Abstract: Chickens fed 200ppb aflatoxin from 10 days of age were evaluated for their immune response to a modified live infectious laryngotracheitis vaccine. Vaccination was administered at age 4 and 12 weeks. Antibody titers to the vaccine were reduced in chickens given dietary aflatoxin. After 7 weeks, aflatoxin feeding was continued for one month in a treated group and was withdrawn in another. Serology indicated significant differences between the two treated groups relative to whether aflatoxin was fed or not. Significant reduction in body weights, antibody titers and elevated SGOT and SGPT levels were found in chickens treated with aflatoxin. The impact of aflatoxin on reduced body weight, decreased SGOT and SGPT levels and lower antibody titers was shown to be significant in the treated group fed on a ration of aflatoxin until throughout the experiment. Clathrin-coated vesicles (CCVs) play critical roles in multiple cellular processes, including nutrient uptake, endosome/lysosome biogenesis, pathogen invasion, regulation of signalling receptors, etc. Saccharomyces cerevisiae Ent5 ( Sc Ent5) is one of the two major adaptors supporting the CCV-mediated TGN/endosome traffic in yeast cells. However, the classification and phosphoinositide binding characteristic of Sc Ent5 remain elusive. Here we report the crystal structures of the Sc Ent5 N-terminal domain, and find that Sc Ent5 contains an insertion α′ helix that does not exist in other ENTH or ANTH domains. Furthermore, we investigate the classification of Sc Ent5-N 31−191 by evolutionary history analyses and structure comparisons, and find that the Sc Ent5 N-terminal domain shows different phosphoinositide binding property from r Epsin1 and r CALM. The versatility of Hsp90 can be attributed to the variety of co-chaperone proteins that modulate the role of Hsp90 in many cellular processes. As a co-chaperone of Hsp90, Cpr7 is essential for accelerating the cell growth in an Hsp90-containing trimeric complex. Here, we report the crystal structure of Cpr7 at a resolution of 1.8 Å. It consists of an N-terminal PPI domain and a C-terminal TPR domain, and exhibits a U-shape conformation. Our studies revealed the aggregation state of Cpr7 in solution and the interaction properties between Cpr7 and the MEEVD sequence from the C-terminus of Hsp90. In addition, the structure and sequence analysis between Cpr7 and homologues revealed the structure basis both for the function differences between Cpr6 and Cpr7 and the functional complements between Cns1 and Cpr7. Our studies facilitate the understanding of Cpr7 and provide decent insights into the molecular mechanisms of the Hsp90 co-chaperone pathway. [ABSTRACT FROM AUTHOR]

Chemoresistance is often referred to as a major leading reason for cancer therapy failure, causing cancer relapse and further metastasis. As a result, an urgent need has been raised to reach a full comprehension of chemoresistance-associated molecular pathways, thereby designing new therapy methods. Many of metastatic tumor masses are found to be related with a viral cause. Although combined therapy is perceived as the model role therapy in such cases, chemoresistant features, which is more common in viral carcinogenesis, often get into way of this kind of therapy, minimizing the chance of survival. Some investigations indicate that the infecting virus dominates other leading factors, i.e., genetic alternations and tumor microenvironment, in development of cancer cell chemoresistance. Therapeutic vaccines are an effective approach in cancer therapy for treating the disease at later stages. The Food and Drug Administration (FDA) recently approved the first therapeutic cancer vaccine, and further studies are ongoing in clinical trials. These are expected to result in the future development of vaccines with relatively improved efficacy. Several vaccination approaches are being studied in pre-clinical and clinical trials, including the generation of anti-cancer vaccines by plant expression systems.This approach has advantages, such as high safety and low costs, especially for the synthesis of recombinant proteins. Nevertheless, the development of anti-cancer vaccines in plants is faced with some technical obstacles.Herein, we summarize some vaccines that have been used in cancer therapy, with an emphasis on plant-based vaccines.
