Abstract Gene expression profiling is an effective method for identifying predictive and prognostic biomarkers. However, measurements are prone to uncertainty and errors due to various pre-analytical variables. Systematic evaluating effects of these variables on gene expression measurements and relative expression orderings (REOs) of gene pairs, is necessary. A total of 18 datasets were collected, comprising over 800 paired samples. These paired samples were utilized to assess the impact of pre-analytical variables on gene expression measurements and REOs, including sampling methods, tumor sample heterogeneity, fixed time delays, preservation conditions, degradation levels, library preparation kits, amplification kits, RNA quantity, measuring platforms, and laboratory sites at single and multi-variable level. Low-quality samples served as the case group, while paired high-quality samples constituted the control group. In both single and multiple variable analyses, comparing each case sample to paired control sample revealed thousands of genes exhibited a twofold change in expression values. In contrast, on average, 82% and 76% of gene pairs keep consistent REO pattern between paired samples in single-variable and multi-variable analyses, respectively. Notably, the rate steadily increased after excluding gene pairs with the closest expression levels. Statistical analyses shown a higher proportion of differentially expressed genes (DEGs) than that of reversed gene pairs between case and control groups in both single-variable and multi-variable analyses. Furthermore, the proportion of reversal gene pairs among all gene pairs involving DEGs remained below 20% in the majority of comparisons. Our research demonstrates that REOs exhibit higher robustness under the influence of pre-analytical variables. These findings indicate the potential of the REOs-based approach in transcriptomics research and its applicability for biomarker studies.

Developing efficient catalysts is of great significance in improving the sluggish kinetics and high desorption temperature of MgH2 hydrogen storage material. Here, ultrathin NiTi-layered double hydroxide (NiTi-LDH) nanosheets are used as precursors to prepare Mg2Ni/TiH1.5 composite catalysts to improve the hydrogen storage properties of MgH2. The variation of Ni/Ti ratio in LDH plays an important role in regulating the composition, morphology and distribution of Mg2Ni/TiH1.5 catalysts, which significantly affect their synergistic catalytic effect. Mg2Ni/TiH1.5 composite catalyst exhibits significantly improved catalytic performance compared with conventional Ni-, Ti- and Ni/Ti-based catalysts. The optimal MgH2/Mg2Ni/TiH1.5 system shows a significantly reduced desorption temperature of 212 °C which is 133 °C lower than that of pure MgH2 (345 °C), and can release 5.97 wt% hydrogen within 300s at 300 °C. Further mechanism analysis reveals that the unique flaky morphology and suitable composition of Ni/Ti LDH can significantly enhance the synergistic effect of Mg2Ni and TiH1.5, which promotes the fracture of the HH and Mg-H bonds.

Abstract Seed coating with fungicides is a common practice in controlling seed-borne diseases, but conventional methods often result in high toxicity to plants and soil. In this study, a nanoparticle formulation was successfully developed using the metal–organic framework UiO-66 as a carrier of the fungicide ipconazole (IPC), with a tannic acid (TA)-ZnII coating serving as a protective layer. The IPC@UiO-66-TA-ZnII nanoparticles provided a controlled release, triggered and regulated by environmental factors such as pH and temperature. This formulation efficiently controlled the proliferation of Fusarium fujikuroi spores, with high penetration into both rice roots and fungal mycelia. The product exhibited high antifungal activity, achieving control efficacy rates of 84.09% to 93.10%, low biotoxicity, and promoted rice growth. Compared to the IPC flowable suspension formula, IPC@UiO-66-TA-ZnII improved the physicochemical properties and enzymatic activities in soil. Importantly, it showed potential for mitigating damage to beneficial soil bacteria. This study provides a promising approach for managing plant diseases using nanoscale fungicides in seed treatment. ObjectiveEndometriosis (EMs) is a chronic inflammatory disease characterized by the presence of endometrial tissue in the non-uterine cavity, resulting in dysmenorrhea, pelvic pain, and infertility. Epidemiologic data have suggested the correlation between EMs and recurrent pregnancy loss (RPL), but the pathological mechanism is unclear. This study aims to investigate the potential biomarkers and immune infiltration in EMs and RPL, providing a basis for early detection and treatment of the two diseases.MethodsTwo RPL and six EMs transcriptomic datasets from the Gene Expression Omnibus (GEO) database were used for differential analysis via limma package, followed by weighted gene co-expression network analysis (WGCNA) for key modules screening. Protein-protein interaction (PPI) network and two machine learning algorithms were applied to identify the common core genes in both diseases. The diagnostic capabilities of the core genes were assessed by receiver operating characteristic (ROC) curves. Moreover, immune cell infiltration was estimated using CIBERSORTx, and the Cancer Genome Atlas (TCGA) database was employed to elucidate the role of key genes in endometrial carcinoma (EC).Results26 common differentially expressed genes (DEGs) were screened in both diseases, three of which were identified as common core genes (MAN2A1, PAPSS1, RIBC2) through the combination of WGCNA, PPI network, and machine learning-based feature selection. The area under the curve (AUC) values generated by the ROC indicates excellent diagnostic powers in both EMs and RPL. The key genes were found to be significantly associated with the infiltration of several immune cells. Interestingly, MAN2A1 and RIBC2 may play a predominant role in the development and prognostic stratification of EC.ConclusionWe identified three key genes linking EMs and RPL, emphasizing the heterogeneity of immune infiltration in the occurrence of both diseases. These findings may provide new mechanistic insights or therapeutic targets for further research of EMs and RPL.

Abstract Background Most patients with severe asthma are sensitized to at least one allergen. Whether local immunoglobulin E (IgE) in induced sputum reflects asthma control status has not been investigated. Methods Patients with asthma were classified as well controlled, partly controlled, and uncontrolled asthma (UCA) according to Global Initiative for Asthma 2022 guidelines. Lung function and fractional exhaled nitric oxide (FeNO) were evaluated. Induced sputum was collected and total IgE and Phadiatop (IgE to common inhalant allergens) measurements were performed. General clinical characteristics and pulmonary inflammation indicators were analyzed between the three groups of asthmatic patients. Univariate and multifactor ordinal logistic regression were used to model the relationship between pulmonary inflammation indicators and asthma control status. The ability of sputum total IgE in identifying different levels of asthma control was assessed by receiver operating characteristic curve (ROC). Results Patients with UCA had worse lung function and airway inflammation as indicated by lower levels of forced expiratory volume in 1s (FEV1)%pred, FEV1/FVC, MEF75%pred, MEF50%pred and MEF25%pred, and higher levels of FeNO and sputum eosinophil% compared with the WCA group. In addition, higher levels of total sputum IgE and Phadiatop were found in patients with UCA than in patients with WCA and PCA. BackgroundEmerging evidence underscores the comorbidity mechanisms among autoimmune diseases (AIDs), with innovative technologies such as single-cell RNA sequencing (scRNA-seq) significantly advancing the explorations in this field. This study aimed to investigate the shared genes among three AIDs—Multiple Sclerosis (MS), Systemic Lupus Erythematosus (SLE), and Rheumatoid Arthritis (RA) using bioinformatics databases, and to identify potential biomarkers for early diagnosis.MethodsWe retrieved transcriptomic data of MS, SLE, and RA patients from public databases. Weighted Gene Co-Expression Network Analysis (WGCNA) was employed to construct gene co-expression networks and identify disease-associated modules. Functional enrichment analyses and Protein-Protein Interaction (PPI) network was constructed. We used machine learning algorithms to select candidate biomarkers and evaluate their diagnostic value. The Cibersort algorithm was and scRNA-seq analysis was performed to identify key gene expression patterns and assess the infiltration of immune cells in MS patients. Finally, the biomarkers’ expression was validated in human and mice experiments.ResultsSeveral shared genes among MS, SLE, and RA were identified, which play crucial roles in immune responses and inflammation regulation. PPI network analysis highlighted key hub genes, some of which were selected as candidate biomarkers through machine learning algorithms. Receiver Operating Characteristic (ROC) curve analysis indicated that some genes had high diagnostic value (Area Under the Curve, AUC >0.7). Immune cell infiltration pattern analysis showed significant differences in the expression of various immune cells in MS patients. scRNA-seq analysis revealed clusters of genes that were significantly upregulated in the single cells of cerebrospinal fluid in MS patients. The expression of shared genes was validated in the EAE mose model. Validation using clinical samples confirmed the expression of potential diagnostic biomarkers.ConclusionThis study identified shared genes among MS, SLE, and RA and proposed potential early diagnostic biomarkers. These genes are pivotal in regulating immune responses, providing new targets and theoretical basis for the early diagnosis and treatment of autoimmune diseases.

BackgroundAcupoint catgut embedding (ACE) is a traditional Chinese medicine technique commonly used for managing various disorders, including chronic inflammatory pain and allergic asthma. Despite its growing use, the neuroimmunological mechanisms underlying ACE treatment effects remain unclear.MethodsThis study investigated the roles and potential mechanisms of the effects of ACE in treating experimental autoimmune encephalomyelitis (EAE), a frequently used animal model of autoimmune neuroinflammation. The effects of ACE treatment were evaluated by monitoring body weight and EAE severity scores. Behavioral tests, histopathological analysis, ELISA, and flow cytometry were conducted to assess the therapeutic efficacy of ACE. RNA sequencing was performed to uncover ACE-associated transcriptional signatures in the spinal cords of EAE mice.ResultsThe results were validated through western blotting, qRT-PCR, and immunofluorescence (IF) staining. In ACE-treated mice, EAE disease severity was significantly ameliorated, along with improvements in anxiety-like behaviors and reduced inflammation and demyelination. The ACE treatment restored immune imbalance in the EAE mice by decreasing Th17 and Th1 cells, while increasing Treg cells in peripheral immune organs and reducing serum inflammatory cytokine levels. RNA sequencing revealed significant suppression of the genes and pathways associated with reactive microglial and astrocytic activation, corroborated by IF studies. Additionally, ACE treatment could suppress the ERK and JNK signaling pathways at both RNA and protein levels.ConclusionThese findings confirm the protective role of ACE in mitigating EAE symptoms by modulating microglial and astrocytic activity and regulating inflammatory cytokines.

ObjectiveThis study aimed to systematically evaluate the clinical efficacy of Jingjin (muscle region of the meridian, sinew/tendon/fascia) acupuncture therapy in treating peripheral facial paralysis.MethodsA computerized search of PubMed, EMBASE, Cochrane Central Register of Controlled Clinical Studies, SCOPUS, Web of Science, PEDro, China Knowledge, Wanfang, and Wipu databases was performed for published randomized controlled trials (RCTs) on the treatment of peripheral facial paralysis using Jingjin acupuncture therapy from the beginning of the construction of the databases until 2 April 2024. After a two-person independent extraction of data, the studies were assessed for paper quality and then analyzed for meta-analysis using RevMan5.4 software.ResultsA total of 19 randomized controlled trials involving 1,436 patients were included. Abstract Background To investigate the role of lncRNA LINC00665 in modulating ovarian cancer stemness and its influence on treatment resistance and cancer development. Methods We isolated ovarian cancer stem cells (OCSCs) from the COC1 cell line using a combination of chemotherapeutic agents and growth factors, and verified their stemness through western blotting and immunofluorescence for stem cell markers. Employing bioinformatics, we identified lncRNAs associated with ovarian cancer, with a focus on LINC00665 and its interaction with the CNBP mRNA. In situ hybridization, immunohistochemistry, and qPCR were utilized to examine their expression and localization, alongside functional assays to determine the effects of LINC00665 on CNBP. Results LINC00665 employs its Alu elements to interact with the 3’-UTR of CNBP mRNA, targeting it for degradation. This molecular crosstalk enhances stemness by promoting the STAU1-mediated decay of CNBP mRNA, thereby modulating the Wnt and Notch signaling cascades that are pivotal for maintaining CSC characteristics and driving tumor progression. These mechanistic insights were corroborated by a series of in vitro assays and validated in vivo using tumor xenograft models. Furthermore, we established a positive correlation between elevated CNBP levels and increased disease-free survival in patients with ovarian cancer, underscoring the prognostic value of CNBP in this context. Conclusions lncRNA LINC00665 enhances stemness in ovarian cancer by mediating the degradation of CNBP mRNA, thereby identifying LINC00665 as a potential therapeutic target to counteract drug resistance and tumor recurrence associated with CSCs.

Abstract Background Autosomal recessive polycystic kidney disease (ARPKD) is a rare inherited cystic disease characterized by bilateral renal cyst formation and congenital liver fibrosis. Cardiovascular disorders such as noncompaction of ventricular myocardium (NVM) have not been reported with ARPKD. Case presentation A 5-month-old girl was examined after presenting with a fever and turbid urine for one day and was diagnosed as urinary tract infection. Urinary ultrasound showed multiple round, small cysts varying in size in both kidneys. Genetic testing revealed two heterozygous mutations and one exon deletion in the polycystic kidney and hepatic disease 1 gene, indicating a diagnosis of ARPKD. During hospitalization, she was found to have chronic heart failure after respiratory tract infection, with an ejection fraction of 29% and fraction shortening of 13%. When the patient was 15 months old, it was found that she had prominent trabeculations and deep intertrabecular recesses with the appearance of blood flow from the ventricular cavity into the intertrabecular recesses by echocardiography. The noncompaction myocardium was 0.716 cm and compaction myocardium was 0.221 cm (N/C = 3.27), indicating a diagnosis of NVM. Liver and kidney function remained normal during four-year follow-up. Conclusions This is the first report of NVM in a patient with ARPKD. It is unsure if the coexistence of NVM and ARPKD is a coincidence or they are different manifestations of ciliary dysfunction in the heart and kidneys.

Citrinin (CTN) is a mycotoxin commonly found in contaminated foods and feed, posing health risks to both humans and animals. However, the mechanism by which CTN damages the intestine remains unclear. In this study, a model of intestinal injury was induced by administering 1.25 mg/kg and 5 mg/kg of CTN via gavage for 28 consecutive days in 6-week-old Kunming mice, aiming to explore the potential mechanisms underlying intestinal injury. The results demonstrate that CTN can cause structural damage to the mouse jejunum. Additionally, CTN reduces the protein expression of Claudin-1, Occludin, ZO-1, and MUC2, thereby disrupting the physical and chemical barriers of the intestine. Furthermore, exposure to CTN alters the structure of the intestinal microbiota in mice, thus compromising the intestinal microbial barrier. Meanwhile, the results showed that CTN exposure could induce excessive apoptosis in intestinal cells by altering the expression of proteins such as CHOP and GRP78 in the endoplasmic reticulum and Bax and Cyt c in mitochondria. The mitochondria and endoplasmic reticulum are connected through the mitochondria-associated endoplasmic reticulum membrane (MAM), which regulates the membrane. We found that the expression of bridging proteins Fis1 and BAP31 on the membrane was increased after CTN treatment, which would exacerbate the endoplasmic reticulum dysfunction, and could activate proteins such as Caspase-8 and Bid, thus further inducing apoptosis via the mitochondrial pathway. Taken together, these results suggest that CTN exposure can cause intestinal damage by disrupting the intestinal barrier and inducing excessive apoptosis in intestinal cells.

Citrinin (CTN) has been reported to induce renal failure and structural damage, but its nephrotoxic effects and mechanisms are not fully understood. Therefore, we established a model by orally administering CTN (0, 1.25, 5, or 20 mg/kg) to mice for 21 consecutive days. Histological and biochemical analyses revealed that CTN caused structural damage to renal tubules, increased inflammatory cell infiltration, and elevated levels of serum markers of renal function (creatinine, urea, and uric acid). Moreover, mRNA transcript levels of the inflammatory factors TNF-α, IL-1β, and IL-6 were increased, indicating the occurrence of an inflammatory response. Furthermore, exposure to CTN induced renal oxidative stress by decreasing antioxidant GSH levels, antioxidant enzyme (SOD, CAT) activities, and increasing oxidative products (ROS, MDA). In addition, CTN increased the expression of proteins associated with endoplasmic reticulum (ER)stress and apoptotic pathways. ER stress has been shown to be involved in regulating various models of kidney disease, but its role in CTN-induced renal injury has not been reported. We found that pretreatment with the ER stress inhibitor 4-PBA (240 mg/kg, ip) alleviated CTN-induced oxidative stress, NF-κB pathway mediated inflammatory response, and apoptosis. Interestingly, 4-PBA also partially alleviated renal structural damage and dysfunction. Thus, ER stress may be a novel target for the prevention and treatment of CTN-induced renal injury.

AimThe correlation between type 2 diabetes mellitus (T2DM) and the occurrence of liver fibrosis is well-established. However, the longitudinal association between body composition and liver fibrosis progression in patients with T2DM remains incompletely explored.MethodsTotal of 390 patients with T2DM underwent body composition assessments, followed by a median duration of 2.13 years. The calculated parameters included body mass index (BMI), fat mass index (FMI), trunk fat mass index (TFMI), appendicular skeletal muscle mass index (ASMI), muscle/fat mass ratio (M/F) and appendicular skeletal muscle mass/trunk fat mass ratio (A/T). Liver fibrosis was evaluated through liver stiffness measurement (LSM). Patients were classified according to BMI and body composition, followed by a comprehensive investigation into the impact of body composition changes on liver fibrosis outcomes.ResultsAmong 72 patients with incident advanced liver fibrosis at readmission, ΔBMI, ΔFMI and ΔTFMI increased, while ΔM/F and ΔA/T decreased. Individuals who kept obese had a dramatically elevated hazard of incident advanced liver fibrosis compared to those who kept non-obese, with an adjusted odds ratio of 3.464. When TFMI heightened, the hazard of incident advanced liver fibrosis was 3.601 times higher compared to the decreased group. Additionally, individuals in increased ASMI and A/T groups showed a slight advantage in preventing incident advanced liver fibrosis compared to the stable groups.ConclusionStable obesity was associated with a greater hazard of liver fibrosis advancement, and an increase in TFMI may promote the progression of liver fibrosis. Maintaining a balanced muscle/fat ratio appeared to help prevent the progression.

With rapidly increasing levels of renewable energy penetration, flexibility resources play an ever more critical role in the future power system. This paper describes a two-stage stochastic mixed integer second order cone (MISOC) method for flexibility resource planning (VFRP) considering source-grid-load-storage interactions. Considering renewable energy penetration levels and flexibility resource characteristics, this study quantifies the extent to which the share of renewable energy generation affects cost performance, and assesses the priority and potential of investing in flexibility resources. Moreover, the multi-type flexibility resource portfolio allocation method and energy storage only approach are compared in terms of socio-economic benefits. A multi-cut Benders-based approach is used for decomposing large-scale stochastic expansion planning problems to reduce the computational burden. Numerical simulation tests on the modified energy test systems I39G20 and F213G20 verified the correctness and effectiveness of the proposed flexible resource planning models and solution algorithms. The simulation results show that retrofit of coal-fired units for better flexibility has a higher investment return. Still, energy storage presents a tremendous investment prospect and potential. Finally, the multi-type flexibility resource portfolio allocation method is more cost-effective than an energy storage only approach.

Diatoms (> 2 μm) have traditionally been identified as the primary biological agents linking the carbon (C) and silicon (Si) cycles. However, recent research has shown that picophytoplankton species (< 2 μm) also play a crucial role in the intertwined Si-C biogeochemical cycling in marine ecosystems. In this study, we examined the spatial distribution and vertical variation of micro/nano-diatoms and picophytoplankton in the eastern Indian Ocean (EIO), aimed to differentiate the contributions of living carbon and biogenic silica (bSi) stocks between diatoms and picophytoplankton. The study revealed that the abundance of picophytoplankton surpasses that of diatoms by four to seven orders of magnitude. Synechococcus was predominantly presented in the upper layer, while Prochlorococcus, picoeukaryotes, and diatoms were primarily located in the middle layer. Aggregation Boosted Tree (ABT) and Generalized Additive Models (GAM) analyses revealed that temperature and silicate (DSi) levels strongly influenced the diatom and picophytoplankton communities in the EIO. The fractions smaller than 2 μm made substantial contributions of 86.20%, 55.69%, and 96.86% to chlorophyll a (Chl-a), particulate organic carbon (POC), and living carbon biomass, respectively, underscoring the ecological significance of picophytoplankton in the carbon cycle of oligotrophic regions. Picophytoplankton represented a 33.06% of bSi stocks in the area, comparable to the contribution of diatoms (> 20 μm). Moreover, estimated contributions of diatom living carbon and silicon quota averaged 0.47% and 0.66%, respectively, while that of Synechococcus stood at 2.58% and 1.77%, indicating the predominance of Synechococcus as a weakly siliceous organism with high cell abundance in oligotrophic seas. Overall, this study draws on data from diatom and picophytoplankton biomass in the EIO to offer insights into the disproportionate carbon and silicon budgets in oligotrophic oceans from a biological perspective.

Abstract The excellent dielectric properties and tunable structural design of metal sulfides have attracted considerable interest in realizing electromagnetic wave (EMW) absorption. However, compared with traditional monometallic and bimetallic sulfides that are extensively studied, the unique physical characteristics of solid‐solution‐type sulfides in response to EMW have not been revealed yet. Herein, a unique method for preparing high‐purity solid‐solution‐type sulfides is proposed based on solid‐phase in situ exsolution of different metal ions from hybrid precursors. Utilizing CoAl‐LDH/MIL‐88A composite as a precursor, Fe0.8Co0.2S single‐phase nanoparticles are uniformly in situ formed on an amorphous substrate (denoted as CoAl), forming CoAl/Fe0.8Co0.2S heterostructure. Combing with density functional theory (DFT) calculations and wave absorption simulations, it is revealed that Fe0.8Co0.2S solid solution has stronger intracrystal polarization and electronic conductivity than traditional monometallic and bimetallic sulfides, which lead to higher dielectric properties in EM field. Therefore, CoAl/Fe0.8Co0.2S heterostructure exhibits significantly enhanced EMW absorption ability in the low‐frequency region (2–6 GHz) and can achieve frequency screening by selectively absorbing EMW of specific frequency. This work not only provides a unique method for preparing high‐purity solid‐solution‐type sulfides but also fundamentally reveals the physical essence of their excellent EMW absorption performance.

ObjectiveTo study the association between the concentration of calcium, iron, zinc, copper, and magnesium and their ratios in whole blood and the risk of recurrent pneumonia in infants and young children, and to provide guidelines for the prevention and control of recurrent pneumonia in the population. MethodsA case-control study was conducted among 86 children (< 36 months of age) with recurrent pneumonia who were hospitalized between September 2018 and February 2019, and 174 age-matched healthy children who underwent physical examinations during the same period at Tongshan County People's Hospital in Hubei province. Relevant information on the participants was obtained from the hospital's electronic medical records, and venous blood samples were collected to measure the concentrations of the five metal elements and their ratios. Multivariate logistic regression analysis was used to assess risk factors for recurrent pneumonia in infants and young children; and receiver operating cure (ROC) analysis was used to determine the significance of ratios of mental elements in predicting recurrent pneumonia. ResultsThe cases had a lower calcium concentration than the controls (1.59 ± 0.22 vs. 1.66 ± 0.17 mmol/L), with a deficiency rate of 22.09%, but a higher copper concentration (17.62 ± 6.18 vs. 14.11 ± 2.97 μmol/L) and a higher magnesium/calcium ratio (0.95 ± 0.14 vs. 0.90 ± 0.12) and copper/zinc ratio (0.25 ± 0.07 vs. 0.20 ± 0.04). Multivariate logistic regression analysis showed that low blood calcium (odds ratio [OR] = 0.120, 95% confidence interval [CI] : 0.027 to 0.530), high blood copper (OR = 1.209, 95%CI: 1.127 to 1.298), high magnesium/calcium ratio (OR = 1.039, 95%CI: 1.015 to 1.063), and high copper/zinc ratio (OR = 1.216, 95%CI: 1.137 to 1.301) were associated with an increased risk of recurrent pneumonia in infants and young children. ROC curve analysis showed that the area under the curve (AUC) of magnesium/calcium ratio, copper/zinc ratio, and their combination in predicting recurrent pneumonia in infants were 0.608, 0.707, and 0.758, respectively (P < 0.05 for all). ConclusionThe children less than 36 months of age with recurrent pneumonia have reduced whole blood calcium but increased calcium deficiency rate, copper concentration, magnesium/calcium ratio, and copper/zinc ratio, and magnesium/calcium and copper/zinc ratios could predict the risk of recurrent pneumonia in the children to some extent.

A dysphagia diet is a special dietary programme. The development and design of foods for dysphagia should consider both swallowing safety and food nutritional quality. In this study, we investigated the rheological properties (viscosity, thixotropy, and viscoelasticity), textural properties, and swallowing behaviour of commercially available natural, pregelatinised, acetylated, and phosphorylated maize starch and tapioca starch. The results showed that all the samples belonged to food grade 3 in the framework of the International Dysphagia Dietary Standardization Initiative (IDDSI) and exhibited shear-thinning behaviour in favour of dysphagia patients, except for the sample containing pregelatinised starch, which was grade 2. Rheological tests showed that the samples had good structural recovery properties. At the same starch concentration, the elastic modulus of phosphorylated cassava starch FSMP was significantly greater than that of the starch solution, whereas that of acetylated starch was significantly less than that of the starch solution, and the combination of acetylated starch and protein led to a significant viscosity reduction phenomenon, resulting in FSMPs with good stability and fluidity; this may provide an opportunity for the incorporation of more high-energy substructures. The textural results showed that all the samples possessed textural properties of low hardness, low adhesion, and high cohesion, all of which could be used as food for dysphagia patients. This study may provide a theoretical basis for the creation and design of novel nutritional foods for dysphagia.

BackgroundThyroid metastasis represents a rare occurrence, with commonly observed primary tumors originating from renal cell carcinoma, malignant neoplasms of the gastrointestinal tract, lungs, and breast. However, the metastasis of hepatocellular carcinoma to the thyroid gland remains infrequent. Previous investigations have consistently demonstrated an unfavorable prognosis for patients with malignancies that have metastasized to the thyroid. In this context, we present a noteworthy case of thyroid metastasis from hepatocellular carcinoma (HCC), characterized by a distinct ultrasonographic manifestation of diffuse thyroid lesion, deviating from the previously documented imaging presentations of thyroid metastases in HCC.Case presentationA 62-year-old Chinese female patient was diagnosed with hepatocellular liver cancer in 2019, following which she underwent a radical hepatic resection. Pathological examination revealed HCC located in the right lobe (stage T3bN0M0 IIIB). No additional interventions were administered subsequent to the surgery. After a span of 15 months, the patient presented with dyspnea. Ultrasonographic findings showed diffuse solid infiltration within the thyroid gland, along with tumor thrombi in both internal jugular veins. Computed tomography (CT) scans demonstrated malignant thyroid lesions infiltrating the retropharyngeal space, prevertebral space, and esophageal wall. The subsequent pathology report from the puncture biopsy confirmed the malignant nature of the tumor, and immunohistochemical analysis definitively established its hepatic origin.ConclusionsPatients with a history of HCC should be subjected to long-term monitoring and habitual thyroid ultrasonography. Newly detected thyroid nodules in such patients should be immediately regarded suspect for potential metastatic disease. Even when a nodule doesn’t exhibit malignant characteristics on ultrasound, FNAB should be administered promptly to elucidate the pathological condition. Larger, swiftly multiplying thyroid masses should warrant an immediate CNB. The identification of thyroid metastases, particularly when coupled with peripheral tissue invasion, typically signifies a bleak prognosis.

Abstract Objective Our current study aimed to assess the relationship between TNF-related apoptosis-inducing ligand (TRAIL) and ferroptosis in non-small cell lung cancer (NSCLC) development. Methods The expression of TRAIL was detected by western blot, RT-qRCR and immunohistochemistry. The viability of NSCLC cells was analyzed by CCK-8 kit. The migration and invasion of NSCLC cells were detected by wound healing assay and transwell assay, respectively. Labile iron pool (LIP) was detected based on the calcein-acetoxymethyl ester method. Ferrous iron (Fe2+) and iron levels were assessed by detection kits. The levels of superoxide dismutase (SOD), catalase (CAT), and malondialdehyde (MDA) were measured using corresponding detection kits. Mice tumor xenograft models were used for the in vivo research. Results The expression of TRAIL was reduced in H1299, NCL-H1395, and A549 cells compared with BEAS-2B cells. The up-regulation of TRAIL expression significantly reduced cell viability, invasion, and migration of H1299 and A549 cells. TRAIL reduced the expression of ferroptosis-related genes (FTH1, GPX4, and SLC7A11), increased the levels of LIP, iron, and Fe2+, and promoted lipid peroxidation, thereby predisposing NSCLC cells to ferroptosis. TRAIL up-regulated the expression of phosphate modification of ASK-1 and JNK. ASKI-1 inhibitor GS-4977 attenuated the effects of TRAIL on the viability, migration, invasion, and ferroptosis of H1299 cells. Furthermore, TRAIL further suppressed tumor growth and ferroptosis in mice tumor xenograft models. Conclusion We indicated that overexpression of TRAIL induced ferroptosis in NSCLC cells and exerted anti-tumor effects. Mechanistically, TRAIL promoted ferroptosis by the activation of the ASK-1/JNK1 pathway. Our results may provide new therapeutic strategies for NSCLC.

Abstract Hydrogen-isotope storage materials are essential for the controlled nuclear fusion. However, the currently used smelting-ZrCo alloy suffers from rapid degradation of performance due to severe disproportionation. Here, we reveal a defect-derived disproportionation mechanism and report a nano-single-crystal strategy to solve ZrCo’s problems. Single-crystal nano-ZrCo is synthesized by a wet-chemistry method and exhibits excellent comprehensive hydrogen-isotope storage performances, including ultrafast uptake/release kinetics, high anti-disproportionation ability, and stable cycling, far superior to conventional smelting-ZrCo. Especially, a further incorporation of Ti into nano-ZrCo can almost suppress the disproportionation reaction. Moreover, a mathematical relationship between dehydrogenation temperature and ZrCo particle size is established. Additionally, a microwave method capable of nondestructively detecting the hydrogen storage state of ZrCo is developed. The proposed disproportionation mechanism and anti-disproportionation strategy will be instructive for other materials with similar problems.

Available polarization-difference imaging techniques face serious challenges in imaging speed and application range. To address these issues, this paper proposes an underwater dynamic polarization-difference imaging method with greater applicability. First, the intensity distribution of backscattered light is estimated via the Stokes vector. Afterward, the differential operation between the total intensity of light and the amplified estimation result of backscattered light makes clear imaging immediately accessible. Regardless of the movement states and polarization characteristics of the target, experimental results consistently demonstrate that the backscattered light can be eliminated to a great extent, and imaging quality and applicability are significantly enhanced. Meanwhile, the proposed method is immune to unexpected factors such as uneven illumination and has good stability. More importantly, there are also apparent advantages in terms of imaging time.

Sinapine is a natural polyphenol from the cruciferous plant family that has anti-aging effects but is low in bioavailability. To improve the bioavailability and therapeutic effect of sinapine, sinapine-crosslinked dual-protein-based hydrogels were prepared using soy protein isolate as a cross-linking agent. The preparation conditions were optimized by single-factor experiments, and the optimal ratios were obtained as follows: the concentration of sinapine was 300 μg/mL; the water–oil ratio was 1:3. The encapsulation rate was greater than 95%, and the drug loading capacity was 3.5 mg/g. In vitro, digestion experiments showed that the dual-protein-based hydrogels as a drug carrier stabilized the release of sinapine and improved the bioavailability of sinapine by 19.3%. The IC50 of DPPH antioxidants was 25 μg/mL as determined by in vitro digestion, and the antioxidant capacity of ABTS was about 20% higher than that of glutaraldehyde control. This is due to the addition of sinapine to enhance the antioxidant properties of the system. It can be seen that the developed hydrogels have potential applications in related fields, such as food nutrition fortification and drug delivery.

Enzymatic hydrolysis preparation process of angiotensin-I converting enzyme (ACE) inhibitory peptides from Antarctic krill (Euphausia superba) was optimized and its stability was evaluated. Using the degreased Antarctic krill meal as the substrate and the ACE inhibition rate of enzymatic hydrolysates as the evaluation index, alkaline protease was selected as the optimal protease for preparing ACE inhibitory peptides from Antarctic krill among six types of proteases. The optimum enzymatic hydrolysis conditions were determined through single factor and response surface experiments, which were finally confirmed as follows: Enzymatic hydrolysis time of 3.4 h, material liquid ratio of 1:7 (g/mL) and enzyme dosage of 1.6%. Under these conditions, the ACE inhibition rate of enzymatic hydrolysates was 74.37%±0.87%. The obtained ACE inhibitory peptides exhibited good thermal stability at temperatures ranging from 20 ℃ to 100 ℃ and were relatively stable under neutral and weak alkaline conditions. BackgroundPerfluoroalkyl and polyfluoroalkyl substances (PFASs) exposure may affect male reproductive health. There are regional differences in PFASs exposure levels among men of childbearing age in China, and current research results on associated influencing factors are inconsistent. ObjectiveTo investigate the levels of PFASs in serum and their determinants among men of childbearing age. MethodsThe participants (n=113, 22-45 years old) were from a cross-sectional study of exposure to environmental pollutants and male reproductive health damage in Hubei Province conducted from April to June 2013 at the Reproductive Medicine Center of Tongji Hospital, Wuhan, Hubei Province. Eleven kinds of PFASs were measured in serum by isotopic dilution-high performance liquid chromatography-tandem mass spectrometry. The included PFASs were prefluorohexanoic acid (PFHxA), perfluoroheptanoic acid (PFHpA), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoroundecanoic acid (PFUdA), perfluorododecanoic acid (PFDoA), perfluorotridecanoic acid (PFTrDA), perfluorotetradecanoic acid (PFTeDA), perfluorohexane sulfonate (PFHxS), and perfluorooctane sulfonate (PFOS). Information about participants' demographic characteristics, lifestyle, and habits was collected by a set of self-designed questionnaires. The associations of demographic characteristics, lifestyle, and habits with exposure to PFASs were analyzed by linear regression. ResultsThe major components of PFASs were PFOS and PFOA, and the concentrations expressed as M (P25, P75) were 8.31 (4.90, 17.79) ng·mL−1 and 2.77 (2.18, 3.46) ng·mL−1, respectively. The positive rates of six PFASs (PFOA, PFNA, PFDA, PFUdA, PFHxS, and PFOS) were 100%, followed by PFDoA and PFTrDA (87.61% and 88.59%, respectively). Abstract Metal–organic frameworks (MOFs) are promising candidates for room‐temperature hydrogen storage materials after modification, thanks to their ability to chemisorb hydrogen. However, the hydrogen adsorption strength of these modified MOFs remains insufficient to meet the capacity and safety requirements of hydrogen storage systems. To address this challenge, a highly defective framework material known as de‐MgMOF is prepared by gently annealing Mg‐MOF‐74. This material retains some of the crystal properties of the original Mg‐MOF‐74 and exhibits exceptional hydrogen storage capacity at above‐ambient temperatures. The MgO5 knots around linker vacancies in de‐MgMOF can adsorb a significant amount of dissociated and nondissociated hydrogen, with adsorption enthalpies ranging from −22.7 to −43.6 kJ mol−1, indicating a strong chemisorption interaction. By leveraging a spillover catalyst of Pt, the material achieves a reversible hydrogen storage capacity of 2.55 wt.% at 160 °C and 81 bar. Additionally, this material offers rapid hydrogen uptake/release, stable cycling, and convenient storage capabilities. A comprehensive techno‐economic analysis demonstrates that this material outperforms many other hydrogen storage materials at the system level for on‐board applications.

Persister cells are transiently tolerant to antibiotics and are associated with recalcitrant chronic infections due to recolonization of host cells after antibiotic removal. Brucella spp. are facultative pathogens that establish intracellular infection cycles in host cells which results in chronic persistent infections. Brucella abortus forms multi-drug persister cells which are promoted by the (p)ppGpp synthetase Rsh during rifampicin exposure. Here, we confirmed that Rsh promoted persister cells formation in B. abortus stationary phase treated with rifampicin and enrofloxacin. Deletion of the gene for Rsh decreased persister cells level in the presence of these drugs in different growth phases. However, persister cells formation by deletion strain varied in different growth phases in the presence of other antibiotics. Rsh also was involved in persister cells formation during rifampicin treatment under certain stress conditions, including acidic conditions, exposure to PBS, and heat stress. Moreover, Rsh impacted persister cell levels during rifampicin or enrofloxacin treatment in RAW264.7 macrophages. Certain typeIItoxin-antitoxin modules were upregulated under various stress conditions in B. abortus. We established that Rsh positively regulated the type II toxin-antitoxin mbcTA. Moreover, rifampicin-tolerant persister cells formation was elevated and ATP levels were decreased when mbcTA promoter was overexpressed in Rsh deletion background in stationary phase. Our results establish that (p)ppGpp synthetase Rsh plays a key role in B. abortus persistence and may serve as a potent novel target in combination with rifampicin in the development of new therapeutic approaches and prevention strategies to treat chronic infections of Brucella.

Abstract Background Previous studies have shown that IL-25 levels are increased in patients with asthma with fixed airflow limitation (FAL). However, the mechanism by which IL-25 contributes to airway remodeling and FAL remains unclear. Here, we hypothesized that IL-25 facilitates pro-fibrotic phenotypic changes in bronchial epithelial cells (BECs) and circulating fibrocytes (CFs), orchestrates pathological crosstalk from BECs to CFs, and thereby contributes to airway remodeling and FAL. Methods Fibrocytes from asthmatic patients with FAL and chronic asthma murine models were detected using flow cytometry, multiplex staining and multispectral imaging analysis. The effect of IL-25 on BECs and CFs and on the crosstalk between BECs and CFs was determined using cell culture and co-culture systems. Results We found that asthmatic patients with FAL had higher numbers of IL-25 receptor (i.e., IL-17RB)+-CFs, which were negatively correlated with forced expiratory volume in 1 s/forced vital capacity (FEV1/FVC). The number of airway IL-17RB+-fibrocytes was significantly increased in ovalbumin (OVA)- and IL-25-induced asthmatic mice versus the control subjects. BECs stimulated with IL-25 exhibited an epithelial-mesenchymal transition (EMT)-like phenotypic changes. CFs stimulated with IL-25 produced high levels of extracellular matrix (ECM) proteins and connective tissue growth factors (CTGF). These profibrotic effects of IL-25 were partially blocked by the PI3K-AKT inhibitor LY294002. In the cell co-culture system, OVA-challenged BECs facilitated the migration and expression of ECM proteins and CTGF in CFs, which were markedly blocked using an anti-IL-17RB antibody. Conclusion These results suggest that IL-25 may serve as a potential therapeutic target for asthmatic patients with FAL.

Abstract Introduction The overall survival of patients with mesothelioma is poor and heterogeneous. At present, the prediction model for Chinese patients needs to be improved. We sought to investigate predictors of survival in malignant pleural mesothelioma and develop prognostic prediction models. Methods This Two-center retrospective cohort study recruited patients with pathologically diagnosed mesothelioma at Beijing Chao-Yang Hospital and Beijing Tong-Ren Hospital. We developed a new prognostic prediction model based on COX multivariable analysis using data from patients who were recruited from June 1, 2010 to July 1, 2021 in Beijing Chao-Yang Hospital (n = 95, development cohort) and validated this model using data from patients recruited from July 18, 2014 to May 9, 2022 in Beijing Tong-Ren Hospital (n = 23, validation cohort). Receiver operating characteristic analysis was used to estimate model accuracy. Results The parameters in this new model included PLT > 289.5(10^9/L) (1 point), Lymphocyte > 1.785(10^9/L) (-1point), Age > 73 years old (1 point), Calcium > 2.145(mmol/L) (-1point), Eastern Cooperative Oncology Group performance status (ECOG PS) > 2 (2 points). Abstract NEDD4 family represent an important group of E3 ligases, which regulate various cellular pathways of cell proliferation, cell junction and inflammation. Emerging evidence suggested that NEDD4 family members participate in the initiation and development of tumor. In this study, we systematically investigated the molecular alterations as well as the clinical relevance regarding NEDD4 family genes in 33 cancer types. Finally, we found that NEDD4 members showed increased expression in pancreas cancer and decreased expression in thyroid cancer. NEDD4 E3 ligase family genes had an average mutation frequency in the range of 0-32.1%, of which HECW1 and HECW2 demonstrated relatively high mutation rate. Breast cancer harbors large amount of NEDD4 copy number amplification. NEDD4 family members interacted proteins were enriched in various pathways including p53, Akt, apoptosis and autophagy, which were confirmed by further western blot and flow cytometric analysis in A549 and H1299 lung cancer cells. In addition, expression of NEDD4 family genes were associated with survival of cancer patients. Our findings provide novel insight into the effect of NEDD4 E3 ligase genes on cancer progression and treatment in the future.

Despite being indispensable devices in the electronic manufacturing industry, printed circuit boards (PCBs) may develop various soldering defects in the production process, which seriously affect the product’s quality. Due to the substantial background interference in the soldering defect image and the small and irregular shapes of the defects, the accurate segmentation of soldering defects is a challenging task. To address this issue, a method to improve the encoder–decoder network structure of UNet is proposed for PCB soldering defect segmentation. To enhance the feature extraction capabilities of the encoder and focus more on deeper features, VGG16 is employed as the network encoder. Moreover, a hybrid attention module called the DHAM, which combines channel attention and dynamic spatial attention, is proposed to reduce the background interference in images and direct the model’s focus more toward defect areas. Additionally, based on GSConv, the RGSM is introduced and applied in the decoder to enhance the model’s feature fusion capabilities and improve the segmentation accuracy. The experiments demonstrate that the proposed method can effectively improve the segmentation accuracy for PCB soldering defects, achieving an mIoU of 81.74% and mPA of 87.33%, while maintaining a relatively low number of model parameters at only 22.13 M and achieving an FPS of 30.16, thus meeting the real-time detection speed requirements.

BackgroundMachine learning is a potentially effective method for predicting the response to platinum-based treatment for ovarian cancer. However, the predictive performance of various machine learning methods and variables is still a matter of controversy and debate. ObjectiveThis study aims to systematically review relevant literature on the predictive value of machine learning for platinum-based chemotherapy responses in patients with ovarian cancer. MethodsFollowing the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines, we systematically searched the PubMed, Embase, Web of Science, and Cochrane databases for relevant studies on predictive models for platinum-based therapies for the treatment of ovarian cancer published before April 26, 2023. The Prediction Model Risk of Bias Assessment tool was used to evaluate the risk of bias in the included articles. Concordance index (C-index), sensitivity, and specificity were used to evaluate the performance of the prediction models to investigate the predictive value of machine learning for platinum chemotherapy responses in patients with ovarian cancer. ResultsA total of 1749 articles were examined, and 19 of them involving 39 models were eligible for this study. The most commonly used modeling methods were logistic regression (16/39, 41%), Extreme Gradient Boosting (4/39, 10%), and support vector machine (4/39, 10%). The training cohort reported C-index in 39 predictive models, with a pooled value of 0.806; the validation cohort reported C-index in 12 predictive models, with a pooled value of 0.831. Support vector machine performed well in both the training and validation cohorts, with a C-index of 0.942 and 0.879, respectively. The pooled sensitivity was 0.890, and the pooled specificity was 0.790 in the training cohort. ConclusionsMachine learning can effectively predict how patients with ovarian cancer respond to platinum-based chemotherapy and may provide a reference for the development or updating of subsequent scoring systems.

Introduction: The present study presents the development and validation of a clinical prediction model using random survival forest (RSF) and stepwise Cox regression, aiming to predict the probability of pelvic inflammatory disease (PID) progressing to sepsis. Methods: A retrospective cohort study was conducted, gathering clinical data of patients diagnosed with PID between 2008 and 2019 from the Medical Information Mart for Intensive Care (MIMIC)-IV database. Patients who met the Sepsis 3.0 diagnostic criteria were selected, with sepsis as the outcome. Univariate Cox regression and stepwise Cox regression were used to screen variables for constructing a nomogram. Moreover, an RSF model was created using machine learning algorithms. To verify the model's performance, a calibration curve, decision curve analysis (DCA), and receiver operating characteristic (ROC) curve were utilized. Furthermore, the capabilities of the two models for estimating the incidence of sepsis in PID patients within 3 and 7 days were compared. Results: A total of 1064 PID patients were included, of whom 54 had progressed to sepsis. The established nomogram highlighted dialysis, reduced platelet (PLT) counts, history of pneumonia, medication of glucocorticoids, and increased leukocyte counts as significant predictive factors. The areas under the curve (AUCs) of the nomogram for prediction of PID progression to sepsis at 3-day and 7-day (3-/7-day) in the training set and the validation set were 0.886/0.863 and 0.824/0.726, respectively, and the C-index of the model was 0.8905. The RSF displayed excellent performance, with AUCs of 0.939/0.919 and 0.712/0.571 for 3-/7-day risk prediction in the training set and validation set, respectively. Conclusion: The nomogram accurately predicted the incidence of sepsis in PID patients, and relevant risk factors were identified. While the RSF model outperformed the Cox regression models in predicting sepsis incidence, its performance exhibited some instability. On the other hand, the Cox regression-based nomogram displayed stable performance and improved interpretability, thereby supporting clinical decision-making in PID treatment.

Introduction: Gliomas, the most prevalent tumors of the central nervous system, are known for their aggressive nature and poor prognosis. The heterogeneity among gliomas leads to varying responses to the same treatments, even among similar glioma types. In our study, we efferocytosis-related subtypes and explored their characteristics in terms of immune landscape, intercellular communication, and metabolic processes, ultimately elucidating their potential clinical implications.Methods and Results: We first identified efferocytosis-related subtypes in Bulk RNA-seq using the NMF algorithm. We then preliminarily demonstrated the correlation of these subtypes with efferocytosis by examining enrichment scores of cell death pathways, macrophage infiltration, and the expression of immune ligands. Our analysis of single-cell RNA-seq data further supported the association of these subtypes with efferocytosis. Through enrichment analysis, we found that efferocytosis-related subtypes differ from other types of gliomas in terms of immune landscape, intercellular communication, and substance metabolism. Moreover, we found that the efferocytosis-related classification is a prognostic factor with robust predictive performance by calculating the AUC values. We also found that efferocytosis-related subtypes, when compared with other gliomas in drug sensitivity, survival, and TIDE scores, show a clear link to the effectiveness of chemotherapy, radiotherapy, and immunotherapy in glioma patients.Discussion: We identified efferocytosis-related subtypes in gliomas by analyzing the expression of 137 efferocytosis-associated genes, exploring their characteristics in immune landscape, intercellular communication, metabolic processes, and genomic variations. Moreover, we discovered that the classification of efferocytosis-related subtypes has a strong prognostic predictive power and holds potential significance in guiding clinical treatment.

Objectives: The spread of SARS-Cov-2 remains a global concern along with the emergence of variants. This study aims to characterize the epidemiological and clinical features of hospitalized patients who were dragonized with five different variants of SARS-CoV-2 during the past 3 years. Methods: This retrospective study recruited 432 COVID-19 patients who were hospitalized in the First Hospital of Changsha from January 2020 to August 2022. Clinical records on clinical symptoms, laboratory profiles, and chest CT images was collected. The epidemiological and clinical features were compared between COVID-19 patients infected with either the wild-type, Omicron variant or pre- Omicron variants (e.g., Alpha, Beta, Delta). Results: A total of 432 laboratory-confirmed COVID-19 inpatients were dialogized during three waves, including 247 cases during the wild-type transmission period, 65 cases during the transmission period of pre-Omicron variants, and 119 cases during the transmission period of Omicron variants. The proportion of moderately or severely ill inpatients showed a gradual decline from the wild-type transmission period to the Omicron transmission period. The common symptoms of inpatients infected with SARS-CoV-2 wildtype strains included fever (67.61 %), cough (57.89 %), fatigue (33.60 %), and shortness of breath (12.15 %). In contrast, patients infected with other variants mostly showed upper respiratory symptoms. AbstractObjective To compare the clinical efficacy of High intensity-focused ultrasound(HIFU) for different types of adenomyosis classified by magnetic resonance imaging (MRI).Methods A total of 227 patients with adenomyosis who underwent HIFU treatment in Suining Central Hospital from January 2014 to December 2019 were included. Based on the preoperative pelvic MRI examination and Kishi isometric classification method, the patients were divided into 4 types according to the location of lesions. There were 82 cases of type I (endogenous type), 75 cases of type II (exogenous type), 34 cases of type III (nodular type), and 36 cases of type IV (heterogeneous type). The results of HIFU treatment for four types of adenomyosis and intraoperative adverse effects were analyzed. The changes of lesion size, dysmenorrhea, and menstrual volume were analyzed at 3 and 6 months after the operation.Result (1) adverse effects: All patients successfully accepted HIFU treatment under sedation and analgesia. Grade A adverse effects occurred in 51 cases (22.47%) and grade B adverse effects in 3 cases (1.32%). The incidence of adverse effects in type I–IV patients was 21.95%, 22.66%, 23.53%, and 30.56%, respectively. Abstract Polycystic ovary syndrome (PCOS) is a common endocrine disorder in women of childbearing age. The etiology of PCOS is multifactorial, and current treatments for PCOS are far from satisfactory. Recently, an imbalanced autonomic nervous system (ANS) with sympathetic hyperactivity and reduced parasympathetic nerve activity (vagal tone) has aroused increasing attention in the pathogenesis of PCOS. In this paper, we review an innovative therapy for the treatment of PCOS and related co-morbidities by targeting parasympathetic modulation based on non-invasive transcutaneous auricular vagal nerve stimulation (ta-VNS). In this work, we present the role of the ANS in the development of PCOS and describe a large number of experimental and clinical reports that support the favorable effects of VNS/ta-VNS in treating a variety of symptoms, including obesity, insulin resistance, type 2 diabetes mellitus, inflammation, microbiome dysregulation, cardiovascular disease, and depression, all of which are also commonly present in PCOS patients. We propose a model focusing on ta-VNS that may treat PCOS by (1) regulating energy metabolism via bidirectional vagal signaling; (2) reversing insulin resistance via its antidiabetic effect; (3) activating anti-inflammatory pathways; (4) restoring homeostasis of the microbiota-gut-brain axis; (5) restoring the sympatho-vagal balance to improve CVD outcomes; (6) and modulating mental disorders. ta-VNS is a safe clinical procedure and it might be a promising new treatment approach for PCOS, or at least a supplementary treatment for current therapeutics.

Antarctic krill (Euphausia superba), an important group of marine zooplankton in the Southern Ocean, is the only fishery resource with extremely rich reserves and a low degree of development in the world. Antarctic krill is considered to be the greatest potential source of high-quality marine protein resources due to its abundant biomass and high protein content. Peptides prepared from Antarctic krill exhibit multiple physiological activities, including osteoporosis relief, glucose metabolism regulation, blood pressure amelioration, antioxidation, fatigue alleviation, and anti-aging activity. The production and development of Antarctic krill peptides has recently become an industry focus; however, existing research has been limited to the optimization of enzymatic hydrolysis processes, mainly involving the screening of suitable enzymes and the optimization of enzymatic hydrolysis conditions. Due to the high mineral content of Antarctic krill and the introduction of buffer salt in the process of enzymatic hydrolysis, current Antarctic krill peptides products have a high salt content, which leads to poor sensory experience and health risks. Hence, a process for desalination of Antarctic krill peptides is needed. Desalination methods for bioactive substances include dialysis, ultrafiltration, nanofiltration, electrodialysis, and macroporous resin adsorption. In the field of membrane separation, nanofiltration technology has been widely used in the purification, concentration, and desalination of food components owing to its advantages: low operation cost, no introduction of exogenous substances, no destruction of materials, and low rejection rate of monovalent ions. In order to improve product quality and ensure market expansion, the process of desalination of Antarctic krill peptides using nanofiltration technology was studied and optimized in this study.Defatted Antarctic krill powder was enzymatically hydrolyzed by alkaline protease to obtain Antarctic krill peptides for further use. The main factors affecting the desalination effect of Antarctic krill peptides (peptides concentration, nanofiltration pressure, and cycle times) were optimized by single-factor and orthogonal tests, using the desalination rate and protein loss rate as evaluation indexes. The experimental optimization ranges included peptides concentration of 1%~5%, nanofiltration pressure of 0.6~1.4 MPa and cycle times of 1~5. The salt contents of the samples before and after desalination were quantified using the silver nitrate titration method; the protein contents of the experimental samples were quantified using the Lowry colorimetric method. The quality indexes of the Antarctic krill peptides after treatment (including the basic nutritional composition: moisture content, protein content, ash content, salt content; amino acid composition; and molecular weight distribution) were systematically evaluated by the corresponding national standard methods. All experiments were performed in triplicate, and data were expressed as mean ± standard deviation. Excel 2016, IBM SPSS 20.0, and Origin 2018 were used for data analysis and chart drawing.Single-factor tests revealed that peptides concentration of 3%, nanofiltration pressure of 1.0 MPa and a cycle time of 2 could be selected as the design basis for the L9 (33) orthogonal test. The range value of the orthogonal test indicated that the degree of influence of the three factors on the desalination effect was as follows: peptides concentration > cycle times > nanofiltration pressure. The optimum conditions for desalting Antarctic krill peptides obtained by k value analysis were as follows: peptides concentration of 3.0%, nanofiltration pressure of 1.2 MPa and a cycle time of 3. Under the optimal condition, the desalination rate of the Antarctic krill peptides reached up to (86.35±2.11)%, and the protein loss rate was controlled at (9.10±0.35)%, demonstrating the feasibility of the process. The salt content of the Antarctic krill peptides after desalination was reduced to (1.14±0.12)% and the protein content was (92.73±2.29)%. The molecular weights of the Antarctic krill peptides after desalination were mainly distributed between 189 Da and 6500 Da, of which the proportion of peptides with molecular weight less than 3000 Da was (88.91±2.19)%, conforming to the molecular weight distribution range of bioactive peptides. The amount of essential amino acids in the Antarctic krill peptides after desalination accounted for (40.06±0.10)% of the total amino acids, and the ratio of essential amino acids to nonessential amino acids was (66.82±0.28)%. The amino acid compositions of the Antarctic krill peptides after desalination were ideal and met the standard stipulated by the FAO/WHO. The established nanofiltration desalination process presented good treatment effects, and the obtained peptides were of good quality and high nutritional value.The production of Antarctic krill protein-related products may be the next key development for the processing industry, since the sole high-value products of Antarctic krill at present are Antarctic krill oil and its derivatives. The established nanofiltration desalination process has practical application value and would provide technical support for the development of high-quality Antarctic krill peptides. This research provides scientific support for the efficient utilization of Antarctic krill resources.

Abstract Environmental problems such as greenhouse effect, the consumption of fossil energy, and the increase of human demand for energy are becoming more and more serious, which force researcher to turn their attention to the reduction of CO2 and the development of renewable energy. Unsafety, easy to lead to secondary environmental pollution, cost inefficiency, and other problems limit the development of conventional CO2 capture technology. In recent years, many microorganisms have attracted much attention to capture CO2 and synthesize valuable products directly. Fatty acid derivatives (e.g., fatty acid esters, fatty alcohols, and aliphatic hydrocarbons), which can be used as a kind of environmentally friendly and renewable biofuels, are sustainable substitutes for fossil energy. In this review, conventional CO2 capture techniques pathways, microbial CO2 concentration mechanisms and fixation pathways were introduced. Then, the metabolic pathway and progress of direct production of fatty acid derivatives from CO2 in microbial cell factories were discussed. The synthetic biology means used to design engineering microorganisms and optimize their metabolic pathways were depicted, with final discussion on the potential of optoelectronic–microbial integrated capture and production systems.

In order to optimize the decolorization process of Antarctic krill peptides and improve the product quality, activated carbon adsorption method was used to remove the pigment from the Antarctic krill peptides solution. Taking the decolorization rate and protein retention rate as the evaluation indexes, the influence of four independent variables including dosage of activated carbon, pH, decolorization temperature and decolorization time on the decolorization effect were investigated respectively. On the basis of single factor experiment, three independent variables including dosage of activated carbon, pH and decolorization time were optimized by the response surface methodology under the decolorization temperature of 50 ℃. Results showed that powdered activated carbon was more suitable for the removal of pigment from Antarctic krill peptides when compared with granular activated carbon, and the optimal conditions determined by response surface methodology were as follows: dosage of activated carbon 4.0%, pH1.5 and decolorization time 1.0 h. Under the optimal conditions, the decolorization rate was 82.19%±0.20% and the protein retention rate was 90.93%±2.28%. The decolorization treatment had no obvious effects on the proportion of essential amino acids and nonessential amino acids in the amino acid compositions as well as the molecular weight distribution of Antarctic krill peptides. This research would provide scientific support for the development of high-quality Antarctic krill peptides products.

BackgroundPatients with Parkinson’s disease (PD) and sarcopenia often exhibit resilience, frailty, disability, and depression, highlighting the complex and interrelated nature of these conditions.ObjectiveDespite the presence of clinical manifestations of muscle atrophy in both PD and sarcopenia, accurately discerning the coexistence of sarcopenia in PD patients remains a challenging task with significant implications for treatment strategies and prognostic assessments. This study aims to elucidate the specific ultrasonic diagnostic parameters associated with PD accompanied by sarcopenia through a comparative analysis of muscle ultrasound parameters in patients with PD, thereby presenting a novel approach for rapid identification of this condition.MethodsA total of 110 participants were enrolled in this study, including patients with PD and control subjects. Demographic data, clinical characteristics, physical performance tests, appendicular skeletal muscle mass index (ASMI), bioelectrical impedance analysis and muscle ultrasound measurements were collected from all participants. The muscle ultrasound measurements encompassed assessments of muscle thickness, pennation angle and shear wave elastography at various anatomical sites.ResultsParkinson’s disease patients exhibited decreased muscle strength and physical performance, and increased shear wave elastography value. In PD patients with sarcopenia, body circumference, including calf circumference, mid-arm circumference, Waist-to-Hip Ratio and body mass index (BMI) were all significantly decreased. Biceps brachii muscle thickness (MT) and gastrocnemius MT decreased in PD patients with sarcopenia and low ASMI. Binary logistic regression analysis revealed that male PD patients, BMI and gastrocnemius MT were predictive factors for ASMI in PD patients.ConclusionBiceps brachii MT and gastrocnemius MT are important indicators for distinguishing whether PD patients have sarcopenia. Male patients, low BMI and gastrocnemius MT were identified as valid predictors of low ASMI in PD patients. The findings of this study provide important insights into the use of muscle ultrasound in the diagnosis of PD with sarcopenia.

The purpose of this study was to explore antecedent factors that contribute to employee engagement in the human services sector, using a convenience sample of staff in human service agencies in the New York City region. Organizational trust, satisfaction with supervisor, and coworker support were significant predictors of employee engagement.

Abstract Obesity is one of the major public health problems threatening the world, as well as a potential risk factor for chronic metabolic diseases. There is growing evidence that iron metabolism is altered in obese people, however, the highly refined regulation of iron metabolism in obesity and obesity-related complications is still being investigated. Iron accumulation can affect the body’s sensitivity to insulin, Type 2 diabetes, liver disease and cardiovascular disease. This review summarized the changes and potential mechanisms of iron metabolism in several chronic diseases related to obesity, providing new clues for future research.

Abstract Ubiquitination is a critical type of post-translational modifications, of which K63-linked ubiquitination regulates interaction, translocation, and activation of proteins. In recent years, emerging evidence suggest involvement of K63-linked ubiquitination in multiple signaling pathways and various human diseases including cancer. Increasing number of studies indicated that K63-linked ubiquitination controls initiation, development, invasion, metastasis, and therapy of diverse cancers. Here, we summarized molecular mechanisms of K63-linked ubiquitination dictating different biological activities of tumor and highlighted novel opportunities for future therapy targeting certain regulation of K63-linked ubiquitination in tumor.

Abstract Background Quality is the most important factor in satisfaction. However, the existing satisfaction index model of urban and rural resident-based basic medical insurance scheme (SIM_URRBMI) lacks the segmentation of perceived quality elements, it couldn’t provide a reference for quality improvement and satisfaction promotion. This study aims to construct a revised SIM_URRBMI that can accurately and detailly measure perceived quality and provide feasible and scientific suggestions for improving the satisfaction of urban and rural residents' basic medical insurance scheme (URRBMI) in China. Methods Based on the theoretical framework of the American Customer Satisfaction Index, the elements of perceived quality were refined through literature review and expert consultation, and a pool of alternative measurement variables was formed. A three-stage randomized stratified cluster sampling was adopted. The main decision makers of URRBMI in the families of primary school students in 8 primary schools in Changsha were selected. Both the classic test theory and the item response theory were used for measurement variables selection. The reliability and validity of the model were tested by partial least squares (PLS)-related methods. Results A total of 1909 respondents who had URRBMI for their children were investigated. The SIM_URRBMI1.0 consists of 11 latent variables and 28 measurement variables with good reliability and validity. Among the three explanatory variables of public satisfaction, perceived quality had the largest total effect (path coefficient) (0.737). The variable with the greatest effect among the five first-order latent variables on perceived quality was the quality of the medical insurance policy (0.472). Conclusions The SIM_URRBMI1.0 consists of 28 measurement variables and 11 latent variables. It is a reliable, valid, and standard satisfaction measurement tool for URRBMI with good prediction ability for public satisfaction. In addition, the model provides an accurate evaluation of the perceived quality, which will greatly help with performance improvement diagnosis. The most critical aspects of satisfaction improvement are optimizing the scope and proportion of reimbursement as well as setting appropriate level of deductible and capitation of URRBMI.

Abstract Background Although 20–60% of patients with chronic rhinosinusitis with nasal polyps (CRSwNP) have asthma, the risk factors associated with comorbid asthma are not clear. The aim of the study was to investigate the factors associated with asthma, and develop a practical scoring system to screen asthma comorbidity in CRSwNP patients. Methods This report describes a cross-sectional study with consecutive CRSwNP patients. Two cohorts of CRSwNP patients named “modelling” group and “validation” group were investigated respectively. Logistic regression analysis was performed based on demographic and clinical data collected from patients in the modelling group to determine the risk factors associated with asthma, and establish a scoring system for screening comorbid asthma. Receiver operating characteristic curve was constructed to evaluate the screening system; the optimal cut-off point was established by means of the Yoden Index. The consistency between the diagnosis of asthma by the Global Initiative for Asthma (GINA) criteria and by the screening system was assessed by Kappa value in the validation group.
