This study is aimed at examining the mesoscopic mechanical response and crack development characteristics of asphalt mixtures using the three-dimensional discrete element approach via particle flow code (PFC). The material is considered an assembly of three phases of aggregate, mortar, and voids, for which three types of contact are identified and described using a modified Burgers model allowing for bond failure and crack formation at contact. The laboratory splitting test is conducted to determine the contact parameters and to provide the basis for selecting three different load levels used in the indirect tensile fatigue test and simulation. The reliability of the simulation is verified by comparing the fatigue lives and dissipated energies against those from the test. Under cyclic loading, the internal tensile and compressive force chains vary dynamically as a response to the cyclic loading; both are initially concentrated beneath the top loading strip and then extend downward along the loading line. The compressive chains are oriented roughly vertically and develop an elliptic shape as damage grows, while the tensile chains are mostly horizontal and become denser. An analysis based on the histories of the numbers of different contact types indicates that damage mainly originates from bond failures among the aggregate particles and at the aggregate–mortar interfaces. In terms of location, cracking is initiated below the loading point (consistent with observations from the force chains) and propagates downward and laterally, leading to the macrocrack along the vertical diameter. The optical features of mineral composition and texture in petrographic thin sections are an important basis for rock identification and rock evolution analysis. However, the efficiency and accuracy of human visual interpretation of petrographic thin section images have depended on the experience of experts for a long time. The application of image-based computer vision and deep-learning algorithms to the intelligent analysis of the optical properties of mineral composition and texture in petrographic thin section images (in plane polarizing light) has the potential to significantly improve the efficiency and accuracy of rock identification and classification. This study completed the transition from simple petrographic thin image classification to multitarget detection, to address more complex research tasks and more refined research scales that contain more abundant information, such as spatial, quantitative and category target information. Oolitic texture is an important paleoenvironmental indicator that widely exists in sedimentary records and is related to shallow water hydraulic conditions. We used transfer learning and image data augmentation in this paper to identify the oolitic texture of petrographic thin section images based on the faster region-based convolutional neural network (Faster RCNN) method. In this study, we evaluated the performance of Faster RCNN, a two-stage object detection algorithm, using VGG16 and ResNet50 as backbones for image feature extraction. Our findings indicate that ResNet50 outperformed VGG16 in this regard. Specifically, the Faster RCNN model with ResNet50 as the backbone achieved an average precision (AP) of 92.25% for the ooids test set, demonstrating the accuracy and reliability of this approach for detecting ooids. The experimental results also showed that the uneven distribution of training sample images and the complexity of images both significantly affect detection performance; however, the uneven distribution of training sample images has a greater impact. Our work is preliminary for intelligent recognition of multiple mineral texture targets in petrographic thin section images. We hope that it will inspire further research in this field. A memristor is a kind of nonlinear two-port circuit element with memory characteristics, whose resistance value is subject to being controlled by the voltage or current on both its ends, and thus it has broad application prospects. At present, most of the memristor application research is based on the change of resistance and memory characteristics, which involves how to make the memristor change according to the desired trajectory. Aiming at this problem, a resistance tracking control method of memristors is proposed based on iterative learning controls. This method is based on the general mathematical model of the voltage-controlled memristor, and uses the derivative of the error between the actual resistance and the desired resistance to continuously modify the control voltage, making the current control voltage gradually approach the desired control voltage. Furthermore, the convergence of the proposed algorithm is proved theoretically, and the convergence conditions of the algorithm are given. Theoretical analysis and simulation results show that the proposed algorithm can make the resistance of the memristor completely track the desired resistance in a finite time interval with the increase of iterations. This method can realize the design of the controller when the mathematical model of the memristor is unknown, and the structure of the controller is simple. Impact craters with layered ejecta deposits are widespread on Mars. Prevailing views suggest that such ejecta were formed due to the involvement of target water and/or water ice in the impact excavation and/or the post-deposition movement of the impact ejecta. The long-runout landslides and lobate debris aprons that are likely formed due to the involvement of water ice are used as analogs to compare roughness at multiple scales, considering that these three landforms share some similarities in their geomorphology. Analog studies of the morphological similarities and differences of layered ejecta deposits with different emplacement mechanisms are an important approach to untangling how layered ejecta deposits might form on Mars and beyond. Earlier morphological comparisons were usually based on qualitative descriptions or one-dimensional topographic roughness characteristics at given azimuths; however, the emplacement processes of layered deposits are recorded in two-dimensional topography and at multiple scales. In this study, we designed a multiwavelet algorithm to characterize the multi-scale topographic roughness of different forms of Martian layered deposits. Our comparisons show that the inner facies of the layered ejecta deposits and long-runout landslides exhibited similar roughness characteristics, and the outer facies of the layered ejecta deposits were more similar in roughness to lobate debris aprons. This study highlights the importance of the spatial resolution of digital terrain models in characterizing fine topographic fluctuations on layered ejecta deposits, providing additional insights into the possible emplacement mechanisms of different parts of layered ejecta deposits. Simple Summary: Salinity can affect the physiological, reproductive, and developmental characteristics of marine molluscs. The Chinese razor clam (Sinonovacula constricta) is a marine bivalve that inhabits estuarine and intertidal habitats where salinity is usually lower when compared to open sea waters. The species is also an economically important subject of aquaculture in China. Here, RNA sequencing was used to study the internal mechanism of salt stress response in the Chinese razor clam. Differentially expressed RNAs were identified and lncRNA/circRNA-miRNA-mRNA regulatory networks were successfully constructed. Furthermore, the expression of four candidate genes in the Chinese razor clam subjected to different salinity stress were examined by qRT-PCR. The results of this study will improve our understanding of genetic-level responses to salinity changes in the Chinese razor clam. Moreover, the acquired information may be useful for optimization of the artificial breeding of the species under aquaculture conditions. The Chinese razor clam (Sinonovacula constricta) is an important for Chinese aquaculture marine bivalve that naturally occurs across intertidal and estuarine areas subjected to significant changes in salinity level. However, the information on the molecular mechanisms related to high salinity stress in the species remain limited. In this study, nine gill samples of S. constricta treated with 20, 30, and 40 ppt salinity for 24 h were used for whole-transcriptome RNA sequencing, and a regulatory network of competing endogenous RNAs (ceRNAs) was constructed to better understand the mechanisms responsible for adaptation of the species to high salinity. A total of 83,262 lncRNAs, 52,422 mRNAs, 2890 circRNAs, and 498 miRNAs were identified, and 4175 of them displayed differential expression pattern among the three groups examined. The KEGG analyses of differentially expressed RNAs evidenced that amino acid synthesis and membrane transport were the dominant factors involved in the adaptation of the Chinese razor clam to acute salinity increase, while lipid metabolism and signaling played only a supporting role. In addition, lncRNA/circRNA-miRNA-mRNA regulatory networks (ceRNA network) showed clearly regulatory relationships among different RNAs. Moreover, the expression of four candidate genes, including tyrosine aminotransferase (TAT), hyaluronidase 4 (HYAL4), cysteine sulfinic acid decarboxylase (CSAD), and ∆1-pyrroline-5-carboxylate synthase (P5CS) at different challenge time were detected by qRT-PCR. The expression trend of TAT and HYAL4 was consistent with that of the ceRNA network, supporting the reliability of established network. The expression of TAT, CSAD, and P5CS were upregulated in response to increased salinity. This might be associated with increased amino acid synthesis rate, which seems to play an essential role in adaptation of the species to high salinity stress. In contrast, the expression level of HYAL4 gene decreased in response to elevated salinity level, which is associated with reduction Hyaluronan hydrolysis to help maintain water in the cell. Our findings provide a very rich reference for understanding the important role of ncRNAs in the salinity adaptation of shellfish. As the silicon photonics field matures and a data-hungry future looms ahead, new technologies are required to keep up pace with the increase in capacity demand. In this paper, we review current developments in the near-IR and mid-IR group IV photonic modulators that show promising performance. We analyse recent trends in optical and electrical co-integration of modulators and drivers enabling modulation data rates of 112 GBaud in the near infrared. We then describe new developments in short wave infrared spectrum modulators such as employing more spectrally efficient PAM-4 coding schemes for modulations up to 40 GBaud. Finally, we review recent results at the mid infrared spectrum and application of the thermo-optic effect for modulation as well as the emergence of new platforms based on germanium to tackle the challenges of modulating light in the long wave infrared spectrum up to 10.7 μ m with data rates of 225 MBaud. Recycle and reuse of waste asphalt materials in the pavement industry has brought tremendous contributions to the infrastructure sustainability and environmental preservation. The recent literature has suggested a great potential of plasticizers to be used for rejuvenating the oxidated paving asphalts. This study was aimed at assessing the rejuvenating effectiveness by rheological characterizations of two typical plasticizers, dibutyl phthalate (DBP) and tributyl citrate (TBC), selected based on the molecular structural differences. The underlying rejuvenating mechanisms were approached using molecular dynamics (MD) simulation, for probing the interactions between the plasticizers and oxidized asphaltenes and examining the outcomes in terms of deagglomeration. The results indicated that both plasticizers were highly effective in restoring the stiffness and elasticity properties as well as fatigue resistance of the aged asphalt. According to the simulations, the two plasticizers were able to deagglomerate the asphaltene associations. Owing to the high polarity and hydroxyl group, TBC appeared to be slightly more efficient in dissociating the asphaltenes, which explained its higher effectiveness in restoring the rheological properties as compared to DBP. Oxidative aging is an inevitable environmental factor that accelerates asphalt pavement deterioration. This study employed a molecular dynamics simulation to investigate the impact of aging on asphalt cement from the perspectives of thermodynamic properties, and diffusion and adhesion characteristics. Results indicate that aging increased bulk density from 1.008 to 1.081 g/cm3 and cohesive energy density by 15.6%, which was attributed to the promoted molecular polarity and intermolecular attractiveness. The enhanced molecular interactions also reduced molecular mobility, which led to an increase in the glass transition temperature by 30 K, suggesting that aging diminished the resistance of asphalt to thermal cracking. Simulations of the diffusion behaviors across different temperatures demonstrated that the Arrhenius relationship described well the temperature dependence of the diffusion coefficient, and that aging considerably slowed down the diffusion process as represented by Arrhenius prefactor D0, which dropped by 38.2%. The asphalt–aggregate adhesion was assessed using layered models with and without a water interlayer of different thicknesses. The adhesion was enhanced upon aging due to the significantly improved electrostatic interactions at the interface. Evaluation of the residual adhesion with the presence of interfacial water suggested that aging would raise the moisture susceptibility of asphalt pavement. The increase in molecular polarity was considered to be highly responsible for these aging consequences, and was thus further investigated via the electrostatic potential surface and dipole moment. Hematite concentration is an important climatic proxy for environmental (climatic) studies of soils and sediments. However, the accurate quantification of naturally occurring hematite has always been a difficult question, especially for those areas with lower hematite concentrations. Diffuse reflectance spectroscopy (DRS) is an effective method for hematite identification and quantification with lower detection limits. In this study, we synthesized a set of samples with well-determined concentrations to explore the exact detectable range of hematite and propose the most effective transfer function between the DRS proxy and hematite concentration. In addition, natural sediments from Inland Asia and the Western Pacific Ocean were used to further test the feasibility of the new transfer function. Results show that the lowest DRS detection limit for hematite could reach ~0.00078%, but is affected by the natural matrix. We also find that the second derivative of the Kubelka–Munk (K–M) function is monotonically correlated with the hematite concentration (0.00078%–100%), but ambiguities exist for the first derivative. Therefore, the second derivative of the K–M function is highly suggested for the hematite quantification, especially when concentration exhibits a wide range of variations. LiDAR (Light Detection And Ranging) technology is an important means to obtain three-dimensional information of trees and vegetation. However, due to the influence of scanning mode, environmental occlusion and mutual occlusion between tree canopies and other factors, a tree point cloud often has different degrees of data loss, which affects the high-precision quantitative extraction of vegetation parameters. Aiming at the problem of a tree laser point cloud being missing, an individual tree incomplete point cloud restoration method based on local features of the point cloud is proposed. The L1-Median algorithm is used to extract key points of the tree skeleton, then the dominant direction of skeleton key points and local point cloud density are calculated, and the point cloud near the missing area is moved based on these features to gradually complete the incomplete point cloud compensation. The experimental results show that the above repair method can effectively repair the incomplete point cloud with good robustness and can adapt to the individual tree point cloud with different geometric structures and correct the branch topological connection errors. Isovalent doping offers a method to enhance the thermoelectric properties of semiconductors, yet its influence on the phonon structure and propagation is often overlooked. Here, we take C d X ( X = T e ,   S e ) compounds as an example to study the role of isovalent doping in thermoelectrics by first-principles calculations in combination with the Boltzmann transport theory. The electronic and phononic properties of C d 8 S e 8 , C d 8 S e 7 T e , C d 8 T e 8 , and C d 8 T e 7 S e are compared. The results suggest that isovalent doping with C d X significantly improves the thermoelectric performance. Due to the similar properties of S e and T e atoms, the electronic properties remain unaffected. Moreover, doping enhances anharmonic phonon scattering, leading to a reduction in lattice thermal conductivity. Our results show that optimized p-type(n-type) ZT values can reach 3.13 (1.33) and 2.51 (1.21) for C d 8 T e 7 S e and C d 8 S e 7 T e at 900 K, respectively. The COVID-19 pandemic significantly disrupted global healthcare systems. The impacts of SARS-CoV-2 infection on urticaria and its management are unknown. This study aimed to collect information about patients with urticaria infected with SARS-CoV-2 and to investigate the impact of SARS-CoV-2 infection on urticaria severity, course, and treatment to better support recovery. This was a questionnaire-based study of patients with urticaria infected with SARS-CoV-2. Changes in urticaria severity (measured with the urticaria activity score (UAS)), course, and treatment were assessed before, during, and after SARS-CoV-2 infection. The mean (±SD) UAS scores were 5.17 ± 1.67, 4.23 ± 1.98, and 4.37 ± 1.93 before, during, and after SARS-CoV-2 infection, respectively (F = 8.839, p < 0.01). The median (IQR) wheal score was 0.464 (0.464, 0.763), 0.464 (0.138, 0.763), and 0.464 (0.138, 0.763) before, during, and after infection, respectively (Kruskal–Wallis H-test, H = 12.230, p = 0.02). The median (IQR) pruritus score was 0.695 (0.395, 0.695), 0.394 (0.123, 0.695), and 0.394 (0.123, 0.695) before, during, and after infection, respectively (Kruskal–Wallis H-test, H = 21.001, p < 0.01). Within the limitations of a questionnaire study, urticaria appears to improve during SARS-CoV-2 infection and worsens slightly after recovery, and the frequency of Western medicine use increases. Since the 1970s, certain areas within the Three-Rivers Headwater Region (TRHR) of China have faced severe land degradation due to the combined effects of climate change and human activities, leading to restricted ecological service functions and hindering the achievement of sustainable development goals (SDGs). Land degradation in the TRHR has received widespread attention. However, the current research mainly focuses on single-dimensional degradation and lacks a comprehensive evaluation of patterns and structures, as well as above-ground and underground assessments. To address this gap, this study employed the SDG indicator 15.3.1 framework, comprehensively considering fragmentation and habitat quality index based on land cover changes, grassland degradation index, and soil water erosion index. These indexes represent the three land degradation pathways of landscape degradation, vegetation degradation, and soil erosion. This study assessed land degradation patterns in the TRHR from 2000 to 2020. Results show that approximately 44.67% of the TRHR experienced land degradation during this period, mainly in meadow-dominated regions. Additionally, 5.64% of the regions experienced the superimposition of two or more land degradation pathways, with the frequent coexistence of soil erosion and grassland degradation, accounting for 4.1% of the affected areas. Landscape degradation affected approximately 2.39% of the regions, characterized by increased grassland fragmentation or habitat quality degradation. In terms of grassland degradation, 22.26% of the regions showed medium degradation, while 7.21% and 5.63% experienced moderate and severe degradation, respectively. Moreover, approximately 13.36% of the region faced a worsening situation of soil erosion. Approximately 55.34% of the study area underwent land improvement, with significant enhancements mainly concentrated in the western and eastern regions. The regrowth of grassland in the western region and the enhancement and homogenization of grassland productivity in the eastern region played pivotal roles in promoting land improvement. This study provides critical insights into the land degradation pattern in the TRHR over the past 20 years, offering valuable references for formulating and implementing measures to protect and construct the ecological security barrier of the plateau. Simple Summary: Previous studies have reported how the silk production ability of Bombyx mori can be enhanced, but the mechanism that regulates silk protein genes remains unclear. We performed co-expression network analysis using networkz, an in-house program, which led to the identification of 91 transcription factors were co-expressed with silk protein genes. Of them, 13 transcripts were identified to be novel regulatory factors by time-course expression analysis during the fifth instar larvae stage. Their expression patterns were highly relevant to those of silk protein genes. Our results suggest that the two-step expression screening was robust and highly sensitive to screen relative genes, and a complex mechanism regulates silk protein production in B. mori. The novel candidates that were identified herein can serve as key genes to develop methods to enhance the silk protein production ability of B. mori. Bombyx mori is an important economic insect and an animal model in pharmacomedical research. Although its physiology has been studied for many years, the mechanism via which silk protein genes are regulated remains unclear. In this study, we performed two-step expression screening, namely co-expression network and time-course expression analyses to screen silk protein regulation factors. A co-expression network analysis using RNA-seq data that were obtained from various tissues, including the silk glands of B. mori, was performed to identify novel silk protein regulatory factors. Overall, 91 transcription factors, including some known ones, were found to be co-expressed with silk protein genes. Furthermore, time-course expression analysis during the fifth instar larvae stage revealed that the expression pattern of 13 novel transcription factors was highly relevant to that of silk protein genes and their known regulatory factor genes. In particular, the expression peak of several transcription factors (TFs) was detected before the expression of silk protein genes peak. These results indicated that a larger number of genes than expected may be involved in silk protein regulation in B. mori. The dynamic modulus is a key property determining the short- and long-term performance of asphalt pavement, and its strong dependence on confining pressure and material density (mixture compactness) has been clearly indicated in the literature. It is always challenging to reproduce three-dimensional in situ stress conditions in the laboratory. To alleviate this difficulty, in this study, a convenient experimental setup was developed, in which the lateral confinement was made present and variable as a concomitant reaction of the surrounding materials to the vertical loading. Three dense-graded mixtures were prepared to a set of four different densities and then subjected to the confined dynamic modulus test. The results indicated a significant dependence of the confined modulus on the three factors of temperature, frequency, and compactness and that the mixture with coarser gradation demonstrated a less sensitivity to these parameters. A mathematical model was developed for the dynamic modulus master curve unifying these factors by means of horizontal shifting due to the time–temperature superposition principle (validated against the variable confinement at different compactness) and the vertical shift factor as a function of reduced frequency and compactness. Biomedical alloys are essential parts of modern biomedical applications. However, they cannot satisfy the increasing requirements for large-scale production owing to the degradation of metals. Physical surface modification could be an effective way to enhance their biofunctionality. The main goal of this review is to emphasize the importance of the physical surface modification of biomedical alloys. In this review, we compare the properties of several common biomedical alloys, including stainless steel, Co–Cr, and Ti alloys. Then, we introduce the principle and applications of some popular physical surface modifications, such as thermal spraying, glow discharge plasma, ion implantation, ultrasonic nanocrystal surface modification, and physical vapor deposition. The importance of physical surface modifications in improving the biofunctionality of biomedical alloys is revealed. In recent years, deep learning has become the mainstream development direction in the change-detection field, and its accuracy and speed have also reached a high level. However, the change-detection method based on deep learning cannot predict all the change areas accurately, and its application is limited due to local prediction defects. For this reason, we propose an interactive change-detection network (ICD) for very high resolution (VHR) based on a deep convolution neural network. The network integrates positive- and negative-click information in the distance layer of the change-detection network, and users can correct the prediction defects by adding clicks. We carried out experiments on the open source dataset WHU and LEVIR-CD. By adding clicks, their F1-scores can reach 0.920 and 0.912, respectively, which are 4.3% and 4.2% higher than the original network. To better evaluate the correction ability of clicks, we propose a set of evaluation indices—click-correction ranges, which is suitable for evaluating clicks, and we carry out experiments on the above models. Current cell-based bone tissue regeneration strategies cannot cover large bone defects. K-carrageenan is a highly hydrophilic and biocompatible seaweed-derived sulfated polysaccharide, that has been proposed as a promising candidate for tissue engineering applications. Whether κ-carrageenan can be used to enhance bone regeneration is still unclear. In this study, we aimed to investigate whether κ-carrageenan has osteogenic potential by testing its effect on pre-osteoblast proliferation and osteogenic differentiation in vitro. Treatment with κ-carrageenan (0.5 and 2 mg/mL) increased both MC3T3-E1 pre-osteoblast adhesion and spreading at 1 h. K-carrageenan (0.125–2 mg/mL) dose-dependently increased pre-osteoblast proliferation and metabolic activity, with a maximum effect at 2 mg/mL at day three. K-carrageenan (0.5 and 2 mg/mL) increased osteogenic differentiation, as shown by enhanced alkaline phosphatase activity (1.8-fold increase at 2 mg/mL) at day four, and matrix mineralization (6.2-fold increase at 2 mg/mL) at day 21. K-carrageenan enhanced osteogenic gene expression (Opn, Dmp1, and Mepe) at day 14 and 21. In conclusion, κ-carrageenan promoted MC3T3-E1 pre-osteoblast adhesion and spreading, metabolic activity, proliferation, and osteogenic differentiation, suggesting that κ-carrageenan is a potential osteogenic inductive factor for clinical application to enhance bone regeneration. Surface charges of catalysts have important influences on the thermodynamics and kinetics of electrochemical reactions. Herein, we develop a modified version of the grand-canonical potential kinetics (GCP-K) method based on density functional theory (DFT) calculations to explore the effect of surface charges on reaction thermodynamics and kinetics. Using the hydrogen evolution reaction (HER) on the Pt(111) surface as an example, we show how to track the change of surface charge in a reaction and how to analyze its influence on the kinetics. Grand-canonical calculations demonstrate that the optimum hydrogen adsorption energy on Pt under the standard hydrogen electrode condition (SHE) is around −0.2 eV, rather than 0 eV established under the canonical ensemble, due to the high density of surface negative charges. By separating the surface charges that can freely exchange with the external electron reservoir, we obtain a Tafel barrier that is in good agreement with the experimental result. During the Tafel reaction, the net electron inflow into the catalyst leads to a stabilization of canonical energy and a destabilization of the charge-dependent grand-canonical component. Brain hypoxia is associated with a wide range of physiological and clinical conditions. Although oxygen is an essential constituent of maintaining brain functions, our understanding of how specific brain cell types globally respond and adapt to decreasing oxygen conditions is incomplete. In this study, we exposed mouse primary neurons, astrocytes, and microglia to normoxia and two hypoxic conditions and obtained genome-wide transcriptional profiles of the treated cells. Analysis of differentially expressed genes under conditions of reduced oxygen revealed a canonical hypoxic response shared among different brain cell types. In addition, we observed a higher sensitivity of neurons to oxygen decline, and dissected cell type-specific biological processes affected by hypoxia. Three-dimensional (3D) hepatocyte models have become a research hotspot for evaluating drug metabolism and hepatotoxicity. Compared to two-dimensional (2D) cultures, 3D cultures are better at mimicking the morphology and microenvironment of hepatocytes in vivo. However, commonly used 3D culture techniques are not suitable for high-throughput drug screening (HTS) due to their high cost, complex handling, and inability to simulate cell–extracellular matrix (ECM) interactions. This article describes a method for rapid and reproducible 3D cell cultures with ECM–cell interactions based on 3D culture instrumentation to provide more efficient HTS. We developed a microsphere preparation based on a high-voltage electrostatic (HVE) field and used sodium alginate- and collagen-based hydrogels as scaffolds for 3D cultures of HepG2 cells. The microsphere-generating device enables the rapid and reproducible preparation of bioactive hydrogel microspheres. This 3D culture system exhibited better cell viability, heterogeneity, and drug-metabolizing activity than 2D and other 3D culture models, and the long-term culture characteristics of this system make it suitable for predicting long-term liver toxicity. This system improves the overall applicability of HepG2 spheroids in safety assessment studies, and this simple and controllable high-throughput-compatible method shows potential for use in drug toxicity screening assays and mechanistic studies. Studies on genetic diversity analysis based on different types and numbers of molecular markers have yielded inconsistent results. In this study, we obtained 2,610,157 high-quality SNPs and 494,698 InDels through whole-genome resequencing of two farmed populations and one wild population. Both farmed populations showed consistent levels of genomic polymorphism and a slight increase in linkage compared with wild populations. The population structure of the two farmed populations was distinct from that of the wild population, but the degree of genetic differentiation was low (overall average Fst = 0.015). Selective sweep analysis showed that 523,529 genes were selected in the two farmed populations, and KEGG enrichment analysis showed that the selected genes were related to amino acid metabolism, which might be caused by artificial feeding. Seasonal freezing depth change is important in many environmental science and engineering applications. However, such changes are rare at region scales, especially over China, in the long time series. In this study, we evaluated the annual changes in seasonal maximum freezing depth (MFD) over China from 1971 to 2020 using an ensemble-modeling method based on support vector machine regression (SVMR) with 600 repetitions. Remote sensing data and climate data were input variables used as predictors. The models were trained using a large amount of annual measurement data from 600 meteorological stations. The main reason for using SVMR here was because it has been shown to perform better than random forests (RF), k-nearest neighbors (KNN), and generalized linear regression (GLR) in these cases. The prediction results were generally consistent with the observed MFD values. Cross validation showed that the model performed well on training data and had a better spatial generalization ability. The results show that the freezing depth of seasonally frozen ground in China decreased year by year. The average MFD was reduced by 3.64 cm, 7.59 cm, 5.54 cm, and 5.58 cm, in the 1980s, 1990s, 2000s, and 2010s, respectively, compared with the decade before. In the last 50 years, the area occupied by the freezing depth ranges of 0–40 cm, 40–60 cm, 60–80 cm, 80–100 cm, and 120–140 cm increased by 99,300 square kilometers, 146,200 square kilometers, 130,300 square kilometers, 115,600 square kilometers, and 83,800 square kilometers, respectively. In addition to the slight decrease in freezing depth range of 100–120 cm, the reduced area was 29,500 square kilometers. Freezing depth ranges greater than 140 cm showed a decreasing trend. The freezing depth range of 140–160 cm, which was the lowest range, decreased by 89,700 square kilometers. The 160–180 cm range decreased by 120,500 square kilometers, and the 180–200 cm range decreased by 161,500 square kilometers. The freezing depth range greater than 200 cm, which was the highest reduction range, decreased by 174,000 square kilometers. Considering the lack of data on the change in MFD of seasonally frozen ground in China in recent decades, machine learning provides an effective method for studying meteorological data and reanalysis data in order to predict the changes in MFD. Grassland gross primary productivity (GPP) is an important part of global terrestrial carbon flux, and its accurate simulation and future prediction play an important role in understanding the ecosystem carbon cycle. Machine learning has potential in large-scale GPP prediction, but its application accuracy and impact factors still need further research. This paper takes the Mongolian Plateau as the research area. Six machine learning methods (multilayer perception, random forest, Adaboost, gradient boosting decision tree, XGBoost, LightGBM) were trained using remote sensing data (MODIS GPP) and 14 impact factor data and carried out the prediction of grassland GPP. Then, using flux observation data (positions of flux stations) and remote sensing data (positions of non-flux stations) as reference data, detailed accuracy evaluation and comprehensive trade-offs are carried out on the results, and key factors affecting prediction performance are further explored. The results show that: (1) The prediction results of the six methods are highly consistent with the change tendency of the reference data, demonstrating the applicability of machine learning in GPP prediction. (2) LightGBM has the best overall performance, with small absolute error (mean absolute error less than 1.3), low degree of deviation (root mean square error less than 3.2), strong model reliability (relative percentage difference more than 5.9), and a high degree of fit with reference data (regression determination coefficient more than 0.97), and the prediction results are closest to the reference data (mean bias is only −0.034). (3) Enhanced vegetation index, normalized difference vegetation index, precipitation, land use/land cover, maximum air temperature, potential evapotranspiration, and evapotranspiration are significantly higher than other factors as determining factors, and the total contribution ratio to the prediction accuracy exceeds 95%. They are the main factors influencing GPP prediction. Citric acid is widely used in the food, chemical and pharmaceutical industries. Aspergillus niger is the workhorse used for citric acid production in industry. A canonical citrate biosynthesis that occurred in mitochondria was well established; however, some research suggested that the cytosolic citrate biosynthesis pathway may play a role in this chemical production. Here, the roles of cytosolic phosphoketolase (PK), acetate kinase (ACK) and acetyl-CoA synthetase (ACS) in citrate biosynthesis were investigated by gene deletion and complementation in A. niger. The results indicated that PK, ACK and ACS were important for cytosolic acetyl-CoA accumulation and had significant effects on citric acid biosynthesis. Subsequently, the functions of variant PKs and phosphotransacetylase (PTA) were evaluated, and their efficiencies were determined. Finally, an efficient PK-PTA pathway was reconstructed in A. niger S469 with Ca-PK from Clostridium acetobutylicum and Ts-PTA from Thermoanaerobacterium saccharolyticum. The resultant strain showed an increase of 96.4% and 88% in the citrate titer and yield, respectively, compared with the parent strain in the bioreactor fermentation. These findings indicate that the cytosolic citrate biosynthesis pathway is important for citric acid biosynthesis, and increasing the cytosolic acetyl-CoA level can significantly enhance citric acid production. Osteoarthritis (OA) is an inflammation-driven degenerative joint disease. Human salivary peptide histatin-1 (Hst1) shows pro-healing and immunomodulatory properties. but its role in OA treatment is not fully understood. In this study, we investigated the efficacy of Hst1 in the inflammation modulation-mediated attenuation of bone and cartilage damage in OA. Hst1 was intra-articularly injected into a rat knee joint in a monosodium iodoacetate (MIA)-induced OA model. Micro-CT, histological, and immunohistochemical analyses showed that Hst1 significantly attenuates cartilage and bone deconstruction as well as macrophage infiltration. In the lipopolysaccharide-induced air pouch model, Hst1 significantly reduced inflammatory cell infiltration and inflammation. Enzyme-linked immunosorbent assay (ELISA), RT-qPCR, Western blot, immunofluorescence staining, flow cytometry (FCM), metabolic energy analysis, and high-throughput gene sequencing showed that Hst1 significantly triggers M1-to-M2 macrophage phenotype switching, during which it significantly downregulated nuclear factor kappa-B (NF-κB) and mitogen-activated protein kinases (MAPK) signaling pathways. Furthermore, cell migration assay, Alcian blue, Safranin O staining, RT-qPCR, Western blot, and FCM showed that Hst1 not only attenuates M1-macrophage-CM-induced apoptosis and matrix metalloproteinase expression in chondrogenic cells, but it also restores their metabolic activity, migration, and chondrogenic differentiation. Heart failure is a serious and life-threatening disease worldwide. Cadherin-11 (Cad-11) is highly expressed in the heart and closely associated with inflammation. There is currently limited understanding on how Cad-11 contributes to cardiac remodeling and its underline molecular mechanism. We found an increased expression of Cad-11 in biopsy heart samples from heart failure patients, suggesting a link between Cad-11 and heart failure. To determine the role of Cad-11 in cardiac remodeling, Cad-11-deficient mice were used in a well-established mouse transverse aortic constriction (TAC) model. Loss of Cad11 greatly improved pressure overload-induced LV structural and electrical remodeling. IL (interleukin)-6 production was increased following TAC in WT mice and this increase was inhibited in cadherin-11−/− mice. We further tested the effect of IL-6 on myocyte hypertrophy and fibrosis in a primary culture system. The addition of hCad-11-Fc to cultured cardiac fibroblasts increased IL-6 production and fibroblast cell activation, whereas neutralizing IL-6 with an IL-6 antibody resulted in alleviating the fibroblast activation induced by hCad-11-Fc. On the other hand, cardiomyocytes were promoted to cardiomyocyte hypertrophy when cultured in condition media collected from cardiac fibroblasts stimulated by hCad-11-Fc.Similarly, neutralizing IL-6 prevented cardiomyocyte hypertrophy. Finally, we found that MAPKs and CaMKII–STAT3 pathways were activated in both hCad-11-Fc stimulated fibroblasts and cardiomyocytes treated with hCad-11-Fc stimulated fibroblast condition medium. IL-6 neutralization inhibited such MAPK and CaMKII-STAT3 signaling activation. These data demonstrate that Cad-11 functions in pressure overload-induced ventricular remodeling through inducing IL-6 secretion from cardiac fibroblasts to modulate the pathophysiology of neighboring cardiomyocytes. Because of easy functionalization, low cost, and large-scale fabrication, pure organic fluorescent polymers are widely applied in light-emitting display, bio-fluorescence-enhanced imaging, explosive detection, and other fields. Among these applications, due to their unique optical rotation characteristics, chiral fluorescent polymer materials are part of fluorescent polymers which could be used in chiral molecular detection and separation, biological target detection, etc. In this work, we designed and synthesized the first chiral organic fluorescent polysulfate materials through sulfur fluoride exchange polymerization (new click chemistry) by asymmetric binaphthol molecular. The chiral fluorescent polysulfate were synthesized by R/S [1,1′-binaphthalene]-2,2′-diol(Binol.), propane-2,2-diylbis(4,1-phenylene) bis(sulfurofluoridate) (FO2S–BA–SO2F) and 4,4′-(propane-2,2-diyl)diphenol(BA.) through step-by-step polymerization reaction under alkali present. It was found that the local crystallization of pure bisphenol A polysulfate was broken by the asymmetric axial chiral BINOL molecule inserted in it and let the polymer into the amorphous state. Fluorescent chiral molecules are uniformly dispersed in the polymer; the 120 µm film prepared by the film scraper was transparent and had good luminescence characteristics under ultraviolet light. Osteosarcoma (OS) is the most common malignant bone tumor. However, treatment strategies have not changed over the past 30 years. The relationship between OS and the immune microenvironment may provide a basis for the establishment of novel therapeutic targets. In this study, a large-scale gene expression dataset (GSE42352) was used to identify key genes in OS. A Target-OS dataset from the Cancer Genome Atlas was used as a validation set. Ecotropic viral integration site 2B (EVI2B) was significantly upregulated in OS tumor samples. Differentially expressed genes (DEGs) were identified between samples with high and low EVI2B expression in both the test and validation cohorts. The top three functions of DEGs determined by a gene set enrichment analysis (GSEA) were chemokine signaling, cytokine–cytokine receptor interaction, and Human T-cell leukemia virus 1 infection. A prognostic prediction model including EVI2B, DOCK2, and CD33 was constructed by a Cox regression analysis. This model indicated that EVI2B is an independent protective prognostic marker in OS. An analysis of immune infiltration further showed that high EVI2B expression levels were correlated with high levels of macrophage infiltration. Protein expression data derived from the Human Protein Atlas suggested EVI2B to be highly expressed in monocytes. Finally, we validated the elevated expression of EVI2B in OS cell lines and OS tissue samples; these results were consistent with those of the analyses of the GSE42352 and Target-OS datasets. Our integrative bioinformatics analysis and experimental results provide clear evidence for the prognostic value of EVI2B in OS and its close relationship with monocyte and macrophage infiltration. Fish cell lines are an important research tool with the advantages of low assay conditions, accurate control of assay results, and ease of operation, and thus have been widely used in virology research. We established a new cell line Japanese flounder spleen (JFSP) from the spleen tissue of the Japanese flounder (P. olivaceus). We explored the optimal growth conditions, chromosome morphology, and number and transfection experiment of JFSP cells in vitro. The susceptibility of JFSP cells to different viruses, including Bohle virus (BIV), Viral hemorrhagic septicemia virus (VHSV), Hirame rhabdovirus (HIRRV), Infectious hematopoietic necrosis virus (IHNV), and Lymphocystis disease virus (LCDV) and the expression changes of immune-related genes after virus infection were detected. The established JFSP cell line has enriched the fish cell resource bank and can be used as an excellent tool for genetic manipulation, studying host-virus interactions, and the development of potential vaccines. A new cell line Japanese flounder spleen (JFSP) derived from the spleen of Japanese flounder (Paralichthys olivaceus) was established and characterized in this study. The JFSP cells grew rapidly at 29 °C, and the optimum fetal bovine serum concentration in the L-15 medium was 15%. Cells were subcultured for more than 80 passages. The JFSP cells have a diploid chromosome number of 2n = 68, which differs from the chromosome number of normal diploid Japanese flounder. The established cells were susceptible to Bohle virus (BIV), Viral hemorrhagic septicemia virus (VHSV), Hirame rhabdovirus (HIRRV), Infectious hematopoietic necrosis virus (IHNV), and Lymphocystis disease virus (LCDV), as evidenced by varying degrees of cytopathic effects (CPE). Replication of the virus in JFSP cells was confirmed by qRT-PCR and transmission electron microscopy. In addition, the expression of four immune-related genes, TRAF3, IL-1β, TNF-α, and TLR2, was differentially altered following viral infection. The results indicated that the cells underwent an antiviral immune response. JFSP cell line is an ideal tool in vitro for virology. Sixteen dihydroartemisinin-5-methylisatin hybrids 6a–c and 7a–m tethered via different carbon spacers were assessed for their antiproliferative activity against MCF-7, MDA-MB-231, MCF-7/ADR and MDA-MB-231/ADR breast cancer cell lines as well as cytotoxicity towards MCF-10A cells to investigate the influence of the length of carbon spacers on the activity. The preliminary results illustrated that the length of the carbon spacer was the main parameter which affected the activity, and hybrids tethered via the two-carbon linker showed the highest activity. Amongst the synthesized hybrids, the representative hybrid 7a (IC50: 15.3–20.1 µM) not only demonstrated profound activity against both drug-sensitive and drug-resistant breast cancer cell lines, but also possessed excellent safety and selectivity profile. The disease has an effect on honeybee productivity and reproduction. Antibiotic fumagillin is still used in some countries for the treatment of Nosema sp. infection. However, using fumagillin has adverse effects on human health, as well as on honey bee physiology. Therefore, there are trends to develop non-antibiotic alternatives with already existing therapeutics. The present work attempts to emphasize the natural compounds now available for treating nosemosis. The honey bee is an important economic insect due to its role in pollinating many agricultural plants. Unfortunately, bees are susceptible to many pathogens, including pests, parasites, bacteria, and viruses, most of which exert a destructive impact on thousands of colonies. The occurrence of resistance to the therapeutic substances used against these organisms is rising, and the residue from these chemicals may accumulate in honey bee products, subsequently affecting the human health. There is current advice to avoid the use of antibiotics, antifungals, antivirals, and other drugs in bees, and therefore, it is necessary to develop alternative strategies for the treatment of bee diseases. In this context, the impact of nosema diseases (nosemosis) on bee health and the negative insults of existing drugs are discussed. Photocatalysis represents a promising technology that might alleviate the current environmental crisis. One of the most representative photocatalysts is graphitic carbon nitride (g-C3N4) due to its stability, cost-effectiveness, facile synthesis procedure, and absorption properties in visible light. Nevertheless, pristine g-C3N4 still exhibits low photoactivity due to the rapid recombination of photo-induced electron-hole (e−-h+) pairs. To solve this drawback, Z-scheme photocatalysts based on g-C3N4 are superior alternatives since these systems present the same band configuration but follow a different charge carrier recombination mechanism. To contextualize the topic, the main drawbacks of using g-C3N4 as a photocatalyst in environmental applications are mentioned in this review. Then, the basic concepts of the Z-scheme and the synthesis and characterization of the Z-scheme based on g-C3N4 are addressed to obtain novel systems with suitable photocatalytic activity in environmental applications (pollutant abatement, H2 production, and CO2 reduction). Focusing on the applications of the Z-scheme based on g-C3N4, the most representative examples of these systems are referred to, analyzed, and commented on in the main text. A novel nidovirus, CSBV Bces-Po19, was isolated from the marine fish, Japanese flounder (Paralichthys olivaceus). The viral genome was 26,597 nucleotides long and shared 98.62% nucleotide identity with CSBV WHQSR4345. PacBio Sequel and Illumina sequencing were used to perform full-length transcriptome sequencing on CSBV Bces-Po19-sensitive (S) and -resistant (R) Japanese flounder. The results of negative staining revealed bacilliform and spherical virions. There were in total 1444 different genes between CSBV Bces-Po19 S and R groups, with 935 being up-regulated and 513 being down-regulated. Metabolism-, immune-, and RNA-related pathways were significantly enriched. Furthermore, CSBV Bces-Po19 infection induced alternative splicing (AS) events in Japanese flounder; the S group had a higher numbers of AS events (12,352) than the R group (11,452). The number of long non-coding RNA (lncRNA) in the S group, on the other hand, was significantly lower than in the R group. In addition to providing valuable information that sheds more light on CSBV Bces-Po19 infection, these research findings provide further clues for CSBV Bces-Po19 prevention and treatment.
