Climate change literacy (CCL) is recognized as a critical component in formulating and implementing effective climate change policies and actions. However, the degree of CCL among those entrusted with these very responsibilities – public officials, remains relatively unexplored. This study pioneers an investigation into the CCL of public officials in Taiwan through a national survey. Our findings indicate that while a majority of these officials are cognizant of climate change issues, their personal commitment to climate action leaves considerable room for growth. Furthermore, their general understanding of climate change, its impacts, and the necessary adaptation strategies is found wanting. A similar deficit is observed in their comprehension of the relevance of the United Nations’ Sustainable Development Goals (SDGs) to climate change. Our study, by establishing a framework for assessing CCL among public officials and providing preliminary data on the CCL of Taiwanese officials, not only fills a significant knowledge gap but also lays the groundwork for comparative studies across nations. Moreover, our findings underscore the need for governments to invest in enhancing the CCL of their personnel, thereby equipping them with the requisite knowledge and motivation to contribute effectively to global climate change mitigation and adaptation efforts. Tight junctions (TJs) are the most apical components of junctional complexes in epithelial and endothelial cells. Barrier function is one of the major functions of TJ, which restricts the ions and small water-soluble molecules from passing through the paracellular pathway. Adherens junctions (AJs) play an important role in cell–cell adhesion and cell signaling. Gap junctions (GJs) are intercellular channels regulating electrical and metabolic signals between cells. It is well known that TJ integral membrane proteins, such as claudins and occludins, are the molecular building blocks responsible for TJ barrier function. However, recent studies demonstrate that proteins of other junctional complexes can influence and regulate TJ barrier function. Therefore, the crosstalk between different cell junctions represents a common means to modulate cellular activities. In this review, we will discuss the interactions among TJ, AJ, and GJ by focusing on how AJ and GJ proteins regulate TJ barrier function in different biological systems. [ABSTRACT FROM AUTHOR]

Background: Research has shown that epigenetic modification are involved the regulation of diapause in bivoltine silkworms (Bombyx mori), but it remains unclear how epigenetic modification in response to environmental signals precisely to regulate the diapause processing of bivoltine B. mori. Methods and results: In this study, the diapause terminated eggs of bivoltine B. mori, Qiufeng (QF) were divided into two groups: a QFHT group incubated at 25 °C with a natural day/night cycle to produce diapause eggs, and a QFLT group incubated at 16.5 °C in darkness to produce non-diapause eggs. On the 3rd day of the pupal stage, the total RNAs of the eggs were extracted and their N6-adenosine methylation (m6A) abundances were analyzed to explore the effects of m6A methylation on diapause in the silkworm. The results showed that 1984 m6A peaks are shared, 1563 in QFLT and 659 in QFHT. The m6A methylation level of the QFLT group was higher than that of the QFHT one in various signaling pathways. The m6A methylation rate of mevalonate kinase (MK) in the insect hormone synthesis pathway was significantly different between the two groups. The knockdown of MK by RNA interference in the pupae of QFLT resulted in females laying diapause eggs rather than non-diapause eggs after mating. Conclusions: m6A methylation involves in the diapause regulation of bivoltine B. mori by changing the expression levels of MK. A total of 3 607 surface soil samples in the cultivated land of Quanzhou City were collected and analyzed. The spatial distribution of available boron content in the cultivated soil and its influencing factors were studied by u- sing the statistical analysis method. The results showed that: the content of available boron in the cultivated soil of Quanzhou City was relatively deficient on the whole. The content of available boron ranged from 0.01 to 1.90 mg ⋅ kg-1, and the average content of available boron in the soil was 0.33 mg ⋅ kg-1. The content of available boron in 79.5% of cultivated soil was at a low or very low level. The land use types, soil types, altitude, soil physical and chemical properties had different effects on the content of available boron in soil. The content of available boron in soil varied greatly, and the content of available boron in the high altitude area was significantly lower than that in the low altitude area. Boron was generally deficient in the cultivated soil in Quanzhou City, so it was necessary to apply boron fertilizer. [ABSTRACT FROM AUTHOR]

The high frequency of chemotherapy resistance is ultimately responsible for clinical relapse in acute lymphoblastic leukemia (ALL). Nevertheless, the molecular mechanism relevant to glucocorticoid (GC) resistance remains ambiguous. Quantitative real-time polymerase chain reaction and Western blot were performed to detect the expressions of paraoxonase 2 (PON2), Bcl-2 and Bax. shRNA was used to knockdown PON2 expression in SUP-B15 and REH cell. CCK-8 and flow cytometry assay were conducted to monitor the changes of proliferation and apoptosis in ALL cells. The growth of ALL REH cells in vivo was determined using transplanted tumor model. This study was designed to identify GC resistance-associated genes by means of the transcriptome chip from the public Gene Expression Omnibus database, and preliminarily investigation of dexamethasone (DEX)-resistance mechanism in ALL. We disclosed that PON2 expression was elevated in ALL patients and especially higher in DEX-resistance ALL patients. Then, cell apoptosis assay suggested that silencing of PON2 dramatically promoted in DEX-resistant ALL cells apoptosis and the activity of Caspase 3 induced by DEX administration. In xenograft tumor model, PON2 knockdown significantly reduced DEX-resistant ALL cells growth in immunodeficient mice. Collectively, inhibition of PON2 may represent a novel method to restore the sensitivity of treatment-resistant ALL to GC-induced cell death. [ABSTRACT FROM AUTHOR]

Background: The Maastricht V/Florence Consensus Report recommends amoxicillin‐fluoroquinolone triple or quadruple therapy as a second‐line treatment for Helicobacter pylori infection. An important caveat of amoxicillin‐fluoroquinolone rescue therapy is poor eradication efficacy in the presence of fluoroquinolone resistance. The study aimed to investigate the efficacies of tetracycline‐levofloxacin (TL) quadruple therapy and amoxicillin‐levofloxacin (AL) quadruple therapy in the second‐line treatment of H. pylori infection. Methods: Consecutive H. pylori‐infected subjects after the failure of first‐line therapies were randomly allocated to receive either TL quadruple therapy (tetracycline 500 mg QID, levofloxacin 500 mg QD, esomeprazole 40 mg BID, and tripotassium dicitrato bismuthate 300 mg QID) or AL quadruple therapy (amoxicillin 500 mg QID, levofloxacin 500 mg QD, esomeprazole 40 mg BID, and tripotassium dicitrato bismuthate 300 mg QID) for 10 days. Post‐treatment H. pylori status was assessed 6 weeks after the end of therapy. Results: The study was early terminated after an interim analysis. In the TL quadruple group, 50 out of 56 patients (89.3%) had successful eradication of H. pylori infection. Cure of H. pylori infection was achieved only in 39 of 52 patients (69.6%) receiving AL quadruple therapy. Intention‐to‐treat analysis showed that TL quadruple therapy achieved a markedly higher eradication rate than AL quadruple therapy (95% confidence interval: 4.8% to 34.6%; p = 0.010). Further analysis revealed that TL quadruple therapy had a high eradication rate for both levofloxacin‐susceptible and resistant strains (100% and 88.9%). In contrast, AL quadruple therapy yielded a high eradication for levofloxacin‐susceptible strains (90.9%) but a poor eradication efficacy for levofloxacin‐resistant strains (50.0%). The two therapies exhibited comparable frequencies of adverse events (37.5% vs 21.4%) and drug adherence (98.2% vs 94.6%). Conclusions: Ten‐day TL quadruple therapy is more effective than AL quadruple therapy in the second‐line treatment of H. pylori infection in a population with high levofloxacin resistance. A simple, efficient, and cost-effective extended graphite as a supporting platform further supported the MnO2 growth for the construction of hierarchical flower-like MnO2/extended graphite. MnO2/extended graphite exhibited an increase in sp2 carbon bonds in comparison with that of extended graphite. It can be expected to display better electrical conductivity and further promote electron/ion transport kinetics for boosting the electrochemical performance in supercapacitors and glucose sensing. In supercapacitors, MnO2/extended graphite delivered an areal capacitance value of 20.4 mF cm−2 at 0.25 mA cm−2 current densities and great cycling stability (capacitance retention of 83% after 1000 cycles). In glucose sensing, MnO2/extended graphite exhibited a good linear relationship in glucose concentration up to about 5 mM, sensitivity of 43 μA mM−1cm−2, and the limit of detection of 0.081 mM. It is further concluded that MnO2/extended graphite could be a good candidate for the future design of synergistic multifunctional materials in electrochemical techniques. [ABSTRACT FROM AUTHOR]

Cataract is the main cause of visual impairment and blindness worldwide while the only effective cure for cataract is still surgery. Consecutive phacoemulsification under topical anesthesia has been the routine procedure for cataract surgery. However, patients often grumbled that they felt more painful during the second-eye surgery compared to the first-eye surgery. The intraoperative pain experience has negative influence on satisfaction and willingness for second-eye cataract surgery of patients with bilateral cataracts. Intraoperative ocular pain is a complicated process induced by the nociceptors activation in the peripheral nervous system. Immunological, neuropsychological, and pharmacological factors work together in the enhancement of intraoperative pain. Accumulating published literatures have focused on the pain enhancement during the second-eye phacoemulsification surgeries. In this review, we searched PubMed database for articles associated with pain perception differences between consecutive cataract surgeries published up to Feb. 1, 2024. We summarized the recent research progress in mechanisms and interventions for pain perception enhancement in consecutive second-eye phacoemulsification cataract surgeries. The increasing use of nitrogen fertilizers exerts extreme pressure on the environment (e.g., greenhouse gas emissions, GHGs) for winter wheat-summer maize rotation systems in the North China Plain. The application of controlled-release fertilizers is considered as an effective measure to improve crop yield and nitrogen fertilizer utilization efficiency. To explore the impact of one-time fertilization of controlled-release blended fertilizer on crop yield and GHGs of a wheat-maize rotation system, field experiments were carried out in Dezhou Modern Agricultural Science and Technology Park from 2020 to 2022. Five treatments were established for both winter wheat and summer maize, including no nitrogen control (CK), farmers' conventional nitrogen application (FFP), optimized nitrogen application (OPT), CRU1 (the blending ratio of coated urea and traditional urea on winter wheat and summer maize was 5:5 and 3:7, respectively), and CRU2 (the blending ratio of coated urea and traditional urea on winter wheat and summer maize was 7:3 and 5:5, respectively). The differences in yield, nitrogen fertilizer utilization efficiency, fertilization economic benefits, and GHGs among different treatments were compared and analyzed. The results showed that nitrogen application significantly increased the single season and annual crop yields of the wheat-maize rotation system ( P < 0.05). Compared with those of FFP, the CRU1 and CRU2 treatments increased the yields of summer maize by 0.4% to 5.6%, winter wheat by -5.4% to 4.1%, and annual yields by -1.1% to 3.9% ( P > 0.05). N recovery efficiency (NRE), N agronomic efficiency (NAE), and N partial factor productivity (NPFP) were increased by -8.6%-43.4%, 2.05-6.24 kg·kg -1 , and 4.24-10.13 kg·kg -1 , respectively. Annual net income increased by 0.2% to 6.3%. Nitrogen application significantly increased the annual emissions of soil N 2 O and CO 2 in the rotation system ( P < 0.05) but had no effect on the annual emissions of CH 4 (except for in the FFP treatment in the first year). The annual total N 2 O emissions under the CRU1 and CRU2 treatments were significantly reduced by 23.4% to 30.2% compared to those under the FFP treatment ( P < 0.05). Additionally, nitrogen application significantly increased the annual global warming potential (GWP) of the rotation system ( P < 0.05), but the intensity of greenhouse gas emissions was reduced due to the increase in crop yields. Compared with that under FFP, the annual GWP under the CRU1 and CRU2 treatments decreased by 9.6% to 11.5% ( P < 0.05), and the annual GHGs decreased by 11.2% to 13.8% ( P > 0.05). In summary, the one-time application of controlled-release blended fertilizer had a positive role in improving crop yield and economic benefits, reducing nitrogen fertilizer input and labor costs, and GHGs, which is an effective nitrogen fertilizer management measure to promote cleaner production of food crops in the North China Plain.

To elucidate the agronomic and environmental effects of single basal application of controlled-release blended fertilizer in summer maize, and optimize management measures of nitrogen fertilizer for grain production in North China Plain, we conducted a field experiment in Dezhou Modern Agricultural Science and Technology Park in Shandong Province. There were four treatments: CK (no N fertilizer), FFP (farmer's fertilizing practice, 240 kg N·hm -2 ), OPT (optimized nitrogen application, 210 kg N·hm -2 ), and CRBF (controlled-release blended fertilizer with single basal application, 210 kg N·hm -2 ). We compared maize yield and reactive nitrogen loss, and quantitatively evaluated the carbon and nitrogen footprints by using life cycle assessment method. The results showed that nitrogen application significantly increased summer maize yield. Compared with FFP, OPT and CRBF increased summer corn yield by 0.7% and 2.9%, respectively, decreased the total amount of ammonia volatilization, N 2 O emission, and nitrate leaching by 13.0% and 72.7%, 13.3% and 37.5%, 20.5% and 23.5% respectively. Compared with CK, nitrogen application significantly increased the global warming potential (GWP) of summer maize production. Compared with FFP, GWP and greenhouse gas emission intensity of OPT decreased by 3.8% and 4.2%, while the reduction of CRBF were 8.7% and 12.0%, respectively. Compared with CK, nitrogen application significantly increased the carbon and nitrogen footprint of summer maize production. The production and transportation of nitrogen fertilizer and soil greenhouse gas emission were the main contributing factors of the carbon footprint, with contribution rates of 54%-60% and 24%-31%, respectively. Nitrate leaching was the main contributing factor of nitrogen footprint, with contribution rate of 57%-94%. Compared with FFP, the carbon and nitrogen footprints of OPT and CRBF were reduced by 11.0% and 16.5%, 19.6% and 28.4%, respectively. Considering the yield, reactive nitrogen loss and carbon and nitrogen footprint, we recommended the single basal application of controlled-release blended fertilizer as an effective nitrogen fertilizer management measure to promote grain clean production in the North China Plain.

We explored the effects of addition of sorghum stubble rhizosphere soil on the growth of continuous cropping cucumber and rhizosphere microbial community in a pot experiment. The diffe-rences in soil bacterial and fungal community composition were analyzed with fluorescence quantitative PCR and high-throughput sequencing technology. There were four treatments: CK (no fertilization), T 1 (fertilizer only), T 2 (optimized fertilization), and T 3 (optimized fertilization + rhizosphere soil of sorghum stubble). The results showed that compared with other treatments, T 3 promoted the growth and development of cucumber, and increased the abundance of 16S rRNA and ITS rRNA genes in soil. Compared with the T 1 treatment, T 2 and T 3 significantly increased the richness and diversity of bacterial communities. There was no significant difference in fungal community richness and diversity among different treatments. Adding rhizosphere soil of sorghum stubble changed the composition of bacterial and fungal communities at both phylum and genus levels. For bacteria, it increased the abundances of Acidobacteria and Bacteroides, but decreased that of Proteobacteria, Firmicutes, Nitrospira and Bacillus. For fungi, it increased the abundance of Basidiomycota, Trichoderma and Pseudurotium , but decreased that of Fusarium and Metarhizium . Results of redundancy analysis showed that soil nitrate and organic matter were the key factors affecting the difference of bacterial and fungal community composition, respectively. In conclusion, addition of sorghum stubble rhizosphere soil improved the total abundance of soil microorganisms and bacterial diversity for continuous cropping cucumber. It increased the abundance of beneficial bacteria Trichoderma , reduced that of pathogenic Fusarium , and maintained the survival rate of cucumber, thus provided a feasible solution for alleviating the barriers for the continuous cropping of cucumber.
