As a new tumor therapeutic strategy, adaptive therapy involves utilizing the competition between cancer cells to suppress the growth of drug-resistant cells, maintaining a certain tumor burden. However, it is difficult to determine the appropriate time and drug dose. In this paper, we consider the competition model between drug-sensitive cells and drug-resistant cells, propose the problem of drug concentration, and provide two state constraints: the upper limit of the maximum allowable drug concentration and the tumor burden. Using relevant theories, we propose the best treatment strategy. Through a numerical simulation and quantitative analysis, the effects of drug concentrations and different tumor burdens on treatments are studied, and the effects of cell-to-cell competitive advantage on cell changes are taken into account. The clinical dose titration method is further simulated; the results show that our therapeutic regimen can better suppress the growth of drug-resistant cells, control the tumor burden, limit drug toxicity, and extend the effective treatment time. It is of critical importance to comprehend the biological environment and core tumor populations when trying to design successful therapeutic solutions for fighting cancers. In several diseases, G9a has been recognized as a novel epigenetic therapeutic target, and its blockage can shift tumor cells (TCs) toward tumor propagating cells (TPCs). This study combines mathematical modeling based on ordinary differential equations and dynamical analysis to quantitatively and qualitatively understand the interactions among G9a, TCs, and TPCs, denoted as G9a-TC-TPC. We propose four different dynamical systems with the impact of the strong Allee effect, named the Hill–Hill system, Logistic–Logistic system, Hill–Logistic system and Logistic–Hill system, to simulate different biological processes through the Hill functions and the Logistic functions that are often used in the models of biological systems. Based on theoretical analysis of these models, including the positivity, boundedness and stability of equilibria, we find that the Hill–Logistic system can display bistable states that correspond to the wild-type tumors and the aggressive tumors. Consequently, we use bifurcation analysis and numerical simulations to illustrate the complicated dynamical behavior of this system. It has been shown that under a specific therapy that changes the relative apoptotic rate of TCs (G9a suppresses the apoptosis of TCs), which can affect the bistability and instability of the system, the wide-type state can be obtained. We also discover that the relative handling time of TCs and TPCs can cooperatively enhance bistability, whereas the cooperative coefficient of feedback can contribute to all tumor cells moving from high-level monostability to bistable states in a restricted region, then to low-level stable states. Tumor is a complex and aggressive type of disease that poses significant health challenges. Understanding the cellular mechanisms underlying its progression is crucial for developing effective treatments. In this study, we develop a novel mathematical framework to investigate the role of cellular plasticity and heterogeneity in tumor progression. By leveraging temporal single-cell data, we propose a reaction-convection-diffusion model that effectively captures the spatiotemporal dynamics of tumor cells and macrophages within the tumor microenvironment. Through theoretical analysis, we obtain the estimate of the pulse wave speed and analyze the stability of the homogeneous steady state solutions. Notably, we employe the AddModuleScore function to quantify cellular plasticity. One of the highlights of our approach is the introduction of pulse wave speed as a quantitative measure to precisely gauge the rate of cell phenotype transitions, as well as the novel implementation of the high-plasticity cell state/low-plasticity cell state ratio as an indicator of tumor malignancy. Furthermore, the bifurcation analysis reveals the complex dynamics of tumor cell populations. Our extensive analysis demonstrates that an increased rate of phenotype transition is associated with heightened malignancy, attributable to the tumor's ability to explore a wider phenotypic space. The study also investigates how the proliferation rate and the death rate of tumor cells, phenotypic convection velocity, and the midpoint of the phenotype transition stage affect the speed of tumor cell phenotype transitions and the progression to adenocarcinoma. Radiomics, providing quantitative data extracted from medical images, has emerged as a critical role in diagnosis and classification of diseases such as glioma. One main challenge is how to uncover key disease-relevant features from the large amount of extracted quantitative features. Many existing methods suffer from low accuracy or overfitting. We propose a new method, Multiple-Filter and Multi-Objective-based method (MFMO), to identify predictive and robust biomarkers for disease diagnosis and classification. This method combines a multi-filter feature extraction with a multi-objective optimization-based feature selection model, which identifies a small set of predictive radiomic biomarkers with less redundancy. Taking magnetic resonance imaging (MRI) images-based glioma grading as a case study, we identify 10 key radiomic biomarkers that can accurately distinguish low-grade glioma (LGG) from high-grade glioma (HGG) on both training and test datasets. Using these 10 signature features, the classification model reaches training Area Under the receiving operating characteristic Curve (AUC) of 0.96 and test AUC of 0.95, which shows superior performance over existing methods and previously identified biomarkers.

Cancer is a serious threat to human health and life. Using anti-tumor drugs is one of the important ways for treating cancer. A large number of experiments have shown that the hormesis appeared in the dose-response relationship of various anti-tumor drugs. Modeling this phenomenon will contribute to finding the appropriate dose. However, few studies have used dynamical models to quantitatively explore the hormesis phenomenon in anti-tumor drug dose-response. In this study, we present a mathematical model and dynamical analysis to quantify hormesis of anti-tumor drugs and reveal the critical threshold of antibody dose. Firstly, a dynamical model is established to describe the interactions among tumor cells, natural killer cells and M2-polarized macrophages. Model parameters are fitted through the published experimental data. Secondly, the positivity of solution and bounded invariant set are given. The stability of equilibrium points is proved. Thirdly, through bifurcation analysis and numerical simulations, the hormesis phenomenon of low dose antibody promoting tumor growth and high dose antibody inhibiting tumor growth is revealed. Furthermore, we fit out the quantitative relationship of the dose-response of antibodies. Finally, the critical threshold point of antibody dose changing from promoting tumor growth to inhibiting tumor growth is obtained. These results can provide suggestions for the selection of appropriate drug dosage in the clinical treatment of cancer.
