Umbilical venous (UV) catheters (UVCs) are commonly used in severely ill neonates. Complications associated with UVC often result from an inappropriate UVC position. Calcification of the UV, a rare complication, was observed in an extremely low-birth-weight infant born at 23 weeks of gestation. After birth, the infant experienced respiratory and circulatory dysfunction, followed by disseminated intravascular coagulation (DIC). A UVC was inserted, and circulatory agonists and blood transfusions were administered, as well as a calcium gluconate infusion for hypocalcemia and hyperkalemia. Ten days after birth, calcification was detected in the UV, likely due to a tunica intima injury caused by UVC, a hypercoagulable state due to DIC, and a high-dose calcium gluconate infusion. Additionally, proximal port malpositioning of the double-lumen catheter might have contributed to calcification within the UV. To prevent such complications, real-time ultrasound confirmation with agitated saline contrast during UVC placement is recommended; in the absence of the facility or skills for ultrasonography, X-rays should be performed in the lateral and anteroposterior views. Furthermore, when using multi-lumen catheters, physicians should not only verify the tip position but also ensure proper placement of proximal ports and carefully select medications administered through the ports. Developing experimental animal models that show clinical symptoms and methods for quantitative and objective evaluation are important for understanding food allergies. Therefore, this study aimed to develop an ovalbumin (OVA)-induced mouse model of food allergy and a useful method to evaluate the symptoms of food allergy. Mice were sensitized via intraperitoneal injection of OVA. Subsequently, local sensitization was performed once weekly by oral administration of OVA. Itching and nasal symptoms were observed after oral administration of the antigen. First, we examined the dose-dependency of the antigen. Symptoms were checked weekly. In order to confirm food allergy symptoms, the effect of histamine H 1 receptor antagonist was examined. Finally, we measured antigen-specific IgE antibody levels in the serum. Scratching behavior, sneezing and nasal rubbing were increased. Both itching and rhinitis symptoms increased steadily, after which, the number of symptoms remained almost constant. No difference was observed between the results of 3- and 5-week-old mice. Cetirizine inhibited these symptoms in a dose-dependent manner. In addition, antigen-specific IgE antibodies were produced in both 3- and 5-week-old mice. This method may be useful for evaluating the symptoms of skin and rhinitis that could not be assessed in the conventional food allergy model and could be induced with a low dose of antigen. In particular, the developed method, which measures the number of itching and nasal symptoms, may enable quantitative, objective, and noninvasive evaluation of food allergy severity. Cases: We present 2 cases (19- and 40-year-old women) of lumbosacral spinal deformity with sacroiliac joint destruction. Reconstruction surgery using a fibular strut autograft was performed in both patients, and spinal reconstruction and fusion were successfully maintained without bone resorption for at least 7 years. Conclusion: This is the first report of reconstructive surgery using a fibular strut bone graft for lumbosacral deformity with destruction of the sacroiliac joint in patients with neurofibromatosis type 1 (NF-1). These findings suggest that reconstruction with a fibular strut autograft is useful for lumbosacral spinal deformity that includes destruction of the sacroiliac joints in NF-1. In the present study, mass transfer across a gas–liquid interface of acoustic cavitation bubble is numerically investigated. The volume of fluid (VOF) method coupled with a continuous species transfer model is used to simulate the bubble dynamic behavior and mass transfer across the gas–liquid interface. Chemical species are transferred repeatedly between the liquid and gas phases in accordance with the bubble oscillation, and the chemical species are transferred into the bubble during an expansion phase, and to the outside of bubble during a compression phase. Time-averaged Sherwood number is decreased with an amplitude of sound pressure due to area effect and shell effect , and its value becomes negative at high pressure amplitude. The structure of written texts is analyzed by focusing on word sequences. As a method, word sequences in texts are transformed into rank sequences of the occurrence frequency of each word and return maps are drawn. The features of word sequences are extracted by comparing with the surrogate data, i.e., a sequence in which all the words are randomly rearranged. A total of 140 written texts consisting of ten languages are selected for analysis. To characterize the distribution in the return map quantitatively, two characteristic quantities are defined, the distance between the original distribution and surrogate distribution, and the correlation coefficient of the adjacent word ranks. The results show that there is a negative correlation in the rank of adjacent words in almost all languages, and features of return maps of the same language texts are similar. A clustering structure which implies the relation to language (sub)family is observed. A mathematical model is proposed for reproducing features of the return map for multiple languages. An essential aspect of extending safe operation of the world's active nuclear reactors is understanding and predicting the embrittlement that occurs in the steels that make up the Reactor pressure vessel (RPV). In this work we integrate state of the art machine learning methods using ensembles of neural networks with unprecedented data collection and integration to develop a new model for RPV steel embrittlement. The new model has multiple improvements over previous machine learning and hand-tuned efforts, including greater accuracy (e.g., at high-fluence relevant for extending the life of present reactors), wider domain of applicability (e.g., including a wide-range of compositions), uncertainty quantification, and online accessibility for easy use by the community. These improvements provide a model with significant new capabilities, including the ability to easily and accurately explore compositions, flux, and fluence effects on RPV steel embrittlement for the first time. Furthermore, our detailed comparisons show our approach improves on the leading American Society for Testing and Materials (ASTM) E900-15 standard model for RPV embrittlement on every metric we assessed, demonstrating the efficacy of machine learning approaches for this type of highly demanding materials property prediction. Microstructural evolutions in two variants of the F82H 8 wt%Cr tempered martensitic steel under dual (He+ plus Fe3+) ion irradiations at 500 °C have been characterized by transmission electron microscopy. Emphasis is on understanding the combined effects of displacements per atom (dpa) and the helium/dpa ratio (He/dpa) on cavity evolution and void swelling (f v). The dual ion database includes 231 alloy-dpa-He/dpa conditions for irradiations up to ∼ 82 dpa and 3700 appm helium. We hypothesize that the large "scatter" observed in the f v data is primarily due to local microstructural variations, especially those that shield regions at shallower depths from the effects of injected interstitial atoms, which would otherwise reduce f v. More generally, our hypothesis is that the highest f v data, at a specified dpa and He/dpa, provides the most appropriate estimate of swelling for fusion neutron irradiation conditions. The dose dependence of f v can be described by an incubation dpa i , marking the onset of significant void growth, and a post-incubation swelling rate, f v ' = Δf v /Δdpa. Both dpa i and f v ' decrease with increasing He/dpa. The f v ' data are quantitatively consistent with a simple model for defect partitioning between evolving bubble, void, and dislocation sinks strengths and biases. Most notably, the model correctly predicts both the incubation dpa i , and the subsequent f v ', in spite of the fact that they reflect somewhat different physics. Our analysis also suggests that defect recombination is modest at 500 °C. Along with other considerations, this supports the hypothesis that, even at high dpa rates, dual ion irradiations can be used to reasonably emulate void nucleation and growth for a range of fusion-relevant neutron irradiation service conditions. The UCSB database on cavity evolution in 9-12Cr tempered martensitic steels (TMS), includes the results for both dual heavy and helium ion (DII), and High Flux Isotope Reactor (HFIR) in situ helium injection (ISHI) neutron irradiations at 500°C. These results were combined with literature single ion and fission neutron irradiation data to derive a model for the void volume fraction, f v , as a function of displacements per atom (dpa) and transmutant helium concentrations in atomic parts per million (appm). The scientific foundation for the paper is described in a companion paper entitled "Cavity Evolution and Void Swelling in Dual Ion Irradiated Tempered Martensitic Steels ". Here, we show that f v (dpa, He/dpa) is described by the incubation dose, dpa i , for the onset of void growth, and the post-incubation growth rate, f v '(%/dpa). Both dpa i and f v ' decrease with increasing He/dpa at > ∼ 5. The dpa i is also lower for the ISHI neutron irradiations at the same He/dpa. Single heavy ion and fission reactor neutron irradiations, with low He/dpa ratios, have a much larger dpa i. Based on a combined analysis of DII, single ion, ISHI and fission neutron data, we further show that the post-incubation f v data analyzed here have a common empirical curve shape, with f v ' reaching up to ∼ 0.2%/dpa at very high dpa. We also show that f v ' can be predicted based on a physical model of defect partitioning between evolving sinks. At 500°C and fusion relevant He/dpa ≈ 10, the best-fit model predicts nominal swelling, S = f v /(1-f v), of ∼ 1.1, 4.9 and 16% at 50, 100 and 200 dpa, respectively. Recent trends in the aluminum industry toward increasing the recycling rate of aluminum alloys have led to difficulties in controlling the formation of intermetallic compounds (IMCs). The up-to-date investigations have revealed that ultrasonic irradiation is a promising tool to refine and modify the intermetallic compounds formed in the melt before or during casting, and the reported mechanisms include ultrasound-assisted nucleation and fragmentation of compounds. This work presents results concerning the modification effect of ultrasound on model Al 3 Zr compounds with emphasis on the effect of vibration amplitudes. The results reveal that needle-like Al 3 Zr compounds can be significantly modified to thicker shape when amplitude increases from 38 µm to 48 µm, and then change to more rounded shape with the further increase of amplitude. Aiming at reducing energy consumptions in data writing, the development of new memory materials is highly required. To develop new phase change materials (PCMs) with extremely low operation energy used in non-volatile memory, such as storage-class memory, we performed Bayesian optimization for the physical properties of PCMs through numerical simulations. In this numerical simulation, the electrical potential and temperature distribution were solved simultaneously. It was found that a PCM with low thermal conductivity, low melting temperature, and low ratio of contact resistance to volumetric resistance results in a low operation energy of a PCM-based memory application. Finally, we developed a design strategy for PCMs. New PCMs should be developed by lowering the operation energy E , described as E = κ (1 + C)Δ T /Δ z , where κ is the thermal conductivity of the PCM, Δ T is the melting temperature, C is the ratio of contact resistance to the volumetric resistance, and Δ z is the thickness of the PCM. The present results clarify the relationship between thermal and electrical properties for lowering operation energy ever hidden in previous studies. The proliferation and differentiation of skeletal muscle cells are usually controlled by serum components. Myogenic differentiation is induced by a reduction of serum components in vitro. It has been recently reported that serum contains not only various growth factors with specific actions on the proliferation and differentiation of myogenic cells, but also exogenous exosomes, the function of which is poorly understood in myogenesis. We have found that exosomes in fetal bovine serum are capable of exerting an inhibitive effect on the differentiation of C2C12 myogenic cells in vitro. In this process of inhibition, the downregulation of Tceal5 and Tceal7 genes was observed. Expression of these genes is specifically increased in direct proportion to myogenic differentiation. Loss- or gain- of function studies with Tceal5 and Tceal7 indicated that they have the potential to regulate myogenic differentiation via exosomes in fetal bovine serum. [ABSTRACT FROM AUTHOR]

• Investigation on gas injection during ACAP process is conducted. • Argon provides a better efficiency of Rhodamine B degradation compared to air. • Hydrogen and oxygen-containing radicals are generated in larger amount during argon injection. • Argon dissolves in water that lowers breakdown voltage and improves cavitation treatment. Underwater plasma has been long recognized as a "green" alternative to conventional chemicals in the wastewater treatment processes. However, practical application of underwater plasma is still challenging due to insufficient treatment performance. Recently, we proposed a novel process named ACAP utilizing acoustic cavitation in order to stabilize the plasma generation and to enlarge the plasma processing region. This work continues our investigation regarding the ACAP treatment process focusing on effects of gas injection. Experiments were performed using an ultrasonic installation equipped with a specially designed sonotrode (Diam. 48 mm) operated at a frequency of 20 kHz and acoustic power of 120 W. The results revealed that the degradation efficiency of Rhodamine B, which was used as a model wastewater pollutant, remains almost unchangeable in the case of air injection, but it is doubled when argon is injected into the ACAP reactor. It was found that the argon injection enhances the degradation efficiency significantly even without ultrasound irradiation. Results of additional measurements suggest that the effect of argon is attributed to its ability to yield high temperature during cavitation, comparatively good solubility in water and a better ability to reduce the breakdown voltage in water compared to the air case. In our previous study, a novel method combining underwater high-voltage plasma discharge with acoustic cavitation (ACAP) was developed and implemented using rhodamine B (RhB) as a model organic pollutant. Results revealed that injecting argon gas into the ACAP reactor positively influences RhB decomposition efficiency, but there is still further potential for improvement. The aim of this study was therefore to further improve the efficiency of the ACAP process through Fenton reactions. Two options for ferrous ion supply were considered: the addition of FeCl2 or the dissolution of iron from ACAP reactor steel parts into the RhB-containing solution. The results revealed that the degradation efficiency is increased by 20% due to the Fenton reactions when the concentration of ferrous ions reaches an optimal value. Lower pH was found to be desirable for the effect of Fenton reactions. Based on measurements using high performance liquid chromatography, a plausible mechanism of RhB degradation by the ACAP process assisted by Fenton reactions is additionally proposed and discussed. Human pluripotent stem cells (hPSCs) are grouped into two cell types; embryonic stem cells (hESCs) and induced pluripotent stem cells (hiPSCs). hESCs have provided multiple powerful platforms to study human biology, including human development and diseases; however, there were difficulties in the establishment of hESCs from human embryo and concerns over its ethical issues. The discovery of hiPSCs has expanded to various applications in no time because hiPSCs had already overcome these problems. Many hPSC-based studies have been performed using two-dimensional monocellular culture methods at the cellular level. However, in many physiological and pathophysiological conditions, intra- and inter-organ interactions play an essential role, which has hampered the establishment of an appropriate study model. Therefore, the application of recently developed technologies, such as three-dimensional organoids, bioengineering, and organ-on-a-chip technology, has great potential for constructing multicellular tissues, generating the functional organs from hPSCs, and recapitulating complex tissue functions for better biological research and disease modeling. Moreover, emerging techniques, such as single-cell transcriptomics, spatial transcriptomics, and artificial intelligence (AI) allowed for a denser and more precise analysis of such heterogeneous and complex tissues. Here, we review the applications of hPSCs to construct complex organs and discuss further prospects of disease modeling and drug discovery based on these PSC-derived organs. [ABSTRACT FROM AUTHOR]

Acoustic cavitation occurs in ultrasonic treatment causing various phenomena such as chemical synthesis, chemical decomposition, and emulsification. Nonlinear oscillations of cavitation bubbles are assumed to be responsible for these phenomena, and the neighboring bubbles may interact each other. In the present study, we numerically investigated the dynamic behavior of cavitation bubbles in multi-bubble systems. The results reveal that the oscillation amplitude of a cavitation bubble surrounded by other bubbles in a multi-bubble system becomes larger compared with that in the single-bubble case. It is found that this is caused by an acoustic wake effect, which reduces the pressure near a bubble surrounded by other bubbles and increases the time delay between the bubble contraction/expansion cycles and sound pressure oscillations. A new parameter, called "cover ratio" is introduced to quantitatively evaluate the variation in the bubble oscillation amplitude, the time delay, and the maximum bubble radius. It is well known that solidification of molten metals occurs under the influence of transient unsteady phenomena, which are difficult to control in actual casting operation. Examples include buoyancy flows, natural heat convection, repeated remelting and solidification in mushy zone and solid shell at the mold interface. This paper presents the results suggesting that ultrasound waves, irradiated into the hot-top mold offers an attractive way to control the transient phenomena in DC casting of aluminum alloy billets. A novel mathematical model was developed to simulate the DC casting with considering transient melt flow, heat transfer, ultrasound propagation, acoustic streaming and solidification. It was found that ultrasound irradiation alters the solidification behavior of melt and sump evolution especially in the earlier stage of casting, however as the billet length increases, the sump profile becomes almost the same for both the ultrasonic and conventional castings. In this condition, the casting speed becomes the key parameter influencing the sump evolution and mushy zone volume. The results reveal that ultrasound-driven acoustic streaming and turbulent oscillations result in a suppression of buoyancy flow in the sump and lead to an increase of frequency of the melt temperature oscillations at the solidification interface. One of the useful results of these phenomena can be a more frequent repetition of remelting/solidification cycle and improvement of the billet quality, particularly the billet surface morphology. Although the mRNA SARS-CoV-2 vaccine has improved the mortality rate in the general population, its efficacy against rapidly mutating virus strains, especially in kidney transplant recipients, remains unclear. We examined the anti-SARS-CoV-2 spike protein IgG antibody and neutralizing antibody titers and cellular immunity against B.1.1, BA.1, and BA.5 antigens in 73 uninfected kidney recipients and 16 uninfected healthy controls who received three doses of an mRNA SARS-CoV-2 vaccine. The IgG antibody titers were significantly lower in recipients than in healthy controls. Similarly, neutralizing antibody titers against three viral variants were significantly lower in recipients. When the virus was mutated, the neutralizing antibody titers decreased significantly in both groups. In cellular immunity analysis, the number of spike-specific CD8 + non-naïve T cells against three variants significantly decreased in recipients. Conversely, the frequency of spike-specific Th2 CD4 + T-cells in recipients was higher than that in healthy controls. Nineteen recipients and six healthy controls also received a bivalent omicron-containing booster vaccine, leading to increase IgG and neutralizing antibody titers in both groups. After that, eleven recipients and five healthy controls received XBB.1.5 monovalent vaccines, increasing the neutralizing antibody titers against not only XBB.1.5, but also EG.5.1 and BA.2.86 antigens in kidney recipients. Although kidney recipients did not gain sufficient immunity against Omicron BA.5 with the third dose of vaccine, humoral response against mutant SARS-CoV-2 lineages significantly increased after bivalent Omicron-containing booster vaccine and the XBB.1.5 monovalent vaccine. Accumulating evidence suggests that endothelial cells can be useful therapeutic targets. One of the potential targets is an endothelial cell-specific protein, Roundabout4 (ROBO4). ROBO4 has been shown to ameliorate multiple diseases in mice, including infectious diseases and sepsis. However, its mechanisms are not fully understood. In this study, using RNA-seq analysis, we found that ROBO4 downregulates prostaglandin-endoperoxide synthase 2 (PTGS2), which encodes cyclooxygenase-2. Mechanistic analysis reveals that ROBO4 interacts with IQ motif-containing GTPase-activating protein 1 (IQGAP1) and TNF receptor-associated factor 7 (TRAF7), a ubiquitin E3 ligase. In this complex, ROBO4 enhances IQGAP1 ubiquitination through TRAF7, inhibits prolonged RAC1 activation, and decreases PTGS2 expression in inflammatory endothelial cells. In addition, Robo4-deficiency in mice exacerbates PTGS2-associated inflammatory diseases, including arthritis, edema, and pain. Thus, we reveal the molecular mechanism by which ROBO4 suppresses the inflammatory response and vascular hyperpermeability, highlighting its potential as a promising therapeutic target for inflammatory diseases. To gain insight into how immunity develops against SARS-CoV-2 from 2020 to 2022, we analyzed the immune response of a small group of university staff and students who were either infected or vaccinated. We investigated the levels of receptor-binding domain (RBD)-specific and nucleocapsid (N)-specific IgG and IgA antibodies in serum and saliva samples taken early (around 10 days after infection or vaccination) and later (around 1 month later), as well as N-specific T-cell responses. One patient who had been infected in 2020 developed serum RBD and N-specific IgG antibodies, but declined eight months later, then mRNA vaccination in 2021 produced a higher level of anti-RBD IgG than natural infection. In the vaccination of naïve individuals, vaccines induced anti-RBD IgG, but it declined after six months. A third vaccination boosted the IgG level again, albeit to a lower level than after the second. In 2022, when the Omicron variant became dominant, familial transmission occurred among vaccinated people. In infected individuals, the levels of serum anti-RBD IgG antibodies increased later, while anti-N IgG peaked earlier. The N-specific activated T cells expressing IFN γ or CD107a were detected only early. Although SARS-CoV-2-specific salivary IgA was undetectable, two individuals showed a temporary peak in RBD- and N-specific IgA antibodies in their saliva on the second day after infection. Our study, despite having a small sample size, revealed that SARS-CoV-2 infection triggers the expected immune responses against acute viral infections. Moreover, our findings suggest that the temporary mucosal immune responses induced early during infection may provide better protection than the currently available intramuscular vaccines. Background: Instrumentation failure (IF) is a major complication associated with growth-sparing surgery for pediatric spinal deformities; however, studies focusing on IF following each surgical procedure are lacking. We aimed to evaluate the incidence, timing, and rates of unplanned return to the operating room (UPROR) associated with IF following each surgical procedure in growth-sparing surgeries using traditional growing rods (TGRs) and vertical expandable prosthetic titanium ribs (VEPTRs). Methods: We reviewed 1,139 surgical procedures documented in a Japanese multicenter database from 2015 to 2017. Of these, 544 TGR and 455 VEPTR procedures were included for evaluation on a per-surgery basis. IF was defined as the occurrence of an implant-related complication requiring revision surgery. Results: The surgery-based incidences of IF requiring revision surgery in the TGR and VEPTR groups were 4.3% and 4.0%, respectively, with no significant intergroup difference. Remarkably, there was a negative correlation between IF incidence per surgical procedure and the number of lengthening surgeries in both groups. In addition, rod breakage in the TGR group and anchor-related complications in the VEPTR group tended to occur relatively early in the treatment course. The surgery-based rates of UPROR due to IF in the TGR and VEPTR groups were 2.0% and 1.5%, respectively, showing no statistically significant difference. Conclusions: We found that IF, such as anchor related-complications and rod breakage, occurs more frequently earlier in the course of lengthening surgeries. Life extension of light water reactors requires robust models to predict the embrittlement of reactor pressure vessel (RPV) steels under long-term neutron irradiation. Embrittlement is due to nanoscale precipitation hardening by phases containing Cu, Mn, Ni, and Si, resulting in increases of the ductile to brittle transition temperature. Embrittlement data for low flux, high fluence extended life conditions, up to 80 years or more, are largely not available. Thus, higher flux, accelerated test reactor irradiations have been used to help develop models to predict low flux embrittlement at high fluence. Here flux, or dose rate, is expressed in units of displacements per atom per second (dpa/s). The dose rate can significantly influence precipitate evolution in a way that depends on fluence (dpa), temperature, and alloy composition, as well as flux itself. Here, we explore precipitation in surveillance steels with varying compositions at high fluxes (10−5–10−6 dpa/s) produced using 70 MeV Fe2+ ion irradiations. Atom probe tomography was used to characterize the dose dependence of precipitation at very high ion irradiation dose rates, and to evaluate corresponding similarities or differences compared to neutron irradiated steels at a much lower dose rate of ≈ 5 × 10−9 dpa/s. The data show that, on average, the ion irradiation precipitate volume fractions increase by factors of ∼ 1.5 to 2 between 0.3 dpa and 1.2 dpa. Limited data show that the major effect of neutron versus ion irradiation is the former has lower precipitate number densities and larger sizes. The maternal liver is challenged by metabolic demands throughout pregnancy. However, hepatocyte dynamics and their physiological significance in pregnancy remain unclear. Here, we show in mice that hepatocyte proliferation is spatiotemporally regulated in each liver lobular zone during pregnancy, with transient proliferation of periportal and pericentral hepatocytes during mid and late gestation, respectively. Using adeno-associated virus (AAV)−8-mediated expression of the cell cycle inhibitor p21 in hepatocytes, we show that inhibition of hepatocyte proliferation during mid, but not late, gestation impairs liver growth. Transcriptionally, genes involved in glucose/glycogen metabolism are downregulated in late pregnancy when midgestational hepatocyte proliferation is attenuated. In addition, hepatic glycogen storage is abolished, with concomitant elevated blood glucose concentrations, glucose intolerance, placental glycogen deposition, and fetal overgrowth. Laser capture microdissection and RNA-seq analysis of each liver lobular zone show zone-specific changes in the transcriptome during pregnancy and identify genes that are periportally expressed at midgestation, including the hyaluronan-mediated motility receptor (Hmmr). Knockdown of Hmmr in hepatocytes by AAV8-shHmmr suppresses periportal hepatocyte proliferation at midgestation and induces impaired hepatic glycogen storage, glucose intolerance, placental glycogen deposition and fetal overgrowth. In-service exposure to high-energy neutrons embrittles reactor pressure vessel (RPV) steels. An increase in the yield stress (Δσ y) results in a corresponding increase in the brittle to ductile transition temperature (ΔT c). Most existing models underpredict ΔT c at higher fluence following accelerated irradiations in test reactors. High fluence, up to 1020 n/cm2 in some cases, will be reached over extended RPV vessel operation of 80 years, or more, at low service flux. Embrittlement has been extensively studied in accelerated, higher flux test reaction irradiations. However, the use of test reactor data naturally raises the question of flux effects. This study used a machine learning approach trained on a set of hardening data, covering a wide range of flux, fluence, and steel compositions to determine the interactive effects of both irradiation and material variables on Δσ y. The analysis included machine learning-based cross-plots of the variable dependence of Δσ y for six core steels (i.e., CM6, LC, LD, LG, LH, and LI), with controlled differences in their Cu and Ni contents. A primary objective is to evaluate an effective to actual fluence (ɸt e /ɸt) ratio, as a function of flux, fluence, and steel composition. This is information critical to properly use intermediate flux-high fluence data in calibrating a low flux-high fluence embrittlement model. However, their synthesis remains underexplored. Here, we investigated macro‐rotaxane formation by the topological trapping of multicyclic polydimethylsiloxanes (mc‐PDMSs) in silicone networks. mc‐PDMS with different numbers of cyclic units and ring sizes was synthesized by cyclopolymerization of a α,ω‐norbornenyl‐functionalized PDMS. Silicone networks were prepared in the presence of 10–60 wt % mc‐PDMS, and the trapping efficiency of mc‐PDMS was determined. In contrast to monocyclic PDMS, mc‐PDMSs with more cyclic units and larger ring sizes can be quantitatively trapped in the network as macro‐rotaxanes. Development of a universal influenza vaccine that can provide robust and long-lasting protection against heterologous infections is a global public health priority. A variety of vaccine antigens are designed to increase the antigenicity of conserved epitopes to elicit cross-protective antibodies that often lack virus-neutralizing activity. Given the contribution of antibody effector functions to cross-protection, adjuvants need to be added to modulate antibody effector functions as well as to enhance antibody quantity. We previously showed that post-fusion influenza vaccine antigens elicit non-neutralizing but cross-protective antibodies against conserved epitopes. Here, using a murine model, we comparably assessed the adjuvanticity of the newly developed SA-2 adjuvant containing a synthetic TLR7 agonist DSP-0546 and squalene-based MF59 analog as representative Th1- or Th2-type adjuvants, respectively. Both types of adjuvants in the post-fusion vaccine comparably enhanced cross-reactive IgG titers against heterologous strains. However, only SA-2 skewed the IgG subclass into the IgG2c subclass in association to its Th1-polarizing nature. SA-2-enhanced IgG2c responses exhibited antibody-dependent cellular cytotoxicity against heterologous virus strains, without cross-neutralizing activity. Eventually, the SA-2-adjuvanted vaccination provided protection against lethal infection by heterologous H3N2 and H1N1 viruses. Together, we conclude that the combination with a SA-2 is advantageous for enhancing the cross-protective capability of post-fusion HA vaccines that elicit non-neutralizing IgG antibodies. Lipid metabolism is one of energy metabolic pathways that produce adenosine triphosphate (ATP). In this pathway, lysosomal acid lipase (LAL) encoded by Lipase A (LIPA), plays an important role in catalyzing lipids to fatty acids (FAs), which drive oxidative phosphorylation (OXPHOS) and generate ATP. Previously, we found that a LIPA single nucleotide polymorphism rs143793106, which decreases the LAL activity, suppressed the cytodifferentiation of human periodontal ligament (HPDL) cells. However, the mechanisms underlying that suppression are still not fully clarified. Thus, we aimed to investigate the mechanisms regulating the cytodifferentiation of HPDL cells by LAL in terms of energy metabolism. We performed the osteogenic induction of HPDL cells with or without Lalistat-2, a LAL inhibitor. To visualize lipid droplet (LD) utilization, we performed confocal microscopy on HPDL cells. We also performed real-time PCR to analyze the gene expression of calcification-related and metabolism-related genes. Furthermore, we measured the ATP production rate from two major energy production pathways, OXPHOS and glycolysis, and OXPHOS-related parameters of HPDL cells during their cytodifferentiation. We found that LDs were utilized during the cytodifferentiation of HPDL cells. Alkaline phosphatase (ALPL), collagen type 1 alpha 1 chain (COL1A1), ATP synthase F1 subunit alpha (ATP5F1A), and carnitine palmitoyltransferase 1A (CPT1A) mRNA expressions were upregulated, whereas lactate dehydrogenase A (LDHA) mRNA expression was downregulated. Additionally, total ATP production rate was significantly increased. In contrast, in the presence of Lalistat-2, LD utilization was inhibited and ALPL , COL1A1 , and ATP5F1A mRNA expression was downregulated. Additionally, ATP production rate and spare respiratory capacity of the OXPHOS pathway were decreased in HPDL cells during their cytodifferentiation. Collectively, the defect of LAL in HPDL cells decreased LD utilization and OXPHOS capacity, resulting in reduced energy to sustain the adequate ATP production required for the cytodifferentiation of HPDL cells. Recently, experimental investigations of a class of temperature-responsive polymers tethered to oligooxyethylene side chains terminated with alkyl groups have been conducted. In this study, aqueous solutions of poly(glycidyl ether)s (PGE) with varying numbers of oxyethylene units, poly(methyl(oligooxyethylene)n glycidyl ether) (poly(Me(EO)nGE)), and poly(ethyl(oligooxyethylene)n glycidyl ether) (poly(Et(EO)nGE) (n = 0, 1, and 2) were investigated by all-atom molecular dynamics simulations, focusing on the thermal responses of their chain extensions, the recombination of intrapolymer and polymer–water hydrogen bonds, and water-solvation shells around the alkyl groups. No clear relationship was established between the phase-transition temperature and the polymer-chain extensions unlike the case for the coil–globule transition of poly(N-isopropylacrylamide). However, the temperature response of the first water-solvation shell around the alkyl group exhibited a notable correlation with the phase-transition temperature. In addition, the temperature at which the hydrophobic hydration shell strength around the terminal alkyl group equals the bulk water density ( T CRP ) was slightly lower than the cloud point temperature ( T CLP ) for the methyl-terminated poly(Me(EO)nGE) and slightly higher for the ethyl-terminated poly(Et(EO)nGE). Human in vitro oogenesis provides a framework for clarifying the mechanism of human oogenesis. To create its benchmark, it is vital to promote in vitro oogenesis using a model physiologically close to humans. Here, we establish a foundation for in vitro oogenesis in cynomolgus (cy) monkeys (Macaca fascicularis): cy female embryonic stem cells harboring one active and one inactive X chromosome (Xa and Xi, respectively) differentiate robustly into primordial germ cell‐like cells, which in xenogeneic reconstituted ovaries develop efficiently into oogonia and, remarkably, further into meiotic oocytes at the zygotene stage. This differentiation entails comprehensive epigenetic reprogramming, including Xi reprogramming, yet Xa and Xi remain epigenetically asymmetric with, as partly observed in vivo, incomplete Xi reactivation. In humans and monkeys, the Xi epigenome in pluripotent stem cells functions as an Xi‐reprogramming determinant. We further show that developmental pathway over‐activations with suboptimal up‐regulation of relevant meiotic genes impede in vitro meiotic progression. Cy in vitro oogenesis exhibits critical homology with the human system, including with respect to bottlenecks, providing a salient model for advancing human in vitro oogenesis. Endothelial cells (ECs) line blood vessels and serve as a niche for hematopoietic stem and progenitor cells (HSPCs). Recent data point to tissue-specific EC specialization as well as heterogeneity; however, it remains unclear how ECs acquire these properties. Here, by combining live-imaging-based lineage-tracing and single-cell transcriptomics in zebrafish embryos, we identify an unexpected origin for part of the vascular HSPC niche. We find that islet1 (isl1)-expressing cells are the progenitors of the venous ECs that constitute the majority of the HSPC niche. These isl1 -expressing cells surprisingly originate from the endoderm and differentiate into ECs in a process dependent on Bmp-Smad signaling and subsequently requiring npas4l (cloche) function. Single-cell RNA sequencing analyses show that isl1 -derived ECs express a set of genes that reflect their distinct origin. This study demonstrates that endothelial specialization in the HSPC niche is determined at least in part by the origin of the ECs. The redox behaviors of macrocyclic molecules with an entirely π-conjugated system are of interest due to their unique optical, electronic, and magnetic properties. In this study, defect-free cyclic P3HT with a degree of polymerization (DPn) from 14 to 43 was synthesized based on our previously established method, and its unique redox behaviors arising from the cyclic topology were investigated. Cyclic voltammetry (CV) showed that the HOMO level of cyclic P3HT decreases from –4.86 eV (14 mer) to –4.89 eV (43 mer), in contrast to the linear counterparts increasing from –4.94 eV (14 mer) to –4.91 eV (43 mer). During the CV measurement, linear P3HT suffered from electro-oxidation at the chain ends, while cyclic P3HT was stable. ESR and UV–Vis–NIR spectroscopy suggested that cyclic P3HT has stronger dicationic properties due to the interactions between the polarons. On the other hand, linear P3HT showed characteristics of polaron pairs with multiple isolated polarons. Background: Hypoglycemia is a significant problem for all neonates and requires minimally invasive and reliable monitoring. The primary objective of this study was to verify the safety and accuracy of the continuous glucose monitoring (CGM) of full‐term neonates using Freestyle Libre, a flash glucose monitoring (FGM) device. Methods: The study was conducted on 20 neonates. Shortly after birth, we placed the FGM sensor on the outside of the neonates' thighs. We scanned the CGM values at 60, 120, 180, and 360 min after birth and simultaneously obtained blood glucose values with plantar capillaries by heel puncture. The neonates wore the sensors for up to 6 h and then they were removed. Results: Of the 75 data points to be measured, 65 points (86.7%) were obtained by scan. There was no change in the sensor attachment site in 12 of 18 completed cases in this study but we observed slight induration in four cases (22.2%) and slight redness in one case (5.5%) at the sensor puncture site. A moderate correlation was observed between the CGM and blood glucose values. The CGM values tended to be low at 120, 180, and 360 min after birth, and tended to be high only at 60 min after birth. Conclusions: The CGM device was safe to wear on the neonate and the CGM data correlated well with blood glucose levels. Ameloblastic fibrosarcoma (AFS) is the most common odontogenic sarcoma, but the incidence is relatively low, and its molecular biology is poorly understood. We experienced a young female patient with a rapidly growing soft tissue tumor of the left maxilla, which eventually occupied the left side of the oral cavity. Histologically, the tumor mainly consisted of a proliferation of atypical spindle to polygonal cells without any specific differentiation, but a small number of benign odontogenic epithelial foci mainly in the tumor periphery were also noted; thus, a diagnosis of AFS was made. We performed whole-exome sequencing (WES) on the tumor to investigate its molecular features and identify therapeutic options. We found that the tumor harbored EGFR exon 20 insertions and MDM2 amplification; the former may be a target for newly developed tyrosine kinase inhibitors in case of recurrence. To the best of our knowledge, this is the first case of AFS for which WES was performed and with EGFR mutation. Our case provides new genetic information on AFS and suggests that comprehensive genetic analysis can clarify the molecular biology in rare cancers, potentially leading to the proposal of therapeutic strategies. Silver nanoparticles (AgNPs) are used in a wide range of applications, and the size control and stability of the nanoparticles are crucial aspects in their applications. In the present study, cyclized poly(ethylene glycol) (c-PEG) with various molecular weights, along with linear PEG with hydroxy chain ends (HO–PEG–OH) and methoxy chain ends (MeO–PEG–OMe) were applied for the Tollens' synthesis of AgNPs. The particle size was significantly affected by the topology and end groups of PEG. For example, the size determined by TEM was 40 ± 7 nm for HO–PEG5k–OH, 21 ± 4 nm for c-PEG5k, and 48 ± 9 nm for MeO–PEG5k–OMe when the molar ratio of PEG to AgNO3 (ω) was 44. The stability of AgNPs was also drastically improved by cyclization; the relative UV–Vis absorption intensity (A/A0 × 100%) at λmax to determine the proportion of persisting AgNPs in an aqueous NaCl solution (37.5 mM) was 58% for HO–PEG5k–OH, 80% for c-PEG5k, and 40% for MeO–PEG5k–OMe, despite the fact that AgNPs with c-PEG5k were much smaller than those with HO–PEG5k–OH and MeO–PEG5k–OMe. Aluminum contaminations, particularly iron, present a serious challenge to aluminum recycling technology. This is why many studies have focused on the reduction of detrimental effects of iron-containing contaminations through addition of elements such as manganese and chromium. However, the desirable modifying effect is often difficult to achieve because it would require concentrations of added elements greater than the allowable limits for many aluminum alloys. Thus, an alternative way to obtain the modifying effect, by using a much smaller amount of the modifying elements which are added to the aluminum melt as solid particles, was proposed in this study. The main goal of the present study was to investigate the possibilities of fabricating an aluminum master alloy by adding CrSi2 particles onto the surface of the vortex formed during mechanical agitation of molten aluminum. Two kinds of CrSi2 powder were used: one was commercial powder, and the other was self-synthesized CrSi2 via mechanical alloying by planetary ball milling. The results revealed that CrSi2 particles with a larger size penetrate the melt better. Particles of three kinds were found to exist in the Al melt after the addition of CrSi2 powder: (1) inclusions of eutectic origin formed at the last stage of crystallization, (2) mixtures of Al-Cr compounds and original CrSi2 particles and (3) original CrSi2 particles. Low melt temperatures and short treatment times were found to favor the fabrication of master alloys because they impeded the dissolution of CrSi2 particles into the Al melt and, thus, allowed one to fabricate the master alloy containing the particles of the second and third types. Simple Summary: Pituitary adenoma pathophysiology has been studied mainly using murine cell lines, animal models, and pituitary tumor samples. However, the lack of human pituitary cell line is a significant limiting factor in studying the molecular mechanisms of human pituitary tumors. Recently, pituitary induction methods from human-induced pluripotent stem cells (hiPSCs) have been established. These methods can induce human pituitary hormone-producing cells that retain physiological properties. hiPSCs in which tumor-causing gene mutations are introduced using genome-editing techniques, such as CRISPR/Cas9 systems, provide great opportunities to establish in vitro human pituitary adenoma disease models. The models will be a novel platform to discover novel drugs and investigate tumorigenesis and pathophysiology. The purpose of this review is to provide an overview of the applications of iPSCs for pituitary and neoplastic disorder research and genome-editing technologies to create strategies for developing pituitary adenoma models using iPSCs. Pituitary adenomas are characterized by abnormal growth in the pituitary gland. Surgical excision is the first-line treatment for functional (hormone-producing) pituitary adenomas, except for prolactin-producing adenomas; however, complete excision is technically challenging, and many patients require long-term medication after the treatment. In addition, the pathophysiology of pituitary adenomas, such as tumorigenesis, has not been fully understood. Pituitary adenoma pathophysiology has mainly been studied using animal models and animal tumor-derived cell lines. Nevertheless, experimental studies on human pituitary adenomas are difficult because of the significant differences among species and the lack of reliable cell lines. Recently, several methods have been established to differentiate pituitary cells from human pluripotent stem cells (hPSCs). The induced pituitary hormone-producing cells retain the physiological properties already lost in tumor-derived cell lines. Moreover, CRISPR/Cas9 systems have expedited the introduction of causative gene mutations in various malignant tumors into hPSCs. Therefore, hPSC-derived pituitary cells have great potential as a novel platform for studying the pathophysiology of human-specific pituitary adenomas and developing novel drugs. This review presents an overview of the recent progresses in hPSC applications for pituitary research, functional pituitary adenoma pathogenesis, and genome-editing techniques for introducing causative mutations. Early diagnosis of pancreatic ductal adenocarcinoma (PDAC) is essential for improving prognosis; however, diagnosing PDAC at an early stage is challenging. In patients with localized high‐grade pancreatic intraepithelial neoplasia (HG‐PanIN), whose tumorous lesion is undetectable on cross‐sectional images such as computed tomography or magnetic resonance image, long‐term survival is expected. Pancreatic cystic lesions or main pancreatic duct (MPD) dilatation are important indirect findings for the initial diagnosis of HG‐PanIN. Magnetic resonance cholangiopancreatography (MRCP) and endoscopic ultrasound (EUS) should play important roles in detecting abnormal image findings, such as local irregular MPD stenosis, caliber MPD changes, small cystic lesions, or branch duct dilatation. Additionally, EUS could detect hypoechoic areas around the MPD stenosis in some patients with HG‐PanIN. Subsequently, endoscopic retrograde cholangiopancreatography (ERCP) and its associated pancreatic juice cytology, including serial pancreatic juice aspiration cytologic examination (SPACE) after placement of an endoscopic nasopancreatic drainage (ENPD) tube, may have high diagnostic accuracy for confirming the malignancy in HG‐PanIN. Although ERCP and its associated pancreatic cytology, including SPACE, may be associated with post‐ERCP pancreatitis (PEP), a recent randomized trial suggested that a 4‐Fr ENPD tube may reduce the incidence of PEP. In the future, further prospective multicenter studies are required to establish a standard method of SPACE. Additionally, further studies for novel biomarkers could help to establish evolutionary methods with duodenal fluid and pancreatic juice for the early and accurate diagnosis of early‐stage PDAC. The development of an in vitro cell model that can be used to study severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) research is expected. Here we conducted infection experiments in bronchial organoids (BO) and an BO-derived air-liquid interface model (BO-ALI) using 8 SARS-CoV-2 variants. The infection efficiency in BO-ALI was more than 1,000 times higher than that in BO. Among the bronchial epithelial cells, we found that ciliated cells were infected with the virus, but basal cells were not. Ciliated cells died 7 days after the viral infection, but basal cells survived after the viral infection and differentiated into ciliated cells. Fibroblast growth factor 10 signaling was essential for this differentiation. Block copolymers (BCPs) have garnered considerable interest due to their ability to form microphase-separated structures suitable for nanofabrication. For these applications, it is critical to achieve both sufficient etch selectivity and a small domain size. To meet both requirements concurrently, we propose the use of oligosaccharide and oligodimethylsiloxane as hydrophilic and etch-resistant hydrophobic inorganic blocks, respectively, to build up a novel BCP system, i.e., carbohydrate-inorganic hybrid BCP. The carbohydrate-inorganic hybrid BCPs were synthesized via a click reaction between oligodimethylsiloxane with an azido group at each chain end and propargyl-functionalized maltooligosaccharide (consisting of one, two, and three glucose units). In the bulk state, small-angle X-ray scattering revealed that these BCPs microphase separated into gyroid, asymmetric lamellar, and symmetric lamellar structures with domain-spacing ranging from 5.0 to 5.9 nm depending on the volume fraction. Additionally, we investigated microphase-separated structures in the thin film state and discovered that the BCP with the most asymmetric composition formed an ultrafine and highly oriented gyroid structure as well as in the bulk state. After reactive ion etching, the gyroid thin film was transformed into a nanoporous-structured gyroid SiO2 material, demonstrating the material's promising potential as nanotemplates. The topology effects of cyclization on thermal phase transition behaviors were investigated for a series of amphiphilic Pluronic copolymers of both hydrophilic–hydrophobic–hydrophilic and hydrophobic–hydrophilic–hydrophobic block sequences. The dye solubilization measurements revealed the lowered critical micelle temperatures (TCMT) along with the decreased micellization enthalpy (ΔHmic) and entropy (ΔSmic) for the cyclized species. Furthermore, the transmittance and dynamic light scattering (DLS) measurements indicated a block sequence-dependent effect on the clouding phenomena, where a profound decrease in cloud point (Tc) was only found for the copolymers with a hydrophilic–hydrophobic–hydrophilic block sequence. Thus, the effect of cyclization on these critical temperatures was manifested differently depending on its block sequence. Finally, a comparison of the linear hydroxy-terminated, methoxy-terminated, and cyclized species indicated the effect of cyclization to be unique from a simple elimination of the terminal hydrophilic moieties. Naked mole-rats (NMRs) have a very low spontaneous carcinogenesis rate, which has prompted studies on the responsible mechanisms to provide clues for human cancer prevention. However, it remains unknown whether and how NMR tissues respond to experimental carcinogenesis induction. Here, we show that NMRs exhibit extraordinary resistance against potent chemical carcinogenesis induction through a dampened inflammatory response. Although carcinogenic insults damaged skin cells of both NMRs and mice, NMR skin showed markedly lower immune cell infiltration. NMRs harbour loss-of-function mutations in RIPK3 and MLKL genes, which are essential for necroptosis, a type of necrotic cell death that activates strong inflammation. In mice, disruption of Ripk3 reduced immune cell infiltration and delayed carcinogenesis. Therefore, necroptosis deficiency may serve as a cancer resistance mechanism via attenuating the inflammatory response in NMRs. Our study sheds light on the importance of a dampened inflammatory response as a non-cell-autonomous cancer resistance mechanism in NMRs. Naked mole rats are found to be resistant to cancer development through dampened inflammatory response due to genetically determined impaired necroptosis, with essential necroptosis genes RIPK3 and MLKL containing mutations causing premature stop codons. In pregnant mice, the maternal liver expands drastically during gestation, which is believed to be essential to accommodate various metabolic demands caused by physiological changes and fetal growth. Although hepatocyte proliferation and hypertrophy have been reported, little is known about the dynamics of biliary epithelial cells (BECs), which comprise the bile duct epithelium in the liver. Here, we show that BECs transiently proliferate during the early stage of gestation. Lineage tracing revealed that BEC progeny were retained in the bile duct epithelium and did not differentiate into hepatocytes, indicating BEC self‐replication during pregnancy. RNA‐sequencing analysis of BECs identified their early pregnancy‐signature transcriptomes, which highlighted Yes‐associated protein (YAP) signaling‐related genes. Nuclear accumulation of YAP was enhanced in BECs during pregnancy but was barely detectable in hepatocytes. In addition, the pharmacological inhibition of YAP attenuated BEC proliferation and liver weight gain during pregnancy. Herein, we report the Suzuki–Miyaura catalyst-transfer polycondensation (SCTP) of triolborate-type carbazole monomers, i.e., potassium 3-(6-bromo-9-(2-octyldodecyl)-9H-carbazole-2-yl)triolborate (M1) and potassium 2-(7-bromo-9-(2-octyldodecyl)-9H-carbazole-2-yl) triolborate (M2), as an efficient and versatile approach for precisely synthesizing poly[9-(2-octyldodecyl)-3,6-carbazole] (3,6-PCz) and poly[9-(2-octyldodecyl)-2,7-carbazole] (2,7-PCz), respectively. The SCTP of triolborate-type carbazole monomers was performed in a mixture of THF/H2O using an initiating system consisted of 4-iodobenzyl alcohol, Pd2(dba)3•CHCl3, and t-Bu3P. In the SCTP of M1, cyclic by-product formation was confirmed, as reported for the corresponding pinacolboronate-type monomer. By optimizing the reaction temperature and reaction time, we successfully synthesized linear end-functionalized 3,6-PCz for the first time. The SCTP of M2 proceeded with almost no side reaction, yielding 2,7-PCz with a functional initiator residue at the α-chain end. Kinetic and block copolymerization experiments demonstrated that the SCTP of M2 proceeded in a chain-growth and controlled/living polymerization manner. This is a novel study on the synthesis of 2,7-PCz via SCTP. By taking advantage of the well-controlled nature of this polymerization system, we demonstrated the synthesis of high-molecular-weight 2,7-PCzs (Mn = 5–38 kg mol−1) with a relatively narrow ÐM (1.35–1.48). Furthermore, we successfully synthesized fluorene/carbazole copolymers as well as 2,7-PCz-containing diblock copolymers, demonstrating the versatility of the present polymerization system as a novel synthetic strategy for well-defined polycarbazole-based materials. We assess the effects of neutron irradiation on the deformation and stress evolution of large-scale fusion energy structures. This is accomplished through non-linear finite element structural analysis of the coupled thermal and mechanical fields at the Beginning-Of-Life (BOL), at 45 dpa, and at 90 dpa. Radiation effects include volumetric swelling and the influence of radiation on the mechanical properties. The system studied here is a large section of a full inboard module of an integrated structure comprising the First Wall and Blanket (FW/B) of a Dual Cooled Lithium-Lead (DCLL) energy conversion unit in the Fusion Nuclear Science Facility (FNSF). The structural material is the ferritic/martensitic steel F82H. We analyze several radiation effects phenomena that can lead to significant impact on the mechanical design and lifetime of the structure. These include volumetric swelling effects on the assembly and disassembly of modules and the impact of spatial gradients in neutron damage and helium generation on deformation, stress evolution, and plastic strain accumulation. We show that the stress state is strongly influenced by the accumulation of swelling strains, radiation hardening and softening, and by spatial gradients in displacement damage and helium gas generation. Several key regions in the FW/B structure are identified where severe plastic strains accumulate and may be potential sites for failure, especially during cyclic reactor operations. We finally show that minimum gaps between Inboard (IB) blanket sectors must be present during assembly so as to avoid excessive stress and strain in FW/B modules critical regions and raising chances of failure. A range of structural displacements are shown, from as low as 4 mm at BOL for free side walls to as high as 46 mm at 90 dpa for fully constrained side walls. [ABSTRACT FROM AUTHOR]

Our group has reported that poly(glycidyl ether) block copolymers exhibited complex temperature response in aqueous solution. In this study, we investigated the two chains of the block copolymer structure and configurations at different temperatures by using MD simulation. The results showed that at low temperatures, the polymer structure is linear and can form micelles, but above a certain temperature, the polymer structure changes to a U-shape and the micelle structure would be destroyed, which is responsible for the complex temperature response. We have investigated magnetic properties for reactor pressure vessel model alloys with variable Cu contents, subjected to neutron irradiation up to a fluence of 9 × 10 19 n cm − 2. Unlike a monotonic increase of microhardness with neutron fluence, the major-loop coercivity decreases at a higher fluence and the decrease becomes larger for the alloy containing a higher amount of Cu. The measurements of first-order reversal curves (FORCs) for the high-Cu alloy show that the position of the FORC distribution peak shifts toward a lower coercivity just after neutron irradiation, followed by a slight increase, associated with the broadening along both the coercivity and interaction field axes. The results can be explained by the enhancement of magnetic inhomogeneity in a matrix due to Cu precipitation and an increasing magnetostatic interaction between local magnetic regions with different coercivity. The magnetic method using FORCs can be a possible technique which provides in-depth information on microstructural changes due to neutron irradiation, which is not obtained by measurements of a conventional major hysteresis loop. Isolation of antigen (Ag)-specific T cells is an important step in the investigation of T-cell immunity. Activation-induced markers (AIMs), such as CD154/tumor necrosis factor (TNF)/CD107A/CD134/CD137 enable the sorting of Ag-specific T cells without using human leukocyte antigen (HLA)-multimers. However, optimal conditions suitable for simultaneous detection of both Ag-specific CD4 and CD8 T cells have not been investigated. Here, conditions were optimized to simultaneously detect the maximum number of activated CD4 and CD8 T cells in a TCR-dependent manner. First, the frequency of total pools of AIM-positive cells induced by superantigen, staphylococcal enterotoxin B (SEB), stimulation in various culture conditions was monitored and compared side-by-side. The total amount of AIM-positive CD4 T cells, but not CD8 T cells, was significantly abrogated by addition of brefeldin A. TNF-alpha converting enzyme inhibitor treatment effectively increased the TNF-positive population, without affecting other markers' positivity. AIM-positive CD4 T cells and CD8 T cells were detected at least 3 h after stimulation. Furthermore, examination of the multiple combination of each marker revealed that minimum contribution of CD134 on the total pool of AIM-positive cells at this setting, suggesting the essential and non-essential AIMs to maximize the detected number of AIM-positive cells. Taken together, this optimized method will be a useful tool for the simultaneous monitoring the T-cell receptor stimulation-dependent activation of CD4 and CD8 T cells using inducible markers on the cell surface including Ag-specific T cells. Interleukin-27 (IL-27) is a cytokine that suppresses human immunodeficiency virus (HIV)-1 infection in macrophages and is considered as an immunotherapeutic reagent for infectious diseases. It is reported that IL-27 suppresses autophagy in Mycobacterium tuberculosis-infected macrophages; however, a role for IL-27 on autophagy induction has been less studied. In this study, we investigated the impact of IL-27 in both autophagy induction and HIV-1 infection in macrophages. Primary human monocytes were differentiated into macrophages using human AB serum (huAB) alone, macrophage-colony stimulating factor (M-CSF) alone, or a combination of IL-27 with huAB or M-CSF. Electron microscopy and immunofluorescence staining demonstrated that a 20-fold increase in autophagosome formation was only detected in IL-27 + huAB-induced macrophages. Western blot analysis indicated that the autophagosome induction was not linked to either dephosphorylation of the mammalian target of rapamycin (mTOR) or lipidation of microtubule-associated protein 1A/1B-light chain 3 (LC3), an autophagosomal marker, implying that IL-27 can induce autophagy through a novel non-canonical pathway.
