Abstract

Multiple myeloma (MM), the seco nd most common haematological malignancy, is currently incurable because patients often develop multiple drug resistance and experience subsequent relapse of the disease. This study aims to identify a potential therapeutic agent that can counter bortezomib (BTZ) resistance in MM. DCZ0358, a novel alkaloid compound, is found to exert potent cytotoxic effects against BTZ-resistant MM cells in vivo and in vitro. The anti-myeloma activity of DCZ0358 is associated with inhibition of cell proliferation, promotion of cell apoptosis via caspase-mediated apoptotic pathways, and induction of G0/G1 phase arrest via downregulation of cyclin D1, CDK4, and CDK6. Further investigation of the molecular mechanism shows that DCZ0358 suppresses the JAK2/STAT3 signaling pathway. In conclusion, DCZ0358 can successfully counter BTZ resistance in MM cells. This study provides evidence that warrants future preclinical assessments of DCZ0358 as a therapeutic agent against BTZ resistance in MM.

Abstract

Streptococcus agalactiae (GBS) is an important Gram-positive pathogen of fish aquaculture worldwide. In this study, we performed a comparative analysis of GBS S03 (serotype III) and S07 (serotype Ia) isolated from Schizothorax spp.. We explored the association between phenotypic antibiotic resistance, virulence, and genomic characteristics. Antimicrobial sensitivity tests on 12 common antibiotics using the disc diffusion method revealed that the GBS S03 resisted seven antibiotics, while GBS S07 showed sensitivity to eleven antibiotics. Pathogenicity analysis demonstrated greater virulence of GBS S07 than S03. Then, the occurrence of antibiotic resistance and virulence genes were identified using the whole-genome sequence (WGS) of GBS S03 and GBS S07. Besides just GBS S03 had gyrA and parC gene mutations, there were mefE, tetO, lnuB, lsaE, APH3', and sat-4 resistance genes present only in GBS S03 genome. In addition to 51 virulence genes in both GBS S03 and S07 genomes, GBS S07 carried virulence genes associated with invasion, such as SAN1519, rfbA and cylE_ genes. There was complete concordance between genotypic evidence and phenotypic characteristics. Virulence factors and phylogenetic analysis showed that GBS S03 and human sources shared an extremely close evolutionary relationship. Our findings also provide important proof for using WGS as an effective tool for phenotypic predictions of GBS.

Abstract

Gut health is essential for animal growth because it can absorb nutrients more efficiently, promote growth, and protect the body from invasion by pathogenic microorganisms. Temperature, dissolved oxygen, water quality, nutrition level, pathogen, net fishing, and so on will affect the gut health of aquatic animals. Intestinal diseases in aquatic animals were often not easily detected, making them difficult to prevent and control. Therefore, establishing a stable and scientific evaluation standard of gut health was of great significance. This study evaluated the effects of gross pathological and histological parameters of largemouth bass and channel catfish infected by Aeromonas hydrophila. A total of 150 largemouth bass and 150 channel catfish were randomly divided into 5 groups: 1 control group (NC) and 4 challenge groups (CH, fish were infected with A. hydrophila by intraperitoneal injection with a concentration of 200 µL 1x108 CFU/mL, 1x107 CFU/mL, 1x106 CFU/mL, and 1x105 CFU/mL, respectively). After the challenge, largemouth bass' gross pathological and histological changes were observed on the 2nd, 6th, 9th, 12th, and 14th days. In contrast, channel catfish's gross pathological and histological changes were observed on the 3rd, 6th, 9th, 12th, and 15th day. In this study, regarding the “I See Inside” (ISI) methodology of poultry, a pathological evaluation method of fish intestinal health was established to comprehensively evaluate the intestinal health status of fish by gross pathological and histopathological score. In gross pathology, it was scored by the body surface and intestinal lesions. In histology, it was a metric evaluation of histological alterations in the intestine, converting macroscopic and microscopic alterations into numbers and then scoring. 27. Visible Light‐Responsive Crystalline B←N Host Adducts with Solvent‐Induced Allosteric Effect for Guest Release.

Abstract

Photo‐responsive organic crystals, capable of converting light energy into chemical energy to initiate conformational transitions, present an emerging strategy for developing lightweight and versatile smart materials. However, visible light‐triggered tailored guests capture and release behaviors in all‐organic solids are rarely reported. Here, we introduce a photoreactive crystalline boron‐nitrogen (B←N) host adduct with the ability to undergo [2+2] photocycloaddition upon 447 nm light exposure. This process facilitates single‐crystal‐to‐single‐crystal (SCSC) photodimerization in the mother liquor, maintaining the original B←N host structure. Weakened intermolecular interactions within the photodimer host contribute to fast guest release in air under irradiation. Furthermore, the dynamic B←N bonds enable reversible transformations between organic host adducts and adduct cocrystals under the solvent‐induced allosteric effect. As a result, four B←N host adduct crystals containing individual alkane guest are easily obtained and exhibited the ability of photo‐controlled alkane release. Therefore, the integration of photo reactivity and structural transformation within B←N host adduct enables customized capture and release of guest molecules. [ABSTRACT FROM AUTHOR]
