp embryogenesis.Detached embryos are rounder and often show aberrant morphologies. When a part of the oogonial cell wall remains attached to the zygote, the apical–basal patterning is rescued. Furthermore, the absence of contact with maternal tissue increases parthenogenesis, highlighting the critical role of maternal signals in the initial stages of development.These results show a key role for the connection to the maternal oogonial cell wall in apical–basal patterning in kelps. This observation is reminiscent of another brown alga, Fucus, where the cell wall directs the cell fate. Our findings suggest a conserved mechanism across phylogenetically distant oogamous lineages, where localised secretion of sulphated F2 fucans mediates the establishment of the apical–basal polarity. This study aimed to assess the anti-inflammatory properties of a bioactive glutamic-alanine rich glycoprotein (GP) derived from Undaria pinnatifida on both LPS-stimulated RAW264.7 cells, peritoneal macrophages, and mouse models of carrageenan- and xylene-induced inflammation, investigating the underlying molecular mechanisms. In both in-vitro and in-vivo settings, GP was found to reduce the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) while also inhibiting the production of nitric oxide (NO) and prostaglandin E2 (PGE2) in response to lipopolysaccharide (LPS) stimulation. GP treatment significantly impeded the nuclear translocation of the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) pathway by blocking the phosphorylation of IKKα and IκBα, leading to a reduction in proinflammatory cytokines such as tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6). Additionally, GP effectively inhibited the activation of mitogen-activated protein kinases (MAPKs), with specific inhibitors of p38 and extra-cellular signal regulated kinase (ERK) enhancing GP's anti-inflammatory efficacy. Notably, GP administration at 10 mg/kg/day (p.o.) markedly reduced carrageenan-induced paw inflammation and xylene-induced ear edema by preventing the infiltration of inflammatory cells into targeted tissues. GP treatment also downregulated key inflammatory markers, including iNOS, COX-2, IκBα, and NF-κB, by suppressing the phosphorylation of p38 and ERK, thereby improving the inflammatory index in both carrageenan- and xylene-induced mouse models. These findings suggest that marine resources, particularly seaweeds like U. pinnatifida, could serve as valuable sources of natural anti-inflammatory proteins for the effective treatment of inflammation and related conditions. This study set out to provide a preliminary assessment of nutritional profiles of five selected New Zealand macroalgae to investigate the potential of landcultivated Ulva spp. The New Zealand seaweeds species, selected from a limited range, were either collected from the wild (Pyropia plicata from Tauranga and Kaikōura), or were sourced from commercial wild harvest suppliers (Macrocystis pyrifera and Undaria pinnatifida), and were compared to land-based cultivated Ulva species (Ulva ralfsii and Ulva stenophylloides). Species were assessed for their content of crude protein, total lipids, carbohydrates, fibers, amino acids, mineral profiles, heavy metals and antioxidant capacity (Oxygen Radical Absorbance Capacity, ORAC). The two cultivated Ulva spp. had the highest crude protein content of the six seaweeds analyzed (approximately 21% dw, N×6.25), as expected with nutrient supplemented cultivation. They also had the highest total, essential and branched chain amino acid quantities (151, 70, and 29 mg/g for U. stenophylloides and 138, 62, and 27 mg/g for U. ralfsii). Though the two Ulva spp. were grown to the same cultivation specifications, they varied in carbohydrate and total fiber content (U. stenophylloides: 63% and 62%; U. ralfsii: 39% and 40%, respectively). Pyropia plicata collected in Kaikōura had the highest carbohydrate levels (65%) though not the highest total fiber. Both P. plicata had the highest trace minerals (417 mg/kg and 720 mg/kg for Kaikōura and Tauranga P. plicata respectively). All seaweeds analyzed showed no lipophilic antioxidants, though the two Phaeophyceae had the highest hydrophilic antioxidant content at 115 TE and 168 TE for Macrocystis pyrifera and Undaria pinnatifida, respectively. This paper provides a preliminary indication of the relative nutritional attributes of a range of potential New Zealand seaweed aquaculture targets benchmarked against sealettuce (Ulva spp.). The subcritical water extraction of Undaria pinnatifida (blade, sporophyll, and root) was evaluated to determine its chemical properties and biological activities. The extraction was conducted at 180 °C and 3 MPa. Root extracts exhibited the highest phenolic content (43.32 ± 0.19 mg phloroglucinol/g) and flavonoid content (31.54 ± 1.63 mg quercetin/g). Sporophyll extracts had the highest total sugar, reducing sugar, and protein content, with 97.35 ± 4.23 mg glucose/g, 56.44 ± 3.10 mg glucose/g, and 84.93 ± 2.82 mg bovine serum albumin (BSA)/g, respectively. The sporophyll contained the highest fucose (41.99%) and mannose (10.37%), whereas the blade had the highest galactose (48.57%) and glucose (17.27%) content. Sporophyll had the highest sulfate content (7.76%). Key compounds included sorbitol, glycerol, L-fucose, and palmitic acid. Root extracts contained the highest antioxidant activity, with IC50 values of 1.51 mg/mL (DPPH), 3.31 mg/mL (ABTS+), and 2.23 mg/mL (FRAP). The root extract exhibited significant α-glucosidase inhibitory activity with an IC50 of 5.07 mg/mL, indicating strong antidiabetic potential. The blade extract showed notable antihypertensive activity with an IC50 of 0.62 mg/mL. Hence, subcritical water extraction to obtain bioactive compounds from U. pinnatifida, supporting their use in functional foods, cosmetics, and pharmaceuticals is highlighted. This study uniquely demonstrates the variation in bioactive compound composition and bioactivities across different parts of U. pinnatifida, providing deeper insights. The consumption of seaweed products has gained the public's attention due to various bioactive compounds therein, which are known to have health benefits in obesity-associated metabolic disturbances, cardiovascular diseases, hyperglycemia, and insulin resistance. This review discusses the biological functions of bioactive compounds, such as peptides, polyphenols (phlorotannins and bromophenols), polysaccharides (alginate, fucoidans, laminarin), carotenoids (fucoxanthin), and phytosterols, abundant in Undaria pinnatifida (Sea Mustard, Miyeok, Wakame), Saccharina japonica (formerly Laminaria japonica; Kelp, Dasima, Kombu), and Saccharina latissima (Sugar kelp). Although seaweed products show promising preventive and therapeutic effects of seaweed products, it is crucial to consider potential health risks. Therefore, this review addresses health concerns related to heavy metal contamination and high iodine contents, particularly associated with the consumption of brown seaweed.

To study the graded alcohol precipitation components of fucoidan from Undaria pinnatifida and its hypoglycemic activity in vitro, this study used Undaria pinnatifida as raw material, extracted Undaria pinnatifida fucoidan gum through CaCl2 solution, and precipitated it step by step to obtain four sets of polysaccharide samples (UPP-45, UPP-55, UPP-65, and UPP-75). Determined their molecular weight, total sugar content, uronic acid content, protein content, sulfate content and monosaccharide composition and analyzed by ultraviolet spectrum (UV), Fourier transform infrared spectra(FT-IR) and scanning electron microscopy(SEM). The inhibition of polysaccharides on α-amylase and α-glucosidase was determined to explore their hypoglycemic activity in vitro. The results showed that the higher the concentration of alcohol precipitation, the higher the proportion of small molecular weight polysaccharide. Background: Snakebite envenomation (SBE) causes diverse toxic effects in humans, including disability and death. Current antivenom therapies effectively prevent death but fail to block local tissue damage, leading to an increase in the severity of envenomation; thus, seeking alternative treatments is crucial. Methods: This study analyzed the potential of two fucoidan sulfated polysaccharides extracted from brown seaweeds Fucus vesiculosus (FVF) and Undaria pinnatifida (UPF) against the fibrinogen or plasma coagulation, proteolytic, and phospholipase A2 (PLA2) activities of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom. The toxicity of FVF and UPF was assessed by the hemocompatibility test. Results: FVF and UPF did not lyse human red blood cells. FVF and UPF inhibited the proteolytic activity of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom by approximately 25%, 50%, and 75%, respectively, while all venoms led to a 20% inhibition of PLA2 activity. UPF and FVF delayed plasma coagulation caused by the venoms of B. jararaca and B. neuwiedi but did not affect the activity of B. jararacussu venom. FVF and UPF blocked the coagulation of fibrinogen induced by all these Bothropic venoms. Conclusion: FVF and UPF may be of importance as adjuvants for SBE caused by species of Bothrops, which are the most medically relevant snakebite incidents in South America, especially Brazil. Brown algae have gained attention as a sustainable feedstock for biorefineries due to their ability to sequester carbon dioxide, rapid growth, and high carbohydrate content. The carbohydrate content in brown algae has only been analyzed for a few species, and in most cases, access to fundamental data such as sugar composition is limited, which hinders the assessment of brown algal biomass-based biorefining potential. In this study, the carbohydrate composition of brown algae (Undaria pinnatifida, Saccharina japonica, Ecklonia cava, and Ecklonia stolonifera) was analyzed in detail and application directions were proposed. As a result, alginate and glucan were detected in all resources, and the contents (alginate and glucan wt%) were as follows: U. pinnatifida (39.6 and 4.9 wt%), S. japonica (34.0 and 6.3 wt%), E. cava (24.3 and 7.7 wt%), and E. stolonifera (39.1 and 9.7 wt%). All feedstocks contain trace amounts (2.9–4.9 wt%) or no xylan-mannan-galactan. Mannitol was detected only in S. japonica (26.7 wt%) in rich, showing high potential as a biorefinery feedstock. We highlight that the carbohydrate composition of E. cava and E. stolonifera was analyzed for the first time and the potential use of brown algal biomass in a biorefinery approach. Fucoidan refers to a group of sulphated polysaccharides obtained from brown seaweed, with numerous biological activities. In this study, fucoidan was fortified into Chinese steamed bread (CSB) at different concentrations (0, 1%, 3% and 5%) and the effect of fucoidan on the dough properties, structure properties and bioactivity were investigated. The results showed that fucoidan could change the viscosity of unfermented dough, and a high concentration of fucoidan could remove the free radicals produced by the SH–SS exchange reaction (GS-) in the dough, which significantly reduced the content of disulfide bond and reduced the expanded volume of fermented dough (p < 0.05). In addition, fucoidan forms a physical barrier on the surface of starch particles and hinders the reaction between protein-to-protein; therefore, fucoidan increased the hardness, gumminess and chewiness in CSB, and reduced the specific volume in CSB. Furthermore, the fucoidan-fortified CSB samples were found to have both the ability to significantly reduce the predicted glycemic index (pGI) (p < 0.05) and improve antioxidant activity (p < 0.05). Undaria pinnatifida can effectively deal with organotin pollution through its excellent accumulation and degradation capabilities found under laboratory conditions. However, nothing is known regarding its accumulation, degradation performance, and related impact factors in the wild farming area. In this study, we monitored triphenyltin chloride (TPTCL) contents and degradation products in different algal parts (blades, stipes, sporophylls, and holdfasts) of cultivated U. pinnatifida from December 2018 to May 2019. Our results showed that sporophytes had an accumulation and degradation capacity for TPTCL. The TPTCL contents and degradation products varied with the algal growth stages and algal parts. TPTCL accumulated in the blades at the growth stage and the blades, stipes, sporophylls, and holdfasts at the mature stage. The TPTCL content among algal parts was blades (74.92 ± 2.52 μg kg−1) > holdfasts (62.59 ± 1.42 μg kg−1) > sporophylls (47.24 ± 1.41 μg kg−1) > stipes (35.53 ± 0.55 μg kg−1). The primary degradation product DPTCL accumulated only in the blades at any stage, with a concentration of 69.30 ± 3.89 μg kg−1. The secondary degradation product MPTCL accumulated in the blades at the growth stage and in the blades, stipe, and sporophyll at the mature stage. The MPTCL content among algal parts was blades (52.80 ± 3.48 μg kg−1) > sporophylls (31.08 ± 1.53 μg kg−1) > stipes (20.44 ± 0.85 μg kg−1). The accumulation pattern of TPTCL and its degradation products seems closely related to nutrient allocation in U. pinnatifida. Summary: Rich in numerous nutrients, undaria pinnatifida (UP) exhibited a broad spectrum of biological properties, including anti‐inflammatory, anti‐bacterial, anti‐cancer, and antioxidant properties. A sexual pigment protein complex known as phycocyanin (PC) was the perfect photosensitiser and had no negative effects. Due to their superior qualities, hydrogels were also widely employed in tissue engineering, medication administration, and food packaging at the same time. The aim of this work was to investigate the impact of UVB irradiation on the gel characteristics of PC‐UP hydrogels using UVB irradiation technology rather than chemical crosslinking approach. The results showed that the G′ value of PC‐UP hydrogel increased from 114.17 ± 0.58 Pa to 156.72 ± 0.82 Pa and the G″ value increased from 36.80 ± 0.79 Pa to 58.05 ± 1.06 Pa after UVB irradiation. At the same time, the texture properties of hydrogel were significantly improved, and the hardness, adhesion and chewiness were increased to 214.02 ± 3.85, 198.51 ± 4.25 and 195.81 ± 5.69, respectively. After UVB irradiation, hydrogen bond was formed in PC‐UP hydrogel to improve its thermal stability. In conclusion, the results show that PC decomposes with alginic acid, potassium salt, and sodium salt in UP under UVB irradiation to form a gel network structure, which enhances its gel properties. This study provides guidance for the photo response of PC‐UP hydrogels, provides ideas for the application of PC‐UP hydrogels, and broadens the application field of future hydrogels. Background: Allergic rhinitis (AR) is the most prevalent form of atopic disease. Undaria pinnatifida has potent antioxidative, antidiabetic, and anti‐inflammatory properties. Aims: We investigated the immunomodulatory effect of Undaria pinnatifida extract (UPE) on allergic inflammation in an AR mouse model. Materials & Methods: Mice were sensitized and intranasally challenged with ovalbumin (OVA), and the Th1/Th2 and Th17/Treg‐related cytokines and histopathology were exanimated after UPE treatments. Enzyme‐linked immunosorbent assay was performed using serum samples and NALF to detect OVA‐specific immunoglobulins and inflammatory cytokines. Mitogen‐activated protein kinases (MAPKs) were measured by western blotting analysis, and an in vitro study measured mast cell activation induced by compound 48/80. Results: After UPE treatment, nasal and lung allergy symptoms, nasal mucosal swelling, and goblet cell hyperplasia were ameliorated. Oral UPE regulated the balance of Th1/Th2 and Th17/Treg cell differentiation in AR mice in a dose‐dependent manner. In addition, UPE attenuated the migration of eosinophils and mast cells to the nasal mucosa by suppressing nuclear factor kappa B (NF‐κB)/MAPKs. The levels of anti‐OVA IgE and IgG1 were also decreased. Discussion: UPE inhibited inflammation by regulating the NF‐κB/MAPKs signaling pathway and supressing the activation of critical immune cells such as eosinophils and mast cells. The immune system acts as a defense mechanism against foreign antigens. Impairment of the immune system leads to the development of chronic diseases such as respiratory infections, cancer, cardiovascular diseases, and neurodegeneration. Macrophages, natural scavengers that are part of innate immunity, are known to directly participate in scavenging foreign antigens. The functional modulation of macrophages could be an effective treatment for pathogens. Seaweeds are marine macroalgae known to exhibit multiple bioactive properties. Thus, this study evaluated the immune-enhancing properties of marine brown algae extracts of Sargassum horneri (SH), Undaria pinnatifida (UP), and Sargassum fusiforme (SF) on murine macrophage cells. The results showed that all three algal extracts stimulated cell proliferation. SH and UP outshined SF in enhancing the expression levels of IL-1β, TNF-α, and IL-6 at almost all the concentrations tested as compared to SF which showed similar effects only at 200 or 400 μg/mL. A similar trend was seen in TNF-α, NO, and PGE2 production. Additionally, only SH and SF could enhance the mRNA expression levels of IL-12, and only SH upregulated the mRNA expression level of IL-10. The algal extracts also enhanced the phagocytosis activity of macrophages at 50–400 μg/mL for SH and 100–400 μg/mL for UP and SF. The flavor of algae was one of the key factors for consumer acceptance. The objective of this study was to investigate the characteristic volatile compounds in cooking and seasoned cooking edible brown seaweeds (Undaria pinnatifida and Laminaria japonica). The gas chromatography-ion mobility spectrometry (GC-IMS) and electronic nose (E-nose) analysis showed that baking resulted in significant difference in flavor of brown seaweeds. However, the overall effect of cooking was not as significant as that of the seasoning solution treatment. Additionally, brown seaweeds treated with the seasoning solution were more acceptable. Undaria pinnatifida was found to contain 72 volatile flavor compounds, while Laminaria japonica had a total of 70. This study proved the applicability of GC-IMS combined with E-nose technology to detect the changes of volatile components of brown seaweeds after processing, providing beneficial knowledge and basic theory for the deep processing of brown seaweeds.

In recent years, carotenoids, as photosynthetic pigments, have drawn increasing interest due to their health benefits. This is because carotenoids have possessed enormous potential in lowering the risk of cardiovascular, cataracts, macular degradation and malignant tumours. Although many carotenoids can be produced via chemical synthesis, the commonly used petroleum-derived extraction method can be problematic for the environment and potentially contaminate the isolated carotenoids. Therefore, in contrast to chemosynthesis, microalgae can be considered an excellent alternative natural source to synthetic ones. Varying in microalgal strains, a variety of different carotenoids can be produced, for instance, the β-carotene from Dunaliella salina, the fucoxanthin from Undaria pinnatifida and the astaxanthin from Haematococcus pluvialis. Besides, the pharmacological action of carotenoids is highly associated with their bioaccessibility and bioavailability, which is one of the research directions and affecting factors of the carotenoid application in the food, pharmaceutical, nutraceutical and cosmeceutical industries. Despite there being numerous publications about carotenoids, a comprehensive review on the microalgal carotenoids and their triggered bioactivities is still lacking. Considering the significance of bioavailability, this review presents extensive information on microalgal carotenoids, including their classifications, bioaccessibilities, absorption process mechanisms, bioactivities and relevant health benefits. Alginate, a promising biopolymer in the food, biomedical, pharmaceutical, and electronic materials industries, is characterized by its biodegradability, biocompatibility, low toxicity, and gel-forming properties. It is most abundantly found in brown algae. However, conventional dilute acid and alkali extraction methods face limitations in commercialization due to their long processing time, low throughput, and high solvent requirements. In this study, a microwave-assisted extraction (MAE) process for sodium alginate was designed to improve extraction efficiency. The solid/liquid ratio, extraction temperature, and extraction solvent concentration were major variables affecting sodium alginate extraction from Undaria pinnatifida (sea mustard). They were then statistically optimized using response surface methodology. Under optimal conditions (13.27 g/L, 91.86 °C, 2.51% (w/v), and 15 min), the yield was 38.41%, which was 93.43% of the theoretical content of sodium alginate in Undaria pinnatifida. Our work has confirmed the productivity and industrial feasibility of the efficient extraction of sodium alginate from marine biomass, and we hope that it will serve as an encouraging case for the application of biopolymers as one of the desirable options for alternative petrochemicals to construct a sustainable society. Population dynamics can be influenced by physical and biological factors, particularly in stressful environments. Introduced species usually have great physiological plasticity, resulting in populations with different traits. Undaria pinnatifida, a macroalga originally described from northeast Asia, was introduced in Northern Patagonia, Argentina (San Matías Gulf) around 2010. To describe the spatio‐temporal variability in population structure and morphometry of U. pinnatifida, we conducted monthly field samplings for 2 years at the intertidal area of two contrasting sites in the San Matías Gulf. Individuals of U. pinnatifida were classified by developmental stage, and their morpho‐gravimetric variables were measured. In both intertidal sites juveniles were found in higher proportion during austral autumn and grew and matured during the autumn‐winter months (from May onwards), and individuals senesced during early austral summer (December and January). Conversely, density and biomass were largely different between sites, and individuals showed slight morphological variability between sites. Environmental (e.g., nutrient concentration, available substrate) and biological factors (e.g., facilitation, competition) may explain the observed differences. Since there is not a macroalga with U. pinnatifida morphometrical characteristics in the intertidal environments of San Matías Gulf, studying this recent introduction gives us a better understanding of its potential ecological effects. The phenology of the alga-dwelling amphipod Ceinina japonica Stephensen, 1933 (Amphipoda: Eophliantidae) was studied at Rishiri Island, Hokkaido, Japan, from May 2016 to March 2017. Seasonal shifting between the host algal species was confirmed through observations at a coastal field site and a nearby aquaculture facility for kelp. Amphipods mainly occurred in the farmed biennial kelp Saccharina japonica var. ochotensis and wild algae Undaria pinnatifida, with Agarum clathratum, Costaria costata, Saccharina cichorioides, and Sargassum boreale as new host algal records. Amphipods occurred in sporophytes of U. pinnatifida at the field site from February to March and from May to July, but they were found in the kelp of S. j. var. ochotensis during the rest of the year, from August to the following January. Individuals of C. japonica infected sporophytes of the large brown alga Undaria pinnatifida from February to July, and C. japonica reproduced from July to September, a period during which the amphipods switch to a different host in July, the sporophytes of the saccharinan kelp S. j. var. ochotensis, where they stay until January. The amphipods then switched back to U. pinnatifida in January or February. Experimental evidence from a kelp-culturing facility also confirmed the timing of this host shift. The removal of U. pinnatifida from algal culture ropes in such facilities is suggested as a method to prevent amphipod damage, which is important for commercially farmed S. j. var. ochotensis. The lipidic extract of Undaria pinnatifida, one of the worst invasive species, was investigated for its potential exploitation in biotechnological applications. The antimicrobial activity of the lipidic extract in three different portions (blade, sporophyll, and holdfast) was assessed by using the Kirby–Bauer method, while the antioxidant activity was evaluated by the TEAC, ORAC, and Folin–Ciocalteu assays. NMR spectroscopy and thin-layer chromatography were employed for the chemical characterization. The extracts showed antibacterial activity against several of the tested Vibrio species: V. aestuarinus, V. fischeri, V. furnisii, V. inusitatus, V. litoralis, and V. mediterranei, including some pathogens for farmed fish. Intriguing antioxidant activity was recorded, with the highest value in the blade (126.907 ± 28.993 mmol Trolox equivalent/g TEAC). Free, saturated, unsaturated, and polyunsaturated fatty acids were highlighted by 1D and 2D NMR spectroscopy. The presence of ω-3 and ω-6 PUFAs indicates the importance of this algal species in the food industry. We suggest the employment of U. pinnatifida as source of new and safer therapeutic agents to control fish and shellfish diseases due to vibriosis, as well as a source of natural antioxidants that are useful for human health, considering the growing interest in the development of strategies for invasive seaweed control. Undaria pinnatifida, a marine biological resource from which antioxidants such as polysaccharides can be obtained, is primarily distributed in the coastal areas of East Asia. Reactive oxygen species (ROS) are essential for physiological processes; however, excess ROS levels in the body result in cellular oxidative damage. Several extraction methods exist; however, factors such as long extraction times and high temperatures degrade polysaccharides. Therefore, this study aimed to increase the yield of U. pinnatifida sporophyll extract (UPE), a U. pinnatifida byproduct, using ultrasonication, an environmentally friendly extraction method, and identify UPE components with antioxidant activity. UPE_2, 4, 6, and 8 extracts were obtained at extraction times of 2, 4, 6, and 8 h, respectively. UPE_8 had the highest yield (31.91%) and polysaccharide (69.22%), polyphenol, (8.59 GAE μg/mg), and fucoxanthin contents (2.3 μg/g). UPE_8 showed the greatest protective and inhibitory effects on ROS generation in H2O2-damaged Vero cells. Ethanol precipitation of UPE_8 confirmed that UPE_8P (precipitate) had superior antioxidant activity in Vero cells compared to UPE_8S (supernatant). UPE_8P contained a large amount of polysaccharides, a major contributor to the antioxidant activity of UPE_8. Abstract Background Allergic rhinitis (AR) is the most prevalent form of atopic disease. Undaria pinnatifida has potent antioxidative, antidiabetic, and anti‐inflammatory properties. Aims We investigated the immunomodulatory effect of Undaria pinnatifida extract (UPE) on allergic inflammation in an AR mouse model. Materials & Methods Mice were sensitized and intranasally challenged with ovalbumin (OVA), and the Th1/Th2 and Th17/Treg‐related cytokines and histopathology were exanimated after UPE treatments. Enzyme‐linked immunosorbent assay was performed using serum samples and NALF to detect OVA‐specific immunoglobulins and inflammatory cytokines. Mitogen‐activated protein kinases (MAPKs) were measured by western blotting analysis, and an in vitro study measured mast cell activation induced by compound 48/80. Results After UPE treatment, nasal and lung allergy symptoms, nasal mucosal swelling, and goblet cell hyperplasia were ameliorated. Oral UPE regulated the balance of Th1/Th2 and Th17/Treg cell differentiation in AR mice in a dose‐dependent manner. In addition, UPE attenuated the migration of eosinophils and mast cells to the nasal mucosa by suppressing nuclear factor kappa B (NF‐κB)/MAPKs. The levels of anti‐OVA IgE and IgG1 were also decreased. Discussion UPE inhibited inflammation by regulating the NF‐κB/MAPKs signaling pathway and supressing the activation of critical immune cells such as eosinophils and mast cells. Conclusion UPE may have therapeutic potential for AR.

Undaria pinnatifida is a brown alga with high economic value. Its annual production is second only to Saccharina japonica among economically important brown algae in China. Due to climate change and increased cultivation density, the occurrence of diseases in cultivated seaweeds has become more frequent and serious in recent years. Most diseases are directly or indirectly related to the interactions between the seaweed host and the epiphytic microorganisms such as bacteria. There is a close relationship between algae and epiphytic microorganisms. When the phycosphere niche maintains a dynamic balance, the two rely positively on each other growing and developing together. When the balance is disturbed, the structure of the epiphytic microbial community may change, possibly resulting in algal diseases. The absence of certain microbes may also lead to the failure of key biological processes such as the morphogenesis of the host algae. Therefore, understanding the composition of the epiphytic microbial community is of great significance for the study of the interaction between U. pinnatifida and epiphytic microorganisms and the better control of U. pinnatifida diseases. In addition, stock resources of U. pinnatifida are usually conserved in the form of gametophytes, which can persist for a long time under controlled conditions. Hence, understanding the composition of the epiphytic microbial community will also facilitate the development of efficient conservation methods and help remove microbial contaminations when axenic cultures need to be established. The life history of U. pinnatifida consists of the alternation between heteromorphic macroscopic sporophytes and microscopic gametophytes. The stark morphological and physiological differences between these two phases suggest that the composition of epiphytic microbial communities between them is likely different. However, studies on the composition of epiphytic microbial communities, especially comparison studies between sporophytes and gametophytes in U. pinnatifida are limited. The advent of high-throughput sequencing provides robust and efficient tools for studying the composition and relative abundance of the microbial community associated with the algae. In the present study, sporophytes and gametophytes (each with three biological replicates) of U. pinnatifida derived from the cultivated population in Dalian China were selected as the study objects. After DNA extraction and PCR amplification of the v3–v4 region of 16S rRNA gene and the v4 region of 18S rRNA gene, Illumina HiSeq 2500 high-throughput sequencing platform was used to sequence these specific regions. We identified and classified the composition of epiphytic microbial communities of the gametophytes and sporophytes of U. pinnatifida based on the sequencing results. A total of 446 932 effective reads were obtained through 16S rRNA gene sequencing. The raw reads have been submitted to the GenBank database (https://www.ncbi.nlm.nih.gov/) with the accession number PRJNA823903. The bacterial community composition of gametophyte and sporophyte was revealed to be significantly different, and the diversity of the bacterial community in gametophyte samples was higher than that in sporophyte samples. In gametophytes, Proteobacteria (66.67%) was the most dominant phylum, followed by Bacteroidetes (13.48%) and Cyanobacteria (11.13%). At the class level, Alphaproteobacteria (34.58%) was the most abundant, followed by Gammaproteobacteria (31.01%), Bacteroidia (13.16%), and Oxyphotobacteria (11.13%). Cyanobacteria (95.67%) was predominantly detected in sporophytes, followed by Actinobacteria (1.65%) and Firmicutes (1.48%). The distribution of Oxyphotobacteria, Alphaproteobacteria, Bacteroidia, Gammaproteobacteria, Negativicutes, OM190, Acidimicrobiia, Erysipelotrichia, Planctomycetacia, and Verrucomicrobiae were found to be different between gametophyte and sporophyte samples, among which OM190, Acidimicrobiia and Planctomycetacia were unique to gametophytes. The genus-level bacterial groups detected in gametophyte samples were Lewinella (10.06%), Leucothrix (5.99%), Sulfitobacter (4.06%), Bifidobacterium (0.02%), while Bifidobacterium accounted for 1.41% of the bacterial genus of sporophyte samples. There were 57.37% and 95.68% uncultured bacterium in gametophytes and sporophytes, respectively. We obtained a total of 473 770 effective reads through 18S rRNA gene sequencing. A major share (97.22%) of microeukaryotes in gametophytes were unclassified, while in sporophytes, the number was 94.95%. Streptophyta, Intramacronucleata, Basidiomycota, Apicomplexa, Arthropoda, Bacillariophyta, Chordata, Gastrotricha, Ascomycota, and Mucoromycota were detected both in the gametophytes and sporophytes. Among them, Basidiomycota, Ascomycota, and Mucoromycota belong to fungi. The community abundance of sporophyte samples was higher than that of gametophytes. In gametophyte samples, except for Phaeophyceae (88.77%) to which U. pinnatifida belongs, Copepoda of Arthropoda, Mediophyceae of Bacillariophyta, Mammalia of Chordata, Prostomatea of Intramacronucleata, and Liliopsida were dominant, with a proportion of 0.62%, 0.50%, 0.25%, 0.23%, and 0.23%, respectively. In addition to Phaeophyceae (94.49%), Conoidasida of Protozoa, Agaricomycetes of Basidiomycota, Spirotrichea of Intramacronucleata and Mammalia of Chordata were predominant in sporophytes, accounting for 0.91%, 0.83%, 0.51% and 0.24% of OTUs, respectively. Chytridiomycetes, Nassophorea, Colpodea, Tremellomycetes, Sordariomycetes, Conoidasida, Agaricomycetes, Arachnida, and Chromadorea were only detected in the sporophytes, and there was a significant difference in Spirotrichea abundance between gametophytes and sporophytes. In conclusion, the composition of epiphytic microbial communities and the relative abundance of different bacteria and microeukaryotes in the sporophytes and gametophytes of U. pinnatifida were determined through high-throughput sequencing of the amplicons of the v3–v4 region of 16S rRNA gene and the v4 region of 18S rRNA gene. Remarkable differences were revealed between the two life stages, indicating that their growth and development are associated with different microbial communities. The results of this study provide valuable information for sustainable cultivation and stock culture conservation of this important kelp species.

The subcritical water extraction of Undaria pinnatifida (blade, sporophyll, and root) was evaluated to determine its chemical properties and biological activities. The extraction was conducted at 180 °C and 3 MPa. Root extracts exhibited the highest phenolic content (43.32 ± 0.19 mg phloroglucinol/g) and flavonoid content (31.54 ± 1.63 mg quercetin/g). Sporophyll extracts had the highest total sugar, reducing sugar, and protein content, with 97.35 ± 4.23 mg glucose/g, 56.44 ± 3.10 mg glucose/g, and 84.93 ± 2.82 mg bovine serum albumin (BSA)/g, respectively. The sporophyll contained the highest fucose (41.99%) and mannose (10.37%), whereas the blade had the highest galactose (48.57%) and glucose (17.27%) content. Sporophyll had the highest sulfate content (7.76%). Key compounds included sorbitol, glycerol, L-fucose, and palmitic acid. Root extracts contained the highest antioxidant activity, with IC50 values of 1.51 mg/mL (DPPH), 3.31 mg/mL (ABTS+), and 2.23 mg/mL (FRAP). The root extract exhibited significant α-glucosidase inhibitory activity with an IC50 of 5.07 mg/mL, indicating strong antidiabetic potential. The blade extract showed notable antihypertensive activity with an IC50 of 0.62 mg/mL. Hence, subcritical water extraction to obtain bioactive compounds from U. pinnatifida, supporting their use in functional foods, cosmetics, and pharmaceuticals is highlighted. This study uniquely demonstrates the variation in bioactive compound composition and bioactivities across different parts of U. pinnatifida, providing deeper insights. Significant correlations between chemical properties and biological activities emphasize the use of U. pinnatifida extracts for chronic conditions.

It has been optimized thermal acid hydrolytic pretreatment and enzymatic saccharification (Es) in flask culture of Undaria pinnatifida seaweed, which is a prebiotic. The optimal hydrolytic conditions were a slurry content of 8% (w/v), 180 mM H2SO4, and 121°C for 30 min. Es using Celluclast 1.5 L at 8 U/mL produced 2.7 g/L glucose with an efficiency of 96.2%. The concentration of fucose (a prebiotic) was 0.48 g/L after pretreatment and saccharification. The fucose concentration decreased slightly during fermentation. Monosodium glutamate (MSG) (3%, w/v) and pyridoxal 5′-phosphate (PLP) (30 μM) were added to enhance gamma-aminobutyric acid (GABA) production. To further improve the consumption of mixed monosaccharides, adaptation of Lactobacillus brevis KCL010 to high concentrations of mannitol improved the synbiotic fermentation efficiency of U. pinnatifida hydrolysates. Vegetated habitats play a key role in the structure and function of ecosystems by modifying local environmental conditions and providing food and habitat to a great range of organisms. The aim of this study is to assess the relative importance of two widely distributed macroalgae species in providing ecosystem functions in coastal areas where they are prevalent (Patagonia, Argentina, Lat 43.73°, Lon 64.92°) in comparison to low-vegetated areas without these two species. In October 2018 and May 2019, we tested whether Ulva spp. and Undaria pinnatifida perform a series of ecosystem functions commonly associated with vegetated coastal environments. Our study suggests that Ulva spp. decreases water current and provides habitat for epifauna, as the adjacent low-vegetated areas without Ulva spp. present higher gypsum block dissolution rates, lower biomass and richness of total epifauna, and lower grazer biomass than areas dominated by Ulva spp. U. pinnatifida, on the other hand, only showed a positive effect on epifauna biomass relative to low-vegetated areas, and no effect on the rest of the variables measured, despite providing similar levels of coverage to Ulva spp., and having a larger and more complex structure. None of the two macroalgae influence the habitat use by fishes. Despite smaller size and a labile structure, Ulva spp. presence had an effect on more variables than U. pinnatifida, and the function provided by U. pinnatifida was also provided by Ulva spp., namely U. pinnatifida did not provide new functions to the system. Macroalgae are one of the main producers in marine environments. However, only a few toxicity test methods have been established that use reference strains of macroalgae to evaluate the effects of chemicals on the growth and reproduction of macroalgae to monitor water quality. We selected reference strains of Chlorophyta, Ulva aragoënsis; Phaeophyceae, Ectocarpus siliculosus; and wakame, Undaria pinnatifida, as test species to establish a microplate-based method to investigate the toxicity of potassium dichromate, 3,5-dichlorophenol, and two common herbicides (diuron and simazine). We determined the growth of the three macroalgae in their early life stages and during the sporangia formation stage in E. siliculosus under laboratory conditions. We observed that the growth and sporangia formation in these algae were impaired in a dose-dependent manner. Additionally, we investigated the sensitivity of these macroalgae by comparing the toxicity values of toxicants used in this study with those obtained from a database. Compared to other microalgae and plant species, macroalgae showed a relatively high sensitivity to organic compounds, including herbicides. Growth tests using U. aragoënsis and E. siliculosus produced reliable results at 0–32 and 25–32 practical salinity units (PSU), respectively. The tests established in this study could test the toxicity of chemical substances in macroalgae and are thus expected to contribute to a better understanding of the environmental risks of chemical substances on aquatic biota. Global expansion of bivalve aquaculture can drive sustainable protein production. Inland culture of mussel spat can play an important role in supporting extensive mussel farming. Nursery culture of bivalves is, however, dependent on nutritious, cost‐efficient, and more reliable diets for spat. The aim of this study was to assess the impact of dietary alternatives to commercial algal feeds (Shellfish Diet 1800) on the survival and growth of Mytilus galloprovincialis spat, widely farmed in Europe. Spat (6.8 ± 1.1 mm) were supplied with different diets for 6 weeks: commercial microalgal diet (A), microencapsulated feeds containing a 1:1 blend of the macroalga Undaria pinnatifida and the microalga Schizochytrium (BioBullets; BB), or commercial microalgae and BioBullets combined (ABB). Unsupplemented spat showed no growth and little change in body condition (CI). Spat fed microcapsules grew at comparable rates, and body condition rose at higher levels (shell growth rates: 8.5 ± 3.7 μm day−1; ΔCI: 6.1 ± 1.1%) relative to those fed commercial microalgae (8.5 ± 5.7 μm day−1; ΔCI: 3.3 ± 0.8%). Supplementing microencapsulated feeds with the commercial microalgal diet did not significantly improve growth performances (9.3 ± 2.3 μm day−1; ΔCI: 4.7 ± 1.4%) relative to mussels fed microcapsules alone. Microencapsulated feeds for M. galloprovincialis spat production can significantly reduce nursery costs compared with commercial feeds or cultured microalgae. Background: Snakebite envenomation (SBE) causes diverse toxic effects in humans, including disability and death. Current antivenom therapies effectively prevent death but fail to block local tissue damage, leading to an increase in the severity of envenomation; thus, seeking alternative treatments is crucial. Methods: This study analyzed the potential of two fucoidan sulfated polysaccharides extracted from brown seaweeds Fucus vesiculosus (FVF) and Undaria pinnatifida (UPF) against the fibrinogen or plasma coagulation, proteolytic, and phospholipase A2 (PLA2) activities of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom. The toxicity of FVF and UPF was assessed by the hemocompatibility test. Results: FVF and UPF did not lyse human red blood cells. FVF and UPF inhibited the proteolytic activity of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom by approximately 25%, 50%, and 75%, respectively, while all venoms led to a 20% inhibition of PLA2 activity. UPF and FVF delayed plasma coagulation caused by the venoms of B. jararaca and B. neuwiedi but did not affect the activity of B. jararacussu venom. FVF and UPF blocked the coagulation of fibrinogen induced by all these Bothropic venoms. Conclusion: FVF and UPF may be of importance as adjuvants for SBE caused by species of Bothrops, which are the most medically relevant snakebite incidents in South America, especially Brazil.

Fucoidan refers to a group of sulphated polysaccharides obtained from brown seaweed, with numerous biological activities. In this study, fucoidan was fortified into Chinese steamed bread (CSB) at different concentrations (0, 1%, 3% and 5%) and the effect of fucoidan on the dough properties, structure properties and bioactivity were investigated. The results showed that fucoidan could change the viscosity of unfermented dough, and a high concentration of fucoidan could remove the free radicals produced by the SH–SS exchange reaction (GS-) in the dough, which significantly reduced the content of disulfide bond and reduced the expanded volume of fermented dough (p < 0.05). In addition, fucoidan forms a physical barrier on the surface of starch particles and hinders the reaction between protein-to-protein; therefore, fucoidan increased the hardness, gumminess and chewiness in CSB, and reduced the specific volume in CSB. Furthermore, the fucoidan-fortified CSB samples were found to have both the ability to significantly reduce the predicted glycemic index (pGI) (p < 0.05) and improve antioxidant activity (p < 0.05). Collectively, these findings could provide a theoretical basis for the applications of fucoidan as a functional component in fermented foods.

Undaria pinnatifida can effectively deal with organotin pollution through its excellent accumulation and degradation capabilities found under laboratory conditions. However, nothing is known regarding its accumulation, degradation performance, and related impact factors in the wild farming area. In this study, we monitored triphenyltin chloride (TPTCL) contents and degradation products in different algal parts (blades, stipes, sporophylls, and holdfasts) of cultivated U. pinnatifida from December 2018 to May 2019. Our results showed that sporophytes had an accumulation and degradation capacity for TPTCL. The TPTCL contents and degradation products varied with the algal growth stages and algal parts. TPTCL accumulated in the blades at the growth stage and the blades, stipes, sporophylls, and holdfasts at the mature stage. The TPTCL content among algal parts was blades (74.92 ± 2.52 μg kg−1) > holdfasts (62.59 ± 1.42 μg kg−1) > sporophylls (47.24 ± 1.41 μg kg−1) > stipes (35.53 ± 0.55 μg kg−1). The primary degradation product DPTCL accumulated only in the blades at any stage, with a concentration of 69.30 ± 3.89 μg kg−1. The secondary degradation product MPTCL accumulated in the blades at the growth stage and in the blades, stipe, and sporophyll at the mature stage. The MPTCL content among algal parts was blades (52.80 ± 3.48 μg kg−1) > sporophylls (31.08 ± 1.53 μg kg−1) > stipes (20.44 ± 0.85 μg kg−1). The accumulation pattern of TPTCL and its degradation products seems closely related to nutrient allocation in U. pinnatifida. These results provide the basis for applying cultivated U. pinnatifida in the bioremediation of organotin pollution and the food safety evaluation of edible algae.

This study analyzes the impact of the addition of three different types of algae (White Chlorella vulgaris, Himanthalia elongate (sea spaghetti), and Undaria pinnatifida (wakame)) at two concentrations (1.5% and 3%) on the sensory characteristics of caseless sausages (cevap) served at two different temperatures. The aim of this study was to compare the color, smell, taste, texture, juiciness, and overall acceptability of the reheated samples with those served at room temperature. The results show that cevap with wakame had the lowest scores, while white Chlorella had no significant effect on the sensory parameters compared to the control sample. The overall acceptability of the three cevap types was significantly higher when they were served warm than when served cold, while the color, taste, texture, and juiciness parameters were different for one of the tested samples, proving that cevap needs to be served warm. Species traits have been used extensively in invasion science, providing common metrics across taxa and ecosystems that enable comparisons based on the functional responses and effects of biota. However, most work on traits in invasion science has focused on terrestrial plants, despite the vulnerability of aquatic ecosystems to invasive species, such as invasive seaweeds. Research that focuses on individual species of invasive seaweeds has intensified in recent years, yet few studies have synthesised the information learned on species traits to identify commonalities or knowledge gaps in invasion science. Through a systematic review of 322 papers that investigate the traits of seaweed species from across the globe, here we ask - what are the trends and gaps in research that investigates traits of invasive seaweeds? To address this question, we aimed to: (1) identify publication rates and characteristics of the studies examining traits of invasive seaweeds; (2) clarify which and how many species have been investigated; and (3) assess which traits have been measured and how those traits have been used. Our review revealed that study regions for research on invasive seaweed traits were concentrated in Europe and North America. In addition, we found a total of 158 species that have been investigated, with a large proportion of studies (35%) focusing on just two species, Sargassum muticum and Undaria pinnatifida. Our study revealed that the most researched traits were morphological, which were used to address a wide range of research questions. Key research gaps included relatively few studies from Africa, Asia and South America, a lack of papers researching more than one species and a lack of measurements of biomechanical traits. Altogether, this review provides a thorough overview of research progress on species traits of invasive seaweeds and highlights the existing knowledge gaps that may lead to new ways in which the traits of invasive seaweeds can be used to answer important ecological questions. Fucoidans are discussed as antiviral agents, and fucoidan from Undaria pinnatifida (UpF), in particular has gained interest as potential food additive in antinoroviral strategies. As the competitive blocking activity of antinoroviral agents increases with the valency of terminal nonreducing fucose on the competitor, an effective processing of fucoidans to inhibitory oligosaccharides will depend on basic structural features of the polysaccharide. We demonstrate increased antiviral binding activity of processed low-mass UpF generated by hydrothermal degradation contrasting with decreased efficacy of low-mass fucoidan from Fucus vesiculosus. As this finding is in conflict with current structural models of UpF, we undertook a re-investigation of the glycan backbone in UpF. Applying solvolytical desulfation combined with enzymatic cleavage of low-mass fucoidan by endo-β6-galactanase and terminal labeling of oligosaccharides by deutero-reduction and bis-5-phenyl-3-methyl-1-pyrazolone (PMP) substitution, evidence from mass spectrometry and methylation linkage analysis of the oligosaccharides indicates that fucoses and galactoses in the glycan backbone are organized in homomeric blocks, where oligo-fucoses branch off from a galactane-type core: Fuc(1-3Fuc)n1-3[Gal(1-6Gal)n1-6]Gal(1-6Gal)n. Fucoxanthin (Fx) has been proven to exert numerous biological properties, which makes it an interesting molecule with diverse industrial applications. In this study, the kinetic behavior of Fx was studied to optimize three variables: time (t—3 min to 7 days), temperature (T—5 to 85 °C), and concentration of ethanol in water (S—50 to 100%, v/v), in order to obtain the best Fx yield from Undaria pinnatifida using conventional heat extraction. The Fx content (Y1) was found through HPLC-DAD and expressed in µg Fx/g of algae sample dry weight (AS dw). Furthermore, extraction yield (Y2) was also found through dry weight analysis and was expressed in mg extract (E)/g AS dw. The purity of the extracts (Y3) was found and expressed in mg Fx/g E dw. The optimal conditions selected for Y1 were T = 45 °C, S = 70%, and t = 66 min, obtaining ~5.24 mg Fx/g AS; for Y2 were T = 65 °C, S = 60%, and t = ~10 min, obtaining ~450 mg E/g AS; and for Y3 were T = 45 °C, S = 70%, and t = 45 min, obtaining ~12.3 mg Fx/g E. In addition, for the selected optimums, a full screening of pigments was performed by HPLC-DAD, while phenolics and flavonoids were quantified by spectrophotometric techniques and several biological properties were evaluated (namely, antioxidant, antimicrobial, and cholinesterase inhibitory activity). These results could be of interest for future applications in the food, cosmetic, or pharmaceutical industries, as they show the Fx kinetic behavior and could help reduce costs associated with energy and solvent consumption while maximizing the extraction yields. Bacterial and fungal infections are a challenging global problem due to the reported increasing resistance of pathogenic microorganisms to conventional antimicrobials. Nanomaterials are a promising strategy to fight infections caused by multidrug-resistant microbes. In this work, gold (Au@UP) and silver (Ag@UP) nanoparticles were produced for the first time by green synthesis using an aqueous extract of the invasive macroalgae Undaria pinnatifida (UP). The nanoparticles were characterized by a wide range of physicochemical techniques. Au@UP and Ag@UP demonstrated to be spherical and crystalline with an average size of 6.8 ± 1.0 nm and 14.1 ± 2.8 nm, respectively. Carbohydrates and proteins of the UP extract may participate in the synthesis and capping of the nanoparticles. The UP extract, Ag@UP, and Au@UP were assessed for their antimicrobial activity against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, and Candida auris. Ag@UP showed the highest antimicrobial activity with very low MIC and MBC values for all the tested bacteria, and Au@UP demonstrated to be very effective against biofilm-producing bacteria. The antifungal properties of both Ag@UP and Au@UP were remarkable, inhibiting hyphae formation. This study points towards a very promising biomedical exploitation of this invasive brown algae. This study aimed to investigate the effects of different relative humidities (%) on the microbial safety, antioxidant activity, ascorbic acid, fucoxanthin, and tocopherol contents of Undaria pinnatifida sporophyll powder (UPSP) stored for 4 weeks. The caking phenomenon did not occur in the 11–53% relative humidity conditions, but it did in the 69%, 81%, and 93% relative humidity conditions with caking index values of 88.30%, 99.75%, and 99.98%, respectively. The aerobic bacterial contents increased drastically in samples stored at 69–93% relative humidity. Ascorbic acid was unstable at high relative humidity, but fucoxanthin and tocopherol were more unstable at low relative humidity. Therefore, it was most stable at intermediate relative humidity. The 69% relative humidity sample had higher DPPH (12.57 g BHAE/kg), ABTS (4.87 g AAE/kg), and FRAP (4.60 g Fe (II)/kg) than the other samples. This study could be helpful for the storage and transport of UPSP under optimum relative humidity conditions, which can significantly prevent quality losses. [ABSTRACT FROM AUTHOR]

The spread of non‐indigenous and invasive seaweeds has increased worldwide, and their potential effects on native seaweeds have raised concern. Undaria pinnatifida is considered among the most prolific non‐indigenous species. This species has expanded rapidly in the Northeast Pacific, overlapping with native communities such as the iconic giant kelp forests (Macrocystis pyrifera). Canopy shading by giant kelp has been argued to be a limiting factor for the presence of U. pinnatifida in the understory, thus its invasiveness capacity. However, its physiological plasticity under light limitation remains unclear. In this work, we compared the physiology and growth of juvenile U. pinnatifida and M. pyrifera sporophytes transplanted to the understory of a giant kelp forest, to juveniles growing outside of the forest. Extreme low light availability compared to that outside (~0.2 and ~4.4 mol photon ⋅ m−2 ⋅ d−1, respectively) likely caused a "metabolic energy crisis" in U. pinnatifida, thus restricting its photoacclimation plasticity and nitrogen acquisition, ultimately reducing its growth. Despite M. pyrifera juveniles showing photoacclimatory responses (e.g., increases in photosynthetic efficiency and lower compensation irradiance, Ec), their physiological/vegetative status deteriorated similarly to U. pinnatifida, which explains the low recruitment inside the forest. For heteromorphic algae with alternating generations, the thallus and gametophyte phases are different morphologies in free‐living life history. The thalli are popular used as traditional vegetables and herbal drugs, whereas the gametophyte phases are little involved. To better understand the functional lipids in the gametophyte phase of three commercial kelps, Saccharina japonica, Undaria pinnatifida, and Costaria costata, the contents of total lipids (TLs), fatty acid (FA) profiles, and transcriptomic analysis were performed. For the studied kelps, the TL contents in gametophyte phase were always almost twice more than those in the thallus, and the kelp species, their life stage, and the gender were critical factors affecting lipid accumulation. The gametophyte phases of U. pinnatifida and C. costata were rich in essential FA C18:2 n − 6 and C18:3 n − 3. The S. japonica gametophyte phase contained abundant C20:5 n − 3 and C18:4 n − 3, possessed an ideal ratio of n − 6/n − 3 polyunsaturated fatty acid below 1.0, and was supported by the transcriptome data which showed that the key sjD12/15 (n − 3) gene of gametophyte partially upregulated than sporophyte. The results suggested that S. japonica gametophyte phase was the worthiest of further development and utilization as a functional food. Practical Application: It is the first report on the fatty acid characteristics of three gametophyte phases of Saccharina japonica, Undaria pinnatifida, and Costaria costata and find that the S. japonica was worthy of further development and utilization as a functional food owing to its satisfactory fatty acid composition. Several bioactive compounds that hold a potential interest in the food industry as phenolic compounds, polysaccharides, proteins and vitamins, among others, are present in seaweeds. Green extraction technologies are the preferred way to obtain these compounds. Pressurized hot water extraction, from 160 to 220 °C, was tested to achieve high yields of these components from the edible brown seaweed, Undaria pinnatifida. The maximum fucoidan content was recovered at 160 °C, while the phloroglucinol content and antioxidant activity were maximum at 220 °C. The possibility of encapsulating these bioactive fractions using mannitol was assessed. The highest production yield of the polymeric particles was found using the 220 °C fraction (close to 75%). In order to formulate microparticles with bioactive potential, several ratios of liquid phases were assessed, 3:1, 1:1 and 1:3 (w:w), using the liquid fractions obtained at 160 °C and 220 °C. The yield production was always above 67%, being in the 1:3 ratio (160 °C:220 °C) and close to 75%. The rheological results indicated that the presence of microparticles enhanced the apparent viscosity of the aqueous dispersions with non-Newtonian profiles, achieving the highest viscosity for those formulated with microparticles from 160 °C:200 °C (3:1). The clinical manifestation of primary hyperoxaluria includes hyperoxaluria and recurrent urinary calculi. In this study, an oxidative damage model was constructed based on oxalate damage to the human renal proximal tubular epithelial cells (HK-2), and a comparative study was carried out on four different sulfated levels of Undaria pinnatifida polysaccharides (UPP0, UPP1, UPP2, and UPP3 with sulfate group [–OSO3−] contents of 1.59%, 6.03%, 20.83%, and 36.39%, respectively) on the repair of oxidatively damaged HK-2 cells. The results showed that after repair by UPPs, cell viability was enhanced, healing ability was improved, the intracellular superoxide dismutase level and mitochondrial membrane potential were increased, malondialdehyde, reactive oxygen species, and intracellular Ca2+ levels were reduced, cellular autophagy was reduced; lysosomal integrity was improved, and cytoskeleton and cell morphology were restored. The ability of repaired cells to endocytose nano-calcium oxalate dihydrate crystals (nano−COD) was enhanced. The activity of UPPs was closely related to their –OSO3− content. A too high or too low –OSO3− content was not conducive to polysaccharide activity, and only UPP2 exhibited the best cell repair ability and strongest ability to promote the cell endocytosis of crystals. Macroalgae are an extensive resource for the obtention of bioactive compounds, mainly phenolic compounds, phlorotannins, and pigments. Fucoxanthin (Fx) is the most abundant pigment present in brown algae and has shown several useful bioactivities that can be used to fortify products in the food and cosmetic industries. Nevertheless, to date, there is still insufficient literature reporting on the extraction yield of Fx from U. pinnatifida species from green technologies. In this regard, the present study aims to optimize the extraction conditions to obtain the highest Fx yield from U. pinnatifida through emerging techniques, namely microwave-assisted extraction (MAE) and ultrasound-assisted extraction (UAE). These methods will be compared with the conventional methodologies of heat-assisted extraction (HAE) and Soxhlet-assisted extraction (SAE). According to our results, even though the extraction yield could be slightly higher when using MAE than UAE, the Fx concentration obtained from the alga was double when using UAE. Thus, the Fx ratio in the final extract reached values of 124.39 mg Fx/g E. However, the optimal conditions should also be considered since UAE needed 30 min to perform the extraction, whereas MAE was able to obtain 58.83 mg Fx/g E in only 3 min and 2 bar, meaning less energy expenditure and minimum cost function. To our knowledge, this study obtains the highest concentrations of Fx ever reported (58.83 mg Fx/g E for MAE and 124.39 mg Fx/g E for UAE), with low energy consumption and short times (3.00 min for MAE and 35.16 min for UAE). Objective: This study aimed to improve both the drying rate and quality of Undaria pinnatifida. Methods: Microwave vacuum drying experiment was carried out under different microwave power density, vacuum degree and pulse intermittent conditions. The thin layer drying model was established and the mass and heat transfer parameters were calculated. The drying process parameters were optimized by L9（34） orthogonal test with indicators of drying time, sensory quality, rehydration rate, energy consumption and material temperature. Results: Page model was found fit the drying kinetics well, and the change of drying kinetics showed obvious dependence on energy contribution of drying conditions. The effective moisture diffusion coefficient ranged from 2.38×10-9 to 1.49×10-8 m2/s and increased with the microwave power density. The enthalpy and entropy decreased, while the Gibbs free energy increased with the increase of drying temperature. The drying activation energy of U. pinnatifida was 4.51 kJ/mol. When the microwave power density was 4.5 W/g, with vacuum degree 80 kPa, the moisture content was reduced to 12.30% within 16 min, and the energy consumption was 0.04 kW·h/g H2O, with rehydration rate of 1 885.69% at an average drying temperature of 26.71 ℃. The leaves of U. pinnatifida were in dark green, flat and free of burning spots and bubbles. Conclusion: Microwave vacuum drying was a suitable method for drying U. pinnatifida with high efficiency, controllable quality. Most importantly, few difference between rehydrated specimen and fresh materials was found.

Fucoidanase is an unstable enzyme with high specificity that requires a large about of time to screen it from microorganisms. In this study, enzymatic hydrolysis was used to produce low-molecular-weight fucoidan from microorganisms via the degradation of high-molecular-weight fucoidan without damage to the sulfate esterification structure of oligosaccharide. The microbial strain HN-25 was isolated from sea mud and was made to undergo mutagenicity under ultraviolet light. Fucoidanase was extracted via ultrasonication and its enzymatic activity was improved via optimization of the ultrasonic conditions. The enzymatic properties and degradation efficiency of fucoidanase were characterized. The microbial strain HN-25 is a Gram-negative aerobic and rod-shaped-cell bacterium, and therefore was identified as Cobetia amphilecti via 16s rDNA. The results proved that fucoidanase is a hydrolytic enzyme with a molecular weight of 35 kDa and with high activity and stability at 30 °C and pH 8.0. The activity of fucoidanase was significantly enhanced by sodium and calcium ions and inhibited by a copper ion and ethylenediaminetetraacetate (EDTA). There was a significant decrease in the molecular weight of fucoidan after enzymatic hydrolysis. The low-molecular-weight fuicodan was divided into four fractions, mainly concentrated at F3 (20~10 kDa) and F4 (≤6 kDa). These consequences suggest that fucoidanase obtained from Cobetia amphilecti is stable and efficient and could be a good tool in the production of bioactive compounds. [ABSTRACT FROM AUTHOR]

Seaweeds are considered to be third-generation renewable biomasses, the comprehensive utilization of which has drawn increasing attention in recent years. A novel cold-active alginate lyase (VfAly7) was identified from Vibrio fortis and biochemically characterized for brown seaweed utilization. The alginate lyase gene was high-level expressed in Pichia pastoris, with an enzyme yield of 560 U/mL and a protein content of 9.8 mg/mL by high-cell density fermentation. The recombinant enzyme was most active at 30 °C and pH 7.5, respectively. VfAly7 was a bifunctional alginate lyase with both poly-guluronate and poly-mannuronate hydrolysis activities. On the basis of VfAly7, a bioconversion strategy for the utilization of brown seaweed (Undaria pinnatifida) was developed. The obtained AOSs showed stronger prebiotic activity towards tested probiotics when compared to that of commercial fructooligosaccharides (FOSs), while the obtained protein hydrolysates displayed strong xanthine oxidase inhibitory activity with IC50 of 3.3 mg/mL. Different from the traditional knowledge about kelp, three sexual phenotypes (female, male, and monoecious) exist in the haploid gametophytes of Undaria pinnatifida. However, the sex‐determining mechanisms remain unknown. Genetic linkage mapping is an efficient tool to identify sex‐linked regions. In the present study, we resequenced a segregating gametophyte family based on the male genome of U. pinnatifida. A high‐density genetic linkage map was constructed using 9887 SNPs, with an average distance of 0.41 cM between adjacent SNPs. On the basis of this genetic map and using the composite interval mapping method, we identified 62 SNPs significantly linked with the sexual phenotype. They were located at a position of 67.67 cM on the linkage group 23, corresponding to a physical range of 14.67 Mbp on the HiC_Scaffold_23 of the genome. Reanalysis of the previous specific length amplified fragment sequencing data according to the reference genome led to the identification of a sex‐linked genomic region that encompassed the above‐mentioned 14.67 Mbp region. Hence, this overlapped genomic range was likely the sex‐determining region. Within this region, 129 genes were retrieved and 39 of them were annotated with explicit function, including the potential male sex‐determining gene‐encoding high mobility group (HMG) domain protein. Relative expression analysis of the HMG gene showed that its expression was higher in male gametophytes during the vegetative phase and monoecious gametophytes during both the vegetative and gametogenesis phases, but significantly lower in male gametophytes during the gametogenesis phase.
