Fungi in the genus Trichoderma are widespread in the environment, mainly in soils. They are used in agriculture because of their mycoparasitic potential; Trichoderma have the ability to increase plant health and provide protection against phytopathogens, making them desirable plant symbionts. We isolated, identified, and characterized Trichoderma from different regions of Saudi Arabia and evaluated the ability of Trichoderma to promote plant growth. Morphological and molecular characterization, along with phylogenetic studies, were utilized to differentiate between Trichoderma species isolated from soil samples in the Abha and Riyadh regions, Saudi Arabia. Then, plant growth-promoting traits of the isolated Trichoderma species were assessed. Eight Trichoderma isolates were characterized via morphological and molecular analysis; six (Trichoderma koningiopsis, Trichoderma lixii, Trichoderma koningii, Trichoderma harzianum, Trichoderma brevicompactum, and Trichoderma velutinum) were from Abha and two (T. lixii and T. harzianum) were from Riyadh. The isolated Trichoderma strains belonged to three different clades (Clade 1: Harzianum, Clade 2: Brevicompactum, and Clade 3: Viride). The Trichoderma isolates varied in plant growth-promoting traits. Seeds treated with most isolates exhibited a high percentage of germination, except seeds treated with the T3-T. koningii isolate. 100% germination was reported for seeds treated with the T4-T. harzianum and T6-T. brevicompactum isolates, while seeds treated with the T1-T. koniniopsis and T5-T. lixii isolates showed 91.1% and 90.9% germination, respectively. Seeds treated with the T8-T. velutinum, T2-T. lixii, and T7-T. harzianum isolates had germination rates of 84.1%, 82.2%, and 72.7%, respectively. The Trichoderma isolate T5-T. lixii stimulated tomato plant growth the most, followed by T7-T. harzianum, T8-T. velutinum, T4-T. harzianum, T1-T. koniniopsis, T2-T. lixii, and T6-T. brevicompactum; the least effective was T3-T. koningii. A maximum fresh weight of 669.33 mg was observed for the T5-T. lixii-treated plants. The Abha region had a higher diversity of Trichoderma species than the Riyadh region, and most isolated Trichoderma spp. promoted tomato growth. [ABSTRACT FROM AUTHOR]

Cordyceps militaris, a medicinal fungus, has gained considerable attention owing to its potential health benefits, notably the production of bioactive compounds such as cordycepin. Cordycepin possesses significant antifungal, antibacterial, and antiviral properties. The present study focused on optimizing the fermentation conditions for C. militaris to boost the production of mycelia and cordycepin, alongside investigating its antifungal properties using in silico and in vitro approaches. The optimal conditions, yielding the highest cordycepin and mycelial biomass, were a temperature of 20°C and a pH range of 4–6, with glucose and sucrose as carbon sources and yeast extract and casein hydrolysate as nitrogen sources. Under these conditions, cordycepin production peaked at low pH (600–1000 mg/L) and with carbon and maltose (400–500 mg/L). The low temperature favored cordycepin production (400 mg/L), whereas casein hydrolysate as a nitrogen source boosted cordycepin yield (600 mg/L). The docking analysis indicated that cordycepin had the highest binding affinity for the tubulin beta chain 2 (−10.4 kcal/mol) compared to the fungicide tebuconazole (−7.9 kcal/mol for both targets). The in silico results were corroborated by in vitro studies, where the mycelial extract of C. militaris inhibited approximately 75% of fungal growth at a concentration of 6000 ppm. These findings suggest that optimizing fermentation conditions significantly enhances cordycepin production, and cordycepin shows antifungal solid activity, making it a promising agent for biocontrol in agriculture. The current study aims to ascertain the chemical composition and biological activities of thyme leaves essential oil (TEO), with an emphasis on its in silico molecular docking studies targeting the antibacterial properties of its constituents. TEO was extracted from dried leaves using a standardized procedure, with a yield of 0.75%. The chemical constituents of TEO were identified using Gas Chromatography-Mass Spectrometry (GC-MS). The primary compounds detected included carvacrol, o-acetyl thymol, α-pinene, and spathulenol. Subsequently, the antioxidant activity of the isolated oil was measured, and the results showed that the total phenolic contents (TPC) was 4.69 ± 0.17 GAE mg/100 g, the total flavonoid contents (TFC) was 1.45 ± 0.04 CE mg/100 g, the ferric reducing antioxidant potential (FRAP) ranged from 0.07-0.81, and the DPPH radical scavenging activity was 42.88-86.94%. TEO exhibited notable antibacterial activity against certain strains of Staphylococcus aureus and Escherichia coli. The molecular docking studies were then conducted to further investigate the interaction of TEO's potent metabolites with key bacterial enzymes, identifying spathulenol and caryophyllene as the best-interacting ligands. Hemolytic activity tests of TEO indicated negligible cytotoxic effects. These findings support that TEO holds great potential for the development of functional food and nutra-pharmaceuticals. Citrus limon (L.) Osbeck is considered as one of the most significant horticultural as well as essential oil crop. However, the significance of the fruit peels left after the utilization of the sour fruit find limited importance and these leftover peels are rich in essential oil. Lemon peel essential oil (LPEO) finds used in the fragrance, food, cosmetic, and pharmaceutical industries. In this study lemon peel were used for isolation of essential oil by hydro-distillation, analyzed by GC-FID and GC-MS and was further subjected to in vitro anti-oxidant (DPPH, ABTS), anti-inflammatory, anti-tyrosinase, genotoxicity and anti-diabetic assays. The essential oil analysis has revealed limonene as major compound (38.08%) along with other minor compounds namely, α-citral (12.02%), decanal (8.14%) and neral (7.36%). The total amount of citral (α-citral and neral) was found to be 19.38%, comparatively higher compared to previous reports. LPEO was found to exhibit very strong tyrosinase inhibition activity along with good alpha-amylase inhibition. The results from this study further strengthen the immense potential of LPEO as an anti-diabetic and skin whitening agent. The results framed from this study will further strengthen the position of earlier works on biological activities of C. limon besides providing a descriptive work on the potential of Assam LPEO. The root-knot nematode (RKN) causes significant yield loss in tomatoes. Understanding the interaction of biocontrol agents (BCAs)-nematicides-soil microbiomes and RKNs is essential for enhancing the efficacy of biocontrol agents and nematicides to curb RKN damage to crops. The present study aimed to evaluate the in vitro effectiveness of BACa and nematicide against RKN and to apply the amplicon sequencing to assess the interaction of Bacillus velezensis (VB7) and Trichoderma koningiopsis (TK) against RKNs. Metagenomic analysis revealed the relative abundance of three phyla such as Proteobacteria (42.16%), Firmicutes (19.57%), and Actinobacteria (17.69%) in tomato rhizospheres. Those tomato rhizospheres treated with the combined application of B. velezensis VB7 + T. koningiopsis TK and RKN had a greater frequency of diversity and richness than the control. RKN-infested tomato rhizosphere drenched with bacterial and fungal antagonists had the maximum diversity index of bacterial communities. A strong correlation with a maximum number of interconnection edges in the phyla Proteobacteria, Firmicutes, and Actinobacteria was evident in soils treated with both B. velezensis VB7 and T. koningiopsis TK challenged against RKN in infected soil. The present study determined a much greater diversity of bacterial taxa observed in tomato rhizosphere soils treated with B. velezensis VB7 and T. koningiopsis TK than in untreated soil. It is suggested that the increased diversity and abundance of bacterial communities might be responsible for increased nematicidal properties in tomato plants. Hence, the combined applications of B. velezensis VB7 and T. koningiopsis TK can enhance the nematicidal action to curb RKN infecting tomatoes. [ABSTRACT FROM AUTHOR]

The phosphate solubilizing properties of Lysinibacillus macroides ST-30, Pseudomonas pelleroniana N-26, and Bacillus cereus ST-6 were tested for the chickpea crop of the Tarai region of Uttarakhand. These microbially inoculated plants have shown significant (p > 0.05) improvement in the plant health and crop health parameters, viz., root length, shoot length, fresh weight, dry weight, nodule number, nodule fresh weight, nodule dry weight, chlorophyll content, and nitrate reductase. The highest shoot length (46.10 cm) and chlorophyll content (0.57 mg g-1 fresh weight) were observed in ST-30 at 75 DAS with 20 kg P2O5/ha. Similarly, for plant P content, an increase of 90.12% over control was recorded in the same treatment. Treatments consisting of Lysinibacillus macroides ST-30 along with 20 kg/ha P2O5 were found to be most suitable as phosphatic fertilizer. Conclusively, sustainable agriculture practices in the Tarai as well as the field region may be developed based on a strategy of exploring microbial inoculants from the pristine region of the Western Himalayas. The presence and abundance of bacterial inoculants were confirmed through qRT-PCT. We conclude that the effective plant growth-promoting bacterium Lysinibacillus macroides ST-30 broadens the spectrum of phosphate solubilizers available for field applications and might be used together with 20 Kg/ha P2O5. Fusarium‐induced wilt significantly affects the cultivation and yield of pigeon peas. This warrants sustainable disease management while promoting plant growth. The present study investigated the biopotential of coinoculation of Streptomyces pseudogriseolus S‐9 and Rhizophagus irregularis for plant growth promotion and mitigation of the impact of Fusarium wilt on pigeon pea over three seasons at pot and field levels. Pigeon pea plants were subjected to Fusarium wilt stress and treated with different inoculation strategies, including single and combined applications of S. pseudogriseolus S‐9 and R. irregularis. Plant growth parameters and yields were assessed to evaluate the efficacy of the coinoculation. In the pot experiment, T‐6 treatment resulted in the longest root (62.56 ± 0.01 cm) and shoot (70.24 ± 0.01 cm) lengths compared to the application of commercial biofungicide T‐8 (Trichoderma). This treatment also significantly influenced the yield of potted plants. It resulted in the highest fresh root weight (62.27 ± 0.01 g), fresh shoot weight (70.24 ± 0.02 g), maximum root (55.25 ± 0.01 g) and shoot dry weights (52.25 ± 0.01 g). In the field experiment, pigeon pea plants treated with the bioinoculant also demonstrated a substantial increase (р ≤ 0.05) in total grain yield, the weight of 100 grains, and the number of filled grains compared to the control group in all experimental seasons. In vitro, antagonism assay of compatibility of mycorrhizae and bacteria showed good activity using powder formulation. Thus, the consortium application inspired the broad application of Streptomyces and Trichoderma as effective bioinoculants for wilt management and yield improvement in pigeon peas. A new bidentate Schiff base ligand (C16H16Cl2N4), condensation product of ethylene diamine and 4-chloro N-phenyl formamide, and its metal complexes [M(C16H16Cl2N4)2(OAc)2] (where M = Mn(II) and Zn(II)) were synthesized and characterized using various analytical and spectral techniques, including high-resolution mass spectrometry (HRMS), elemental analysis, ultraviolet-visible (UV-vis), Fourier-transform infrared (FTIR) spectroscopy, AAS, molar conductance, 1H NMR, and powder XRD. All the compounds were non-electrolytes and nanocrystalline. The synthesized compounds were assessed for antioxidant potential by DPPH radical scavenging and FRAP assay, with BHT serving as the positive control. Inhibitory concentration at 50% inhibition (IC50) values were calculated and used for comparative analysis. Furthermore, the prepared compounds were screened for antibacterial activity against two Gram-negative bacteria (Staphylococcus aureus and Bacillus subtilis) and two Gram-positive bacteria (Escherichia coli and Salmonella typhi) using disk-diffusion methods, with amikacin employed as the standard reference. The comparison of inhibition zones revealed that the complexes showed better antibacterial activity than the ligand. To gain insights into the molecular interactions underlying the antibacterial activity, the ligand and complexes were analyzed for their binding affinity with S. aureus tyrosyl-tRNA synthetase (PDB ID: 1JIL) and S. typhi cell membrane protein OmpF complex (PDB ID: 4KR4). Mentha longifolia (Lamiaceae) is regarded as highly valuable aromatic medicinal species with wide range of pharmaceutical application. In the present work, leaf essential oil of M. longifolia was applied for the assessment of in vitro pharmacological potentials and compositional analysis. A sum of 15 compounds were detected by GC-FID and GC-MS analysis of M. longifolia essential oil with total area of 99.98% where major compounds were carvone (59.56%), D-limonene (20.81%), eucalyptol (10.08%). The anti-oxidant potency of M. longifolia was estimated using two methods viz., ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl) assay where M. longifolia essential oil showed moderate anti-oxidant potency. M. longifolia essential oil revealed high anti-inflammatory and tyrosinase inhibitory activity with an IC50 value 3.68 μL/mL and 1.22 μL/mL respectively compared to standard sodium diclofenac (5.33 μL/mL) and kojic acid (2.28 μL/mL). The acetylcholinesterase (AchE) inhibitory and genotoxic assay was performed, where M. longifolia essential oil revealed strong anti-acetylcholinesterase and anti-genotoxic activity. Anti-microbial potential of M. longifolia essential oil was also where strong anti-bacterial activity was shown against Salmonella typhimurium, Bacillus cereus, Staphylococcus aureus, Micrococcus luteus. For anti-fungal activity, M. longifolia essential oil showed no zone of inhibition against any used strains except Aspergillus flavus. From the present research it was established that M. longifolia essential oil possesses pharmaceutical potential with strong AchE inhibitory, anti-inflammatory, anti-microbial, anti-tyrosinase, anti-genotoxic and anti-oxidant activity which can be used for plant-based drug development in future endeavor. Introduction: Developing organic herbal crops provides chemical-free herbs to support environmentally friendly and sustainable farming. One way in agricultural biotechnology to grow herbal organic crops is to use rhizobacteria. The herbal plant used in this study is the Javanese ginseng plant (Talinum paniculatum), which contains phytochemicals that increase stamina. Methods: The study used four rhizobacteria to enhance the growth and phytochemistry of T. paniculatum leaves; the target phytochemical compounds analyzed in Javanese ginseng leaves were total flavonoids, total phenols, and antioxidants (IC50%). The four selected rhizobacteria can produce indole-3-acetic acid (IAA), fix nitrogen, and dissolve phosphate. Thus, high-quality T. paniculatum leaves were obtained as herbal tea ingredients. The pattern used is a random group pattern in the greenhouse. Results: The results of the study showed that the use of rhizobacteria at 2% concentrations had a real effect on plant growth parameters such as plant height, leaf area, root length, wet weight and dry weight of the plant, and phytochemical content like total flavonoids, total phenols, IC50%, total chlorophyll, content of T. paniculatum leave plant when compared to the control. The four rhizobacteria used can produce the IAA, fix nitrogen, and dissolve phosphorus. Discussion: The four rhizobacteria can also colonize the root of the T. paniculatum plant. The four Rhizobzcteria used significantly affected the growth and phytochemical content of T. paniculatum leaves as an ingredient in herbal tea at a concentration of 2% compared to controls. The four rhizobacteria can produce IAA hormones, fix nitrogen, and dissolve phosphorus. All four rhizobacteria can colonize the roots of T. paniculatum plants. These four rhizobacteria can be used as alternative methods in developing organic farming systems and can also be used practically in the field by farmers. Increased levels of the non-essential hazardous metalloid arsenic (As) in rice grains pose a threat to human health and the sustainability of the rice industry. In several counties, the average As contamination in polished rice has been detected to range from 0.002 to 0.39 mg/kg, which is above the safe limit of 1 mg/kg as recommended by the World Health Organisation. Beyond this limit, the digestive tract, circulatory system, skin, liver, kidney, nervous system and heart can be affected. Humans can develop cancer from consuming or inhaling As. In addition, long-term exposure to drinking water contaminated with arsenic has also been linked to a dose-response relationship with an increased risk of hypertension and diabetes mellitus. Rice has been shown to be an indirect source of arsenic accumulation in human bodies. Under flooded paddy soil, trivalent arsenate (AsIII) occupies 87-94% of the total As, while under non-flooded soil, pentavalent arsenate (AsV) predominates (73-96% of the total As). This review aims to provide a thorough and interdisciplinary understanding of the behaviour of As in the paddy soil and transportation to rice grain and further investigate efficient ways to limit arsenic contamination. Supplementation of soil with specific mineral nutrients such as iron (Fe), sulphur (S) and silicon (Si) can significantly decrease the arsenic accumulation in rice grain by minimising its uptake and translocation. The hydrogen bonding potentials of uronic acids, proteins and amino sugars on the extracellular surface of soil microorganisms facilitate the detoxification of arsenic species. Further, rice is absorbed less when exposed to aerobic water management practices than anaerobic ones since it reduces the build-up of As in rice, and the solution is immobilised as in the soil. Bacillus species appearas the most attractive plant growth-promoting rhizobacteria (PGPR) and alternative to synthetic chemical pesticides. The present study examined the antagonistic potential of spore forming-Bacilli isolated from organic farm soil samples of Allahabad, India. Eighty-seven Bacillus strains were isolated and characterized based on their morphological, plant growth promoting traits and molecular characteristics. The diversity analysis used 16S-rDNA, BOX-element, and enterobacterial repetitive intergenic consensus. Two strains, PR30 and PR32, later identified as Bacillus sp., exhibited potent in vitro antagonistic activity against Ralstonia solanaceorum. These isolates produced copious amounts of multiple PGP traits, such as indole-3-acetic acid (40.0 and 54.5 μg/mL), phosphate solubilization index (PSI) (4.4 and 5.3), ammonia, siderophore (3 and 4 cm), and 1-aminocyclopropane-1-carboxylate deaminase (8.1and 9.2 μM/mg//h) and hydrogen cyanide. These isolates were subjected to the antibiotic sensitivity test. The two potent isolates based on the higher antagonistic and the best plant growth-promoting ability were selected for plant growth-promoting response studies in tomatoe, broccoli, and chickpea. In the pot study, Bacillus subtilis (PR30 and PR31) showed significant improvement in seed germination (27–34%), root length (20–50%), shoot length (20–40%), vigor index (50–75%), carotenoid content (0.543–1.733), and lycopene content (2.333–2.646 mg/100 g) in tomato, broccoli, and chickpea. The present study demonstrated the production of multiple plant growth-promoting traits by the isolates and their potential as effective bioinoculants for plant growth promotion and biocontrol of phytopathogens. Background: Microorganisms are crucial in our ecosystem, offering diverse functions and adaptability. The UNGA Science Summit has underscored the importance of understanding microbes in alignment with the UN Sustainable Development Goals. Bacillus anthracis poses significant challenges among various microorganisms due to its harmful effects on both soil and public health. Our study employed computational techniques to investigate the inhibitory effects of curcumin and mangiferin on Bacillus anthracis, with the aim of presenting a novel bio-based approach to microbial management. Methods: Employing high-throughput screening, we identified potential binding sites on B. anthracis. Molecular docking revealed that curcumin and mangiferin, when synergistically combined, exhibited strong binding affinities at different sites on the bacterium. Our findings demonstrated a significant drop in binding free energy, indicating a stronger interaction when these compounds were used together. Findings: Results of Molecular docking indicated binding energies of -8.45 kcal/mol for mangiferin, -7.68 kcal/mol for curcumin, and a notably higher binding energy of -19.47 kcal/mol for the combination of mangiferin and curcumin with CapD protein. Molecular dynamics simulations further validated these interactions, demonstrating increased stability and structural changes in the bacterium. Conclusion: This study highlights the effectiveness of natural compounds like curcumin and mangiferin in microbial management, especially against challenging pathogens like B. anthracis.
