Introduction Home-based, virtually-supported care models may represent the most efficient and scalable approach to delivering prehabilitation services. However, virtual approaches to prehabilitation are understudied. This manuscript describes the protocol for an internal pilot randomised controlled trial of a virtually-delivered, multimodal prehabilitation intervention.Methods and analysis We will conduct a pragmatic, individual patient, internal pilot randomised controlled trial of home-based, virtually supported, multimodal prehabilitation compared with standard perioperative care in adults undergoing elective, inpatient thoracic, abdominal, pelvic and vascular surgery at five Canadian hospitals. Participants will be partially blinded; clinicians and outcome assessors will be fully blinded. The intervention consists of 3–12 weeks of a home-based, multimodal (exercise, nutrition and psychosocial support) prehabilitation programme supported through an online platform. The primary feasibility outcomes and their progression targets are (1) monthly recruitment of>6 participants at each centre, (2) intervention adherence of>75%, (3) retention of>90% of participants at the patient-reported primary outcome point of 30-days after surgery and (4) elicitation of patient, clinician and researcher-identified barriers to our pragmatic trial. A sample size of 144 participants will be adequate to estimate recruitment, adherence and retention rates with acceptable precision. All participants will be followed to either death or up to 1 year. As an internal pilot, if no substantive changes to the trial or intervention design are required, pilot participant outcome data will migrate, unanalysed by allocation, to the future full-scale trial.Ethics and dissemination Ethical approval has been granted by Clinical Trials Ontario (Project ID: 4479) and our ethics review board (Protocol Approval #20230399–01T). Results will be disseminated through presentations at scientific conferences, peer-reviewed publications, partner organisations and engagement of social and traditional media.Trial registration number ClinicalTrials.gov identifier NCT06042491. Background: Exposure to antiretrovirals at or early after HIV acquisition can suppress viral replication and blunt antibody (Ab) responses; a reduced HIV detectability could impact diagnosis and blood donation screening. Methods: We used three antigen (Ag)/Ab assays and one nucleic acid test (NAT) to analyze samples collected in pre-exposure prophylaxis (PrEP) trials (iPrEx; Partners PrEP) before infection detection by Ab-only rapid diagnostic tests (RDTs), and in early antiretroviral treatment (ART) initiation studies (RV254; SIPP). Results: Reactivity using NAT and Ag/Ab assays in samples collected up to 8 weeks prior to the first reactive RDT from 251 PrEP trials participants varied between 49-61% for active PrEP users and between 27-37% for placebo users. Abstract Modern height systems are based on the combination of satellite positioning and gravity field models of high resolution. However, in many regions, especially developing or newly industrializing countries, there is no (reliable) regional gravity model at all, due to challenges such as limited data availability, unknown/low data quality, and missing metadata. This paper addresses this issue in a case study of Colombia, where eight decades of historical terrestrial and airborne gravity measurements are available but widely contain systematic errors, outliers, and biases. Correspondingly, processing strategies and structures are proposed and applied to validate and improve the quality of old gravity datasets. A novel method is developed based on spherical radial basis functions (SRBFs) for estimating biases, which are found in different airborne surveys with values exceeding 40 mGal. The validity of this bias estimation method is demonstrated both by a simulation test and by the evaluation of the airborne data in comparison to the SATOP (SAtellite-TOPography) model, which merges the satellite-only global gravity model GOCO06s with the Earth2014 topography model. The terrestrial and airborne data are then combined with a global gravity model (GGM), ultra-high-resolution topography models, as well as altimetry-derived gravity anomalies from DTU21GRA for the offshore areas. The results are presented in terms of height anomalies (QGeoidCOL2023), and they are thoroughly validated using GPS/leveling data both in the absolute and relative manner. The standard deviation in comparison to the GPS/leveling data after applying a correction surface to account for the datum inconsistencies amounts to 15.76 cm, which is 27% smaller compared to the mean standard deviation value given by five recent high-resolution GGMs, and 36% smaller than the one delivered by the latest South American quasi-geoid model QGEOID2021. The relative validation results show that QGeoidCOL2023 performs better, i.e., delivers lower RMS errors than the GGMs and QGEOID2021 in all the baseline length groups. These results indicate the validity and benefits of the developed methods and procedures, which can be used for other data-challenging areas to facilitate the realization of geopotential-based height systems. Abstract The heterogeneity of the whole-exome sequencing (WES) data generation methods present a challenge to a joint analysis. Here we present a bioinformatics strategy for joint-calling 20,504 WES samples collected across nine studies and sequenced using ten capture kits in fourteen sequencing centers in the Alzheimer’s Disease Sequencing Project. The joint-genotype called variant-called format (VCF) file contains only positions within the union of capture kits. The VCF was then processed specifically to account for the batch effects arising from the use of different capture kits from different studies. We identified 8.2 million autosomal variants. 96.82% of the variants are high-quality, and are located in 28,579 Ensembl transcripts. 41% of the variants are intronic and 1.8% of the variants are with CADD > 30, indicating they are of high predicted pathogenicity. Here we show our new strategy can generate high-quality data from processing these diversely generated WES samples. The improved ability to combine data sequenced in different batches benefits the whole genomics research community.

Abstract In recent years, the laser has become an important tool in hospitals. Laser surgery in particular has many advantages. However, there is still a lack of the understanding of the influence of the relevant parameters for laser surgery. In order to fill this gap, the parameters pulse frequency, use of an exhaustion system, air cooling, laser power, laser scan speed, laser line energy and waiting time between cuts were analysed by ANOVA using inter-animal variation as a benchmark. The quality of the cuts was quantized by a previously published scoring system. A total of 1710 cuts were performed with a $$\text {CO}_2$$ CO 2 laser. Of the parameters investigated, laser power and scan speed have the strongest influence. Only the right combination of these two parameters allows good results. Other effects, such as the use of pulsed or continuous wave (CW) laser operation, or air cooling, show a small or negligible influence. By modulating only the laser power and scan speed, an almost perfect cut can be achieved with a $$\text {CO}_2$$ CO 2 laser, regardless of the external cooling used or the laser pulse duration or repetition rate from CW to nanosecond pulses.

Sialic acids are found as terminal sugars on glycan structures on cellular surfaces. T cells carry these sialoglycans abundantly, and they are thought to serve multiple functions in cell adhesion, cell migration, and protection from complement attack. We studied the role of sialoglycans on T cells in a mouse model with a T cell-specific deletion of cytidine monophosphate-sialic acid synthase (CMAS), the enzyme that is crucial for the synthesis of sialoglycans. These mice showed a T-cell deficiency in peripheral lymphoid organs. Many T cells with an undeleted Cmas allele were found in the periphery, suggesting that they escaped the Cre-mediated deletion. The remaining peripheral T cells of T cell-specific Cmas KO mice had a memory-like phenotype. Additional depletion of the complement factor C3 could not rescue the phenotype, showing that the T-cell defect was not caused by a host complement activity. Cmas-deficient T cells showed a high level of activated caspase 3, indicating an ongoing apoptosis. In bone marrow chimeric cellular transfer experiments, we observed a strong competitive disadvantage of Cmas-deficient T cells compared to wild-type T cells. These results show that sialoglycans on the surface of T cells are crucial for T-cell survival and maintenance. This function has not been recognized before and is similar to the function of sialoglycans on B cells.

In recent years, the topic of beam shaping for improved laser material processing has rapidly grown influencing metal laser powder bed fusion (PBF-LB/M). Given the need to reduce the cost and improve control of the PBF-LB/M process to make it more competitive with traditional manufacturing methods, increasing productivity of PBF-LB/M is critical. When research reports a new beam profile (e.g. ring profile) to improve productivity on a specific material, it is often generalised, and assumed to have the capability to improve productivity in PBF-LB/M across the board. In this work, we use both low-fidelity simulations and experimental work to investigate the difference between Gaussian and ring/spot beam profiles on metals with very different thermal properties (a stainless steel and an aluminium alloy). We show that the two materials have opposite responses to the change in beam profile (both in terms of melt pool dimensions and thermal gradients); further, the most beneficial intensity distribution is dependent on the energy input to the material. This exemplifies yet another way in which the PBF-LB/M process is non-linear and contradicts the idea that a ring/spot laser profile is beneficial for all laser processing technologies. This highlights the need for further research into the non-linear effect of varying intensity distributions on laser processing before the benefits of dynamic beam shaping can be truly realised.

A promising approach for producing parts with outstanding properties in directed energy deposition (DED-LB/M) provides the application of tailored powder mixtures processed by applying in situ alloying strategies. In this work, DED-LB/M was used to manufacture multilayer specimens from AISI H11 steel powders enriched with carbon nanoparticles (C-np) in concentrations of 0.1 wt.-% and 0.2 wt.-%. The scientific aim was to investigate the impact of C-np on the microstructural (particularly retained austenite content (RA-c) and grain size) and mechanical properties (specifically hardness and compression yield strength) of the manufactured specimens. It was shown that the addition of C-np to the H11 powder leads to a stronger distortion of martensite as well as significantly enhancing the RA-c. Furthermore, the C-np seem to favor the formation of finer martensite, as can be verified with XRD and EBSD. Under as-built conditions, the mean hardness increases from 653 ± 10 HV1 for the H11 sample to 770 ± 14 HV1 for the sample reinforced with 0.2 wt.-% C-np. At the same time, Y0.2% rises up from 1839 ± 61 MPa to 2134 ± 68 MPa. The hardness- and strength-increasing effect of the added C-np is retained even after heat treatment, similarly to the industrial standard.

Abstract This paper presents an approach for achieving full optical photoacoustic imaging with enhanced resolution utilizing speckle pattern analysis. The proposed technique involves projecting patterns derived from binary masks corresponding to orthogonal functions onto the target to elicit a photoacoustic signal. The resulting signal is then recorded using a high-speed camera and analyzed using correlation analysis of the speckle motion. Our results demonstrate the feasibility of this optical approach to achieve imaging with enhanced resolution without the need for physical contact with the target, opening up new possibilities for non-invasive medical imaging and other applications.

Laser-based directed energy deposition of metals (DED-LB/M) supports the synthesis of functional materials with tailored properties and performance through in-situ modification of the alloying composition within the processing zone. In this investigation, a low-alloyed steel was modified stepwise to analyse the influence of carbon and tungsten carbide (WC) addition on the resulting material properties. A moderate carbon concentration of 0.4 wt.-% improved the average hardness (520 HV0.5). WC particles on the other hand were dissolved within the matrix and resulted in a fine microstructure with high hardness (780 HV0.5). A combined addition of carbon and WC led to the highest material hardness (840 HV0.5). Scratch tests showed that the wear resistance rises with increasing hardness but is improved the most by the addition of hard particles. Furthermore, these tests revealed an anisotropic abrasive wear resistance which correlates with the direction of the weld tracks. Loading the material parallel to the weld track direction led to a homogeneous wear. When the material is scratched perpendicularly to the weld tracks, an inhomogeneous wear with periodic characteristics occurred. The periodicity can be explained by the different microstructural characteristics and hardness at the transition zone between adjacent weld tracks deposited in DED-LB/M. For all materials, the transition between two weld tracks was characterized by a columnar microstructure with low microhardness while the adjacent weld tracks possessed a finer microstructure and higher microhardness. These microstructural differences were mirrored in scratch testing since wear peaks can be observed at the transition zone between two weld tracks.

Case-hardening steels are gaining increasing interest in the field of laser powder bed fusion (PBF-LB/M) due to their excellent weldability. In combination with post-process carburization heat treatment, the surface properties can be improved to generate high-strength products. When manufacturing larger products by means of PBF-LB/M, the in situ heat accumulation and the altered cooling rates affect the resulting material properties. Therefore, the fabrication of larger products requires an understanding on the influencing factors that affect the material properties. This work investigates the effect of different volumetric energy densities (VED) on the resulting microstructural and mechanical properties. It is found that the hardness decreases continuously along the build direction. The gradient depends on the applied energy and is stronger for higher energy inputs due to heat accumulation and lowered cooling rates. Furthermore, countering strategies are investigated to avoid process-specific hardness reduction along the build direction. This includes a reduced number of parts within the build job as well as a modified inter-layer time (ILT) between consecutive layers of the specimen. Applying a moderate inter-layer time helps to counter process-specific overheating, which is indicated by an almost homogeneous material hardness and melt pool size along the build direction.

Treated municipal wastewater (TMW) can provide a reliable source of irrigation water for crops, which is especially important in arid areas where water resources are limited or prone to drought. Nonetheless, TMW may contain residual antibiotics, potentially exposing the crops to these substances. The goal of this study was to investigate the dissemination of antibiotics resistance genes (ARGs) in the soil–plant–earthworm continuum after irrigation of spinach and radish plants with TMW containing trimethoprim, sulfamethoxazole, and sulfapyridine in a greenhouse experiment, followed by feeding of earthworms with harvested plant materials. Our results showed that antibiotic resistance genes (ARGs) were enriched in the soil–plant–earthworm microbiomes irrigated with TMW and TMW spiked with higher concentrations of antibiotics. The number of ARGs and antibiotic-resistant bacteria (ARB) enrichment varied with plant type, with spinach harboring a significantly higher amount of ARGs and ARB compared to radish. Our data showed that bulk and rhizosphere soils of spinach and radish plants irrigated with MilliQ water, TMW, TMW10, or TMW100 had significant differences in bacterial community (p 90 %) of bacterial MAG of genus Leclercia adecarboxylata B from the fecal microbiome of earthworm that was fed harvested radish tubers and spinach leaves grown on TMW10 irrigated waters, and this bacterium has been shown to be an emerging pathogen causing infection in immunocompromised patients that may lead to health complications and death. Therefore, crops irrigated with TMW containing residual antibiotics and ARGs may lead to increased incidences of enrichment of ARB in the soil–plant–earthworm continuum.

Abstract Dynamic phase-only beam shaping with a liquid crystal spatial light modulator is a powerful technique for tailoring the intensity profile or wave front of a beam. While shaping and controlling the light field is a highly researched topic, dynamic nonlinear beam shaping has hardly been explored so far. One potential reason is that generating the second harmonic is a degenerate process as it mixes two fields at the same frequency. To overcome this problem, we propose the use of type II phase matching as a control mechanism to distinguish between the two fields. Our experiments demonstrate that distributions of arbitrary intensity can be shaped in the frequency-converted field at the same quality as for linear beam shaping and with conversion efficiencies similar to without beam shaping. We envision this method as a milestone toward beam shaping beyond the physical limits of liquid crystal displays by facilitating dynamic phase-only beam shaping in the ultraviolet spectral range.

Abstract We investigate if the projected high-precision measurements of the cross section of the Higgsstrahlung process $$e^+ e^- \rightarrow Zh$$ e + e - → Z h at a future electron–positron collider can be utilised to indirectly probe the fermionic Seesaw models. We consider the two centre-of-mass energies $$\sqrt{s}=240$$ s = 240 GeV and 365 GeV, and compare the collider reaches to constraints from electroweak observables, probes of lepton flavour universality and the existing and prospective bounds from searches for lepton flavour violation. For the analysis we assume the limit of an exactly conserved lepton-number symmetry. We find that while any appreciable correction to the Higgsstrahlung cross section is already strictly constrained in the Type-I Seesaw model, effects of up to $${\mathcal {O}}(10\%)$$ O ( 10 % ) are possible within Type-III Seesaw.

Objectives: In this early retrospective cohort study, a total of 26 patients with SARS-CoV-2 were treated with bamlanivimab or casirivimab/imdevimab, and the reduction of the viral load associated with the developed clinical symptoms was analyzed. Methods: Patients in the intervention groups received bamlanivimab or casirivimab/imdevimab. Patients without treatment served as control. Outcomes were assessed by clinical symptoms and change in log viral load from baseline based on the cycle threshold over a period of 18 days. Results: Median log viral load decline was higher in both intervention groups after 3 and 6 days compared to control. However, at later time points, the decline of the viral load was more distinct in the control group. Mild symptoms of COVID-19 were observed in 6.3% of the intervention groups and in no patient of the control. No patients treated with bamlanivimab, 18.8% treated with casirivimab/imdevimab, and 14.2% in the control group developed moderate symptoms. Severe symptoms were recorded only in the control group (14.2%), including one related death. Conclusion: Treatment with monoclonal SARS-CoV-2 antibodies seems to accelerate decline of virus loads, especially in the first 6 days after administration, compared to control. This may be associated with a reduced likeliness of a severe course of COVID-19.

A planar volume dielectric barrier discharge (DBD) in pure carbon dioxide (CO2) for the formation of carbon monoxide (CO) is examined by combined electrical and CO density measurements. The influence of the type of electrode, the barrier material, the barrier thickness, and the discharge gap on the plasma power and the CO formation is analyzed systematically. The electrical characterization by means of charge-voltage plots is based on the simplest equivalent circuit model of DBDs, extended by the so-called partial surface discharge effect and the presence of parallel parasitic capacitances. The stackable discharge arrangement in this study enables one to elucidate the influence of parasitic capacitances, which can be overlooked in the application of such plasma sources. The determination of the discharge voltage from charge-voltage plots and the validity of the so-called Manley power equation are revised by taking into account non-uniform coverage as well as parasitic capacitances. The energy yield (EY) of CO is analyzed and compared with the literature. No correlations of EY with the mean reduced electric field strength or the geometric parameters of the DBD arrangement are observed.

The laser-based directed energy deposition of polymers (DED-LB/P) is an additive manufacturing process which offers the possibility to build up 3D structures with a high level of individualization on free-form surfaces. In this article, an absorber free DED-LB/P process with a thulium-doped fiber laser is presented for the first time to produce dielectric image lines (DIL) consisting of polypropylene (PP) for high frequency applications. This type of transmission line has advantages over the commonly used stripline technology due to its low signal attenuation, especially at high frequencies, but is currently hardly found in mass-market applications because no suitable manufacturing technology has been established for it yet. The steadily increasing demand of higher bandwidth in telecommunication or increased resolution of radar sensors leads to higher application frequencies and alternative interconnect technologies will become more important as they significantly define the achievable overall system performance. Based on the fabricated low-loss DILs in this study, DED-LB/P could represent such an alternative manufacturing process for future RF-applications.

The ionospheric refraction of GNSS signals can have an impact on positioning accuracy, especially in cases of single-frequency observations. Ionosphere models that are broadcasted by the satellite systems (e.g., Klobuchar, NeQuick-G) do not include enough details to permit them to correct single-frequency observations with sufficient accuracy. To address this issue, regional ionosphere models (RIMs) have been developed in several countries in the western Balkans based on dense Continuous Operating Reference Stations (CORS) observations. Subsequently, a RIM for the western Balkans was built using an artificial neural network that combined regional ionosphere parameters estimated from the CORS data with spatiotemporal (latitude, longitude, hour of day), solar (F10.7) and geomagnetic (Kp, Dst) parameters. The RIMs were tested at the solar maximum (March 2014), a geomagnetic storm (March 2015), and the solar minimum (March 2018). The new RIMs mimic the integrated electron density much more effectively than the Klobuchar model. Furthermore, RIMs significantly reduce the ionospheric effects on single-frequency positioning, indicating their necessity for use in positioning applications.

In this study, the DED-LB/M process of AISI H11 tool steel powder blends modified by adding WC nanoparticles (WC-np) in concentrations of 1, 2.5 and 5 wt.-% was the object of scientific investigations. For this, 30-layer cuboid specimens were manufactured. The overall scientific aim was to examine how the WC-np interact with the steel melt and in the end, influence the processability, microstructure and mechanical properties of produced specimens. The examinations were carried out on both as-built and thermally post-processed specimens. An advanced microstructural analysis (SEM, EDS, EBSD and XRD) revealed that due to the high solubility of WC-np in the molten steel, most of the WC-np appear to have dissolved during the ongoing laser process. Furthermore, the WC-np favor a stronger distortion and finer grain size of martensite in the manufactured specimens. An increase in hardness from about 650 HV1 for the H11 specimen to 780 HV1 for the one manufactured using the powder blend containing 5 wt.-% of WC-np was observed in as-built conditions. In the same way, the compression yield strength enhanced from 1839 MPA to 2188 MPA. The hardness and strength increasing effect of WC-np remained unchanged even after heat treatments similar to those used in industry.

Natural aromas like cinnamaldehyde are suitable solvents to extract curcuminoids, the active ingredients found in the rhizomes of Curcuma longa L. In a pursuit to find other nature-based solvents, capable of solving curcumin, forty fragrances and flavours were investigated in terms of their solubilisation power. Aroma compounds were selected according to their molecular structure and functional groups. Their capabilities of solving curcumin were examined by UV–Vis spectroscopy and COSMO-RS calculations. The trends of these calculations were in accordance with the experimental solubilisation trend of the solubility screening and a list with the respective curcumin concentrations is given; σ-profiles and Gibbs free energy were considered to further investigate the solubilisation process of curcumin, which was found to be based on hydrogen bonding. High curcumin solubility was achieved in the presence of solvent (mixtures) with high hydrogen-bond-acceptor and low hydrogen-bond-donor abilities, like γ- and δ-lactones. The special case of DMSO was also examined, as the highest curcumin solubility was observed with it. Possible specific interactions of selected aroma compounds (citral and δ-hexalactone) with curcumin were investigated via 1H NMR and NOESY experiments. The tested flavours and fragrances were evaluated regarding their potential as green alternative solvents.

Abstract Many modern applications, such as precise point positioning, autonomous driving or precision agriculture would benefit significantly if a high-precision and high-resolution model of electron density in the ionosphere and the plasmasphere would be globally available. Since the development of such a model still relies on data with insufficient and uneven global coverage, the consideration of background information and the introduction of equality and inequality constraints on Chapman key parameters are essential. In this work, we develop a multi-layer Chapman model based on B-spline expansions of selected key parameters of the electron density. The unknown series coefficients of the key parameters are subject to equality and inequality constraints. Finally, the developed model is applied to a combination of real and semi-simulated input data; the results are validated through ionosonde measurements. Graphical Abstract

Abstract Dynamical changes in the ionosphere and thermosphere during geomagnetic storm times can have a significant impact on our communication and navigation applications, as well as satellite orbit determination and prediction activities. Because of the complex electrodynamics coupling processes during storms, which cannot be fully described with the sparse set of thermosphere–ionosphere (TI) observations, it is crucial to accurately model the state of the TI system. The approximation closest to the true state can be obtained by assimilating relevant measurements into physics-based models. Thermospheric mass density (TMD) derived from satellite measurements is ideal to improve the thermosphere through data assimilation. Given the coupled nature of the TI system, the changes in the thermosphere will also influence the ionosphere state. This study presents a quantification of the changes and improvement of the model state produced by assimilating TMD not only for the thermosphere density but also for the ionosphere electron density under storm conditions. TMD estimates derived from a single Swarm satellite and the Coupled Thermosphere Ionosphere Plasmasphere electrodynamics (CTIPe) physics-based model are used for the data assimilation. The results are presented for a case study during the St. Patricks Day storm 2015. It is shown that the TMD data assimilation generates an improvement of the model’s thermosphere density of up to 40% (measured along the orbit of the non-assimilated Swarm satellites). The model’s electron density during the course of the storm has been improved by approximately 8 and 22% relative to Swarm-A and GRACE, respectively. The comparison of the model’s global electron density against a high-quality 3D electron density model, generated through assimilation of total electron content, shows that TMD assimilation modifies the model’s ionosphere state positively and negatively during storm time. The major improvement areas are the mid-low latitudes during the storm’s recovery phase. Graphical Abstract

Abstract The idea of laser surgery is nearly as old as the laser itself. From the first trials to modern laser surgery systems, it was and is the aim to selectively cut the tissue in the focus spot without causing harm to surrounding structures. This is only possible when the correct parameters for the surgical laser are chosen. Usually, this is done by parameter studies. However, the concrete evaluation scheme often differs between groups and more precise approaches require staining and microscopic evaluation. To overcome these issues, a macroscopic scoring system is presented and evaluated. It can be shown that the scoring system works well and, thus, a laser cut can be evaluated within a few seconds. At the same time, the whole cutting front is taken into account. The presented scoring system is evaluated by the intra class correlation (ICC). The final agreement between different raters is more than 0.7. Therefore, the scoring system can be used to optimize and evaluate the cutting process and it should be suitable for comparing the results between different groups. Definitely, it can be applied for scoring within a group to enable e.g., a profound statistical analysis for a parameter study.

Abstract Background Chronic kidney disease (CKD) is a global burden affecting both children and adults. Novel imaging modalities hold great promise to visualize and quantify structural, functional, and molecular organ damage. The aim of the study was to visualize and quantify murine renal vasculature using label-free raster scanning optoacoustic mesoscopy (RSOM) in explanted organs from mice with renal injury. Material and methods For the experiments, freshly bisected kidneys of alpha 8 integrin knock-out (KO) and wildtype mice (WT) were used. A total of n=7 female (n=4 KO, n=3 WT) and n=6 male animals (n=2 KO, n=4 WT) aged 6 weeks were examined with RSOM optoacoustic imaging systems (RSOM Explorer P50 at SWL 532nm and/or ms-P50 imaging system at 532 nm, 555 nm, 579 nm, and 606 nm). Images were reconstructed using a dedicated software, analyzed for size and vascular area and compared to standard histologic sections. Results RSOM enabled mapping of murine kidney size and vascular area, revealing differences between kidney sizes of male (m) and female (f) mice (merged frequencies (MF) f vs. m: 52.42±6.24 mm2 vs. 69.18±15.96 mm2, p=0.0156) and absolute vascular area (MF f vs. m: 35.67±4.22 mm2 vs. 49.07±13.48 mm2, p=0.0036). Without respect to sex, the absolute kidney area was found to be smaller in knock-out (KO) than in wildtype (WT) mice (WT vs. KO: MF: p=0.0255) and showed a similar trend for the relative vessel area (WT vs. KO: MF p=0.0031). Also the absolute vessel areas of KO compared to WT were found significantly different (MF p=0.0089). A significant decrease in absolute vessel area was found in KO compared to WT male mice (MF WT vs. KO: 54.37±9.35 mm2 vs. 34.93±13.82 mm2, p=0.0232). In addition, multispectral RSOM allowed visualization of oxygenated and deoxygenated parenchymal regions by spectral unmixing. Conclusion This study demonstrates the capability of RSOM for label-free visualization of differences in vascular morphology in ex vivo murine renal tissue at high resolution. Due to its scalability optoacoustic imaging provides an emerging modality with potential for further preclinical and clinical imaging applications.

BackgroundRecent data on immune evasion of new SARS-CoV-2 variants raise concerns about the efficacy of antibody-based COVID-19 therapies. Therefore, in this study the in-vitro neutralization capacity against SARS-CoV-2 variant B.1 and the Omicron subvariants BA.1, BA.2 and BA.5 of sera from convalescent individuals with and without boost by vaccination was assessed.Methods and findingsThe study included 313 serum samples from 155 individuals with a history of SARS-CoV-2 infection, divided into subgroups without (n=25) and with SARS-CoV-2 vaccination (n=130). We measured anti-SARS-CoV-2 antibody concentrations by serological assays (anti-SARS-CoV-2-QuantiVac-ELISA (IgG) and Elecsys Anti-SARS-CoV-2 S) and neutralizing titers against B.1, BA.1, BA.2 and BA.5 in a pseudovirus neutralization assay. Sera of the majority of unvaccinated convalescents did not effectively neutralize Omicron sublineages BA.1, BA.2 and BA.5 (51.7%, 24.1% and 51.7%, resp.). Abstract Multi- and hyperspectral endoscopy are possibilities to improve the endoscopic detection of neoplastic lesions in the colon and rectum during colonoscopy. However, most studies in this context are performed on histological samples/biopsies or ex vivo. This leads to the question if previous results can be transferred to an in vivo setting. Therefore, the current study evaluated the usefulness of multispectral endoscopy in identifying neoplastic lesions in the colon. The data set consists of 25 mice with colonic neoplastic lesions and the data analysis is performed by machine learning. Another question addressed was whether adding additional spatial features based on Gauss–Laguerre polynomials leads to an improved detection rate. As a result, detection of neoplastic lesions was achieved with an MCC of 0.47. Therefore, the classification accuracy of multispectral colonoscopy is comparable with hyperspectral colonoscopy in the same spectral range when additional spatial features are used. Moreover, this paper strongly supports the current path towards the application of multi/hyperspectral endoscopy in clinical settings and shows that the challenges from transferring results from ex vivo to in vivo endoscopy can be solved.

A surge in successful Information Security (IS) breaches targeting Research and Education (R&E) institutions highlights a pressing need for enhanced protection. Addressing this, a consortium of European National Research and Education Network (NREN) organizations has developed a unified IS framework. This paper aims to introduce the Security Baseline for NRENs and a security maturity model tailored for R&E entities, derived from established security best practices to meet the specific needs of NRENs, universities, and various research institutions. The models currently in existence do not possess a system to smoothly correlate varying requirement tiers with distinct user groups or scenarios, baseline standards, and existing legislative actions. This segmentation poses a significant hurdle to the community’s capacity to guarantee consistency, congruency, and thorough compliance with a cohesive array of security standards and regulations. By employing taxonomical engineering principles, a mapping of baseline requirements to other security frameworks and regulations has been established. This reveals a correlation across most regulations impacting R&E institutions and uncovers an overlap in the high-level requirements, which is beneficial for the implementation of multiple standards. Consequently, organizations can systematically compare diverse security requirements, pinpoint gaps in their strategy, and formulate a roadmap to bolster their security initiatives.

Predictive control has great potential in the home energy management domain. However, such controls need reliable predictions of the system dynamics as well as energy consumption and generation, and the actual implementation in the real system is associated with many challenges. This paper presents the implementation of predictive controls for a heat pump with thermal storage in a real single-family house with a photovoltaic rooftop system. The predictive controls make use of a novel cloud camera-based short-term solar energy prediction and an intraday prediction system that includes additional data sources. In addition, machine learning methods were used to model the dynamics of the heating system and predict loads using extensive measured data. The results of the real and simulated operation will be presented.

The topographical information of a weld seam bears information about quality relevant characteristics such as humping or spatter. Optical coherence tomography (OCT) can be used for inline scanning the weld topography coaxially mounted at a laser scanning optic. Feature extraction from this topographical information is challenging due to finding mathematical representations for the identification of relevant features. Feature extraction based on scalable hypothesis tests (FRESH) allows for feature extraction by a combination of various time series characterization methods. FRESHs feature selection is supported with an automatically configured hypothesis test and hence allows for quick extraction of significant features from sensing data in laser welding processes. In this work, a proof-of-concept is demonstrated for weld result categorization from OCT data by feature extraction using the FRESH algorithm. Changes in weld topography are characterized in a vast variety of process parameters for weld categories such as spatter, deep penetration welding, humping and heat conduction welding. As a result, a quantified separation of weld categories is possible and shows the feasibility of the FRESH algorithm for future quality assessments with different sensing technologies in laser welding.

Abstract Various clinically applicable scores and indices are available to help identify the state of a microcirculatory disorder in a patient. Several of these methods, however, leave room for interpretation and only provide clues for diagnosis. Thus, a measurement method that allows a reliable detection of impending or manifest circulatory malfunctions would be of great value. In this context, the optical and non-invasive method of shifted position-diffuse reflectance imaging (SP-DRI) was developed. It allows to determine the capillary diameter and thus to assess the state of the microcirculation. The aim of the present study is to investigate how the quantification of capillary diameters by SP-DRI behaves in different individuals, i.e. for a wide range of optical properties. For this, within Monte-Carlo simulations all optical properties (seven skin layers, hemoglobin) were randomly varied following a Gaussian distribution. An important finding from the present investigation is that SP-DRI works when the optical properties are chosen randomly. Furthermore, it is shown that appropriate data analysis allows calibration-free absolute quantification of the capillary diameter across individuals using SP-DRI. This underpins the potential of SP-DRI to serve as an early alert system for the onset of microcirculatory associated diseases.

Context: Recurrent caries are the leading cause of composite resin failure. Aims: The purpose of this pilot study was to test the efficacy of a novel copper iodide (CuI) containing dental adhesive in an in vitro caries model. Subjects and Methods: Streptococcus mutans and Lactobacillus acidophilus were grown individually on the complex medium for 48 h at 37°C. The pH of the mixed medium was 7.0 initially and tested every 24 h. 40 extracted teeth were prepared with standardized cavity preparations and coated with control or experimental CuI adhesives and imaged using a micro-computed tomography (microCT). Four study groups were evaluated: (1) control (2) 0.5 μg/ml CuI (3) 1.0 μg/ml CuI, 4) 5.0 μg/ml CuI. After incubation, the teeth were re-imaged using the microCT. Utilizing AnalyzePro software the three-dimensional data sets were overlaid and demineralization was measured and statistics were run. Statistics: Stratified ANOVA models were run to determine if there were differences between the control and experimental adhesive groups. Similarly, pH and bacterial concentrations were evaluated to ensure the viability of polymicrobial specimen. Results and Conclusions: Significant differences were found between the control group and the 1.0 and 5.0 CuI adhesive groups. No differences in pH were noted between the groups. Overlaid changes in demineralization were recorded as volume loss. CuI adhesives with 5 mg/ml or higher have the potential to limit tooth demineralization after bacterial penetration of a dental restoration in an in vitro caries model. Further testing is needed.

BackgroundWaterpipe (WP) use is rapidly increasing among young people worldwide due to the widespread misperception that it is safer than cigarette smoking. Health warning labels (HWLs) can effectively communicate tobacco-related health risks but have yet to be developed for WP. This study aimed to optimize and adapt a set of 16 pictorial WP-specific HWLs, developed by an international Delphi study, to the Tunisian context. HWLs were grouped into four themes: WP health risks, WP harm to others, WP-specific harms, and WP harm compared to cigarettes.MethodsUsing a mixed method approach, we conducted ten focus groups combined with a survey among young WP users and nonusers (N = 63; age 18-34 years). In the survey, participants rated the HWLs on several communication outcomes (e.g., reaction, harm perception, effectiveness) and were then instructed to view all HWLs in each theme and rank them in the order of overall perceived effectiveness, from the most to the least effective. Afterward, participants provided in-depth feedback on HWLs and avenues for improvement. Mean effectiveness rating scores and percentages of participants' top-ranked HWLs were calculated. Discussions were audio-taped, transcribed verbatim, and analyzed thematically.ResultsThe top-ranked HWLs were those showing oral cancers, orally transmitted diseases, and a sick child. Focus group discussion illustrated that these selections were based on participants' reactions to the direct impact of WP on a person's physical appearance and evoking guilt over children's exposure to WP smoke. Suggestions for improvement highlighted the need to use the local dialect and more affirmative statements (e.g., avoiding "may" or "can").ConclusionsThis study is the first in North Africa to attempt to advance HWLs policy as the World Health Organization recommended. The results of this study can be used as a basis for implementing WP-specific health messages in the Eastern Mediterranean Region.

BACKGROUND Results of many randomized trials on COVID-19 convalescent plasma (CCP) have been reported, but information on long-term outcome after CCP treatment is limited. The objectives of this extended observation of the randomized CAPSID trial are to assess long-term outcome and disease burden in patients initially treated with or without CCP.METHODS Of 105 randomized patients, 50 participated in the extended observation. Quality of life (QoL) was assessed by questionnaires and a structured interview. CCP donors (n = 113) with asymptomatic to moderate COVID-19 were included as a reference group.RESULTS The median follow-up of patients was 396 days, and the estimated 1-year survival was 78.7% in the CCP group and 60.2% in the control (P = 0.08). The subgroup treated with a higher cumulative amount of neutralizing antibodies showed a better 1-year survival compared with the control group (91.5% versus 60.2%, P = 0.01). Medical events and QoL assessments showed a consistent trend for better results in the CCP group without reaching statistical significance. There was no difference in the increase in neutralizing antibodies after vaccination between the CCP and control groups.CONCLUSION The trial demonstrated a trend toward better outcome in the CCP group without reaching statistical significance. A predefined subgroup analysis showed a significantly better outcome (long-term survival, time to discharge from ICU, and time to hospital discharge) among those who received a higher amount of neutralizing antibodies compared with the control group. Multi-material additive manufacturing (AM) attempts to utilize the full benefits of complex part production with a comprehensive and complementary material spectrum. In this context, this research article presents new processing strategies in the field of polymer- and metal-based multi-material AM. The investigation highlights the current progress in powder-based multi-material AM based on three successfully utilized technological approaches: additive and formative manufacturing of hybrid metal parts with locally adapted and tailored properties, material-efficient AM of multi-material polymer parts through electrophotography, and the implementation of UV-curable thermosets within the laser-based powder bed fusion of plastics. Owing to the complex requirements for the mechanical testing of multi-material parts with an emphasis on the transition area, this research targets an experimental shear testing set-up as a universal method for both metal- and polymer-based processes. The method was selected based on the common need of all technologies for the sufficient characterization of the bonding behavior between the individual materials.

Glioblastoma multiforme (GBM) is at present an incurable disease with a 5-year survival rate of 5.5%, despite improvements in treatment modalities such as surgery, radiation therapy, chemotherapy [e.g., temozolomide (TMZ)], and targeted therapy [e.g., the antiangiogenic agent bevacizumab (BEV)]. Diffusing alpha-emitters radiation therapy (DaRT) is a new modality that employs radium-224-loaded seeds that disperse alpha-emitting atoms inside the tumor. This treatment was shown to be effective in mice bearing human-derived GBM tumors. Here, the effect of DaRT in combination with standard-of-care therapies such as TMZ or BEV was investigated. In a viability assay, the combination of alpha radiation with TMZ doubled the cytotoxic effect of each of the treatments alone in U87 cultured cells. A colony formation assay demonstrated that the surviving fraction of U87 cells treated by TMZ in combination with alpha irradiation was lower than was achieved by alpha- or x-ray irradiation as monotherapies, or by x-ray combined with TMZ. The treatment of U87-bearing mice with DaRT and TMZ delayed tumor development more than the monotherapies. Unlike other radiation types, alpha radiation did not increase VEGF secretion from U87 cells in culture. BEV treatment introduced several days after DaRT implantation improved tumor control, compared to BEV or DaRT as monotherapies. The combination was also shown to be superior when starting BEV administration prior to DaRT implantation in large tumors relative to the seed size. BEV induced a decrease in CD31 staining under DaRT treatment, increased the diffusive spread of 224Ra progeny atoms in the tumor tissue, and decreased their clearance from the tumor through the blood. Taken together, the combinations of DaRT with standard-of-care chemotherapy or antiangiogenic therapy are promising approaches, which may improve the treatment of GBM patients.

The sudden increase of X-radiation and EUV emission following solar flares causes additional ionization and increased absorption of electromagnetic (EM) waves in the Earth’s atmosphere. The solar flare impact on the ionosphere above Europe on 05 and 06 December 2006 was investigated using ground-based (ionosonde and VLF) and satellite-based data (Vertical Total Electron Content (VTEC) derived from GNSS observations and VLF measurements from DEMETER satellite). Based on the Kp and Dst indices, 05 December 2006 was a quiet day, while there was a geomagnetic storm on 06 December 2006. The total fade-out of the EM waves emitted by the ionosondes was experienced at all investigated stations during an X9 class flare on 05 December 2006. The variation of the fmin parameter (first echo trace observed on ionograms, it is a rough measure of the “non-deviative” absorption) and its difference between the quiet period and during the flares have been analyzed. A latitude dependent enhancement of fmin (2–9 MHz) and Δfmin (relative change of about 150%–300%) was observed at every station at the time of the X9 (on 05 December) and M6 (on 06 December) flares. Furthermore, we analyzed VTEC changes during and after the flare events with respect to the mean VTEC values of reference quiet days. During the X9 solar flare, VTEC increased depending on the latitude (2–3 TECU and 5%–20%). On 06 December 2006, the geomagnetic storm increased ionization (5–10 TECU) representing a “positive” ionospheric storm. However, an additional peak in VTEC related to the M6 flare could not be detected. We have also observed a quantifiable change in transionospheric VLF absorption of signals from ground transmitters detected in low Earth orbit associated with the X9 and M6 flare events on 05 and 06 December in the DEMETER data. Moreover, amplitude and phase of ground-based, subionospherically propagating VLF signals were measured simultaneously during the investigated flares to analyze ionosphere reaction and to evaluate the electron density profile versus altitude. For the X9 and M6 flare events we have also calculated the ionospheric parameters (sharpness, reflection height) important for the description and modelling of this medium under forced additional ionization.

The commonly used Gaussian intensity distribution during the laser-based processing of metals can negatively affect melt pool stability, which might lead to defects such as porosity, hot cracking, or poor surface quality. Hot cracking is a major factor in limiting production rates of high-strength aluminium alloys in laser-based processes such as welding or the powder bed fusion of metals (PBF-LB/M). Going away from a Gaussian intensity distribution to ring-shaped profiles allows for a more even heat distribution during processing, resulting in more stable melt pools and reduced defect formations. Therefore, the aim of this study is to investigate the influence of different laser beam profiles on the processing of high-strength aluminium alloys by using a multicore fiber laser, allowing for in-house beam shaping. Single weld tracks on the aluminium alloy EN AW-5083 are produced with varying laser powers and weld speeds, as well as different beam profiles, ranging from Gaussian intensity distribution to point/ring profiles. The molten cross sections are analyzed regarding their geometry and defects, and the surface roughness of the weld tracks is measured. By using point/ring beam profiles, the processing window can be significantly increased. Hot cracking is considerably reduced for weld speeds of up to 1000 mm/s compared to the Gaussian beam profile. Furthermore, the melt pool width and depth are more stable, with varying parameters for the point/ring profiles, while the Gaussian beam tends to keyhole formation at higher beam powers. Finally, a strong decrease in surface roughness for the point/ring profiles, accompanied by a significantly reduced humping effect, starting even at lower beam powers of 200 W, can be observed. Therefore, these results show the potential of beam shaping for further applications in laser-based processing of high-strength aluminium alloys.

X-ray-computed tomography (CT) is today’s gold standard for the non-destructive evaluation of internal component defects such as cracks and porosity. Using automated standardized evaluation algorithms, an analysis can be performed without knowledge of the shape, location, or size of the defects. Both the measurement and the evaluation are based on the fact that the component has no internal structures or cavities. However, additive manufacturing (AM) and hybrid subtractive procedures offer the possibility of integrating internal structures directly during the building process. The examination of powder bed fusion (PBF) samples made of Ti64 and PA12 showed that the standardized evaluation methods were not able to identify internal structures correctly. Different evaluation methods for the CT-measured values were analyzed and recommendations on a procedure for measuring internal structures are given.

In this work, an innovative manufacturing approach that includes a fully linked and integrated manufacturing system consisting of a laser-based directed energy deposition (DED-LB/M) module and a forming press is presented. The alternating additive manufacturing (AM) process is based on a combination of a DED-LB/M process using a laser power of 600 W and a feed rate of 400 mm/min and a subsequent forming process, in which the structure is upset with a hydraulic press using a constant forming force of 500 kN in order to smooth the surface and influence the accuracy of the components. For the generation of a fundamental process understanding, a cuboid, basic shape was chosen as geometry for the investigations. The aim is to improve part properties by applying the process steps to generate part properties, which are superior to solely additive manufactured material. It is shown that the geometry of additive manufactured structures can be adapted, and the top surface can be smoothed due to the forming operation. The mean roughness value Rz decreases up to 50% after the forming operation. The hardness can be increased by work hardening. Of special interest is that the higher hardness can be kept up even though a further DED-LB/M process step and forming operation are applied to the additively manufactured and formed structure again. Finally, an analysis of the new manufacturing approach regarding its potential is given.

Spatial and temporal knowledge of temperature evolution is crucial in laser beam welding of low-melting materials such as aluminum alloys. One major advantage of additive manufacturing is the high freedom of design, which supports the fabrication of complex structures. However, geometrical features such as combined massive volumes and cellular structures in such parts can lead to an uneven heat distribution during processing, resulting in different material properties throughout the part. In this study, we demonstrate these effects, using a complex structure consisting of three conic shapes with narrow cylinders in between hindering heat flux. We manufacture the parts via powder bed fusion of Ti6Al4V by applying a laser beam (PBF-LB/M) as well as an electron beam (PBF-EB). We investigate the impact of the different thermal regimes on the part density, microstructure and mechanical properties aided by finite element simulations as well as by thermography and X-ray computed tomography measurements. Both simulations and thermography show an increase in inter-layer temperature with increasing part radius, subsequently leading to heat accumulation along the build direction. While the geometry and thermal history have a minor influence on the relative density of the parts, the microstructure is greatly affected by the thermal history in PBF-LB/M. The acicular martensitic structure in the narrow parts is decomposed into a mix of tempered lath-like martensite and an ultrafine α + β microstructure with increasing part radius. The EBM part exhibits a lamellar α + β microstructure for both the cylindric and conic structures. The different microstructures directly influence the hardness of the parts. For the PBF-LB part, the hardness ranges between 400 HV0.5 in the narrow sections and a maximum hardness of 450 HV0.5 in the broader sections, while the PBF-EB part exhibits hardness values between 280 and 380 HV0.5.

SARS-CoV-2 serosurveillance is important to adapt infection control measures and estimate the degree of underreporting. Blood donor samples can be used as a proxy for the healthy adult population. In a repeated cross-sectional study from April 2020 to April 2021, September 2021, and April/May 2022, 13 blood establishments collected 134,510 anonymised specimens from blood donors in 28 study regions across Germany. These were tested for antibodies against the SARS-CoV-2 spike protein and nucleocapsid, including neutralising capacity. Seroprevalence was adjusted for test performance and sampling and weighted for demographic differences between the sample and the general population. Seroprevalence estimates were compared to notified COVID-19 cases. The overall adjusted SARS-CoV-2 seroprevalence remained below 2% until December 2020 and increased to 18.1% in April 2021, 89.4% in September 2021, and to 100% in April/May 2022. Neutralising capacity was found in 74% of all positive specimens until April 2021 and in 98% in April/May 2022. Our serosurveillance allowed for repeated estimations of underreporting from the early stage of the pandemic onwards. Underreporting ranged between factors 5.1 and 1.1 in the first two waves of the pandemic and remained well below 2 afterwards, indicating an adequate test strategy and notification system in Germany.

BackgroundThe search for health information from web-based resources raises opportunities to inform the service operations of health care systems. Google Trends search query data have been used to study public health topics, such as seasonal influenza, suicide, and prescription drug abuse; however, there is a paucity of literature using Google Trends data to improve emergency department patient-volume forecasting. ObjectiveWe assessed the ability of Google Trends search query data to improve the performance of adult emergency department daily volume prediction models. MethodsGoogle Trends search query data related to chief complaints and health care facilities were collected from Chicago, Illinois (July 2015 to June 2017). We calculated correlations between Google Trends search query data and emergency department daily patient volumes from a tertiary care adult hospital in Chicago. A baseline multiple linear regression model of emergency department daily volume with traditional predictors was augmented with Google Trends search query data; model performance was measured using mean absolute error and mean absolute percentage error. ResultsThere were substantial correlations between emergency department daily volume and Google Trends “hospital” (r=0.54), combined terms (r=0.50), and “Northwestern Memorial Hospital” (r=0.34) search query data. The final Google Trends data–augmented model included the predictors Combined 3-day moving average and Hospital 3-day moving average and performed better (mean absolute percentage error 6.42%) than the final baseline model (mean absolute percentage error 6.67%)—an improvement of 3.1%. ConclusionsThe incorporation of Google Trends search query data into an adult tertiary care hospital emergency department daily volume prediction model modestly improved model performance. Further development of advanced models with comprehensive search query terms and complementary data sources may improve prediction performance and could be an avenue for further research.

We present the discovery of TOI-1420b, an exceptionally low-density ( ρ = 0.08 ± 0.02 g cm ^−3 ) transiting planet in a P = 6.96 days orbit around a late G-dwarf star. Using transit observations from TESS, LCOGT, Observatoire Privé du Mont, Whitin, Wendelstein, OAUV, Ca l’Ou, and KeplerCam, along with radial velocity observations from HARPS-N and NEID, we find that the planet has a radius of R _p = 11.9 ± 0.3 R _⊕ and a mass of M _p = 25.1 ± 3.8 M _⊕ . TOI-1420b is the largest known planet with a mass less than 50 M _⊕ , indicating that it contains a sizeable envelope of hydrogen and helium. We determine TOI-1420b’s envelope mass fraction to be ${f}_{\mathrm{env}}={82}_{-6}^{+7} \% $ , suggesting that runaway gas accretion occurred when its core was at most four to five times the mass of the Earth. TOI-1420b is similar to the planet WASP-107b in mass, radius, density, and orbital period, so a comparison of these two systems may help reveal the origins of close-in low-density planets. With an atmospheric scale height of 1950 km, a transmission spectroscopy metric of 580, and a predicted Rossiter–McLaughlin amplitude of about 17 m s ^−1 , TOI-1420b is an excellent target for future atmospheric and dynamical characterization.

Laser welding of copper is being used with increasing demand for contacting applications in electric components such as batteries, power electronics, and electric drives. With its local, non-contact energy input and high automation capability enabling reproducible weld quality, this joining technology represents a key enabler of future mobility systems. However, a major challenge in process design is the combination of energy efficiency and precise process guidance in terms of weld seam depth and defect prevention (i.e., spatter and melt ejections) due to the high electrical and thermal conductivity of copper. High-power lasers in the near infrared wavelength range (𝜆 ≈ 1 μm) and excellent beam quality provide an established joining solution for this purpose; nevertheless, the low absorptivity (≤5%) advocates novel beam sources at visible wavelengths due to altered absorptivity (40% at 515 nm) characteristics as an improved tool. In order to understand the influence of laser wavelength and process parameters on the vapor capillary geometry, in situ synchrotron investigations on Cu-ETP with 515 nm and 1030 nm laser sources with the same spot diameter are compared. The material phase contrast analysis was successfully used to distinguish keyhole and melt pool phase boundaries during the welding process. A significantly different sensitivity of the keyhole depth in relation to the feed rate was found, which is increased for the infrared laser. This behavior could be attributed to the increased effect of multiple reflections at 1030 nm.

In laser beam fusion cutting of metals, the interaction of the gas jet with the melt determines the dynamics of the melt extrusion and the quality of the resulting cutting kerf. The gas-dynamic phenomena occurring during laser beam cutting are not fully known, especially regarding temporal fluctuations in the gas jet. The observation of gas and melt dynamics is difficult because the gas flow is not directly visible in video recordings and access to the process zone for observation is limited. In this study, the problem of imaging the gas jet from the cutting nozzle is addressed in a novel way by utilizing the striation pattern formed at the cutting kerf as a background pattern for background-oriented Schlieren imaging (BOS). In this first feasibility study, jets of different gas nozzles were observed in front of a solidified cutting kerf, which served as a background pattern for imaging. The results show that imaging of the characteristic shock diamonds of cutting nozzles is possible. Furthermore, the resulting shock fronts from an interaction of the gas jet with a model of a cutting front can be observed. The possibility of high-speed BOS with the proposed method is shown, which could be suitable to extend the knowledge of gas-dynamic phenomena in laser beam fusion cutting.

In-process monitoring of weld penetration depth is possible with optical coherence tomography (OCT). The weld depth can be identified with OCT by statistical signal processing of the raw OCT signal and keyhole mapping. This approach is only applicable to stable welding processes and requires a time-consuming keyhole mapping to identify the optimal placement of a singular OCT measuring beam. In this work, we use an OCT measurement line for the identification of the weld depth. This approach shows the advantage that the calibration effort can be reduced as the measurement line requires only calibration in one dimension. As current literature focuses on weld depth measurement with a singular measurement point in the keyhole, no optimal algorithm exists for weld depth measurement with an OCT measurement line. We developed seven different weld depth processing pipelines and tested these algorithms under different weld conditions, such as stable deep penetration welding, unstable deep penetration welding, and heat conduction welding. We analyzed the accuracy of the weld depth processing algorithms by comparing the measured weld depth with metallographic weld depths. The intensity accumulation approach is identified as the most accurate algorithm for successful weld depth measurement with a scanning OCT measurement line.
