Mansonellosis is a neglected and emerging tropical disease. Among all zoonotic filarial diseases, it is probably the most prevalent and least studied, with approximately 114 million people infected. The parasites of Mansonella spp. are among the most common blood parasitemias and are widely found in Africa and Latin America. Through molecular analysis of blood samples from free-ranging primates Sapajus nigritus (n 33) and Alouatta guariba clamitans (n 5) in the southern states of Brazil (Santa Catarina and Rio Grande do Sul), we identified samples positive for Mansonella perstans in two specimens of A. guariba clamitans. A fragment of 578 bp from the ITS intergenic region (5.8S-ITS2-28S) was targeted for an initial PCR screening. Subsequently, positive samples were subjected to other PCR assays targeting a fragment of the 12S and the 18S genes. This is the first record of molecular detection of the agent in this host in the Pampa Biome. With a wide distribution across Brazil and Argentina, these primates may represent a potential wild reservoir for the zoonotic agent of mansonellosis. Entomological and transmission studies are essential to avoid the urbanization of mansonellosis and to understand the cycles of agents in different environmental scenarios.

Generalized and fatal felid alphaherpesvirus-1 (FeHV-1) natural infection with liver involvement is rarely reported in cats, and the occurrence of herpesvirus viraemia with internal organ histologic lesions in adult cats is unknown. A 1.5-year-old cat, female, mixed breed, positive for feline leukaemia virus (FeLV) presented in a veterinary teaching hospital with sneezing, nasal discharge, anorexia, and diarrhoea after two weeks, evolving to inspiratory dyspnoea. Complete blood count and serum biochemistry analysis showed marked leukopenia and thrombocytopenia. After clinical worsening and lack of treatment response, the cat was euthanized. Pathological findings included hepatic necrosis, fibrinonecrotic tracheitis, and bronchointerstitial pneumonia. Marked amounts of coccobacillary bacteria were observed covering the necrotic tracheal and bronchial mucosa, at the cytoplasm of alveolar macrophages, and free in alveoli lumen, mimicking a primary bacterial tracheitis and pneumonia. Both lung and tracheal bacteria exhibited marked immunolabeling in anti-Escherichia coli immunohistochemistry. In addition, rare epithelial cells of bronchi contained round, eosinophilic, intranuclear viral inclusion bodies (4-7 µm) that marginate the chromatin, characteristic of FeHV-1 infection. Strong multifocal anti-FeHV-1 immunolabeling was observed in necrotic epithelial cells of the liver, trachea, and lungs. Generalized herpesvirus infection with the occurrence of acute hepatic necrosis and severe respiratory illness is a potential differential diagnosis in FeLV-positive cats with respiratory signs. The immunodepression in these cats probably favours a FeHV-1 viraemia in addition to the development of opportunistic bacterial infections, such as Escherichia coli, and it is associated with a poor outcome.

BACKGROUND Anthropogenic disturbances are the main threats to nonhuman primates conservation, and infectious diseases may also play a key role in primate population decline. This study aimed to determine the main causes of death in neotropical primates. METHODS A retrospective study of post-mortem examinations was conducted on 146 neotropical primates between January 2000 and December 2018. RESULTS Conclusive diagnoses were obtained in 68.5% of the cases, of which 59 corresponded to non-infectious causes and 41 to infectious diseases. Trauma was the main cause of death (54/100), with anthropogenic stressors caused by blunt force trauma injuries (collision with vehicles) and puncture wound injuries associated with interspecific aggression (dog predation) were the most common factors. Other causes of death included bacterial diseases (27%), followed by parasitic diseases (12%), neoplasms (2%), and viral diseases (2%). CONCLUSIONS Free-ranging primates were mostly affected by non-infectious causes, while captive primates were by infectious conditions.

The increase in sheep production is directly related to the health status of the flock. Brazil is one of the largest sheep producers in the world, and the sheep flock is concentrated in southern and northeast regions. Infectious diseases are responsible for severe economic losses resulting from a decrease in milk and meat production, deaths, and cost of treatment. Among infectious diseases, viral diseases are described chiefly in case reports or retrospective studies. This study aimed to review the main features of viral diseases that affect sheep in Brazil and their current situation in the Brazilian territory. We included eight viral diseases described in Brazil: rabies, bluetongue, contagious ecthyma, foot and mouth disease, visna-maedi, enzootic nasal tumor, ovine pulmonary adenocarcinoma, and border disease. We review the etiological, epidemiological, clinical, and pathological findings for each agent and included differential diagnoses, information on recommended diagnostic methods to confirm the disease etiology, and control measures. This study served as quick consultation material for field veterinarians for an accurate diagnosis. RESUMO: O aumento da produção ovina está diretamente relacionado ao status sanitário do rebanho. O Brasil é um dos maiores produtores mundiais de ovinos, e o rebanho está concentrado nas regiões nordeste e sul do país. As doenças infecciosas são responsáveis por perdas econômicas severas que resultam da diminuição na produção de leite e carne, mortes e custos com tratamentos. Entre os agentes infecciosos, as doenças de origem viral são, em sua maioria, descritas em relatos de caso ou estudos retrospectivos. Este estudo teve como objetivo revisar as principais características das doenças virais que afetam ovinos no território brasileiro, e qual sua situação atual. Nós incluímos oito doenças virais diagnosticadas no Brasil: raiva, língua azul, ectima contagioso, febre aftosa, maedi-visna, tumor enzoótico nasal, adenocarcinoma pulmonar ovino, e pestivirose. Revisamos os achados etiológicos, epidemiológicos, clínicos e patológicos de cada agente e incluímos seus respectivos diagnósticos diferenciais, informações acerca dos métodos diagnósticos para confirmação da etiologia da doença e medidas de controle e prevenção. Esse estudo tem o propósito de servir como objetivo servir de material de consulta rápida, especialmente para veterinários de campo realizar um diagnóstico acurado.

Summary Pneumonia in cats may cause severe lung injury and consequent death. We describe the post-mortem findings and aetiologies of naturally fatal pneumonia in 78 domestic cats, using gross and histopathological examinations, immunohistochemistry and microbiological techniques. Morphological patterns found were bronchopneumonia (27/78), interstitial (15/78), bronchointerstitial (13/78), granulomatous (8/78), aspiration (8/78) and pyogranulomatous (5/78) pneumonia, and pleuropneumonia (2/78). Bacterial pneumonia was identified as the most common cause (32/78), followed by viral (15/28 feline calicivirus, 10/28 felid alphaherpesvirus 1 and 3/28 both viruses), aspiration (8/78), fungal (5/78) and parasitic pneumonia (5/78). Co-infection with feline immunodeficiency virus and feline leukaemia virus was found in 54 cats. Viral infections involved cats of all ages, indicating the importance of investigating viral causes in cats with respiratory diseases, including in adult and ageing cats.

Toxoplasma gondii infections are usually asymptomatic in pigs, and an acute clinical disease is rare in this host. This study aimed to determine the pathological and molecular aspects of an outbreak of fatal systemic toxoplasmosis in finishing pigs in Brazil. The outbreak occurred on a commercial finishing pig farm in the state of Santa Catarina in southern Brazil. The farm had 1500 pigs and 3.8% of mortality rate during the outbreak. The pigs had fever, anorexia, apathy, and locomotor deficits. Seven pigs were necropsied. Gross findings included multifocal to coalescent pale areas in skeletal muscles, lymphadenomegaly, hepatosplenomegaly, and non-colapsed lungs. The histological findings included granulomatous lymphadenitis, hepatitis and splenitis, necrotizing myositis, and lymphoplasmacytic interstitial pneumonia. Lung and liver lesions were occasionally accompanied by T. gondii parasitic structures. Positive immunolabeling for T. gondii tachyzoites and encysted bradyzoites was detected in all examined pigs. PCR-RFLP (11 markers) and microsatellite analysis (15 markers) identified the non-archetypal genotype #278 in pigs. This is the first report of systemic toxoplasmosis in pigs with muscle lesions and additionally shows the diversity of disease-causing T. gondii genotypes circulating in animals in Brazil. Resumo As infecções por Toxoplasma gondii são geralmente assintomáticas em suínos, e uma doença clínica aguda é rara nessa espécie. Este estudo teve como objetivo determinar os aspectos patológicos e moleculares de um surto de toxoplasmose sistêmica fatal em suínos em terminação no Brasil. O surto ocorreu em uma granja comercial de suínos em terminação no estado de Santa Catarina, no sul do Brasil. A granja tinha 1500 suínos e a taxa de mortalidade durante o surto foi de 3,8%. Os suínos apresentaram febre, anorexia, apatia e déficits locomotores. Sete suínos foram necropsiados. Os achados macroscópicos incluíram áreas pálidas multifocais a coalescentes nos músculos esqueléticos, linfadenomegalia, hepatoesplenomegalia e pulmões não colapsados. Os achados histológicos incluíram linfadenite, hepatite, esplenite granulomatosa e miosite necrosante, assim como pneumonia intersticial linfoplasmocítica. Lesões pulmonares e hepáticas foram ocasionalmente acompanhadas por estruturas parasitárias de T. gondii. A imunomarcação positiva para taquizoítos e bradizoítos encistados de T. gondii foi observada em todos os suínos examinados. PCR-RFLP (11 marcadores) e análise de microssatélites (15 marcadores) identificaram o genótipo não arquetípico #278 em suínos. Este é o primeiro relato de toxoplasmose sistêmica em suínos com lesões musculares e, adicionalmente, demonstra a diversidade de genótipos de T. gondii causadores de doenças circulantes em animais no Brasil.

Cutaneous mast cell tumor (MCT) is one of the most frequent cutaneous neoplasms of dogs and may vary from well-differentiated to aggressive tumors with metastasis. The authors retrospectively described the gross and histologic aspects of metastatic MCT in 49 dogs. Primary MCT was most commonly identified in the inguinal region (14/35; 40%), and at necropsy multiple, cutaneous nodules were frequently reported (23/49; 47%). All primary MCT were classified as high-grade neoplasms, and metastases involved the lymph nodes (47/49; 96%), spleen (33/49; 67%), liver (29/49; 59%), bone marrow (20/49; 41%), kidneys (16/49; 33%), and heart (14/49; 29%), while the lungs were less commonly affected (9/49; 18%). The main gross findings included lymphadenomegaly in 47 cases; splenomegaly in 28 cases, with splenic nodules in 13 dogs; hepatomegaly in 28 cases, with white pinpoint foci in 9 cases; nodules on the capsular surface of the kidneys in 9 dogs; and epicardial nodules in 6 cases. Histologically, the lymph nodes were largely obliterated by neoplastic mast cells, while in the spleen, neoplastic cells were multifocally scattered (16/33; 48%), arranged in nodules (10/33; 30%), or obliterated the parenchyma (9/33; 27%). In the liver, the neoplastic cells mainly infiltrated the sinusoids (24/29; 83%), but were also arranged in random nodules (10/29; 34%). Interstitial and nodular metastases were observed in the kidneys and the heart. Grossly unapparent metastases were common in the heart (6/14; 43%), kidneys (4/16; 25%), and lungs (6/9). KIT III and KIT II staining patterns were observed in 29 and 20 cases, respectively.

Background: The peafowl is an ornamental bird that has the habit of eating directly from the earthy soil, which makes this bird more susceptible to endoparasites. One important endoparasite is Eucoleus contortus, which leads to inflammatory processes that alter the local microbiota, potentializing disease. By the other way, a member of the bird’s microbiota there is the genus Lactobacillus, but when occurs some imbalance, these bacteria can overgrowth and even cause some infection. This report describes the pathological and microbiological findings of chronic necrotizing pneumonia and aerossacolitis caused by Lactobacillus agilis in a peafowl, associated with parasitism by E. contortus. Case: A peafowl (Pavo cristatus), adult, male, who lived on a farm with contact with other species of animal, was submitted to post-mortem examination due to sudden death. This animal lived in an extensive system on the property and was the only one of its species. During the gross evaluation, the air sacs were filled with solid yellowish crumbly material. The same material was observed forming well-defined nodules that occupied > 50% of the lung parenchyma. Histological analysis showed multiple parabronchi dilated and filled with caseous necrosis, characterized by abundant cellular debris and fibrin deposition. These areas were surrounded by the proliferation of fibrous connective tissue and inflammatory infiltrate of macrophages, giant cells, lymphocytes, and plasma cells. The air sacs parenchyma showed fibrin deposition and mixed inflammatory infiltrate. Multiple gram-positive bacilli were observed within the caseous foci in Gram-stained slides. In the crop and esophageal mucosa, cross-sections of filiform nematodes morphologically compatible with E. contortus were associated with chronic inflammatory infiltrate and epidermal hyperkeratosis. A lung section was submitted to Gram-Brown-Hopps and Ziehl-Neelsen (ZN) stains for bacterial investigation, and Grocott's methenamine silver (GMS) stain for fungal investigation. Short gram-positive bacilli bacteria are observed within the caseous foci in Gram-stained slides. By the other way, no agents were identified on the ZN and GMS stains. Following the analysis, lung fragments were cultivated at aerobic and microaerophilic conditions on sheep blood agar and McConkey agar. All the microbial cultures were incubated at 37°C to 48 h. Pure culture, in microaerophilic condition, of Gram-positive bacilli, was observed. The isolated bacterium was identified by MALDI-TOF MS as L. agilis.Discussion: Although uncommon, L. agilis was the single bacterium identified and therefore, associated as a primary cause of necrotic pneumonia and aerossacolitis in the studied peafowl. The presence of E. contortus could induce the aspiration of regurgitated of little amount of material from the gastrointestinal tract with sufficient bacterial load to initiate an infection, but not enough to smother the animal. The gradual aspiration can induce a chronic inflammatory condition. Infections by bacteria from the host microbiota have been observed in animals and humans with immunodeficiency. In summary, both the parasitosis and the inflammation could be resulted by the parasite and the aspiration of gastric product, which probably interfered in the immune response and allowed the overgrowth of L. agilis. In the current case report, based on macroscopic, microscopic and bacteriological results, we have provided insights to understand how the parasitosis made possible a pneumonia from a bacterium from the host's microbiota. Finally, to the best of our knowledge, this is the first report of L. agilis as causal agent of fatal necrotic pneumonia and aerossacolitis in peafowl.Keywords: respiratory infection, ornamental birds, secondary infection, parasitosis.

Although the previous report of Coxiella burnetii in humans and animals, the knowledge about the occurrence of this pathogen in livestock in Brazil is scarce. This study aimed to search C. burnetii and possible coinfections in tissues of aborted bovine fetuses from Brazil. Tissue samples from seventy-six aborted bovine fetuses sent to the laboratory of molecular diagnosis of infectious diseases from 2013 to 2019 were evaluated by real-time PCR for C. burnetii. Overall, 9.2% (7/76) of the samples were positive for C. burnetii. Moreover, the molecular diagnostic history of our lab revealed the coinfection with Neospora spp. in three fetuses and the presence of histopathological features suggestive with fetal neosporosis in another one. The previous report of C. burnetii in humans and animals in the country, with the detection of C. burnetii from tissues of aborted bovine fetuses reported here, reinforces the neglected state of the disease in Brazil and raises the question of the role of the pathogen in reproductive disorders in national livestock. Respiratory problems due to tracheobronchial foreign bodies (FBs) are unusual in horses; although equines kept in pastures eventually inhale FBs, as conifer twigs of Araucaria angustifolia. A 1,5-year-old Criolle foal was presented with hemoptysis, dyspnea, restlessness and fever (40.9 ºC rectal temperature). Complete blood count showed intense neutropenia, monocytosis, thrombocytopenia and hypoproteinemia. Treatment was carried out but no clinical improvement was observed. At the post-mortem examination, marked amount of dark red liquid was observed in the thoracic cavity (hemothorax). The lung parenchyma was diffusely consolidated, predominantly in the cranioventral area, associated with mild pleural fibrin deposition. The right primary bronchus was obliterated by a Araucaria angustifolia pine branch measuring 18 cm in length, with adjacent darkened areas (lung consolidation). Microscopically, there was diffuse necrosis with severe hemorrhage in the lungs, associated with marked neutrophilic inflammatory infiltrate, numerous coccoid bacterial aggregates, and fibrinous pleuritis. Additionally, there was diffuse alveolar edema and multifocal thrombosis. Lung fragments were submitted for bacterial culture and mixed bacterial growth was observed with a predominance of Streptococcus equi subsp. zooepidemicus. Inhalation of branches is not commonly reported in horses, but it must be included in the differential diagnoses of pneumonia, and attention should be taken when allowing horses to graze in areas where the plant occurs. RESUMO: Problemas respiratórios devido a corpos estranhos (CEs) traqueobrônquicos são incomuns em equinos, embora cavalos em pastagem possam eventualmente aspirar CEs, como galhos de Araucaria angustifolia. Um potro Crioulo, 1,5 anos, apresentou hemoptise, dispneia, inquietação e temperatura retal de 40,9 ºC. O hemograma revelou intensa neutropenia, monocitose, trombocitopenia e hipoproteinemia. O tratamento foi realizado, mas sem sucesso. Na cavidade torácica, foi observada grande quantidade de líquido avermelhado livre (hemotórax). Os pulmões estavam difusamente consolidados, predominantemente cranioventral e com discreta deposição de fibrina sobre a superfície pleural. O brônquio principal direito estava obliterado por um ramo de pinheiro de Araucaria angustifolia com 18 cm de comprimento. Microscopicamente, notou-se necrose de coagulação pulmonar difusa com hemorragia severa, infiltrado inflamatório neutrofílico marcado, numerosos agregados bacterianos cocoides e pleurite fibrinosa. Fragmentos de pulmão foram submetidos ao isolamento bacteriológico e abundante crescimento misto com predominância de Streptococcus equi subsp. zooepidemicus foi observado. A inalação de grimpas de pinheiro não é comumente relatada em equinos, mas deve ser incluída nos diagnósticos diferenciais de pneumonia e deve-se ter atenção ao introduzir cavalos no campo com a presença da planta.

A mortalidade de porcas está diretamente relacionada a perdas financeiras e à eficiência produtiva das granjas. Apesar disso, as doenças associadas à morte de porcas são pouco conhecidas. Este estudo teve como objetivo determinar as principais causas de morte de porcas em granjas suinícolas brasileiras. Para a realização desta pesquisa, três granjas brasileiras foram visitadas e necropsias foram realizadas em todas as porcas que morreram espontaneamente ou foram submetidas à eutanásia. Fragmentos de tecidos foram coletados para exame histopatológico e bacteriológico. Além disso, foram registrados os sinais clínicos, estágio reprodutivo, ordem de parto e tipo de morte (espontânea ou eutanásia). Um total de 138 necropsias foram realizadas e 132 tiveram um diagnóstico conclusivo. Os estágios produtivos mais frequentes foram gestação e lactação (33,3 e 31,9%, respectivamente), seguidos por porcas parturientes (17,4%), porcas de descarte (9,4%), intervalo desmame-estro (IDE) (4,3%) e leitoas vazias (3,6%). Os sistemas orgânicos mais afetados foram reprodutor (28%), digestivo (25%), locomotor (22%), cardiovascular (9,1%) e hematopoiético (6,1%). As condições mais frequentemente diagnosticadas foram prolapso uterino (16/132; 12,1%), úlcera gástrica (13/132; 9,8%), artrite supurativa (11/132; 8,3%), torção do lobo hepático (11/132; 8,3%), insuficiência cardíaca (9/132; 6,8%), prolapso vaginal ou prolapso vaginal e retal (9/132; 6,8%) e pododermatite (8/132; 6,1%). Embora 58,2% dos óbitos tenham ocorrido por uma dessas sete doenças, houve grande variabilidade de diagnósticos. This study aimed to determine the main causes of death of sows in Brazilian pig farms. To perform this research, three Brazilian pig farms were visited, and necropsies were performed on all sows that had died spontaneously or were subjected to euthanasia. Tissue fragments were collected for histopathological and bacteriological examination. In addition, the clinical signs, productive stage, parity, and type of death (spontaneous or euthanasia) were recorded. A total of 138 necropsies were performed, and 132 had a conclusive diagnosis. The most frequent productive stages were gestation and lactation (33.3 and 31.9%, respectively), followed by parturient sows (17.4%), cull sows (9.4%), weaning-to-estrus interval (WEI) (4.3%), and unmated gilts (3.6%). The most affected organ systems were reproductive (28%), digestive (25%), locomotor (22%), cardiovascular (9.1%), and hematopoietic (6.1%). The most frequently diagnosed conditions were uterine prolapse (16/132; 12.1%), gastric ulcer (13/132; 9.8%), suppurative arthritis (11/132; 8.3%), liver lobe torsion (11/132; 8.3%), heart failure (9/132; 6.8%), vaginal or vaginal and rectal prolapse (9/132; 6.8%), and pododermatitis (8/132; 6.1%). Although 58.2% of the deaths were due to one of these seven diseases, there was a great variability in diagnoses.

No. of bitstreams: 0 Previous issue date: 2022-05-01 Holoprosencephaly is a central nervous system malformation, characterized by incomplete or total lack of division of prosencephalon hemispheres, which is commonly accompanied by craniofacial malformations. A 9-month-gestation aborted American Quarter Horse fetus was submitted for postmortem examination. The fetus lacked haircoat and had severe facial malformations including marked shortening/absence of the maxillary, incisive and nasal bones, bilateral anophthalmia, and pre-maxillary agenesis. The prosencephalon was small and nearly spherical, represented by a single lobe, with no visible separation between cerebral hemispheres. The olfactory bulbs, piriform lobes, and the optic chiasm were absent. At cross sectioning of the prosencephalon, the inner structures of the brain were completely absent, and replaced by a monoventricle lined by the remaining compressed cortex, and the thalami were fused. Since mutations in the sonic hedgehog (SHH) gene have been associated with human holoprosencephaly, the three coding SHH exons were sequenced using liver DNA of the aborted foal. The obtained SHH sequence was similar to the Equus caballus SHH mRNA sequence deposited in Genbank (XM_023640069.1); therefore, no polymorphism in the coding region of this gene justifying the phenotype was observed. This is the first report of alobar holoprosencephaly in horses.
