In the grinding process of crankshafts, the grinding parameters were improperly selected, which easily caused the high temperature in the grinding zone and led to burns on the crankshaft journal surface. To address this issue, referencing previous studies, the suitability of the Gaussian heat source model was verified by comparing it with three different heat source models. The Gaussian heat source model was applied to predict the temperature on the surface of the crankshaft connecting rod journal. Transient thermal analysis and investigation of material phase transformation were employed to explore the mechanism of grinding burns. The interaction effects among grinding parameters were analyzed using the Box-Behnken design, and a multiple linear regression equation was established for response surface optimization. The results demonstrated that applying the Gaussian heat source model and response surface optimization yielded the optimal solution, with a maximum deviation of 1.37% between the simulation and optimization results. By selecting a grinding depth (ap) of 0.40 mm, wheel speed (vs) of 40.00 m/s, and feed rate (vx) of 0.036 mm/s as the processing parameters, the temperature in the grinding zone was reduced to 659.37 °C, effectively mitigating grinding burns. We study studied the alpine meadow soil in Gannan Prefecture. Geostatistical analysis showed that Circular [soil total nitrogen (STN), soil total potassium (STK), soil available phosphorus (SAP)] model, index [bulk density (BD), soil water content (SWC), soil organic carbon (SOC), soil available nitrogen (SAN)] model and Gaussian [soil total phosphorus (STP), soil available potassium (SAK)] model is universal Kriging Method semi-variation function optimization fitting model. In the model, the spatial correlation is weak to medium, and SOC, STN, STP, STK, SAN, SAP, SAK, and SWC have specific spatial trend effects. Then principal component analysis was used to obtain three key factors (SOC, BD, and SWC) affecting soil quality, which accounted for 81.91% of the total variation of soil quality. These factors were taken as the minimum data set (MDS) and soil quality index (SQI) was calculated. According to SQI, the whole region was divided into three soil management zones (SMZs). Aims: Within root systems, framework pioneer roots mainly function in soil exploration and resource transport, while fibrous roots and their associated fungi extend the foraging soil space for resource absorption. However, the plasticity of pioneer and fibrous roots in response to elevated nutrient availability has received relatively little attention. Methods: For three hardwood species, newly formed root branches were harvested in order to compare pioneer and fibrous roots for variation in morphology, mycorrhizal colonization and anatomy, as well as their trait shifts in response to fertilization. Results: Compared with the fibrous roots, pioneer roots were generally larger in diameter, stele and cortex size, root tissue density, and hydraulic conductivity, but lower in specific root length (SRL) and mycorrhizal colonization rate (MCR). Fertilization generally decreased SRL and MCR in fibrous roots, and increased conduit diameters in pioneer roots. Additionally, fertilization enhanced hydraulic conductivity for all three species: 162, 45 and 263% in pioneer roots, and 46, 13 and 172% in fibrous roots for Juglans mandshurica Maxim., Fraxinus mandshurica Rupr. and Phellodendron amurense Rupr., respectively. Conclusions: Pioneer and fibrous roots responded differently to fertilization. The resulting enhanced resource transportation capacity may serve to mitigate the toxic effect in fibrous roots and avoid pioneer roots becoming a bottleneck for absorbed water and nutrient in an expanding root system, suggesting tightly associated traits and functional equilibrium within an individual root and even the root branches. Objective: To analyze the prognostic significance of plasma total cholesterol (TC) and high-density lipoprotein (HDL) in patients with gastric cancer receiving oxaliplatin combination with S-1 (SOX) chemotherapy after radical resection and to establish models of relevant prognostic influencing factors. Method: The clinicopathologic features of 301 patients treated with SOX after radical gastrectomy were retrospectively analyzed. Univariate analysis, multivariate analysis, and the Kaplan–Meier survival curve were used to analyze the prognostic value of TC and HDL in patients undergoing adjuvant SOX chemotherapy after curative gastric surgery. Based on the results of multivariate Cox regression, we constructed nomograms to predict 1 and 3-year CSS (cancer-specific survival time) and DFS (disease-free survival time) in patients with adjuvant chemotherapy after radical gastrectomy. We assessed the model's accuracy using the consistency index (C index) and calibration curve, and the ROC curve and DCA curve were applied to compare it with TNM staging. Results: Multivariate analysis revealed that TC and HDL were independent influencing factors of CSS, while HDL was an independent factor unique to DFS. According to Kaplan–Meier curves, low TC and HDL suggested poor survival (P < 0.001). The relevant prognostic factors from the multivariate study were used to build nomograms for DFS and CSS. Both DFS and CSS had C index and AUC values that were higher than 0.71. The calibration curves demonstrated that the predicted results are consistent with the observed results. The AUC valves for DFS and CSS in our models were higher than TNM staging. The decision curve analysis indicated that net benefits were moderately positive. Significant differences in survival were seen between the high and low-risk groups according to the nomogram risk score. Conclusions: TC and HDL have a certain significance for the prognosis of patients with gastric cancer after radical resection receiving adjuvant SOX chemotherapy. Lower TC and HDL suggested poor DFS and CSS. Both prediction models for CSS and DFS demonstrated good predictive ability and had a higher predictive value than the TNM staging system. With the prevalence of service composition, how to recommend API services that meet the Mashup requirements for developers has become a challenging in the field of service computing. Existing works on Web service recommendation typically conduct service recommendation based on the semantic requirements and interaction relationships of Web services, while ignoring the construction intent of Mashup creators. Moreover, they often only focus on the impact of single intent on recommendation, neglecting the diverse requirements of Mashup creators at different intent-levels. As a result, the recommendation models fail to comprehensively understand the construction intent of Mashup creators, which affects the quality and effectiveness of Web service recommendation. To address this problem, this paper proposes an intent recognition-based API recommendation method, denoted as ARIR. This method utilizes the annotation information of Mashup and API to analyse the creation intent of Mashup and the functional intent of API, thereby providing more accurate API with high-quality to Mashup creators. Firstly, it decouples the representations of Mashup and API at different intent-levels and independently initializes the node representations at each intent-level. Secondly, it trains the vector representations at each intent-level using a decoupled graph convolutional neural network module and obtains the representation vectors of Mashup/API at different intent-levels with attention weights. Then, it aggregates the intents of Mashup and API using the Mashup-API interaction relationships, resulting in the final node representations of Mashup/API. Furthermore, it constructs a similarity heterogeneous network by calculating the Mashup-to-Mashup similarity and API-to-API similarity, updating the node representations by using the Mashup and API feature matrices and adjacency matrices, and obtaining the final recommendation prediction results by using a fully connected layer. Finally, the experimental results conducted on real-world Web service datasets demonstrate that the ARIR outperforms the best-performing baseline method with Recall@20 of 1.1% and NDCG@20 of 3.3%. Aims: Functional traits play key role in plant resource-use strategy, but the intraspecific variation of root traits particularly of anatomy along large-scale environmental gradient has been poorly understood. Methods: Here, we examined 16 morphological, anatomical, and chemical traits of leaves and absorptive roots (i.e., the first-order roots) for two wide-planted tree species, Ginkgo biloba and Eucommia ulmoides, sampled at five locations from subtropical, temperate, and cold-arid regions in China. Results: Morphological traits in leaves tended to shift from conservative at resource-poor sites to acquisitive at resource-rich sites, showing lower leaf thickness and larger specific leaf area, while roots showed a reverse pattern. All anatomical traits in leaves and roots inclined to be more conservative in resource-poor sites. Leaves and roots generally showed negative relationships in analogous morphological traits, but positive in anatomical and chemical traits across both species. Intraspecific variation of leaf traits confirmed the existence of one-dimensional economics spectrum, whereas root traits displayed multidimensional variation, showing an independent dimension of cortex to stele size ratio. Conclusions: Leaf and root traits in both species showed considerable intraspecific variations under changing environments, which improved whole-plant-level adaptions to climate and soil constraints, and exhibited different above- and belowground intraspecific economics spectra. Background and aims: The rate of mycorrhizal colonization in plant roots generally decreases with increased soil nutrient availability but the underlying mechanism is still poorly understood. Our aims were to explore the responses of root mycorrhizal colonization, passage cells, and penetration points under nitrogen (N) and phosphorus (P) addition and their potential linkages in woody and herbaceous plants. Methods: N and P were added to the pots of 14 temperate species (eight woody and six herbaceous) in the greenhouse, and the distribution of mycorrhizal colonization, passage cells and penetration points for each species were observed by staining and microscopy of first-order roots. Results: The average density and proportion of mycorrhizal colonization, passage cells and penetration points of 14 species were significantly decreased under N and P addition. The N addition had a stronger effect on the plants than the P addition. More importantly, the mycorrhizal colonization density and proportion showed significant positive correlations with passage cell density and proportion, and with penetration point density and proportion of woody and herbaceous plants under control and nutrient treatments. Conclusions: The density and proportion of mycorrhizal colonization were closely related with passage cells and penetration points in both woody and herbaceous species in either control or nutrient treatments. This study fabricated four kinds of CoCrMnFeNiX0.1 (X = Al, Cu, Mo, Ti) high-entropy alloys through rolling with subsequent thermal treatment, to investigate the alloying effects on the microstructural evolution, mechanical properties, and electrochemical corrosion performance. High-density parallel twinning bands were observed in the as-rolled CoCrMnFeNiX0.1 alloys, excluding the Cu0.1 variant, signifying that plastic deformation was dominated by twinning. However, detwinning and thickening of twin layers was observed after annealing. This effect was more pronounced in the rolling direction-normal direction (RD-ND) plane due to the stress state induced by rolling. The introduction of Cu altered the deformation mechanism to a combination of twinning and slipping, resulting in fewer twinning bands. Notably, annealing induced the formation of abundant bamboo-shaped twinning bands (width: ~ 10 nm) in the Cu0.1 alloy, which were correlated with nano precipitated particles. Additionally, the incorporation of Mo or Al elements activated more twinning directions, thereby enhancing plasticity. For the Ti0.1 alloy, the primary factors influencing the altered tensile and electrochemical performances were the formation and growth of few precipitated particles during annealing. Consequently, the strengthening effect can be ranked in the order of Mo > Ti > Al > Cu. Therefore, the addition of Al and Ti is recommended for improving the anticorrosive property and thermal stability of recrystallized grains. [ABSTRACT FROM AUTHOR]

Purpose: This study aimed to assess the pharmacokinetics (PK) and pharmacodynamics (PD) of ivosidenib in Chinese patients with relapsed or refractory acute myeloid leukemia (R/R AML) carrying the mIDH1 mutation. Methods: A bridging study (NCT04176393) was conducted involving 29 Chinese patients who received a daily dose of ivosidenib 500 mg in 28-day cycles. Plasma concentrations of ivosidenib and D-2-hydroxyglutarate (2-HG) were measured before and after treatment. Non-compartmental analysis (NCA) was employed to evaluate the PK, and an established population pharmacokinetic (popPK) model developed from non-Chinese patients was externally validated. Results: The findings revealed comparable PD effects of ivosidenib in Chinese patients with mIDH1 R/R AML. After adjusting for concomitant drug effects, PK characteristics were similar between Chinese and non-Chinese patients. Furthermore, the popPK model offered additional insights into the possible causes of the apparent ethnic difference in PK exposure. Conclusion: The study indicates that ivosidenib can be used effectively in Chinese patients, and the observed ethnic differences in PK exposure can be explained by concomitant drug effects. In this study, five types of economic forest plots were selected as sample plots (Malus pumila, Juglans regia, Zanthoxylum bungeanum, Prunus persica, and Prunus armeniaca) in Gangu County, Gansu Province, and wasteland was used as a control to investigate the changes of soil organic carbon content in 0–100 cm of different economic forests in Gangu County. The results showed that the soil organic carbon content of different economic forests in Gangu County ranged from 12.65 ± 0.09...7.20 ± 0.13 g/kg in layer 0–10 cm. The soil organic carbon content at 0–100 cm depth ranged from high to low: apple (Malus pumila), pepper (Zanthoxylum bungeanum), walnut (Juglans regia), apricot (Prunus armeniaca), peach (Prunus persica) and wasteland. The SOC content of the five forest types and the wasteland showed apparent surface aggregation, with the highest SOC content of 17.20 g/kg for Malus pumila and the lowest SOC content of 12.65 g/kg for the wasteland in the 0–10 cm surface layer; in the 80–100 cm deep layer, the highest SOC content of 14.90 g/kg for Juglans regia and the lowest SOC content of 9.17 g/kg for the wasteland. SOC content was positively correlated (p < 0.05, n = 126) with soil water content, soil enzyme activity and soil microbial population, and significantly negatively correlated (p < 0.05, n = 126) with pH and soil bulk density. Overall, the mean value of SOC content in 0–100 cm soils of Malus pumila and Zanthoxylum bungeanum is higher among the five economic forest species in Gangu County. The mean SOC content of Malus pumila and Zanthoxylum bungeanum in 100 cm soil is relatively high among the five economic forest species, therefore Malus pumila is the preferred choice for economic forest planting in Gangu County, and in the future economic development and ecological restoration practice of Gangu County, it is recommended to plant a mixed forest planting pattern of Malus pumila and Zanthoxylum bungeanum, which is conducive to improving the potential carbon sink function of economic forests in the area. The rational design of efficient electrocatalysts with favorable electronic structures and geometries can significantly influence their analytical performance for non-enzymatic glucose detection. In this work, nickel hydroxide nanocages on nickel foam (Ni(OH)2 NCs/NF) were fabricated by an ion-assisted etching method using zinc-based imidazole zeolite framework (ZIF-8) as a structural template. Due to the hollow nanocage structure of Ni(OH)2 NCs/NF, more exposed active sites and larger void space were obtained. The prepared hollow Ni(OH)2 NCs were used as a working electrode for electrochemical sensing glucose with high electro-catalysis performance under alkaline conditions. Under the optimal conditions, the Ni(OH)2 NCs/NF presents a short response time (less than 3 s) and a wide linear range from 1 μM to 3.45 mM for glucose assay. A low detection of limit (LOD) of 0.1 μM (S/N = 3) and an excellent sensitivity of 7413 μA mM−1 cm−2 were obtained by the prepared non-enzymatic electrochemical glucose sensor. In addition, the constructed Ni(OH)2 NCs exhibits good long-term stability and high selectivity for glucose detection. Objective: Although human umbilical cord-derived mesenchymal stem cells (HU-MSCs) have attracted increasing attention because of their pivotal functions in the process of wound healing, the underlying molecular mechanisms have been poorly understood. It has been shown that the TGF-β/Smad signaling pathway plays an important role in the process of scar formation. The present study focused on exploring whether HU-MSCs improve uterine incision healing after cesarean delivery in rats via the TGF-β/Smad signaling pathway. Study Design: Pregnant rats were randomly assigned to three groups, including the NP group, incision-injected group (HU-MSCs1 group), and tail vein-injected group (HU-MSCs2 group), and 30 days after cesarean section, sampling was carried out to further explore the specific mechanisms from tissue and protein levels. Results: HU-MSCs secretion could inhibit the fibrosis of scar tissue. We observed that the TGF-β induced expression of TGF-β1, Smad2, and Smad3 was attenuated upon HU-MSCs treatment in scar tissue, while the decrease in TGF-β3 expression was enhanced by HU-MSCs. Furthermore, HU-MSCs treatment accelerated wound healing and attenuated collagen deposition in a damaged uterine rat model, leading to the promoting of uterine incision scarring. In addition, the expression of alpha-smooth muscle actin (a-SMA) was enhanced by HU-MSCs treatment. Growing evidences have revealed that a histone deacetylase inhibitor (HDACi), suberoylanilide hydroxamic acid (SAHA) has anti-fibrotic effect in different diseases. In this study, we first evaluated whether SAHA could suppress cardiac fibrosis. Mice with MI-induced cardiac fibrosis were treated with SAHA by intraperitoneal injection and their cardiac function was improved after SAHA treatment. Results of western blotting and qRT-PCR in heart tissues suggested that TGFβ1/P38 pathway was activated in MI mice, and this effect was reversed by SAHA. Cell proliferation assay suggested that SAHA could suppress TGF-β1-induced cardiac fibroblasts proliferation. Furthermore, results of western blotting and qRT-PCR in cardiac fibroblasts depicted that SAHA may exert its anti-fibrotic effect through inhibiting TGF-β1-induced P38 phosphorylation by promoting DUSP4 expression. Single-molecule localization microscopy enables three-dimensional fluorescence imaging at tens-of-nanometer resolution, but requires many camera frames to reconstruct a super-resolved image. This limits the typical throughput to tens of cells per day. While frame rates can now be increased by over an order of magnitude, the large data volumes become limiting in existing workflows. Here we present an integrated acquisition and analysis platform leveraging microscopy-specific data compression, distributed storage and distributed analysis to enable an acquisition and analysis throughput of 10,000 cells per day. The platform facilitates graphically reconfigurable analyses to be automatically initiated from the microscope during acquisition and remotely executed, and can even feed back and queue new acquisition tasks on the microscope. We demonstrate the utility of this framework by imaging hundreds of cells per well in multi-well sample formats. Our platform, implemented within the PYthon-Microscopy Environment (PYME), is easily configurable to control custom microscopes, and includes a plugin framework for user-defined extensions. Vibration signals of rolling element bearings (REBs) contain substantial bearing motion state information. However, the property of nonlinear and nonstationary vibration signals decreases the diagnostic accuracy of REBs. To improve the accuracy of fault diagnosis for REBs, an ensemble approach based on ensemble empirical mode decomposition (EEMD), multi-scale permutation entropy (MPE), and backpropagation (BP) neural network optimized by genetic algorithm (GA) is proposed. Firstly, the REBs are decomposed into a set of intrinsic mode functions (IMFs) that contain various fault features by EEMD. The fault features of the first four IMFs are extracted by MPE, and the feature vectors are formed. Then, the BP neural network optimized by GA is utilized as a classifier for fault diagnosis to train and test the feature vector set, and the fault diagnosis of the REBs is realized in the form of probability output. Experimental results show that the proposed method can identify the fault pattern of the vibration signals of REBs precisely. Compared with the existing fault diagnosis methods, the proposed method can realize the fault diagnosis of REBs with 16 fault patterns, and demonstrates an excellent accuracy rate. Patients treated with immune checkpoint inhibitors (ICIs) often experience unique immune-related adverse events (irAEs), and the previous studies demonstrated an association between irAEs and better outcomes in patients with ICI treatment for advanced non-small cell lung cancer (NSCLC). However, the correlation between the occurrence of mild and severe irAEs and prognosis remains unclear. Additionally, little is known regarding the association between the timing of mild and severe irAEs and clinical outcomes. We retrospectively conducted a multicenter study of advanced NSCLC patients treated with ICI monotherapy. Of the 222 patients, 79 patients (35.6%) experienced at least one irAE, and most were of grade 1 or 2 (mild) (26.6%). The most common irAEs were pneumonitis (n = 21, 9.5%) and skin-related adverse reactions (n = 19, 8.6%). The median progression-free survival of all patients treated with ICIs was 3.2 months. Patients experiencing irAEs had a better prognosis than those without such events (6.5 vs. 2.6 months, p = 0.004), and mild irAEs were associated with the best prognosis. The difference in overall survival between mild and severe irAEs was significant (34.3 vs. 17.3 months, p = 0.021). We further analyzed differences between patients with irAEs occurring at 3 or 6 weeks, and found that the earlier the occurrence of mild irAEs, the better the prognosis; however, the opposite was true for severe irAEs. In summary, patients with early occurring mild irAEs showed better clinical outcomes, whereas those with early severe irAEs tended to show poorer clinical outcomes. [ABSTRACT FROM AUTHOR]

Objectives: To prospectively investigate the capability of intravoxel incoherent motion (IVIM) and conventional diffusion tensor imaging (DTI) to identify early kidney function injury in type 2 diabetes.Methods: Forty-one diabetes patients (normoalbuminuria: n = 27; microalbuminuria: n = 14) and 28 volunteers were recruited. All participants were examined using DTI and IVIM with 3.0-T MRI. DTI parameters (mean diffusivity [MD], fractional anisotropy [FA]), and IVIM parameters (true diffusion coefficient [D], pseudo-diffusion coefficient [D*], and pseudo-diffusion component fraction [f]) were measured in the renal parenchyma (cortex and medulla) by two experienced radiologists independently. Image features were compared among the groups using separate one-way analyses of variance. Diagnostic performances of various diffusion parameters for predicting diabetic renal damage were compared.Results: The medullary D and FA values were significantly different among the microalbuminuria subgroup, normoalbuminuria subgroup, and control group (all p < 0.001). In medulla, area under the curve (AUC) values for combined FA and D were significantly higher than single FA (AUC = 0.938, 0.769, respectively; p = 0.003), and the combined AUC of FA and D was numerically higher than that of single D (0.938 vs 0.878, p > 0.05). AUC of combined FA and D was 0.985, not significantly different from individual AUC for FA and D (AUC = 0.909 and 0.952, respectively; all p > 0.05) in differentiating the microalbuminuria subgroup from the control group.Conclusion: IVIM-derived D and DTI-derived FA values were better than other parameters for evaluating early kidney impairment of diabetes.
