Abstract

Abstract Psychological distress, including anxiety or mood disorders, emanates from the onset of chronic/unpredictable stressful events. Symptoms in the form of maladaptive behaviors are learned and difficult to treat. While the origin of stress-induced disorders seems to be where learning and stress intersect, this relationship and molecular pathways involved remain largely unresolved. The hippocampus, studied for its role in learning, is divided into regions that designate the passage of neuronal signaling during memory formation, including dentate gyrus (DG), CA3, CA2, and CA1. Inputs into these hippocampal subregions, like those from hypothalamic orexinergic neurons, may modify learning outcomes. We have previously shown the orexin system to balance stress states, where receptor subtypes prompt opposing actions on behavior. Here, we explore the connection between hippocampal orexin receptors and learning during stress. In a social stress/learning paradigm separating mice into stress resilient and vulnerable populations, hippocampal Orx1R and Orx2R transcription is regulated in a phenotype-dependent fashion. We further identified Orx1R as highly expressed in the hilus of DG, while Orx2R is abundant in CA2. Finally, we designed an experiment where mice were provided prior exposure to a stressful environment, which ultimately modified behavior, as well as transcription of hippocampal orexin receptors.

3. Presynaptic depolarization differentially regulates dual neurotransmitter release from starburst amacrine cells in the mouse retina

Abstract

The retina is comprised of diverse neural networks, signaling from photoreceptors to ganglion cells to encode images. The synaptic connections between these retinal neurons are crucial points for information transfer; however, the input-output relations of many synapses are understudied. Starburst amacrine cells in the retina are known to contribute to retinal motion detection circuits, providing a unique window for understanding neural computations. We examined the dual transmitter release of GABA and acetylcholine from starburst amacrine cells by optogenetic activation of these cells, and conducted patch clamp recordings from postsynaptic ganglion cells to record excitatory and inhibitory postsynaptic currents (EPSCs and IPSCs). As starburst amacrine cells exhibit distinct kinetics in response to objects moving in a preferred or null direction, we mimicked their depolarization kinetics using optogenetic stimuli by varying slopes of the rising phase. The amplitudes of EPSCs and IPSCs in postsynaptic ganglion cells were reduced as the stimulus rising speed was prolonged. However, the sensitivity of postsynaptic currents to the stimulus slope differed. EPSC amplitudes were consistently reduced as the steepness of the rising phase fell. By contrast, IPSCs were less sensitive to the slope of the stimulus rise phase and maintained their amplitudes until the slope became shallow. These results indicate that distinct synaptic release mechanisms contribute to acetylcholine and GABA release from starburst amacrine cells, which could contribute to the ganglion cells’ direction selectivity.

Abstract

Metal polymer composites combining low density, high strength composites with highly ductile and tough metals have gained traction over the last few decades as lightweight and high-performance materials for industrial applications. However, the mechanical properties are limited by the interfacial bonding strength between metals and polymers achieved through adhesives, welding, and surface treatment processes. In this paper, a novel manufacturing process combining additive manufacturing and compression molding to obtain hybrid metal polymer composites with enhanced mechanical properties is presented. Additive manufacturing enabled deposition of polymeric material with fibers in a predetermined pattern to form tailored charge or preform for compression molding. A grade 300 maraging steel triangular lattice is first fabricated using AddUp FormUp350 laser powder bed system and compression overmolded with additively manufactured long carbon fiber-reinforced polyamide-6,6 (40% wt. CF/PA66) preform. The fabricated hybrid metal polymer composites showed high stiffness and tensile strength. The stiffness and failure characteristics determined from the uniaxial tensile tests were correlated to a finite element model within 20% deviation. Fractographic analyses was performed using microscopy to investigate failure mechanisms of the hybrid structures.

Abstract

Varroa destructor is a major threat for apiculture worldwide. A successful approach to control this parasite must include the application of effective treatments at the correct time. To understand the effect that treatment timing has on Varroa populations at different seasons, we conducted an experiment using a dataset comprising two separate field trials over multiple years, both trials containing four apiary sites composed of 20 honey bee colonies across an area representative of north central Florida environments. Before the start of the season, colonies were treated with two acaricides simultaneously to bring the Varroa populations to ∼0.25 mites/100 bees. Following treatment, we monitored the mite populations monthly via alcohol washes. Our results show that the temporal efficacy of Varroa treatments varies across seasons. We observed that it takes about 4–5 months after treatment in winter and spring for mite populations to return to the standard economical threshold (3 mites/100 bees). An analog crystal filter based cavity emulator is modified with reverse biased varactor diodes to provide a tuning range of around 160 Hz. The piezo drive voltage of the resonance controller is used to detune the cavity through the bias voltage. A signal conditioning and summing circuit allows the introduction of microphonics disturbance from a signal source or using real microphonics data from cavity testing. 7. LLRF System for the Fermilab PIP-II Superconducting LINAC

Abstract

PIP-II is an 800 MEV superconducting linac that is in the initial acceleration chain for the Fermilab accelerator complex. The RF system consists of a warm front-end with an ion source, RFQ and buncher cavities along with 25 superconducting cryo-modules comprised of five different acceleration \(\beta\). The LLRF system for the LINAC has to provide field and resonance control for a total of 125 RF cavities.The LLRF system design is in the final design review phase and will enter the production phase next year. The PIP-II project is an international collaboration with various partner labs contributing subsystems. Abstract

Microphonics causes the cavity to detune. This study discusses the microphonics of sixteen 1.3 GHz cryomodules, 14 for LCLS-II and 2 for LCLS-II HE tested at CMTF. The peak detuning, as well as the RMS detuning for each cryomodule, will be discussed. For each cryomodule, the data was taken with enough soaking time to prevent any thermalization effects which can show up in the detuning. Each data capture taken was 30 minutes or longer and sampled at 1 kHz.

Abstract

An eight-cavity, 1.3 GHz, SLAC linac coherent light source II High Energy cryomodule was assembled and tested at Fermilab to verify performance before the start of production. Its cavities were processed with a novel nitrogen doping treatment to improve gradient performance. The cryomodule was tested with a modified protocol to process sporadic quenches, which were observed in Linac Coherent Light Source II production cryomodules and are attributed to multipacting. Dedicated vertical test experiments support the attribution to multipacting. The verification cryomodule achieved an acceleration voltage of 200 MV in continuous wave mode, corresponding to an average accelerating gradient of 24.1 MV/m, significantly exceeding the specification of 173 MV. The average Q0 (3.0×1010) also exceeded its specification (2.7×1010). After processing, no field emission was observed up to the maximum gradient of each cavity. This paper reviews the cryomodule performance and discusses operational issues and mitigations implemented during the several month program.

Abstract

The thermal hysteresis exhibited in plastic crystal compounds greatly reduces their cyclic efficiency, limiting their potential for replacing current environmentally harmful refrigerants. A mechanistic understanding of the origins of this hysteresis has yet to be established. Here, we systematically investigate the transformation kinetics of the model plastic crystal, neopentyl glycol (NPG), through microscopic and calorimetric techniques. We reveal an asymmetry between the forward (heating) and reverse (cooling) transitions. We also demonstrate that the forward transformation is rate-limited by the rate of growth of rotationally disordered domains. In contrast, the reverse transformation is rate-limited by the nucleation of the ordered crystal domain, demonstrated by the sharp exothermic peaks in calorimetry and rapid self-nucleation phenomena observed optically. This nucleation limitation is largely responsible for the large thermal hysteresis in NPG, which we observe to be as large as 16.7 °C for an approximately 10 mg sample cooled at 0.5 °C min−1. These findings demonstrate the underlying origin of the thermal hysteresis and introduce a direction to mitigate hysteresis in plastic crystal transformations. [ABSTRACT FROM AUTHOR]

Abstract

Maximum dose bioassays were conducted to assess the efficacy of multiple registered active ingredients for diamondback moth (DBM), Plutella xylostella (L.), control in Georgia and Florida from 2021 to 2023 as a follow-up to an earlier study. Low efficacy (<40% mortality) was recorded for the highest labeled rate of Bacillus thuringiensis var. kurstaki strain ATBS-351 in Georgia, as well as chlorantraniliprole, cyantraniliprole, and cyclaniliprole in Georgia and Florida. The active ingredients with the highest efficacy (>80% mortality) in both states were naled, emamectin benzoate, and spinetoram. Independent analysis of data by state indicated that the efficacy of bifenthrin, chlorantraniliprole, cyantraniliprole, tolfenpyrad, and methomyl was higher in Florida populations than in Georgia populations. In addition, a comparison of these data to a recent DBM maximum dose survey in the same region suggested that these DBM populations have rapidly developed high levels of resistance to cyantraniliprole and cyclaniliprole. This work provides growers in the region with a recent ranking of insecticide efficacy that documents the loss of control for certain active ingredients, which assists pest managers in the planning of ongoing insecticide rotations for DBM resistance management. Abstract

Gerontology, the study of aging and older adulthood, is a vast field consisting of many intersecting disciplines, such as medicine, nutrition, public policy, psychology, social work, and professional counseling. As the older adult population rapidly increases, so too will the prevalence of mental health concerns among this population, including risk for suicide. Older adults presently comprise 16.5% of the U.S. population, yet account for 19.3% of all deaths by suicide (Drapeau & McIntosh, 2020). Professional counselors are newly eligible Medicare providers and can close gaps in healthcare access for older adults who rely on Medicare to receive mental health services, which might include suicide risk assessment and intervention. The Gerontological Imagination is an integrative paradigm to understand the process of aging across multiple disciplines. In this manuscript, we use The Gerontological Imagination as a guiding framework to discuss a professional counselor’s role in identifying and responding to suicide among older adults. Abstract

Electrical transmission rights-of-way are ubiquitous and critical infrastructure across the landscape. Active vegetation management of these rights-of-way, a necessity to deliver electricity more safely, maintains these landscape features as stages of early successional habitat, a rarity in many regions, making these areas viable movement corridors for many taxa. The goals of this study were to (i) evaluate the effects of different electrical transmission landscape management practices on flowering plant and flower-visiting insect diversity parameters and (ii) generate conservation management inferences for these landscapes. In this study we tested the impact of three vegetation management levels across 18 electrical transmission sites. We evaluated the effects of treatment on bloom abundance and species richness as well as flower-visiting insect abundance and family richness. We identified 76541 flowers/inflorescences across 456 transects, including 188 species in 56 plant families. Additionally, we obtained data on 11361 flower-visitoring insects representing 33 families from 2376 pan trap sets. High vegetation management favored the reduction of coarse woody debris in the sites and harbored the highest level of abundance and richness of both floral resources and flower-visiting insects. We discuss that we can align social and ecological values of rights-of-way, ensuring their sustainability by applying regular and targeted integrated vegetation management. Thus, we can use rights-of-way landscapes not only as an effective management strategy for the delivery of essential human services, but also to provide conservation benefits for wild pollinators. Abstract

Background: As gene-targeted therapies are increasingly being developed for Parkinson's disease (PD), identifying and characterizing carriers of specific genetic pathogenic variants is imperative. Only a small fraction of the estimated number of subjects with monogenic PD worldwide are currently represented in the literature and availability of clinical data and clinical trial-ready cohorts is limited. Objective: The objectives are to (1) establish an international cohort of affected and unaffected individuals with PD-linked variants; (2) provide harmonized and quality-controlled clinical characterization data for each included individual; and (3) further promote collaboration of researchers in the field of monogenic PD. Methods: We conducted a worldwide, systematic online survey to collect individual-level data on individuals with PD-linked variants in SNCA, LRRK2, VPS35, PRKN, PINK1, DJ-1, as well as selected pathogenic and risk variants in GBA and corresponding demographic, clinical, and genetic data. All registered cases underwent thorough quality checks, and pathogenicity scoring of the variants and genotype–phenotype relationships were analyzed. Results: We collected 3888 variant carriers for our analyses, reported by 92 centers (42 countries) worldwide. Of the included individuals, 3185 had a diagnosis of PD (ie, 1306 LRRK2, 115 SNCA, 23 VPS35, 429 PRKN, 75 PINK1, 13 DJ-1, and 1224 GBA) and 703 were unaffected (ie, 328 LRRK2, 32 SNCA, 3 VPS35, 1 PRKN, 1 PINK1, and 338 GBA). In total, we identified 269 different pathogenic variants; 1322 individuals in our cohort (34%) were indicated as not previously published. Abstract

The Manitoba Geological Survey, in collaboration with the University of Alberta, is developing a model of the lithospheric structure beneath the province of Manitoba. Information about deep lithospheric structure cannot be obtained from drilling and requires deep sounding geophysical exploration. One of the most useful methods for this task is magnetotelluric exploration, which uses natural low-frequency electromagnetic signals to determine the electrical resistivity structure of the Earth to depths in excess of 200 km. A long-period magnetotelluric survey was conducted at 22 stations in June and July of 2024 in west-central Manitoba. The new data will be combined with long-period magnetotelluric data previously collected in Saskatchewan to produce a 3-D resistivity model of the lithosphere of the Trans-Hudson orogen beneath west-central Manitoba and central Saskatchewan. This 3-D resistivity model will improve the understanding of the factors that control the spatial distribution of mineralization and may also guide future exploration in the region. Abstract

Supramolecular polymers allow access to dynamic materials, where noncovalent interactions can be used to offer both enhanced material toughness and stimuli-responsiveness. The versatility of self-assembly has enabled these supramolecular motifs to be incorporated into a wide array of glassy and elastomeric materials; moreover, the interaction of these noncovalent motifs with their environment has shown to be a convenient platform for controlling material properties. In this Viewpoint, supramolecular polymers are examined through their self-assembly chemistries, approaches that can be used to control their self-assembly (e.g., covalent cross-links, nanofillers, etc.), and how the strategic application of supramolecular polymers can be used as a platform for designing the next generation of smart materials. This Viewpoint provides an overview of the aspects that have garnered interest in supramolecular polymer chemistry, while also highlighting challenges faced and innovations developed by researchers in the field.

43. Off Starburst Amacrine Cells in the Retina Trigger Looming-Evoked Fear Responses in Mice

Abstract

A rapidly approaching dark object evokes an evolutionarily conserved fear response in both vertebrates and invertebrates, young to old. A looming visual stimulus mimics an approaching object and triggers a similarly robust fear response in mice, resulting in freeze and flight. However, the retinal neural pathway responsible for this innate response has not been fully understood. We first explored a variety of visual stimuli that reliably induced these innate responses, and found that a looming stimulus with 2-d acclimation consistently evoked fear responses. Because the fear responses were triggered by the looming stimulus with moving edges, but not by a screen flipping from light to dark, we targeted the starburst amacrine cells (SACs), crucial neurons for retinal motion detection. We used intraocular injection of diphtheria toxin (DT) in mutant mice expressing diphtheria toxin receptors (DTR) in SACs. The looming-evoked fear responses disappeared in half of the DT-injected mice, and the other mice still exhibited the fear responses. The optomotor responses (OMRs) were reduced or eliminated, which occurred independent of the disappearance of the fear responses. A histologic examination revealed that ON SACs were reduced in both mouse groups preserved or absent fear responses. In contrast, the number of OFF SACs was different among two groups. The OFF SACs were relatively preserved in mice exhibiting continued fear responses, whereas they were ablated in mice lacking fear response to looming stimulation. These results indicate that OFF SACs and the direction-selective pathway in the retina play a role in looming-induced fear behaviors.

45. The True and the Good : A Strong Virtue Theory of the Value of Truth

Abstract

Philosophical thinking about truth has revolved around two key questions. First, what is the nature of truth? Second, why should we value it? It has proven difficult to answer both questions at once. Some theories analyse truth in terms of goodness. Truth is a specific kind of goodness for beliefs. They have an easy explanation of truth's value, but they obscure the connection between truth and how things are. What does belief have to do with whether it's true that there is water on Mars? Other theories treat truth as a descriptive feature: a claim is true when things are as it says they are. Such theories face a version of G. E. Moore's open question problem. How could a claim's purely descriptive features make it good or right to believe? The problem of truth's value is the problem of reconciling a good theory of what truth is with a good theory of why truth is valuable. This is the first monograph focused on solving it for theories that construe truth in the purely descriptive way. Drawing on minimal assumptions about truth's nature, it proposes a virtue-theoretic understanding of truth's value. The attitude of valuing truth is a morally laudable character trait. That is, it's a virtue. But its virtuousness doesn't stem from the nature of truth. It arises because, in manifesting the trait, we promote the overall good. A good society needs to members who value truth over falsehood and ignorance, for themselves and for others.

Bipolar cells are the second-order neurons in the retina that are less accessible for investigating their synaptic responses. Here, we present a protocol to conduct patch clamp recordings from bipolar cells in the wholemount retina from Ai32 mutant mice. We detail whole-cell patch-clamp recording from bipolar cells to examine their light-evoked responses to optogenetic stimulation, followed by imaging terminals of recorded cells to determine bipolar cell type. We describe light stimulus information to activate channelrhodopsin-2 (ChR2). For complete details on the use and execution of this protocol, please refer to Hellmer et al. (2021).

Abstract

Previous literature suggests that Democratic candidates of color receive less support than white Democrats on Election Day. It has been suggested that this reduced support is due to voters' ideological perceptions of people of color being more liberal than whites, which translates into perceptions of candidates. However, these studies are frequently limited to white voters' perceptions of Democratic candidates of color, and the full relationship linking race/ethnicity, to ideological placement, to vote choice has rarely been tested. In the current study, we examine how both the race/ethnicity and party affiliation of Senate candidates shape perceptions of their ideology and vote choice among people of all race and ethnicities. Using the 2006–2018 Cooperative Congressional Election Study, we find support for our hypothesis that Black Democrats are generally perceived as more liberal than white Democrats. These perceptions have real consequences, resulting in fewer votes. Conversely, Black Republican candidates are perceived to be just as conservative—and Hispanic Republicans even more conservative—than white Republican candidates. Findings suggest that the link between race/ethnicity, perceptions of ideology, and electoral success vary according to the race/ethnicity and party of candidates. [ABSTRACT FROM AUTHOR]

50. Getting "Primaried" in the Senate: Primary Challengers and the Roll-Call Voting Behavior of Sitting senators.

Abstract

Primary challenges are semi-regular occurrences for incumbent members of Congress, and while many of these primary challengers never pose a serious threat to the incumbent, there are occasions when the incumbent faces a difficult primary challenge. How do incumbent members of Congress, particularly members of the Senate, act in response to primary challengers? This study examines the impact a primary challenger has on the voting behavior of incumbent senators to determine if incumbents change how they vote in Congress to counter their primary threat. In response to a primary challenger incumbent I find that senators shift their voting behavior and vote more often with their party's leadership, but this is conditional on the threat-level posed by the primary challenger. High-quality primary challengers elicit the greatest shift in the incumbent's behavior, according to this study.
