Photothermal therapy (PTT) of nanomaterials is an emerging novel therapeutic strategy for breast cancer. However, there exists an urgent need for appropriate strategies to enhance the antitumor efficacy of PTT and minimize damage to surrounding normal tissues. Piezo1 might be a promising novel photothermal therapeutic target for breast cancer. This study aims to explore the potential role of Piezo1 activation in the hyperthermia therapy of breast cancer cells and investigate the underlying mechanisms. Results showed that the specific agonist of Piezo1 ion channel (Yoda1) aggravated the cell death of breast cancer cells triggered by heat stress in vitro. Reactive oxygen species (ROS) production was significantly increased following heat stress, and Yoda1 exacerbated the rise in ROS release. GSK2795039, an inhibitor of NADPH oxidase 2 (NOX2), reversed the Yoda1-mediated aggravation of cellular injury and ROS generation after heat stress. The in vivo experiments demonstrate the well photothermal conversion efficiency of TiCN under the 1,064 nm laser irradiation, and Yoda1 increases the sensitivity of breast tumors to PTT in the presence of TiCN. Our study reveals that Piezo1 activation might serve as a photothermal sensitizer for PTT, which may develop as a promising therapeutic strategy for breast cancer.

Objective: This study aims to assess the clinical effectiveness of combining mindfulness‐based stress reduction (MBSR) with exercise intervention in improving anxiety, depression, sleep quality and mood regulation in patients with non‐small cell lung cancer (NSCLC). Methods: A total of 60 patients with NSCLC who had not received surgical treatment were selected using convenience sampling and divided into an intervention group and control group, with 30 patients in each group. The control group received conventional psychological nursing care, whereas the intervention group received a combination of MBwSR and exercise therapy. Before the intervention, a questionnaire was completed to collect the basic data of the two groups. Further questionnaires were administered at 6 and 8 weeks after treatment to assess anxiety, depression, sleep quality and other items included in the five‐item Brief Symptom Rating Scale (BSRS‐5). Results: No significant differences between the intervention and control groups were identified in terms of personal and clinical characteristics (p > 0.05). No significant differences were determined in the BSRS‐5, Self‐Rating Anxiety Scale (SAS), Self‐Rating Depression Scale (SDS) or Pittsburgh Sleep Quality Index (PSQI) scores between the intervention and control groups before the intervention. However, 6 and 8 weeks after the intervention, scores were significantly lower in both groups (p < 0.001). Significant differences in the BSRS‐5, SAS, SDS and PSQI scores were identified between the two groups at different time points (p < 0.001). Conclusion: The combination of MBSR and exercise intervention demonstrated improvements in anxiety, depression, sleep quality and BSRS‐5 scores in patients with NSCLC. [ABSTRACT FROM AUTHOR]

In order to understand the function of cyclophilin family OsCYP22 from Oryza sativa L., the yeast two hybrid bait vector of OsCYP22 was constructed and proteins interacting with OsCYP22 were screened by yeast two hybrid. The results showed that noself-activating activity and toxicity of OsCYP22 bait vector to yeast cells were detected., among which 38 positive colonies were obtained by yeast two hybrid screening. Combining sequencing analysis and bioinformatics methods, 20 candidates that may interact with OsCYP22 were screened. Further validation of the full-length interactions between OsCYP22 and OsCSN5 and OsRUB1 related to the regulation of plant root growth was conducted. The results showed that OsCSN5 interacted with OsCYP22 in yeast. This study provides a theoretical basis for further study of the biological function of OsCYP22. [ABSTRACT FROM AUTHOR]

Multiwalled carbon nanotubes (MWCNTs) have numerous applications in the field of carbon nanomaterials. However, the associated toxicity concerns have increased significantly because of their widespread use. The inhalation of MWCNTs can lead to nanoparticle deposition in the lung tissue, causing inflammation and health risks. In this study, celastrol, a natural plant medicine with potent anti-inflammatory properties, effectively reduced the number of inflammatory cells, including white blood cells, neutrophils, and lymphocytes, and levels of inflammatory cytokines, such as IL-1β, IL-6, and TNF-α, in mice lungs exposed to MWCNTs. Moreover, celastrol inhibited the activation of the NF-κB-signaling pathway. This study confirmed these findings by demonstrating comparable reductions in inflammation upon exposure to MWCNTs in mice with the deletion of NF-κB (P50−/−). These results indicate the utility of celastrol as a promising pharmacological agent for preventing MWCNT-induced lung tissue inflammation. [ABSTRACT FROM AUTHOR]
