Abstract

Identifying and discovering drug-target interactions(DTIs) are vital steps in drug discovery and development. They play a crucial role in assisting scientists in finding new drugs and accelerating the drug development process. Recently, knowledge graph and knowledge graph embedding (KGE) models have made rapid advancements and demonstrated impressive performance in drug discovery. However, such models lack authenticity and accuracy in drug target identification, leading to an increased misjudgment rate and reduced drug development efficiency. To address these issues, we focus on the problem of drug-target interactions, with knowledge mapping as the core technology. Specifically, a causal intervention-based confidence measure is employed to assess the triplet score to improve the accuracy of the drug-target interaction prediction model. Experimental results demonstrate that the developed confidence measurement method based on causal intervention can significantly enhance the accuracy of DTI link prediction, particularly for high-precision models. The predicted results are more valuable in guiding the design and development of subsequent drug development experiments, thereby significantly improving the efficiency of drug development.

Abstract

With the continuous development of business process management technology, the increasing business process models are usually owned by large enterprises. In large enterprises, different stakeholders may modify the same business process model. In order to better manage the changeability of processes, they adopt configurable business process models to manage process variants. However, the process variants will vary with the change in enterprise business demands. Therefore, it is necessary to explore the co-evolution of the process variants so as to effectively manage the business process family. To this end, a novel framework for co-evolution between business process variants through a configurable process model is proposed in this work. First, the mapping relationship between process variants and configurable models is standardized in this study. A series of change operations and change propagation operations between process variants and configurable models are further defined for achieving propagation. Then, an overall algorithm is proposed for achieving co-evolution of process variants. Next, a prototype is developed for managing change synchronization between process variants and configurable process models. Finally, the effectiveness and efficiency of our proposed process change propagation method are verified based on experiments on two business process datasets. Abstract

Gut microbiota plays a crucial role in modulating pig development and health, and gut microbiota characteristics are associated with differences in feed efficiency. To answer open questions in feed efficiency analysis, biologists seek to retrieve information across multiple heterogeneous data sources. However, this is error-prone and time-consuming work since the queries can involve a sequence of multiple sub-queries over several databases. We present an implementation of an ontology-based Swine Gut Microbiota Federated Query Platform (SGMFQP) that provides a convenient, automated, and efficient query service about swine feeding and gut microbiota. The system is constructed based on a domain-specific Swine Gut Microbiota Ontology (SGMO), which facilitates the construction of queries independent of the actual organization of the data in the individual sources. This process is supported by a template-based query interface. A Datalog+-based federated query engine transforms the queries into sub-queries tailored for each individual data source, and an automated workflow orchestration mechanism executes the queries in each source database and consolidates the results. The efficiency of the system is demonstrated on several swine feeding scenarios.

37. Fine and combinatorial regulation of key metabolic pathway for enhanced β‐alanine biosynthesis with non‐inducible Escherichia coli.

Abstract

β‐Alanine is the only β‐amino acid in nature and one of the most important three‐carbon chemicals. This work was aimed to construct a non‐inducible β‐alanine producer with enhanced metabolic flux towards β‐alanine biosynthesis in Escherichia coli. First of all, the assembled E. coli endogenous promoters and 5′‐untranslated regions (PUTR) were screened to finely regulate the combinatorial expression of genes panDBS and aspBCG for an optimal flux match between two key pathways. Subsequently, additional copies of key genes (panDBSK104S and ppc) were chromosomally introduced into the host A1. On these bases, dynamical regulation of the gene thrA was performed to reduce the carbon flux directed in the competitive pathway. Finally, the β‐alanine titer reached 10.25 g/L by strain A14‐R15, 361.7% higher than that of the original strain. Under fed‐batch fermentation in a 5‐L fermentor, a titer of 57.13 g/L β‐alanine was achieved at 80 h. This is the highest titer of β‐alanine production ever reported using non‐inducible engineered E. coli. Abstract

Summary: Sesame seed hull contains a substantial concentration of minerals. During the roasting, these minerals assume a catalytic function in the degradation of the sesame seed hull pectin. In this work, the effect of four inorganic chloride salts on the structure and thermal degradation products of sesame hull pectin during heating was investigated. The results indicated that, during roasting, chloride salts catalysed the degradation of pectin polysaccharide side chain regions more than the linear main chain. The introduction of chloride salts reduced the rate of mass loss from sesame hull pectin and resulted in a shift of the main peak towards a lower temperature range. The introduction of KCl and NaCl resulted in a significant decrease in the yield of furans and an increase in the proportion of small molecules such as acids and aldehydes in the thermal degradation products of sesame peel pectin, suggesting that the presence of KCl and NaCl facilitated the fragmentation of the sugar units. In contrast, MgCl2 and CaCl2 clearly facilitated the formation of furans and exhibited a discernible catalytic effect on the degradation of acidic substances. Abstract

Summary: In this study, sesame hull oil (SHO) was extracted to characterise its composition. SHO was heated to determine the volatiles and their effect on the quality of cold‐pressed dehulled sesame oil (SO). Seventeen fatty acids, four lignans (1088.65 mg/100 g), tocopherols (284.17 mg/100 g) mainly in the γ‐form, and sterol (1722.16 mg/100 g, 4.67 times more than in sesame kernel oil) were detected in SHO. After heating, the volatiles found in SHO were mainly aldehydes and acids, represented by hexanal (fatty, green aroma) and 3‐methyl‐butanoic acid (fatty, rancid aroma), respectively. The acid value, peroxide value, anisidine value, and turbidity of SO increased significantly after adding SHO, and the scavenging ability of 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) was enhanced. The oxidation induction times of SO with the addition of SHO ranged from 8.93 to 9.53 h. The present study provides information about SHO and a new direction for the utilisation of sesame hulls. Abstract

A novel bismuth(III) trifluoro‐methanesulfonate‐catalyzed and environmentally benign synthetic strategy for the construction of a wide range of structurally diverse, sophisticated [5,6,5]‐oxygen‐containing tricyclic frameworks with easy‐to handle propargylic alcohols and 2‐allylphenols as substrates in the presence of Bi(OTf)3 and AgOTf is described. This Lewis acid catalyzed [3+2] annulation protocol, which tolerates a great deal of functional groups, proceeds through a sequential Meyer‐Schuster rearrangement, nucleophilic substitution, 5‐exo‐trig cyclization, 5‐endo‐trig cyclization, and proton exchange sequences, affording a versatile approach for accessing oxygen‐containing tricyclic skeletons in moderate‐to‐excellent yields. In addition, most of the obtained compounds exhibited anti‐tumor activities against three types of human cancer cell lines in vitro, including Caco‐2 colon cancer cells, MCF‐7 breast cancer cells, and Hepg‐2 liver cancer cells. Abstract

We report the application of the resistive anode readout method on a two dimensional imaging GEM detector. The resistive anode consists of $6\times6$ cells with the cell size $6~\mathrm{mm}\times6~\mathrm{mm}$. New electronics and DAQ system are used to process the signals from the 49 readout channels. The detector has been tested by using the X-ray tube (8~keV). The spatial resolution of the detector is about $103.46~\mathrm{{\mu}m}$ with the detector response part about $66.41~\mathrm{{\mu}m}$. The nonlinearity of the detector is less than $1.5\%$. A quite good two dimensional imaging capability is achieved as well. The performances of the detector show the prospect of the resistive anode readout method for the large readout area imaging detectors., Comment: 6 Pages, 10 figures
