application/pdf, Whilst molecular biological articles have been extensively described in high school-level textbook guidelined by MEXT, it is, however, commonly deemed to be practically difficult to perform molecular biological experiments in high schools. We herein report an ingenious protocol for molecular biological experiments best suitable for biology classes in Japanese high schools, comprising DNA extraction, DNA detection, PCR and agarose electrophoresis, all of which can be completed substantially within no more than two classes. In order to exploit the protocol, we cautiously cared the following points: sets of the experiments can be accomplished in typical period in high schools (50 minutes); and most of the listed experiments can be performed in standard high school facilities. For DNA extraction, “silica monolith” spin column (Animos Co. Ltd.) was employed, much facilitating and shortening the procedure (completed within ~10 minutes). DNA from rice powder (O. sativa) and flour (T. aestivum) was successfully detected by means of agarose electrophoresis, followed by PCR amplification by Taq polymerase reaction mixture (Quick Taq® HS DyeMix; Toyobo Co. Ltd.), targeted for RuBisCO small subunit gene through O. sativa specific primers. The amplification was confirmed by agarose electrophoresis, non-hazardously stained by Midori Green (Nippon Genetics Co. Ltd.). To further develop the experimental protocol, it is thought to be necessary not only to supply the equipment required for routine molecular biological experiments in the high schools, but also to manufacture either illustrated or visualised protocol, expecting for future facilitation of molecular biological learning in high schools.

Artikel ini bertujuan untuk melihat strategi dakwah BAZNAS Provinsi Bengkulu dalam pengentasan kemiskinan. Metode dalam penelitian ini ialah mixed metode dengan pendekatan fenomenologi. Teknik pengumpulan data yaitu wawancara, observasi dan dokumentasi, dengan teknik analisis model interaktif. Hasil penelitian ini menunjukan bahwa dalam mengoptimalkan pengelolaan zakat harus berorientasi pada nilai-nilai dakwah berbasis maqosyid syariah dalam mewujudkan misi dakwahnya sebagai pengentasan kemiskinan. Strategi yang digunakan ialah dengan menyusur pada menejemen dakwah yaitu tahap perencanaan. Pada tahap ini ada beberapa strategi yang digunakan oleh BAZNAS Provinsi Bengkulu dalam pengelolaan organisasi yang menangani zakat. Diantaranya dengan berbagai metode dakwah bil-lisan, bil-qolam dan bil hal dengan pemberdayaan masyarakat dengan program ekonomi, sosial, pendidikan dan dakwah. Namun, strategi perencanaan BAZNAS Provinsi Bengkulu mengalami beberapa kendala, terutama dalam hal anggaran.

Remaja putri sering mengalami masalah kesehatan salah satunya anemia. Anemia adalah kondisi jumlah konsentrasi hemoglobin di dalamnya lebih rendah dari normalnya. Secara global prevalensi anemia di dunia tahun 2019 adalah 29.9 % (WHO, 2020). Prevalensi anemia di Indonesia tahun 2018 yaitu 48,9% . Sebagian besar anemia pada remaja Indonesia diakibatkan kekurangan zat besi. Kacang merah mengandung zat besi, mudah di dapat di daerah Kokap dan harganya terjangkau, masyarakat setempat terbiasa mengkonsumsi kacang merah dalam bentuk hidangan masakan atau jus. Untuk mengetahui pengaruh jus kacang merah terhadap kadar hemoglobin pada remaja putri di, penelitian ini menggunakan metode eksperimental semu (quasy experimental) dengan pendekatan non equivalent control group. Teknik pengambilan sampel menggunakan Purposive Sample dengan jumlah sampel sebanyak 30 orang remaja putri dibagi menjadi kelompok kontrol dan kelompok intervensi masing-masing 15 orang. Kelompok Intervensi diberikan kacang merah 50 gram dalam bentuk jus sebanyak 250 ml sebanyak 1 kali sehari selama 14 hari. Dan kelompok kontrol tidak diberikan perlakuan jus kacang merah. Pengukuran kadar hemoglobin dilakukan sebelum dan sesudah pemberian jus kacang merah menggunakan cyanmethoglobin oleh petugas dan di uji dengan Wilcoxon Test. Berdasarkan data kejadian anemia pada responden sebanyak 30 orang dengan anemia ringan 23 orang, anemia sedang 4 orang dan tidak anemia 3 orang. Hasil penelitian menunjukkan Rata-rata kadar hemoglobin sebelum pada kelompok kontrol yaitu 11.17 g/dl dan setelah yaitu 11.14 g/dl. Rata-rata kadar hemoglobin sebelum pemberian jus kacang merah pada kelompok intervensi yaitu 11.14 g/dl dan setelah pemberian yaitu 13.16 g/dl. Hasil penelitian menggunakan uji Wilcoxon diperoleh nilai signifikan 0,001 (p< 0,05).Terdapat pengaruh pemberian jus kacang merah terhadap kadar hemoglobin pada remaja putri di Pondok Pesantren Nurul Quran Kecamatan Kokap Kabupaten Kulon Progo Provinsi Daerah Istimewa Yogyakarta.

This study aims to examine and analyze the effect of liquidity, solvency, activity, and firm size on the profitability of companies listed on the Indonesia Stock Exchange for the 2016-2021. Profitability is proxied by ROA, liquidity is proxied by CR, solvency is proxied by DER, activity is proxied by Asset turnover, and company size is proxied by Ln (total assets). This study uses an associative quantitative approach with a causal nature. The population in this study are companies listed on the Indonesia Stock Exchange. There are 15 companies selected as samples using purposive sampling method. In this study, the analytical technique used is multiple linear regression analysis. Based on the results of simultaneous hypothesis testing, obtained a significant effect on profitability, with the results of the coefficient of determination test of 87%. Based on the results of partial hypothesis testing, liquidity, solvency, firm size affect profitability while activity has no effect on profitability.

This study aims to determine public perceptions of advertising to prevent the spread of the Covid-19 virus. To describe this perception, the researcher focused on several sub-categories, namely applications through the sense of sight and sense of hearing so as to produce people's perceptions of messages in advertisements. The research method in this study used a qualitative approach, namely descriptive analysis. The subjects in this study were 3 people. Data collection techniques through in-depth interviews, observation, documentation and internet searching. The results of the study illustrate that, the application of perception through the sense of sight is very positive, where people consider the visuals displayed in this advertisement to be very attractive so that people do not get bored seeing it. The application of perception through the sense of hearing is also very good, because the audio presentation displayed in this advertisement uses language that is easy to understand and is accompanied by music. The public's perception of this message as a whole is positive. The conclusion of this research is the public's perception is that this advertisement attracts people's attention and changes people's behavior to follow the contents of the message in the advertisement, namely steps to protect themselves from the spread of the Covid-19 virus.

OBJECTIVE Identifying current occupant travel patterns can inform decision making regarding the design, regulation, and occupant protection systems helpful for automated vehicle systems and mobility services. METHODS Two travel data sets were analyzed to quantify travel patterns: the 2017 National Household Travel Survey (NHTS), which provides data on household trips logged for a 24-h period, and the 2011-2015 National Automotive Sampling System-General Estimates System (NASS-GES), which contains data sampled from police-reported crashes. Analysis identified trends with driver age and gender, occupant age and gender, time of day, day of week, trip purpose, trip duration, vehicle type, as well as occupant role as solo driver, driver of others, single passenger, or multiple passengers. RESULTS In NHTS, the median trip duration is 15 min; only 10% of trips last longer than 40 min. Trip duration does not vary with occupant role or vehicle type. Variations with trip time of day and day of week show a unimodal pattern for weekends, as well as weekday trips for those aged 55 years and older and non-solo occupants aged 18 to 29 years. Other occupant groups have a bimodal weekday travel pattern with peak trips corresponding to morning and evening rush hours.In GES, approximately half of occupants are solo drivers. Female drivers aged 55 and older travel alone 60% of the time, and drivers under age 18 and female drivers aged 30 to 54 drive alone on less than 45% of trips. Approximately 13% of occupants are single passengers, and 16% travel with a driver and at least 1 other passenger. About 16% of occupants are front seat passengers. CONCLUSIONS This analysis of vehicle occupancy provides insights on what ridership of future automated vehicles and expanded ride-hailing services may look like. Because half of occupants are solo drivers, only 16% are multiple passengers, and median trip length is just 15 min, proposed alternative seating arrangements intended to promote comfort and passenger interaction may not represent the typical future vehicle use case in the United States. Knowledge of current occupancy patterns can help automated vehicle designers and regulators develop safe seating scenarios that meet customer needs.
