With the advancement of global maritime transport, the carriage of foreign organisms by vessels is inciting ecological catastrophes such as red tides, posing a grave threat to China's marine environmental security. Clarifying the geographical distribution of invasive species and their relationship with climate change can provide scientific basis for their prevention and control. In this discourse, the study centers on the typical alien microalgae, Nitzschiella closterium. By integrating environmental variables and distribution data and employing an optimized Maxent model, we delve into its potential distribution in current and future ecological conditions in the coastal waters of China, as well as the response relationship to environmental variables. After model optimization, the results reveal that the Area Under the Curve (AUC) value of the training dataset for Nitzschiella closterium stands at 0.993, denoting a high degree of model accuracy and bestowing credibility upon the simulated outcomes. Currently, the high suitability area constitutes 0.74% of the total suitability area, with an area of 0.44×104 km², and is mainly distributed in the sea area corresponding to Jiangsu Province, Shanghai City, and Zhejiang Province. The most significant environmental factor affecting the geographical distribution of Nitzschiella closterium is the annual temperature variation range (bio24). In the context of future climate changes, there is an overall trend of substantial expansion in the high suitability area for Nitzschiella closterium, which is profoundly affected by climate change, suggesting that the future control will be more severe. This research comprehensively understands the response relationship of the invasive species Nitzschiella closterium to leading environmental variables and its potential geographical distribution in the context of climate change through macro prediction and proposes control countermeasures. The three leading modes of the North Atlantic atmospheric circulation explain about 70% of the winter climate variability. Although climate models generally can capture these modes, biases may induce large uncertainties in regional climate predictions. Here, we evaluate the leading winter modes simulated by CMIP5‐PMIP3 and CMIP6‐PMIP4 models from the last millennium to future scenarios in comparison with historical reanalysis and paleo‐reconstructions. The models generally have a good representation of the average spatial pattern of the North Atlantic Oscillation (NAO) while showing a larger spread in performance for the East Atlantic and Scandinavian patterns. In contrast to historical reanalysis, the simulated NAO pattern tends to be rather stationary under various climate states over the years 861–2100. Such underestimated spatial variability in the simulated NAO is directly related to the biased spatial shifts in NAO‐related regional temperature and precipitation changes, inducing uncertainties in climate projections over the North Atlantic sector. Plain Language Summary: The North Atlantic Oscillation (NAO) is the most dominant weather pattern over the North Atlantic‐European sector with profound influence on regional climate. The NAO pattern describes the changes in a low‐pressure system over Iceland and a high‐pressure system over the Azores, which are commonly known as the NAO centers of action. However, the NAO centers of action are not always fixed in these two locations, but show spatial movement over time. In this study, we compare the winter weather patterns over the North Atlantic region from the last millennium to future scenarios in climate models with reanalysis products and paleoclimate reconstructions. We find that models generally can capture the spatial structure of the NAO very well but with little changes in its location and shape over time in contrast to historical reanalysis. These biased NAO patterns in climate models can shift the modeled regional temperature and precipitation patterns associated with the NAO. This remains a challenge for the reliability of future climate projections over the North Atlantic region. Rice bran dietary fiber (RBDF) is a byproduct of rice milling that has potential applications in breadmaking. This study investigated the effects of adding RBDF at different soluble and insoluble fractions on bread quality and storage properties. RBDF was separated into soluble (RBSDF) and insoluble (RBIDF) fractions and added to the bread at various concentrations. The results showed that adding an appropriate amount of RBSDF (6%) improved the texture, specific volume, sensory evaluation, and shelf life of bread while adding too much RBSDF (12%) or RBIDF (>8%) reduced the quality of bread and increased its hardening and moisture loss during storage. The bread quality was improved when the addition of RBSDF and RBIDF were 6% and 5%, respectively, which could enhance the fiber intake, meeting the diversified dietary needs of consumers. Decision response and feedback in gambling are interrelated. Different decisions lead to different ranges of feedback, which in turn influences subsequent decisions. However, the mechanism underlying the continuous decision-feedback process is still left unveiled. To fulfill this gap, we applied the hidden Markov model (HMM) to the gambling electroencephalogram (EEG) data to characterize the dynamics of this process. Furthermore, we explored the differences between distinct decision responses (i.e. choose large or small bets) or distinct feedback (i.e. win or loss outcomes) in corresponding phases. We demonstrated that the processing stages in decision-feedback process including strategy adjustment and visual information processing can be characterized by distinct brain networks. Moreover, time-varying networks showed, after decision response, large bet recruited more resources from right frontal and right center cortices while small bet was more related to the activation of the left frontal lobe. Concerning feedback, networks of win feedback showed a strong right frontal and right center pattern, while an information flow originating from the left frontal lobe to the middle frontal lobe was observed in loss feedback. Taken together, these findings shed light on general principles of natural decision-feedback and may contribute to the design of biologically inspired, participant-independent decision-feedback systems. Water scarcity is a growing global concern, resulting from drastic climate and societal changes witnessed over the past few decades. Previous attention has predominantly focused on inadequate water quantity as the main cause of water scarcity, but pollution-induced water scarcity has been largely overlooked. Furthermore, there is a lack of understanding about the effectiveness of policy interventions in mitigating water scarcity and its social impact. To address these knowledge gaps, we developed an ecosystem service-based approach to measure quantity- and quality-related water scarcity by comparing supply of and demand for water provision and water purification services. We also developed several water and land management scenarios to examine their potential in alleviating water scarcity under different climate conditions and water quality requirements. Our case study in the Taihu Basin of southeastern China identified four types of water scarcity status across different sub-basins, including quantity-based, quality-based, quantity- and quality-based, and no water scarcity. The degradation of water quality in the Taihu Basin has exacerbated the scarcity of clean water supply. Water conservation measures effectively alleviated quantity-based water scarcity, while nitrogen reduction and grain for green (i.e., conversion of cropland into forest land) exhibited similar efficacy in alleviating quality-based water scarcity. Integrating these measures contributed to alleviating both quantity- and quality-based water scarcity. However, the effectiveness of these measures decreased under drier climate conditions and higher water quality requirements, highlighting the importance of climate and water quality in determining the magnitude, extent, and severity of water scarcity. Color-switch electrochemiluminescence (ECL) sensing platform based on a dual-bipolar electrode (D-BPE) is reported in this work. The D-BPE was composed of a cathode filled with buffer and two anodes filled with [Ru(bpy) 3 ]2+-TPrA and luminol-H 2 O 2 solutions, respectively. Both anodes were modified with capture DNA and served as ECL reporting platforms. After introducing ferrocene-labeled aptamer (Fc-aptamer) on both anodes, the ECL emission signal of the [Ru(bpy) 3 ]2+ was difficult to be observed (anode 1), while luminol emitted a strong and visible ECL signal (anode 2). Ferrocene (Fc) did not only prevent the oxidation of [Ru(bpy) 3 ]2+ due to its lower oxidation potential, its oxidation product Fc+ also quenched the [Ru(bpy) 3 ]2+ ECL through efficient energy transfer. For luminol, Fc+ catalyzes the accelerated formation of the excited-state of the luminol anion radical, which leads to the enhancement of the luminol ECL. In the presence of food-borne pathogens, the aptamer was assembled with them, leading to the leaving of Fc from the surface of the D-BPE anodes. The ECL intensity of [Ru(bpy) 3 ]2+ was enlarged, meanwhile, the blue emission signal of luminol became weakened. By self-calibrating the ratio of the two signals, 1–106 CFU mL−1 food-borne pathogenic bacteria can be sensitively detected with a detection limit of 1 CFU mL−1. Ingeniously, the color-switch biosensor can be used to detect S. aureus , E. coli and S. typhimurium by assembling the corresponding aptamers onto the D-BPE anodes. [ABSTRACT FROM AUTHOR]

After discovery of electron transfer from bacteria, most bacteria known to be electrochemically active are utilized as a self-regenerable catalyst at the anode of microbial fuel cells (MFCs). However, the reverse phenomenon, cathodic catalysts is not so widely researched. This present study demonstrated that E. coli O157:H7 was electrochemically active, and it was able to catalyze oxygen reduction at the cathode of bipolar electrode (BPE). Applying a constant potential to the BPE, E. coli O157:H7 can catalyze electrochemical reduction of O 2 , decrease the overpotential of O 2 reduction at the cathode, which in turn generates an electrochemiluminescence (ECL) reporting intensity change at the anode. Significantly, a majority of food matrix does not exhibit catalytic activity for electrochemical reduction of O 2. Meanwhile, due to the physically separation of two poles of closed BPE, complex food matrix at the cathode does not interfere with the ECL reaction at the anode. Therefore, the effect of food matrix is negligible when measuring E. coli O157:H7 levels in food. A low detection limit of 10 CFU mL−1 E. coli O157:H7 could be identified within 1 h. A series of novel 4-arylaminoquinazoline derivatives were synthesized by five-step reactions, the structure was characterized by IR, 1H NMR, MS and single crystal X-ray diffraction. The result of 5aX crystal was as follows: the crystal belongs to the monoclinic system, space group P21/c with a = 12.3637(4) Å, b = 12.7902(2) Å, c = 13.2240(5) Å, α = 90°, β = 117.030(5)°, γ = 90°, V = 1862.75(12) Å3, Z = 2, F(000) = 804.0, µ = 0.095 mm-1, S = 1.039, R = 0.0569, wR = 0.1535 for 8417 observed reflections with I > 2σ(I). The compound 5cX exhibited an excellent inhibitory effect on the cell line A549 (IC50 < 2.5 µM) and the drug-resistant cell line H1975 (IC50 < 2.5 µM) by the biological activity, which were superior to the positive control Zorifertinib (against A549: IC50 = 31.08 µM; against H1975: IC50 = 64.17 µM). Molecular docking revealed that the better inhibitory activity of compound 5cX was owing to the combining EGFRWT and EGFRL858R/T790M by hydrogen bonding. • A temporal sensing platform of Staphylococcus aureus based on Ag@C hybrid BPE are established. • Anodic dissolution can improve sensitivity and extend the detection pattern to the time dimension. • The interference of food matrix can be ignored in this strategy. An Ag@C hybrid bipolar electrode (BPE) sensing platform has been established for the temporal detection of Staphylococcus aureus (S. aureus) in food. Combining the advantages of anodic dissolution of Ag and cathodic biocatalysis of oxygen (O 2) reduction, this strategy showed an ultralow detection limit down to 10 CFU mL−1. As the formation of Ag@C completely quenched the electrochemiluminescence (ECL) emission of luminol, the ECL emission recovery reflected the extent of anodic dissolution. Meanwhile, S. aureus catalyzed the electrochemical reduction of O 2 at the cathode, reducing the overpotential for cathodic O 2 reduction and thus increasing the rate of anodic electron loss, facilitating Ag dissolution and restoring the ECL emission of luminol. When a constant potential was applied, through monitoring the ECL recovery time before and after the incubation of S. aureus on the cathode, S. aureus could be quantified due to the slight difference of the conductivity. • An integrated framework for ecological protection and restoration is proposed. • Recreational service is estimated by considering spatial accessibility. • Priority areas of protection are identified based on trade-off of ecosystem service. • Ecological protection and restoration pattern is identified at the two scales. • Land management measures are proposed for ecological protection and restoration. Ecological protection and restoration is an important bridge to adjust the contradiction between ecological protection and economic development, and it is essential for ecological security and sustainable development. An integrated evaluation framework for multiscale ecological protection and restoration based on multi-scenario trade-offs of ecosystem services was proposed in this study, and the city of Nanjing was taken as an example to identify key areas and land strategies for ecological protection and restoration. In our framework, carbon storage, habitat quality, nitrogen retention, and water conservation services were quantified by InVEST model, and recreational service was quantified as the total areas of green spaces within ecological land accessibility. The ordered weighted averaging model was used to simulate the priority areas of ecological protection under 11 scenarios of trade-offs of ecosystem services, and ecological sources, ecological corridors, ecological pinch points and ecological barriers at the Nanjing city scale and the main urban scale were identified based on circuit theory. We also nested the ecological conservation and restoration patterns at two scales and analyzed the coincidence degree between two scales. Our results showed that the S7 scenario had the highest average conservation efficiency among the 11 scenarios. The priority areas for ecological protection in this scenario had an area of 1582 km2 and are mainly distributed in the central and southern parts of Nanjing. In our study, 50 ecological sources (with an area of 1153.36 km2), 77 ecological corridors (with a length of 443.04 km), 25 key nodes for ecological protection and 31 key nodes for ecological restoration were identified in Nanjing. By nesting the ecological protection and restoration patterns of two scales, we found that the overlap of ecological sources and ecological pinch points at two scales is 100%, and the overlap of ecological corridors and ecological barriers is 45.28% and 57.14%, respectively, which means that the ecological protection and restoration patterns at the two scales in Nanjing were well connected, but the ecological network at the large scale cannot fully reflect the real ecological problems and needs at the small scale. We suggest regarding the ecological protection and restoration pattern at large scale as the basis and the pattern at small scale as the supplement. We also propose that priority should be given to the protection and restoration of key areas where the two scales overlap, and strictly control their land development to promote sustainable development. In order to solve the problem of insufficient supply of aggregate for cemented filling material, the experimental research on the mix ratio of cemented filling material using construction waste as recycled coarse aggregate was carried out. A ternary quadratic regression model was created using the Box-Behnken design (BBD) based on the response surface methodology (RSM), using slump, bleeding ratio, and 28 d uniaxial compressive strength of the filling material as response variables, and the model's accuracy and reliability were confirmed. The findings of the regression model reveal that the response value is influenced by a single component as well as the interaction between the two factors. Finally, based on filling material cost optimization model, an optimal mix ratio is given, in which the wet fly ash/aggregate ratio, Talbol gradation index of recycled aggregate, and dosage of water reducing agent to cement are 0.507, 0.5, and 0.678%, respectively. The filling material which adopted the optimal mix ratio exhibited a good performance with slump of 215 mm and bleeding ratio of 4.00%, uniaxial compressive strength increased significantly within 3 d, and 28 d uniaxial compressive strength was 4.08 MPa, which meet the requirements of field construction. Background: The 5-5-1 criteria (> 5 minutes - 5 cannulation attempts - 1 unintended pancreas duct cannulation) were proposed by the European Society of Gastrointestinal Endoscopy to define difficult biliary cannulation. However, the criteria may be inappropriate for trainee-involved procedures. We developed criteria for difficult cannulation in trainee-involved procedures.Methods: Patients undergoing biliary cannulation with or without trainee involvement were eligible. Procedures that might be too easy (e. g. fistula) or too difficult (e. g. altered anatomy) were excluded. The primary outcome was difficult cannulation, defined as cannulation time, attempts, or inadvertent pancreatic duct (PD) cannulation exceeding the 75 % percentile of each variable. Propensity score matching (PSM) analysis was used.Results: After PSM, there were 1596 patients in each group. Trainee-involved procedures had longer median (interquartile range [IQR]) cannulation time (7.5 [2.2-15.3] vs. 2.0 [0.6-5.2] minutes), and more attempts (5 [2-10] vs. 2 [1-4]) and inadvertent PD cannulation (0 [0-2] vs. 0 [0-1]) vs. procedures without trainee involvement (all P < 0.001). The 15-10-2 criteria for difficult cannulation were proposed for trainee-involved cannulation and the 5-5-1 criteria were nearly confirmed for cannulation without trainee involvement. The proportions of difficult cannulation using these respective criteria were 35.5 % (95 % confidence interval [CI] 33.2 %-37.9 %) and 31.8 % (95 %CI 29.5 %-34.2 %), respectively (odds ratio 1.18 [95 %CI 1.02-1.37]). Incidences of post-ERCP pancreatitis following difficult cannulation were comparable (7.8 % [95 %CI 5.7 %-10.3 %] vs. 9.8 % [95 %CI 7.4 %-12.8 %], respectively).Conclusion: By using the 75 % percentiles as cutoffs, the proposed 15-10-2 criteria for difficult cannulation could be appropriate in trainee-involved procedures. Synthetic lethality was proposed nearly a century ago by geneticists and recently applied to develop precision anti-cancer therapies. To exploit the synthetic lethality concept in the design of chemical anti-cancer agents, we developed a bio-orthogonally catalyzed lethality (BCL) strategy to generate targeting anti-tumor metallodrugs both in vitro and in vivo. Metallodrug Ru-rhein was generated from two non-toxic species Ru-N3 and rhein-alkyne via exclusive endogenous copper-catalyzed azide alkyne cycloaddition (CuAAC) reaction without the need of an external copper catalyst. The non-toxic species Ru-arene complex Ru-N3 and rhein-alkyne were designed to perform this strategy, and the mitochondrial targeting product Ru-rhein was generated in high yield (>83%) and showed high anti-tumor efficacy in vitro. This BCL strategy achieved a remarkable tumor suppression effect on the tumor-bearing mice models. It is interesting that the combination of metal-arene complexes with rhein via CuAAC reaction could transform two non-toxic species into a targeting anti-cancer metallodrug both in vitro and in vivo , while the product Ru-rhein was non-toxic towards normal cells. This is the first example that exclusive endogenous copper was used to generate metal-based anti-cancer drugs for cancer treatment. The anti-cancer mechanism of Ru-rhein was studied and autophagy was induced by increased reactive oxygen species and mitochondrial damage. The generality of this BCL strategy was also studied and it could be extended to other metal complexes such as Os-arene and Ir-arene complexes. Compared with the traditional methods for cancer treatment, this work presented a new approach to generating targeting metallodrugs in vivo via the BCL strategy from non-toxic species in metal-based chemotherapy. Context. The immune system of piglets is not fully developed at weaning. The immature adaptive immune system along with the change in the feed types and the living conditions make piglets susceptible to pathogenic infections, and result in diarrhoea and decreased growth. However, the underlying mechanisms require further research. Aims. This study was conducted to investigate the effects of weaning on the growth performance and intestinal immune function of piglets. Methods. In total, 40 piglets with similar bodyweights were selected in pairs at 21 days old and divided into the suckling group (SG, breastfed by their mothers) and weaning group (WG, weaned at 21 days old). Eight piglets from each group were randomly selected and sacrificed at 24 days (SG3 and WG3) and 28 days of age (SG7 and WG7). The growth performance, T lymphocyte subpopulations, the concentration of cytokines and immunoglobulins, and the expression of Notch2 signalling proteins were determined. Key results. The weaning caused a decrease in bodyweight (P < 0.01) and the ratio of CD3+CD4+/CD3+CD8+ T cells in the thymus (P < 0.05). Compared with SG3, the concentration of secretory immunoglobulin A (sIgA) in the jejunum was decreased, and that of interleukin 2 (IL-2) in serum and ileum, IL-1β and IL-2 in jejunum were upregulated (P < 0.01), while IL-10 in the small intestine was downregulated (P < 0.05) in WG3. Weaning downregulated gene expression of IL-4 and upregulated gene expression of IL-1β, IL-12, and interferon γ (IFN-γ) in small intestine (P < 0.05). Weaning downregulated protein expression of Notch2 and Hes1 but upregulated Jagged1 expression in small intestine of piglets (P < 0.05). Conclusions. Weaning caused an imbalance in T lymphocyte distribution, thus impairing the intestinal immune function of piglets, which might be associated with the Notch2 signalling. Furthermore, weaning caused adverse effects on piglets especially on the first 3 days, which were partly recovered after 3 days, but still existed at 7 days after weaning. Implications. Preventing pathogenic viral and bacterial transmission in the human environment is critical, especially in potential outbreaks that may be caused by the release of ancient bacteria currently trapped in the permafrost. Existing commercial disinfectants present issues such as a high carbon footprint. This study proposes a sustainable alternative, a bioliquid derived from biomass prepared by hydrothermal liquefaction. Results indicate a high inactivation rate of pathogenic virus and bacteria by the as-prepared bioliquid, such as up to 99.99% for H1N1, H5N1, H7N9 influenza A virus, and Bacillus subtilis var. niger spores and 99.49% for Bacillus anthracis. Inactivation of Escherichia coli and Staphylococcus epidermidis confirmed that low-molecular-weight and low-polarity compounds in bioliquid are potential antibacterial components. High temperatures promoted the production of antibacterial substances via depolymerization and dehydration reactions. Moreover, bioliquid was innoxious as confirmed by the rabbit skin test, and the cost per kilogram of the bioliquid was $0.04427, which is notably lower than that of commercial disinfectants. To clarify the path of the temporal branch of facial nerve (TB) crossing the zygomatic arch (ZA). Eighteen fresh adult heads specimens were carefully dissected in the zygomatic region, with the location of TB as well as its number documented. The hierarchical relationship between the temporal branch and the soft tissue in this region was observed on 64 P45 plastinated slices. 1. TB crosses the ZA as type I (21.8 %), type II (50.0 %,), and type III (28.1 %) twigs. 2. At the level of the superior edge of the ZA, the average distance between the anterior trunk of TB and the anterior part of the auricle is 36.36±6.56 mm, for the posterior trunk is 25.59±5.29 mm. At the level of the inferior edge of the ZA, the average distance between the anterior trunk of TB and the anterior part of the auricle is 25.77±6.19 mm, for the posterior trunk is 19.16±4.71 mm. 3. The average length of ZA is 62.06±5.36 mm. TB crosses the inferior edge of the ZA at an average of 14.67±6.45 mm. TB crosses the superior edge of the ZA at an average of 9.08±4.54 mm. 4. At the level of the ZA, TB passes on the surface of the pericranium while below the SMAS. The TB obliquely crosses the middle 1/3 part of the superior margin of the ZA and the junction of the middle 1/3 part and the posterior 1/3 part of the inferior margin of the ZA below the SMAS while beyond the periosteum. Value perception is an important basis for behavior coordination in collaborative governance. Two variables, collaborative ability and relationship quality, were introduced to form the theoretical framework of "perceived-competency-relations-behavior". A chain-mediated model was constructed to explore the mediating effects of collaborative ability, relationship quality, and their joint influence on the relationship between value perception and behavior coordination. The results show that: value perception can significantly enhance behavior coordination in atmospheric collaborative governance; collaborative ability and relationship quality respectively serve as a mediator between value perception and behavior coordination; collaborative ability and relationship quality jointly play a chain mediating role between value perception and behavior coordination. The conclusions enrich the research on the intrinsic mechanism of value perception affecting behavior coordination, and can provide a reference for decision-making in exploring behavior coordination paths. Backgrounds: Many studies suggest that both psychotherapy and drug therapy are effective in the treatment of bipolar disorders (BDs). However, the pathophysiology of both types of intervention has not been established definitively. Methods: An activation likelihood estimation meta-analysis was performed to identify the distinct brain activity alterations between psychotherapy and drug therapy for the treatment of BDs. Articles were identified by searching databases including PubMed, Embase, Cochrane Library, and Web of Science databases. Eligible studies on BDs were published up until 10 June 2021. Results: 21 studies were included and we conducted a meta-analysis for different therapies and imaging tasks. After receiving psychotherapy, BD patients showed increased activation in the inferior frontal gyrus (IFG) and superior temporal gyrus. While after taking drug therapy, BD patients displayed increased activation in the anterior cingulate cortex, medial frontal gyrus, IFG, and decreased activation in the posterior cingulate cortex. The regions of brain activity changes caused by psychotherapy were mostly focused on the frontal areas, while drug therapy mainly impacted on the limbic areas. Different type of tasks also affected brain regions which were activated. Conclusions: Our comprehensive meta-analysis indicates that these two treatments might have effect on BD in their own therapeutic modes. Psychotherapy might have a top-down effect, while drug therapy might have a bottom-up effect. Planned urban form has become an important strategy to improve air quality in urban agglomerations (UAs), especially pollution due to PM 2.5 , but the influencing mechanisms are not yet clear. This study explores the relationship between four metrics of urban form (size, fragmentation, shape, and dispersion) as determined by analysis of remotely sensed images at 30-m resolution and PM 2.5 concentrations in 19 Chinese UAs. The influence of level of urban development and season is examined. Five control variables, including population density, temperature, precipitation, wind speed, and the normalized difference vegetation index (NDVI) are selected for use in multiple linear regression models. Size, fragmentation, and shape of urban form, but not dispersion, were found to have significant effects on PM 2.5 concentrations of different urbanization-level UAs. Urban size and fragmentation have stronger impacts on PM 2.5 concentrations in UAs with lower urbanization levels while urban shape has a greater impact in higher-level UAs. In terms of seasonal variation in all UAs, urban form is more pronouncedly associated with PM 2.5 concentrations during spring and autumn than summer and winter. Urban size and fragmentation are positively associated with PM 2.5 concentrations whereas urban shape and dispersion are on the contrary. The relationships between urban form and PM 2.5 uncovered here underscore the importance of urban planning as a tool to minimize PM 2.5 pollution. Specifically, local government should encourage polycentric urban form with lower fragmentation in urban agglomerations. UAs with lower urbanization levels should control the disordered expansion of construction land and higher-level UAs should promote the mix of green land and construction land. Both alternative sigma factor RpoS and ubiquitous secondary messenger c-di-GMP participate in the biofilm forming of Salmonella Pullorum; however, the relationship between RpoS and c-di-GMP-regulated genes during biofilm forming remains unclear. In this study, nine genes related with c-di-GMP regulation were found to be differentially expressed (P<0.01) by RNA-seq analysis when compared with S. Pullorum strain S9 and its rpoS deletion strain S9S. Specifically, the rpoS deletion strain S9S had higher transcription level of gene STM1703 and lower c-di-GMP concentration and biofilm-forming ability than S9 (P<0.01). The STM1703 gene deletion in strains S9 and S9S significantly enhanced the c-di-GMP concentration and the biofilm-forming ability (P<0.01). qRT–PCR analysis showed that rpoS deletion or P193L substitution in RpoS increased the transcription level of the STM1703 gene by decreasing the transcription levels of the csrA and STM1344 genes (P<0.01). Context: Obesity, one of the major public health problems worldwide, has attracted increasing attention. Ginsenoside Rb1 is the most abundant active component of Panax ginseng C.A.Mey (Araliaceae) and is reported to have beneficial effects on obesity and diabetes. However, the mechanisms by which Rb1 regulates obesity remain to be explored. Objective: This paper intends to further explore the mechanism of Rb1 in regulating obesity. Materials and methods: The C57BL/6 obese mice were divided into two groups: the control (CTR) and Rb1. The CTR group [intraperitoneally (ip) administered with saline] and the Rb1 group (ip administered with Rb1, 40mg/kg/d) were treated daily for four weeks. In vitro, Rb1 (0, 10, 20, 40 lM) was added to differentiated C2C12 cells and Rb1 (0, 20, 40 lM) was added to 3T3-L1 cells. After 24 h, total RNA and protein from C2C12 cells and 3T3-L1 cells were used to detect myostatin (MSTN) and fibronectin type III domain-containing 5 (FNDC5) expression. Results: Rb1 reduced the body weight and adipocyte size. Improved glucose tolerance and increased basic metabolic activity were also found in Rb1 treated mice. MSTN was downregulated in differentiated C2C12 cells, 3T3-L1 cells and adipose tissues upon Rb1 treatment. FNDC5 was increased after Rb1 treatment. However, MSTN overexpression attenuated Rb1-mediated decrease accumulation of lipid droplets in differentiated 3T3-L1 adipocytes. Discussion & Conclusions: Rb1 may ameliorate obesity in part through the MSTN/FNDC5 signalling pathway. Our results showed that Rb1 can be used as an effective drug in the treatment of human obesity. Background and Aim: Functional dyspepsia symptom diary (FDSD) is a newly designed questionnaire of functional dyspepsia (FD). The relationships between FDSD and other FD‐related questionnaires and patient‐reported severity remain unclear. This study aims to investigate the correlations between FDSD and other questionnaires and to determine the relationships between FDSD and FD severity. Methods: Consecutive outpatients with FD were prospectively enrolled in four tertiary hospitals. All patients were evaluated by six FD‐related questionnaires, including FDSD, Dyspepsia Symptom Severity Index (DSSI), Gastrointestinal Symptom Rating Scale, Short‐Form Nepean Dyspepsia Index, and Hamilton Depression and Anxiety Scale (HAMD and HAMA). The severity of FD was also graded as mild, moderate, and severe by patients themselves. Correlations between different scores were assessed by Spearman correlation coefficient (ρ), and risk factors for patient‐reported severity were identified. Results: For 512 enrolled FD patients, the overall median FDSD score was 19 (2–42). FDSD was well correlated with DSSI (ρ = 0.64) and fairly correlated with the other four scores (ρ = 0.32–0.55) (all P < 0.001). Mild, moderate, and severe FD were reported by 18.5%, 55.9%, and 25.6% of patients, respectively. There were seven factors associated with the severe FD, including education level, duration, and subtypes of FD, prior treatment, FDSD, HAMD, and HAMA scores (all P < 0.10). FDSD ≥ 20 (odds ratio [OR] 3.3, 95% confidence interval [CI]: 2.0–5.2, P < 0.001) and HAMD ≥ 13 (OR 2.9, 95% CI: 1.8–4.6, P < 0.001) were independently associated with patient‐reported severe FD. Conclusions: This study firstly revealed that the newly developed FDSD correlated with other FD‐related questionnaires. INTRODUCTION: Up to 60% of patients with common bile duct stone (CBDS) recurrence suffer from further recurrence after endoscopic retrograde cholangiopancreatography (ERCP). There are no effective methods to prevent recurrence in most patients. In this study, we aimed to assess the short-term and long-term efficacies of endoscopic papillary large balloon dilation (EPLBD) for the management of recurrent CBDS in a randomized controlled trial. METHODS: Consecutive patients with recurrent CBDS were eligible and randomly assigned in a 1:1 ratio to the EPLBD group or the control group. The primary outcome was the CBDS recurrence rate within 2 years after ERCP. The analysis followed the intention-to-treat principle. RESULTS: From 2014 to 2021, 180 patients with recurrent CBDS were included, with 90 in each group. All patients underwent complete CBDS clearance by 1 or several sessions of ERCP. The rate of complete clearance in 1 session was significantly higher with EPLBD treatment (95.6% vs 85.6%, P = 0.017). During the follow-up, the CBDS recurrence rate within 2 years was significantly lower in the EPLBD group than in the control group (21.1% [19/90] vs 36.7% [33/90], relative risk 0.58, 95% confidence interval 0.36–0.93, P = 0.021). At a median follow-up of approximately 56 months, CBDS recurrence was found in 34.4% of the patients (31/90) in the EPLBD group and 51.1% (46/90) in the control group (hazard ratio 0.57, 95% confidence interval 0.36–0.89, P = 0.012). Multiple recurrences (≥2) were also decreased in the EPLBD group (4.4% vs 18.9%, P = 0.020). DISCUSSION: During the long-term follow-up, nearly half of the patients with recurrent CBDS experienced stone recurrence after traditional ERCP. Polyploidy plays an important role in the evolution of eukaryotes, especially for flowering plants. Many of ecologically or agronomically important plant or crop species are polyploids, including sycamore maple (tetraploid), the world second and third largest food crops wheat (hexaploid) and potato (tetraploid) as well as economically important aquaculture animals such as Atlantic salmon and trout. The next generation sequencing data enables to allocate genotype at a sequence variant site, known as genotyping by sequencing (GBS). GBS has stimulated enormous interests in population based genomics studies in almost all diploid and many polyploid organisms. DNA sequence polymorphisms are codominant and thus fully informative about the underlying genotype at the polymorphic site, making GBS a straightforward task in diploids. However, sequence data may usually be uninformative in polyploid species, making GBS a far more challenging task in polyploids. This paper presents novel and rigorous statistical methods for predicting the number of sequence reads needed to ensure accurate GBS at a polymorphic site bared by the reads in polyploids and shows that a dozen of reads can ensure a probability of 95% to recover all constituent alleles of any tetraploid genotype but several hundreds of reads are needed to accurately uncover the genotype with probability confidence of 90%, subverting the proposition of GBS using low coverage sequence data in the literature. The theoretical prediction was tested by use of RAD‐seq data from tetraploid potato cultivars. Cerebral autosomal dominant arteriopathy, subcortical infarcts, and leukoencephalopathy (CADASIL) is the most common monogenic form of small vessel disease characterized by migraine with aura, leukoaraiosis, strokes, and dementia. CADASIL mutations cause cerebrovascular dysfunction in both animal models and humans. Here, we showed that 2 different human CADASIL mutations (Notch3 R90C or R169C) worsen ischemic stroke outcomes in transgenic mice; this was explained by the higher blood flow threshold to maintain tissue viability compared with that in wild type (WT) mice. Both mutants developed larger infarcts and worse neurological deficits compared with WT mice, regardless of age or sex after filament middle cerebral artery occlusion. However, full-field laser speckle flowmetry during distal middle cerebral artery occlusion showed comparable perfusion deficits in mutants and their respective WT controls. Circle of Willis anatomy and pial collateralization also did not differ among the genotypes. In contrast, mutants had a higher cerebral blood flow threshold, below which infarction ensued, suggesting increased sensitivity of brain tissue to ischemia. Electrophysiological recordings revealed a 1.5- to 2-fold higher frequency of peri-infarct spreading depolarizations in CADASIL mutants. Higher extracellular K+ elevations during spreading depolarizations in the mutants implicated a defect in extracellular K+ clearance. Altogether, these data reveal a mechanism of enhanced vulnerability to ischemic injury linked to abnormal extracellular ion homeostasis and susceptibility to ischemic depolarizations in CADASIL. Background and Aim: Rectal indomethacin and pancreatic duct stenting (PDS) are recommended for the prevention of post‐endoscopic retrograde cholangiopancreatography pancreatitis (PEP). However, the effects of the combination of the two methods on preventing PEP are controversial. We hypothesized that some group of difficult patients might benefit from the combination of indomethacin plus PDS (IP) compared with indomethacin alone (IN). Methods: Patients with native papilla who underwent endoscopic retrograde cholangiopancreatography in eight tertiary hospitals were screened. They were enrolled if the cannulation proved difficult and post‐procedure indomethacin was administered. Propensity score matching (PSM) was used to balance the baseline characteristics between IP and IN groups. The primary outcome was PEP. Results: Among 4456 patients with available cannulation‐related data, 1889 (42.4%) patients had difficult cannulation and received indomethacin. After PSM, both IP and IN groups included 332 patients. PEP was comparable between the two groups (12.7% vs 10.2%, P = 0.329). By subgroup analysis, the PEP rate was found to be lower in the IP group than in the IN group (7.3% vs 18.2%, P = 0.026) in patients undergoing double‐guidewire technique (DGT). The results of an additional analysis using PSM in DGT patients were consistent with the subgroup analysis results (7.8% vs 19.4%, P = 0.036). Conclusions: The current study indicated that the combined prevention of PEP with indomethacin plus PDS was useful in PEP prevention in patients undergoing DGT. • A functional BPE-ECL platform based on biocathode reducing oxygen was constructed. • Electrochemically active bacteria were able to catalyze oxygen reduction at the cathode of BPE. • A low detection limit of 30 CFU (30 mL system) could be identified within 30 min. A functional bipolar electrode-electrochemiluminescence (BPE-ECL) platform based on biocathode reducing oxygen was constructed for detecting electrochemically active bacteria (EAB) in this paper. Firstly, thiolated trimethylated chitosan quaternary ammonium salt (TMC-SH) layer was assembled on the gold-plated cathode of BPE. TMC-SH contains quaternary ammonium salt branch chain, which can inhibit the growth of microorganisms on the surface or in the surrounding environment while absorbing bacteria. Then, the peristaltic pump was used to flow all of the samples through the cathode, and the EAB was electrostatically adsorbed on the electrode surface. Finally, applying a constant potential to the BPE, bacteria can catalyze electrochemical reduction of O 2 , and decrease the overpotential of O 2 reduction at the cathode, which in turn generates an ECL reporting intensity change at the anode. Although it is challenging to integrate supply and demand to comprehensively understand urbanization effects on ecosystem services, this knowledge is essential, especially in rapidly urbanizing areas. A spatially explicit approach was developed here to synthesize supply and demand dynamics of the two most important hydrologic services in the rapidly urbanizing Taihu Lake Basin (TLB) of eastern China. The supply of water purification (WP S) and flood mitigation (FM S) were measured as nitrogen removal and runoff retention, respectively. Whereas the demand for water purification (WP D) was quantified as the difference between total and permitted nitrogen loading based on relevant water quality standards, the demand for flood mitigation (FM D) was estimated as the vulnerability to potential flood damage, including economic losses and causalities. We found a spatial mismatch where high WP S and FM S occurred in mountainous areas while high WP D and FM D concentrated in urban and agricultural areas across the basin. WP S and WP D decreased by 10% and 20%, respectively during 2000–2015, due mainly to loss of croplands to urban expansion. This was also the main cause of decreased FM S by 7% but increased FM D by 67%, which underscored the importance of conserving croplands in rapidly urbanizing regions of the TLB. Overall, land use composition had strong associations with WP D (r 2 ≥ 0.57) and FM S (r 2 ≥ 0.38) at the sub-basin scale, while the configuration of multiple land uses, such as urban sprawl, cropland fragmentation, and riparian buffers were crucial in influencing WP S. In comparison, FM D was most sensitive to urban expansion (r 2 = 0.74), economic development (r 2 = 0.81), and population growth (r 2 = 0.93). These findings provide new insights into sustainable land management for coordinating supply of and demand for hydrologic services in the TLB and other urbanizing watersheds of the world. • Supply and demand dynamics of hydrologic services are quantified in an urbanizing watershed. • Spatial mismatches between supply of and demand for hydrologic services are revealed. • Urbanization and land use have strong effects on hydrologic services at the sub-basin scale. • Land management measures are proposed to coordinate hydrologic service supply and demand.
