A field experiment was carried out during two successive seasons of 2018/2019 and 2019/2020 in the Floriculture Farm of the Horticulture Department, Faculty of Agriculture, Benha University, Egypt to study the effects of spraying some growth substances (kinetin, salicylic acid, calcium thiosulfate and potassium silicate as well as, N.P.K chemical fertilization) on vegetative growth and chemical composition of Matthiola incana plants to increase and improve the quality of this plant as a bed gardens ornamentation. The obtained results showed that the tallest plants and the thickest plant stems were recorded by 100 ppm kinetin combined with NPK fertilization at high level treatment in both seasons. The heaviest fresh and dry weights of leaves/plant were recorded by 100 ppm kinetin-sprayed plants enriched with NPK fertilization at the highest level in both seasons. The highest number of branches and leaves/plant were recorded by 100 ppm kinetin-sprayed plants combined with NPK fertilization at high level in both seasons. The highest leaf N and total carbohydrates % were recorded by 100 ppm kinetin- sprayed plants provided with NPK fertilization at the highest level, whereas the richest leaf P % was registered by 200 ppm salicylic-sprayed plants combined with NPK fertilization at the highest level. While, the greatest leaf K % was scored by 6 cm3 potassium silicate-sprayed plants, supplemented with NPK fertilization at the highest level in both seasons. Conclusively, spraying Matthiola incana plants with kinetin at 100 ppm in addition to chemical fertilizer at 100 N: 200 P: 200 K kg/fed produced the best vegetative growth and quality of this plant

Stock (Matthiola incana (L.) R. Br.) is a famous annual ornamental plant with important ornamental and economic value. The lack of DNA molecular markers has limited genetic analysis, genome evolution, and marker-assisted selective breeding studies of M. incana. Therefore, more DNA markers are needed to support the further elucidation of the biology and genetics of M. incana. In this study, a high-quality genome of M. incana was initially assembled and a set of effective SSR primers was developed at the whole-genome level using genome data. A total of 45,612 loci of SSRs were identified; the di-nucleotide motifs were the most abundant (77.35%). In total, 43,540 primer pairs were designed, of which 300 were randomly selected for PCR validation, and as the success rate for amplification. In addition, 22 polymorphic SSR markers were used to analyze the genetic diversity of 40 stock varieties. Clustering analysis showed that all varieties could be divided into two clusters with a genetic distance of 0.68, which were highly consistent with their flower shape (potted or cut type). Moreover, we have verified that these SSR markers are effective and transferable within the Brassicaceae family. In this study, potential SSR molecular markers were successfully developed for 40 M. incana varieties using whole genome analysis, providing an important genetic tool for theoretical and applied research on M. incana. Garden stock [Matthiola incana (L.) R. Br.] is a commercially important horticultural crop owing to its ornamental effect. There are different stock cultivars varied in color and shape, especially flowered phenotype is an essential index evaluating its commercial value, because double flowered individuals have more brilliant flowers compared to single flowered one. The present work aimed: (1) to make superior cultivars with different colors, high fertility, being capable of early selecting only double flowered seedlings by leaf color and to investigate morphological characteristics and (2) to select RAPD and ISSR primers for the cultivar certification and identification to culture and produce good commercial stock cultivars. Here, in this study new stock cultivars were obtained with different colors including pink, pale pink and white, through outcrossing between "white" cultivar (high fertile but unable to select double flower phenotype) and "pink" cultivar (vice. versa). Among newly obtained stock cultivars, single and double flowered seedlings are distinguishable from single ones by leaf color, having about 70% of fertility. Moreover RAPD and ISSR markers selected in this study can be applied to identify different stock cultivars in seed production, culture and to establish cultivar certification system. The ornamental plant stock flower (Matthiola incana (L.) R. Br.) has been widely cultivated in China. In 2021, stem rot on M. incana , which caused plant death, was first observed in Shanghai, China. Field surveys indicated a disease incidence of 15%–20%. Following pathogen isolation, identification was performed by combining morphological observation of colonies, sclerotia and apothecia, with molecular methods, including DNA sequencing and phylogenetic analysis. Morphological characteristics of the isolates, including white cottony mycelium, blackish globular to irregularly shaped sclerotia and apothecia formation, were consistent with Sclerotinia sclerotiorum. Results of sequence comparison and phylogenetic analysis based on the internal transcribed spacer (ITS) sequences supported morphological data. Pathogenicity assays were conducted successfully according to Koch's postulates. Considering the results of the morphological and molecular identification, along with the pathogenicity assessment, S. sclerotiorum was confirmed as the causal agent of stem rot of stock flower in Shanghai, China. A combination of cultural practices, rotation of resistant ornamentals, and biological and chemical treatments is recommended to control stem rot of stock. • Matthiola incana was susceptible to Sclerotinia sclerotiorum. • Pathogen was identified as S. sclerotiorum by morphological and molecular methods. • This is the first report of S. sclerotiorum causing stem rot of M. incana in China. • An integrated approach to the management of stem rot is recommended.
