Background: The synthetic liver X receptor ligand (LXR) T0901317 (T0) has been reported to attenuate atherosclerosis (AS) without hyperglyceridemia due to innovative drug combination or nano-sized drug delivery. Given the key roles of mangiferin (MGF) in lipid metabolism and atherogenesis, it is critical to investigate progression of atherosclerotic lesion after combined treatment of MGF and T0. Methods: Atherosclerotic plaque formation and hepatic lipid accumulation were compared in Apoe−/− mice among T0 and/or MGF treatment. The in vitro functions of MGF and T0 were analyzed by Oil-red O staining, cholesterol efflux assay, transmission electron microscopy and western blot analyses with or without acetylated low density lipoprotein. Results: The combination therapy are effective regulators for atherosclerotic plaque formation in Apoe−/− mice, due to upregulation of ABCA1 and ABCG1 induced by LXR activation. Subsequently, we identified autophagy promoted by MGF and T0 treatment establishes a positive feedback loop that increases cholesterol efflux, resulted from LXRα activation. Under atherogenic conditions, the autophagy inhibitor CQ abolished the enhancement effect on cholesterol outflow of MGF and T0. Mechanically, MGF and T0 promotes LXRα and mTOR/AMPK signaling cascade in macrophage, and promotes AMPK signaling cascade in hepatocyte, leading to lipid metabolic homeostasis. Conclusions: Altogether, our findings reveal that MGF and T0 engages in AS therapy without side effects by activating AMPK-dependent autophagy to promote macrophage cholesterol efflux, and MGF might serve as a natural compound to assist T0 in AS via targeting autophagy. Fezolinetant (VEOZAH™) is an oral, small molecule, neurokinin 3 receptor (NK3R) antagonist, which is being developed by Astellas Pharma Inc. for the treatment of moderate to severe vasomotor symptoms (VMS) or hot flashes due to menopause. Inhibiting NK3R-mediated signalling in the central nervous system is a non-hormonal strategy to modulate the activity of neurones that are associated with thermoregulation, thereby reducing the frequency and severity of VMS. Fezolinetant received its first approval in the USA in May 2023 for the treatment of moderate to severe VMS due to menopause. Polyurethane foams (PUFs) are a significant group of polymeric foam materials. Thanks to their outstanding mechanical, chemical, and physical properties, they are implemented successfully in a wide range of applications. Conventionally, PUFs are obtained in polyaddition reactions between polyols, diisoycyanate, and water to get a CO2 foaming agent. The toxicity of isocyanate has attracted considerable attention from both scientists and industry professionals to explore cleaner synthesis routes for polyurethanes excluding the use of isocyanate. The polyaddition of cyclic carbonates (CCs) and polyfunctional amines in the presence of an external blowing agent or by self-blowing appears to be the most promising route to substitute the conventional PUFs process and to produce isocyanate-free polyurethane foams (NIPUFs). Especially for polyhydroxyurethane foams (PHUFs), the use of a blowing agent is essential to regenerate the gas responsible for the creation of the cells that are the basis of the foam. In this review, we report on the use of different blowing agents, such as Poly(methylhydrogensiloxane) (PHMS) and liquid fluorohydrocarbons for the preparation of NIPUFs. Furthermore, the preparation of NIPUFs using the self-blowing technique to produce gas without external blowing agents is assessed. In this study, fluorine-19 nuclear magnetic resonance spectroscopy (19F NMR) served as a highly specific tool for identification of fluorinated new psychoactive substances (NPS) as well as a suitable analytical method for the accurate quantification of fluorinated NPS in different seized samples. In the first part of the study, 19F NMR spectroscopy of a number of different fluorinated NPS, including 51 synthetic cannabinoids, 8 synthetic cathinones, 7 phenethylamines, 8 fentanyl analogues, and 9 other types of compounds was conducted. The chemical shifts and multiplet of the primary fluorides (RCH2F), fluorobenzenes (ortho-ArF, meta-ArF, and para-ArF), and trifluoromethylbenzenes (ArCF3) were discussed in detail to illustrate the role of 19F signals as special fingerprints in assisting the structure identification of fluorine-containing NPS. To the best of our knowledge, this study is the largest evaluation of fluorinated NPS compounds by 19F NMR. The second part of this study dealt with the problems encountered in the 19F quantification procedure and the criteria to be considered for successful quantification by 19F NMR. General high field (HF)- and low field (LF)- 19F qNMR methods for the quantification of fluorinated NPS were established after the thorough discussion of NMR spectrum acquisition and processing parameters such as: transmitter frequency offset (O1P), spin-lattice relaxation time (T1), and different baseline correction methods. The limit of quantifications (LOQs) for HF-19F qNMR varied between 0.1 mg/mL and 0.2 mg/mL, and for LF-19F qNMR varied between 1.0 mg/mL and 2.0 mg/mL. The limit of detections (LODs) for HF-19F qNMR varied between 0.03 mg/mL and 0.06 mg/mL, and for LF-19F qNMR varied between 0.3 mg/mL and 0.6 mg/mL. Finally, the developed methods were applied for the quantification of fluorinated-NPS in seventeen herbal blends, e-liquid, tablet, and powder NPS seizures. [ABSTRACT FROM AUTHOR]
