Two experiments tested the effect of a bacterial 6-phytase (PhyG) supplemented to a vegetable broiler diet without or with added trace minerals (TM), on growth performance and TM utilization. Each tested 12 treatments in a 2 × 6 factorial arrangement with 3,360 Ross 308 males (35 birds/pen, 8 pens/treatment) in a randomized complete block design. Phytase levels comprised no PhyG or PhyG at 2,000, 1,500 and 1,000 FTU/kg during 0 to 10, 10 to 20 and (Experiment 2) 20 to 35 d of age, respectively; TM diets comprised: no added TM (diet 1); 10, 15, 3 and 10, or 20, 30, 6 and 20 mg/kg of Zn, Fe, Cu and Mn as sulphate (diets 2 and 3); 15, 3 and 10 mg/kg of Zn, Cu and Mn as oxide and 15 mg/kg Fe as sulphate, or 30, 6 and 20 mg/kg of Zn, Cu and Mn as oxide and 30 mg/kg Fe as sulphate (diets 4 and 5), and; 10, 3, 15 and 10 mg/kg of organic Zn, Cu, Fe and Mn (diet 6). Apart from no added TM, basal diets were nutritionally adequate; phytase-supplemented diets were reduced in Ca and retainable P. All diets were pelleted and fed ad libitum. In both experiments during all phases there were interactions (P < 0.05) between phytase and TM on BW, BW gain (BWG), feed intake (FI), FCR (experiment 2 only) and tibia Zn. Without phytase, TM improved (P < 0.05) these measures. Phytase without TM increased (P < 0.05) d 35 BW, overall BWG and FI, equivalent to or beyond the level achieved by TM without phytase. Tibia Zn at d 10 and 20 in Experiment 2 was increased by phytase beyond the level achieved by TM without phytase (P < 0.05). Phytase also increased (P < 0.05) liver Zn, Fe, and Mn at d 10 and 20 and plasma Zn at d 20 d. There was no consistent effect of TM source or dose on bird responses. In conclusion, PhyG phytase replaced the effect of added TM on performance during 0 to 35 d of age and could support a reduction in commercial dose levels of added TM in all-vegetable broiler diets., Competing Interests: Declaration of competing interest Authors declare they have no conflicts of interests., (Copyright © 2024. An experiment was conducted to evaluate the effects of increasing the dose of a novel consensus bacterial 6-phytase variant expressed in Trichoderma reesei (PhyG) in broilers fed complex diets highly deficient in minerals, dig AA, and energy. Diets were a nutrient-adequate control (PC); a nutrient-reduced control (NC) formulated with a reduction in available P (avP) by 0.199%, Ca by 0.21%, crude protein by 0.72-1.03%, dig Lys by 0.064-0.084%, Na by 0.047%, and ME by 87.8 kcal/kg, respectively; and NC supplemented with PhyG at 500, 1000, and 2000 FTU/kg feed. BW was decreased and FCR increased in the NC vs. PC, while the PhyG treatments were similar to the PC. Carcass yield and bone ash were also maintained with PhyG supplementation. Phytase provided economic benefit on a feed cost per kg of weight basis for 1 to 35 d; the cost reductions equated to USD 0.006, 0.016, and 0.02/kg BWG at 500, 1000, and 2000 FTU/kg. In conclusion, this trial demonstrated that supplementation with a novel consensus phytase variant in diets highly deficient in minerals, dig AA, and energy maintained growth performance and provided economic benefit, with production benefits being maximized at inclusion levels of 2000 FTU/kg.

Polycyclic aromatic hydrocarbons (PAHs) were studied in the soils of three proglacial areas in France (Noir and Chardon Glaciers) and Italy (Miage Glacier). PAH contents, PAH stocks and PAH contents normalized to the total organic carbon contents (PAHs/TOC ratio) were investigated along proglacial soil chronosequences to infer their evolutions with soil age (from 3 to 4200 years), where the PAH contamination was only related to long-range atmospheric transport. Evolutions of PAH and TOC contents, PAHs/TOC ratio and PAH stock were fitted with exponential and logarithmic relations. For the three proglacial areas, PAH contents increased rapidly during the first 150 years of soil development, ranged from 4 to 152 ng·g -1 , and showed a strong relationship with total organic carbon (TOC) contents (r = 0.83, p < 0.05). The joint increase of PAH and TOC contents suggested that PAH accumulation in soils were not only driven by PAH inputs but also by the capacity of soils to store these contaminants. PAH contents in the oldest soils (from 1200 BCE and 2200 BCE) were similar than for soils from 1850 CE. The period 1850-2019 CE corresponded to a decrease in the PAHs/TOC ratio suggesting both a faster accumulation of TOC than PAHs and a dilution effect of PAHs already present in soils. For the oldest soils, the PAHs/TOC ratio appeared similar to those for soils from 1850 CE, with values ranging from 0.48 to 2.06 ng·mg -1 , suggesting an equilibrium between both parameters for soils older than 170 years. Finally, PAH stocks ranged from 0.41 mg·m -2 to 6.80 mg·m -2 in the youngest and oldest soils, respectively. The digestive capacity of organic compounds by the black soldier fly (BSF, Hermetia illucens, Diptera: Stratiomyidae, Linnaeus, 1758) is known to rely on complex larva-microbiota interactions. Although insect development is known to be a driver of changes of bacterial communities, the fluctuations along BSF life cycle in terms of composition and diversity of bacterial communities are still unknown. In this work, we used a metabarcoding approach to explore the differences in bacterial diversity along all four BSF developmental stages: eggs, larvae, pupae, and adult. We detected not only significant differences in bacterial community composition and species richness along the development of BSF, but also nine prevalent amplicon single variants (ASVs) forming the core microbiota. Out of the 2010 ASVs identified, 160 were significantly more abundant in one of the life stages. Moreover, using PICRUSt2, we inferred 27 potential metabolic pathways differentially used among the BSF life cycle. This distribution of metabolic pathways was congruent with the bacterial taxonomic distribution among life stages, demonstrating that the functional requirements of each phase of development are drivers of bacterial composition and diversity. This study provides a better understanding of the different metabolic processes occurring during BSF development and their links to changes in bacterial taxa. Pachyonychia congenita (PC) is a rare keratinizing disorder characterized by painful palmoplantar keratoderma for which there is no standard current treatment. PC is caused by dominant mutations in keratin (K) K6A, K6B, K6C, K16, or K17 genes involved in stress, wound healing, and epidermal barrier formation. Mechanisms leading to pain and painful palmoplantar keratoderma in PC remain elusive. In this study, we show overexpression of EGFR ligands epiregulin and TGF-α as well as HER1‒EGFR and HER2 in the upper spinous layers of PC lesions. EGFR activation was confirmed by upregulated MAPK/ERK and mTOR signaling. Abnormal late terminal keratinization was associated with elevated TGM1 activity. In addition, the calcium ion permeable channel TRPV3 was significantly increased in PC-lesional skin, suggesting a predominant role of the TRPV3/EGFR signaling complex in PC. We hypothesized that this complex contributes to promoting TGM1 activity and induces the expression and shedding of EGFR ligands. To counteract this biological cascade, we treated three patients with PC with oral erlotinib for 6‒8 months. The treatment was well-tolerated and led to an early, drastic, and sustained reduction of neuropathic pain with a major improvement of QOL. The effect of a biosynthetic bacterial 6-phytase (PhyG) on the digestibility and excretion of crude protein (CP), phosphorus (P), and phytate-P (PP) in midlactating dairy cows was investigated. Thirty Holstein-Friesians were assigned to three treatments with 10 cows per treatment in a randomized block design. Cows were fed forage (grass and corn silage) provided ad libitum, and a concentrate (without added inorganic phosphate) administered separately in amounts individualized per cow according to milk production, supplemented with phytase according to treatment. The formulated forage-to-concentrate-ratio was ~65%:35%. Dietary treatments comprised the control diet (CON) and CON supplemented with 2,000 (PhyG2,000) or 5,000 (PhyG5,000) phytase units (FTU)/kg DM in the total diet. The experiment comprised an 18-d preperiod for the collection of data to facilitate the allocation of cows to the treatments, followed by a 19-d experimental period comprising a 14-d diet adaptation period and 5 d of twice daily feces collection. Fecal samples were analyzed for the determination of apparent total tract digestibility (ATTD) of chemical constituents in the diet. The ATTD of PP was 92.6% in CON suggesting a high but incomplete degradation of phytate by ruminal microbial phytases. Cows fed PhyG2,000 exhibited increased ATTD of CP and PP [68.4% (2.7% points above CON) and 95.1% (2.5% points above CON), respectively] whilst PhyG5,000 further increased ATTD PP and also increased ATTD P [54.1% (7.8% points above CON)]; ATTD of Ca tended to be increased in PhyG5,000 vs. CON. Linear dose-response relationships were observed for ATTD of DM, CP, P, Ca, and PP. In addition, fecal excretion of P, and PP linearly reduced and that of Ca and CP tended to linearly reduce with increasing PhyG dose level. No difference was observed for DM intake and milk composition was unaffected except for milk protein which tended to be higher in cows fed PhyG5,000 than CON. In summary, the addition of exogenous phytase at 2,000 FTU/kg or higher to diets of lactating dairy cows improved P, PP, Ca, and CP digestibility and reduced fecal excretion of P, PP, and CP in a dose-dependent manner., (© The Author(s) 2023. Paramylon is a linear β-1,3-glucan, similar to curdlan, produced as intracellular granules by the microalga Euglena gracilis, a highly versatile and robust strain, able to grow under various trophic conditions, with valorization of CO 2, wastewaters, or food byproducts as nutrients. This review focuses in particular on the various processing routes leading to new potential paramylon based products. Due to its crystalline structure, involving triple helices stabilized by internal intermolecular hydrogen bonds, paramylon is neither water-soluble nor thermoplastic. The few solvents able to disrupt the triple helices, and to fully solubilize the polymer as random coils, allow non derivatizing shaping into films, fibers, and even nanofibers by a specific self-assembly mechanism. Chemical modification in homogeneous or heterogeneous conditions is also possible. The non-selective or regioselective substitution of the hydroxyl groups of glucosidic units leads to water-soluble ionic derivatives and thermoplastic paramylon esters with foreseen applications ranging from health to bioplastics., (Copyright © 2022 Elsevier Ltd. Nitrogen limitation and changing solar conditions are both known to affect triacylglycerol (TAG) production in microalgae. This study investigates the optimization of TAG production with a continuous nitrogen-limited culture of Nannochloropsis gaditana in simulated day-night cycles (DNc). The effect of DNc was first investigated in nitrogen-deprived condition (i.e., batch culture), emphasizing a significant change in mechanical resistance of the strain during the night. The concept of released TAG, which shows how much of the TAG produced is actually recovered in the downstream stages, that is, after cell disruption, was shown here of interest. For a maximum released TAG, the optimum harvesting time was suggested as being 4 h into the night period, which minimizes the losses due to a too great cell mechanical resistance. The protocol for continuous nitrogen-limited culture was then optimized, and a continuous nitrogen addition was compared to a pulsed-addition. For the latter, nitrogen was supplied in a single pulse at the beginning of the light periods, while the bulk medium was supplied separately at a slow but constant dilution rate of 0.005 h - 1 $0.005\,{{\rm{h}}}^{-1}$ . The pulse dose was calculated after the study of nitrogen consumption and TAG production/consumption during the DNc. The estimated released TAG for the pulsed-addition of 1.4 × 10 -3  kg/m 2 d found significantly higher than the one achieved in batch culture (0.3 × 10 -3  kg/m 2 d) but lower than for continuous nitrogen addition which obtained the highest released TAG of 3 × 1 0 - 3 kg ∕ m 2   d $3\times 1{0}^{-3}\,\text{kg}\unicode{x02215}{{\rm{m}}}^{2}\unicode{x0200A}{\rm{d}}$ ., (© 2022 Wiley Periodicals LLC.)

Fucoxanthin is a brown-colored pigment from algae, with great potential as a bioactive molecule due to its numerous properties. This review aims to present current knowledge on this high added-value pigment. An accurate analysis of the biological function of fucoxanthin explains its wide photon absorption capacities in golden-brown algae. The specific chemical structure of this pigment also leads to many functional activities in human health. They are outlined in this work and are supported by the latest studies in the literature. The scientific and industrial interest in fucoxanthin is correlated with great improvements in the development of algae cultures and downstream processes. The best fucoxanthin producing algae and their associated culture parameters are described. The light intensity is a major influencing factor, as it has to enable both a high biomass growth and a high fucoxanthin content. This review also insists on the most eco-friendly and innovative extraction methods and their perspective within the next years. The use of bio-based solvents, aqueous two-phase systems and the centrifugal partition chromatography are the most promising processes. The analysis of the global market and multiple applications of fucoxanthin revealed that Asian companies are major actors in the market with macroalgae. In addition, fucoxanthin from microalgae are currently produced in Israel and France, and are mostly authorized in the USA.

We observed differences in lhc classification in Chromista. We proposed a classification of the lhcf family with two groups specific to haptophytes, one specific to diatoms, and one specific to seaweeds. Identification and characterization of the Fucoxanthin and Chlorophyll a/c -binding Protein (FCP) of the haptophyte microalgae Tisochrysis lutea were performed by similarity analysis. The FCP family contains 52 lhc genes in T. lutea . FCP pigment binding site candidates were characterized on Lhcf protein monomers of T. lutea , which possesses at least nine chlorophylls and five fucoxanthin molecules, on average, per monomer. The expression of T. lutea lhc genes was assessed during turbidostat and chemostat experiments, one with constant light (CL) and changing nitrogen phases, the second with a 12 h:12 h sinusoidal photoperiod and changing nitrogen phases. RNA-seq analysis revealed a dynamic decrease in the expression of lhc genes with nitrogen depletion. Introduction/aims: While the peripheral nervous system has the inherent ability to recover following injury, results are often unsatisfactory, resulting in permanent functional deficits and disability. Therefore, methods that enhance regeneration are of significant interest. The present study investigates an injectable nerve-tissue-specific hydrogel as a biomaterial for nerve regeneration in a rat nerve crush model., Methods: Nerve-specific hydrogels were injected into the subepineurial space in both uninjured and crushed sciatic nerves of rats to assess safety and efficacy, respectively. The animals were followed longitudinally for 12 wk using sciatic functional index and kinematic measures. At 12 wk, electrophysiologic examination was also performed, followed by nerve and muscle histologic assessment., Results: When the hydrogel was injected into an uninjured nerve, no differences in sciatic functional index, kinematic function, or axon counts were observed. A slight reduction in muscle fiber diameter was observed in the hydrogel-injected animals, but overall muscle area and kinematic function were not affected. Hydrogel injection following nerve crush injury resulted in multiple modest improvements in sciatic functional index and kinematic function with an earlier return to normal function observed in the hydrogel treated animals as compared to untreated controls. While no improvements in supramaximal compound motor action potential were observed in hydrogel treated animals, increased axon counts were observed on histologic assessment., Discussion: These improvements in functional and histologic outcomes in a rapidly and fully recovering model suggest that injection of a nerve-specific hydrogel is safe and has the potential to improve outcomes following nerve injury., (© 2021 Wiley Periodicals LLC.)

The effect of a novel consensus bacterial 6-phytase variant (PhyG) on apparent ileal digestibility (AID) of amino acids (AA) and phosphorus (P) utilization in young broilers when added to diets with high phytate-P (PP) content without added inorganic phosphate (Pi) and deficient in digestible (dig) AA and metabolizable energy (ME) was investigated. A total of 256 Ross 308 male broilers were assigned to 4 treatments (8 birds/cage, 8 cages/treatment) in a completely randomized design. Treatments comprised a positive control (PC, 2,975 kcal/kg ME, 3.7 g/kg dig P, 2.83 g/kg PP, 8.4 g/kg Ca, 10.6 g/kg dig lysine), a negative control (NC) without added Pi (ME -68 kcal/kg, crude protein -10 g/kg, dig AA -0.1 to -0.4 g/kg, Ca -2.0 g/kg, dig P -2.2 g/kg, Na -0.4 g/kg vs. PC), and NC plus 500 or 1,000 FTU/kg of PhyG. Test diets were corn/soy/rapeseed-meal/rice-bran-based and fed from 5 to 15 d of age. Ileal digesta and tibias were collected on day 15. Excreta was collected during days 12 to 15 to determine P retention. The NC (vs. PC) reduced (P < 0.05) P retention (-10.4% units), tibia ash (-14.3% units), weight gain (-109 g), feed intake (-82 g) and increased FCR (from 1.199 to 1.504), confirming that the NC was extremely deficient in nutrients and energy. Phytase addition to the NC linearly (P < 0.001) improved performance, but did not fully recover it to the level of the PC due to the severe nutrients/energy reduction in NC. Phytase linearly increased P retention (P < 0.001), tibia ash (P < 0.001), AID of dry matter (P < 0.05), nitrogen (P < 0.01), gross energy (P < 0.05), and all 17 individual AA (P < 0.01). At 1,000 FTU/kg, phytase increased (P < 0.05) P retention vs. PC and NC (+14.5 and +24.9% units, respectively) and increased tibia ash vs. NC (+13.8% units), equivalent to PC. The NC decreased AID of Cys, Gly, Thr, and Met vs. PC (P < 0.05). At 1,000 FTU/kg, phytase increased AID of all 17 AA vs. NC (P < 0.01), equivalent to PC. At 1,000 FTU/kg, AID AA responses (above NC) ranged from +4.5% (Met) to +15.0% (Cys), being maximal for essential Thr (+10.4%) and Val (+8.2%) and non-essential Cys (+15.0%) and Gly (+10.4%). The results highlight the efficacy of PhyG at a dose level of 500 to 1,000 FTU/kg in young broilers for improving the ileal digestibility of nitrogen, AA, and energy alongside P retention and tibia ash.
