Phytoremediation is a plant-based approach to extract, stabilise, eliminate, or render pollutants into less harmful form. The study highlights the use of a native polar microalga as a means of phytoremediation in Antarctica where imported microbes are prohibited. Since 1959, Antarctica has been a protected region to preserve its dynamic ecosystems, but it is increasingly vulnerable to climate change and pollution. One of the anthropogenic disturbances in the continent is diesel spillage. Due to the extreme polar environment, natural attenuation of spilled diesel is severely hindered; hence, the problem calls for an effective and sustainable solution. This laboratory study proved that Antarctic microalga was capable of removing diesel (57.6%) through biodegradation and biosorption in the span of nine days. Meanwhile, mixotrophic cultivation triggered the vacuolar activities and potentially stimulated lipid assimilation in the cells. The microalgal-based process offers a cheap alternative in water decontamination while bearing the economic potential through the secretion of valuable products, such as biolipids.

Phenol is an important pollutant widely discharged as a component of hydrocarbon fuels, but its degradation in cold regions is challenging due to the harsh environmental conditions. To date, there is little information available concerning the capability for phenol biodegradation by indigenous Antarctic bacteria. In this study, enzyme activities and genes encoding phenol degradative enzymes identified using whole genome sequencing (WGS) were investigated to determine the pathway(s) of phenol degradation of Arthrobacter sp. strains AQ5-05 and AQ5-06, originally isolated from Antarctica. Complete phenol degradative genes involved only in the ortho-cleavage were detected in both strains. This was validated using assays of the enzymes catechol 1,2-dioxygenase and catechol 2,3-dioxygenase, which indicated the activity of only catechol 1,2-dioxygenase in both strains, in agreement with the results from the WGS. Both strains were psychrotolerant with the optimum temperature for phenol degradation, being between 10 and 15 °C. This study suggests the potential use of cold-adapted bacteria in the bioremediation of phenol pollution in cold environments.

Oil pollution such as diesel poses a significant threat to the environment. Due to this, there is increasing interest in using natural materials mainly from agricultural waste as organic oil spill sorbents. Oil palm’s empty fruit bunch (EFB), a cost-effective material, non-toxic, renewable resource, and abundantly available in Malaysia, contains cellulosic materials that have been proven to show a good result in pollution treatment. This study evaluated the optimum screening part of EFB that efficiently absorbs oil and the physicochemical characterisation of untreated and treated EFB fibre using Fourier Transform Infrared Spectroscopy (FTIR) and Scanning Electron Microscopy (SEM). The treatment conditions were optimised using one-factor-at-a-time (OFAT), which identified optimal treatment conditions of 170 °C, 20 min, 0.1 g/cm3, and 10% diesel, resulting in 23 mL of oil absorbed. The predicted model was highly significant in statistical Response Surface Methodology (RSM) and confirmed that all the parameters (temperature, time, packing density, and diesel concentration) significantly influenced the oil absorbed. The predicted values in RSM were 175 °C, 22.5 min, 0.095 g/cm3, and 10%, which resulted in 24 mL of oil absorbed. Using the experimental values generated by RSM, 175 °C, 22.5 min, 0.095 g/cm3, and 10%, the highest oil absorption achieved was 24.33 mL. This study provides further evidence, as the data suggested that RSM provided a better approach to obtain a high efficiency of oil absorbed.

Fucoxanthin is one of the light-harvesting pigments in brown microalgae, which is increasingly gaining attention due to its numerous health-promoting properties. Currently, the production of microalgal fucoxanthin is not yet feasible from an economic perspective. However, the cultivation of microalgae at favourable conditions holds great potential to increase the viability of this fucoxanthin source. Hence, this study aimed to review the fucoxanthin production of microalgae under different conditions systematically. A literature search was performed using the Web of Science, Scopus and PubMed databases. A total of 188 articles were downloaded and 28 articles were selected for the current review by two independent authors. Microalgae appeared to be a more reliable fucoxanthin source compared to macroalgae. Overall, a consensus fucoxanthin production condition was obtained and proposed: light intensity ranging from 10 to 100 µmol/m2/s could achieve a higher fucoxanthin content. However, the optimal light condition in producing fucoxanthin is species-specific. The current review serves as an antecedent by offering insights into the fucoxanthin-producing microalgae response to different culture factors via a systematic analysis. With the current findings and recommendations, the feasibility of producing fucoxanthin commercially could be enhanced and possibly achieve practical and sustainable fucoxanthin production.

The CRISPR/Cas9 system is a precise and versatile genetic tool used to induce site-specific manipulation in the targeted gene. This study elaborates on the strategy to select the gRNAs targeting the oil palm phytoene desaturase (EgPDS) gene, which complements CRISPR-GE and sequence analysis. Sixty-one gRNAs were selected from 167 gRNA candidates generated by the CRISPR-GE tool. These gRNA candidates were further analysed to meet the specific criteria, including the guanine (G) and cytosine (C) content of 35%-60% and the presence of preferable gRNA features. From 61 gRNAs, seven gRNAs were selected, which represent three different parts of the EgPDS gene in the genome. The efficiency of each gRNA candidate to cause cleavage on the target gene was confirmed by in vitro cleavage analysis. All seven gRNA candidates were then cloned into the pYLsgRNA-OsU6 cassette before being incorporated seamlessly into the final transformation vector, pYLCRISPR/Cas9P35S-H, via Gibson assembly. Three CRISPR/Cas9 transformation vectors targeting the EgPDS gene, namely pYLEgPDS1-35S-H, pYLEgPDS2-35S-H and pYLEgPDS3-35S-H, were successfully constructed. These constructs will be transformed into oil palm protoplasts via polyethylene glycol (PEG)- mediated transformation and subsequently into oil palm embryogenic calli via biolistic and Agrobacteriummediated transformation to determine the efficacy of the multiplex CRISPR/Cas9 system in oil palm genome.

Some cyanobacteria species have a high capacity for accumulating phycobiliprotein contents in their cells. However, there is a lack of information on screening tropical freshwater cyanobacteria, particularly phycobiliproteins. In addition, it is unclear which characteristics of cyanobacteria (morphological and/or growth) could affect phycobiliprotein contents. This study aimed to screen and characterise Malaysian indigenous freshwater cyanobacteria for the growth, biomass, and pigment contents and determine the major characteristic that contributed to the variation of phycobiliproteins. The surface/volume (S/V) ratio, specific growth rate, biomass productivity, and pigment contents of the isolated cyanobacteria were analysed. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) were applied to distinguish the factor responsible for phycobiliprotein variations in cyanobacteria. For the phycobiliprotein contents, Arthrospira sp., Pseudanabaena sp., and Synechococcus elongatus showed significantly higher (p < 0.05) amounts of cyanobacterial phycocyanin (C-PC), phycoerythrin (C-PE), and allophycocyanin (C-APC), respectively, than other studied cyanobacteria. This study showed no apparent trend of similarity and difference between the unicellular and filamentous cyanobacteria. In addition, carotenoid contents demonstrated a positive correlation with total phycobiliproteins, C-PC and C-APC. Based on the current findings, Arthrospira sp., Pseudanabaena sp. and Synechococcus elongatus might be the promising candidate to be the C-PC, C-PE and C-APC sources, respectively, for commercial production purposes. The selection of optimal cyanobacterial strain is crucial for efficient phycobiliprotein production. This study underlined the potential of freshwater cyanobacteria in producing respective and total phycobiliproteins.
