Background: Two-line hybrid wheat technology system is one way to harness wheat heterosis both domestically and internationally. Seed vigor is a crucial parameter for assessing seed quality, as enhanced seed vigor can lead to yield increments of over 20% to a certain extent. MicroRNAs (miRNAs) were known to participate in the development and vigor of seed in plants, but its impact on seed vigor in two-line hybrid wheat remains poorly elucidated. Results: The hybrid (BS1453/11GF5135) wheat exhibited superiority in seed vigor and anti-aging capacity, compared to its male parent (11GF5135, MP) and female parent (BS1453, FP). We identified four miRNAs associated with seed vigor, all of which are novel miRNAs. The majority of targets of miRNAs were related to ubiquitin ligases, kinases, sucrose synthases and hydrolases, involving in starch and sucrose metabolism, hydrolysis, catalysis, plant hormone signal transduction, and other pathways, which played crucial roles in seed development. Additionally, we also found miR531 was differentially expressed in both male parent and hybrid, and its target gene was a component of the E1 subunit of α-ketoate dehydrogenase complex, which interacted with dihydrolipoamide acetyltransferase (E2) and dihydrolipoyl dehydrogenase (E3). Finally, We established a presumptive interaction model to speculate the relationship of miR531 and seed vigor. Conclusions: This study analyzed the seed vigor of two-line hybrid wheat, and screened seed vigor-related miRNAs. Meanwhile speculated the genetic relationship of hybrid and parents, in terms of miRNAs. Consequently, the present study provides new insights into the miRNA-mediated gene and protein interaction network that regulates seed vigor. These findings hold significance for enhancing the yield and quality of two-line hybrid wheat, facilitating its future applications. [ABSTRACT FROM AUTHOR]

The phenomenon of droplet coalescence and jumping has received increasing attention due to its potential applications in the fields of condensation heat transfer and surface self-cleaning. Basic research on the process and mechanism of coalescence-induced droplet jumping has been carried out, and some universal laws have been established. However, it is found that the focus of these studies is based on two identical droplets, and the coalescence-induced jumping with different radii is rarely investigated, which is commonly encountered in nature. Therefore, it is essential to proceed with the research of coalescence and jumping of droplets with unequal radii. In this paper, molecular dynamics (MD) simulations are performed to reveal the effects of radius ratio and radius of small droplets on jumping velocity. The results show that as the increasing of radius ratio with an unchanged small droplet radius of 8.1 nm, the jumping velocity increases then decreases, which indicates there is an optimal radius ratio to maximize the jumping velocity. Additionally, it is found that if the small droplet radius is changed, the critical radius ratio for characterizing whether the coalesced droplet jumping increases with increasing the small droplet radius. Furthermore, according to energy conservation, the conversion efficiency of energy is discussed. The results show that when the radius ratio is greater than 1.3 with three different small droplet radii, the energy conversion efficiency rapidly decreases to below 1.0%; and the critical radius ratios are consistent with the result obtained from the velocity analysis. This work broadens the understanding of the more general phenomenon of coalescence-induced droplet jumping and can better guide industrial applications. [ABSTRACT FROM AUTHOR]

Two new dinor-eudesmane sesquiterpenoids, named multistalin A (1), and multistalin B (2), together with three sesquiterpene glycosides (3–5), and a norlabdane-type diterpene (6) were isolated from the root extract of Chloranthus multistachys Pei. Their structures were elucidated on the basis of spectroscopic analysis including 1D, 2D NMR techniques and HR-ESI-MS. Vascular calcification and vascular ageing are "silent" diseases but are highly prevalent in patients with end stage renal failure and type 2 diabetes, as well as in the ageing population. Melatonin (MT) has been shown to induce cardiovascular protection effects. However, the role of MT on vascular calcification and ageing has not been well-identified. In this study, the aortic transcriptional landscape revealed clues for MT related cell-to-cell communication between endothelial cells (ECs) and vascular smooth muscle cells (VSMCs) in vascular calcification and vascular ageing. Furthermore, we elucidated that it was exosomes that participate in the information transportation from ECs to VSMCs. The exosomes secreted from melatonin-treated ECs (MT-ECs-Exos) inhibited calcification and senescence of VSMCs. Mechanistically, miR-302d-5p was highly enriched in MT-ECs-Exos, while depletion of miR-302d-5p blocked the ability of MT-ECs-Exos to suppress VSMC calcification and senescence. Notably, Wnt3 was a bona fide target of miR-302d-5p and modulated VSMC calcification and senescence. Furthermore, we found that maturation of endothelial derived exosomal miR-302d-5p was promoted by WTAP in an N 6 -methyladenosine (m 6 A)-dependent manner. Interestingly, MT alleviated vascular calcification and ageing in 5/6-nephrectomy (5/6 NTP) mice, a chronic kidney disease (CKD) induced vascular calcification and vascular ageing mouse model. MT-ECs-Exos was absorbed by VSMCs in vivo and effectively prevented vascular calcification and ageing in 5/6 NTP mice. ECs-derived miR-302d-5p mediated MT induced anti-calcification and anti-ageing effects in 5/6 NTP mice. Chronic kidney disease (CKD), one of the common clinical urological diseases, is increasingly more prevalent in recent years and has emerged as a major concern of public health around the globe. The continuous recurrence of CKD caused by renal function impairment leads eventually to irreversible renal failure and severe systemic complications, which causes severe negative impact on the quality of life of the patient. As an essential component of human microbiome, oral microbiota plays a major role in maintaining health, and there has been research suggesting close association between oral dysbiosis and CKD. It is therefore of great clinical significance to understand the correlation between CKD and oral microbiota. Herein, we reviewed the characteristics of oral microbiota of CKD patients, the possible mechanisms of oral microbiota's involvement in the pathogenesis and development of CKD, and the latest research findings on oral dysbiosis and CKD, with a view to finding new approaches to early prevention and control of CKD through oral microbial targets., (Copyright© by Editorial Board of Journal of Sichuan University (Medical Sciences).)

Purpose: To investigate the related factors of black tooth stain in primary teeth of 3~5 years old children and caries status of primary dentition in these children., Methods: From December 2019 to August 2020, 182 3~5 years old children with black tooth stain and 200 children without pigmentation were investigated by oral examination and questionnaire survey to their guardians, while caries status , distribution range of the pigment, factors associated with black tooth stain were evaluated. SPSS 20.0 software package was used for data analysis., Results: Children with black tooth stain had fewer dental caries. The lingual surfaces of the mandibular anterior teeth were the most affected sites. Factors associated with black tooth stain were foods with soy sauce and brushing teeth with parents' help., Conclusions: There is a negative correlation between the occurrence of primary dentition caries and blacktooth stain, but there is no significant correlation between formation of black tooth stain and most environmental factors.

Purpose: To explore the differences between oral and intestinal microorganisms of 860 children aged 1-6 years in Nanjing city., Methods: Eight hundred and sixty children aged 1-6 years who met the inclusion criteria were enrolled. 860 saliva samples and equivalent stool samples were collected. Microbial DNA was extracted and amplified using PCR. High-throughput sequencing was performed using Miseq sequencer, and bioinformatics analysis was performed to compare the differences of oral and intestinal microflora. SPSS 20.0 software package was used to perform the statistical analysis., Results: At phylum level, Bacteroidetes (39.98%), Proteobacteria (25.32%) and Firmicutes (21.78%) were the most common microbes in oral cavity, while Firmicutes (45.21%) and Bacteroidetes (37.21%) were the most abundant microbes in the gut. At genus level, the top three microbes in the oral cavity included Prevotella(26.11%), Neisseria (12.39%), Porphyromonas(10.13%), while the top three microbes in the gut included Bacteroidetes(20.11%), Prevotella (9.13%), and Faecalibacterium (5.13%). There were significant alpha and beta differences in oral and intestinal microbial diversity., Conclusions: Bacteroidetes and Proteobacteria are the dominant species in the oral cavity, while Firmicutes and Bacteroidetes are the dominant species in the gut. There are significant differences in species composition and abundance between oral and intestinal microorganisms.
