Objective: A method for determining the residues of 30 antiparasitic drugs in animal-derived food using ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was established. Methods: The samples were extracted with acetonitrile and 1% ammonia acetic ether and purified by QuEChERS. After purification, Waters ACQUITY UPLCTM BEH C18 column was used for separation, and a gradient elution of 10 mmol/L ammonium formate (containing 0.1% formic acid) aqueous solution, acetonitrile:methanol (50:50, v:v) was performed as the mobile phase. Detection was carried out using electrospray ionization (ESI) in both positive and negative ion modes using multiple reaction monitoring (MRM). Matrix-matched external standard quantification was used. Results: Under the optimized conditions, the 30 antiparasitic drugs showed good linearity within their respective linear ranges, with coefficient of determination (r2) greater than 0.99. The recoveries ranged from 70.1% to 111%, and the relative standard deviations were between 0.10% and 9.1% (n=6). The method detection limit ranged from 0.001 to 0.3 μg/kg, and the method quantification limit ranged from 0.004 to 1 μg/kg. Conclusion: The method is sensitive, accurate, and exhibits good repeatability and stability, making it suitable for detecting various antiparasitic drug residues in animal-derived food.

[Objectives] The paper was to establish an ultra high performance liquid chromatography-quadrupole/linear ion trap complex mass spectrometry for the determination of 10 kinds of a2-receptor agonists in animal derived food. [Methods] The samples were extracted with sodium carbonate buffer solution and ethyl acetate, and analyzed by mass spectrometry after solid phase extraction and high performance liquid chromatography separation. [Results] Ten kinds of a2-receptor agonists showed a good linear relationship in the range of 1 -100 p,g/mL, with the average recovery of over 69% and the relative standard deviation less than 8.32%. The detection limit of 10 kinds of a2-receptor agonists was up to 1 jxg/kg. [Conclusions] The method has good selectivity and strong anti-interference ability, and can meet the requirements of 10 kinds of a2-receptor agonists residues in animal derived food. On the basis of a brief description of the common types of antibiotics in animal derived food, the current status of antibiotic residues and hazards, focusing on antibiotic residue detection methods and research progress in animal source foods, it was pointed out that the main types of antibiotics were fluoroquinolones, β-lactams, macrolides, tetracyclines, nitrofurans, sulfonamides, amphenicols and aminoglycosides. Currently, antibiotic abuse caused serious residual problems, and the accumulation of antibiotics in the body could trigger allergic reactions, drug resistance and other symptoms. Antibiotic detection methods mainly included chromatography, spectrometry, electrochemistry, enzyme-linked immunosorbent assay (ELISA) and chemical sensing, which respectively suffered from the problems of complicated pre-treatment process, weak anti-interference ability, susceptibility to false positives, fewer types of detection, and difficulty in realising the simultaneous detection of a variety of antibiotics, and so on. Compared with other methods, chemical sensing had the advantages of simple and rapid process, obvious color change reaction, and enormous potential for application in the detection of antibiotic residues. In the future, the structural characteristics of antibiotics can be analysed in depth, and sensors detecting a variety of antibiotics simultaneously can be developed, thus further promoting the development of rapid and accurate detection of antibiotic residues in animal derived food. With the improvement of people's living standards and health awareness, the safety of animal-derived food has attracted worldwide attention. Chemical contamination (melamine, veterinary drug residues, food additive residues, etc.) and pathogenic microorganisms are the primary causes of food safety issues in animal-derived food products, potentially threatening human health and socioeconomic development. Therefore, it is essential to establish rapid detection methods for animal-derived food safety. Surface-enhanced Raman spectroscopy (SERS) has the advantages of rapid detection, good selectivity, and high sensitivity. It has shown great application potential in food safety, which has become one of the research hotspots. This paper reviews the application of SERS in the safety of animal-derived food detection and comprehensively introduces the use of SERS to detect melamine, foodborne pathogens, pesticide and veterinary drug residues, and food additives. Furthermore, it prospects the application of SERS technology in rapid residue detection, which would be believed to provide valuable insights into the field of SERS-based food safety evaluation.

A detection method for the determination of 20 kinds of cephalosporins in animal derived food was established with multiple stable isotope internal standards by liquid chromatography-tandem mass spectrometry. Cephalosporins were extracted from samples with methanol, and purified with octadecylsilane sorbent. An Ascentis C8 column was used for analyzing with 0.1% formic acid aqueous solution and acetonitrile as mobile phases. Analysis of cephalosporins were respectively performed on the positive and negative scan mode by multiple reactions monitoring. Results showed that the linear range of the methods was 5~100 μg/L for 17 kinds of cephalosporins with highly responsive, and 25~500 μg/L for 3 kinds of cephalosporins with lowly responsive. The determination coefficients of the linear equations ranged from 0.9913 to 0.9998. The limit of detection for 20 kinds of cephalosporins ranged from 1.0 to 20.0 μg/kg, and the limit of quantification ranged from 3.0 to 60.0 μg/kg in animal derived sample. The average recovery rate of 20 cephalosporins ranged from 78.08% to 115.47%, and the relative standard deviation ranged from 3.07% to 12.44%. The validated method was simple and accurate. It was suitable for the determination of 20 cephalosporins in animal food.

Food safety has long been one of the greatest problems in public health. Among all the food safety problems, antibiotic residues in animal derived foods are one of the most serious food contaminations that threaten human health seriously. However, the conventional techniques for monitoring and detection of antibiotics have shortcomings such as complicated sample pretreatment procedures, complex and time-consuming operations, dependence on expensive and bulky instruments, need for skilled professional personnel as well as poor selectivity and sensitivity, thus limiting their applications. The emergence of aptamers provides a new novel probe for the establishment of aptasensors to circumvent those drawbacks because of their outstanding superiorities. Thus, the application of aptamers for monitoring and detection of antibiotic residues has attracted increasing attention and has become one of the research hotspots in the past years, showing a great potential for practical transformation and commercialization. In this review, the recent progresses of aptamer selection against various antibiotics were first presented. Then, the design and fabrication of various nanomaterials based aptasensors for monitoring and detection of antibiotic residues in animal derived foods were systematically summarized, with anticipation to avoid repeated work on aptamer selection and provide new insight and point out the future direction into the development of aptasensors for antibiotic detection. Mycotoxins are common toxic metabolites present in cereals and vegetal raw materials, which are commonly included in animal feed. Ochratoxin A (OTA) has generally been detected in plant origin foodstuffs such as cereals, coffee, dried fruits, nuts, among others. However, it has been also detected in meat and meat by-products, especially those derived from pork, which is the most sensitive specie to OTA exposure. The exposure of farm animals to mycotoxins could lead to undesirable residues in food products of animal origin, which constitute an important part of daily diets. Thus, although contents reported in animal by-products are lower than those reported in products of vegetal origin, there is also a public health concern about the possible ingestion of edible animal products contaminated by mycotoxins, as their entry into the food chain may cause adverse effects on human health. No maximum levels have been set for OTA in animal by-products, although its presence in meat and meat products made from contaminated raw materials has been widely reported, reaching high levels in some cases. A method for the analysis of 120 drugs in animal derived food was developed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). These analytes belong to 12 families of veterinary anti-microbial agents (quinolones, macrolides, β-lactams, nitroimidazoles, sulfonamides, lincomycines, chloramphenicols, quinoxalines, tetracyclines, polypeptides, and antibacterial synergists) as well as other compounds not assigned to a particular drug family. The animal derived food samples include muscle and liver of swine, bovine, sheep, and chicken, as well as hen eggs and dairy milk. The sample preparation included ultrasound-assisted extraction (UAE) with acetonitrile-water and a final clean-up with auto solid-phase extraction (SPE) on HLB cartridges. The detection and quantification of 120 anti-microbial agents was performed using LC-MS/MS in positive and negative ion mode. The chromatographic separation was performed on a C18 column using acetonitrile and 0.1% formic acid as the mobile phase. The limit of detection (LOD) and limit of quantification (LOQ) of all drugs in food-producing animals were 0.5–3.0 μg/kg and 1.5–10.0 μg/kg, respectively. The developed method was successfully utilized to monitor real samples, which demonstrated that it is a simple, fast, and robust method, and could be used as a regulatory to screen for the presence of residues from veterinary anti-microbial drugs in animal-derived foods. A liquid chromatography-tandem mass spectrometry method, coupled with a dispersive solid phase extraction (DSPE) procedure for sample preparation, was developed to determine 13 classes of 42 veterinary drugs in four representative animal-derived foods. The analytes were dispersed with water, extracted with acetonitrile containing 5% (v/v) formic acid, salted out by salts, and purified by DSPE. The analytes were separated on a C18 column (100 mm×2.1 mm, 2 μ m) with gradient elution using the mobile phase containing methanol and 0.1% (v/v) formic acid aqueous solution. Electrospray ionization-mass spectrometry was performed in multiple reaction monitoring mode for analysis of the 42 compounds. The correlation coefficients of the standard calibration curves for the 42 veterinary drugs were all above 0.995. Most recoveries at three spiked levels in the four representative matrixes ranged from 65.8% to 135.5%, with relative standard deviations of 0.5%-14.2% ( n =6). The limits of detection (LODs, S/N =3) and the limits of quantification (LOQs, S/N =10) were 0.01-1.68 μ g/kg and 0.01-5.62 μ g/kg, respectively. The method is simple, rapid, sensitive, and suitable for the simultaneous determination of the 42 veterinary drugs in animal-derived food.
