17. In Silico Screening of Bioactive Peptides in Stout Beer and Analysis of ACE Inhibitory Activity.

Abstract

Stout beer was selected as the research object to screen angiotensin-converting enzyme (ACE) inhibitory peptides. The peptide sequences of stout beer were identified using ultra-performance liquid chromatography-quadrupole-Orbitrap mass spectrometry with de novo, and 41 peptides were identified with high confidence. Peptide Ranker was used to score the biological activity and six peptides with a score ≥ 0.5 were screened to predict their potential ACE inhibitory (ACEI) activity. The toxicity, hydrophilicity, absorption, and excretion of these peptides were predicted. In addition, molecular docking between the peptides and ACE revealed a significant property of the peptide DLGGFFGFQR. Furthermore, molecular docking conformation and molecular dynamics simulation revealed that DLGGFFGFQR could be tightly bound to ACE through hydrogen bonding and hydrophobic interaction. Lastly, the ACEI activity of DLGGFFGFQR was confirmed using in vitro evaluation and the IC50 value was determined to be 24.45 μM. [ABSTRACT FROM AUTHOR]

21. New molecular insights into ferroptosis in lung adenocarcinoma progression and pharmacological compounds for targeted therapy.

Abstract

Background: The involvement of ferroptosis has been found in many pathological conditions of the lung. The genetic engineering of ferroptosis‐related genes may provide a potential target for the treatment of lung adenocarcinoma (LUAD). Methods: Nine ferroptosis regulators and markers were collected from FerrDb and their somatic mutations and expressions were analyzed based on The Cancer Genome Atlas (TCGA)‐LUAD cohort data. Least absolute shrinkage and selection operator (LASSO) and Cox regression analysis were performed to screen genes significantly associated with ferroptosis. The ferroptosis‐related gene signature was constructed using TCGA‐LUAD cohort data and was verified using the GSE cohort with pooled data for GSE30219, GSE31210, GSE37745 and GSE50081. Immune microenvironment component and mutation analysis were performed for genes in the ferroptosis‐related gene signature. Results: All nine ferroptosis regulators and markers were differentially expressed between normal LUAD tumor tissues and adjacent normal tissues and were related to copy number variation. The expression of 1329 genes were significantly associated with nine ferroptosis regulators and markers in the TCGA‐LUAD dataset, five (ALDOA, PLK1, CD47, CENPC and TMOD3) of which were integrated into a ferroptosis‐related gene signature to calculate the risk score of LUAD samples, showing a significant correlation with the abundance of immune cell infiltration and the immune score. Molecular docking showed the binding activity of natural active compound quercetin to target proteins ALDOA and CD47, as well as the binding activity of aristolochic acid to PLK1 protein and TMOD3 protein. Conclusions: In the present study, a ferroptosis‐related gene signature with predictive value for LUAD prognosis was constructed, in which the gene was a potential therapeutic target for LUAD. Quercetin and aristolochic acid were potential candidates for inhibiting these targets by directly binding to them and showing high affinity and strong stability. [ABSTRACT FROM AUTHOR]

Neodigitodesmium cheirosporum W. H. Tian & Maharachch. sp. nov. (FIG. 2) MycoBank: MB 844433 Etymology: The name refers to the cheiroid conidia Saprobic on submerged decayed wood in aquatic habitats. Sexual morph: Undetermined. Asexual morph: Hyphomycetous. Conidiomata sporodochial on natural substrate, superficial, compact, dark brown to black, with base attached on the surface of substrate. Conidiophores micronematous, reduced to conidiogenous cell. Conidiogenous cells 7–12 × 5–11 μm (x̅ = 9 × 7 μm, n = 15), holoblastic, at the base of the spore, hyaline. Conidia 35–70 × 9–24 μm (x̅ = 57 ×19 μm, n = 30), solitary, dictyosporous, conical shaped, cheiroid, not complanate, consisting of 1–4 rows with each row composed of 6–10 cells, with a hyaline basal connecting cell, not easy to separate, brown, subhyaline at the tip of peripheral rows. Culture characteristics: Conidia germinating on PDA within 12 h at 25 °C in the dark. Colonies on PDA circular, woolly at the margin, raised at the center, reverse yellow to brown center with a white margin. Material examined: CHINA, Sichuan Province, Chengdu, Baiyungou, on submerged decayed wood, 103°24′19″ E, 30°47′52″ N, 27 September 2021, W. H. Tian BY112_3 (HKAS 124014, holotype), ex-type culture CGMCC 3.23623 = UESTCC 22.0020. Notes: Combined sequences of LSU, ITS, SSU and tef1 confirmed the isolate of Neodigitodesmium formed a distinct clade in Dictyosporiaceae. However, it could not assign to any known genera (FIG. 1). In the combined gene tree, Neodigitodesmium is phylogenetically related to the Vikalpa australiensis (HKUCC 8797), Aquadictyospora lignicola (MFLUCC 17-1318) and Aquadictyospora clematidis (MFLUCC 17-2080) (FIG. 1), but they are not clustered on one branch and have obvious morphological differences. Neodigitodesmium is morphologically similar to Digitodesmium, both having sporodochial conidiomata and acrogenous, cheiroid, digitate conidia (Kirk 1981; Nóbrega et al. 2021). However, Neodigitodesmium differs from Digitodesmium and other Dictyosporiaceae members in conicalshaped conidia, composed of 1–4 compactly arranged rows of dark brown cells, with a hyaline basal connecting conidiogenous cells (FIG. 2). Digitodesmium tectonae (Rajeshkumar, R. K. Verma, Boonmee, K. D. Hyde, Chandrasiri & Wijayaw.) W. H. Tian & Maharachch, comb. nov. MycoBank: MB 844526 Basionym: Paradictyocheirospora tectonae Rajeshkumar, R. K. Verma, Boonmee, K. D. Hyde, Chandrasiri & Wijayaw, Phytotaxa 509 (3): 259–271 (2021). Notes: The genus Paradictyocheirospora was introduced by Rajeshkumar et al. (2021), with P. tectonae as the type species. The phylogenetic analysis based on a combined dataset of LSU, ITS, SSU and tef1 sequences show that P. tectonae is clustered within Digitodesmium (FIG. 1). In addition, Paradictyocheirospora possesses key characteristics of Digitodesmium such as punctiform, sporodochial conidiomata, acrogenous, cheiroid, digitate conidia, with an apical gelatinous cap (Kirk 1981; Rajeshkumar et al. 2021; Nóbrega et al. 2021). Thus, Paradictyocheirospora and Digitodesmium are congeneric. 30. Construction of N-doped mesoporous carbon viamicelle-induced chitosan for enhancing capacitive storage

Abstract

Poor rate performance of traditional porous carbon originating from slow ion transport in deep and tortuous pore structures seriously limits its practical application in the field of capacitive storage. In this regard, a novel N-doped mesoporous carbon with excellent rate performance is fabricated through micellar induced chitosan self-assembly, followed by carbonization and activation. The resultant N-doped porous carbon exhibits high specific surface area of 1130 m2 g−1and interconnected mesoporous structure, which offers short electron and ion transfer pathways improved wettability, and meanwhile provides sufficient interfacial active sites, and enhanced capacity of ion adsorption. These unique advantages endow the prepared porous carbon with a superior capacitance of 287 F g−1at 1 A g−1as well as an outstanding rate performance of 189 F g−1at 10 A g−1under 10 mg cm−2in aqueous supercapacitor. Impressively, the assembled Zn-ion hybrid capacitor device achieves an energy density of 121.95 Wh kg−1at 900 W kg−1and maintains 65.25 Wh kg−1at 18000 W kg−1. Meanwhile, this device also achieves excellent cycle stability. This research provides a cost-effective strategy for the construction of carbon-based materials in high-rate energy storage.

31. Facile synthesis of dense porous carbon derived from Linum usitatissimumL. root for high mass loading supercapacitors

Abstract

Achieving carbon electrodes with high gravimetric/volumetric/areal capacitances and rate performance, especially at high mass loadings, is crucial for practical applications in supercapacitors. In this regard, a novel dense porous carbon derived from Linum usitatissimumL. root was prepared through a facile in-situ activation method. The prepared porous carbon displays unique pore structure, i.e. abundant sub-nanopores (0.64–0.91 nm) and large size micropores (1.07–1.48 nm), which endow it with large surface area, good electric conductivity, and high packing density. Besides, the prepared porous carbon exhibits abundant active sites on the surface, which promote efficient surface utilization and ion transport. As a result, the resultant porous carbon presents high gravimetric/volumetric/areal capacitances of 421 F g−1/316 F cm−3/8.0 F cm−2at 1 A g−1. Importantly, the obtained symmetric supercapacitor achieves the high energy densities of 8.16 Wh kg−1/6.12 Wh L−1/163.19 μWh cm−2, even at the high mass loading (20 mg cm−2). This work provides a novel insight for designing and fabricating advanced electrode materials for energy storage application.

32. Multi-frequency power ultrasound (MFPU) pretreatment of crayfish (Procambarus clarkii): Effect on the enzymatic hydrolysis process and subsequent Maillard reaction.

Abstract

Crayfish, as a new consumer trend, has become one of the most popular freshwater aquatic foods in China, and its processed product output has been increasing year by year. In this study, the effect of multi-frequency power ultrasound (MFPU) pretreatment in various working modes including mono-, dual- and tri-frequency on the enzymatic hydrolysis and Maillard reaction of crayfish was investigated. Additionally, the underlying mechanism was also explored. Results showed that the degree of hydrolysis (DH) of crayfish was 23.89 %, while it increased to 40.78 % after MFPU pretreatment at 20/40 kHz dual-frequency, indicating that MFPU pretreatment significantly (p < 0.05) enhanced the enzymatic hydrolysis of crayfish. The crayfish protein after MFPU pretreatment exhibited a decrease in α-helix and increase in β-fold content, and the occurrence in the unfolding of the protein structure and alteration of tertiary structure. According to scanning electron microscopy (SEM) and atomic force microscopy (AFM) analyses, the MFPU pretreatment resulted in the fragmentation of crayfish protein. During the analyses of subsequent Maillard reaction, the composition and quantity of volatiles detected by the electronic nose and GC-MS indicated that MFPU pretreatment enhanced flavor of the Maillard reaction products.
