Key message: Major QTL for grain number per spike were identified on chromosomes 2B and 2D. Haplotypes and candidate genes of QGns.cib-2B.1were analyzed. Grain number per spike (GNS) is one of the main components of wheat yield. Genetic dissection of their regulatory factors is essential to improve the yield potential. In present study, a recombinant inbred line population comprising 180 lines developed from the cross between a high GNS line W7268 and a cultivar Chuanyu12 was employed to identify quantitative trait loci (QTL) associated with GNS across six environments. Two major QTL, QGns.cib-2B.1 and QGns.cib-2D.1, were detected in at least four environments with the phenotypic variations of 12.99–27.07% and 8.50–13.79%, respectively. And significant interactions were observed between the two major QTL. In addition, QGns.cib-2B.1 is a QTL cluster for GNS, grain number per spikelet and fertile tiller number, and they were validated in different genetic backgrounds using Kompetitive Allele Specific PCR (KASP) markers. QGns.cib-2B.1 showed pleotropic effects on other yield-related traits including plant height, spike length, and spikelet number per spike, but did not significantly affect thousand grain weight which suggested that it might be potentially applicable in breeding program. Comparison analysis suggested that QGns.cib-2B.1 might be a novel QTL. Furthermore, haplotype analysis of QGns.cib-2B.1 indicated that it is a hot spot of artificial selection during wheat improvement. Based on the expression patterns, gene annotation, orthologs analysis and sequence variations, the candidate genes of QGns.cib-2B.1 were predicted. Gamma delta (γδ) T cells, a unique T cell subgroup, are crucial in various immune responses and immunopathology1–3. The γδ T cell receptor (TCR), which is generated by γδ T cells, recognizes a diverse range of antigens independently of the major histocompatibility complex2. The γδ TCR associates with CD3 subunits, initiating T cell activation and holding great potential in immunotherapy4. Here we report the structures of two prototypical human Vγ9Vδ2 and Vγ5Vδ1 TCR–CD3 complexes5,6, revealing two distinct assembly mechanisms that depend on Vγ usage. The Vγ9Vδ2 TCR–CD3 complex is monomeric, with considerable conformational flexibility in the TCRγ–TCRδ extracellular domain and connecting peptides. The length of the connecting peptides regulates the ligand association and T cell activation. A cholesterol-like molecule wedges into the transmembrane region, exerting an inhibitory role in TCR signalling. The Vγ5Vδ1 TCR–CD3 complex displays a dimeric architecture, whereby two protomers nestle back to back through the Vγ5 domains of the TCR extracellular domains. Our biochemical and biophysical assays further corroborate the dimeric structure. Importantly, the dimeric form of the Vγ5Vδ1 TCR is essential for T cell activation. Due to global warming and the disturbance of the interannual variability of precipitation, the frequency of extreme drought events has increased. The impact of global climate change on water resources is becoming increasingly apparent, then it is particularly necessary to explore the carrying capacity of water ecological environment under extreme drought conditions, which can guarantee the ecological water security in river basins. This study takes the Guanzhong area of the Wei River Basin as an example, calculating the water environment carrying capacity of 40 areas in the Weihe Guanzhong area in different levels of years under extreme drought conditions by comprehensive evaluation model of carrying capacity and using geographic information system GIS to display the spatial distribution of water environment carrying capacity in 40 regions. According to the results of the spatial distribution of water environmental bearing capacity, four different schemes are designed to improve the bearing capacity. The first plan reduces the industrial water consumption and irrigation quota by 5%, the second plan increases the industrial water and sewage treatment rate on this basis. the third plan further improves the development and utilization rate of surface and groundwater, and the fourth plan, on the basis of the first three plans, supplies 600 million cubic meters of industrial and agricultural water to Guanzhong region. Through comparative analysis, without taking any measures, under the extreme drought conditions, the water environment carrying capacity of the 40 areas in Guanzhong is all in an unbearable state. Overall, plan 4 has the most significant improvement in the water environment-carrying capacity, especially the Dong zhuang Reservoir of the Jing River which has played a very important role in enhancing the water ecological environment carrying capacity of the downstream water of the Wei River. The Jishishan Ms 6.2 earthquake occurred at 23:59 on December 18, 2023 in Gansu Province, China. We conducted a field survey to assess the hazards and damages caused by the earthquake and its associated geo-activities. Subsequently, we organized a seminar to discuss the possible causes of the destruction of a prehistoric site—Lajia Settlement—dated back to four thousand years B.P. and located only several kilometers away from the epicenter of the Jishishan earthquake. The Jishishan earthquake was unique for its hazard and disaster process, which featured ground shaking and a series of complex geological and geomorphological activities: sediment and soil spray piles, liquefaction, collapse, landslide, and mudflow along water channels. We define this phenomenon as the Jishishan earthquake ripple hazard (JERH). The most recent evidence from the JERH suggests that a prehistoric earthquake similar to the JERH, instead of riverine floods or earthquake-induced landslide dam outburst flood, as previously hypothesized, destroyed the Lajia Settlement. Medullary Carcinoma of the Colon (MCC) is a rare histological subtype of colon cancer, and there is currently no recognized optimal treatment plan for it, with its prognosis remaining unclear. The aim of this study is to analyze the independent prognostic factors for MCC patients and develop and validate nomograms to predict overall survival (OS). A total of 760 patients newly diagnosed with MCC from 2004 to 2020 were selected from the Surveillance, Epidemiology, and End Results (SEER) database. All patients were randomly allocated to a training group and a validation group in a 7:3 ratio. Univariate and multivariable Cox regression analyses were conducted to identify prognostic factors and construct nomograms. The nomogram prediction model was evaluated and validated using receiver operating characteristic (ROC) curves, calibration curves, and decision curve analysis (DCA). The study found that elderly women are more susceptible to MCC, and the ascending colon and cecum are the most common sites of involvement. MCC is poorly differentiated, with stages II and III being the most common. Surgery is the primary treatment for MCC. The prognosis for patients with stage IV MCC is poor, with a median survival time of only 10 months. Independent prognostic factors for MCC include age, N stage, M stage, surgery, chemotherapy, and tumor size. Among them, age < 75 years and completion of chemotherapy were protective factors for colon medullary carcinoma, while N2 (HR = 2.18, 95%CI 1.40–3.38), M1 (HR = 3.31, 95%CI 2.01–5.46), no surgery (HR = 27.94, 95%CI 3.69–211.75), and tumor diameter > 7 cm (HR = 1.66, 95%CI 1.20–2.30) were risk factors for colon medullary carcinoma. The results of ROC, AUC, calibration curves, and DCA demonstrate that the nomogram prediction model exhibits good predictive performance. We have updated the demographic characteristics of colon medullary carcinoma and identified age, N staging, M staging, surgery, chemotherapy and tumor size as independent prognostic factors for colon medullary carcinoma. The LAT1-4F2hc complex (SLC7A5-SLC3A2) facilitates uptake of essential amino acids, hormones and drugs. Its dysfunction is associated with many cancers and immune/neurological disorders. Here, we apply native mass spectrometry (MS)-based approaches to provide evidence of super-dimer formation (LAT1-4F2hc)2. When combined with lipidomics, and site-directed mutagenesis, we discover four endogenous phosphatidylethanolamine (PE) molecules at the interface and C-terminus of both LAT1 subunits. We find that interfacial PE binding is regulated by 4F2hc-R183 and is critical for regulation of palmitoylation on neighbouring LAT1-C187. Combining native MS with mass photometry (MP), we reveal that super-dimerization is sensitive to pH, and modulated by complex N-glycans on the 4F2hc subunit. We further validate the dynamic assemblies of LAT1-4F2hc on plasma membrane and in the lysosome. Together our results link PTM and lipid binding with regulation and localisation of the LAT1-4F2hc super-dimer. The amino acid transporter complex LAT1-4F2hc is considered a major drug target for many cancers. For the hard guessing random additive noise decoding Markov order (GRAND-MO) algorithm, it is crucial to develop an efficient noise error patterns (NEPs) generator to facilitate its application in bursty channels. This paper proposes a practical hardware realization by generating the NEPs in a sequential manner. Based on classification of the four types of NEPs, we propose to iteratively calculate the "1" and the "0" permutations in the same time. Then, the novel "0" permutation regularization and bit flipping techniques are employed, through which the generation of the four types of NEPs is uniformed at the same way. Moreover, the proposed NEPs generator can generate all NEPs by using the "1" burst parameters, and is suitable for the guessing decoding of any linear block codes. Built on field programmable gate array (FPGA) implementation and comparison with existing benchmark, we show the proposed NEPs generator is a power-efficient architecture for realization. Photocatalytic oxidation of methane to value-added chemicals is a promising process under mild conditions, nevertheless confronting great challenges in efficiently activating C–H bonds and inhibiting over-oxidation. Herein, we propose a comprehensive strategy for the selective generation of reactive oxygen species (ROS) by regulating the sizes and facets of Au nanoparticles loaded on ZnO. For photocatalytic methane oxidation at ambient temperature, a high oxygenates yield of 36.4 mmol·g−1·h−1 with a nearly 100% selectivity has been achieved over the optimized 1.0% Au/ZnO-9.6 (1% Au with (111) facet and 9.6 nm size on ZnO) photocatalyst, exceeding most reported literatures. Mechanism investigations reveal that 1.0% Au/ZnO-9.6 with the medium size and Au (111) facet guarantees the favourable formation of superoxide radicals (·OOH) through mild oxygen reduction, ultimately leading to excellent photocatalytic methane oxidation performance. Problematic mobile phone use (PMPU) has been revealed to be shaped by multiple factors. Nevertheless, limited studies have focused on sense of control, an intrinsic human motive, and investigated its impact on PMPU and the mechanisms of this influence. On the grounds of the Interaction of Person-Affect-Cognition-Execution model, the present study tested experiential avoidance as a possible mediator and self-concept clarity as a possible moderator in the connection linking sense of control and PMPU. Undergraduate students (N = 1,093; Mage = 18.95, SD = 1.10; 53.2% women) recruited through convenience sampling completed questionnaires in their classrooms. The mediation analysis revealed a significant mediating effect of experiential avoidance on the negative link between sense of control and PMPU. The moderated mediation analysis demonstrated a significant moderating effect of self-concept clarity, which attenuated both the direct path linking sense of control and PMPU and the mediating path through experiential avoidance. The findings contribute to elucidating whether, how, and under what conditions sense of control is connected with PMPU, and offer insights for developing practical intervention strategies to address PMPU. Biochar, as a potential adsorbent, has been widely employed to remove pollutants from sewage. In this study, a lignin-based biochar (CB-800) was prepared by a simple high-temperature pyrolysis using urban green waste (Cinnamomum camphora leaves) as a feedstock to remove norfloxacin (NOR) from water. Batch adsorption test results indicated that CB-800 had a strong removal capacity for NOR at a wide range of pH values. The maximum adsorption achieved in the study was 50.90 ± 0.64 mg/g at 298 K. The pseudo-first and second-order kinetic models and the Dubinin-Radushkevich isotherm fitted the experimental data well, indicating that NOR adsorption by CB-800 was a complex process involving both physi-sorption and chemi-sorption. The physical properties of CB-800 were characterized by SEM and BET. The mesoporous structures were formed hierarchically on the surface of CB-800 (with an average pore size of 2.760 nm), and the spatial structure of NOR molecules was more easily adsorbed by mesoporous structures. Combined with Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) analysis, it was showed that the main NOR adsorption mechanisms by CB-800 included ion exchange, π-electron coordination, hydrogen bonding, and electrostatic adsorption. Meanwhile, the reduction of C = O and pyridine nitrogen, and the presence of C-F2, also indicated the occurrence of substitution, addition, and redox. This study not only determined the reaction mechanism between biochar and NOR, but also provides guidance to waste managers for the removal of NOR from water by biochar. Molecular identification, such as DNA barcoding, is a useful tool that is widely applied in distinguishing species. To identify the cyprinid Acrossocheilus jishouensis, which was previously known to be restricted to only its type locality, we conducted molecular identification of this species based on 23 samples in five localities. Molecular identification based on the mitochondrial COI gene sequence showed that the morphologically similar samples from the five populations were all A. jishouensis, as the mean genetic distances between populations were very small (0.1–1.6%); thus, the distribution of this species was substantially expanded. The whole mitochondrial genome of one sample was also assembled, which was 16,594 bp in length and consisted of 13 protein-coding genes (PCGs), two rRNA genes, 22 tRNA genes, and one control region. All PCGs began with ATG except the COI gene, which started with GTG; seven PCGs used the complete stop codon TAA, while four terminated in T(AA) and two ended with TAG. The overall base composition reflected a higher proportion of A+T than G+C and a positive AT-skew and negative GC-skew pattern except for the opposite in ND6. Phylogenetic relationships inferred using BI and ML methods revealed that both Acrossocheilus and Onychostoma were nonmonophyletic, which indicated that the traditional diagnoses between these two genera need to be assessed further. The results of this study not only expanded the known distribution ranges of A. jishouensis, but also provided a valuable data resource for future molecular and evolutionary studies of Acrossocheilus and other cyprinids in Barbinae. Non-specific endonucleases can be used for the digestion of nucleic acids because they hydrolyze DNA/RNA into 3–5 base pairs (bp) length oligonucleotide fragments without strict selectivity. In this work, a novel non-specific endonuclease from Pseudomonas fluorescens (PfNuc) with high activities for both DNA and RNA was successfully cloned and expressed in Escherichia coli. The production of PfNuc in flask scale could be achieved to 1.73 × 106 U/L and 4.82 × 106 U/L for DNA and RNA by investigation of the culture and induction conditions. The characterization of PfNuc indicated that it was Mg2+-dependent and the catalytic activity was enhanced by 3.74 folds for DNA and 1.06 folds for RNA in the presence of 5 mM Mg2+. The specific activity of PfNuc for DNA was 1.44 × 105 U/mg at pH 8.0 and 40 °C, and 3.93 × 105 U/mg for RNA at pH 8.5 and 45 °C. The Km of the enzyme for both DNA and RNA was close to 43 µM. The Vmax was 6.40 × 105 U/mg and 1.11 × 106 U/mg for DNA and RNA, respectively. There was no observed activity loss when PfNuc was stored at 4 °C and − 20 °C after 28 days or 10 repeated freeze–thaw cycles at − 80 °C. Molecular docking revealed that PfNuc formed 17 and 19 hydrogen bonds with single-stranded RNA and double-stranded DNA, respectively. Background: The efficacy of immune checkpoint inhibitors (ICIs) in hepatocellular carcinoma (HCC) is poor and great heterogeneity among individuals. Chemokines are highly correlated with tumor immune response. Here, we aimed to identify an effective chemokine for predicting the efficacy of immunotherapy in HCC. Methods: Chemokine C‐C motif ligand 21 (CCL21) was screened by transcriptomic analysis in tumor tissues from HCC patients with different responses to ICIs. The least absolute shrinkage and selection operator (LASSO) regression analysis was conducted to construct a predictive nomogram. Neutrophils in vitro and HCC subcutaneous tumor model in vivo were applied to explore the role of CCL21 on the tumor microenvironment (TME) of HCC. Results: Transcriptome analysis showed that CCL21 level was much higher in HCC patients with response to immunotherapy. The predictive nomogram was constructed and validated as a classifier. CCL21 could inhibit N2 neutrophil polarization by suppressing the activation of nuclear factor kappa B (NF-κB) pathway. In addition, CCL21 enhanced the therapeutic efficacy of ICIs. Conclusion: CCL21 may serve as a predictive biomarker for immunotherapy response in HCC patients. High levels of CCL21 in TME inhibit immunosuppressive polarization of neutrophils. Hard carbon is considered to be a viable choice for anode materials in sodium-ion batteries due to its low cost and high specific capacity for sodium storage. The wasted grains derived from Chinese baijiu possess significant potential as biomass-based precursors for the production of hard carbon compounds. However, their use as biomass material has been mostly ignored. Therefore, this research focuses on the use of discarded grains from baijiu as a raw material for the production of porous hard carbon materials, specifically for the anode of sodium-ion batteries. The proposed synthesis procedure involves the use of a ZnCl2 chemical treatment to generate hard carbon materials that possess a porous structure and exhibit graphitic features. In comparison to the direct carbonization synthesis process, the use of ZnCl2 treatment on porous hard carbon results in notable enhancements in electrochemical performance, including specific capacity and rate capability, while simultaneously preserving exceptional long-term cycle stability. Especially, the enhanced capacity mostly manifests in the low-voltage plateau region, and the high plateau capacity can maintained even at higher specific currents, which is suitable for high-rate application, exceeding the typical commercial hard carbon materials used in sodium-ion batteries. Wire hot extrusion was investigated to produce thin wires of metals and metallic matrix nanocomposites, including aluminum (Al), copper (Cu), and copper/carbon nanotube composites (Cu/CNT, 1%wt). A new energy-based analytical solution was developed to predict extrusion force, where friction effect between die and sample was considered. This analytical solution achieved a much better result than the classical slip line theory and other existing analytical solutions, where friction effect was usually not considered. Finite element (FE) models using ALE method were built to validate the analytical solution under different extrusion conditions. A series of FE simulations were run for different die angles (30°, 45°, and 60°) and two different area extrusion ratios (4:1 and 16:1). The comparison results showed a very good correlation between the analytical solution and FE results. Four wire extrusion tests of metals and Cu/CNT composite were conducted under elevated temperatures (about 300 to 700 ℃). Test results further proved the excellent accuracy of the analytical solution and the necessity of considering friction effect. Context: Cities are still expanding in many parts of the world in the 21st century, especially in developing regions, where socioeconomic transition has triggered urbanization. The Qinghai-Tibetan Plateau, characterized by its high elevation, developing economy, and fragile environment, has undergone rapid urbanization in the past twenty years, potentially heightening the region’s risk of geological and hydrometeorological disasters. Objectives: This article examines the process of urbanization on the Qinghai-Tibetan Plateau in order to better understand the relationship between urban expansion and population change and to explore whether geological and hydrometeorological disasters threaten urban expansion. Methods: We created a high-resolution urban built-up boundary dataset from 2000 to 2020 based on remote sensing and GIS. By examining the spatio-temporal pattern of urban expansion and its population dynamics from 2000 to 2020 in 15 cities across the Qinghai-Tibetan Plateau, we quantified the urban expansion characteristics and explored the rising geological and hydrometeorological disaster risks urban areas face. Results: The Qinghai-Tibetan Plateau has experienced rapid and uneven urbanization in the past 20 years. The total expansion rate was over 252% – 1.48 and 2.81 times higher than that of China and the world, respectively. Urban expansion exceeded population growth in most cities throughout the whole period. The urban built-up areas and population of the Qinghai-Tibetan Plateau tended to concentrate in the Yellow River-Huangshui River Valley of Qinghai Province and the Yarlung Zangbo River and its two tributaries of Tibet. High-density urban built-up areas and population distribution were exposed to geological and hydrometeorological disasters. The urban boundary dataset produced in this study has better accuracy than the other five existing datasets, for the given area and time range. Conclusions: This study will not only provide a scientific reference for urban development research and future urban planning on the Qinghai-Tibetan Plateau at the regional scale, but will also contribute to research on the urbanization process of the plateau and mountainous regions on a global scale, raising awareness of geological and hydrometeorological disaster risk prevention in these areas worldwide [ABSTRACT FROM AUTHOR]

This meta-analysis compared the efficacy and safety of different antithrombotic regimens after left atrial appendage closure (LAAC). PubMed, Embase, Medline, Cochrane Library databases were systematically searched from their inception to March 2023. Patients were divided into short-term oral anticoagulation (OAC) group and antiplatelet therapy (APT) group. The incidence of events were performed using RevMan 5.4. The events including device-related thrombus (DRT), ischemic stroke/systemic embolization (SE), major bleeding, any bleeding, any major adverse event and all-cause mortality. Subgroup analysis were based on OAC alone or OAC plus single antiplatelet therapy (SAPT) in OAC group. Oral anticoagulants include warfarin and direct oral anticoagulant (DOAC). Fourteen studies with 35,166 patients were included. We found that the incidence of DRT (OR = 0.49, 95% CI 0.36–0.66, P＜0.0001) and all-cause mortality (OR = 0.71, 95% CI 0.57–0.89, P = 0.002) were significantly lower in OAC group than APT group. However, there was no statistical differences in the incidence rates of ischemic stroke/SE (OR = 0.77, 95% CI 0.49–1.20, P = 0.25), major bleeding (OR = 0.84, 95% CI 0.55–1.27, P = 0.84), any bleeding (OR = 0.83, 95% CI 0.56–1.22, P = 0.34) and any major adverse event (OR = 0.56, 95% CI 0.30–1.03, P = 0.06) in the two groups. Subgroup analysis found that the incidence of DRT, all-cause mortality and any major adverse event in OAC monotherapy were lower than that in APT group (P＜0.05), but not statistically different from other outcome. The incidence of DRT, all-cause mortality, any major adverse event and any bleeding in DOAC were significantly better than APT group (P＜0.05). While warfarin only has better incidence of DRT than APT (P＜0.05), there was no statistical difference between the two groups in other outcome (P＞0.05). The incidence of DRT was significantly lower than APT group (P＜0.05), major bleeding were higher, and the rest of the outcome did not show any statistically significant differences(P＞0.05) when OAC plus SAPT. Based on the existing data, short-term OAC may be favored over APT for patients who undergo LAAC. To advance the full potential of quantum networks one should be able to distribute quantum resources over long distances at appreciable rates. As a consequence, all components in such networks need to have large multimode capacity to manipulate photonic quantum states. Towards this end, a photonic quantum memory with a large multimode capacity, especially one operating at telecom wavelength, remains an important challenge. Here we optimize the preparation of atomic frequency combs and demonstrate a spectro-temporally multiplexed quantum memory in a 10-m-long cryogenically cooled erbium doped silica fibre. Our multiplexing storage has five spectral channels - each 10 GHz wide with 5 GHz separation - with up to 330 temporal modes in each, thus resulting in a simultaneous storage of 1,650 modes of heralded single photons with a 1000-fold increasing in coincidence detection rate with respect to single mode storage. Hydrochar has shown great potential for As adsorption owing to its inherent advantages of having richer functional groups while consuming less energy; however, the effect of iron modification on hydrochar in As adsorption remains unclear. In this paper, Fe-modified hydrochars (Fe@PWC and Fe@CSC) were synthesized by poplar sawdust (PW) and corn straw (CS) with Fe(NO3)3 through one-step hydrothermal carbonization (HTC) and characterized by BET, SEM, XRD, XPS, and FTIR. The various capacities of the two hydrochars for absorbing As in solution and immobilizing As in soil were examined. The findings showed that Fe@PWC exhibited superior performance by increasing the adsorption capacity for As(V) to 6.79 mg/g, which was approximately 6 times that of unmodified PWC, while also increasing the residual As fraction in soil by 55%. However, compared with Fe@PWC, the adsorption and immobilization of As(V) by Fe@CSC were limited. It may be due to the agglomeration phenomenon of Fe@CSC during the HTC process, which resulted in the limited availability of functional groups and thereby restricted the enhancement of its adsorptive properties. DFT calculations confirmed that As(V) primarily formed monodentate complexes with iron oxide on the surface of Fe@HC, and -OH functional groups played a significant role in As adsorption. Furthermore, redox reactions occurred during the adsorption process, and Fe-O groups were transformed into FeOOH. This study elucidated the changes in the surface structure of hydrochar after the Fe-modified HTC process and uncovered the microscopic mechanism by which As is adsorbed on the molecular level of iron-modified hydrochar. Monolayer transition metal dichalcogenides (TMDCs) have become a promising platform in valleytronics due to possessing the regulatable valley degrees of freedom. While, as a result of the rapid intervalley scattering, it is difficult to measure the PL valley polarization of monolayer TMDCs at room temperature, which limits their application in valleytronics devices. Here, we report a room temperature photoluminescence (PL) valley polarization up to 3.73% in an "optical bilayer" WS2 formed by transferring monolayer WS2 onto flat Ag film. Furthermore, in the transient absorption (TA) measurements, a remarkably long valley depolarization lifetime is found. Thus, we demonstrate the valley properties of such "optical bilayer" WS2 resemble actual bilayer WS2, in which the robust valley polarization can be attributed to the phonons depletion effect and the blocked interlayer hopping processes. Key message: A novel major adult-plant stripe rust resistance QTL derived from cultivated emmer wheat was mapped to a 123.6-kb region on wheat chromosome 2BL. Stripe rust, caused by the fungal pathogen Puccinia striiformis f. sp. tritici (Pst), is one of the most devastating diseases of wheat. Identification of new sources of resistance and their utilization in breeding programs is the effectively control strategy. The objective of this study was to identify and genetically characterize the stripe rust resistance derived from the cultivated emmer accession AS286. A recombinant inbred line population, developed from a cross between the susceptible durum wheat line langdon and AS286, was genotyped using the Wheat55K single nucleotide polymorphism array and evaluated in field conditions with a mixture of the prevalent Chinese Pst races (CYR32, CYR33, CYR34, Zhong4, and HY46) and in growth chamber with race CYR34. Three QTLs conferring resistance were mapped on chromosomes 1BS, 2BL, and 5BL, respectively. The QYrAS286-1BS and QYrAS286-2BL were stable with major effects, explaining 12.91% to 18.82% and 11.31% to 31.43% of phenotypic variation, respectively. QYrAS286-5BL was only detected based on growth chamber seedling data. RILs harboring both QYrAS286-1BS and QYrAS286-2BL showed high levels of stripe rust resistance equal to the parent AS286. The QYrAS286-2BL was only detected at the adult-plant stage, which is different from previously named Yr genes and inherited as a single gene. It was further mapped to a 123.6-kb region using KASP markers derived from SNPs identified by bulked segregant RNA sequencing (BSR-Seq). The identified loci enrich our stripe rust resistance gene pool, and the flanking markers developed here could be useful in marker-assisted selection for incorporating QYrAS286-2BL into wheat cultivars. Background: Relapse of hepatocellular carcinoma (HCC) due to vascular invasion is common, but the genomic mechanisms remain unclear, and molecular determinants of high-risk relapse cases are lacking. We aimed to reveal the evolutionary trajectory of microvascular invasion (MVI) and develop a predictive signature for relapse in HCC. Methods: Whole-exome sequencing was performed on tumor and peritumor tissues, portal vein tumor thrombus (PVTT), and circulating tumor DNA (ctDNA) to compare the genomic profiles between 5 HCC patients with MVI and 5 patients without MVI. We conducted an integrated analysis of exome and transcriptome to develop and validate a prognostic signature in two public cohorts and one cohort from Zhongshan Hospital, Fudan University. Results: Shared genomic landscapes and identical clonal origins among tumor, PVTT, and ctDNA were observed in MVI (+) HCC, suggesting that genomic changes favoring metastasis occur at the primary tumor stage and are inherited in metastatic lesions and ctDNA. There was no clonal relatedness between the primary tumor and ctDNA in MVI (–) HCC. HCC had dynamic mutation alterations during MVI and exhibited genetic heterogeneity between primary and metastatic tumors, which can be comprehensively reflected by ctDNA. A relapse-related gene signature named RGSHCC was developed based on the significantly mutated genes associated with MVI and shown to be a robust classifier of HCC relapse. Conclusions: We characterized the genomic alterations during HCC vascular invasion and revealed a previously undescribed evolution pattern of ctDNA in HCC. Objective: Current studies on the association between fecal incontinence (FI) and depression are very limited, and most of them are restricted to women or elderly patients. This study aims to evaluate in detail the association between FI and depression among US adults. Methods: 13,480 adults aged 20 years and older were selected from the National Health and Nutrition Examination Survey 2005–2010. Monthly loss of solid, liquid, or mucous stool was defined as FI. Clinical depression and depression severity were assessed by the validated Patient Health Questionnaide-9 (PHQ-9). Models of multivariate logistic regression were used to calculate adjusted odds ratios (ORs). A subgroup analysis was carried out to ensure that the results were stable. Results: After adjusting for covariates such as demographics, risk behaviors and associated comorbidities, the PHQ-9 score and clinical depression were both significantly associated with FI, with ORs and 95%CIs of 1.11 (1.10–1.13) and 3.01 (2.53–3.57). Depression of all severities was also significantly associated with FI. The ORs and 95%CIs of FI with mild depression, moderate depression, and moderately severe to severe depression were 2.29 (1.96–2.68), 3.44 (2.77–4.27) and 4.65 (3.61–6.00), respectively. Subgroup analyses showed no statistically significant interactions (P > 0.05). Conclusions: In conclusion, FI significantly associated with depression of any severity. Like the elderly or female patients with FI, young and middle-aged or men with FI are also at high risk of depression and should also be the focus of depression screening and early intervention. Purpose: To analyze the long-term overall survival (OS) and influencing factors of patients with esophageal squamous cell cancer (ESCC) under surgical treatment. Method: We collected patients with ESCC who received surgical treatment in Sichuan Cancer Hospital & institute from January 2010 to December 2017, and selected 2,766 patients with thoracic esophageal carcinoma with relatively complete follow-up results as the objects of this study. We analyzed the characteristics, postoperative complications and long-term OS results of those patients. Results: Of the 2766 patients, 81.6% were male, midthoracic esophageal cancer accounted for 53.5%. McKeown was used in 72.0% of patients and Ivor-Lewis was used in 26.4% of patients. About 47.8% of patients received minimally invasive esophagectomy (MIE). The overall complication rate was 25.8%. The 1-year, 3-year and 5-year OS rates were 86.2%, 57.5% and 46.8%, respectively. McKeown had a better long-term OS rate than Ivor-Lewis (49.5% vs 41.2%, P < 0.001), and MIE is superior to open surgery (51.8% vs 42.5%, P < 0.001). Conclusion: McKeown has advantages over Ivor-Lewis. MIE results in better long-term survival outcomes for patients. But more prospective randomized controlled trials with large samples are needed. Producing hydrogen peroxide (H2O2) through an electrochemical oxygen reduction reaction (ORR) is a safe, green strategy and a promising alternative to traditional energy-intensive anthraquinone processes. Air and renewable power could be utilized for on-site and decentralized H2O2 production, demonstrating significant application potential. Currently, single atom catalysts (SACs) have demonstrated significant advantages in the catalytic production of H2O2 in 2e− ORR. However, the selectivity of SACs in ORR once puzzled researchers. This article reviews the research on the development and achievements of H2O2 production by SACs catalysis in recent years. Especially, the structure–performance relationship is a guide to designing new SACs. Combining advanced characterization techniques and theoretical calculation methods, researchers have a clearer and more thorough understanding of the impact of the atomic interface of SACs on ORR catalytic performance. The coordination moiety formed between the active metal center atom and the support seriously determines the selectivity of SACs, mainly manifested in the adsorption of *OOH intermediates. Particularly, the atomic interface of metal atoms together with O/N co-coordination exhibit high selectivity and mass activity, and heteroatoms or functional groups on carbon supports present synergistic effects to promote the production of H2O2 in 2e− ORR. Fine and accurate regulation of the atomic interface of SACs directly affects the 2e− ORR performance of the catalysts. Bear bile powder (BBP) is a valuable animal-derived product with a huge adulteration problem on market. It is a crucially important task to identify BBP and its counterfeit. Electronic sensory technologies are the inheritance and development of traditional empirical identification. Considering that each drug has its own specific odor and taste characteristics, electronic tongue (E-tongue), electronic nose (E-nose) and GC-MS were used to evaluate the aroma and taste of BBP and its common counterfeit. Two active components of BBP, namely tauroursodeoxycholic acid (TUDCA) and taurochenodeoxycholic acid (TCDCA) were measured and linked with the electronic sensory data. The results showed that bitterness was the main flavor of TUDCA in BBP, saltiness and umami were the main flavor of TCDCA. The volatiles detected by E-nose and GC-MS were mainly aldehydes, ketones, alcohols, hydrocarbons, carboxylic acids, heterocyclic, lipids, and amines, mainly earthy, musty, coffee, bitter almond, burnt, pungent odor descriptions. Four different machine learning algorithms (backpropagation neural network, support vector machine, K-nearest neighbor, and random forest) were used to identify BBP and its counterfeit, and the regression performance of these four algorithms was also evaluated. For qualitative identification, the algorithm of random forest has shown the best performance, with 100% accuracy, precision, recall and F1-score. The safety and immunogenicity of a protein-based tetravalent vaccine SCTV01E that contains spike protein ectodomain (S-ECD) of Alpha, Beta, Delta and Omicron BA.1 are assessed and compared with bivalent protein vaccine SCTV01C (Alpha and Beta variants) and monovalent mRNA vaccine (NCT05323461). The primary endpoints are the geometric mean titers (GMT) of live virus neutralizing antibodies (nAb) to Delta (B.1.617.2) and Omicron BA.1 at day 28 post-injection. The secondary endpoints include the safety, day 180 GMTs against Delta and Omicron BA.1, day 28 GMTs to BA.5, and seroresponse rates of neutralizing antibodies and T cell responses at day 28 post-injection. 450 participants, comprising of 449 males and 1 female, with a median age (range) of 27 (18–62) years, are assigned to receive one booster dose of BNT162b2, 20 µg SCTV01C or 30 µg SCTV01E and completed 4-week follow-up. All SCTV01E related adverse events (AEs) are mild or moderate and no Grade ≥3 AE, serious AE or new safety concerns are identified. Day 28 GMT of live virus neutralizing antibodies and seroresponse against Omicron BA.1 and BA.5 with SCTV01E are significantly higher than those with SCTV01C and BNT162b2. These data indicate an overall neutralization superiority with tetravalent booster immunization in men. Pathogenic bacterial infection is one of the principal causes affecting human health and ecosystems. The accurate identification of bacteria in food and water samples is of significant interests to maintain safety and health for humans. Culture-based tests are practically tedious and may produce false-positive results, while viable but non-culturable microorganisms (NCMs) cannot be retrieved. Thus, it requires fast, reliable, and low-cost detection strategies for on-field analysis and point-of-care (POC) monitoring. The standard detection methods such as nucleic acid analysis (RT-PCR) and enzyme-linked immunosorbent assays (ELISA) are still challenging in POC practice due to their time-consuming (several hours to days) and expensive laboratory operations. The optical (surface plasmon resonance (SPR), fluorescence, and surface-enhanced Raman scattering (SERS)) and electrochemical-based detection of microbes (early stage of infective diseases) have been considered as alternative routes in the emerging world of nanostructured biosensing since they can attain a faster and concurrent screening of several pathogens in real samples. Moreover, optical and electrochemical detection strategies are opening a new route for the ability of detecting pathogens through the integration of cellphones, which is well fitted for POC analysis. Although relevant literature investigates the economic value of online sales channel (OSC) from the perspective of the stock market, knowledge on this topic remains insufficient or unclear because existing studies are conducted under an extremely turbulent market environment and have not considered different aspects. This study aims to examine the topic by focusing on the market reactions to OSC investment from three aspects (namely, the innovativeness, business model and goods types) in a relatively stable market environment to fill in the research gap. Empirical results, obtained using 69 firm-level OSC announcements from October 2002 to September 2007, show that the stock market reacts positively to OSC investment by firms. Background: Salt stress seriously impacts the plantation and productivity of global alfalfa (Medicago sativa). Previously, we genetically defined the M. sativa DROUGHT-INDUCED UNKNOWN PROTEIN 1 (MsDIUP1) as a drought tolerance gene of alfalfa. However, whether it is associated with salt tolerance needs to be explored. Results: In this study, we re-classified MsDIUP1 as a putative novel late embryogenesis abundant protein, and found its promoter activity was strongly induced by salt and ABA treatment. Heterologous overexpression of MsDIUP1 in Arabidopsis led to enhanced salt tolerance and ABA insensitivity, with a higher fresh weight, lateral root number, primary root length, and survival rate compared to the wild-type (WT) plants. Similarly, homologous overexpression of MsDIUP1 also enhanced the salt tolerance of alfalfa over-expression (OE) plants, accompanied by reduced oxidative damage, higher K+/Na+ ratio, increased osmoprotectant accumulation, and improved photosynthetic capacity relative to the WT plants. In reverse, disruption of MsDIUP1 expression caused a salt-hypersensitive phenotype of alfalfa RNA interference (RNAi) plants, with higher oxidative damage, less osmoprotectant accumulation, and lower photosynthetic pigment content than that of the WT. These physiological discoveries were in accordance with differential transcript profiles of the relevant genes associated with stress signaling, antioxidant defense, and osmotic adjustment in the salt-treated alfalfa WT, OE, and RNAi lines. Furthermore, we found that alfalfa OE plants showed no apparent alteration in their forage quality compared with the WT plants. Conclusions: Overall, these results reveal that MsDIUP1 improves plant salt tolerance via regulating stress signaling, antioxidant defense, ion homeostasis, osmotic adjustment, and photosynthesis. Load bearing/energy storage integrated devices (LEIDs) allow using structural parts to store energy, and thus become a promising solution to boost the overall energy density of mobile energy storage systems, such as electric cars and drones. Herein, with a new high-strength solid electrolyte, we prepare a practical high-performance load-bearing/energy storage integrated electrochemical capacitors with excellent mechanical strength (flexural modulus: 18.1 GPa, flexural strength: 160.0 MPa) and high energy storage ability (specific capacitance: 32.4 mF cm−2, energy density: 0.13 Wh m−2, maximum power density: 1.3 W m−2). We design and compare two basic types of multilayered structures for LEID, which significantly enhance the practical bearing ability and working flexibility of the device. Besides, we also demonstrate the excellent processability of the LEID, by forming them into curved shapes, and secondarily machining and assembling them into complex structures without affecting their energy storage ability. High-strength composite materials for electrochemical energy storage is attractive for mobile systems. The immunogenicity of SARS-CoV-2 vaccines is poor in kidney transplant recipients (KTRs). The factors related to poor immunogenicity to vaccination in KTRs are not well defined. Here, observational study demonstrated no severe adverse effects were observed in KTRs and healthy participants (HPs) after first or second dose of SARS-CoV-2 inactivated vaccine. Different from HPs with excellent immunity against SARS-CoV-2, IgG antibodies against S1 subunit of spike protein, receptor-binding domain, and nucleocapsid protein were not effectively induced in a majority of KTRs after the second dose of inactivated vaccine. Specific T cell immunity response was detectable in 40% KTRs after the second dose of inactivated vaccine. KTRs who developed specific T cell immunity were more likely to be female, and have lower levels of total bilirubin, unconjugated bilirubin, and blood tacrolimus concentrations. Multivariate logistic regression analysis found that blood unconjugated bilirubin and tacrolimus concentration were significantly negatively associated with SARS-CoV-2 specific T cell immunity response in KTRs. Altogether, these data suggest compared to humoral immunity, SARS-CoV-2 specific T cell immunity response are more likely to be induced in KTRs after administration of inactivated vaccine. Multi-tooth cutting is an effective method for materials machining with high quality and efficiency. To lay a foundation for the feasibility in multi-tooth milling of 2.5D C/SiC composites, combining with a new micro–macro scale numerical model and experimental analysis, the interaction mechanism and damage fracture mechanism between different fiber (0°, 90°, and Z-direction needling punched fiber) and multi-tooth scales mill are studied. Two main mechanisms resulting in fiber fracture are found, namely the extruding flexural fracture and micro-crack propagation fracture. The proposed numerical model is in good agreement with the experimental results in terms of fiber removal, chip forming, and surface topography. Specifically, fiber debonding and micro-crack propagation fracture occur in 0° fiber, and lamellar surface is produced. 90° fiber is mainly broken by shearing action and extruding bending. Correspondingly, the pits appear due to the fiber pulling out. Extruding flexural fracture occurs in Z-direction needle punched fiber, and peeling and lamellar surface is formed. Multi-tooth of the scales mill plays a repairing and grinding effect on the machined surface. And arc micro edge can restrain the formation of the fiber delamination and burr damage in particular. Since masonry structures are prone to collapse in earthquakes, a novel joint reinforcement method with a polypropylene band (PP-band) and cement mortar (CM) has been put forward. Compared with the common reinforcement methods, this method not only facilitates construction but also ensures lower reinforcement cost. To systematically explore the influence of joint reinforcement on the seismic performance of masonry walls, quasi-static tests were carried out on six specimens with different reinforcement forms. The test results show that the joint action of PP-band and CM can significantly improve the specimen's brittle failure characteristics and enhance the integrity of the specimen after cracking. Compared with the specimen without reinforcement, each of the seismic performance indexes of the joint reinforced specimen had obvious improvement. The pandemic of COVID-19 caused by SARS-CoV-2 continues to spread around the world. Mutant strains of SARS-CoV-2 are constantly emerging. At present, Omicron variants have become mainstream. In this work, we carried out a systematic and comprehensive analysis of the reported spike protein antibodies, counting the epitopes and genotypes of these antibodies. We further comprehensively analyzed the impact of Omicron mutations on antibody epitopes and classified these antibodies according to their binding patterns. We found that the epitopes of the H-RBD class antibodies were significantly less affected by Omicron mutations than other classes. Binding and virus neutralization experiments showed that such antibodies could effectively inhibit the immune escape of Omicron. Cryo-EM results showed that this class of antibodies utilized a conserved mechanism to neutralize SARS-CoV-2. Our results greatly help us deeply understand the impact of Omicron mutations. Key message: A major and stable QTL for fertile spikelet number per spike and grain number per fertile spikelet identified in a 4.96-Mb interval on chromosome 2A was validated in different genetic backgrounds. Fertile spikelet number per spike (FSN) and grain number per fertile spikelet (GNFS) contribute greatly to wheat yield improvement. To detect quantitative trait loci (QTL) associated with FSN and GNFS, we used a recombinant inbred line population crossed by Zhongkemai 13F10 and Chuanmai 42 in eight environments. Two Genomic regions associated with FSN were detected on chromosomes 2A and 6A using bulked segregant exome sequencing analysis. After the genetic linkage maps were constructed, four QTL QFsn.cib-2A, QFsn.cib-6A, QGnfs.cib-2A and QGnfs.cib-6A were identified in three or more environments. Among them, two major QTL QFsn.cib-2A (LOD = 4.67–9.34, PVE = 6.66–13.05%) and QGnfs.cib-2A (LOD = 5.27–11.68, PVE = 7.95–16.71%) were detected in seven and six environments, respectively. They were co-located in the same region, namely QFsn/Gnfs.cib-2A. The developed linked Kompetitive Allele Specific PCR (KASP) markers further validated this QTL in a different genetic background. QFsn/Gnfs.cib-2A showed pleiotropic effects on grain number per spike (GNS) and spike compactness (SC), and had no effect on grain weight. Since QFsn/Gnfs.cib-2A might be a new locus, it and the developed KASP markers can be used in wheat breeding. According to haplotype analysis, QFsn/Gnfs.cib-2A was identified as a target of artificial selection during wheat improvement. Based on haplotype analysis, sequence differences, spatiotemporal expression patterns, and gene annotation, the potential candidate genes for QFsn/Gnfs.cib-2A were predicted. Background: The relationship between the ratio of blood urea nitrogen to creatinine (BUN/Cr) and physical frailty in elderly patients remains unclear. The study aims to investigate the association between the BUN/Cr ratio and physical frailty in the elderly Chinese population. Methods: In this cross-sectional analysis, the clinical data of 5213 participants from 2015 were selected from the China Health and Retirement Longitudinal Study (CHARLS). The demographic variables (including age and gender) and health behavior (including smoking and drinking history), anthropometric (including systolic and diastolic blood pressure, waist circumference (WC), etc.), physical performances (i.e., grip strength, repeated chair stands, etc.), and biochemical indicators (i.e., blood urea nitrogen (BUN), creatinine(Cr), total cholesterol (TC), triglycerides (TG), etc.) were measured. The association between the BUN/Cr ratio and physical frailty was analyzed. Results: After adjusting for potential confounding factors, smooth curve fitting showed a linear relationship between the BUN/Cr ratio and grip strength, a non-linear relationship between the BUN/Cr ratio, and repeated chair-rising time. The fully adjusted linear regression results showed a negative association between the BUN/Cr ratio and grip strength. In the multivariate, piecewise linear regression, when the BUN/Cr ratio was greater than 18.60, the repeated chair-rising time increased with the increase in BUN/Cr ratio (β = 0.046, 95%CI 0.025, 0.066; p < 0.001). However, we did not observe a significant correlation when the BUN/Cr ratio was less than 18.60 (β = −0.007, 95%CI −0.046, 0.032; p = 0.717). Welding defects are unavoidable for welded structures, which can lead to fatigue damage even under the random wind load with small amplitude. It is therefore necessary to explore the effect of welding defect on the fatigue properties of welded steel pipes. Three groups of welded steel pipe specimens were designed according to welding defect conditions, i.e. specimens without welding defect (Group I), specimens with incomplete fusion (Group II), and specimens with welding porosity (Group III). Uniaxial tension–compression and torsion high-cycle fatigue tests were carried out. S–N curves of uniaxial tension–compression and torsion tests were obtained by cyclic loading with equal stress amplitude. The test results show that the high-cycle fatigue strength of weldments is obviously lower than that of base metal with the same strength under uniaxial tension–compression and torsion loading. In addition, the welding defects result in a decrease in fatigue strength, while the decrease extent by welding porosity is greater than that by incomplete fusion. Finally, because of the inherent multiaxial loading characteristics of welded structures, the high-cycle multiaxial fatigue life of steel pipe weldments was also predicted by using the modified Wöhler curve method based on the uniaxial fatigue test results. It can be found that when the stress amplitude is constant, the fatigue life of welded steel pipe decreases and the modified Wöhler curves move downward more quickly with the increase of damage parameter defined as the ratio of normal stress amplitude to shear stress amplitude on the critical plane, which means that normal stress amplitude will accelerate the cracks growth and result in faster failure of the weld materials. Background: Tecarfarin (ATI-5923), a structural analog of warfarin, was designed to provide more uniform and stable anticoagulation. Objective: We aimed to evaluate the safety, tolerability, pharmacokinetic and pharmacodynamic profile of tecarfarin when administered in multiple ascending doses (MADs) to healthy Chinese volunteers. Methods: Forty healthy Chinese volunteers were enrolled into four sequential cohorts (10, 20, 30, and 40 mg), with 10 subjects in each cohort. Participants in the MAD study for each sequential cohort were dose-titrated to achieve the target international normalized ratio (INR 1.7–2.0) for 14 days. Safety and tolerability were assessed throughout the study. Results: The pharmacokinetic and pharmacodynamic profile of tecarfarin was investigated in a healthy Chinese population. Dose titration of tecarfarin was necessary to keep the INR in the target range in all subjects in the 20, 30 and 40 mg cohorts and a few subjects (n = 3) in the 10 mg cohort. Tecarfarin was well tolerated without serious adverse events. Only one treatment-related adverse event (hematochezia) resulted in early withdrawal from the MAD 40 mg cohort. Conclusion: Tecarfarin was well-tolerated by Chinese volunteers. Dose titration was needed for tecarfarin doses larger than 20 mg to keep the INR in the target range. Introduction: Evidence regarding the association between body mass index (BMI) and outcomes in non-cardiothoracic, non-obstetric surgery patients with pulmonary hypertension (PHTN) is limited. We aimed to investigate the association between BMI and 30-day mortality in non-cardiothoracic, non-obstetric surgery patients with pulmonary hypertension. Material and Methods: We performed a secondary analysis of non-cardiothoracic, non-obstetric procedures on patients with PHTN between 2007 and 2013 at a single institution. Multivariable logistic regression models were used to estimate the association between BMI and 30-day mortality. Interaction and stratified analyses were conducted according to age, sex, PHTN medical therapy, anticipated inpatient or outpatient post-procedure status, procedure length > 2 h, open surgical approach, and PHTN severity class. Results: A total of 513 participants were included, with men accounting for 55.8% of the sample; the 30-day mortality was 1.9%. In the multivariate regression models, BMI was significantly associated with 30-day mortality after adjusting for potential covariates. A 1 kg/m2 increase in BMI was associated with a 15% decrease in 30-day mortality (adjusted OR = 0.85, 95% CI: 0.73–0.98, P = 0.03). Stratified analyses indicated that this negative correlation was robust. Conclusions: In this study, we found that low BMI is associated with increased risk of death in patients undergoing non-cardiothoracic, non-obstetric surgery requiring general anesthesia. To improve the financing performance, optimization models have been developed to analyze the operation mechanism of crowdfunding. However, network externality, which has proven to exist in crowdfunding, was ignored in previous studies. In order to address this gap, we construct an optimization model incorporating network externality and investigate its impact on crowdfunding operation decisions. Our analysis reveals that network externality does not monotonously influence optimal prices, while it always improves the project quality. In addition, we prove that although network externality increases the creator's profit, it increases the price discrimination level and may hurt consumers, thus reducing social welfare. In extensions, we investigate the effects of demand uncertainty, external financing, and word-of-mouth effect on the optimal decisions. Purpose: To study prognostic values of distal ureter involvement (DUI) and survival outcomes in bladder cancer at T1 and T2 stages. Methods: The national Surveillance, Epidemiology, and End Results database (2004–2015) was applied to obtain bladder cancer patients. We used the Kaplan–Meier method with the log-rank test, subgroup analyses, the multivariable Cox proportional hazard model and propensity score matching (PSM). Results: A total of 490 patients with DUI and 28,498 patients with non-DUI (non-involvement) were enrolled in our study. After 1:1 PSM, 490 matched pairs were picked out. The multivariable Cox regression before and after PSM revealed that the DUI group had a high risk of overall mortality (HR = 1.374, P < 0.001 before PSM; HR = 1.513, P < 0.001 after PSM) and cancer-specific mortality (HR = 1.632, P < 0.001 before PSM; HR = 1.699, P < 0.001 after PSM). The results of survival analyses showed that patients in the DUI group had lower survival probability in OS (P = 0.0011) and CSS (P < 0.0001) analyses. Nevertheless, in the subgroup analysis, significant differences were only observed in the T1 stage in terms of CSS and T2a stage in terms of OS and CSS (all P < 0.05). Conclusion: The prognosis of DUI was poorer than that of non-DUI. DUI was an independent risk factor for OM and CSM in bladder cancer at T1 and T2 stages especially for those at T1 and T2a stages. Exhaled volatile organic compounds (VOCs) have been widely applied for the study of disease biomarkers. Oral exhalation and nasal exhalation are two of the most common sampling methods. However, VOCs released from food residues and bacteria in the mouth or upper respiratory tract were also sampled and usually mistaken as that produced from body metabolism. In this study, exhalation from deep airway was first directly collected through intubation sampling and analyzed. The exhalation samples of 35 subjects were collected through a catheter, which was inserted into the trachea or bronchus through the mouth and upper respiratory tract. Then, the VOCs in these samples were detected by proton transfer reaction mass spectrometry (PTR-MS). In addition, fast gas chromatography proton transfer reaction mass spectrometry (FGC-PTR-MS) was used to further determine the VOCs with the same mass-to-charge ratios. The results showed that there was methanol, acetonitrile, ethanol, methyl mercaptan, acetone, isoprene, and phenol in the deep airway. Compared with that in oral exhalation, ethanol, methyl mercaptan, and phenol had lower concentrations. In detail, the median concentrations of ethanol, methyl mercaptan, and phenol were 7.3, 0.6, and 23.9 ppbv, while those in the oral exhalation were 80.0, 5.1, and 71.3 ppbv, respectively, which meant the three VOCs mainly originated from the food residues and bacteria in the mouth or upper respiratory tract, rather than body metabolism. The research results in our study can provide references for expiratory VOC research based on oral and nasal exhalation samplings, which are more feasible in clinical practice. [ABSTRACT FROM AUTHOR]
