1. When Precedents Clash

Abstract

Consistency of case bases is a way to avoid the problem of retrieving conflicting constraining precedents for new cases to be decided. However, in legal practice the consistency requirements for case bases may not be satisfied. As pointed out in (Broughton 2019), a model of precedential constraint should take into account the hierarchical structure of the specific legal system under consideration and the temporal dimension of cases. This article continues the research initiated in (Liu et al. 2022; Di Florio et al. 2023), which established a connection between Boolean classifiers and legal case-based reasoning. On this basis, we enrich the classifier models with an organisational structure that takes into account both the hierarchy of courts and which courts issue decisions that are binding/constraining on subsequent cases. We focus on common law systems. We also introduce a temporal relation between cases. Within this enriched framework, we can formalise the notions of overruled cases and cases decided per incuriam: such cases are not to be considered binding on later cases. Finally, we show under which condition principles based on the hierarchical structure and on the temporal dimension can provide an unambiguous decision-making process for new cases in the presence of conflicting binding precedents., Comment: 13 pages. Abstract

We consider time dependently perturbed quantum harmonic oscillators in $\mathbb{R}^2$: $$ {\rm i} \partial_t u=\frac12(-\partial_{x_1}^2-\partial_{x_2}^2 + x_1^2+x_2^2)u +V(t, x, D)u, \qquad \ x \in \mathbb{R}^2, $$ where $V(t, x, D)$ is a selfadjoint pseudodifferential operator of degree zero, $2\pi$ periodic in time. We identify sufficient conditions on the principal symbol of the potential $V(t, x, D)$ that ensure existence of solutions exhibiting unbounded growth in time of their positive Sobolev norms and we show that the class of symbols satisfying such conditions is generic in the Fr\'echet space of classical $2\pi$- time periodic symbols of order zero. To prove our result we apply the abstract Theorem of arXiv:2101.09055v1 : the main difficulty is to find a conjugate operator $A$ for the resonant average of $V(t,x, D)$. We construct explicitly the symbol of the conjugate operator $A$, called escape function, combining techniques from microlocal analysis, dynamical systems and contact topology.

This paper examines how a notion of stable explanation developed elsewhere in Defeasible Logic can be expressed in the context of formal argumentation. With this done, we discuss the deontic meaning of this reconstruction and show how to build from argumentation neighborhood structures for deontic logic where this notion of explanation can be characterised. 9. Sorafenib maintenance after allogeneic stem cell transplantation in patients with FLT3+ AML receiving midostaurin during induction and consolidation: a retrospective analysis

Abstract

IntroductionAcute myeloid leukemia (AML) relapse is the main cause of death after allogeneic stem cell transplant (allo-SCT). In AML FLT3+, it was shown that Sorafenib used as maintenance therapy after allo-SCT, significantly reduces the risk of relapse and death. MethodsWe analyzed 29 adult patients with FLT3m AML and underwent allogeneic stem cell transplant from 2019 to 2023. All patients received midostaurin plus conventional CT during induction and consolidation. After transplantation, Sorafenib maintenance was administered in all patients independently from MRD status at transplantation. ResultsSorafenib maintenance was applied in 18 patients out 29 patients (62%). Median time to start sorafenib was 100 days (range 37-225) and median duration of treatment was 775 days (range 140-1064). For the whole population (n=29), 2-year OS, LFS, and CIR was 76%, 68% and 28%, respectively. The median time to relapse was 137 days (range 49-246). For patients treated with sorafenib (n=18), the 2-year OS, LFS, and CIR were 94%, 84% and 11%, respectively. For the whole population, the 100-day NRM was 0% and 1-year NRM was 3%. Death was caused by transplant-associated thrombotic microangiopathy in 1 patient. For patients who were administered with Sorafenib, the 1-y NRM was 5%. Death was caused by transplant associated transplant-associated thrombotic microangiopathy. DiscussionThis retrospective study suggests that sorafenib maintenance seem to be effective even in patients pre-treated with midostaurin.

10. Validation of a PD-1/CD28 chimeric switch receptor to augment CAR-T function in dogs with spontaneous B cell lymphoma

Abstract

Summary: Chimeric antigen receptor (CAR) T cell therapy has achieved unprecedented clinical outcomes in patients with relapsed/refractory B cell leukemias; however, response rates in patients with large B cell lymphoma (LBCL) are less impressive. Expression of PD-1 on activated T cells and PD-L1 on malignant, stromal, and immune cells within the tumor microenvironment (TME) contribute to CAR-T exhaustion, hypofunction, and treatment failures. Here, a comparative approach is taken to develop a chimeric switch receptor (CSR) with potential to augment CAR-T persistence, function, and clinical efficacy in immune competent, pet dogs with spontaneous B cell lymphoma (BCL). We show that similar to human CAR-T cells, expression of a PD-1/CD28 CSR in canine CAR-T cells results in enhanced function against PD-L1+ targets and preserves central memory phenotype. We also demonstrate that these effects depend upon active CSR signaling. This work paves the way for in vivo studies in canine BCL patients to inform human trial design.

14. Modelling and Explaining Legal Case-based Reasoners through Classifiers

Abstract

This paper brings together two lines of research: factor-based models of case-based reasoning (CBR) and the logical specification of classifiers. Logical approaches to classifiers capture the connection between features and outcomes in classifier systems. Factor-based reasoning is a popular approach to reasoning by precedent in AI & Law. Horty (2011) has developed the factor-based models of precedent into a theory of precedential constraint. In this paper we combine the modal logic approach (binary-input classifier, BLC) to classifiers and their explanations given by Liu & Lorini (2021) with Horty's account of factor-based CBR, since both a classifier and CBR map sets of features to decisions or classifications. We reformulate case bases of Horty in the language of BCL, and give several representation results. Furthermore, we show how notions of CBR, e.g. reason, preference between reasons, can be analyzed by notions of classifier system., Comment: 16 pages

Abstract

The use of meta-rules in logic, i.e., rules whose content includes other rules, has recently gained attention in the setting of non-monotonic reasoning: a first logical formalisation and efficient algorithms to compute the (meta)-extensions of such theories were proposed in Olivieri et al (2021) This work extends such a logical framework by considering the deontic aspect. The resulting logic will not just be able to model policies but also tackle well-known aspects that occur in numerous legal systems. The use of Defeasible Logic (DL) to model meta-rules in the application area we just alluded to has been investigated. Within this line of research, the study mentioned above was not focusing on the general computational properties of meta-rules. This study fills this gap with two major contributions. First, we introduce and formalise two variants of Defeasible Deontic Logic with Meta-Rules to represent (1) defeasible meta-theories with deontic modalities, and (2) two different types of conflicts among rules: Simple Conflict Defeasible Deontic Logic, and Cautious Conflict Defeasible Deontic Logic. Second, we advance efficient algorithms to compute the extensions for both variants.

16. Avoiding Pragmatic Oddity: A Bottom-up Defeasible Deontic Logic

Abstract

This paper presents an extension of Defeasible Deontic Logic to deal with the Pragmatic Oddity problem. The logic applies three general principles: (1) the Pragmatic Oddity problem must be solved within a general logical treatment of CTD reasoning; (2) non-monotonic methods must be adopted to handle CTD reasoning; (3) logical models of CTD reasoning must be computationally feasible and, if possible, efficient. The proposed extension of Defeasible Deontic Logic elaborates a preliminary version of the model proposed by Governatori and Rotolo (2019). The previous solution was based on particular characteristics of the (constructive, top-down) proof theory of the logic. However, that method introduces some degree of non-determinism. To avoid the problem, we provide a bottom-up characterisation of the logic. The new characterisation offers insights for the efficient implementation of the logic and allows us to establish the computational complexity of the problem.

17. Acartia spp. (Copepoda: Calanoida) as model organisms to evaluate the toxicity of emerging contaminants : an ecotoxicogenomic approach

Abstract

Copepods are small crustaceans of great ecological and ecotoxicogenomic importance. Among them, Acartia tonsa, invasive species in the Mediterranean Sea, is a model species in ecotoxicology. Acartia clausi, native of the Mediterranean, is a candidate alternative model species in this field. In this PhD project, we compared physiological (naupliar immobilisation, egg hatching success, egg production, faecal pellet production, adult survival) and molecular (quantitative gene expression) responses of both copepod species exposed to nickel chloride (NiCl₂) and nickel nanoparticles (NiNPs), through ecotoxicological bioassays and RT-qPCR. A de novo transcriptome of A. clausi females exposed to elutriates of polluted sediments was also generated. We were able to keep multi-generational cultures of A. clausi, fed the optimal diet of Rhinomonas reticulata, for up to four months, with 26 days from eggs to F2 generation. Bioassays confirmed that nauplii of A. clausi were more sensitive than those of A. tonsa, approximately 6 (NiNPs) and 95 times (NiCl₂). Adults of both species were less affected by these toxicants, with mild effects mostly on egg production. Effects of both toxicants on egg hatching success was mainly due to Ni ions, which are continuously released from NiNPs. The first de novo assembled transcriptome for A. clausi exposed to elutriates of polluted sediments revealed 1,000 differentially expressed genes (743 up-, 257 down-regulated). The response was mostly in up-regulation of proteolysis and detoxification, and down-regulation of ribosomal proteins. Comparative gene expression in copepod females showed greater responses in A. clausi, especially with NiCl₂, mostly with a down-regulation of genes involved in detoxification, indicating an inhibition of molecular defences. For A. clausi, down-regulation of vitellogenin seems correlated with low egg production. Overall, we provided novel information on the physiology and molecular biology of A. clausi and on NiNPs behaviour, which could threaten marine environment in the near future.

Abstract Tracheal pooling for Mycoplasma hyopneumoniae (M. hyopneumoniae) DNA detection allows for decreased diagnostic cost, one of the main constraints in surveillance programs. The objectives of this study were to estimate the sensitivity of pooled-sample testing for the detection of M. hyopneumoniae in tracheal samples and to develop probability of M. hyopneumoniae detection estimates for tracheal samples pooled by 3, 5, and 10. A total of 48 M. hyopneumoniae PCR-positive field samples were pooled 3-, 5-, and 10-times using field M. hyopneumoniae DNA-negative samples and tested in triplicate. Abstract

Summary: Aberrant male germline development can lead to the formation of seminoma, a testicular germ cell tumor. Seminomas are biologically similar to primordial germ cells (PGCs) and many bear an isochromosome 12p [i(12p)] with two additional copies of the short arm of chromosome 12. By mapping seminoma transcriptomes and open chromatin landscape onto a normal human male germline trajectory, we find that seminoma resembles premigratory/migratory PGCs; however, it exhibits enhanced germline and pluripotency programs and upregulation of genes involved in apoptosis, angiogenesis, and MAPK/ERK pathways. Using pluripotent stem cell-derived PGCs from Pallister-Killian syndrome patients mosaic for i(12p), we model seminoma and identify gene dosage effects that may contribute to transformation. As murine seminoma models do not exist, our analyses provide critical insights into genetic, cellular, and signaling programs driving seminoma transformation, and the in vitro platform developed herein permits evaluation of additional signals required for seminoma tumorigenesis.

Abstract

Abstract The increasing prevalence of chronic diseases is a driver for emerging big data technologies for healthcare including digital platforms for data collection, systems for active patient engagement and education, therapy specific predictive models, optimized patient pathway models. Powerful bioelectronic medicine tools for data collection, analysis and visualization allow for joint processing of large volumes of heterogeneous data, which in turn can produce new insights about patient outcomes and alternative interpretations of clinical patterns that can lead to implementation of optimized clinical decisions and clinical patient pathway by healthcare professionals. With this perspective, we identify innovative solutions for disease management and evaluate their impact on patients, payers and society, by analyzing their impact in terms of clinical outcomes (effectiveness, safety, and quality of life) and economic outcomes (cost-effectiveness, savings, and productivity). As a result, we propose a new approach based on the main pillars of innovation in the disease management area, i.e. progressive patient care models, patient-centric approaches, bioelectronics for precise medicine, and lean management that, combined with an increase in appropriate private-public-citizen-partnership, leads towards Patient-Centric Healthcare.
