Abstract

Deep-sea eukaryotic microorganisms play a vital role in degrading organic matter and geochemically cycling elements in the deep ocean. However, the impact of sampling methods on detection of these microorganisms under high hydrostatic pressure remains uncertain. In this study, we compared a traditional water sampling method using a Niskin bottle, an in situ microbial filtration and fixation method (ISMIFF), and a multiple in situ nucleic acid collection (MISNAC) method to exhibit differences in the community structures that were collected at ~590–3100 m in the South China Sea (SCS). The classification and biodiversity indices of 18S rDNA Illumina sequencing reads from the V9 variation region revealed higher diversity for MISNAC DNA absorption column samples compared to others. Importantly, the relative abundance of Ciliophora (19.49%), Copepoda (23.31%), and Diplonemea (10.67%) was higher in MISNAC adsorption column samples, while Retaria (48.86%) were dominant in the MISNAC membrane samples. This indicates that MISNAC columns might collect more DNA in situ for the naked protists, while Retaria with a carbonate shell were more likely retained on the membrane. In conclusion, MISNAC is an effective method for DNA collection of deep-sea eukaryotic microorganisms and provides valuable materials for studying deep-sea microbial ecosystems. 13. A Study on the Optimization of Wind Environment of Existing Villa Buildings in Lingnan Area: A Case Study of Jiangmen’s “Yunshan Poetic” Moon Island Houses

Abstract

Effective natural ventilation reduces humidity, cools the space, and enhances thermal comfort. In light of the frequent ventilation issues in the Lingnan area, this research suggests a successful ventilation technique using Jiangmen’s “Yunshan Poetic” Moon Island houses as an example. With its symmetrical architectural layout of townhouses and its primary courtyard villa product, the community typifies the Lingnan area. First off, we discovered that the district’s average temperature is as high as 30.95 °C and its average humidity is as high as 83.592%RH using actual measurements and simulation of heat and humidity data. The district’s buildings’ issues with dampness, peeling walls, and substance mold are primarily caused by poor ventilation. Secondly, the PHOENICS program was used to provide efficient ventilation solutions for the following six aspects: external wind infusion organization, group orientation layout, planar grouping optimization, building façade combination, monolithic building openings, and indoor ventilation block. In order to determine if the technique is effective, the ventilation variables are compared before and after optimization using the Building Ventilation Effectiveness Test and Evaluation Criteria. The study concluded that the building’s architectural characteristics and the local climate have an impact on natural ventilation’s effectiveness. This serves as a guide for both the scientific layout development of future urban settlements and the optimization of ventilation of existing villa buildings in humid and hot areas.

19. Potential sexual pheromones of sea cucumber Apostichopus japonicusand their functions on aggregative and spawning behavior

Abstract

The surging market demand for sea cucumbers (Apostichopus japonicus) has fueled a thriving growth in the sea cucumber breeding industry. Pheromones are molecules that evolve signals in defined proportions in the case of multicomponent pheromones, emitted by one individual and received by a second individual of the same species to cause a specific response. It remains an enigma as to how the species of sea cucumber, devoid of visual, auditory, and olfactory senses, aggregate and communicate within their groups, as well as coordinate gamete release during the breeding period. We hypothesize that pheromones may serve as the medium for facilitating these behaviors and propose conducting experiments centered around pheromonal investigation. In this study, we aimed to analyze common metabolites of male A. japonicususing LC-MS/MS technology and bioinformatics. Coelomic fluid and ejaculate fluid were analyzed, with aquaculture water serving as the control. Eight target pheromones were analyzed, and eight of metabolite analogs were tested for aggregation and spawning behaviors. Results showed that β-sitosterol at a concentration of 10 μg·L−1significantly promoted aggregation (21.25 % average rate) and induced a significant increase in spawning rate (58 %) of A. japonicus. The significance of this study lies in the fact that by collecting and utilizing pheromones of A. japonicus, farmers can accurately determine the reproductive cycle and optimal breeding season for A. japonicus. This enables fish farmers to implement effective breeding management strategies at the appropriate time, thereby increasing the success rate of reproduction and providing valuable technical support for the cultivation of A. japonicusseedlings.
