Positive-reinforcement training of laboratory pigs can reduce the reliance on forced manual restraint and anaesthesia for sample collection, reducing stress and physiological disruption. Training regimens for laboratory pigs typically rely on specialised equipment for restraint, such as Panepinto slings, with a time investment that may not be justified for short-term studies. These training regimens also commonly rely on pigs being lifted into sling restraints, which is not practical for studies involving large pigs. We developed and assessed a rapid, three-phase, positive-reinforcement training regimen for both individually housed and group-housed laboratory pigs to facilitate the collection of minimally invasive samples consciously and voluntarily. The time to complete each phase of training in both individually housed and group-housed pigs was recorded. The behaviour of the individually housed pigs was assessed via an ethogram of behaviours exhibited during a human approach test, and stress response was assessed by analysing salivary corticosterone. The rapid, positive-reinforcement training regimen successfully facilitated oral swabbing, rectal swabbing and rectal thermometer insertion from individually housed (within 18 days) and group-housed (within 6 days) pigs. The trained pigs displayed increasing positive behaviours, no or very few negative behaviours and corticosterone levels within normal limits throughout the study. This training regimen provides a practical and welfare-positive tool for the collection of minimally invasive samples from both small and large laboratory pigs, with a low time investment of 2–5 min/pig/day without the need for specialised restraint equipment.

Anaesthesia is sometimes required for the effective restraint of laboratory pigs for sample collection. Yet, anaesthesia can initiate a range of physiological disruptions that can increase variability in study data and lead to poorer animal welfare. Judicious use of anaesthesia can mitigate experimental, human safety, and animal welfare concerns, but it does not eliminate the potential for adverse effects. The use of reversal agents can shorten recovery time and reduce the physiological impacts of anaesthesia but can also cause additional side effects. We, therefore, trialled the use of low-dose atipamezole (0.12 mg/kg) for the antagonism of xylazine in laboratory pigs anaesthetised using a combination of xylazine and zolazepam/tiletamine. We measured time to recovery, selected clinical variables, recovery characteristics, and behaviours to investigate if a low dose of antagonist decreased recovery time and reduced the physiological impacts of anaesthesia whilst avoiding adverse negative side effects. We categorised side effects and behaviours as having either a low or high negative welfare impact based on the potential risk of injury and whether behaviours were displayed before or after return to consciousness. Collectively, our results indicated that while the use of low-dose atipamezole decreased recovery time and improved thermoregulation in most pigs, it introduced and exacerbated adverse side effects and behaviours that can lead to poorer welfare outcomes for laboratory pigs.

Enhancing livestock biosecurity is critical to safeguard the livelihoods of farmers, global and local economies, and food security. Vaccination is fundamental to the control and prevention of exotic and endemic high-priority infectious livestock diseases. Successful implementation of vaccination in a biosecurity plan is underpinned by a strong understanding of correlates of protection—those elements of the immune response that can reliably predict the level of protection from viral challenge. While correlates of protection have been successfully characterized for many human viral vaccines, for many high-priority livestock viral diseases, including African swine fever and foot and mouth disease, they remain largely uncharacterized. Current literature provides insights into potential correlates of protection that should be assessed during vaccine development for these high-priority mammalian livestock viral diseases. Establishment of correlates of protection for biosecurity purposes enables immune surveillance, rationale for vaccine development, and successful implementation of livestock vaccines as part of a biosecurity strategy.

Copper processing operations, such as smelters and refineries, can produce airborne particles that may impact the health of workers. At these operations, worker exposure to chemicals are regularly monitored to ensure that regulatory compliance with occupational exposure limit values (OELVs) are maintained. Determining the type of airborne particles present is important for characterizing the composition of dust exposures and better understanding the relationship between worker exposure and health. Routine methods of analysis (e.g., chemical assay) are unable to differentiate between phases containing the same elements and may result in ambiguity. A novel approach of a combination of Quantitative Evaluation of Materials by Scanning Electron Microscope (QEMSCAN) and chemical characterization was used here to evaluate airborne and settled dust collected at key locations throughout a copper smelter in Europe. The copper (Cu) phases present in the airborne dust are indicative of the activities performed at specific locations. In the batch preparation area where Cu concentrate is received, significant amounts of Cu were carried in sulfidic minerals (chalcocite, chalcopyrite/bornite, >40%), whereas near the anode and electric furnace, the majority of Cu in dust was carried in metallic and oxidic phases (60–70%). Particle size analysis of the settled dust indicates that the sulfidic and oxidic Cu minerals are more likely to become airborne over metallic Cu. Furthermore, overall Cu concentrations decreased with particle size where metallic and oxidic Cu dominate, which suggests that differences in the proportion of Cu forms present in the dust will impact how much Cu ends up in the respirable fraction. These results highlight the need to understand the characterization of Cu in dust in order to set better OELVs.

Stress and general anesthesia have an impact on the functional response of the organism due to the detrimental effects on cardiovascular, immunological, and metabolic function, which could limit the organism's response to an infectious event. Animal studies have formed an essential step in understanding and mitigating infectious diseases, as the complexities of physiology and immunity cannot yet be replicated in vivo. Using animals in research continues to come under increasing societal scrutiny, and it is therefore crucial that the welfare of animals used in disease research is optimized to meet both societal expectations and improve scientific outcomes. Everyday management and procedures in animal studies are known to cause stress, which can not only cause poorer welfare outcomes, but also introduces variables in disease studies. Whilst general anesthesia is necessary at times to reduce stress and enhance animal welfare in disease research, evidence of physiological and immunological disruption caused by general anesthesia is increasing. To better understand and quantify the effects of stress and anesthesia on disease study and welfare outcomes, utilizing the most appropriate animal monitoring strategies is imperative. This article aims to analyze recent scientific evidence about the impact of stress and anesthesia as uncontrolled variables, as well as reviewing monitoring strategies and technologies in animal models during infectious diseases.

Systematic manual and algorithmic classification workflows to characterize rock types are increasingly applied in the mineral exploration and mining industry, leveraging large systematically collected datasets. The aim of these are robust and repeatable classifications to aid more traditional visual logging practices. This study uses random forest algorithms to examine the impacts of integrating distinct datasets with complementary characteristics; chemistry to enable compositional distinctions, and photography to enable textural distinctions. We use a random forest classifier to examine the accuracy metrics of models producing rock type classifications using these two data types independently and integrated together. Prediction accuracy, measured using 10-fold cross validation, was 87% for geochemical-only inputs, 85% for photographic-only inputs, and 90% for mixed inputs from both datasets. A mining and exploration project in the Late Miocene to early Pliocene porphyry belt in Chile is the site of this case study, where datasets were systematically acquired using in-field methods on historical drill-cores. Results indicate that classification of lithology is improved by integration of photography-based and composition-based feature inputs. We infer that the benefits of integration would increase in proportion with increasing compositional similarity between rock types. This approach might also be applied to similar geological problems, such as alteration or metallurgical classifications; and with somewhat distinct datatypes, such as geochemical interval data and photographic metric extraction from coincident intervals in core photos.

BackgroundAchieving herd immunity through vaccination depends upon the public’s acceptance, which in turn relies on their understanding of its risks and benefits. The fundamental objective of public health messaging on vaccines is therefore the clear communication of often complex information and, increasingly, the countering of misinformation. The primary outlet shaping public understanding is mainstream online news media, where coverage of COVID-19 vaccines was widespread. ObjectiveWe used text-mining analysis on the front pages of mainstream online news to quantify the volume and sentiment polarization of vaccine coverage. MethodsWe analyzed 28 million articles from 172 major news sources across 11 countries between July 2015 and April 2021. We employed keyword-based frequency analysis to estimate the proportion of overall articles devoted to vaccines. We performed topic detection using BERTopic and named entity recognition to identify the leading subjects and actors mentioned in the context of vaccines. We used the Vader Python module to perform sentiment polarization quantification of all collated English-language articles. ResultsThe proportion of front-page articles mentioning vaccines increased from 0.1% to 4% with the outbreak of COVID-19. The number of negatively polarized articles increased from 6698 in 2015-2019 to 28,552 in 2020-2021. However, overall vaccine coverage before the COVID-19 pandemic was slightly negatively polarized (57% negative), whereas coverage during the pandemic was positively polarized (38% negative). ConclusionsThroughout the pandemic, vaccines have risen from a marginal to a widely discussed topic on the front pages of major news outlets. Mainstream online media has been positively polarized toward vaccines, compared with mainly negative prepandemic vaccine news. However, the pandemic was accompanied by an order-of-magnitude increase in vaccine news that, due to low prepandemic frequency, may contribute to a perceived negative sentiment. These results highlight important interactions between the volume of news and overall polarization. To the best of our knowledge, our work is the first systematic text mining study of front-page vaccine news headlines in the context of COVID-19.
