Een stap naar meer uniformiteit in het UBO-begrip: Nieuwe Europese antiwitwasregels zorgen voor een uitbreiding van verplichtingen voor overheden en bedrijven. Eén van de wijzigingen betreft de definitie van de uiteindelijk belanghebbende (UBO) en de registratie van UBO's in het UBO-register. Voor de wetgever, rechters, toezichthouders, Wwft-instellingen (zoals banken, advocaten, notarissen en accountants) is het van belang kennis te nemen van de fundamentele wijzigingen op dit gebied. Galacto-oligosaccharides (GOS) are non-digestible oligosaccharides characterized by a mix of structures that vary in their degree of polymerization (DP) and glycosidic linkage between the galactose moieties or between galactose and glucose. They have enjoyed extensive scientific scrutiny, and their health-promoting effects are supported by a large number of scientific and clinical studies. A variety of GOS-associated health-promoting effects have been reported, such as growth promotion of beneficial bacteria, in particular bifidobacteria and lactobacilli, inhibition of pathogen adhesion and improvement of gut barrier function. GOS have attracted significant interest from food industries for their versatility as a bioactive ingredient and in particular as a functional component of infant formulations. These oligosaccharides are produced in a kinetically-controlled reaction involving lactose transgalactosylation, being catalyzed by particular β-galactosidases of bacterial or fungal origin. Despite the well-established technology applied for GOS production, this process may still meet with technological challenges when employed at an industrial scale. The current review will cover relevant scientific literature on the beneficial physiological properties of GOS as a prebiotic for the infant gut microbiota, details of GOS structures, the associated reaction mechanism of β-galactosidase, and its (large-scale) production.

Pertussis is a respiratory infection caused by the Gram-negative bacterium Bordetella pertussis . Despite high vaccination coverage this disease remains a public health concern worldwide. A better understanding of the protective immune responses to B. pertussis is required for the development of improved vaccines. The aim of this study was to determine the production of reactive oxygen species (ROS) by human neutrophils in response to B. pertussis and to determine the contribution of opsonizing antibodies from convalescent pertussis patients in this response. The serum samples from convalescent patients were taken at <3, 9, 18 and 36 months after diagnosis of pertussis. Also included were sera from healthy age-matched controls. We show that neutrophils produced high levels of ROS in response to opsonized, compared to non-opsonized, B. pertussis and that this effect was independent of the time the convalescent serum samples were taken. This indicates the presence of functional opsonizing antibodies up to 3 years after B. pertussis infection. While opsonization of B. pertussis with serum samples from uninfected controls also induced ROS production, sera from infected individuals induced significantly higher ROS levels. Spearman correlations analysis showed that IgG antibodies targeting fimbriae3 followed by pertactin, and BrkA correlate with ROS production. Additionally, we observed that neutrophils killed opsonized B. pertussis in a ROS-dependent manner. We AI researchers are concerned about the potential impact of artificially intelligent systems on humanity. In the first half of this essay, I argue that ethics is an evolved body of cultural knowledge that (among other things) encourages individual behavior that promotes the welfare of the society (which in turn promotes the welfare of its individual members). The causal paths involved suggest that trust and cooperation play key roles in this process. In the second half of the essay, I consider whether the key role of trust exposes our society to existential threats. This possibility arises because decision-making agents (humans, AIs, and others) necessarily rely on simplified models to cope with the unbounded complexity of our physical and social world. By selecting actions to maximize a utility measure, a well-formulated game theory model can be a powerful and valuable tool. However, a poorly-formulated game theory model may be uniquely harmful, in cases where the action it recommends deliberately exploits the vulnerability and violates the trust of cooperative partners. Widespread use of such models can erode the overall levels of trust in the society. Cooperation is reduced, resources are constrained, and there is less ability to meet challenges or take advantage of opportunities. Knowledge of the functional roles and interspecies interactions are crucial for improving our understanding of the human intestinal microbiome in health and disease. However, the complexity of the human intestinal microbiome and technical challenges in investigating it pose major challenges. In this proof-of-concept study, we rationally designed, assembled and experimentally tested a synthetic Diet-based Minimal Microbiome (Db-MM) consisting of ten core intestinal bacterial species that together are capable of efficiently converting dietary fibres into short chain fatty acids (SCFAs). Despite their genomic potential for metabolic competition, all ten bacteria coexisted during growth on a mixture of dietary fibres, including pectin, inulin, xylan, cellobiose and starch. By integrated analyses of metabolite production, community composition and metatranscriptomics-based gene expression data, we identified interspecies metabolic interactions leading to production of key SCFAs such as butyrate and propionate. While public goods, such as sugars liberated from colonic fibres, are harvested by non-degraders, some species thrive by cross-feeding on energetically challenging substrates, including the butyrogenic conversion of acetate and lactate.
