Insects attach to various surfaces that differ, among others, in roughness and wettability. Identifying surface characteristics that allow or prevent insects from attaching are an important research avenue of pest control. Here we take an experimental approach to analyse the attachment of common bed bugs, Cimex lectularius Linnaeus (1758), to Perspex (PMMA) substrates. We construct a reliable centrifuge device that allows the measurement of attachment forces at substrate roughnesses, Ra, between 0.02 and 1.3 μm and at two wettabilities. Our results suggest that bed bug attachment to surfaces is minimal at a substrate roughness of 0.2 and 0.4 μm on normal PMMA, where the lowest attachment force was 0.8 mN and the safety factor 15. At lower and higher roughness, attachment forces were higher and the safety factor increased to a maximum of 133. On PMMA that was made superhydrophobic by spray‐coating, attachment was lowest (0.2 mN) at the lowest roughness and continuously increased with increasing roughness, reaching 2.5 mN and a safety factor of 46. For every roughness, attachment forces were lower on superhydrophobic than on normal PMMA. This knowledge may inspire the development of repelling substrates for bed bug control. Simple Summary: Bed bugs have expanded globally over the past two decades, ausing several health risks. Mutations in their genes allow bed bugs to develop insecticide resistance. However, the extent to which gene mutations exist in the bug genome remains largely unknown because the genomes of resistant strains have not been determined. We accurately sequenced the genomes of both susceptible and resistant strains and compared the gene sequences between the two strains. Several genes with resistance-specific mutations have been identified. These mutations can alter gene function and lead to insecticide resistance. Insecticide resistance in the bed bug Cimex lectularius is poorly understood due to the lack of genome sequences for resistant strains. In Japan, we identified a resistant strain of C. lectularius that exhibits a higher pyrethroid resistance ratio compared to many previously discovered strains. We sequenced the genomes of the pyrethroid-resistant and susceptible strains using long-read sequencing, resulting in the construction of highly contiguous genomes (N50 of the resistant strain: 2.1 Mb and N50 of the susceptible strain: 1.5 Mb). Gene prediction was performed by BRAKER3, and the functional annotation was performed by the Fanflow4insects workflow. Next, we compared their amino acid sequences to identify gene mutations, identifying 729 mutated transcripts that were specific to the resistant strain. Among them, those defined previously as resistance genes were included. Additionally, enrichment analysis implicated DNA damage response, cell cycle regulation, insulin metabolism, and lysosomes in the development of pyrethroid resistance. Genome editing of these genes can provide insights into the evolution and mechanisms of insecticide resistance. This study expanded the target genes to monitor allele distribution and frequency changes, which will likely contribute to the assessment of resistance levels. The cryptic behavior that characterizes bed bugs (Cimex lectularius L.) makes them one of the most difficult indoor pests to control. Thus, the use of products with long residual efficacy is a key component of successful bed bug management. Aprehend is a biopesticide for bed bug control, whose active ingredient is the entomopathogenic fungus, Beauveria bassiana. This product shows promise for extended bed bug control against pyrethroid-resistant bed bugs, causing mortality days after contact with the treated surface. However, it is unclear how surface type and distance traveled by bed bugs over a treated surface impact efficacy. Therefore, we evaluated the efficacy of different Aprehend band widths applied to different substrates commonly encountered by bed bugs, looking specifically at average time to mortality and overall mortality. Our findings indicate that exposure to fresh applications of Aprehend resulted in high mortality when bed bugs traveled very short distances (1 cm) over some Aprehend-treated surfaces (vinyl tile and cotton jersey fabric), while exposure to other treated surfaces (unfinished pinewood and painted drywall) resulted in comparatively lower mortality even over longer distances (5 cm). Furthermore, we found that bed bugs crawling on cotton jersey fabric picked up a significantly higher number of spores compared to unfinished pinewood. These results indicate that applications of Aprehend as 5 cm (2 inches) bands according to label recommendations can be effective, although surface type is an important determinator of efficacy. Abstract Over the last two decades, an increase in bed bug infestations has been observed worldwide. Although their definitive role as vectors of infectious agents has not yet been demonstrated, bed bugs have a direct effect on human health through dermatological reactions to their bites and psychological disorders linked to domestic infestations. In this study, the effectiveness of using MALDI-TOF MS to correctly identify these two bed bug species at immature stages was assessed, as well as it effectiveness as discriminating between the immature stages (IS) of C. lectularius and C. hemipterus and their associated developmental stages. A total of 305 specimens were subjected to MALDI-TOF MS analysis, including 153 C. lectularius (28 eggs and 25 nymphs per stage from IS1 to IS5) and 152 C. hemipterus (27 eggs and 25 nymphs per stage from IS1 to IS5). ). MALDI-TOF MS analysis enabled us to obtain 84.97% (130/153) of high-quality MS spectra in terms of reproducibility and profile intensity. Twenty-four spectra including two per stage, from egg to IS5, and per bed bug species - were added to our in-house MS reference arthropod spectra database. All specimens were correctly identified at the species level, independently of the developmental stage, with log score values (LSVs) ranging from 1.75 to 2.79 (mean = 2.29 ± 0.12) and 1.81 to 2.71 (mean = 2.37 ± 0.03) for C. lectularius and C. hemipterus, respectively. MALDI-TOF MS correctly classified 53,33% (104/195) of the Cimex at the correct immature stage. Conversely, an accurate comparison of the profiles with a Genetic Algorithm model underlined that grouping the immature stages in two groups, early (IS1-IS2) and late (IS3-IS4-IS5), made it possible to obtain a cross validation (CV) and recognition capability (RC) greater than 92% and 94%, respectively, for both species. This study holds great promise for the management of bed bug infestations.

The common bed bug, Cimex lectularius , is one of the main human parasites. The world-wide resurgence of this pest is mainly due to globalization, and the spread of insecticide resistance. A few studies have compared the transcriptomes of susceptible and resistant strains; however, these studies usually relied on strains originating from distant locations, possibly explaining their extended candidate gene lists. Here, we compared the transcriptomes of 2 strains originating from the same location and showing low overall genetic differentiation (⁠ F ST = 0.018 ⁠) but varying in their susceptibility to pyrethroids, before and after insecticide exposure. In sharp contrast with previous studies, only 24 genes showing constitutive differential expression between the strains were identified. Interestingly, most of the genes with increased expression in the resistant strain encoded cuticular proteins. However, those changes were not associated with significant difference in cuticular thickness, suggesting that they might be involved in qualitative changes in the cuticle. In contrast, insecticide exposure induced the expression of a multitude of genes, mostly involved in detoxification. Finally, our set of transcriptome candidate loci showed little overlap with a set of loci strongly genetically differentiated in a previous study using the same strains. New geographical and host records of bed bug species of the genus Cimex L. (Hemiptera Cimicidae) are present in the article, and we also document and curate previously known data. In Russia, six species of the genus Cimex (C. dissimilis, C. hemipterus, C. hirundinis, C. lectularius, C. pipistrelli, C. sibiricus) are confirmed. Cimex pipistrelli are newly recorded for northern bat Eptesicus nilssonii and eastern water bat Myotis petax. C. lectularius was registered on greater noctule bat Nyctalus lasiopterus, Nathusius' pipistrelle Pipistrellus nathusii and Turanian serotine Eptesicus serotinus turcomanus for the first time. Our molecular data support the earlier assumption about a low level of genetic differences between C. pipistrelli and C. japonicus, which puts taxonomic status of C. japonicus into question. The common bed bug, Cimex lectularius, is a hemipteran insect that feeds only on blood, and whose bites cause public health issues. Due to globalization and resistance to insecticides, this pest has undergone a significant and global resurgence in recent decades. Blood is an unbalanced diet, lacking notably sufficient B vitamins. Like all strict hematophagous arthropods, bed bugs host a nutritional symbiont supplying B vitamins. In C. lectularius, this nutritional symbiont is the intracellular bacterium Wolbachia (wCle). It is located in specific symbiotic organs, the bacteriomes, as well as in ovaries. Experimental depletion of wCle has been shown to result in longer nymphal development and lower fecundity. These phenotypes were rescued by B vitamin supplementation. Understanding the interaction between wCle and the bed bug may help to develop new pest control methods targeting the disruption of this symbiotic interaction. The objective of this work was thus to quantify accurately the density of wCle over the life cycle of the host and to describe potential associated morphological changes in the bacteriome. We also sought to determine the impact of sex, feeding status, and aging on the bacterial population dynamics. We showed that the relative quantity of wCle continuously increases during bed bug development, while the relative size of the bacteriome remains stable. We also showed that adult females harbor more wCle than males and that wCle relative quantity decreases slightly in adults with age, except in weekly-fed males. Living in groups can provide essential experience that improves sexual performance and reproductive success. While the effects of social experience have drawn considerable scientific interest, commonly used behavioral assays often do not capture the dynamic nature of interactions within a social group. Here, we conducted 3 experiments using a social network framework to test whether social experience during early adulthood improves the sexual competence of bed bugs (Cimex lectularius) when placed in a complex and competitive group environment. In each experiment, we observed replicate groups of bed bugs comprising previously socialized and previously isolated individuals of the same sex, along with an equal number of standardized individuals of the opposite sex. Regardless of whether we controlled for their insemination history, previously isolated males mounted and inseminated females at significantly higher rates than previously socialized males. However, we found no evidence of social experience influencing our other measures of sexual competence: proportion of mounts directed at females, ability to overcome female resistance, and strength of opposite-sex social associations. We similarly did not detect effects of social experience on our female sexual competence metrics: propensity to avoid mounts, rate of successfully avoiding mounts, opposite-sex social association strength, and rate of receiving inseminations. The global resurgence of bed bug infestations, exacerbated by increasing international travel, trade, and insecticide resistance, has significantly impacted Korea. This study identified the bed bug species and performed pyrethroid resistance genotyping of recently resurgent bed bugs in Korea. Thirty-one regional bed bug samples were collected from 5 administrative regions: Gyeonggi-do (n=14), Seoul (n=13), Busan (n=2), Jeonllanam-do (n=1), and Chungcheongbuk-do (n=1). The samples underwent morphological and molecular identification. Twenty-four regional samples (77.4%) were identified as the tropical bed bug, Cimex hemipterus, and the remaining 7 regional samples (22.6%) were identified as the common bed bug, Cimex lectularius. The C. hemipterus regional samples carried at least three mutations associated with knockdown resistance (kdr), including 2 super-kdr mutations. The 7 C. lectularius regional samples possessed at least one of the 3 kdr-related mutations associated with pyrethroid resistance. This study confirms that the prevalent bed bug species recently in Korea is C. hemipterus, replacing the previously endemic C. lectularius. We evaluated lethal temperatures and times for killing bed bugs in diverse covered and uncovered conditions simulating their natural habitats. A total of 5400 adult bed bugs were collected alive from 17 infested locations in Paris. They were morphologically identified in laboratory as Cimex lectularius. They were then distributed in multiple sets of 30 specimens to examine in covered (tissue, furniture, mattress or blanket) and uncovered (direct exposure) conditions and in diverse step-function temperatures (50, 55 and 60°C) and times (15, 30, 60 and 120 minutes), replicated three times. Effective mortality was observed in 1080 specimens exposed directly to 50°C for 60 minutes. In specimens covered by tissue (1080 specimens), furniture (1080) or mattress (1080), all were dead at 60°C within 60 minutes. The specimens covered by blanket (1080) at the same temperature were dead after 120 minutes. Cimex lectularius, known as the common bed bug, is a widespread hematophagous human ectoparasite and urban pest that is not known to be a vector of any human infectious disease agents. However, few studies in the era of molecular biology have profiled the microorganisms harbored by field populations of bed bugs. The objective of this study was to examine the viruses present in a large sampling of common bed bugs and related bat bugs (Cimex pipistrelle). RNA sequencing was undertaken on an international sampling of > 500 field-collected bugs, and multiple workflows were used to assemble contigs and query these against reference nucleotide databases to identify viral genomes. Shuangao bed bug virus 2, an uncharacterized rhabdovirus previously discovered in Cimex hemipterus from China, was found in several bed bug pools from the USA and Europe, as well as in C. pipistrelle, suggesting that this virus is common among bed bug populations. In addition, Shuangao bed bug virus 1 was detected in a bed bug pool from China, and sequences matching Enterobacteria phage P7 were found in all bed bug pools, indicating the ubiquitous presence of phage-derived elements in the genome of the bed bug or its enterobacterial symbiont. However, viral diversity was low in bed bugs in our study, as no other viral genomes were detected with significant coverage. These results provide evidence against frequent virus infection in bed bugs. Nonetheless, our investigation had several important limitations, and additional studies should be conducted to better understand the prevalence and composition of viruses in bed bugs. Most notably, our study largely focused on insects from urban areas in industrialized nations, thus likely missing infrequent virus infections and those that could occur in rural or tropical environments or developing nations. Background: Bed bugs are hematophagous insects with a long history of presence in human communities. Over the last three decades, infestations by bed bugs in human dwellings have drastically increased, leading to a rise in bed bug concerns. Nevertheless, very little is known about the bed bug species and their population diversity in Algeria. Method: A pilot entomological inventory was performed in May 2019 in Tizi Ouzou, in northern Algeria. The gathered bed bug specimens were identified by morphological and molecular approaches, followed by neighbor-joining and network phylogenetic analyses. Results: A total of seven out of 12 requested locations were allowed to inspect for bed bug infestation. Of these, three locations were found with active bed bug infestations. A total of 145 specimens belonging to different life stages [egg (21), nymph (74), adult male (17), and female (33)] were collected and analyzed using morphological and molecular approaches. The adult specimens were identified as Cimex lectularius according to specific morphological criteria, most importantly the pronotum laterally expanded with more flattened extreme margins. Morphological identification of the adults was confirmed further by conventional PCR targeting 450 bp fragment of the COI gene. All the nymphs and eggs were also molecularly identified as C. lectularius. Neighbor-Joining phylogenetic tree reconstructed with the collected specimens provides clues on the presence of two closely phylogenetic groups. The first one gathers our samples of Algeria with previously reported COI haplotype sequences from Asian, European, and North American countries. The second group encompasses a lesser-documented haplotype reported in Europe and Central America. These findings were further confirmed by network analysis. Conclusions: These results provide evidence of established C. lectularius infestation in Algeria and its potential dispersal capacity by travelers or immigrants and will help future management of these ectoparasites.

In late summer of 2023 bed bug (Cimex lectularius) infestations received much media attention especially from Paris Fashion Week (2023–09–25/2023–10–03). Concern in France has grown in recent years and the public may have been sensitised from the recent release of the report Les punaises de lit: impacts, prévention et lutte from the Agence Nationale de Sécurité Sanitaire. Additionally, families returning from summer travel for the start of the school year (2023–09–04) may have brought Cimex spp. with them. A belief, typically false, that they are associated with poor housekeeping and the commercial sensitivity of infestations makes quantitative data on the occurrence and frequency of the insects difficult to find. Often it was based on the number of consultations with physicians and enquiries about bed bugs. Our study has used Google search frequency (Google Trends) to assess the growth and spread of public interest. It found that concern over the Paris outbreak spread to neighbouring countries and was an inverse function of distance. Health issues are a popular topic in science journalism and articles with bad news, threat, continuity and geographic proximity helped generate considerable media activity such that the public perceptions of the problem were enhanced and suggests that government agencies need to collect well standardised data on bed bug occurrence. Google Trends proved a sensitive tool to follow the public concern over an insect that invokes considerable dread.

Simple Summary: Over the past 20 years, there has been a worldwide resurgence in the nuisance biting insects known as bed bugs. The resurgence is due to the development of insecticide resistance, such that modern bed bugs are very hard to kill. This also means that control is expensive, and not everyone has the financial resources to pay for eradication. In an effort to develop cheaper control solutions, we investigated the use of "lethal harborages". Here, pieces of cardboard were treated with different insecticides, two strains of bed bugs (one that is easy to kill and one that is resistant) were allowed to enter the treated cardboards, and mortality was recorded. The cardboard treated with silica dioxide were the most effective, causing 100% of the bugs to die within 14 to 17 days when using the highest dose. This silica dioxide dust was also tested in an artificial environment. By day 21, all the bugs in the treated settings were eliminated. These "lethal harborages" were highly effective in the laboratory and are a potential low-cost solution as part of an overall bed bug control plan. Over the past two decades, there has been a worldwide resurgence in the bed bugs Cimex lectularius L. and Cimex hemipterus (F.). This is primarily due to insecticide resistance, making bed bug management and eradication challenging and expensive. To address the need for more affordable control solutions, "lethal harborages" were explored. Cardboard squares were treated using insecticidal dusts at different dosage levels, including silica dioxide, diatomaceous earth, deltamethrin, permethrin, and fipronil. Two strains of C. lectularius, one susceptible and one resistant, were allowed to enter the treated harborages, and mortality rates were recorded daily. The silica dioxide products proved to be the most efficacious, consistently achieving 100% mortality between 14–17 d at the highest dose. An artificial environment trial using the "new ChinChex®" formulation of silica dioxide resulted in the complete elimination of bed bugs in the treated harborages within 21 d. These findings suggest that lethal harborages, especially those impregnated with silica dioxide, offer a cost-effective solution that could be incorporated into broader integrated bed bug management strategies. Abstract In the last few years, the bed bug Cimex lectularius has been an increasing problem worldwide, mainly due to the development of insecticide resistance to pyrethroids. The characterization of resistance alleles is a prerequisite to improve surveillance and resistance management. To identify genomic variants associated with pyrethroid resistance in Cimex lectularius, we compared the genetic composition of two recent and resistant populations with that of two ancient‐susceptible strains using a genome‐wide pool‐seq design. We identified a large 6 Mb “superlocus” showing particularly high genetic differentiation and association with the resistance phenotype. This superlocus contained several clustered resistance genes and was also characterized by a high density of structural variants (inversions, duplications). The possibility that this superlocus constitutes a resistance “supergene” that evolved after the clustering of alleles adapted to insecticide and after reduction in recombination is discussed.

Bed bugs produce volatile aldehydes that have alarm and aggregation functions. Using two synanthropic bed bug species, Cimex lectularius L. and C. hemipterus (Fabricius), developmental changes were examined for (E)-2-hexenal, 4-oxo-(E)-2-hexenal, (E)-2-octenal, and 4-oxo-(E)-2-octenal, the four most abundant aldehydes shared between the two species. Quantitative analyses of the aldehydes in the nymphal exuviae indicated that the aldehydes' ratio remained similar throughout nymphal development. In general, (E)-2-octenal was most abundant, and (E)-2-hexenal and 4-oxo-(E)-2-octenal were least abundant. The fourth aldehyde, 4-oxo-(E)-2-hexenal, was present in intermediate quantities. The quantities and percent abundances of the aldehydes in nymphal exuviae and the adults were significantly different between C. lectularius and C. hemipterus. The ratio between (E)-2-hexenal and (E)-2-octenal was determined in adult male and female bed bugs of each species. Adult C. hemipterus had a higher proportion of (E)-2-hexenal than C. lectularius, while no sex differences were found. This work provides the first systematic quantification of four aldehydes [(E)-2-hexenal, 4-oxo-(E)-2-hexenal, (E)-2-octenal and 4-oxo-(E)-2-octenal] for all five of the nymphal stages for both C. lectularius and C. hemipterus.

Abstract Background Widespread vector control has been essential in reducing the global incidence and prevalence of malaria, despite now stalled progress. Long-lasting insecticide-treated nets (LLINs) have historically been, and remain, one of the most commonly used vector control tools in the campaign against malaria. LLINs are effective only with proper use, adherence, retention and community adoption, which historically have relied on the successful control of secondary pests, including bed bugs. The emergence of pyrethroid-resistant bed bugs in malaria-endemic communities and failure to control infestations have been suggested to interfere with the effective use of LLINs. Therefore, the behavioral interactions of bed bugs with commonly used bed nets should be better understood. Methods To investigate the interactions between bed bugs (Cimex lectularius L.) and LLINs, insecticide-susceptible and pyrethroid-resistant bed bugs were challenged to pass through two commonly used LLINs in two behavioral assays, namely host (blood meal)-seeking and aggregation-seeking assays. The proportions blood-fed and aggregated bed bugs, aggregation time and mortality were quantified and analyzed in different bed bug life stages. Results Overall, both the insecticide-susceptible bed bugs and highly resistant bed bugs showed a varying ability to pass through LLINs based on treatment status and net design. Deltamethrin-treated nets significantly impeded both feeding and aggregation by the susceptible bed bugs. While none of the tested LLINs significantly impeded feeding (passage of unfed bed bugs through the nets) of the pyrethroid-resistant bed bugs, the untreated bed net, which has small mesh holes, impeded passage of fed bed bugs. Mortality was only seen in the susceptible bed bugs, with significantly higher mortality on deltamethrin-treated nets (63.5 ± 10.7%) than on permethrin-treated nets (2.0 ± 0.9%). Conclusions Commonly used new LLINs failed to prevent the passage of susceptible and pyrethroid-resistant bed bugs in host- and aggregation-seeking bioassays. The overall low and variable mortality observed in susceptible bed bugs during both assays highlighted the potential of LLINs to impose strong selection pressure for the evolution of pyrethroid resistance. Already, the failure to control bed bug infestations has been implicated as a contributing factor to the abandonment or misuse of LLINs. For the first time to our knowledge, we have shown the potential of LLINs in selecting for resistant secondary pest populations and so their potential role in stalling malaria control programs should be further investigated. Abstract Background The common bed bug, Cimex lectularius L., is a hematophagous ectoparasite that was a common pest in poultry farms through the 1960s. Dichlorodiphenyltrichloroethane (DDT) and organophosphates eradicated most infestations, but concurrent with their global resurgence as human ectoparasites, infestations of bed bugs have been reappearing in poultry farms. Although the impact of bed bugs on chicken health has not been quantified, frequent biting and blood-feeding are expected to cause stress, infections and even anemia in birds. Bed bug control options are limited due to the sensitive nature of the poultry environment, limited products labeled for bed bug control and resistance of bed bug populations to a broad spectrum of active ingredients. Veterinary drugs are commonly used to control endo- and ectoparasites in animals. In this study, we evaluated the effects of two common veterinary drugs on bed bugs by treating the host with systemic antiparasitic drugs. Methods We conducted dose–response studies of ivermectin and fluralaner against several bed bug strains using a membrane feeding system. Also, different doses of these drugs were given to chickens and two delivery methods (topical treatment and ingestion) were used to evaluate the efficacy of ivermectin and fluralaner on bed bug mortality. Results Using an artificial feeding system, both ivermectin and fluralaner caused high mortality in insecticide-susceptible bed bugs, and fluralaner was found to be effective on pyrethroid- and fipronil-resistant bed bugs. Ivermectin was ineffective in chickens either by the topical treatment or ingestion, whereas bed bugs that fed on chickens which had ingested fluralaner suffered high mortality when feeding on these chickens for up to 28 days post treatment. Conclusions These findings suggest that systemic ectoparasitic drugs have great potential for practical use to control bed bug infestations in poultry farms. These findings also demonstrate the efficacy of fluralaner (and potentially other isoxazolines) as a potent new active ingredient for bed bug control. Graphical Abstract

In the last few years, the bed bug Cimex lectularius has been an increasing problem worldwide, mainly due to the development of insecticide resistance to pyrethroids. The characterization of resistance alleles is a prerequisite to improve surveillance and resistance management. To identify genomic variants associated with pyrethroid resistance in Cimex lectularius, we compared the genetic composition of two recent and resistant populations with that of two ancient‐susceptible strains using a genome‐wide pool‐seq design. We identified a large 6 Mb "superlocus" showing particularly high genetic differentiation and association with the resistance phenotype. This superlocus contained several clustered resistance genes and was also characterized by a high density of structural variants (inversions, duplications). The possibility that this superlocus constitutes a resistance "supergene" that evolved after the clustering of alleles adapted to insecticide and after reduction in recombination is discussed. Bed bugs are considered a major public health problem in industrialized countries. Usually, bed bug infestations are managed using a combination of physical and chemical methods. In recent years, new strategies for bed bug control have emerged, particularly the use of dusts like diatomaceous earth and silicon dioxide. However, in Europe, the use of silicon dioxide is restricted to professional, while diatomaceous earth can be harmful to the lungs. This study aimed to assess bed bug mortality rates associated with Sommières earth, green clay, talc, and sodium bicarbonate compared to silicon dioxide and diatomaceous earth from a pest management company, diatomaceous earth for litter conditioner, and diatomaceous earth from a supermarket. We tested permanent exposure, short exposure, horizontal transfer and repellent effect on two bed bug colonies. Sommières earth demonstrated efficacy ranging from 75% to 100% in permanent and short exposures, similar to the efficacy of diatomaceous earth from the pest management company. On the contrary, diatomaceous earth for litter conditioner and diatomaceous earth from a supermarket, green clay, talc, and sodium bicarbonate were found to be ineffective. This study demonstrates, for the first time, the efficacy of Sommières earth against bed bugs, but also highlights the variability in efficacy of diatomaceous earths on bed bugs depending on their quality.

This study evaluates the use of chitosan–silver nanocomposites (CSN) as an insecticide against the bed bug (Cimex lectularius). Adult bed bugs were collected from infested residential areas and identified using light microscopy and scanning electron microscopy. CSN were prepared and photographed for characterization using transmission electron microscopy, dynamic light scattering, and zeta potential. The insecticidal effect of different concentrations of CSN (400– 1000 ppm) was compared to that of 0.1% cypermethrin as a positive control and normal saline as a negative control. The bugs (n = 25) were immersed for 20 min in the corresponding medium, dried with filter papers, and then incubated at 27–28°C and 70% RH with a 12:12 h light–dark photoperiod. The mortality rates were recorded at different time intervals (2, 4, 6, 12, and 24 h post-incubation (hPI)), and the entire experiment was repeated five times. Image analysis showed round- to spherical-shaped CSN ranging in size from 34 to 72 nm. The mortality rates were positively associated with increasing concentrations of CSN. The mortality rate first reached 100% for concentrations of 800 ppm at 24 hPI and 1000 ppm at 12 hPI. The calculated LC50 was found at a concentration of 1165 ppm at 2 hPI, and the LC99 was found at a concentration of 1914 ppm at 2 hPI. The positive control, cypermethrin, induced 100% mortality among the bugs at 2 hPI, while the negative control caused no mortality. These results clearly show the potential of CSN as an insecticide against C. lectularius. Future studies on best practices for implementing these particles in clinical settings are recommended. Bed bugs are an important group of medical and urban insect pests. They are obligate blood-feeders. Their bites may cause skin irritation and allergic reactions and, under some circumstances, may lead to mental and other health issues. Despite numerous discoveries on the biology of these obnoxious pests and progress in control strategies over the last two decades, bed bugs continue to preferentially plague those from low socioeconomic communities because the poor generally could not afford effective control options. As a result, such infestations in poorer communities serve as a reservoir for wider society. This Special Collection of the Perspective on Biology and Management of Bed Bugs presents nine original research papers on bed bug detection, insecticide performance and resistance, nonchemical treatment, fungal biopesticides, and pest management procurement and contracts. Modern bed bugs are resistant to multiple insecticide classes, particularly the pyrethroids. The efficacy of pyrethroid-impregnated mattress liners marketed for bed bug management has been variable. This study evaluated the efficacy of a permethrin-impregnated mattress liner, ActiveGuard, against 24 bed bug strains, consisting of both Cimex hemipterus (F.) and Cimex lectularius L. A 'mat assay', employing an allethrin-impregnated mat, was used to establish the pyrethroid resistance profile of all strains. Three experiments were conducted to evaluate the effect of ActiveGuard exposure on bed bug knockdown: 1) exposing the bed bugs continuously on the liner for up to 24 d, 2) holding the bed bugs on the liner for either 4 or 6 h, and 3) placing a noninsecticide treated fabric above the liner with the bed bugs held continuously on top. Our results indicated that all modern strains (collected within the last 15 years during the current resurgence) were pyrethroid-resistant, although the magnitude of resistance was highly variable between strains. In the continuous exposure study, an incomplete knockdown was recorded for most modern bed bug strains, with some having no knockdown even up to 7 d of constant exposure. In the 4 or 6 h exposure study, the level of knockdown was reduced even further, and very few bed bugs were knocked down in the double fabric study. The results of this study indicate that pyrethroid-impregnated mattress liners are not likely to be effective in the management of most modern bed bug infestations involving either C. hemipterus or C. lectularius. Over the past three decades, the bed bug Cimex lectularius has resurged as a prominent indoor pest on a global scale. Knockdown-associated insecticide resistance (kdr) involving the voltage-gated sodium channel, targeted by organochlorine and pyrethroid insecticides, was first reported in C. lectularius within a few years of the widespread use of dichlorodiphenyltrichloroethane (DDT) and has been implicated as a significant factor contributing to the species' recent resurgence. Since then, selection with pyrethroid insecticides has intensified, yet little is known regarding its short-term impacts on the frequency of kdr-associated mutations. Here, we report temporal changes in the frequencies of three kdr-associated mutations in C. lectularius populations collected across the USA from two time periods, sampled approximately a decade apart. The results reveal a significant increase in the frequencies of kdr-associated mutations over this period and the absence of the insecticide-susceptible genotype in recent collections. Furthermore, a significant transition was observed toward infestations possessing multiple kdr-associated mutations. These findings suggest that the persistent use of pyrethroid insecticides over the past decade continues to impose strong selection pressure on C. lectularius populations, driving the proliferation of kdr-associated mutations. Simple Summary: Bed bugs are hematophagous insects that have been described as a cause of nuisance since ancient times. Two species are known to parasitize humans: Cimex lectularius, prevalent in temperate areas, and C. hemipterus, also known as the tropical bed bug. A global increase in infestation has been observed in recent decades. However, data is lacking concerning species distribution, which may be affected by population movements (e.g., travelers, migrants). Moreover, although these insects are not known to harbor and transmit infectious agents to humans, continuous monitoring of their infection status is part of active surveillance. We analyzed bed bug specimens collected from beddings in Mauritania, and assessed MALDI-TOF MS as an identification tool for immature specimens. Moreover, we also conducted bacterial detection in each specimen. Overall, our findings supported MALDI-TOF MS as being a robust tool allowing species level identification of immature Cimex specimens. Human infestations by bed bugs have upsurged globally in recent decades, including in African countries, where recent reports pointed out an increase in infestation. Sympatric dwelling has been described for two species of bed bug parasitizing humans: Cimex hemipterus (the tropical bed bug) and C. lectularius. Identification of these two species is based on morphological characteristics, and gene sequencing, and may also rely on Matrix-Assisted Laser Desorption Ionization Time-Of-Flight Mass Spectrometry (MALDI-TOF MS). The present work aimed to assess whether MALDI-TOF MS was applicable for species level identification of immature stages of Cimex. Arthropods were collected in domestic settings in Nouakchott, Mauritania. Identification used morphological keys and MALDI-TOF MS identification was assessed for immature stages. Quantitative PCR and sequencing assays were used to detect arthropod-associated bacteria in each specimen. A total of 92 arthropods were collected, all morphologically identified as C. hemipterus (32 males, 14 females and 45 immature stages). A total of 35/45 specimens produced good quality MALDI-TOF MS spectra. Analysis allowed species level identification of all immature C. hemipterus after their spectra were entered into our in-house MALDI-TOF MS arthropod spectra database. Molecular screening allowed detection of Wolbachia DNA in each specimen. These results suggested that MALDI-TOF MS is a reliable tool for species level identification of Cimex specimens, including immature specimens. Future studies should assess this approach on larger panels of immature specimens for different Cimex species and focus on the precise staging of their different immature developmental stages. Bed bugs (Hemiptera: Cimicidae) are an important group of obligate hematophagous urban insect pests. The global resurgence of bed bugs, involving the common bed bug, Cimex lectularius L., and the tropical bed bug, Cimex hemipterus (F.), over the past two decades is believed to be primarily due to the development of insecticide resistance, along with global travel and poor pest management, which have contributed to their spread. This review examines and synthesizes the literature on bed bug origins and their global spread and the literature on historical and contemporary control options. This includes bed bug prevention, detection and monitoring, nonchemical and chemical control methodologies (and their limitations), and potential future control options. Future research needs are highlighted, especially the factors behind the modern resurgence, the necessity of identifying differences between the two bed bug species relevant to control, and the need to improve insecticide test protocols and management strategies. Over the past two decades, there has been a worldwide resurgence in the bed bugs Cimex lectularius L. and Cimex hemipterus (F.). This is primarily due to insecticide resistance, making bed bug management and eradication challenging and expensive. To address the need for more affordable control solutions, “lethal harborages” were explored. Cardboard squares were treated using insecticidal dusts at different dosage levels, including silica dioxide, diatomaceous earth, deltamethrin, permethrin, and fipronil. Two strains of C. lectularius, one susceptible and one resistant, were allowed to enter the treated harborages, and mortality rates were recorded daily. The silica dioxide products proved to be the most efficacious, consistently achieving 100% mortality between 14–17 d at the highest dose. An artificial environment trial using the “new ChinChex®” formulation of silica dioxide resulted in the complete elimination of bed bugs in the treated harborages within 21 d. These findings suggest that lethal harborages, especially those impregnated with silica dioxide, offer a cost-effective solution that could be incorporated into broader integrated bed bug management strategies. This approach may help alleviate the burden of bed bug infestations in economically disadvantaged communities.

Background: Widespread vector control has been essential in reducing the global incidence and prevalence of malaria, despite now stalled progress. Long-lasting insecticide-treated nets (LLINs) have historically been, and remain, one of the most commonly used vector control tools in the campaign against malaria. LLINs are effective only with proper use, adherence, retention and community adoption, which historically have relied on the successful control of secondary pests, including bed bugs. The emergence of pyrethroid-resistant bed bugs in malaria-endemic communities and failure to control infestations have been suggested to interfere with the effective use of LLINs. Therefore, the behavioral interactions of bed bugs with commonly used bed nets should be better understood. Methods: To investigate the interactions between bed bugs (Cimex lectularius L.) and LLINs, insecticide-susceptible and pyrethroid-resistant bed bugs were challenged to pass through two commonly used LLINs in two behavioral assays, namely host (blood meal)-seeking and aggregation-seeking assays. The proportions blood-fed and aggregated bed bugs, aggregation time and mortality were quantified and analyzed in different bed bug life stages. Results: Overall, both the insecticide-susceptible bed bugs and highly resistant bed bugs showed a varying ability to pass through LLINs based on treatment status and net design. Deltamethrin-treated nets significantly impeded both feeding and aggregation by the susceptible bed bugs. While none of the tested LLINs significantly impeded feeding (passage of unfed bed bugs through the nets) of the pyrethroid-resistant bed bugs, the untreated bed net, which has small mesh holes, impeded passage of fed bed bugs. Mortality was only seen in the susceptible bed bugs, with significantly higher mortality on deltamethrin-treated nets (63.5 ± 10.7%) than on permethrin-treated nets (2.0 ± 0.9%). Conclusions: Commonly used new LLINs failed to prevent the passage of susceptible and pyrethroid-resistant bed bugs in host- and aggregation-seeking bioassays. Sexual conflict is ubiquitous across the animal kingdom and often involves costly sexual harassment of females by males. An overlooked outcome of sexual conflict is its potential impact on social behaviour. Due to their seemingly harmful mode of copulation (traumatic insemination) and tendency to form aggregations, bed bugs are an ideal model for studying the social implications of sexual conflict. Repeated traumatic inseminations are known to reduce some aspects of female fitness, so we expected the benefits to males and the high costs for females of frequent mating to result in divergent social preferences between the sexes. To examine the impact of sexual harassment on social structure, we devised a novel experimental arena with either two shelters or 12 shelters and continuously tracked sexual and social interactions between individually marked bed bugs over 6 days. By constructing aggregation networks, we examined whether female bed bugs occupied more peripheral network positions compared to males as well as whether females preferentially associated with other females as a strategy to reap the benefits of group living while mitigating the costs of unsolicited sexual attention. We found no evidence that females shape their social environment to evade associating with males. However, when tested individually in a follow-up experiment, mated females showed a strong preference for social cues from females over social cues from males. Our results therefore suggest that males and females may be in conflict over the composition of social associations and highlight the importance of both examining behaviour at the individual level and tracking larger groups of freely interacting populations in more complex environments. • Sexual conflict may differentially influence social behaviour of females and males. • We constructed bed bug social networks to see whether females socially evade males. • Aggregation networks did not show female avoidance through decreased sociality. • Social networks were also not assorted by sex. • However, females preferred social cues associated with other females over males. The presence of fecal spots has often been used to verify the existence of a bed bug (Cimex lectularius) infestation. However, no research has been conducted to determine how much fecal material that a bed bug population produces over time. In this study, the number of fecal spots that each nymphal life stage was capable of producing after a bloodmeal was quantified. Adult fecal spots were also quantified to determine if there was consistent production between feedings. During this study, it was discovered that bed bugs produced visible fecal spots and clear spots that were only visible under ultraviolet light. Therefore, three types of fecal spots were quantified: dark feces, light feces, and clear spots. Clear spots were produced in greater amounts (38.5–55.5%) than either dark spots (27.3–40.7%) or light spots (17.3–21.9%). For example, 5th instar bed bugs were thought to produce an average of 21 spots (dark and light) after a single bloodmeal. However, using the ultraviolet light, it was found that the 5th instars actually produced an average of 44.7 spots. Using the total fecal spot data collected during this study, researchers could project contamination potential for an infestation starting with a single gravid female over 30, 60, and 90 d. In addition, the amount of area covered by these spots was projected to be over 12 m2 in just 3 mo, which could greatly reduce the environmental and aesthetic quality of a home. Over the last two decades, an increase in bed bug infestations has been observed worldwide. Although their definitive role as vectors of infectious agents has not yet been demonstrated, bed bugs have a direct effect on human health through dermatological reactions to their bites and psychological disorders linked to domestic infestations. In this study, the effectiveness of using MALDI-TOF MS to correctly identify these two bed bug species at immature stages was assessed, as well as it effectiveness as discriminating between the immature stages (IS) of C. lectularius and C. hemipterus and their associated developmental stages. A total of 305 specimens were subjected to MALDI-TOF MS analysis, including 153 C. lectularius (28 eggs and 25 nymphs per stage from IS1 to IS5) and 152 C. hemipterus (27 eggs and 25 nymphs per stage from IS1 to IS5). ). MALDI-TOF MS analysis enabled us to obtain 84.97% (130/153) of high-quality MS spectra in terms of reproducibility and profile intensity. Twenty-four spectra including two per stage, from egg to IS5, and per bed bug species - were added to our in-house MS reference arthropod spectra database. All specimens were correctly identified at the species level, independently of the developmental stage, with log score values (LSVs) ranging from 1.75 to 2.79 (mean = 2.29 ± 0.12) and 1.81 to 2.71 (mean = 2.37 ± 0.03) for C. lectularius and C. hemipterus, respectively. MALDI-TOF MS correctly classified 53,33% (104/195) of the Cimex at the correct immature stage. Conversely, an accurate comparison of the profiles with a Genetic Algorithm model underlined that grouping the immature stages in two groups, early (IS1-IS2) and late (IS3-IS4-IS5), made it possible to obtain a cross validation (CV) and recognition capability (RC) greater than 92% and 94%, respectively, for both species. The objective was to evaluate the clinical characteristics, triage information, diagnostic evaluation, and disposition for emergency department patients with and without bed bug infestation. A retrospective case–control study of patients with and without bed bugs was performed from February 1, 2011, through February 1, 2017. Cases (n = 332) and controls (n = 4,952) were matched by age, sex, and emergency department location. Patient characteristics and clinical information were compared between groups. On univariable and multivariable analysis, patients with bed bugs had higher heart rate, lower systolic blood pressure, higher pain scores, and more frequent tobacco use in the past year. They were also more likely to screen positive in triage for an unsafe home; require an abuse consultation; be diagnosed in the emergency department or inpatient setting with malnutrition, marasmus, and/or cachexia; have more emergency department and hospital visits; and have longer hospital encounter lengths of stay (all p ≤ 0.02). This study aimed to review published scientific literature on bed bugs in countries where insecticide resistance has been reported worldwide from 2000 to 2021. Electronic databases, including Scopus, PubMed, and WOS, were searched. Out of 606 articles found in the initial search, we selected 57 articles, of which 40 articles had reported on Cimex lectularius (C. lectularius), and 22 papers had reported on Cimex hemipterus (C. hemipterus). Most studies on insecticide resistance were carried out on C. lectularius in North America (14, 35%) and C. hemipterus in Asia (16, 72.7%). The most common method used to detect bed bug resistance to insecticides was toxicological bioassay with an overall random pooled effect size of 0.38 (95% CI: 0.23–0.53) in C. lectularius and 0.46 (95% CI: 0.27–0.65) in C. hemipterus. Resistance to pyrethroids was reported against C. lectularius with an overall pooled effect size of 0.75 (95% CI: 0.56–0.94) and C. hemipterus with an overall pooled effect size of 0.81 (95% CI: 0.57–0.93) in 33.40 (82.5%) and 19.22 (86.3%) published articles, respectively. A very high resistance level to pyrethroids in both studied species was observed, and resistance ratios at the highest level were 76389.3 and 315.5 in C. lectularius and C. hemipterus, respectively. Resistance mechanisms against pyrethroids were reported from most locations except Iran and Thailand, but these mechanisms were not studied in other insecticide groups. These reports indicate that chemical control options for bed bugs are limited. Therefore, a combination of chemical and non-chemical strategies is recommended for bed bug control. Background and purpose: Control of bedbugs (Cimex lectularius) as important human pests often fails in places where people live in aggregation such as prisons. The aim of this study was to compare the effectiveness of various methods of bedbug control in prisons in Mazandaran province, Iran. Materials and methods: In this descriptive-analytical study performed in 2018, routine control methods of bedbugs including spraying, environmental management, fogging, and flame and combinations of two methods were compared. Data analyses were carried out using two methods. In first method, the number of bites before the control program along with that of four monitoring periods (15 days interval) after the interventions normalized with the population of the prison was analyzed. In second method, we analyzed the data from four monitoring phases after the control operation without considering the population of prisons. Results: Environmental management plus flame were the pair of control intervention with highest impact on bedbug population in prisons. Based on first method of analysis, there was a significant difference in number of individuals bitten between various control methods in all prisons (P0.05). Conclusion: Environmental sanitation of prison wards plus flame was found to have the greatest impact on bedbug population over a period of one month. Where it is not possible to use flame, combined methods, including spraying-environmental sanitation or fogging-environmental sanitation can be used as effective alternatives in reducing bedbug population in prisons.

The worldwide resurgence of bed bugs [both Cimex lectularius L. and Cimex hemipterus (F.)] over the past two decades is believed in large part to be due to the development of insecticide resistance. The transcriptomic and genomic studies since 2010, as well as morphological, biochemical and behavioral studies, have helped insecticide resistance research on bed bugs. Multiple resistance mechanisms, including penetration resistance through thickening or remodelling of the cuticle, metabolic resistance by increased activities of detoxification enzymes (e.g. cytochrome P450 monooxygenases and esterases), and knockdown resistance by kdr mutations, have been experimentally identified as conferring insecticide resistance in bed bugs. Other candidate resistance mechanisms, including behavioral resistance, some types of physiological resistance (e.g. increasing activities of esterases by point mutations, glutathione S-transferase, target site insensitivity including altered AChEs, GABA receptor insensitivity and altered nAChRs), symbiont-mediated resistance and other potential, yet undiscovered mechanisms may exist. This article reviews recent studies of resistance mechanisms and the genes governing insecticide resistance, potential candidate resistance mechanisms, and methods of monitoring insecticide resistance in bed bugs. This article provides an insight into the knowledge essential for the development of both insecticide resistance management (IRM) and integrated pest management (IPM) strategies for successful bed bug management.

The mechanical properties of bed bug (Cimex lectularius L.) tarsi and pretarsi were investigated in order to evaluate their vulnerability to piercing by plant trichomes (sharp microscopic hairs). Nanoindentation was used to measure the force required to insert a sharp probe into the cuticle of these different regions, as well as to determine creep and reduced elastic moduli for the cuticle. Scanning electron microscopy was used to visualize the indents that had been generated by nanoindentation. The force required to insert a cube corner nanoindenter probe into the cuticle was determined for a range of displacements (1 to 9 μm) and strain rates (0.003 to 0.5 s-1). Greater force was required to insert this sharp probe at greater depth or at faster strain rates. A specific region of the pretarsus (membrane with microtrichia) more frequently pierced by trichomes during bed bug locomotion required approximately 20-30% less force, exhibited more creep, and had a lower reduced elastic modulus for the first micron of indentation compared to the other regions; although this pattern was not consistent for greater displacements. These mechanical attributes, which will facilitate the initial stage of puncture in addition to the presence of natural infoldings in the cuticle of this area, may make that area of the pretarsus particularly vulnerable to piercing. This information will help inform development of physical methods for control of insect pests such as bed bugs.

BackgroundThe resistance of bed bugs (Cimex lectularius L.) to chemical insecticides has motivated the development of non-chemical control methods such as heat treatment. However, because bed bugs tend to hide in cracks or crevices, their behavior incidentally generates a thermally insulated microenvironment for themselves. Bed bugs located on the outer surface of luggage are less insulated and potentially more vulnerable to brief heat treatment.ResultsSoft-sided suitcases with adult male bed bugs on the outside were exposed to an air temperature of 70-75 °C. It took 6 min to kill all of the bed bugs, even those that had concealed themselves under zipper flaps or decorative piping. During heating, only one bed bug (out of 250 in total) moved into the luggage (through a closed zipper). Over long periods of time (24 h) at room temperature, adult male bed bugs on the exterior of luggage only infrequently moved inside; only 3% (5/170) had moved inside during 24 h.ConclusionsBrief exterior heat treatment of luggage is a promising way to reduce the spread of bed bugs being transported on the outer surface of luggage. This treatment will not kill bed bugs inside the luggage, but could be a component of integrated management for this pest. © 2016 Society of Chemical Industry.

Bed bugs (Hemiptera: Cimicidae) are widely distributed, obligately blood-feeding insects, but they have never been linked to pathogen transmission in humans. Most other hematophagous insects that frequently bite humans transmit pathogens, and it is unclear why bed bugs do not. One hypothesis is that bed bugs have evolved a highly robust immune system because their mating system, traumatic insemination, exposes females to consistent wounding and bacterial infections. Although this has been proposed, very little is known about the bed bug immune system and how bed bugs respond to microbial challenges introduced by wounding. Similarly, there is little known about how the bed bug immune system responds to human pathogens. Understanding the bed bug immune system could give insight to why bed bugs appear not to transmit disease and under what circumstances they could, while also facilitating biological control efforts involving microbes. To investigate the transcriptomic response of bed bugs to immune challenges, we exposed female bed bugs to three bacterial challenges. 1.) Pseudomonas fluorescens , an entomopathogen known to have harmful effects to bed bugs, 2.) bacteria cultured from a bed bug enclosure (99.9 % Bacillus spp.), likely encountered during traumatic insemination, and 3.) Borrelia duttoni , a human vector-borne pathogen that causes relapsing fever. We compared the transcriptomes of infected bed bugs with uninfected matched controls in a pairwise fashion, focusing on immune-related genes. We found many known antimicrobial effector genes upregulated in response to P. fluorescens and traumatic insemination-associated bacteria, but interestingly, not in response to B. duttoni. In the differentially expressed genes that were shared between experiments, we found significant overlap in the P. fluorescens treatment and the traumatic insemination bacteria treatment, and between the P. fluorescens and B. duttoni treatments, but not between the traumatic insemination bacteria treatment and the B. duttoni treatment. Finally, we identify previously overlooked candidates for future studies of immune function in bed bugs, including a peroxidase-like gene, many putative cuticle-associated genes, a laccase-like gene, and a mucin-like gene.
