The potential effects of combining biochar with hyperaccumulator plants on heavy metal stabilization and uptake in Cd and Pb-contaminated soils have received limited attention. This study used biochar (BC) at levels ranging from 0 to 40 t ha-1 as soil amendments in Cd and Pb contaminated rice paddy soil. Biochar significantly decreased soil bioavailable Cd and Pb concentrations 6.7 to 16.6% and 10.6 to 39.7%, respectively, compared to the control. The shoot and root Cd concentrations in Arabidopsis helleri increased significantly by 33.5% and 133.1%, respectively, in the C3 treatment compared to the control. Similarly, shoot Pb concentrations showed a 57.5% increase compared to the control, but no significant changes were observed in root Pb concentrations. The Arabidopsis helleri bioconcentration factor (BCF) saw an increase of up to 40.5% for Cd and 57.8% for Pb with the C3 treatment. Conversely, the Cd translocation factor (TF) decreased 42.8 to 49.1%, while the Pb TF increased 32.8 to 96.6% with biochar application. The majority of Arabidopsis helleri-biochar Cd and Pb was found in the B3 fraction (organic fraction), constituting over 50%, and even over 80% for Cd. The Arabidopsis helleri-biochar primarily contained organic char binding Cd and exhibited slow-release characteristics.

Sustainable crop production systems should promote large and diverse soil microbial communities to enhance biological soil processes rather than depend solely on chemical interventions that include pesticide applications. Crop rotation increases above-ground temporal diversity which, relative to monoculture, usually increases soil microbial diversity. But comparisons between short and long crop rotations that also include pesticide effects are rare. A 5-yr (2013-2017) field study was conducted to investigate crop rotation and fungicide effects on the soil microbiome and activity. There were nine rotations, with or without fungicide applications, that included four 2-yr rotations (wheat preceded by canola, barley, pea, or flax), four 3-yr rotations where barley or canola were added to the 2-yr rotations, and one rotation where canola and wheat were stacked (canola-canola-wheat-wheat). In 2017, soil microbial biomass, composition, diversity and enzyme activities were measured in the rhizosphere of the final wheat crop in each rotation. Fungicides reduced fungal richness (the number of different fungal taxa) in the wheat rhizosphere (e.g., Chao1 indices of 64.0 vs. 79.9) especially in 2-yr rotations, but rotation length/type and the crops that preceded wheat had different effects on different taxa. Two of the three most predominant prokaryotic phyla, Proteobacteriota and Actinobacteriota, responded differently to rotation length: 3-yr rotations enriched the former (27.4% vs. 20.1% relative abundances), but 2-yr rotations enriched the latter (19.9% vs. 28.3% relative abundances). Relative to oilseed crops preceding the sampled wheat, a field pea preceding crop enriched Actinobacteriota (31.7% vs. 24.8% relative abundances) and the most abundant fungal class, Sordariomycetes (39.1% vs. 22.1% relative abundances), in addition to increasing microbial biomass carbon (MBC) and arylsulphatase activity by 33% and 57%, respectively. Correlations of the relative abundances of fungal or prokaryotic genera with β-glucosidase and arylsulphatase activities were similar (both positive or negative), but they were the opposite of correlations with acid phosphomonoesterase, suggesting a close link between C and S cycling. Besides the nutrient cycling implications of these soil microbial characteristics, there is need to study their biological disease control significance.

Although herbicides have been a dominant and effective weed control strategy for decades in Western Canada, herbicide resistance and the lack of new modes of action have resulted in weed management challenges. Integrated weed management strategies have been shown to be successful in controlling certain weed species that are problematic in cropping systems. The objective of this study was to investigate integrated weed management strategies that have been successful on individual species to determine their applicability to a multiple weed species that may coexist in a field. In addition, harvest weed seed control was incorporated into these integrated weed management strategies to determine its impact in western Canadian cropping systems. A 5-year rotational study was conducted from 2016 to 2020 at Beaverlodge, Lacombe, and Lethbridge, AB; Scott and Saskatoon, SK; and Carman, MB, that incorporated integrated weed management strategies such as rotational crop diversity (including winter annuals and perennials), increased seeding rates, crop silaging, chaff collection, and with or without in-crop herbicides. This research confirmed success in managing some species of weeds such as wild oat when increased seeding rates, 2 years of early cut silage barley, and competitive winter cereals were incorporated into a cropping system, even when no in-crop herbicides were applied. However, some weed growth morphologies (e.g., twining weeds) or life cycles (e.g., facultative winter annuals) were not managed successfully with this combination of strategies. Chaff collection provided incremental weed control benefits but did not serve as a replacement for herbicidal weed control. Weed densities had an apparent impact on the success of these integrated weed management strategies, suggesting that the sooner they are adopted, the more likely they are to be successful at maintaining or reducing weed densities. This study not only showed the ability to reduce reliance on herbicides with strategies that can be effective in Western Canada but also highlighted the need for further understanding of different weed species and their responses to integrated weed management strategies, as well as the complexity of managing a weed community with integrated weed management.

We present a case of a splenic artery (SA) aneurysm (SAA) that had arisen abnormally from the superior mesenteric artery in a 63-year-old man who underwent successful endovascular treatment. Although SAAs characterized by this anatomic abnormality are rare, in all 46 reported cases, the SAAs were located at the root of the SA and had originated abnormally from the superior mesenteric artery. This location is different from that of orthotopic SAAs, which are mostly located in the distal third of the SA. The differences in hemodynamics due to the anatomic abnormalities might play an important role in the formation of the anomalous SAAs.

This study focused on a carbonation treatment that immobilizes trace elements such as lead (Pb) in incineration bottom ash (IBA) using the exhaust gas and the refined carbon dioxide generated (CO2) recovered at the Japan's first incineration facility, which recovers CO2 in the gaseous form. This study describes (1) the bearing capacity of IBA from the municipal solid waste (MSW) obtained through the California bearing ratio (CBR) test to investigate the possibility of using IBA as a base course material, (2) mechanical characteristics of IBA using the cone index test, to investigate the possibility of using as an embankment material, and (3) leaching properties of IBA from MSW obtained through a single batch leaching test and the long-term outdoor exposure of the leaching test to evaluate the effect of carbonation treatments. Results reveal that there was no effect of trace components not included in the separated and recovered CO2, and that the effect of carbonation could be obtained even if the exhaust gas was used directly. The carbonated IBA can be applied as a sub-base course material based on its mechanical properties, regardless of the carbonation conditions. In addition, carbonated IBA can be used as an embankment material owing to its sufficient cone index value. Regarding the heavy metal and metalloid leaching behavior, it was revealed that all the IBA samples satisfied the soil environmental standards set by the Ministry of the Environment, Japan, except for Cr (VI). The long-term leaching characteristics of carbonated IBA showed that Pb could be immobilized over a long period.

Abstract Background Creating a box lesion in the posterior wall of the left atrium from the epicardial side of the beating heart remains a challenge. Although a transmural lesion can be created by applying radiofrequency (RF) energy at clampable sites, it is still difficult to create a transmural lesion at unclampable sites because the inner blood flow in the unclampable free wall weakens the thermal effect on the outside. Our aim was to apply the newly developed infrared coagulator to create linear transmural lesions on the beating heart thoracoscopically to treat atrial fibrillation (AF). Case presentation A 71-year-old male was referred to our hospital with a diagnosis of hypertrophic cardiomyopathy and permanent atrial fibrillation. The patient was first diagnosed with atrial fibrillation 20 years before. Direct current cardioversion had been performed every few years a total of four times, but sinus rhythm restoration had always been temporary. On February 27, 2020, thoracoscopic PV isolation together with infrared roof- and bottom-line ablation to create a box lesion and left atrial appendage amputation (LAAA) were performed. The coagulator could be applied to clinical thoracoscopic surgery to successfully create a box lesion without any complication. The patient restored a regular sinus rhythm, it has been maintained for eleven months, and there have been no adverse events. Conclusions The infrared coagulator might have enough potential to create transmural lesions on the beating heart in thoracoscopic AF surgery.

Aim: Maternal high-fat diet (HFD) is associated with the development of cardiovascular disease (CVD) in adult offspring. Atherosclerotic vascular calcification is well documented in patients with CVD. We examined the effect of maternal HFD on calcified plaque formation. Methods and results: Seven-week-old female apo-E−/− mice (C57BL6/J) were nourished either an HFD or a normal diet (ND) a week before mating, and during gestation and lactation. Offspring of both the groups were fed a high-cholesterol diet (HCD) from 8 weeks of age. Osteogenic activity of the thoracic aorta, assessed using an ex vivo imaging system, was significantly increased after 3 months of HCD in male offspring of HFD-fed dams (O-HFD) as compared with those of ND-fed dams (O-ND). Alizarin-red-positive area in the aortic root was significantly increased after 6 months of HCD in male O-HFD as compared to that of O-ND. Plaque size and Oil Red O-positive staining were comparable between the two groups. Primary cultured vascular smooth muscle cells (VSMCs) of the thoracic aorta were treated with phosphate and interleukinL-1β (IL-1β) to transform them into an osteochondrocytic-like phenotype. Intracellular calcium content and alkaline phosphatase activity were markedly higher in the VSMCs of O-HFD than in O-ND. IL-1β concentration in the supernatant of bone marrow-derived macrophages was markedly higher in O-HFD than in O-ND. Conclusion: Our findings indicate that maternal HFD accelerates the expansion of atherogenic calcification independent of plaque progression. In vitro phosphate- and IL-1β-induced osteochondrocytic transformation of VSMCs was augmented in O-HFD. Inhibition of VSMCs, skewing toward osteochondrocytic-like cells, might be a potential therapeutic strategy for preventing maternal HFD-associated CVD development.

Agroecosystems are comprised of environmental compartments where associated microbial communities interact with one another. These microbial communities, called microbiomes, inhabit livestock, insects, and plants. Microbiomes are also present in the soil and watersheds. Clarifying the nature and extent of microbial interactions between compartments both at intra-farm and global scales can promote sustainable production systems, healthier animals, increased crop yields, and safer meat products. Early research on microbiomes was hindered by a lack of expertise and the high cost of molecular sequencing. However, these limitations have been largely resolved with advances in and reduced costs of sequencing technologies. In this paper, we summarize sequencing and bioinformatics approaches, and review the crucial roles of diverse microbiomes in livestock, plants and soil, as well as pollinators and pest insects. These crucial roles include nutrient cycling, nutrient acquisition, metabolism of toxins and enhanced host immune function. Additionally, we examine potentially undesirable effects of microbiomes associated with climate change and agri-food production such as their role in the release of greenhouse gases from cattle and their impact on meat safety and spoilage. By increasing the awareness of microbiomes and the growing ease with which they can be studied, we hope to foster a greater adoption of microbiome research. Further understanding of the diverse effects and interactions of microbiomes will advance our efforts to increase agricultural production while reducing its negative environmental footprint, thus making the agroecosystems more sustainable.

Depression is an independent risk factor for cardiovascular disease and is significantly associated with the prevalence of abdominal aortic aneurysm (AAA). We investigated the effect of repeated social defeat (RSD) on AAA development. Eight-week-old male wild-type mice were exposed to RSD by being housed with larger CD-1 mice in a shared cage. They were subjected to vigorous physical contact. After the confirmation of depressive-like behavior, calcium chloride was applied to the infrarenal aorta of the mice. At one week, AAA development was comparable between the defeated and control mice, without any differences being observed in the accumulated macrophages or in the matrix metalloproteinase activity. At two weeks, the maximum diameter and circumference of the aneurysm were significantly increased in the defeated mice, and a significant decrease in periaortic fibrosis was also observed. Consistently, the phosphorylation of the extracellular signal-regulated kinase and the incorporation of 5-bromo-2′-deoxyuridine in the primarily cultured aortic vascular smooth muscle cells were significantly reduced in the defeated mice, which was accompanied by a substantial increase in mitogen-activated protein kinase phosphatase-1 (MKP-1). The MKP-1 mRNA and protein expression levels during AAA were much higher in the defeated mice than they were in the control mice. Our findings demonstrate that RSD enhances AAA development by suppressing periaortic fibrosis after an acute inflammatory response and imply novel mechanisms that are associated with depression-related AAA development.

Background/aims We assessed the associations between retinopathy of prematurity (ROP) and continuous measurements of oxygen saturation (SpO2), and developed a risk prediction model for severe ROP using birth data and SpO2 data. Methods This retrospective study included infants who were born before 30 weeks of gestation between August 2009 and January 2019 and who were screened for ROP at a single hospital in Japan. We extracted data on birth weight (BW), birth length, gestational age (GA) and minute-by-minute SpO2 during the first 20 days from the medical records. We defined four SpO2 variables using sequential measurements. Multivariate logistic regression was used to develop a model that combined birth data and SpO2 data to predict treatment-requiring ROP (TR-ROP). The model's performance was evaluated using the area under the receiver operating characteristic curve (AUC). Results Among 350 infants, 83 (23.7%) required ROP treatment. The SpO2 variables in infants with TR-ROP differed significantly from those with non-TR-ROP. The average SpO2 and high SpO2 showed strong associations with GA (r=0.73 and r=0.70, respectively). The model incorporating birth data and the four SpO2 variables demonstrated good discriminative ability (AUC=0.83), but it did not outperform the model incorporating BW and GA (AUC=0.82). Conclusion Data obtained by continuous SpO2 monitoring demonstrated valuable associations with severe ROP, as well as with GA. Differences in the distribution of average SpO2 and high SpO2 between infants with TR-ROP and non-TR-ROP could be used to establish efficient cut-off values for risk determination. From 2012 onward, judo has been a compulsory element in physical education lessons in junior high school in Japan. However, because judo is only introduced in junior high school, it is not part of the systematic curriculum that considers the stages of child development from elementary school on does not exist, unlike other sports, such as apparatus gymnastics and swimming, to which students are exposed in elementary school. Therefore, this study seeks to develop a set of teaching materials that incorporate judo games teaching basic judo moves into the physical education lessons of lower-elementary-school students (7-year-olds), and evaluate its usefulness in ensuring systematic progression between physical education lessons in elementary school and junior high school. The practical implementation of these lessons took place at an elementary school affiliated with a national university in Kagoshima Prefecture. The target students were 18 boys and 17 girls, for a total of 35 second-grade students. Six lessons were planned and implemented in February 2020. When evaluating the usefulness of the teaching materials developed, objective evaluation and subjective evaluation were considered together with tests of physical skills and motor ability. The results of this study show that in formative evaluation, used as a form of objective evaluation (Takahashi, 2003), there was an increase in assessment scores for the three elements--outcome, learning method, and cooperation--together with the total assessment scores. When students were asked to subjectively evaluate themselves in seven categories, including showing more awareness of their own bodies, adjusting the force used, and showing more awareness of other people's bodies, six categories received higher scores in the second half of the teaching unit. In the test of physical skills and motor ability, at the end of the sixth lesson, both male and female students achieved higher scores in all three categories tested. Thus, the teaching materials developed in this study, which incorporate judo games that teach basic judo movements, have a strong possibility of being useful for physical education lessons targeted at 7-yearolds. The learning content developed in this study can be used beyond physical education lessons as it is generic and practical enough to be used as an instructional program in judo clubs targeted at children of similar age.
