Systemic sclerosis (SSc) interstitial lung disease (ILD) is among the leading causes of SSc-related morbidity and mortality. Tocilizumab (TCZ, anti-IL6RA) has demonstrated a reduced rate of pulmonary function decline in two phase 2/3 trials (faSScinate and focuSSced) in SSc-ILD patients. We performed transcriptome analysis of skin biopsy samples collected in the studies to decipher gene networks that were potentially associated with clinical responses to TCZ treatment. One module correlated with disease progression showed pharmacodynamic changes with TCZ treatment, and was characterized by plasma cell (PC) genes. PC signature gene expression levels were also significantly increased in both fibrotic SSc and IPF lungs compared to controls. scRNAseq analyses confirmed that PC signature genes were co-expressed in CD38 and CD138 expressing PC subsets in SSc lungs. These data provide insights into the potential role of PC in disease progression and mechanisms of action of TCZ in fibrotic interstitial lung diseases.

Altered myeloid inflammation and lymphopenia are hallmarks of severe infections. We identified the upregulated EN-RAGE gene program in airway and blood myeloid cells from patients with acute lung injury from SARS-CoV-2 or other causes across 7 cohorts. This program was associated with greater clinical severity and predicted future mechanical ventilation and death. EN-RAGEhi myeloid cells express features consistent with suppressor cell functionality, including low HLA-DR and high PD-L1. Sustained EN-RAGE program expression in airway and blood myeloid cells correlated with clinical severity and increasing expression of T cell dysfunction markers. IL-6 upregulated many EN-RAGE program genes in monocytes in vitro. IL-6 signaling blockade by tocilizumab in a placebo-controlled clinical trial led to rapid normalization of EN-RAGE and T cell gene expression. This identifies IL-6 as a key driver of myeloid dysregulation associated with worse clinical outcomes in COVID-19 patients and provides insights into shared pathophysiological mechanisms in non-COVID-19 ARDS.

Introduction: With depression's growing global prevalence and substantial impact, effective prevention and management strategies are imperative. Our study aims to perform a thorough bibliometric analysis of existing research on the impact of exercise on depression. Methods: A comprehensive analysis of Web of Science Core Collection publications from 2000 to 2020 was performed, highlighting trends, themes, and influential authors. The study focused on subject categories, source journals, countries/regions, institutions, and prolific authors. Co-citation and keyword analyses revealed key themes, hotspots and the thematic evolution. Results: The multidisciplinary nature of this research is evident across psychiatry, psychology, neuroscience, and sports science. Specific populations such as women, the elderly, and those with chronic illnesses were targeted. Mind-body exercises like yoga and tai chi gained prominence. Co-citation clusters showcased the evolution from early investigations on exercise's impact to recent dose-response and protocol studies. Conclusions: This bibliometric analysis provides insights into the dynamic field of exercise interventions for depression. It underscores the importance of individual differences, calls for guidelines considering comorbidities, and points towards future directions such as exploring mind-body exercise mechanisms and well-designed clinical trials. This study contributes to a comprehensive understanding of the research landscape and informs future endeavors aimed at refining depression treatment through exercise interventions.

Although schizophrenia patients exhibit structural abnormalities in the striatum, it remains largely unknown for the role of the striatum subregions in the treatment response of antipsychotic drugs. The purpose of this study was to investigate the associations between the striatal subregions and improved clinical symptoms in first-episode drug-naïve (FEDN) schizophrenia. Forty-two FEDN schizophrenia patients and 29 healthy controls (HCs) were recruited. At baseline, the Positive and Negative Syndrome Scale (PANSS) was used to assess the clinical symptoms of patients, MRI scanner was used to obtain anatomical images of patients and HCs. After 12-week stable doses of risperidone treatment, clinical symptoms were obtained in 38 patients and anatomical images in 26 patients. After 12 weeks of treatment, the left nucleus accumbens volume decreased, whereas the left pallidum volume increased in schizophrenia patients. The decreased left nucleus accumbens volume was positively correlated with cognitive factor improvement measured by PANSS. Intriguingly, greater left nucleus accumbens volume at baseline predicted greater cognitive improvements. Furthermore, the responders who had >50 % improvement in cognitive symptoms exhibited significantly greater baseline left nucleus accumbens volume compared to non-responders. The left striatum volume at baseline and after treatment predicted the cognitive improvements in FEDN schizophrenia, which could be a potential biomarker for the development of precision medicine approaches targeting cognitive function.

Abstract During the past decade, cognitive neuroscience has been calling for population diversity to address the challenge of validity and generalizability, ushering in a new era of population neuroscience. The developing Chinese Color Nest Project (devCCNP, 2013–2022), the first ten-year stage of the lifespan CCNP (2013–2032), is a two-stages project focusing on brain-mind development. The project aims to create and share a large-scale, longitudinal and multimodal dataset of typically developing children and adolescents (ages 6.0–17.9 at enrolment) in the Chinese population. The devCCNP houses not only phenotypes measured by demographic, biophysical, psychological and behavioural, cognitive, affective, and ocular-tracking assessments but also neurotypes measured with magnetic resonance imaging (MRI) of brain morphometry, resting-state function, naturalistic viewing function and diffusion structure. This Data Descriptor introduces the first data release of devCCNP including a total of 864 visits from 479 participants. Herein, we provided details of the experimental design, sampling strategies, and technical validation of the devCCNP resource. We demonstrate and discuss the potential of a multicohort longitudinal design to depict normative brain growth curves from the perspective of developmental population neuroscience. The devCCNP resource is shared as part of the “Chinese Data-sharing Warehouse for In-vivo Imaging Brain” in the Chinese Color Nest Project (CCNP) – Lifespan Brain-Mind Development Data Community ( https://ccnp.scidb.cn ) at the Science Data Bank.

Objective To explore the relationship between regional leptomeningeal collateral (rLMC) score, peripheral blood angiotensin-converting enzyme (ACE), matrix-derived growth factor (SDF)-1α levels, and the prognosis of acute anterior circulation ischemic stroke (AACI) treated with intravenous thrombolysis. Methods A total of 117 patients with AACI admitted to Jinan Third People's Hospital from August 2018 to August 2022 were selected. Based on the modified Rankin Scale (mRS) score at 3 months after intravenous thrombolysis, patients were divided into good prognosis group (mRS score 0-1, n=70) and poor prognosis group (mRS score 2-5, n=47).The Clinical data, rLMC score, ACE, and SDF-1α were compared between the two groups. The correlation between rLMC score, ACE, SDF-1α, and the National Institutes of Health Stroke Scale (NIHSS) score at admission in AACI patients was analyzed. Logistic multivariate analysis was used to analyze the influencing factors on the prognosis of AACI after intravenous thrombolysis, and the predictive efficacy of each indicator for the prognosis of AACI treated with intravenous thrombolysis was evaluated using receiver operating characteristic (ROC) curve. Results Multivariate logistic analysis showed that higher NIHSS score at admission, lower rLMC score, higher ACE level, and lower SDF-1α level were independent risk factors for poor prognosis of AACI after intravenous thrombolysis (P＜0.05). The rLMC score and SDF-1α level in the 117 AACI patients were negatively correlated with the NIHSS score at admission(r=－0.402，－0.577，P＜0.05), while ACE level was positively correlated with the NIHSS score at admission(r=0.357，P＜0.05). The combined use of rLMC score, ACE, and SDF-1α had the highest predictive efficacy for the prognosis of AACI treated with intravenous thrombolysis (AUC=0.824). Conclusion rLMC score, peripheral blood ACE, and SDF-1α levels are closely related to the prognosis of AACI after intravenous thrombolysis and may serve as predictive factors for unfavorable outcomes at three months.
