Abstract

We examine the prevalence of truncated star-forming disks in the Virgo cluster down to $M_* \simeq 10^7 ~\text{M}_{\odot}$. This work makes use of deep, high-resolution imaging in the H$\alpha$+[NII] narrow-band from the Virgo Environmental Survey Tracing Ionised Gas Emission (VESTIGE) and optical imaging from the Next Generation Virgo Survey (NGVS). To aid in understanding the effects of the cluster environment on star formation in Virgo galaxies, we take a physically-motivated approach to define the edge of the star-forming disk via a drop-off in the radial specific star formation rate profile. Comparing with the expected sizes of normal galactic disks provides a measure of how truncated star-forming disks are in the cluster. We find that truncated star-forming disks are nearly ubiquitous across all regions of the Virgo cluster, including beyond the virial radius (0.974 Mpc). The majority of truncated disks at large clustercentric radii are of galaxies likely on first infall. As the intra-cluster medium density is low in this region, it is difficult to explain this population with solely ram-pressure stripping. A plausible explanation is that these galaxies are undergoing starvation of their gas supply before ram-pressure stripping becomes the dominant quenching mechanism. A simple model of starvation shows that this mechanism can produce moderate disk truncations within 1-2 Gyr. This model is consistent with `slow-then-rapid' or `delayed-then-rapid' quenching, where the early starvation mode drives disk truncations without significant change to the integrated star formation rate, and the later ram-pressure stripping mode rapidly quenches the galaxy. Abstract

We present an analysis of Euclid observations of a 0.5 deg$^2$ field in the central region of the Fornax galaxy cluster that were acquired during the performance verification phase. With these data, we investigate the potential of Euclid for identifying GCs at 20 Mpc, and validate the search methods using artificial GCs and known GCs within the field from the literature. Our analysis of artificial GCs injected into the data shows that Euclid's data in $I_{\rm E}$ band is 80% complete at about $I_{\rm E} \sim 26.0$ mag ($M_{V\rm } \sim -5.0$ mag), and resolves GCs as small as $r_{\rm h} = 2.5$ pc. In the $I_{\rm E}$ band, we detect more than 95% of the known GCs from previous spectroscopic surveys and GC candidates of the ACS Fornax Cluster Survey, of which more than 80% are resolved. We identify more than 5000 new GC candidates within the field of view down to $I_{\rm E}$ mag, about 1.5 mag fainter than the typical GC luminosity function turn-over magnitude, and investigate their spatial distribution within the intracluster field. We then focus on the GC candidates around dwarf galaxies and investigate their numbers, stacked luminosity distribution and stacked radial distribution. While the overall GC properties are consistent with those in the literature, an interesting over-representation of relatively bright candidates is found within a small number of relatively GC-rich dwarf galaxies. Our work confirms the capabilities of Euclid data in detecting GCs and separating them from foreground and background contaminants at a distance of 20 Mpc, particularly for low-GC count systems such as dwarf galaxies., Comment: Paper submitted as part of the A&A special issue `Euclid on Sky', which contains Euclid key reference papers and first results from the Euclid Early Release Observations

Abstract

The current standard model of cosmology successfully describes a variety of measurements, but the nature of its main ingredients, dark matter and dark energy, remains unknown. Euclid is a medium-class mission in the Cosmic Vision 2015-2025 programme of the European Space Agency (ESA) that will provide high-resolution optical imaging, as well as near-infrared imaging and spectroscopy, over about 14,000 deg^2 of extragalactic sky. In addition to accurate weak lensing and clustering measurements that probe structure formation over half of the age of the Universe, its primary probes for cosmology, these exquisite data will enable a wide range of science. This paper provides a high-level overview of the mission, summarising the survey characteristics, the various data-processing steps, and data products. Abstract

We use a complete set of deep narrow-band imaging data for 384 galaxies gathered during the VESTIGE survey to derive the first Halpha luminosity function (LF) of the Virgo cluster within R200. The data allow us to cover the whole dynamic range of the Halpha LF (10^3610^14 Mo. The comparison with the IllustrisTNG cosmological hydrodynamical simulations shows a more pronounced decrease at the faint end of the distribution. If Virgo is representative of typical nearby clusters of similar mass, this difference suggests that the stripping process in simulated galaxies in these environments is more efficient than observed., Comment: Accepted for publication on A&A

Abstract

Presently, gamma-ray tracking in germanium segmented detectors is realised by applying two advanced, complex algorithms. While they have already triggered an intensive R&D, they are still subject to further improvements. Making such algorithms effective, online in real time conditions and/or offline for deeper analysis, in data pipelines do require many additional software developments. This review paper gives an overview of the various bricks of software produced so far by the AGATA collaboration. It provides hints of what is foreseen for the next phases of the project up to its full configuration namely with 180 capsules in the array.

Using a compilation of Halpha fluxes for 384 star forming galaxies detected during the VESTIGE survey, we study several important scaling relations for a complete sample of galaxies in a rich environment. Abstract

The Advanced GAmma Tracking Array (AGATA) has been installed at Laboratori Nazionali di Legnaro (LNL), Italy. In this installation, AGATA will consist, at the beginning, of 13 AGATA triple clusters (ATCs) with an angular coverage of 1n,and progressively the number of ATCs will increase up to a 2 pi angular coverage. This setup will exploit both stable and radioactive ion beams delivered by the Tandem-PIAVE-ALPI accelerator complex and the SPES facility. The new implementation of AGATA at LNL will be used in two different configurations, firstly one coupled to the PRISMA large-acceptance magnetic spectrometer and lately a second one at Zero Degrees, along the beam line. These two configurations will allow us to cover a broad physics program, using different reaction mechanisms, such as Coulomb excitation, fusion-evaporation, transfer and fission at energies close to the Coulomb barrier. These setups have been designed to be coupled with a large variety of complementary detectors such as charged particle detectors, neutron detectors, heavy-ion detectors, high-energy gamma-ray arrays, cryogenic and gasjet targets and the plunger device for lifetime measurements. We present in this paper the conceptual design, characteristics and performance figures of this implementation of AGATA at LNL.

Abstract

International audience; Presently, gamma-ray tracking in germanium segmented detectors is realised by applying two advanced, complex algorithms. While they have already triggered an intensive R&D, they are still subject to further improvements. Making such algorithms effective, online in real time conditions and/or offline for deeper analysis, in data pipelines do require many additional software developments. This review paper gives an overview of the various bricks of software produced so far by the AGATA collaboration. It provides hints of what is foreseen for the next phases of the project up to its full configuration namely with 180 capsules in the array.

20. MMP9: Link between neuropathy and colorectal cancer?

Abstract

As chemotherapy is still a cornerstone of colorectal cancer (CRC) treatment, chemotherapy-induced peripheral neuropathy (CIPN) presents significant clinical challenges, affecting millions worldwide. A subset of colon cancer patients (approximately 30%) develop chronic CIPN, with detrimental, untreatable neuropathic pain symptoms. The risk factors of such intractable chronic CIPN are unknown. However, there is growing literature data investigating the intriguing interplay of neurons and cancer (cancer neuroscience). Recent data shows that this interplay might have a key role in the development and severity of CIPN. Given its vast (patho)physiological roles in both colon cancer and neuropathy, MMP9 seems to be a key factor that might drive the development of neuronal damage in colon cancer patients. This review investigates the role of matrix metalloproteinase 9 (MMP9) in linking CRC to neuropathy, aiming to uncover shared mechanisms that could offer new therapeutic targets. By synthesizing insights from a broad range of studies published over the last 20 years, we explore MMP9's involvement in CRC progression, its role in CIPN, and the interconnected pathways influencing both conditions. These studies reveal MMP9 as a pivotal mediator in ECM remodeling, inflammation, and signal transduction pathways, emphasizing its modulation by macrophages. These shared mechanisms of colon cancer and CIPN pathophysiology suggest MMP9's potential contribution to neuropathic conditions in CRC patients, positioning it as a critical factor in disease progression and a promising therapeutic target. Future research should focus on longitudinal studies to assess MMP9's impact on neuropathy outcomes in CRC patients, exploring MMP9 inhibitors, and developing targeted interventions to mitigate the detrimental symptoms of CIPN. 21. Gap junctions fine-tune ganglion cell signals to equalize response kinetics within a given electrically coupled array.

Abstract

Retinal ganglion cells (RGCs) summate inputs and forward a spike train code to the brain in the form of either maintained spiking (sustained) or a quickly decaying brief spike burst (transient). We report diverse response transience values across the RGC population and, contrary to the conventional transient/sustained scheme, responses with intermediary characteristics are the most abundant. Pharmacological tests showed that besides GABAergic inhibition, gap junction (GJ)-mediated excitation also plays a pivotal role in shaping response transience and thus visual coding. More precisely GJs connecting RGCs to nearby amacrine and RGCs play a defining role in the process. These GJs equalize kinetic features, including the response transience of transient OFF alpha (tOFFα) RGCs across a coupled array. 22. Gymnadenia winkeliana -A New Orchid Species to Romanian Flora.

Abstract

A novel species, Gymnadenia winkeliana , has been identified in the Bucegi Natural Park ROSCI0013, located in the Southern Carpathians of Central Romania. Two moderately sized populations of Gymnadenia winkeliana , totalling 120-140 individuals, were discovered inhabiting the alpine grasslands of the park, situated 2.000 m above sea level. To describe this newly found population as comprehensively as possible, 44 vegetative and floral organs/organ parts were directly studied and measured from living plants. Special attention was focused on the characteristics that proved to have taxonomic significance, particularly those involving distinctive details in the morphology of the leaves, perianth, labellum and gynostemium. A total of 223 characteristics were analysed encompassing the morphology of every organ of the plant, cytology and breeding system. Furthermore, comprehensive taxonomic treatment and description, accompanied by colour photographs illustrating the holotype, are provided. Voucher specimens were deposited at the Herbarium of the University of Agriculture and Veterinary Medicine, Bucharest (USAMVB Herbarium barcode: 40102, NEA); Gymnadenia winkeliana , a (micro)endemic species, is characterized as a putative allogamous, facultatively apomict that significantly differs from other Gymnadenia R.Br. species found in Romania. Notably, it distinguishes itself through its smaller habitus (reaching heights of up to 8-10 cm), its two-coloured, rounded/hemispherical inflorescence displaying a gradient of pink hues in an acropetal fashion (ranging from whitish-pink at the base to vivid-pink at the topmost flowers), and its limited distribution in high-altitude areas, encompassing approximately 8-10 km 2 in the central area of the Bucegi Natural Park. This species has been under observation since 2005, with observed population numbers showing a significant increase over time, from ca. 50-55 (counted at the time of its discovery) to 120-140 individuals (counted in June 2023). Additionally, comprehensive information regarding the habitat, ecology, phenology and IUCN conservation assessments of Gymnadenia winkeliana are provided, including maps illustrating its distribution.

23. Enhancing HLA-B27 antigen detection: Leveraging machine learning algorithms for flow cytometric analysis.

Abstract

As the association of human leukocyte antigen B27 (HLA-B27) with spondylarthropathies is widely known, HLA-B27 antigen expression is frequently identified using flow cytometric or other techniques. Because of the possibility of cross-reaction with off target antigens, such as HLA-B7, each flow cytometric technique applies a "gray zone" reserved for equivocal findings. Our aim was to use machine learning (ML) methods to classify such equivocal data as positive or negative. Equivocal samples (n = 99) were selected from samples submitted to our institution for clinical evaluation by HLA-B27 antigen testing. Samples were analyzed by flow cytometry and polymerase chain reaction. Features of histograms generated by flow cytometry were used to train and validate ML methods for classification as logistic regression (LR), decision tree (DT), random forest (RF) and light gradient boost method (GBM). All evaluated ML algorithms performed well, with high accuracy, sensitivity, specificity, as well as negative and positive predictive values. Although, gradient boost approaches are proposed as high performance methods; nevertheless, their effectiveness may be lower for smaller sample sizes. On our relatively smaller sample set, the random forest algorithm performed best (AUC: 0.92), but there was no statistically significant difference between the ML algorithms used. AUC values for light GBM, DT, and LR were 0.88, 0.89, 0.89, respectively. Implementing these algorithms into the process of HLA-B27 testing can reduce the number of uncertain, false negative or false positive cases, especially in laboratories where no genetic testing is available., (© 2024 International Clinical Cytometry Society.)

Abstract

Organoids generated from human pluripotent stem cells provide experimental systems to study development and disease, but quantitative measurements across different spatial scales and molecular modalities are lacking. In this study, we generated multiplexed protein maps over a retinal organoid time course and primary adult human retinal tissue. We developed a toolkit to visualize progenitor and neuron location, the spatial arrangements of extracellular and subcellular components and global patterning in each organoid and primary tissue. In addition, we generated a single-cell transcriptome and chromatin accessibility timecourse dataset and inferred a gene regulatory network underlying organoid development. We integrated genomic data with spatially segmented nuclei into a multimodal atlas to explore organoid patterning and retinal ganglion cell (RGC) spatial neighborhoods, highlighting pathways involved in RGC cell death and showing that mosaic genetic perturbations in retinal organoids provide insight into cell fate regulation., (© 2023. Particle number concentrations and size distributions resulting from the firework displays held in Budapest, Hungary every year on St. Stephen's Day were studied over a period of seven years. In the year most impacted, the total particle number concentration reached its peak measured level of 369 × 10 3  cm -3 5 min after the end of the display, and returned to the pre-event state within 45 min. The fireworks increased hourly mean concentrations by a factor of 5-6, whereas the concentrations in the diameter range of 100-1000 nm, in which the magnitude of the increase was the greatest, were elevated by a factor of 20-25. An extra particle size mode at 203 nm was manifested in the size distributions as result of the fireworks. The PM 10 mass concentrations at peak firework influence and as 1-h mean increased 123 and 58 times, respectively, relative to the concentration before the display. The smoke was characterised by a relatively short overall atmospheric residence time of 25 min. Spatiotemporal dispersion simulations revealed that there were substantial vertical and horizontal concentration gradients in the firework plume. The affected area made up a large part of the city. Not only the spectators of the display at the venue and nearby areas, but the population located further away downwind of the displays and more distant, large and populous urban quarters were affected by the plume and its fallout. The fireworks increased the deposition rate in the respiratory system of females by a factor of 4, as a conservative estimate. The largest surface density deposition rates were seen in the segmental and sub-segmental bronchi, which represents an excessive risk to health. Abstract

Despite the large arsenal of analgesic medications, neuropathic pain (NP) management is not solved yet. Angiotensin II receptor type 1 (AT1) has been identified as a potential target in NP therapy. Here, we investigate the antiallodynic effect of AT1 blockers telmisartan and losartan, and particularly their combination with morphine on rat mononeuropathic pain following acute or chronic oral administration. The impact of telmisartan on morphine analgesic tolerance was also assessed using the rat tail-flick assay. Morphine potency and efficacy in spinal cord samples of treated neuropathic animals were assessed by [ 35 S]GTPγS-binding assay. Finally, the glutamate content of the cerebrospinal fluid (CSF) was measured by capillary electrophoresis. Oral telmisartan or losartan in higher doses showed an acute antiallodynic effect. In the chronic treatment study, the combination of subanalgesic doses of telmisartan and morphine ameliorated allodynia and resulted in a leftward shift in the dose-response curve of morphine in the [ 35 S]GTPγS binding assay and increased CSF glutamate content. Telmisartan delayed morphine analgesic-tolerance development. Our study has identified a promising combination therapy composed of telmisartan and morphine for NP and opioid tolerance. Since telmisartan is an inhibitor of AT1 and activator of PPAR-γ, future studies are needed to analyze the effect of each component.

28. Epipactis bucegensis -A Separate Autogamous Species within the E. helleborine Alliance.

Abstract

A new species of Epipactis from Bucegi Natural Park ROSCI0013, Southern Carpathians, Central Romania is described. Three medium-sized populations of Epipactis bucegensis (65-70 individuals in total) were discovered in the south-eastern, subalpine area of the park. To properly describe and distinguish the newly found taxon from other Romanian Epipactis , 37 morphological characters were measured directly from living plants and flowers. Moreover, a detailed taxonomic treatment and description with corresponding colour photos and line drawings illustrations of the holotype are also included. Epipactis bucegensis is an obligate autogamous species that partially resembles Epipactis muelleri, from which it differs in the basal distribution of leaves on the stem (vs. median distribution); near-erect leaf posture (vs. horizontally spread, arched downwards); lanceolate-acuminate, yellowish-green leaves (vs. oval-elongate, vivid-green leaves); bipartite labellum lacking the mesochile (vs. tripartite labellum); crimson-red, wide, ovoid-elongated, flattened hypochile (vs. dark-brown to black roundish hypochile); triangular, white epichile with a sharply tapering apex (vs. heart-shaped, greenish-yellow epichile with obtuse, roundish apex); and two wide-apart, purple, pyramidal calli (vs. two closely placed, attenuated, mildly wrinkled, greenish-yellow calli). Epipactis bucegensis is easily distinguished from all other European Epipactis taxa by the bipartite, wide labellum that lacks the mesochile. In addition, information regarding its distribution (maps), habitat, ecology, phenology and IUCN conservation assessments are provided.

Key Message: The bs5 resistance gene against bacterial spot was identified by map-based cloning. The recessive bs5 gene of pepper (Capsicum annuum L.) conditions a non-hypersensitive resistance trait, characterized by a slightly swollen, pale green, photosynthetically active leaf tissue, following Xanthomonas euvesicatoria infection. The isolation of the bs5 gene by map-based cloning revealed that the bs5 protein was shorter by 2 amino acids as compared to the wild type Bs5 protein. The natural 2 amino acid deletion occurred in the cysteine-rich transmembrane domain of the tail-anchored (TA) protein, Ca_CYSTM1. The protein products of the wild type Bs5 and mutant bs5 genes were shown to be located in the cell membrane, indicating an unknown function in this membrane compartment. Successful infection of the Bs5 pepper lines was abolished by the 6 bp deletion in the TM encoding domain of the Ca_CYSTM1 gene in bs5 homozygotes, suggesting, that the resulting resistance might be explained by the lack of entry of the Xanthomonas specific effector molecules into the plant cells., (© 2023. 30. Open photoacoustic cell for concentration measurements in rapidly flowing gas.

Abstract

High temporal resolution concentration measurements in rapid gas flows pose a serious challenge for most analytical instruments. The interaction of such flows with solid surfaces can generate excessive aero-acoustic noise making the application of the photoacoustic detection method seemingly impossible. Yet, the fully open photoacoustic cell (OC) has proven to be operable even when the measured gas flows through it at a velocity of several m/s. The OC is a slightly modified version of a previously introduced OC based on the excitation of a combined acoustic mode of a cylindrical resonator. The noise characteristics and analytical performance of the OC are tested in an anechoic room and under field conditions. 32. Remote Teaching of Canine Cardiac Auscultation Using Digital Phonocardiograms.

Abstract

Veterinary students often struggle to correctly interpret heart sounds. This study sought to evaluate if additional online training using digital phonocardiograms (DPCGs) improves students' ability to identify normal and pathologic heart sounds in dogs. Thirty-six randomly assigned veterinary students listened to and interpreted 10 audio recordings of normal heart sounds (2), heart murmurs (4), and arrhythmias (4) at the start and the end of a 4-week period. Twenty-two students participated in training with DPCGs, including those created from these recordings during this period, via a self-study website ( n = 12) or online webinar ( n = 10). Their results were compared with those of a control group ( n = 14) that did not undergo additional training. Although pre- and post-training test scores did not differ between groups, both training groups showed within-group improvement between the two tests ( p = .024, p = .037); the control group did not ( p = .49). Although neither training group showed differences in ability to differentiate normal heart sounds from arrhythmias, both showed increased ability to detect and specify heart murmurs and provide refined diagnoses of detected arrhythmias. These results suggest additional training, even without actual patients, improves students' ability to identify heart murmurs and provide specific diagnoses for arrhythmias. Further study with a larger sample size and an additional group without DPCG-based training would help evaluate the effectiveness of DPCGs regarding arrhythmias. Studying a larger sample size would also allow for a training group participating in both training methods, measuring cumulative effectiveness of both methods.

Abstract

Reports of neurological sequelae related to colon cancer are largely restricted to rare instances of paraneoplastic syndromes, due to autoimmune reactions. Systemic inflammation associated with tumor development influences sensory neuron function in other disease models, though the extent to which this occurs in colorectal cancer is unknown. We induced orthotopic colorectal cancer via orthotopic injection of two colorectal cancer cell lines (MC38 and CT26) in two different mouse strains (C57BL/6 and Balb/c, respectively). Behavioral tests of pain sensitivity and activity did not detect significant alterations in sensory sensitivity or diminished well-being throughout tumor development. However, immunohistochemistry revealed widespread reductions in intraepidermal nerve fiber density in the skin of tumor-bearing mice. Though loss of nerve fiber density was not associated with increased expression of cell injury markers in dorsal root ganglia, lumbar dorsal root ganglia neurons of tumor-bearing animals showed deficits in mitochondrial function. These neurons also had reduced cytosolic calcium levels in live-cell imaging and reduced spontaneous activity in multi-electrode array analysis. Bulk RNA sequencing of DRGs from tumor-bearing mice detected activation of gene expression pathways associated with elevated cytokine and chemokine signaling, including CXCL10. This is consistent with the detection of CXCL10 (and numerous other cytokines, chemokines and growth factors) in MC38 and CT26 cell-conditioned media, and the serum of tumor-bearing mice. Our study demonstrates in a pre-clinical setting that colon cancer is associated with latent sensory neuron dysfunction and implicates cytokine/chemokine signaling in this process.
