The Permian Basin is one of the most prolific, and currently one of the most active, oil and gas basins in the USA. The Lower Permian strata in the Permian Basin have produced more than 14 billion barrels of oil (BBO), making it the largest volume of hydrocarbon in the basin. Sedimentation in the Midland Basin during late Leonardian through early Guadalupian (ca. 272e269 Ma) resulted in progradation of shelf edge and ultimately closure of the basin by Middle Permian time. We analyzed a merged seismic survey covering parts of the Permian Basin (i.e., Central Basin Platform and Midland Basin) in Andrews, Ector, and Midland Counties, Texas. The seismic survey and well logs show the presence of gently dipping (ca. 1°) clinoforms in the Upper San Andres and Grayburg Formations on the eastern edge of the Central Basin Platform and western Midland Basin. The seismic attributes, curvature, and spectral decomposition identify low sinuosity slope channels oriented north-south, but such channels do not appear beyond the slope. The shelf edge shifts from north to south during deposition of the Upper San Andres and Grayburg Formations. We identify five basinward shifts noted by the migration of the shelf edge toward the basin center and the presence of channel features along the depositional slope. The petrophysical analysis indicates that channels cut into carbonate rocks and are filled by siliciclastic sediments; this interpretation is supported by the most negative curvature anomalies along the channel axes caused by the differential compaction between the carbonate and siliciclastic rocks. There are a few channels with a northwest-southeast strike, which matches the direction of the Concho Lineament observed by satellite data. The tannery is one of the leading revenue-generating sectors in developing countries. The ever-increasing demand for leather products in the global market requires converting large amounts of rawhide/skins into resilient non-putrescible finished leather. Only 20% of the raw material is converted into a finished product; the rest 80% is discarded as solid and liquid wastes during leather processing. A heavy discharge of improperly treated solid tannery waste (STW) causes a severe impact on the surrounding environment by polluting soil, surface water, and groundwater resources, posing severe hazards to human and animal health. STW comprises proteinaceous untanned and tanned waste, which requires proper treatment for eco-friendly disposal. Several strategies have been developed over the years for the reduction and recycling of STW for producing renewable energy (biogas and biohydrogen), biofuels (biodiesel and briquettes), construction material, fertilizers, commercial products (adsorbents, animal feeds, proteins, fats, and enzymes), and biodegradable packaging and non-packaging materials. In this review, we discuss various strategies adopted for recycling, reutilization, and reduction of STW in an environment-friendly manner. Furthermore, an overview of the current perspectives toward achieving a zero-waste policy is also presented to reduce the environmental burden using green-clean technology to aid the survival of present-day tanneries. Adeno-associated virus (AAV)-based gene therapies are emerging strategies in Duchenne muscular dystrophy (DMD) treatment. Exposure to wild-type AAV can lead to development of neutralizing antibodies (NAbs) and blocking of AAV transduction, thereby limiting the delivery of AAV vector-based gene therapy. Therefore, it is imperative to check for the presence of AAV NAbs in a patient who is a candidate for gene therapy. We prospectively enrolled 101 genetically confirmed males with DMD (median age 11 years, 48% ambulatory and 59% on steroids) and performed AAV neutralization assays against AAV2, AAV8, AAV9, and AAVrh74 serotypes. The serotype analysis showed that AAV9 (36%) and AAVrh74 (32%) seroprevalence was lower compared with AAV2 (56%) and AAV8 (47%). Interestingly, age was not correlated with NAb titer for any of the capsids. NAb responses were observed at a higher frequency in African American participants and at a lower frequency in Caucasian participants for all four serotypes. Further analysis showed no significant differences in NAb titers regardless of serotype and whether participants were taking steroids or not. Finally, we observed higher AAV8, AAV9, and AAVrh74 seroprevalence and significantly higher AAV2 and AAV8 NAb titers in participants who were ambulatory compared with nonambulatory participants. Overall, these data identify AAV9 and AAVrh74 as the two serotypes with lower pre-existing NAb titers in this study's cohort of 101 males with DMD, possibly showing their utility in future gene therapy applications in treatment of this cohort of patients with DMD. Active macro seepages of methane that occur in between the north bank of the Brahmaputra river and Himalayan foothill region of Assam Arakan Basin, India, indicate the presence of hydrocarbon accumulation in the subsurface, but the hydrocarbon prospects in this region are not well studied. We carried out an extensive field sampling, which included a total of 58 sediment core collections from an active gas seepage location and nearby areas at a depth of 2–2.5 m. Our sample locations are placed at 1 km intervals laterally. We performed laboratory investigations and mapped near-surface chemical alterations associated with active macro seepages and microseepages. The analysis of geochemical composition of hydrocarbon gases in the sediment indicates both the biogenic and thermogenic origins of seeped hydrocarbons. The stable isotope analysis of methane suggests the presence of thermogenic as well as mixed biogenic-thermogenic gases. The presence of such mixing of gases is caused by the secondary alteration processes during their migration through potential faults and fractures. The trace elements of the sediments show anomalous concentrations at different parts of the study area, with a wide range of concentrations for Ba (54 to 492 ppm), Cu (1–25 ppm), Cr (61–329 ppm), Ni (1–42 ppm), Pb (2–48 ppm), Th (2–32 ppm), U (4–39 ppm), V (19–133 ppm) and U (0.87–6.5 ppm). There are higher concentrations of adsorbed gases, trace elements, and microbes along the identified lineaments. Such higher concentration can be triggered by high hydrocarbon-oxidizing bacteria count, which is greater than 104 cfu/gm of soil of bacterial growth around the lineaments. We identified potential hydrocarbon prospects based on the macro and micro seepage analysis using integrated geological, geochemical and microbial techniques in the study area. Metagenomics facilitates the isolation of novel enzymes from the unculturable microorganisms inhabiting a particular ecosystem. In the present study, solid tannery waste (STW) metagenome was explored as a potential source of microbial serine proteases. KEGG annotated functional profile of the UNK metagenome was exploited for in silico mining of candidate serine proteases. A novel serine protease was identified, namely Prt1SU, corresponding to an 1149 bp full-length gene encoding a 383 amino acid protein. The identity of the Prt1SU was evaluated by PSI-BLAST against the databases at NCBI and BLAST against the MEROPS database. Identities ranged from 30–45.7% to their respective nearest homologs of the serine protease family. Utilizing the DNA template isolated from the UNK metagenome, the candidate serine protease gene was cloned in the pET28a(+) vector and expressed in E. coli BL21 (DE3) strain. Although the Prt1SU was overexpressed at 18ºC using 0.1 mM IPTG, it remained inactive because of the formation of inclusion body. After solubilization with 8 M urea, the Prt1SU enzyme was purified to homogeneity using Ni-NTA chromatography under denaturing conditions and then renatured to retain protease activity. The purified Prt1SU with a molecular weight of ~ 40 kDa exhibited the maximum enzyme activity at pH 9.0 and temperature 37ºC. The enzyme activity of the Prt1SU was enhanced by the addition of organic solvents, and the enzyme was found stable in the presence of anionic and non-ionic surfactants and oxidizing agents, suggesting that Prt1SU is suitable for various industrial applications as well as wastewater and STW treatment. Present study aimed to evaluate the changes in proximate composition and physical attributes in brown shrimp (Metapenaeus dobsonii) exposed to different methods of cooking followed by freezing. For this, three different grades (100/200, 200/300, and 300/500 numbers per kg) of brown shrimp were cooked at 90°C till the core temperature of the product reaches 85°C using hot water, steam, and microwave (400W) techniques. The changes in yield, cooking loss, proximate composition, textural, and colour profile were assessed for cooked shrimps. The cooking loss was higher for larger grades of shrimp, whereas shrimp cooked using hot water exhibited the highest cooking loss. Lowest cooking loss was observed for microwave-cooked shrimp. Moisture content decreased after cooking whereas protein, fat, ash, and calorie content increased. After cooking, different grades of shrimp showed an increase in their lightness (L*), redness (a*), and yellowness (b*) values. The smaller grade shrimp exhibited lower value for cohesiveness, hardness, chewiness, and gumminess. Different cooking techniques yielded cooked shrimp of varying hardness values. Several studies have been carried out to investigate the magnitude of forces induced by tornado-like winds on buildings from experimental as well as computational side. The number of experimental studies evaluating the wind loads due to tornado-like vortices are comparatively greater in number than studies based on computational fluid dynamics (CFD). Furthermore, the limited number of CFD studies were often not validated by comparing with experimental measurements. In this work, an attempt is made to validate a CFD model by comparing the pressures induced by tornado-like vortices on a building model with experimental measurements from the tornado simulator at Texas Tech University (TTU). Results of the comparison indicate that the pressure coefficients obtained from the CFD model agree well with TTU experimental datasets. Besides, the effect of building size as well as the effect of flow structure of vortex on pressure coefficients on the building induced by tornado-like vortex is also investigated. It is observed that the pressures on the building can differ by up to 100% when different sizes of building is considered and the vortex with a single celled structure creates the most unfavorable loading conditions on the building model. • A state of the art review on pressures induced by tornado-like vortex on building from existing literature is presented. • Tornado-induced pressures on building computed from CFD model are validated by comparing with TTU experimental datasets. • Influence of size of the building model on the pressures induced by tornado-like vortex is presented. • The influence of different flow structures of vortex on induced pressures on the building is presented. Introduction: Patients with connective tissue disorders (CTD) and pulmonary arterial hypertension (PAH) have a poorer prognosis than those with other PAH etiologies. This study assessed the impact of CTD on healthcare outcomes among PAH patients with and without CTD comorbidities that were treated with oral selexipag. Methods: The study utilized Optum's de-identified Clinformatics® Data Mart Database (2007–2021) from January 1, 2014 to June 30, 2019, and identified patients with PAH without CTD and PAH with CTD treated with oral selexipag. Patients had ≥ 12-month baseline period with no requirement for a minimum follow-up period. Patients were followed until any of the following events: discontinuation of oral selexipag, or health plan disenrollment, or death, or presence of a diagnosis claim for CTEPH, or study end date, whichever occurred first. PAH-related hospitalizations, PAH disease progression, and healthcare utilizations and costs were assessed in the follow-up period. The Cox proportional hazards model was used to evaluate the time to hospitalization and generalized linear models were used to examine healthcare costs and utilization between the two cohorts. Results: In the analysis, 237 PAH without CTD, and 80 PAH patients with CTD comorbidities prescribed oral selexipag were included. The PAH without CTD comorbidities cohort was older (65 vs. 63 years old), had proportionately less females (72 vs. 83%), and higher comorbidity burden than PAH with CTD comorbidities (mean CCI index 3 vs. 2). After adjusting for potential confounders, the risk for PAH-related hospitalization (hazard ratio (HR) 1.13, p value 0.641), all-cause hospitalization (HR 1.09, p value: 0.765), and PAH disease progression (HR 1.14, p value 0.522) between the two cohorts were similar. After adjusting for baseline demographic and clinical characteristics, PAH with CTD comorbidities incurred higher total mean all-cause PAH-related medical care costs compared to PAH without CTD comorbidities. Conclusions: In this real-world study, the risk of hospitalization and PAH disease progression were similar between the two cohorts who received oral selexipag. The results from this study corroborate findings of the GRIPHON post hoc analysis of PAH-associated CTD patients and support oral selexipag use in PAH-CTD patients. [ABSTRACT FROM AUTHOR]

Phenols are ubiquitous pollutants, mainly from industrial effluent, causing pollution of natural water resources. The research focused on screening efficient phenol-degrading bacteria and kinetic modelling of phenol biodegradation and growth. Membrane filtration was used for the isolation of bacteria from the wastewater sample. The screening of phenol-degrading bacteria was based on the efficiency of phenol utilization. The strain with efficient phenol degradation capacity was characterized by 16S rDNA sequencing and designated Acinetobacter baumannii W29. Biomass growth and phenol utilization rate of the strain were evaluated at different initial phenol concentrations (100-800 mgL-1). Specific growth rate data were fitted to five models, i.e. Monod, Haldane, Aiba, Teisser, and Webb model. The yield coefficient at different initial phenol concentrations was calculated from the slope of the specific growth rate (μ) versus the specific phenol utilization rate (q). The strain showed complete phenol degradation potential up to 1000 mgL-1. The maximal growth rate was achieved at 400 mgL-1, which coincided with the maximum substrate utilization rate at the same concentration. The specific growth rate showed the best fit with the Haldane model. The strain had a yield coefficient of 0.70 (mg cell mg-1 phenol). The value of µ and Ks revealed the affinity of the strain for high-concentration phenol and the its ability to withstand high phenol concentrations. The kinetic growth behaviour of the strain fitted well with the Haldane model. The findings of the study could be applied to wastewater treatment with a high phenol load. [ABSTRACT FROM AUTHOR]

The restructuring of power systems and the ever-increasing demand for electricity have given rise to congestion in power networks. The use of distributed generators (DGs) may play a significant role in tackling such issues. DGs may be integrated with electrical power networks to regulate the drift of power in the transmission lines, thereby increasing the power transfer capabilities of lines and improving the overall performance of electrical networks. In this article, an effective method based on the Harris hawks optimization (HHO) algorithm is used to select the optimum capacity, number, and site of solar-based DGs to reduce real power losses and voltage deviation. The proposed HHO has been tested with a complex benchmark function then applied to the IEEE 33 and IEEE 69 bus radial distribution systems. The single and multiple solar-based DGs are optimized for the optimum size and site with a unity power factor. It is observed that the overall performance of the systems is enhanced when additional DGs are installed. Moreover, considering the stochastic and sporadic nature of solar irradiance, the practical size of DG has been suggested based on analysis that may be adopted while designing the actual photovoltaic (PV) plant for usage. The obtained simulation outcomes are compared with the latest state-of-the-art literature and suggest that the proposed HHO is capable of processing complex high dimensional benchmark functions and has capability to handle problems pertaining to electrical distribution in an effective manner. Cytoplasmic and nuclear iron-sulfur (Fe-S) enzymes that are essential for genome maintenance and replication depend on the cytoplasmic Fe-S assembly (CIA) machinery for cluster acquisition. The core of the CIA machinery consists of a complex of CIAO1, MMS19 and FAM96B. The physiological consequences of loss of function in the components of the CIA pathway have thus far remained uncharacterized. Our study revealed that patients with biallelic loss of function in CIAO1 developed proximal and axial muscle weakness, fluctuating creatine kinase elevation, and respiratory insufficiency. In addition, they presented with CNS symptoms including learning difficulties and neurobehavioral comorbidities, along with iron deposition in deep brain nuclei, mild normocytic to macrocytic anemia, and gastrointestinal symptoms. Mutational analysis revealed reduced stability of the variants compared with WT CIAO1. Functional assays demonstrated failure of the variants identified in patients to recruit Fe-S recipient proteins, resulting in compromised activities of DNA helicases, polymerases, and repair enzymes that rely on the CIA complex to acquire their Fe-S cofactors. Lentivirus-mediated restoration of CIAO1 expression reversed all patient-derived cellular abnormalities. Our study identifies CIAO1 as a human disease gene and provides insights into the broader implications of the cytosolic Fe-S assembly pathway in human health and disease. [ABSTRACT FROM AUTHOR]

Introduction Methods Results Conclusions Left bundle branch area (LBBA) pacing (LBBAP) has been proposed as an alternative therapy option in patients indicated for cardiac pacing to treat bradycardia or heart failure. The aim of the study was to evaluate the safety and effectiveness of LBBAP in patients implanted with a Tendril 2088 stylet‐driven lead.The international retrospective data collection registry included 11 sites from 5 countries globally. Patients with attempted implants of the Tendril lead in the LBBA were followed for at least 6 months post the implant attempt. The primary safety and efficacy endpoints were freedom from LBBAP lead‐related serious adverse events and the composite of LBBA capture threshold of ≤2.0 V and R‐wave amplitudes ≥5 mV (or ≥value at implant), respectively.Of 221 patients with attempted implants of the Tendril 2088 lead in the LBBA, 91.4% (202/221) had successful implants for LBBAP. Regardless of the LBBAP implant success, all patients were followed for at least 6 months (8.7 ± 7.3 months). Baseline characteristics: 44% female, 84% ≥65 years old, 34% coronary artery disease, and 86% of primary indications for pacemaker implant. Both primary safety and effectiveness endpoints were met (freedom from LBBAP lead‐related serious adverse device effects of 99.5% and electrical performance composite success rate of 93%). The capture thresholds in LBBAP at implant and 6 months were 0.8 ± 0.3 V@0.4 ± 0.1 ms and 0.8 ± 0.3 V@0.4 ± 0.1 ms. The rate of patients with capture threshold rise ≥1 V was 1.5% through 6 months. The R‐wave amplitudes in LBBAP at implant and 6 months were 9.3 ± 3.2 mV and 10.6 ± 3.0 mV.This large multicenter study demonstrates that the stylet‐driven Tendril™ STS 2088 lead is safe and effective for LBBAP with high success and low complication rates. Introduction/Aims: Objective outcome measures in children undergoing treatment for chronic inflammatory demyelinating polyradiculoneuropathy (CIDP) are lacking. The aim of the study was to record serial grip strength and motor nerve conduction studies to assess interval change. Methods: This was a retrospective review of 16 children (8 females and 8 males; median age, 9.7 years; interquartile range, 6–13 years) with CIDP followed at a tertiary children's hospital from 2013 to 2021. Subjects were treated with intravenous immunoglobulin (IVIG). Right and left grip strength measurements were obtained at each clinic visit using a handheld dynamometer. Annual right median motor nerve conduction study data were recorded during the study period. Results: Mean duration of follow‐up was 2.9 years. Grip strength (right: 0.19 kg/month, p < 0.001; left 0.23 kg/month, p < 0.001) and median F‐wave latencies (−0.23/month, p = 0.015) showed significant improvement over time. Akaike information criterion showed time + IVIG frequency <21 days as best fit for grip strength and distal compound muscle action potential amplitude. Discussion: Our study results indicate serial grip strength measurements are a feasible and objective way to assess motor strength improvement in children with CIDP receiving immunotherapy. Electrocatalytic carbon monoxide reduction has been previously reported to yield a range of carbonaceous products including alcohols, hydrocarbons and carboxylic acids. However, esters, an important family of organic compounds, have not been formed. Herein, we report the electrosynthesis of C3 to C6 acetate esters (H3C−(C=O)−O−R) from carbon monoxide using copper catalysts in a membrane electrode assembly cell. Ethyl acetate and propyl acetate could be produced with an unprecedented total Faradaic efficiency (FE) of ∼22 % and with a current density of up to −55 mA cm−2, alongside minor quantities of methyl acetate and butyl acetate. The esters are produced via the addition reaction of ethenone (H2C=C=O) and alcohols produced during CO reduction. The Pennsylvanian unconformity, which is a detrital surface, separates the beds of the Permian-aged strata from the Lower Paleozoic in the Central Basin Platform. Seismic data interpretation indicates that the unconformity is an angular unconformity, overlying multiple normal faults, and accompanied with a thrust fault which maximizes the region's structural complexity. Additionally, the Pennsylvanian angular unconformity creates pinch-outs between the beds above and below. We computed the spectral decomposition and reflector convergence attributes and analyzed them to characterize the angular unconformity and faults. The spectral decomposition attribute divides the broadband seismic data into different spectral bands to resolve thin beds and show thickness variations. In contrast, the reflector convergence attribute highlights the location and direction of the pinch-outs as they dip south at angles between 2° and 6°. After reviewing findings from RGB blending of the spectrally decomposed frequencies along the Pennsylvanian unconformity, we observed channel-like features and multiple linear bands in addition to the faults and pinch-outs. It can be inferred that the identified linear bands could be the result of different lithologies associated with the tilting of the beds, and the faults may possibly influence hydrocarbon migration or act as a flow barrier to entrap hydrocarbon accumulation. The identification of this angular unconformity and the associated features in the study area are vital for the following reasons: 1) the unconformity surface represents a natural stratigraphic boundary; 2) the stratigraphic pinch-outs act as fluid flow connectivity boundaries; 3) the areal extent of compartmentalized reservoirs' boundaries created by the angular unconformity are better defined; and 4) fault displacements are better understood when planning well locations as faults can be flow barriers, or permeability conduits, depending on facies heterogeneity and/or seal effectiveness of a fault, which can affect hydrocarbon production. The methodology utilized in this study is a further step in the characterization of reservoirs and can be used to expand our knowledge and obtain more information about the Goldsmith Field. Cu catalysts are most apt for reducing CO(2) to multi‐carbon products in aqueous electrolytes. To enhance the product yield, we can increase the overpotential and the catalyst mass loading. However, these approaches can cause inadequate mass transport of CO(2) to the catalytic sites, which will then lead to H2 evolution dominating the product selectivity. Herein, we use a MgAl LDH nanosheet 'house‐of‐cards' scaffold to disperse CuO‐derived Cu (OD‐Cu). With this support‐catalyst design, at −0.7 VRHE, CO could be reduced to C2+ products with a current density (jC2+) of −1251 mA cm−2. This is 14× that of the jC2+ shown by unsupported OD‐Cu. The current densities of C2+ alcohols and C2H4 were also high at −369 and −816 mA cm−2 respectively. We propose that the porosity of the LDH nanosheet scaffold enhances CO diffusion through the Cu sites. Coupling renewable energy with the electrochemical conversion of CO2 to chemicals and fuels has been proposed as a strategy to achieve a new circular carbon economy and help mitigate the effects of anthropogenic CO2 emissions. Liquid‐like Nanoparticle Organic Hybrid Materials (NOHMs) are composed of polymers tethered to nanoparticles and are previously explored as CO2 capture materials and electrolyte additives. In this study, two types of aqueous NOHM‐based electrolytes are prepared to explore the effect of CO2 binding energy (i.e., chemisorption versus physisorption) on CO2 electroreduction over a silver nanoparticle catalyst for syngas production. Poly(ethylenimine) (PEI) and Jeffamine M2070 (HPE) are ionically tethered to SiO2 nanoparticles to form the amine‐containing NOHM‐I‐PEI and ether‐containing NOHM‐I‐HPE, respectively. At less negative cathode potentials, PEI and NOHM‐I‐PEI‐based electrolytes produce CO at higher rates than 0.1 molal. KHCO3 due to favorable catalyst‐electrolyte interactions. Progressive encephalopathy with brain atrophy and thin corpus callosum (PEBAT) is a severe and rare progressive neurodegenerative disease (OMIM 617913). This condition has been described in individuals with pathogenic variants affecting tubulin-specific chaperone protein D (TBCD), which is responsible for proper folding and assembly of tubulin subunits. Here we describe two unrelated infants from Central America presenting with worsening neuromuscular weakness, respiratory failure, polyneuropathy, and neuroimaging findings of severe cerebral volume loss with thin corpus callosum. These individuals harbored the same homozygous variant of uncertain significance in the TBCD gene on whole exome sequencing (WES). Predicted protein modeling of this variant confirmed disruption of the protein helix at the surface of TBCD. The goal of this report is to emphasize the importance of rapid WES, careful interpretation of uncertain variants, prognostication, and family counseling especially when faced with a neurodegenerative clinical course. Aims: The pathophysiological mechanism(s) underlying non-alcoholic fatty liver disease (NAFLD) have yet to be fully delineated and only a single drug, peroxisome proliferator-activated receptor (PPAR) α/γ agonist saroglitazar, has been approved. Here, we sought to investigate the role of Regulator of G Protein Signaling 7 (RGS7) in hyperlipidemia-dependent hepatic dysfunction. Results: RGS7 is elevated in the livers of NAFLD patients, particularly those with severe hepatic damage, pronounced insulin resistance, and high inflammation. In the liver, RGS7 forms a unique complex with transcription factor ATF3 and histone acetyltransferase Tip60, which is implicated in NAFLD. The removal of domains is necessary for ATF3/Tip60 binding compromises RGS7-dependent reactive oxygen species generation and cell death. Hepatic RGS7 knockdown (KD) prevented ATF3/Tip60 induction, and it provided protection against fibrotic remodeling and inflammation in high-fat diet-fed mice translating to improvements in liver function. Hyperlipidemia-dependent oxidative stress and metabolic dysfunction were largely reversed in RGS7 KD mice. Interestingly, saroglitazar failed to prevent RGS7/ATF3 upregulation but it did partially restore Tip60 levels. RGS7 drives the release of particularly tumor necrosis factor α (TNFα) from isolated hepatocytes, stellate cells and its depletion reverses steatosis, oxidative stress by direct TNFα exposure. Conversely, RGS7 overexpression in the liver is sufficient to trigger oxidative stress in hepatocytes that can be mitigated via TNFα inhibition. An extreme chronic wound tissue microenvironment causes epigenetic gene silencing. An unbiased whole-genome methylome was studied in the wound-edge tissue of patients with chronic wounds. A total of 4,689 differentially methylated regions (DMRs) were identified in chronic wound-edge skin compared with unwounded human skin. Hypermethylation was more frequently observed (3,661 DMRs) in the chronic wound-edge tissue compared with hypomethylation (1,028 DMRs). Twenty-six hypermethylated DMRs were involved in epithelial-mesenchymal transition (EMT). Bisulfite sequencing validated hypermethylation of a predicted specific upstream regulator TP53. RNA-Seq analysis was performed to qualify findings from methylome analysis. Analysis of the downregulated genes identified the TP53 signaling pathway as being significantly silenced. Direct comparison of hypermethylation and downregulated genes identified 4 genes, ADAM17, NOTCH, TWIST1, and SMURF1, that functionally represent the EMT pathway. Single-cell RNA-Seq studies revealed that these effects on gene expression were limited to the keratinocyte cell compartment. Experimental murine studies established that tissue ischemia potently induces wound-edge gene methylation and that 5'-azacytidine, inhibitor of methylation, improved wound closure. To specifically address the significance of TP53 methylation, keratinocyte-specific editing of TP53 methylation at the wound edge was achieved by a tissue nanotransfection-based CRISPR/dCas9 approach. Cervical cancer is the most prevalent cancer in females. Melatonin, a neurohormone has been documented as a promising therapeutic molecule for cervical cancer. However, the underlying molecular mechanism is not known. We explored the dose-dependent anti-tumor response of melatonin against cervical cancer cell lines, HeLa (HPV-18 positive) and SiHa (HPV-16 positive). The anti-cancer effect of melatonin was evaluated by MTT assay, cell imaging, colony formation, DAPI, AO/PI, LDH, Flow cytometry, scratch assay, western blot analysis and real-time PCR. Results of DAPI, AO/PI, LDH, and Annexin/PI staining revealed that melatonin induces apoptosis. The results of cell cycle analysis revealed that melatonin arrests the HeLa and SiHa cells in sub-G1 and G1 phases, respectively. Western blot analysis revealed that melatonin downregulated the expression of pro-inflammatory transcription factor, NF-κB and the expression of COX-2 protein, a key mediator in cell proliferation. In addition, melatonin downregulated the expression of an invasive marker, MMP-9, an antiapoptotic protein, Bcl-2, and upregulated the expression of pro-apoptotic protein, Bax at both transcriptional and translational levels. Overall, the results suggest that melatonin exhibited strong anti-cancer therapeutic potential against human cervical cancer cell line progression possibly through inhibition of NF-κB signalling pathway. Shark is a seafood commodity that is a good source of minerals and accumulates heavy metals and trace elements through biomagnification, which can pose health risk if taken above the permissible limit. A study was conducted on commonly landed eleven shark species (Scoliodon laticaudus , Rhizopriodon oligolinx , Sphyrna lewini (CR), Carcharhinus macloti , Carcharinus limbatus , Carcharhinus amblyrhynchoides , Carcharhinus sorrah, Carcharinus falciformes(VU), Glaucostegus granulatus, Chiloscyllium arabicum, Loxodon macrorhinus) and analyzed for their heavy metal content, Hazard Index, Total Hazard Quotient, Metal Pollution Index, and also calculated the health risk associated with the consumption. Most of the heavy metals and trace minerals were found to be within the acceptable limit. The Targeted Hazard Quotient (THQ) and the Hazard Index (HI) of all the species except two were less than 1 (HI ≤ 1.0). The Metal Pollution Index (MPI) is showing either no impact or very low contamination. An overall study on hazard identification and health risk characterization in terms of heavy metals shows contamination of some heavy metals in sharks, but there is no potential human health risk associated with consumption. • Total eleven shark species are evaluated for non-carcinogenic health risk due to selected heavy metals. • THQ, HI and Metal Pollution Index are within the safer limits. • Heavy metal content in the commonly landed shark species, along the Saurashtra coast of India is safe to consume. Therapeutic vascular endothelial growth factor (VEGF) replenishment has met with limited success for the management of critical limb-threatening ischemia. To improve outcomes of VEGF therapy, we applied single-cell RNA sequencing (scRNA-seq) technology to study the endothelial cells of the human diabetic skin. Single-cell suspensions were generated from the human skin followed by cDNA preparation using the Chromium Next GEM Single-cell 3' Kit v3.1. Using appropriate quality control measures, 36,487 cells were chosen for downstream analysis. scRNA-seq studies identified that although VEGF signaling was not significantly altered in diabetic versus nondiabetic skin, phospholipase Cγ2 (PLCγ2) was downregulated. The significance of PLCγ2 in VEGF-mediated increase in endothelial cell metabolism and function was assessed in cultured human microvascular endothelial cells. In these cells, VEGF enhanced mitochondrial function, as indicated by elevation in oxygen consumption rate and extracellular acidification rate. The VEGF-dependent increase in cell metabolism was blunted in response to PLCγ2 inhibition. Follow-up rescue studies therefore focused on understanding the significance of VEGF therapy in presence or absence of endothelial PLCγ2 in type 1 (streptozotocin-injected) and type 2 (db/db) diabetic ischemic tissue. Nonviral topical tissue nanotransfection technology (TNT) delivery of CDH5 promoter-driven PLCγ2 open reading frame promoted the rescue of hindlimb ischemia in diabetic mice. Improvement of blood flow was also associated with higher abundance of VWF+/CD31+ and VWF+/SMA+ immunohistochemical staining. TNT-based gene delivery was not associated with tissue edema, a commonly noted complication associated with proangiogenic gene therapies. Background: Hereditary spastic paraplegia (HSP) encompasses several rare genetic disorders characterized by progressive lower extremity spasticity and weakness caused by corticospinal tract degeneration. Published literature on genetically confirmed pediatric HSP cases is limited.Methods: We conducted a retrospective review of childhood-onset HSP cases followed in the neuromuscular clinics at Children's and Emory Healthcare in Atlanta. Clinical presentation, family history, examination, electrodiagnostic data, neuroimaging, genetic test results, comorbidities, and treatment were recorded.Results: Sixteen patients with HSP (eight males, eight females) with a mean age 19 years ± 15.7 years were included. Ten patients (66%) presented with gait difficulty. Seven (44%) were ambulatory at the last clinic follow-up visit with an average disease duration of 7.4 years. Genetically confirmed etiologies included SPAST (3 patients), MARS (2), KIF1A (2), KIF5A (1), SACS (1), SPG7 (1), REEP1 (1), PNPT1 (1), MT-ATP6 (1), and ATL1 (1). Symptom onset to genetic confirmation on an average was 8.2 years. Sensory motor axonal polyneuropathy was found in seven patients, and two exhibited cerebellar atrophy on magnetic resonance imaging (MRI) of the brain. Neurological comorbidities included developmental delay (n = 9), autism (n = 5), epilepsy (n = 3), and attention-deficit/hyperactivity disorder (n = 2).Conclusions: In our study, a significant proportion (70%) of subjects with childhood-onset HSP had comorbid neurocognitive deficits, polyneuropathy with or without neuroimaging abnormalities, and rare genetic etiology. Genetic diagnosis was established either through inherited genetic neuropathy panel or whole-exome sequencing, which supports the utility of whole-exome sequencing in aiding in HSP diagnosis. Aims: This study was performed to assess the expression pattern of TCF4 in GBC tissue and attempted to correlate its expression with different clinicopathological parameters. Materials and Methods: The study was conducted on 33 surgically resected specimens of gallbladder carcinoma (GBC) and 12 cases of chronic cholecystitis (CC) as control, which had been confirmed from histology. The expression of TCF4 was performed by the reverse transcription polymerase chain reaction and immunohistochemistry. Results: Relative mRNA expression levels of β-catenin and TCF4 in GBC tissues were significantly (P < 0.05) higher than in CC samples. TCF4 protein expression was observed in 81.82% (27/33) GBC cases. Specifically, among GBC samples, 21.21% (7/33) was graded as strongly positive, 60.61% (20/33) graded as moderately positive, whereas 18.18% (6/33) graded as negative. All 12 CC samples graded as negative. Overall, TCF4 expression in GBC tissues was statistically significant over CC samples (P < 0.05). Moreover, we observed that TCF4 expression was significantly higher (P < 0.05) in high tumor grades than low grade, higher (P < 0.05) in Stage 2 and Stage 3 than Stage 1. Conclusion: The present study suggests that TCF4 may exert an oncogenic role in the progression of GBC and may serve as a new potential candidate biomarker for tumor progression, and it might be a potential therapeutic target against GBC. This analysis examined trends in incidence and survival among US adults with myeloproliferative neoplasms, including essential thrombocythemia (ET; n = 14,676), polycythemia vera (PV; n = 15,141), and primary myelofibrosis (PMF; n = 4214), using Surveillance, Epidemiology, and End User Results (SEER) data (SEER 18; 2002–2016). Incidence and survival rates over the study period and by diagnosis year (per 5-year time frames: 2002–2006; 2007–2011; 2012–2016) were assessed. The overall incidence rates (95% CI) were 1.55 (1.52–1.57) for ET, 1.57 (1.55–1.60) for PV, and 0.44 (0.43–0.45) per 100,000 person-years for PMF, with rising ET incidence. Five-year mortality over the study period was 19.2%, 19.0%, and 51.0% for ET, PV, and PMF, respectively. Improved survival over time was observed for PV and PMF, but not for ET. Seismic coherence is of the essence for seismic interpretation as it highlights seismic discontinuity features caused by the deposition process, reservoir boundaries, tectonic movements, etc. Since its appearance in 1995, seismic coherence has become one of the most popular and highly recognized interpretation tools. In the last 25 years, there have been different kinds of approaches to calculate seismic coherence attributes, such as cross-correlation, semblance, and eigen-structure. We have also seen different coherence enhancement techniques and modified ways for coherence calculation to address different problems in diverse field applications. In this survey, we provide a general overview of seismic coherence, which is commonly used to delineate structural and stratigraphic discontinuities. We cover the development of the seismic coherence attributes via introducing existing approaches for coherence calculation, enhancement, spectral band-limited coherence methods, and offset-/azimuth-limited coherence methods. In addition, we also discuss the possible coherence artifacts and pitfalls. To better compare different techniques, field applications are provided, where different disciplinary coherence attributes with enhancement techniques are calculated for the channel, fault, carbonate karst, and volcano interpretation. Objective: There is limited and mixed research describing the memory performance of boys with Duchenne muscular dystrophy (DMD), a progressive disorder that affects the muscle and the brain, presumably due to the absence of dystrophin; however, the literature indicates either the existence of a selective deficit in verbal working memory, or more generalized impairment in both verbal and visual memory. Far less is documented about the neurocognitive profile of boys with Becker muscular dystrophy (BMD), a closely related neuromuscular disorder which allows for at least some functional dystrophin protein to circulate. The Child and Adolescent Memory Profile (ChAMP) is a valid and widely used memory battery that has not been studied in either DMD or BMD. This study aimed to assess the verbal and visual memory performance in boys having either a DMD or a BMD diagnosis using the ChAMP. A working memory measure, the Digit Span subtest from the Wechsler Intelligence Scale for Children-Fifth Edition, was also included for comparison. Participants and Methods: Twenty-one patients (Age M = 12.19 ± 3.60; 100% male; 76% DMD, 24% Becker) were selected from retrospective data collection of neuropsychological performance in children with neuromuscular disorders. Patients were recruited and assessed as part of a larger scale IRB-approved research study designed to better understand the neurocognitive and behavioral trajectories of boys with DMD or BMD with a complete neuropsychological battery. Results: Independent samples f-tests revealed no significant differences between groups across verbal (DMD M = 88.71; BMD M = 100.80; p =.08), visual (DMD M =90.36; BMD M 93.60; p =.33), and working memory (DMD M = 84.69; BMD: M 82.60; p =.40) domains. In additional analyses, a one sample f-test comparing verbal and visual memory within DMD children revealed significantly worse verbal than visual memory scores (verbal memory M = 88.71; visual memory M = 90.36; p = <.001). Conclusions: There were no significant differences between groups in verbal, visual, and working memory performance, though sample size was a significant limitation. However, based on a comparison of means, children with BMD appear to have stronger verbal memory skills than children with DMD. Furthermore, significant differences between verbal and visual memory within DMD children were observed, such that verbal memory skills were weaker. Gallbladder cancer (GBC) is an aggressive malignancy of gastrointestinal tract. Due to uncontrolled growth, GBC cells rapidly synthesize biomolecules including lipids. The lipids are integral component of cell membrane with a wide range of cellular functions. In this study, we measured the clinicopathological features in 40 cases of histologically confirmed GBC and 16 cases of chronic cholecystitis (CC). The female to male ratio in the GBC and CC groups were 3.44:1 and 2.2:1, respectively. The GBC patients exhibited well to poorly differentiated tumor. In the CC group, all patients showed cholecystitis with no evidence of dysplasia or malignancy. The majority of GBC and CC patients reported pain. Using 1H NMR spectroscopy, we observed 4-folds increase in the level of choline containing phospholipids (CCPLs) in the gallbladder of GBC patients as compared to CC patients. Other lipid metabolites such as cholesterol ester, C18-cholesterol and saturated fatty acids were insignificantly changed between GBC and CC patients. Moreover, the level of CCPLs in the GBC patients with BMI <25 kg/m2 was significantly higher as compared to CC patients. Further, a significant increase in the CCPLs level was observed in GBC female patients in comparison to CC patients. From the computational analyses, we observed that the genes involved in the biosynthesis of phosphatidylcholine (PtdCho) indirectly interact with the RELA, which encodes the NF-κB p65 subunit. The genes involved in the PtdCho biosynthesis were also correlated with the overall and disease-free survival of cholangiocarcinoma patients. Load flow analysis is an essential tool for the reliable planning and operation of interconnected power systems. The constant increase in power demand, apart from the increased intermittency in power generation due to renewable energy sources without proportionate augmentation in transmission system infrastructure, has driven the power systems to function nearer to their limits. Though the power flow (PF) solution may exist in such circumstances, the traditional Newton–Raphson based PF techniques may fail due to computational difficulties owing to the singularity of the Jacobian Matrix during critical conditions and faces difficulties in solving ill-conditioned systems. To address these problems and to assess the impact of large-scale photovoltaic generator (PVG) integration in power systems on power flow studies, a derivative-free quasi-oppositional heap-based optimization (HBO) (QOHBO) technique is proposed in the present paper. In the proposed approach, the concept of quasi-oppositional learning is applied to HBO to enhance the convergence speed. The efficacy and effectiveness of the proposed QOHBO-PF technique are verified by applying it to the standard IEEE and ill-conditioned systems. The robustness of the algorithm is validated under the maximum loadability limits and high R/X ratios, comparing the results with other well-known methods suggested in the literature. The results thus obtained show that the proposed QOHBO-PF technique has less computation time, further enhancement of reliability in the presence of PVG, and has the ability to provide multiple PF solutions that can be utilized for voltage stability analysis.
