1. Polydimethylsiloxane-coated macroporous silica adsorbent in thermal desorption gas chromatography

Abstract: Polydimethylsiloxane (PDMS)-coated macroporous silica (MPSi) particles were developed for the extraction of volatile organic compounds (VOCs) from air samples in thermal desorption-gas chromatographic analysis. The gas sample was collected by a gas sampling pump at 100 mL/min. Different amounts of PDMS coating were prepared, and the fundamental extraction and desorption performances were investigated using even numbered n-alkanes of C8–C28. The extraction efficiency increased by increasing PDMS coating, and the desorption efficiency showed more than 99% at a desorption temperature of 310°C for all the investigated adsorbents. This remarkable desorption performance showed a higher peak area of the analytes than that of the Tenax TA-packed thermal desorption tube, especially for higher molecular weight compounds. Furthermore, an activated carbon particle of Carbopack X was introduced on the latter side of the PDMS-MPSi to extract low-molecular weight compounds. This double-bed-type extraction tube showed a successful determination of a wide range of VOCs. The double-bed-type extraction tube was then applied to the extraction of VOCs related to sick building syndrome (SBS). The results for the extraction and desorption performances of the SBS-related VOCs were well agreed with that of alkanes, and the applicability of the extraction tube to the determination of other types of conventional VOCs were confirmed.

2. XAFS Analysis of Sulfuric Acid Solution Related to the Effect of Te and Sb in Suppressing the Disproportionation Reaction of Mn3+ in Mn/Ti Redox Flow Cells

Abstract: In sulfuric acid (H2SO4) aqueous solutions containing manganese (Mn) and titanium (Ti) ions, which were developed to be used for the positive electrolyte in redox flow (RF) batteries, the addition of tellurium (Te) or antimony (Sb) has been found very effective in suppressing the MnO2 precipitation in the charging process from Mn2+ to Mn3+. To investigate its mechanism, x-ray absorption near edge structure (XANES) measurements were carried out for cations in the solution samples at various state of charges (SOCs). In the case of the Te-added solutions, the change rate of Mn ion valence over the SOC was found to very low at the SOCs lower than 30 %. It was also indicated that the valence change of Te ions from Te4+ to Te6+ proceeded at the SOCs below 30 %. On the other hand, no significant change was observed in the valence of Ti at any SOCs. Similar trends were confirmed in the Sb-added solution samples. In conclusion, the supplied charges were considered to be used to increase not only the valence of Mn ions but also that of Te or Sb ions, which resulted in the suppression of the MnO2 precipitation.

3. Neurodevelopmental delay up to the age of 4 years in infants born to women with gestational diabetes mellitus: The Japan Environment and Children's Study

Abstract: Abstract Aims/Introduction This study aimed to investigate the neurodevelopment of infants born to women with gestational diabetes mellitus (GDM). Materials and Methods Data from the National Birth Cohort in the Japan Environment and Children's Study from 2011 to 2014 (n = 81,705) were used. Japan uses the GDM guidelines of the International Association of Diabetes and Pregnancy Study Groups. The Japanese translation of the Ages and Stages Questionnaires, third Edition, was used to assess neurodevelopment in the following domains: communication skills, gross motor skills, fine motor skills, problem‐solving ability, and personal and social skills. The survey was carried out every 6 months from the age of 6 months to 4 years (total of eight times). Generalized estimating equations were used to evaluate the association between maternal GDM and neurodevelopmental delay based on odds ratios (ORs) and 95% confidence intervals (95% CIs). Results Neurodevelopmental delays, particularly in problem‐solving ability, fine motor skills, and personal and social skills, were significantly higher in infants born to women with GDM than in those born to women without GDM (adjusted OR 1.24, 95% CI 1.12–1.36; adjusted OR 1.15, 95% CI 1.03–1.27; and adjusted OR 1.18, 95% CI 1.04–1.33). Furthermore, stratification showed no significant increase in the adjusted ORs (95% CIs) of girls. Conclusions Neurodevelopment was significantly delayed up to 4 years‐of‐age among boys born to women with GDM.

Abstract: Abstract Aims/Introduction We investigated the association between gestational diabetes mellitus (GDM) and perinatal outcomes stratified by pre‐pregnancy body mass index (BMI) and/or gestational weight gain (GWG). Materials and Methods Data from the national birth cohort in the Japan Environment and Children's Study from 2011 to 2014 (n = 85,228) were used. Japan uses the GDM guidelines of the International Association of Diabetes and Pregnancy Study Groups. The odds ratios (ORs) of perinatal outcomes were compared between women with and those without GDM. Results The OR (95% confidence interval) of having a small for gestational age infant in the GDM group with a pre‐pregnancy BMI of ≥25.0 kg/m2 and insufficient GWG (7.25 kg) was 2.04 (1.56–2.67). The OR of hypertensive disorders of pregnancy was higher in women with a BMI ≥18.5 kg/m2 in the GDM group than in the non‐GDM group. Conclusions Large for gestational age and hypertensive disorders of pregnancy were associated with pre‐pregnancy BMI and GWG in either normal weight or overweight/obese women, and the relationship was strengthened when GDM was present. Women with GDM and a BMI of ≥25.0 kg/m2 are at risk of having small for gestational age and large for gestational age infants depending on GWG.

7. XAFS Analysis of Sulfuric Acid Solution Related to the Effect of Bi in Suppressing the Disproportionation Reaction of Mn3+ in Mn/Ti Redox Flow Cells

Abstract: In sulfuric acid (H2SO4) aqueous solutions containing manganese (Mn) and titanium (Ti) ions, which are used for the positive electrolyte in redox flow (RF) batteries, the addition of a small quantity of bismuth (Bi) has been found to be very effective in suppressing the MnO2 precipitation in the charging process from Mn2+ to Mn3+. The effect of the addition of Bi on the structural change in these solutions has been analyzed using x-ray absorption fine structure (XAFS) techniques. X-ray absorption near edge structure (XANES) spectra indicated that some Mn4+ ions were generated only when Bi was not added. It was also confirmed that the Mn-O octahedral coordination was maintained at a high state of charge (SOC). On the other hand, no change was observed in the valence of Ti and Bi ions even at high SOCs. Extended x-ray absorption fine structure (EXAFS) measurements combined with fitting analysis clarified that the addition of Bi resulted in the generation of Mn-O-Bi coordination at the SOCs of 70 and 90 %. Bi-O-Mn coordination may suppress the MnO2 precipitation at a high SOC.

We found that the levels of circulating branched-chain amino acids (BCAA) and their cognate α-ketoacids (BCKA) were significantly correlated with the body weight in humans and mice and that BCAA catabolic defects in BAT were associated with obesity in diet-induced obesity (DIO) mice. Pharmacological systemic enhancement of BCAA catabolic activity reduced plasma BCAA and BCKA levels and protected against obesity; these effects were reduced in BATectomized mice. DIO mice gavaged with Bacteroides dorei and Bacteroides vulgatus exhibited improved BAT BCAA catabolism and attenuated body weight gain, which were not observed in BATectomized DIO mice. Our data have highlighted a possible link between the gut microbiota and BAT BCAA catabolism and suggest that Bacteroides probiotics could be used for treating obesity.

Abstract: Objectives We aimed to clarify the accuracy of pregnant women’s knowledge and understanding regarding infectious disease screening in early pregnancy and clarify the roles that should be played by health care providers in promoting the health of pregnant women and their children. Methods A cross-sectional questionnaire survey was conducted in 25 hospitals across Japan from May 2018 to September 2019. We compared the agreement rates regarding screening results for hepatitis B virus (HBV), hepatitis C virus (HCV), syphilis, human T-cell leukemia virus-1 (HTLV-1), and cervical cytology in the medical records and understanding of their results by pregnant women. We then investigated whether participants had knowledge regarding the risk of mother-to child transmission in these diseases and factors associated with their knowledge. Results We enrolled 2,838 respondents in this study. The rates of agreement for HBV and cervical cancer screening related to human papillomavirus infection were “substantial,” those for syphilis was “moderate,” and those for HCV and HTLV-1 were “fair,” according to the Kappa coefficient. The rate of knowledge regarding mother-to-child transmission of syphilis was highest (37.0%); this rate for the other items was approximately 30%. Increased knowledge was associated with higher educational level and higher annual income. Conclusions for Practice Pregnant women in Japan had generally good levels of understanding regarding their results in early-pregnancy infectious disease screening. However, they had insufficient knowledge regarding mother-to-child transmission of these diseases. Health care providers should raise awareness in infectious disease prevention among pregnant women and the general public, providing appropriate measures and implementing effective perinatal checkups and follow-ups for infectious diseases.

Abstract: Background: Abdominal aortic aneurysm (AAA) is a life-threatening cardiovascular disease characterized by dilated abdominal aorta. Immune cells have been shown to contribute to the development of AAA, and that the gut microbiota is associated with numerous diseases, including cardiovascular diseases, by regulating immune systems or metabolic pathways of the host. However, the interaction between the gut microbiota and AAA remains unknown. Methods: Apolipoprotein E–deficient male mice were fed a high-cholesterol diet and divided into three groups: the control group was maintained under normal water (control group), the oral AVNM group was maintained under drinking water supplemented with ampicillin, vancomycin, neomycin, and metronidazole, and the i.p. AVNM group was injected AVNM intraperitoneally. After 1 week of pretreatment with antibiotics, these mice were administrated Ang II via subcutaneous osmotic pumps for 4 weeks and euthanized to evaluate AAA formation. Results: Depletion of gut microbiota by oral AVNM ameliorated the incidence of AAAs (control group: 58.9% versus oral AVNM group: 28.6% versus i.p. AVNM group: 75.0%, P = 0.0005) and prevented death due to ruptured aneurysms (control group: 11% versus oral AVNM group: 0% versus i.p. AVNM group: 15%). Oral AVNM suppressed monocyte storage in the spleen, but not in other organs. Despite possessing a higher level of cholesterol, recruitment of monocytes into the suprarenal aorta was suppressed in the oral AVNM group. In AVNM drinking mice, NOD1 ligand, a kind of PRR ligands, increased the development of AAAs and accumulation of macrophages in the aortae. Conclusions: The gut microbiota plays a critical role in AAA formation. Therefore, regulation of the microbiota or the immune system can be a therapeutic approach for AAA.

Abstract: Gut microbial lipopolysaccharide (LPS) induces endotoxemia, an independent risk factor for cardiovascular disease (CVD). However, no studies have demonstrated how structural differences in each bacterial LPS contribute to endotoxemia. Here, we investigated the effects of different acyl chains in the lipid A moiety of LPS on endotoxemia and the subsequent immune response and atherosclerotic plaque formation.ApoeE. LPS administration induced endotoxemia, but B. LPS and saline did not. In E. LPS-treated mice, total plaque areas in the aortic root were significantly increased, and neutrophil accumulation and increased formation of neutrophil extracellular traps (NETs) were observed at the plaque lesions, but not in B. LPS-treated mice. A single dose of E. LPS significantly increased the accumulation of neutrophils in plaque lesions on day 3, and NET formation on day 7. E. LPS also increased interleukin-1 beta (IL-1β) production in plaque lesions on day 7. Furthermore, NET formation and IL-1β production were also observed in human coronary plaques.We identified a previously unknown link between structural differences in LPS and atherosclerosis. Lowering microbial LPS activity may reduce NET formation in plaques and prevent CVD progression.

Abstract: The best thromboprophylaxis for pregnant women with congenital antithrombin deficiency (CAD) is controversial.To clarify the effectiveness of a protocol for venous thromboembolism (VTE) prevention in pregnant women with CAD.Women at high risk of VTE were administered antithrombin concentrate and heparin after conception, whereas those at low risk of VTE were administered heparin alone until delivery. All women received antithrombin concentrate at delivery except for one who was diagnosed with CAD.Ten women had CAD, including one in the high-risk group and nine in the low-risk group. No women had VTE at delivery as per the protocol for VTE prevention. Almost all women had increased antithrombin activity before delivery followed by maintenance at ≥ 70% due to antithrombin concentrate administration. VTE prophylaxis during and after delivery was successful in all women with CAD. However, one woman in the low-risk group did not receive heparin and developed VTE induced by severe hyperemesis at 9 gestational weeks, before the diagnosis of CAD. Women in the high-risk group received antithrombin concentrate after delivery but had increased D-dimer levels at postpartum.Our protocol to prevent VTE in pregnant women with CAD is safe and effective.

Abstract: Aims/Introduction We investigated the association between gestational diabetes mellitus (GDM) and perinatal outcomes stratified by pre-pregnancy body mass index (BMI) and/or gestational weight gain (GWG). Materials and Methods Data from the national birth cohort in the Japan Environment and Children's Study from 2011 to 2014 (n = 85,228) were used. Japan uses the GDM guidelines of the International Association of Diabetes and Pregnancy Study Groups. The odds ratios (ORs) of perinatal outcomes were compared between women with and those without GDM. Results The OR (95% confidence interval) of having a small for gestational age infant in the GDM group with a pre-pregnancy BMI of >= 25.0 kg/m(2) and insufficient GWG (7.25 kg) was 2.04 (1.56-2.67). The OR of hypertensive disorders of pregnancy was higher in women with a BMI >= 18.5 kg/m(2) in the GDM group than in the non-GDM group. Conclusions Large for gestational age and hypertensive disorders of pregnancy were associated with pre-pregnancy BMI and GWG in either normal weight or overweight/obese women, and the relationship was strengthened when GDM was present. Women with GDM and a BMI of >= 25.0 kg/m(2) are at risk of having small for gestational age and large for gestational age infants depending on GWG.

Abstract: An analytical method using a porous polytetrafluoroethylene (PTFE) membrane that passes through gas while repelling water, and a needle-type extraction device was developed for purge-and-trap (PT) extraction followed by gas chromatography–mass spectrometric (GC–MS) determination of volatile organic compounds (VOCs) in water samples. The PTFE membrane was fixed in a solid-phase extraction glass cartridge, and 10 mL of water sample and sodium chloride were added. Then, a needle-type extraction device was inserted into the cartridge headspace, and the headspace gas was collected. Due to decompression on the inside of the cartridge, purge gas was introduced from the bottom side of the cartridge through the PTFE membrane. After the sample extraction, VOCs were thermally desorbed by inserting the needle into the heated GC injection port. The limit of quantification of the proposed method for 23 investigated VOCs was less than 0.5 μg/L, and the upper limit of the calibration curve was 500 μg/L for dichloromethane and 1000 μg/L for other compounds. The relative standard deviations of the relative peak area for 23 investigated VOCs were less than 15% (n = 5). The method was used to determine VOCs in tap water samples.

20. Relationship between out-of-facility deliveries and distance and travel time to delivery facilities in Hokkaido, Japan: An ecological study

Abstract: This study aimed to investigate the relationship between the distance and travel time from each municipality to the nearest delivery facilities in the other municipalities and the frequency of out-of-facility deliveries in Hokkaido.Vital statistics from 2016 to 2020 were used. For municipalities without delivery facilities, the distance and travel time from the town office of each municipality to the nearest delivery facility was measured using Google maps. Negative binomial regression with an offset term was used to calculate the relative risks (RRs) and 95% confidence intervals (CIs) of out-of-facility delivery for distance (30, 30-59, ≥60 km), and travel time by car (30, 30-59, and ≥60 min) from the town office to the nearest delivery facility compared with the presence of delivery facilities.The overall rate of out-of-facility deliveries in Hokkaido was 2.1‰; in municipalities with delivery facilities, 1.8‰, and in municipalities without delivery facilities, 3.1‰. The adjusted RRs (95% CIs) for out-of-facility deliveries were significantly higher in municipalities with less than 30 km and travel time of less than 30 min to delivery facilities, 2.63 (1.34-5.17) and 2.76 (1.36-5.58), respectively, compared to municipalities with delivery facilities. However, the adjusted RR of out-of-facility delivery for municipalities ≥30 km was higher, although the difference was not significant.Even in municipalities with a distance to delivery facilities of less than 30 km or travel time of less than 30 min, we should keep in mind the occurrence of out-of-facility deliveries.

22. Relationship Between Plasma Lipopolysaccharides, Gut Microbiota, and Dementia: A Cross-Sectional Study

Abstract: Background: Previous studies have demonstrated associations between gut microbiota, microbial metabolites, and cognitive decline. However, relationships between these factors and lipopolysaccharides (LPS; molecules of the outer membrane of gram-negative bacteria) remain controversial. Objective: To evaluate associations between plasma LPS, gut microbiota, and cognitive function. Methods: We performed a cross-sectional sub-analysis of data of 127 participants (women: 58%, mean age: 76 years) from our prospective cohort study regarding the relationship between gut microbiota and cognitive function. We enrolled patients who visited our memory clinic and assessed demographics, dementia-related risk factors, cognitive function, brain imaging, gut microbiomes, and microbial metabolites. We evaluated relationships between cognitive decline and plasma LPS using multivariable logistic regression analyses. Results: Plasma LPS concentration increased with increasing degree of cognitive decline and total cerebral small vessel disease (SVD) score (Kruskal-Wallis test; p = 0.016 and 0.007, respectively). Participants with high plasma LPS concentrations tended to have lower concentrations of gut microbial metabolites, such as lactic acid and acetic acid, and were less likely to consume fish and shellfish (44.7% versus 69.6%, p = 0.027) than those with low plasma LPS concentrations. Multivariable analyses revealed that plasma LPS concentration was independently associated with the presence of mild cognitive impairment in participants without dementia (odds ratio: 2.09, 95% confidence interval: 1.14–3.84, p = 0.007). Conclusion: In this preliminary study, plasma LPS concentration was associated with both cognitive decline and cerebral SVD and significantly correlated with beneficial gut microbial metabolites. Plasma LPS may be a risk factor for cognitive decline.

24. Decrease Rate of Platelet Count as a Marker for Complications of Preeclampsia: A Cross-Sectional Study

Abstract: BackgroundThrombocytopenia is considered a sign of maternal organ damage of preeclampsia. However, platelet count distribution during pregnancy shifts downward even in uncomplicated women, and platelet counts in some uncomplicated women falls below the cutoff value of thrombocytopenia. Thus, not only absolute platelet counts number but also platelet count reduction rate may relate with adverse outcomes of preeclampsia. This study aimed to evaluate whether the overall platelet count reduction rate was associated with adverse outcomes of preeclampsia. MethodsThis retrospective study included 240 singleton pregnant women with preeclampsia. Overall platelet count reduction ≥30.0% was used as a cut-off value because 10–20% platelet counts reduction can be observed in uncomplicated pregnancy. The association between the overall platelet count reduction from the first trimester ≥30.0% and adverse outcomes; severe hypertension, uteroplacental dysfunction, maternal organ damage except for thrombocytopenia, and preterm delivery ResultsA total of 95/240 (39.6%) of women had ≥30.0% of overall platelet count reduction from the first trimester. The incidences of maternal organ damage except for thrombocytopenia (12.4%, 22.1%; p = 0.047), and preterm delivery p = 0.005) were higher in women with ≥30.0% of overall platelet count reduction than those otherwise. Even in women without thrombocytopenia (platelet counts ≥150 × 109/L), severe hypertension (66.2%, 85.4%; p = 0.018), and preterm delivery p = 0.017) were more frequently observed in women with ≥30.0% of overall platelet count reduction than those otherwise.ConclusionsA ≥30.0% of overall platelet count reduction from the first trimester was associated with adverse outcomes of preeclampsia even in women without thrombocytopenia.

26. Needle extraction device for rapid and quantitative gas chromatographic determination of volatile chlorinated hydrocarbons and benzene in soil

Abstract: A rapid and quantitative analytical method for volatile chlorinated hydrocarbons and benzene, such as chloroethylene, dichloromethane, and carbon tetrachloride, in soil samples was developed using a needle-type extraction device packed with activated carbon-based adsorbents. The soil sample of 10 g was placed in a glass cartridge and fixed with quartz wool to make sure not to move during the extraction process. After the internal standard solution was spiked into the soil sample, the cartridge was sealed with a polytetrafluoroethylene adapter, and the needle extraction device was used to collect the headspace gas. Analytes were purged from the soil sample and collected on the adsorbent, while pure nitrogen gas was introduced to the soil sample during the sample collection. The sampling time was approximately 10 min for 100 mL of gas sampling. After the sampling process, the extraction needle was directly inserted into a heated injection port of gas chromatography equipped with flame ionization detector (GC-FID) or that hyphenated to mass spectrometry (GC-MS). The extracted VOCs were thermally desorbed and simultaneously introduced to a GC capillary column for subsequent separation and determination. The limit of detection and limit of quantification of all the examined analytes was less than 20 and 60 ng/10 g soil in GC-FID and less than 2.5 and 7.5 ng/10 g soil in the case of GC-MS. Linear calibration curves were obtained up to 10 μg/10 g soil for all the analytes. The relative standard deviations of the ratio of analyte peak area and internal standard peak area were less than 10%. The recovery and sensitivity of analytes confirmed in the proposed method were compared with that obtained with a standard method conventionally employed.

Abstract: Non-destructive depth profile evaluation of multi-layer thin film stacks using simultaneous analysis of angle-resolved X-ray photoelectron spectroscopy data from multiple instruments is demonstrated. The data analysis algorithm, called the maximum smoothness method, was originally designed to handle data from a single XPS instrument with a single X-ray energy; in this work, the algorithm is extended to provide a simultaneous analysis tool which can handle data from multiple instruments with multiple X-ray energies. The analysis provides depth profiles for multilayer stacks that cannot be obtained by conventional data analysis methods. 29. XPS and HAXPES analyses for pre-sputtered InP surface and InP/Pt interface

Abstract: The state of the pre-sputtered indium phosphide (InP) surface was analyzed by X-ray photoelectron spectroscopy, employing synchrotron-based relatively low-energy X-ray. It was found that the pre-sputtering treatment induced the phosphorus vaporization and made the surface composition In-rich, which was thought to promote oxidation of InP surface in atmosphere. The state of the interface between InP and Pt was also investigated nondestructively, by using hard X-ray photoemission spectroscopy. As a result, it was demonstrated that the interfacial layer was composed of the native oxide (In–O, P–O) and metallic state (In–Pt or In–In, P–P) and that the pre-sputtering treatment increased significantly the amount of the P–O, In–Pt or In–In, and P–P. From a simplified calculation, assuming a Pt/In–Pt/In–P layer stacking structure and neglecting the In–O and P–O components, the thickness of the interfacial layer was estimated to be approximately 3.0 nm.

Abstract: Summary The gut microbiome has emerged as a key regulator of obesity; however, its role in brown adipose tissue (BAT) metabolism and association with obesity remain to be elucidated. We found that the levels of circulating branched-chain amino acids (BCAA) and their cognate α-ketoacids (BCKA) were significantly correlated with the body weight in humans and mice and that BCAA catabolic defects in BAT were associated with obesity in diet-induced obesity (DIO) mice. Pharmacological systemic enhancement of BCAA catabolic activity reduced plasma BCAA and BCKA levels and protected against obesity; these effects were reduced in BATectomized mice. DIO mice gavaged with Bacteroides dorei and Bacteroides vulgatus exhibited improved BAT BCAA catabolism and attenuated body weight gain, which were not observed in BATectomized DIO mice. Abstract: PURPOSE/OBJECTIVE(S) While the non-inferiority of hypofractionation (HF) to conventional fractionation for prostate cancer had been investigated in some randomized clinical trials, we started a multi-institutional phase II study to confirm the safety with the efficacy of HF intensity-modulated radiotherapy (IMRT) with image-guided technique for Japanese patients. MATERIALS/METHODS Eligibility criteria of the study included: pathologically proven adenocarcinoma; low or intermediate risk (cT1-T2c, PSA ≤ 20 and Gleason score (GS) ≤ 7) or high risk having one of the following factors, cT3a, PSA > 20/ ≤ 30, or GS = 8/9; cN0M0; age 50-80; performance status (PS) 0-1; written informed consent. HF IMRT with 70 Gy/28 fr (2.5 Gy/fr) was performed. The position of the prostate gland or inserted fiducial markers was verified with a full bladder before each treatment. Neoadjuvant hormonal therapy (HT) for 4-8 months was allowed for patients with intermediate or high risk. The GTV was defined as the prostate (+ extracapsular invasion for cT3a). The CTV was defined as the GTV for low risk, and the GTV plus proximal seminal vesicles for intermediate and high risk. The PTV was defined as the CTV plus 4-8 mm margins for organ motions and set-up errors. The bladder, rectum, small and large intestines, and femoral heads were defined as the organs at risk (OAR) as the solid organ. The dose prescription was D50% for the PTV, with Dmax ≤ 105% and Dmin ≥ 95%. The dose constraints for OARs were defined as follows: bladder V70/V65/V60/V55, 15%/25%/35%/50%; rectum V65/V55/V50/V45, 15%/25%/35%/50%; small intestine V55 < 0.5 cc; large intestine V60 < 0.5 cc; femoral heads Vmax ≤ 50 Gy. The primary endpoint was the incidence of late toxicities at 5 years. The secondary endpoints were the incidence of acute toxicities, biochemical failure-free, clinical failure-free, and overall survivals at 5 years. The sample size was calculated to 130 patients using alpha/beta errors of 0.05/0.2, with the threshold/expectation values of ≥ grade 2 late toxicities as 15%/7%. RESULTS From Jul/2012 to Oct/2014, 134 patients from 20 institutions were registered. Patients and tumor characteristics were as follows: median age 71 (54 - 79); cT1c/T2a/T2b/T2c/T3a, 53/50/7/19/5; GS 6/7/8/9, 32/80/16/6; median pretreatment PSA 9.0 ng/ml (3.15 - 28.6); low/intermediate/high risk, 20 (15%)/80 (60%)/34 (25%); PS 0/1, 67/67. HT was used for 121 patients (90%). The median follow-up time was 5.16 years. The incidences of grade 2/ ≥ 3 gastrointestinal and genitourinary late toxicities at 5 years (primary endpoint) were 5.3%/0% and 5.3%/0%, respectively. The corresponding figures of acute toxicities were 3.8%/0% and 14.5%/0%, respectively. The biochemical failure-free, clinical failure-free, and overall survivals at 5 years were 90.4%, 93.8%, and 94.5%, respectively. CONCLUSION The primary endpoint was met, and the safety and efficacy of HF IMRT was confirmed in this trial, and accepted as one of the standard of care for Japanese patients with prostate cancer.

However, AT(N) profiles may vary depending on the markers chosen and the target population.Methods We stratified 177 individuals who participated in the Japanese Alzheimer’s Disease Neuroimaging Initiative by AT(N) classification according to cerebrospinal fluid (CSF) biomarkers. We compared the frequency of AT(N) profiles between the classification using total tau and neurofilament light chain (NfL) as N markers (AT(N)tau and AT(N)NfL). Baseline characteristics, and longitudinal biological and clinical changes were examined between AT(N) profiles.Results We found that 9% of cognitively unimpaired subjects, 49% of subjects with mild cognitive impairment, and 61% of patients with Alzheimer’s disease (AD) dementia had the biological AD profile (ie, A+T+) in the cohort. The frequency of AT(N) profiles substantially differed between the AT(N)tau and AT(N)NfL classifications. When we used t-tau as the N marker (AT(N)tau), those who had T− were more frequently assigned to (N)−, whereas those who had T+were more frequently assigned to (N)+ than when we used NfL as the N marker (AT(N)NfL). During a follow-up, the AD continuum group progressed clinically and biologically compared with the normal biomarker group in both the AT(N)tau and AT(N)NfL classifications. More frequent conversion to dementia was observed in the non-AD pathological change group in the AT(N)tau classification, but not in the AT(N)NfL classification.Conclusions AT(N)tau and AT(N)NfL in CSF may capture different aspects of neurodegeneration and provide a different prognostic value. The AT(N) classification aids in understanding the AD continuum biology in various populations.
