A recent surge in textbooks studies has revealed a closer link with neoliberalism and the way they construct neoliberal subjects. This paper uses Foucauldian governmentality as the conceptual lens to analyze the neoliberal discourses in EFL textbooks used in English Access Microscholarship (EAM)--one of the US-aided global ELT programs in Pakistan. English language learners' views on course outcomes and textbooks were also examined. The study shows that among others, English as a neoliberal life skill, celebrity culture, consumerism, entrepreneurship, and individual and corporate social responsibility dominate textbooks. It is thus found that textbooks play an important role in neoliberalizing learners. Moreover, English language learners perceive English as a key to economic success. They also value consumerism, branding, and personal responsibility. In light of the study findings, we suggest a decolonial option, reflective activism, and post-method pedagogy as possible alternatives at the micro-, meso-, and macro-levels to resist the discourses of neoliberalism and colonial power patterns entrenched in a postcolonial society like Pakistan.

Abstract Background In gut ecosystems, there is a complex interplay of biotic and abiotic interactions that decide the overall fitness of an individual. Divulging the microbe-microbe and microbe-host interactions may lead to better strategies in disease management, as microbes rarely act in isolation. Network inference for microbial communities is often a challenging task limited by both analytical assumptions as well as experimental approaches. Even after the network topologies are obtained, identification of important nodes within the context of underlying disease aetiology remains a convoluted task. We therefore present a network perspective on complex interactions in gut microbial profiles of individuals who have multiple sclerosis with and without Mycobacterium avium subspecies paratuberculosis (MAP) infection. Our exposé is guided by recent advancements in network-wide statistical measures that identify the keystone nodes. We have utilised several centrality measures, including a recently published metric, Integrated View of Influence (IVI), that is robust against biases. Results The ecological networks were generated on microbial abundance data (n = 69 samples) utilising 16 S rRNA amplification. Using SPIEC-EASI, a sparse inverse covariance estimation approach, we have obtained networks separately for MAP positive (+), MAP negative (-) and healthy controls (as a baseline). Using IVI metric, we identified top 20 keystone nodes and regressed them against covariates of interest using a generalised linear latent variable model. Our analyses suggest Eisenbergiella to be of pivotal importance in MS irrespective of MAP infection. For MAP + cohort, Pyarmidobacter, and Peptoclostridium were predominately the most influential genera, also hinting at an infection model similar to those observed in Inflammatory Bowel Diseases (IBDs). In MAP- cohort, on the other hand, Coprostanoligenes group was the most influential genera that reduces cholesterol and supports the intestinal barrier. Conclusions The identification of keystone nodes, their co-occurrences, and associations with the exposome (meta data) advances our understanding of biological interactions through which MAP infection shapes the microbiome in MS individuals, suggesting the link to the inflammatory process of IBDs. The associations presented in this study may lead to development of improved diagnostics and effective vaccines for the management of the disease.

Background: In supraclavicular brachial plexus block, to prolong the duration of analgesia, many adjuvants have been tried in the past in many studies but an ideal adjuvant remains yet to be discovered. Dexmedetomidine, a selective Alfa-2 adrenergic agonist when added to local anesthetic has been reported to prolong the block duration and post-operative analgesia in various regional blocks. Aims and Objectives: The aims and objectives are to study the onset and duration of sensory and motor blockade, postoperative analgesia, and hemodynamic effects of addition of dexmedetomidine with ropivacaine in supraclavicular brachial plexus block. Materials and Methods: Sixty patients aged between 18 and 60 years, American Society of Anesthesiologists class I and II, of both sexes, scheduled for upper limb surgery under supraclavicular brachial plexus block were randomly allocated into 2 groups. Group-A received 20 mL of 0.5% ropivacaine in brachial plexus block with 1 μg/kg dexmedetomidine as adjuvant perineurally and Group-B received 20 mL 0.5% ropivacaine in brachial plexus block with dexmedetomidine intravenous infusion at 1 μg/kg over 10 min. Intraoperatively non-invasive blood pressure, heart rate, SpO2, and sedation were recorded every 5 min for the first 10 min and every 15 min thereafter till the end. Time of first rescue analgesic, intensity of postoperative pain, and total analgesic required were recorded. Results: Onset of sensory and motor block was faster in Group-A than Group-B. Duration of analgesia was prolonged in Group-A than Group-B. Hemodynamic stability was better maintained in Group-A than Group-B. Sedation was better in Group B. Conclusion: Dexmedetomidine as adjuvant to ropivacaine in supraclavicular brachial plexus block is more efficacious in providing faster onset of motor and sensory blocks and prolonging duration of postoperative analgesia with better hemodynamic stability.

This study estimates the influence of three significant concepts on the international trade flows between countries participating in the One Belt One Road (OBOR) initiative and Regional Comprehensive Economic Partnership (RCEP). These concepts, which have been previously examined individually in literature, include the effects of intranational trade, international borders and more importantly, regional trade agreements, on international trade flows. This study employs the structural gravity model with data for various variables such as international trade, intranational trade and regional trade agreements (RTAs) ranging between 1999 and 2019. The approach used in this study produces econometrically robust estimates of the impact of RTAs on trade among OBOR member countries, while various sensitivity analyses including phasing-in effect of the RTAs, endogeneity, reverse causality, and multilateral trade resistances. The findings of this study indicate that the impact of RTAs on trade between OBOR and RCEP countries becomes insignificant when the effects of globalization are isolated. Additionally, the declining influence of international borders on trade flows between OBOR and RCEP countries suggests that trade is facilitated to a greater extent between open economies.

Stunting is a serious public health problem, especially among under-fives, which can cause serious short- and long-term impacts. Efforts to tackle stunting in Indonesia involve national strategies and development priorities. Therefore, this study aims to compare the performance of machine learning regression algorithms in predicting stunting prevalence in Indonesia. The data collected is secondary data. The data collection was done carefully, taking explicit details regarding the source, scope, extent, and analysis of the dataset, and using a careful sampling methodology. The model evaluation results show that the Random Forest Regression algorithm has the best performance, with a success rate of 90.537%. The application of this model to the new dataset shows that East Nusa Tenggara province has the highest percentage of stunting at 31.85%, while Bali has the lowest percentage at 12.07%. Visualization of the dashboard using Tableau provides a clear picture of the distribution of stunting in Indonesia. In conclusion, this research contributes to the development of science, especially in the field of machine learning and public health, and provides policy recommendations for tackling stunting in Indonesia.

The prevalence of cotton leaf curl disease (CLCuD) has significantly hampered chili (Capsicum spp.) production, presenting a formidable challenge in Pakistan. During a chili field survey in 2018, distinct symptoms, including stunted growth, yellowing, and severe leaf curling, were observed on several plants. Subsequently, a comprehensive sampling effort was undertaken, collecting a total of 39 symptomatic samples from diverse locations across Multan, Punjab. The DNA extraction from these samples was conducted at the plant virology laboratory at Bahauddin Zakariya University, Multan, marking a crucial step in the investigation of this debilitating disease and its impact on chili production in the region. Molecular analysis with PCR using Av/Ac Core, Beta 01/02, and CLCuMuBF11/R33 primers confirmed begomovirus infection in chili plants. Positive amplification demonstrated a 71.79% infection rate, with 579 bp, 1.4 kb, and 481 bp amplicons for Av/Ac Core, Beta 01/02, and CLCuMuBF11/R33, respectively. Sequencing identified cotton leaf curl Multan beta-satellite (MT668934) infecting the chili plant. Effectively managing these begomoviruses is crucial to curbing their multiplication and protecting vital crops like chili. Addressing the distributions of beta-satellites in agricultural fields, particularly chili, is imperative to prevent further viral spread.

Abstract Background Therapeutic management of locally advanced and metastatic triple negative breast cancer (TNBC) is often limited due to resistance to conventional chemotherapy. Metastasis is responsible for more than 90% of breast cancer-associated mortality; therefore, the clinical need to prevent or target metastasis is immense. The epithelial to mesenchymal transition (EMT) of cancer stem cells (CSCs) is a crucial determinant in metastasis. Doxorubicin (DOX) is the frequently used chemotherapeutic drug against TNBC that may increase the risk of metastasis in patients. After cancer treatment, CSCs with the EMT characteristic persist, which contributes to advanced malignancy and cancer recurrence. The latest developments in nanotechnology for medicinal applications have raised the possibility of using nanomedicines to target these CSCs. Hence, we present a novel approach of combinatorial treatment of DOX with dietary indole 3,3′-diindolylmethane (DIM) which is an intriguing field of research that may target CSC mediated EMT induction in TNBC. For efficient delivery of both the compounds to the tumor niche, advance method of drug delivery based on exosomes sheathed with mesoporous silica nanoparticles may provide an attractive strategy. Results DOX, according to our findings, was able to induce EMT in CSCs, making the breast cancer cells more aggressive and metastatic. In CSCs produced from spheres of MDAMB-231 and 4T1, overexpression of N-cadherin, Snail, Slug, and Vimentin as well as downregulation of E-cadherin by DOX treatment not only demonstrated EMT induction but also underscored the pressing need for a novel chemotherapeutic combination to counteract this detrimental effect of DOX. To reach this goal, DIM was combined with DOX and delivered to the CSCs concomitantly by loading them in mesoporous silica nanoparticles encapsulated in exosomes (e-DDMSNP). These exosomes improved the specificity, stability and better homing ability of DIM and DOX in the in vitro and in vivo CSC niche. Furthermore, after treating the CSC-enriched TNBC cell population with e-DDMSNP, a notable decrease in DOX mediated EMT induction was observed. Conclusion Our research seeks to propose a new notion for treating TNBC by introducing this unique exosomal nano-preparation against CSC induced EMT. Graphical Abstract

The sudden shift to online learning due to the COVID-19 pandemic left many instructors wondering how to minimize anxiety while keeping students engaged in their virtual courses. In this study, we explored (i) specific online instructional tasks that caused students to experience anxiety, (ii) factors that hindered student engagement with online instruction, and (iii) changes in student anxiety and engagement between spring 2020 and fall 2020. Students enrolled in STEM classes were surveyed at the end of spring 2020 (N = 425) and fall 2020 (N = 347) semesters. Our results show that the majority of student respondents had more anxiety in fall 2020 than in spring 2020 with online learning in general, and less anonymous class activities tended to cause the greatest anxiety. Distractions from the environment and personal technologies commonly prevented engagement in both semesters, but no significant differences were observed between the spring and fall. In contrast, more students reported that health-related stress, work-related stress, and issues with technology prevented participation in fall 2020 than in spring 2020. As institutions consider expanding their online course offerings post-pandemic, these data provide valuable insight into the challenges students experienced with online instruction that can inform future pedagogical choices.

Water deficiency in semiarid regions is a limiting factor that affects crop quality and yield. In Punjab, Pakistan, a 27% decline in maize yield was detected over the past two decades just because of water scarcity. Currently, no studies have reported the effects of organically activated biochar (AB) on crop productivity under natural field conditions. For this purpose, a field experiment in a split-split-plot design was conducted with three amendment levels (0, 2, and 4 tonnes ac−1), and three maize hybrids (DK-9108, DK-6321, and Sarhaab) under 100%, 70%, and 50% irrigation water (IW) of crop evapotranspiration (ETc). The AB significantly improved the soil’s physical and chemical properties, and maximum improvement was recorded in 4 tonnes ac−1 AB amendment in organic matter (16.6%), total organic carbon (17%), phosphorus (11.43%), and available potassium (29.27%). The 4 tonnes ac−1AB amendment in soil had a significant impact on total chlorophyll content (0.3–1-fold in DK-6321), carotenoid content (3.9–4.4-fold in Sarhaab), and relative water content (30% and 21% in Sarhaab) under 50% irrigation water (IW) of ETc at V14th and R3 stages, respectively. Moreover, a significant decline in stress markers (proline content and sugar content) was detected at both growth stages in all maize hybrids in AB amended soil. The analysis of plant metabolites indicated increased intensities of phenolics, alkyl esters, and carbohydrates by 2.5–7%, 17–80%, and 40–43% in DK-6321 under 50% IW in 2–4 tonnes ac−1 AB amended soil, respectively. The highest improvement in growth and yield attributes among maize hybrids was detected in the order DK-6321¿DK-9108¿Sarhaab in 2–4 tonnes ac−1AB amended soil under 70% and 50% IW of ETc, respectively. Hence, this research might help to develop an effective soil amendment to restore degraded soils and improve maize growth under arid climatic conditions.

The current understanding of the formation mechanism and kinetics of the σ phase in DSS remains ambiguous. This study investigates the in-situ evolution of the σ phase in DSS through a multi-faceted approach. Distinct σ phase formation mechanisms were identified at 850 °C and 950 °C, with short-range diffusion of eutectoid decomposition at 850 °C and long-range diffusion of nucleation and growth at 950 °C. In-situ X-ray diffraction (XRD) revealed that the lattice parameter of the γ phase peaks at ∼ 600 °C. The thermal expansion coefficient (CTE) of the γ phase is 1.8 times higher than that of the α phase at low temperatures. Interestingly, the convergence of the CTEs of the α and γ phases above 600 °C triggers σ nucleation. The strain field diagram shows maximum impinging strain at the lattice parameter maxima just before the σ formation range. The adverse effects of the σ phase on DSS heat transport properties were also explored, showing an initial increase in thermal conductivity with an increasing γ volume fraction, followed by an 18 % decrease upon σ evolution. First-principle calculations highlight the correlation between Ni enrichment and lattice parameter maxima, supporting these findings.

Introduction: Laryngoscopy and tracheal intubation are powerful noxious stimuli that should be attenuated. The present study is to compare the efficacy of lignocaine nebulisation vs topical lignocaine spray in attenuation of haemodynamic surge in patients undergoing surgery under general anaesthesia. Methodology: After ethical committee clearance, a single blind randomized control study was done on sixty patients of either sex aged between 18-55 years of ASA status I undergoing elective surgery under general anaesthesia with endotracheal intubation. Patients were randomly allocated into 2 groups: GROUP A:received nebulised Lignocaine Hydrochloride 4% at 2 mg/kg. GROUP B: received Topical Lignocaine spray 10% at 2mg/kg. Background: In this era of laparoscopic surgery, with the use of carbon dioxide insufflation in creating pneumoperitoneum for ease of trocar insertion and visualization of anatomy, it comes certain systemic changes in the patient due to the compression effect of the gas as well as resorption of it through peritoneum. Aims and Objectives: The aim of the study was to assess baseline standards of hepatic and renal functional status in pre-operative patients in comparison with post-operative patients undergoing laparoscopic cholecystectomy (LC). Materials and Methods: Cases were chosen among the persons who presented with chronic calculus cholecystitis to the surgical out-patient department of Ramakrishna Mission Seva Pratishthan, Vivekananda Institute of Medical Sciences, Kolkata. Pre-operative liver function tests (LFT) and renal function tests (RFT) values were recorded. Again, LFT and RFT values were evaluated on post-operative day 1 (POD1) and POD12. Results: Serum conjugated and unconjugated bilirubin, aspartate amino transaminase, alanine amino transaminase, urea, estimated glomerular filtration rate values increases immediate postoperatively on POD1 but returns to normal pre-operative levels on POD 12. Serum alkaline phosphatase, albumin, and creatinine values decrease on POD1 and return to normal pre-operative values in POD 12. Conclusion: Hepatic and renal function changes in the immediate post-operative period of 24 h following LC. This change is transient and returns to normal levels within POD 12.

Stress cardiomyopathy was noted to occur at a higher incidence during coronavirus disease of 2019 (COVID-19) pandemic. This database analysis has been done to compare the in-hospital outcomes in patients with stress cardiomyopathy and concurrent COVID-19 infection with those without COVID-19 infection. The National Inpatient Sample database for the year 2020 was queried to identify all admissions diagnosed with stress cardiomyopathy. These patients were then stratified based on whether they had concomitant COVID-19 infection or not. A 1:1 propensity score matching was performed. Multivariate logistic regression analysis was done to identify predictors of mortality. We identified 41,290 hospitalizations for stress cardiomyopathy, including 1665 patients with concurrent diagnosis of COVID-19. The female preponderance was significantly lower in patients with stress cardiomyopathy and COVID-19. Patients with concomitant COVID-19 were more likely to be African American, diabetic and have chronic kidney disease. After propensity matching, the incidence of complications, including acute kidney injury (AKI), AKI requiring dialysis, coagulopathy, sepsis, cardiogenic shock, cases with prolonged intubation of >24 h, requirement of vasopressor and inpatient mortality, were noted to be significantly higher in patients with COVID-19. Concomitant COVID-19 infection was independently associated with worse outcomes and increased mortality in patients hospitalized with stress cardiomyopathy.

Silver nanoparticles (AgNPs) are known for their large surface area, which enhances their energy and antimicrobial activity. This study focused on producing silver nanoparticles using leaves from the Beta maritima (vulgaris) plant. Various techniques such as Fourier transform infrared (FT-IR) spectroscopy, ultraviolet–visible (UV–vis) spectroscopy, energy-dispersive X-ray spectroscopy (EDX), scanning electron microscopy (SEM), and X-ray diffraction analysis (XRD) to characterize the synthesized AgNPs. The biologically synthesized AgNPs were then evaluated for their antimicrobial (antibacterial and antifungal) and antioxidant properties using different agents such as hydrogen peroxide (H2O2), 1,1-Diphenyl-2-picrylhydrazyl (DPPH), 2,2 azobis, 3-ethyl benzothiazoline-6-sulphonic acid (ABTS), and phosphomolybdate. The results showed that the AgNPs exhibited strong antifungal activity, particularly against fluconazole-resistant strains of Aspergillus niger and Aspergillus flavus, with a 61 % inhibition rate. Moreover, the AgNPs demonstrated significant antibacterial activity against various strains. At a concentration of 40 µg/mL, they exhibited 26 % inhibition against Staphylococcus aureus, 22 % against Micrococcus luteus, 18 % against E. coli, and 24 % against Klebsiella pneumonia, outperforming clarithromycin antibiotics. SEM analysis revealed spherical AgNPs, sized using nano measurer 1.2, EDX showed the presence of silver having composition at 80.38 %, and XRD confirmed a face-centered cubic crystalline structure. In addition to their characterization, the AgNPs also displayed strong antioxidant activity. At the same concentration, they showed percentages of 80.78 % (DPPH), 85.38 % (H2O2), 81.78 % (ABTS), and 80.66 % (phosphomolybdate), surpassing the antioxidant activity of the plant leaf extract alone. The synthesis of AgNPs using the extract of the Beta maritima (vulgaris) plant offers several advantages compared to other methods, including ease of handling, cost-effectiveness, wide availability, and eco-friendliness.

Introduction Despite the evidence against drain placement after thyroidectomy, there is a lack of consensus on drain use in patients with substernal goiter.

Introduction: Curcuma spp. is renowned for its therapeutic properties and holds prominence in traditional Chinese medicine (TCM). Popularly, the plant is referred to as Ézhú (莪术) in China. Traditional Chinese medicine holds enormous potential in addressing several diseases, including Diabetes Mellitus (DM), with a multipronged approach. The present study evaluates the anti-diabetic potency of Curcuma amada Roxb. While C. amada is not explicitly cited in traditional Chinese medicine (TCM) texts, TCM integrates various Curcuma spp. with comparable properties for their potential health benefits. Methods: In-vitro α-amylase and α-glucosidase enzyme inhibition was performed on bioactivity-guided fractions of crude ethyl acetate C. amada rhizome extract to determine the most bioactive fraction. Furthermore, a 2-NBDG (2-(7-Nitro-2,1,3-benzoxadiazol-4-yl)-d-glucosamine) mediated glucose uptake assay was carried out through a flow cytometric approach to corroborate the anti-diabetic property of the lead fraction. GC-MS-based metabolomic profiling was considered to determine the possible bioactive phytochemicals in the most effective bioactive fraction. Extensive in-silico computational and bioinformatics studies, including molecular dynamic simulations, were performed to depict the possible mechanism behind this anti-diabetic efficacy. Results: The comprehensive in-vitro enzyme inhibition screening of fractions identified fraction 43 as the most potent anti-diabetic fraction, corroborated by glucose uptake studies. An extensive in-silico molecular docking study against 50 target receptors associated with DM, with the 21 unique phytocompounds obtained through the GC–MS-based metabolomic profiling study, identified neohesperidin as the most potent bioactive phytochemical. Computational chemistry employing DFT (Density Functional Theory) analysis identified neohesperidin as the strongest chemically reactive compound species in the most bioactive fraction. The molecular dynamic study highlighted that neohesperidin possibly remediated diabetes through TNFα (Tumor Necrosis Factor α) and IGF1R (Insulin-like Growth Factor 1 Receptor) pathways, as represented by the stable dynamic simulations, RMSD (Root Mean Square Deviation), and δG (Gibbs Free Energy) values. Discussion: The in-vitro enzyme inhibition, glucose uptake, in-silico computational and bioinformatics studies reveal that neohesperidin resourced from Curcuma amada Roxb. has enormous potential as a future anti-diabetic drug.

Energy harvesting systems, including piezoelectric (PENG), triboelectric (TENG), and pyroelectric (PYNG) nanogenerator technologies, have emerged as one of the major future energy solutions. Energy harvesting eliminates the need for conventional batteries and encourages eco-friendly alternatives. This study reports hydrothermally synthesized BaTiO3 (BTO) particles with a tetragonal symmetry for hybrid energy harvesting. BTO particles are incorporated with PDMS at various wt% to form a flexible composite film. The 15 wt% BTO-PDMS composite/Al hybrid device (PENG-TENG) produces a peak voltage of 100 V, a current of 980 nA, and a charge of 17 nC, generating a peak power output of 33.64 μW at 100 MΩ. Furthermore, integrating this HNG (external hybridization) yielded an output of 101 V and 980 nA, demonstrating practical applicability. HNG is also employed to interact by touching various objects at different temperatures. The pyroelectric behavior of BTO allows direct thermal sensing of the object. The signals produced are processed using a convolutional neural network (CNN)-based object recognition system, which achieved a remarkable classification accuracy of 99.27% for various objects. External hybridization improves energy efficiency, representing a huge step forward in sustainable technology applications. This research paves the way for developing hybrid energy harvesters and can be employed further for extremely precise battery-free object recognition systems. This unique hybrid nanogenerator, which combines pyroelectric, piezoelectric, and triboelectric components, represents a new method of self-powered object detection. External hybridization improves energy efficiency, representing a huge step forward in sustainable technology applications.

Background: Information technology (IT) is a new way of teaching and learning. One of the promising media of information technology is e-Learning, which is used to enhance knowledge and skills among users. A student gains better and deep knowledge through a useful tool. This survey aimed to determine practices among medical students for e-Learning. Methods: This cross-sectional survey was conducted on 184 students amongst the 500 students currently enrolled in medical college. Data was collected using questionnaires and were analyzed through SPSS version 22. Chi-square was used for qualitative values. Results: Majority 90.80% (n=167) students were aware of e-Learning and were statistically high in first year students (p-value: 0.018). The student did not show statistically significant results for content learned through e-Learning with a p-value of 0.063. Different resources were used for e-Learning in which videos had the highest percentage (87.60%) and audios were used as the least resource for e-Learning (29.20%). Daily, 56% of the students use e-Learning for 1 hour or less and only 3% of the students used it for more than 4 hours. Conclusion: Majority of undergraduate medical students were aware of the use of e-Learning and most of them preferred e-Learning for their course work and studies showing a significant increase in understanding and use, compared to studies conducted earlier. Gastrointestinal stromal tumors are commonly developed in gastrointestinal tract but in duodenum is less than 3% of all gastrointestinal neoplasm. The early diagnostic criteria are very difficult in these tumors in worldwide and being a challenge for medical team due to difficult anatomical structure of duodenum. The study’s aim is to know the incident rate of duodenal gastrointestinal stromal tumors. The average age of these tumors is more than 40 years but in the current case, 19-years old female patient present with complaint of melena and anemia. Her family history was negative. She was diagnosed by scope test of oral gastric duodenum with biopsy, abdominal x-rays, computerized tomography scan and histopathology. The patient was treated by Classic Whipple’s procedure. According to the oncologist, patient has low risk of duodenal tumor from gastrointestinal stromal tumor. These tumors have no need to treat with adjuvant therapy: chemotherapy (Glavic) and radiation therapy after surgical resection.

Objective: To explore the therapeutic effects of the novel wild edible mushroom Astraeus hygrometricus (Pers.) Morgan (A. hygrometricus) on human acute lymphoblastic leukemia cells. Methods: Extensive screening of the antiproliferative and chemopreventive potential of different extracts from 5 wild mushrooms, A. hygrometricus, Phallus sp., Lentinus sp., Tricholoma sp., and Serpula sp. was performed against a panel of 6 cancer cell lines and normal cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Apoptosis determination, cell cycle profiling, intracellular reactive oxygen species (ROS) and reactive nitrogen species (RNS), and mitochondrial membrane potential were analyzed by flow cytometry. The activity of caspases was measured colorimetrically, and the expression pattern of mitochondrial proteins was analyzed. Results: The methanol extract of A. hygrometricus and MOLT-4 cells were identified as the most potent extract exhibiting antiproliferative activity and most sensitive cell line, respectively. The mushroom extract induced robust selective apoptosis in MOLT-4 cells and arrested cell cycle progression at the G0/G1 stage. The extract disrupted the mitochondrial membrane potential and enhanced ROS production in MOLT-4 cells. The methanol extract induced apoptosis by downregulating the expression of Bcl-2, increasing the expression of Bax, and activating the caspase cascade. Conclusion: The novel wild edible mushroom is a potential repository of biomolecules for the development of antileukemic drugs.
