Non-HLA antibodies against heterogeneous targets on endothelial cells have been associated with allograft injuries. The endothelial cell crossmatch (ECXM) is used in the detection of non-HLA antibodies but remains non-discriminatory for specific antibody identification. The primary objective of this study was to delineate the specific non-HLA antibody signatures associated with ECXM positivity and to determine the correlation of ECXM status and non-HLA antibody signatures on allograft health. Serum specimens from 25 lung transplant recipients (LTRs) and 13 renal transplant recipients (RTRs) were collected as part of clinical evaluation, and testing for angiotensin II receptor type 1 (AT1R) and donor-specific MHC class I chain-related gene A (MICA) antibodies and ECXM was performed. Remnant sera were tested for non-HLA antibodies using the LABScreen™ Autoantibody (LSAUT) Group 1, 2, and 3 kits (One Lambda, Inc., Los Angeles, CA, USA). In both cohorts, the concordance of AT1R and MICA together or individually with ECXM+ status was poor (<0.7), suggesting the presence of other unaccounted antibodies. Autoantibody profiling revealed three distinct clusters targeting fibrotic products, cytoskeletal proteins, and cell signaling molecules. A comparative analysis of ECXM+ and ECXM− specimens identified nine and five differentially expressed antibodies in the LTR and RTR cohorts, respectively. Employing machine learning techniques (variable importance, feature selection, ROC-AUC), we derived a five-antibody panel (TNFα, collagen V, CXCL11, GDNF, GAPDH) and a two-antibody panel (TNFα, CXCL9) that effectively discriminated between ECXM+ and ECXM− status in the LTR and RTR cohorts, respectively. This paper explores 'near‐peer' gender and sexuality workshops taking place in UK secondary schools in support of the nationally required relationships and sex education curriculum. In line with UK law, these workshops promote respect, kindness and communication in a framework that explicitly validates LGBTQI+ identities, relationships and family structures. Based on interviews with 24 gender workshop facilitators, we explore the kind of practices that are needed to create spaces in which young people can openly reflect on their views about gender and sexuality, and advance the above‐noted goals of promoting respect, kindness and empathy for people of all genders and sexualities. We argue that gender workshops constitute an important and under‐explored space of 'everyday' activism. We further argue that through practices of atmosphere curation and the near‐peer relational dynamic between facilitators and participants, these workshops create valuable spaces for young people to critically reflect on their views and attitudes about gender and sexuality in a way that does not exist elsewhere. This work extends understanding about the kinds of spatial practices that might be needed to bring forth more positive gender cultures, and deepens our understanding about the experiences and functioning of 'everyday' work for social change that takes place outside of 'high profile' activist sites such as marches, demonstrations and protests. This paper explores and theorises education-based workshops delivered in secondary schools in support of a relationships and sex education curriculum that aims to bring forth more positive understandings and experiences of gender and sexuality. We cast this work as a form of sexual health activism, with our paper deepening understanding of how the motivations of those engaged in this form of activism interface with the decision to invest time in this work. Based on interviews with 40 workshop facilitators in England and Wales we argue that this form of sexual health activism is motivated by facilitators' life experiences as well as the desire to make the world a better place. As such, this form of work can function as a means of 'caring for' both past selves and future generations, thus functioning simultaneously as a form of self-care and a form of 'societal care work'. Ultimately, these activities may be understood as a form of 'extra-clinical' healthcare practice, with leading gender and sexual health workshops serving as an important means of solidifying health students' identities as both healthcare providers and activists for social change. Aim Bw4 and Bw6 are mutually exclusive HLA Class I “public” epitopes encoded by amino acids 77–83. We sought to explore selective reactivity patterns of sera with antigens which only differed for Bw4 or Bw6. Methods We tested 6726 sera from 2953 transplant candidates using Labscreen ® Single Antigen (One Lambda, Canoga Park, CA). We focused on target alleles B ∗ 27:05(Bw4) vs B ∗ 27:08(Bw6) and B ∗ 53:01(Bw4) vs B ∗ 35:01(Bw6). Reactions were called Positive (MFI > 3000) or Negative (MFI < 1000) and a correlation table constructed on the combinations of bead reactivity. The sera which had anti-Bw4 or anti-Bw6 based on selective reactivity within the above allele pairs were examined for reactivity with the other 96 assay beads bearing either Bw4 or Bw6. Results Table 1 shows the excellent correlation between the two sets of allele pairs we used to detect anti-Bw4/Bw6 antibody. Fig. 1 shows that the mean MFI against Bw4 or Bw6 on the other target alleles in the kit were strongly predicted based only on the deduction from the two B27 alleles. Similar predictive value was found for the B35/B53 allele pair. Conclusion Use of selective reactivity of sera with allele targets which only differ in the Bw4 or Bw6 epitopes is a definitive method for detecting such antibodies without presence of masking antibodies against private antigens.
