In this paper, a simple formula is derived for the modal damping ratio of the bridge using the correlation between the instantaneous amplitudes of the related front and rear contact responses of a two-axle test vehicle by the Hilbert transform (HT). To start, closed-form solutions were derived for the dynamic response of the damped bridge and vehicle-bridge contact responses. Next, the HT was employed to generate the instantaneous amplitudes of the two contact points. Based on their correlation, a simple formula is derived for the bridge damping ratio. Finally, the reliability of the derived formula was verified in the numerical study. It was demonstrated that the proposed formula can be successfully used to determine the first bridge damping ratio, even in the presence of rough pavement, but with the aid of random traffic. Background: Common bean (Phaseolus vulgaris L.) is a major food legume with high nutritional value and economic importance globally. A remarkable attribute of this plant is its propensity to generate adventitious roots from petioles of detached leaves when moisture is appropriate. This distinctive feature presents promising prospects for harnessing petioles in the facilitation of transgenic hairy root production. Methods: We achieved the successful induction of transgenic hairy roots from common bean petioles through the utilization of Agrobacterium rhizogenes strain K599. Our experimentation encompassed a diverse array of common bean varieties and leaves of varying ages. Subsequently, we refined and optimized the procedure for hairy root induction. Result: Our investigations have revealed that the most conducive conditions for hairy root transformation with K599 are attained using 5-day-old leaves from the cultivar ‘Honghuaqingjia’, exhibiting a remarkable induction efficiency of 59%. The usefulness of this system was demonstrated through a subcellular localization analysis of the transcription factor PvTCP2 protein in combination with GFP (Green Fluorescent Protein). This paper investigates the use of straight-beam elements to simulate the vibration of curved beams that is mathematically more challenging. First, analytical solutions of the out-of-plane and in-plane dynamic responses of the curved beam are derived for reference. Then, for the straight beam approach, the elastic stiffness matrix, consistent mass matrix, consistent nodal loads, and transformation matrix are derived and included in a procedure for computing the dynamic responses of the curved beam. The effectiveness of the straight beam approach in analyzing the free vibration and forced vibration (induced by moving vehicle) of the curved beam is validated, with the results compared with the theoretical solutions and those by Abaqus. It can be concluded that the straight beam approach is a simple and efficient means for analyzing the out-of-plane and in-plane vibrations of curved beams with high precision. [ABSTRACT FROM AUTHOR]

Real-time hybrid simulation (RTHS) is an economical and reliable method for the evaluation of structural dynamic performances, and the fixed analytical substructure model is often used in RTHS which may affect the accuracy of results. In this study, a real-time hybrid simulation platform (RTHSP) developed by configuring a generic National Instruments (NI) controller with hybrid programming strategy is presented in detail. The dynamic performances of a scaled base isolated structure, where the unscented Kalman filter (UKF) was used to update the analytical substructure Bouc–Wen model during the RTHSs was evaluated by presented RTHSP. RTHS of a base-isolated structure was performed where a lead rubber bearing (LRB) was tested physically as the experimental substructure of a part of the isolation layer and the superstructure with the rest of the isolation layer model updated by UKF was considered as the analytical substructure. Under the excitation of three natural earthquakes, the RTHSs with and without UKF updating were compared and analyzed the differences between the two. The results indicated that the displacements of experimental substructure generated by RTHS with UKF updating are the largest, while the relative displacements and acceleration of superstructure are the smallest overall, and the dynamic characteristics of the isolation layer of the analysis substructure updated by UKF are different from that without updated, which reflected the more authentic dynamic mechanical performance of the base-isolated structure under earthquake excitation. In addition, the RTHSP and the hybrid programming strategy are verified to be reliable in tests and experiments, and the components and implementation of a RTHSP for base-isolated structures is described in detail, providing a reference for research on RTHS method. Salmonella 4,[5],12:i:-, a monophasic variant of Salmonella Typhimurium, has emerged as a global cause of multidrug-resistant salmonellosis and has become endemic in many developing and developed countries, especially in China. Here, we have sequenced 352 clinical isolates in Guangdong, China, during 2009-2019 and performed a large-scale collection of Salmonella 4,[5],12:i:- with whole genome sequencing (WGS) data across the globe, to better understand the population structure, antimicrobial resistance (AMR) genomic characterization, and transmission routes of Salmonella 4,[5],12:i:- across Guangdong. Salmonella 4,[5],12:i:- strains showed broad genetic diversity; Guangdong isolates were found to be widely distributed among the global lineages. Of note, we identified the formation of a novel Guangdong clade (Bayesian analysis of population structure lineage 1 [BAPS1]) genetically diversified from the global isolates and likely emerged around 1990s. BAPS1 exhibits unique genomic features, including large pan-genome, decreased ciprofloxacin susceptibility due to mutation in gyrA and carriage of plasmid-mediated quinolone resistance (PMQR) genes, and the multidrug-resistant IncHI2 plasmid. Furthermore, high genetic similarity was found between strains collected from Guangdong, Europe, and North America, indicating the association with multiple introductions from overseas. These results suggested that global dissemination and local clonal expansion simultaneously occurred in Guangdong, China, and horizontally acquired resistance to first-line and last-line antimicrobials at local level, underlying emergences of extensive drug and pan-drug resistance. Our findings have increased the knowledge of global and local epidemics of Salmonella 4,[5],12:i:- in Guangdong, China, and provided a comprehensive baseline data set essential for future molecular surveillance.IMPORTANCE Salmonella 4,[5],12:i:- has been regarded as the predominant pandemic serotype causing diarrheal diseases globally, while multidrug resistance (MDR) constitutes great public health concerns. This study provided a detailed and comprehensive genome-scale analysis of this important Salmonella serovar in the past decade in Guangdong, China. Our results revealed the complexity of two distinct transmission modes, namely global transmission and local expansion, circulating in Guangdong over a decade. Using phylogeography models, the origin of Salmonella 4,[5],12:i:- was predicted from two aspects, year and country, that is, Salmonella 4,[5],12:i:- emerged in 1983, and was introduced from the UK, and subsequently differentiated into the local endemic lineage circa 1991. Additionally, based on the pan-genome analysis, it was found that the gene accumulation rate in local endemic BAPS 1 lineage was higher than in other lineages, and the horizontal transmission of MDR IncHI2 plasmid associated with high resistance played a major role, which showed the potential threat to public health., Competing Interests: The authors declare no conflict of interest.

Superhydrophobic TC4 Ti alloy is essential for economical use in modern machinery. Herein, a novel superhydrophobic pattern textured TC4 Ti alloy surface with a self-assembled molecular film was fabricated by an efficient nanosecond laser ablation (laser surface texturing (LST)) and a facile chemical modification method. Further, the transformation of the surface morphology and chemical composition was analysed. Upon adjusting the laser fluence, scanning velocity and scanning distance, the maximum water and saline contact angles of the laser surface textured and chemically treated surface reached 155.5 ±0.8° and 152.5 ±1.3°, respectively. The tribological behaviour of the patterned surfaces was investigated under dry-sliding and liquid-sliding conditions. High density textures exhibited the greatest potential for lubrication under water and saline lubrication conditions. In a dry environment, a scattered-line pattern textured surface also showed a friction reducing effect, similar to that under saline sliding conditions. OBJECTIVE: This study aimed to investigate the therapeutic effects and underlying mechanisms of Sankudiwan (SKDW) on myocardial ischemia-reperfusion injury (MIRI) in a rat model. MATERIALS AND METHODS: Rats were subjected to MIRI and treated with varying doses of SKDW. The myocardial infarct size, cardiac function, histological changes, apoptosis, and inflammation were assessed using TTC staining, echocardiography, Hematoxylin and Eosin (HE) staining, TUNEL staining, and ELISA assays. We further explored SKDW's influence on cardiomyocyte mitochondria and inflammatory factor expression. Moreover, oxidative stress-related parameters and differentially expressed genes were analyzed using bioinformatics approaches. RESULTS: SKDW significantly reduced myocardial infarct size and improved cardiac function, demonstrating a dose-dependent therapeutic potential. It ameliorated myocardial tissue damage at the histological level, inhibited cardiomyocyte apoptosis, and mitigated inflammatory response. SKDW also enhanced mitochondrial energy metabolism and suppressed the levels of oxidative stress markers. Bioinformatics analysis identified key differentially expressed genes (DEGs), including cbln1, Tgm1, Trh, and Ccl27, possibly mediating the therapeutic effects of SKDW. CONCLUSIONS: SKDW exerts its therapeutic effects on MIRI through the modulation of several genes and pathways related to inflammation, apoptosis, mitochondrial function, and oxidative stress. This article discusses a study that identified a recurrent splicing mutation (c.2442 + 5G>A) of the Colony-stimulating factor 1 receptor (CSF1R) gene in a Chinese family with leukoencephalopathy. The mutation was found to result in intron 12 not being cut, leading to a premature stop codon in the CSF1R gene. This mutation has previously been reported in a Japanese patient with diffuse leukoencephalopathy. The study confirms the pathogenicity of this mutation and provides further understanding of the genetic profile and clinical diagnosis of patients with leukoencephalopathy. OBJECTIVE: The crosstalk between age and immunity in the context of ulcerative colitis (UC) remains incompletely understood. Our objective is to elucidate the specific age-associated genetic factors that modulate immune cell infiltration in UC, with the aim of identifying innovative therapeutic targets for the treatment of this disease. MATERIALS AND METHODS: Potential batch effects between samples were removed by R package "inSilicoMerging". Unsupervised clustering analysis via the "ConsensusClusterPlus" R package was utilized to perform consensus molecular subtyping of immune subtypes in UC. The construction of a heat map was accomplished through the utilization of the R package "pheatmap", while functional enrichment analysis was executed by means of the Metascape database. The identification of the age-related gene module was achieved by performing weighted gene co-expression network analysis (WGCNA) analysis using the R package "WGCNA". The support vector machine (SVM), least absolute shrinkage and selector operation (LASSO), and random forest algorithms were performed via the "e1071", "glmnet" and "randomForest" packages in R, respectively. The diagnostic performance of the parameter was assessed using the receiver operating characteristic (ROC) curve. Correlation analysis was performed by Spearman correlation. The "XSum" package in R was employed to identify potential small-molecule drugs for UC utilizing the Connectivity Map (CMap) database. Molecular docking was performed with Autodock Vina molecular docking software. RESULTS: A significantly greater frequency of UC patients aged below 40 years was observed in the group with extensive disease extent as compared to those with non-extensive disease extent (70% vs. 47%; Chi-square test, p = 0.02). The application of unsupervised clustering analysis allowed for the stratification of UC patients into two distinct immune subtypes, namely cluster C1 and cluster C2. The distribution of immune subtypes was significantly different between different age categories (Chi-square test, p = 0.00219). The UC samples that were grouped under cluster C1 were distinguished by a higher abundance of macrophages and an elevated number of neutrophils relative to those in cluster C2. Based on both WGCNA and Limma analysis, 146 age-related genes were identified, which exhibited a predominant enrichment in the biological process of cellular senescence. Two age-related genes (MIDN, and PLD6) affecting the immune cell infiltration in UC were identified based on machine learning algorithms (SVM, LASSO, and random forest). The diagnostic performance of MIDN (AUC = 0.93) and PLD6 (AUC = 0.90) in discerning UC patients belonging to cluster C1 was found to be satisfactory, as demonstrated by ROC curve analysis. MIDN demonstrated a positive correlation (r = 0.50, p < 0.0001) with Neutrophil, while PLD6 exhibited a negative correlation (r = -0.52, p < 0.0001) with Neutrophil levels. The "XSum" algorithm revealed that Entinostat has therapeutic potential for UC. The docking glide score between Entinostat and MIDN, and PLD6 protein was -8.9 kcal/mol and -6.8 kcal/mol, respectively. CONCLUSIONS: We have identified two age-related genes, MIDN and PLD6, that are involved in immune cell infiltration in patients with ulcerative colitis. Furthermore, a small molecule drug (Entinostat) with potential therapeutic effects for UC was screened out. Vehicle's wheels were mostly modeled as a point, which can touch the valleys of pavement roughness, creating unrealistic high-frequency oscillation. This can be avoided by using the disk model for the wheels, which however adds significant complexity to the vehicle–bridge interaction (VBI) analysis. In this paper, a refined roughness formula is generated to account for the wheel size effect such that it can be used by point model. Still, the low-frequency part of the roughness presents some masking effect on the bridge scanning by the test vehicle. To this end, two countermeasures are suggested, i.e. residual response and traffic flows. This study has demonstrated that: (1) the roughness generated by the refined formula can reflect the trace of the disk model; (2) the refined formula facilitates the VBI analysis by using the point model; and (3) the two countermeasures for roughness are effective for improving the scanning of bridge frequencies. OBJECTIVE: Studies of possible implications of the maternal hepatitis C virus (HCV) infection in terms of intrauterine fetal growth restriction (IUGR), preterm birth (PTB), low birth weight (LBW) infants, premature rupture of membranes (PROM), maternal and neonatal mortality are limited and inconclusive. Our study aims to assess the impact of HCV on maternal and neonatal outcomes. MATERIALS AND METHODS: Systematic literature search was done in PubMed, Scopus, and Google Scholar, Cochrane Library, and TRIP databases for all observational studies published from 1st January 1950 to 15th October 2022. The pooled odds ratio (OR) or risk ratio (RR) with a 95% confidence interval (CI) was estimated. STATA version 12.0 software was used for analysis. Heterogeneity among the included articles was evaluated by sensitivity, meta-regression, and publication bias analyses. RESULTS: A total of 14 studies involving 12,451 HCV (+) and 56,42,910 HCV (-) pregnant women were included in our meta-analysis. Maternal HCV during pregnancy was significantly associated with the increased risk of PTB (OR=1.66, 95% CI: 1.59-1.74), IUGR (OR=2.09, 95% CI: 2.04-2.14) and LBW (OR=1.96, 95% CI: 1.63-2.36) as compared to healthy pregnant women. Subgroup analysis based on ethnicity also suggested a strong association between maternal HCV infection and a higher risk of PTB in Asian and Caucasian populations. Maternal (RR=3.44, 95% CI: 1.85-6.41), as well as neonatal (RR=1.54, 95% CI: 1.18-2.02) mortality was significantly higher in HCV (+) cases. CONCLUSIONS: Mothers with HCV infection had a markedly increased probability of PTB and/or IUGR and/or LBW. In clinical practice, standard care of treatment and proper monitoring are needed for the pregnant population with HCV infection.
