Wakes produced by underwater vehicles, particularly submarines, in density-stratified fluids play a pivotal role across military, academic, and engineering domains. In comparison to homogeneous fluid environments, wakes in stratified flows exhibit distinctive phenomena, including upstream blocking, pancake eddies, internal waves, and variations in hydrodynamic performance. These phenomena are crucial for optimizing the operation of underwater vehicles. This review critically assesses the hydrodynamic and thermodynamic aspects of these wakes through an integration of theoretical, experimental, and numerical approaches. The hydrodynamic wake evolution, comprising near-wake, non-equilibrium, and quasi-two-dimensional regimes, is scrutinized. The underlying physics, encompassing energy transformation, vertical motion suppression, and momentum dissipation, are analyzed in detail. Special emphasis is placed on numerical methods, encompassing diverse approaches and turbulence models and highlighting their differences in fidelity and computational cost. Numerical simulations not only provide insights into the intricate interplay among various factors but also emerge as a crucial focal point for future research directions. In the realm of thermodynamic wakes, we delve into the thermal wake induced by the discharge of high-temperature cooling water and the cold wake resulting from the stirring of seawater. The generation, evolution, and ascent to the free surface of these wakes are explored. Additionally, this review identifies and analyzes current research shortcomings in each aspect. By systematically addressing existing knowledge gaps, our study contributes novel insights that propel academic progress and bear significant implications for submarine engineering. Simple Summary: Adventitious root formation is a significant limiting factor in the vegetative propagation of economically important plants. Owing to the low germination rates and survival of conventional seed propagation, vegetative propagation remains the best option for large-scale production. Generally, there is a lack of competency for elite plant species cuttings or explants to form adventitious roots. The molecular-level studies of the adventitious root formation have not been explored in many species, such as ramie. As the WUSCEL-related homeobox gene family plays a critical role in promoting vegetative organs and stem cell functioning, we investigated their potential role in adventitious root formation in ramie using genome-wide characterization, gene structure, and expression analysis. The overall result indicated their possible involvement in rooting ramie cuttings. This study thus increases our understanding of the role of these genes and lays a foundation for further studies. A WUSCHEL-related homeobox (WOX) gene family has been implicated in promoting vegetative organs to embryonic transition and maintaining plant embryonic stem cell identity. Using genome-wide analysis, we identified 17 candidates, WOX genes in ramie (Boehmeria nivea). The genes (BnWOX) showed highly conserved homeodomain regions typical of WOX. Based on phylogenetic analysis, they were classified into three distinct groups: modern, intermediate, and ancient clades. The genes displayed 65% and 35% collinearities with their Arabidopsis thaliana and Oryza sativa ortholog, respectively, and exhibited similar motifs, suggesting similar functions. Furthermore, four segmental duplications (BnWOX10/14, BnWOX13A/13B, BnWOX9A/9B, and BnWOX6A/Maker00021031) and a tandem-duplicated pair (BnWOX5/7) among the putative ramie WOX genes were obtained, suggesting that whole-genome duplication (WGD) played a role in WOX gene expansion. Expression profiling analysis of the genes in the bud, leaf, stem, and root of the stem cuttings revealed higher expression levels of BnWOX10 and BnWOX14 in the stem and root and lower in the leaf consistent with the qRT-PCR analysis, suggesting their direct roles in ramie root formation. Analysis of the rooting characteristics and expression in the stem cuttings of sixty-seven different ramie genetic resources showed a possible involvement of BnWOX14 in the adventitious rooting of ramie. The protein phosphatase 2C (PP2C), a key regulator of the ABA signaling pathway, plays important roles in plant growth and development, hormone signaling, and abiotic stress response. Although the PP2C gene family has been identified in many species, systematic analysis was still relatively lacking in ramie (Boehmeria nivea L.). In the present study, we identified 63 BnPP2C genes from the ramie genome, using bioinformatics analysis, and classified them into 12 subfamilies, and this classification was consistently supported by their gene structures and conserved motifs. In addition, we observed that the functional differentiation of the BnPP2C family of genes was restricted and that fragment replication played a major role in the amplification of the BnPP2C gene family. The promoter cis-regulatory elements of BnPP2C genes were mainly involved in light response regulation, phytohormone synthesis, transport and signaling, environmental stress response and plant growth and development regulation. We identified BnPP2C genes with tissue specificity, using ramie transcriptome data from different tissues, in rhizome leaves and bast fibers. The qRT-PCR results showed that the BnPP2C1, BnPP2C26 and BnPP2C27 genes had a strong response to drought, high salt and ABA, and there were a large number of stress-responsive elements in the promoter region of BnPP2C1 and BnPP2C26. The results suggested that BnPP2C1 and BnPP2C26 could be used as the candidate genes for drought and salt tolerance in ramie. These results provide a reference for further studies on the function of the PP2C gene and advance the development of the mechanism of ramie stress response, with a view to providing candidate genes for the molecular breeding of ramie for drought and salt tolerance. Carbonate rocks exhibit significant heterogeneity as both a source rock and reservoir. Stylolite formation plays a crucial role in the enrichment of organic matter and the migration of geofluids within carbonate rocks. In order to study the enrichment mechanism of organic matter and the geofluid migration mode in the stylolites developed in carbonate rocks, stylolite-bearing core samples from the Dachigan structural belt in the eastern Sichuan Basin were collected. The stylolites and matrix were subjected to the total organic carbon (TOC) test and Rock-Eval pyrolysis, thin-section observation under fluorescent light, whole-rock X-ray diffraction, carbon and oxygen isotope analysis, and scanning electron microscopy. The organic matter occurring in the stylolites is mainly in the form of three types: soluble organic matter, pyrobitumen, and bitumen. This suggests that the organic matter within the stylolites mainly consists of secondary migrated organic matter. The stylolites also exhibit well-developed secondary dolomite and pyrite resulting from late-diagenetic recrystallization. These minerals contribute to the preservation of intercrystalline pore spaces and fractures, providing favorable conditions for oil and gas accumulation and migration. The strong cementation observed at the contact between the stylolites and matrix, along with the presence of secondary minerals nearby, may be attributed to the fractionation of light and heavy oil components during the migration of hydrocarbon fluids from the matrix to the stylolites. The thicknesses of the stylolites vary within the bulk, indicating severe diagenesis in thinner areas. Consequently, this leads to significant fractionation effects. The fractionation of crude oil components by stylolites poses challenges for the study of definitive oil–source correlations. To overcome these challenges, future research could investigate biomarker compounds to attempt oil–source correlations. Additionally, future efforts should take into consideration the spatial variation in the crude oil properties. Understanding the role of stylolites in organic matter enrichment and geofluid migration is crucial for optimizing exploration strategies in the Sichuan Basin, a region of growing importance in the energy industry. Moreover, our findings shed light on the complex interactions within stylolite-bearing rocks, which are not limited to this specific basin. As the exploration of unconventional reservoirs progresses, characterizing challenging formations like tight sandstone becomes increasingly complex. Anisotropic parameters play a vital role in accurately characterizing these unconventional reservoirs. In light of this, this paper introduces an approach that uses a dual-constraint anisotropic rock physics model based on compressional and shear wave velocities. The proposed method aims to enhance the precision of anisotropic parameter calculations, thus improving the overall accuracy of reservoir characterization. The paper begins by applying a convolutional neural network (CNN) to predict shear wave velocity, effectively resolving the issue of incomplete shear wave logging data. Subsequently, an anisotropic rock physics model is developed specifically for tight sandstone. A comprehensive analysis is conducted to examine the influence of quartz, clay porosity aspect ratio, and fracture density on compressional and shear wave velocities. Trial calculations using the anisotropic model data demonstrated that the accuracy of calculating anisotropic parameters significantly improved when both compressional and transverse wave velocity constraints were taken into account, as opposed to relying solely on the compressional wave velocity constraint. Furthermore, the rationality of predicting anisotropic parameters using both the shear wave velocity predicted by the convolutional neural network and the measured compressional wave velocity was confirmed using the example of deep tight sandstone in the Junggar Basin. Compared to conventional reservoirs, the prediction of pressure in ultra-deep tight sandstone formations is difficult. The prediction of seismic pressure is more challenging than well-logging pressure prediction. The main methods for seismic pressure prediction include the equivalent depth method, Eaton method, Fillippone formula, and modified versions. Among them, the Eaton method is widely used and has good effectiveness. However, this method relies on difficult-to-obtain normal compaction trend lines, which leads to low prediction accuracy in space. To address this issue, a method combining the Eaton method and collocated cokriging is proposed. Herein, the Eaton formula is used to predict formation pressure at the well, with compressional wave velocity as the covariate for predicting the main variable—formation pressure. By simulating the shear wave velocity based on seismic compressional wave velocity, the influence of various parameters on the prediction results is analyzed, and the accuracy of this method is verified by comparing it with other methods. The proposed method is then applied to predict formation pressure in the ultra-deep formations of the Junggar Basin. The simulation results show that the collocated cokriging method achieves higher planar accuracy and better matches the experimental expectations in terms of prediction results. The directional optimization correction (DOC) method is proposed to reduce the performance error between desired and fabricated terahertz (THz) devices. Three 340 GHz terahertz filters with a bandwidth of 20 GHz are designed and fabricated. The traditional global optimization correction (GOC) method and the proposed DOC method are used to optimize and reduce the performance error, respectively. It is garnered that the center frequency error and bandwidth error of the fabricated terahertz filter optimized by the GOC method are reduced to 3.5 GHz (~1.03%) and 2.2 GHz (~11%), respectively. Meanwhile, the center frequency error and bandwidth error of the fabricated terahertz filter optimized by the DOC method are reduced to 0.2 GHz (~0.06%) and 0.4 GHz (~2.0%), respectively, which has fewer optimization parameters and higher accuracy than the GOC method. Furthermore, the in-band return loss (RL) of two optimized terahertz filters based on the DOC and GOC methods is less than 15 dB, and the in-band insertion loss (IL) is less than 2.3 dB. In recent years, the incidence of prostate cancer in the male population has been increasing year by year. Transrectal ultrasound (TRUS) is an important means of prostate cancer diagnosis. The accurate segmentation of the prostate in TRUS images can assist doctors in needle biopsy and surgery and is also the basis for the accurate identification of prostate cancer. Due to the asymmetric shape and blurred boundary line of the prostate in TRUS images, it is difficult to obtain accurate segmentation results with existing segmentation methods. Therefore, a prostate segmentation method called DSU-Net is proposed in this paper. This proposed method replaces the basic convolution in the U-Net model with the improved convolution combining shear transformation and deformable convolution, making the network more sensitive to border features and more suitable for prostate segmentation tasks. NAC (NAM, ATAF1/2, and CUC2) transcription factors (TFs) are one of the most prominent plant-specific TF families and play essential roles in plant growth, development and adaptation to abiotic stress. Although the NAC gene family has been extensively characterized in many species, systematic analysis is still relatively lacking in Apocynum venetum (A. venetum). In this study, 74 AvNAC proteins were identified from the A. venetum genome and were classified into 16 subgroups. This classification was consistently supported by their gene structures, conserved motifs and subcellular localizations. Nucleotide substitution analysis (Ka/Ks) showed the AvNACs to be under the influence of strong purifying selection, and segmental duplication events were found to play the dominant roles in the AvNAC TF family expansion. Cis-elements analysis demonstrated that the light-, stress-, and phytohormone-responsive elements being dominant in the AvNAC promoters, and potential TFs including Dof, BBR-BPC, ERF and MIKC_MADS were visualized in the TF regulatory network. Among these AvNACs, AvNAC58 and AvNAC69 exhibited significant differential expression in response to drought and salt stresses. The protein interaction prediction further confirmed their potential roles in the trehalose metabolism pathway with respect to drought and salt resistance. The clinical diagnosis of gastrointestinal stromal tumors (GISTs) requires time-consuming tumor localization by physicians, while automated detection of GIST can help physicians develop timely treatment plans. Existing GIST detection methods based on fully supervised deep learning require a large amount of labeled data for the model training, but the acquisition of labeled data is often time-consuming and labor-intensive, hindering the optimization of the model. However, the semi-supervised learning method can perform better than the fully supervised learning method with only a small amount of labeled data because of the full use of unlabeled data, which effectively compensates for the lack of labeled data. Therefore, we propose a semi-supervised gastrointestinal stromal tumor (GIST) detection method based on self-training using the new selection criterion to guarantee the quality of pseudo-labels and adding the pseudo-labeled data to the training set together with the labeled data after linear mixing. In addition, we introduce the improved Faster RCNN with the multiscale module and the feature enhancement module (FEM) for semi-supervised GIST detection. The multiscale module and the FEM can better fit the characteristics of GIST and obtain better detection results. The experiment results showed that our approach achieved the best performance on our GIST image dataset with the joint optimization of the self-training framework, the multiscale module, and the FEM. In the current study, the total content from two Apocynum species leaves (Apocynum venetum and Apocynum hendersonii) collected from the Ili River Valley Region were extracted, and their bioactivities were investigated. The results showed a significant variation in the total flavonoid contents in the leaf samples collected at different periods (June, July, August, and September), with the highest content in August (60.11 ± 0.38 mg RE/g DW for A. venetum and 56.56 ± 0.24 mg RE/g DW for A. hendersonii), and the lowest in June (22.36 ± 0.05 mg RE/g DW for A. venetum and 20.79 ± 0.02 mg RE/g DW for A. hendersonii). The total flavonoid content was comparably higher in A. venetum than in A. hendersonii. Leaves extracts from the two species demonstrated strong bioactivity, which positively correlated with the total flavonoid contents. The anti-oxidative activity of A. venetum was higher than that of A. hendersonii in tandem with its higher flavonoid contents; the antibacterial activity, however, was conversely opposite. Furthermore, a total of 83 flavonoid metabolites were identified in the two species based on UPLC-ESI-MS/MS, out of which 24 metabolites were differentially accumulated. The variability in these metabolites might be the reason for the different bioactivities displayed by the two species. The present study provides insight into the optimal harvest time for Apocynum species planted in the major distribution area of the Ili River Valley and the specific utilization of A. venetum and A. hendersonii. Continental faulted basins are widely distributed in eastern China. Many of these basins, in which the faults block oil and gas reservoirs, have been explored. The heterogeneity of the reservoirs in fault block is very strong, shich restricts the further efficient development of these kinds of oil and gas fields. In this study, porosity and permeability tests, the use of thin sections of rock, mercury injection experiment and CT scan were used to investigate reservoir quality characteristics and control factors. The results showed that the content of quartz, feldspar, and debris in rock had a significant control function on the quality of the reservoir. Reservoir performance improved with increase of quartz and feldspar content, and worsened with increase of debris content. Taking the Ek1 reservoir in the Wang Guantun area as the specific research object, we developed the following understanding. On the one hand, the main compaction in the study area was mechanical compaction. When the compaction rate was greater than 60%, the porosity and permeability were inversely proportional to the compaction rate. On the other hand, dissolution pores were relatively developed in the study area, and the main types of dissolution were intragranular and intergranular dissolution pores. When the surface porosity of the dissolution pore was over 9.2%, porosity increased significantly the increase of dissolution surface porosity. Apocynum hendersonii is a traditional medicinal plant used primarily as tea. It has a potential health benefit from its rich bioactive substances. This study investigated the reactivity of solvents of different polarities (ethanol, ethyl acetate, n-hexane, methanol, and water) extracts of the A. hendersonii leaf. The phytochemical composition of the extracts was evaluated using a Fourier Transform Infrared spectrophotometer (FT-IR), Gas Chromatography-Mass Spectrometry (GC-MS), UHPLC-MS, and Higher Performance Liquid Chromatography (HPLC). The result revealed the presence of medicinally important bioactive constituents, including phenols, flavonoids, and polysaccharides. Methanol extracts exhibited the highest flavonoid contents (20.11 ± 0.85 mg QE/g DW) and the second-highest in terms of phenolic (9.25 ± 0.03 mg GAE/g DW) and polysaccharide (119.66 ± 2.65 mg GE/g DW). It also had the highest antioxidant capacity with 60.30 ± 0.52% and 4.60 ± 0.02 µmol Fe2+ per g DW based on a 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and ferric reducing antioxidant power (FRAP), respectively. Ethanol extract displayed the maximum antibacterial action against Gram-negative and Gram-positive bacteria and the highest inhibition activity against the enzymes tyrosinase and acetylcholinesterase, followed by methanol extract. The principal component analysis revealed a positive correlation between the constituents, bioactivities, and extracts. The overall result showed A. hendersonii as a rich natural source of antimicrobial and antioxidant bioactive compounds and may be used for future applications in pharmaceuticals and food industries. Lacustrine shale, represented by the Middle Permian Lucaogou Formation in the Jimsar Depression in the eastern Junggar Basin, has become one of the main areas of shale oil exploration in China. In this study, we used 137 samples of shale from the Lucaogou Formation, drawn from 14 wells in the Jimsar Depression, to investigate their characteristics of pyrolysis, organic carbon and soluble organic matter content, biomarkers, organic microscopic composition, and vitrinite reflectance. Basin simulation and hydrocarbon generation thermal simulation experiments were also conducted in a closed system. The results of this study indicate that the input of an algae source was dominant in the source rocks of the Lucaogou Formation, that the water in which the rocks were deposited had high salinity and strong reducibility, and that the source rocks were oil-prone. The Lucaogou source rocks generally had good hydrocarbon generation capability, but showed significant heterogeneity. At the end of the Cretaceous period, the shales in the Lucaogou Formation entered the oil-generation window as a whole. Currently, the shales of the Lucaogou Formation are generally in the high-maturity stage in the deep part of the depression, producing a large amount of high-maturity oil and condensate gas, while those in the shallow part have relatively low maturity and can only produce a large amount of conventional crude oil. The maximum crude oil generation rate of the Lucaogou Formation shale obtained from the thermal simulation results was 220.2 mg/g of the total organic carbon (TOC), and the maximum hydrocarbon expulsion efficiency was estimated to be 59.3–76.4%. Coconut cake is an abundant and good potential edible protein source. However, until now it has not been extensively used in the food industry. To promote its usage, the characterization, nutrition value and antioxidant activity of coconut cake protein fractions (albumin, globulin, prolamine, glutelin-1 and glutelin-2) were studied. Results revealed that all the albumin, globulin, glutelin-1 and glutelin-2 fractions showed a high nutrition value. The prolamine, glutelin-1 and glutelin-2 all exhibited good radical scavenging activity and reducing power, and the globulin and prolamine showed high ion chelating ability (89.14–80.38%). Moreover, all the fractions except glutelin-2 could effectively protect DNA against oxidative damage. Several peptides containing five to eight amino acids with antioxidant activity were also identified by LC-MS/MS from the globulin and glutelin-2 fractions. Curcuma is of high economic value, credited to its medicinal, edible, and ornamental properties, which possess all signatures of adaptability, and rapid radiation, especially species of Curcuma (Chinese Curcuma, a recent Qinghai–Tibetan Plateau diversification genera) scattered in China. However, little is known about the incongruent phylogenetic signals within this genera from different inheritance patterns that will militate against the further development of this genera. In this research, we applied complete chloroplast genome data together with double-digest restriction site-associated DNA sequencing data (ddRAD-seq) strategy to investigate phylogenetic signals of Chinese Curcuma species, clustering using two RAD analysis pipelines (STACKS and pyRAD). Phylogenetic trees were obtained from each locus based on the maximum likelihood (ML) and multispecies coalescent (BEAST) methods. For visual comparison, multi-method and different datasets were used to infer the phylogeny. Deep Neural Networks (DNNs) are commonly used methods in computational intelligence. Most prevalent DNN-based image classification methods are dedicated to promoting the performance by designing complicated network architectures and requiring large amounts of model parameters. These large-scale DNN-based models are performed on all images consistently. However, since there are meaningful differences between images, it is difficult to accurately classify all images by a consistent network architecture. For example, a deeper network is fit for the images that are difficult to be distinguished, but may lead to model overfitting for simple images. Therefore, we should selectively use different models to deal with different images, which is similar to the human cognition mechanism, in which different levels of neurons are activated according to the difficulty of object recognition. To this end, we propose a Hierarchical Convolutional Neural Network (HCNN) for image classification in this paper. HCNNs comprise multiple sub-networks, which can be viewed as different levels of neurons in humans, and these sub-networks are used to classify the images progressively. Specifically, we first initialize the weight of each image and each image category, and these images and initial weights are used for training the first sub-network. Then, according to the predicted results of the first sub-network, the weights of misclassified images are increased, while the weights of correctly classified images are decreased. Furthermore, the images with the updated weights are used for training the next sub-networks. Similar operations are performed on all sub-networks. In the test stage, each image passes through the sub-networks in turn. If the prediction confidences in a sub-network are higher than a given threshold, then the results are output directly. Otherwise, deeper visual features need to be learned successively by the subsequent sub-networks until a reliable image classification result is obtained or the last sub-network is reached. Experimental results show that HCNNs can obtain better results than classical CNNs and the existing models based on ensemble learning. HCNNs have 2.68% higher accuracy than Residual Network 50 (Resnet50) on the ultrasonic image dataset, 1.19% than Resnet50 on the chimpanzee facial image dataset, and 10.86% than Adaboost-CNN on the CIFAR-10 dataset. Computational intelligence has been widely used in medical information processing. The deep learning methods, especially, have many successful applications in medical image analysis. In this paper, we proposed an end-to-end medical lesion segmentation framework based on convolutional neural networks with a dual attention mechanism, which integrates both fully and weakly supervised segmentation. The weakly supervised segmentation module achieves accurate lesion segmentation by using bounding-box labels of lesion areas, which solves the problem of the high cost of pixel-level labels with lesions in the medical images. In addition, a dual attention mechanism is introduced to enhance the network's ability for visual feature learning. The dual attention mechanism (channel and spatial attention) can help the network pay attention to feature extraction from important regions. Compared with the current mainstream method of weakly supervised segmentation using pseudo labels, it can greatly reduce the gaps between ground-truth labels and pseudo labels. The final experimental results show that our proposed framework achieved more competitive performances on oral lesion dataset, and our framework further extended to dermatological lesion segmentation. Seismic wave exhibits the characteristics of anisotropy and attenuation while propagating through the fluid-bearing fractured or layered reservoirs, such as fractured carbonate and shale bearing oil or gas. We derive a linearized reflection coefficient that simultaneously considers the effects of anisotropy and attenuation caused by fractures and fluids. Focusing on the low attenuated transversely isotropic medium with a vertical symmetry axis (Q-VTI) medium, we first express the complex stiffness tensors based on the perturbation theory and the linear constant Q model at an arbitrary reference frequency, and then we derive the linearized approximate reflection coefficient of P to P wave. It decouples the P- and S-wave inverse quality factors, and Thomsen-style attenuation-anisotropic parameters from complex P- and S-wave velocity and complex Thomsen anisotropic parameters. By evaluating the reflection coefficients around the solution point of the interface of two models, we analyze the characteristics of reflection coefficient vary with the incident angle and frequency and the effects of different Thomsen anisotropic parameters and attenuation factors. Moreover, we realize the simultaneous inversion of all parameters in the equation using an actual well log as a model. We conclude that the derived reflection coefficient may provide a theoretical tool for the seismic wave forward modeling, and again it can be implemented to predict the reservoir properties of fractures and fluids based on diverse inversion methods of seismic data. In the current study, the active flavonoids from Apocynum venetum and Apocynum hendersonii leaf were efficiently characterized using UPLC-ESI-MS/MS, and yielding the highest content of 15.35 mg/g (A. venetum) and 13.28 mg/g (A. hendersonii) respectively. The antioxidant assay in vitro showed that the isolated flavonoid ingredient groups exhibited free radical scavenging activities to DPPH, ABTS and linoleic acid. The antimicrobial assay revealed the isolated flavonoid ingredient from both A. venetum and A. hendorsonii have exerted anti-MRSA and anti-P. aeruginosa effect through disrupting cell integrity and declining ATP. In vivo assay demonstrated that these flavonoid ingredients effectively accelerated MRSA-infected and P. aeruginosa-infected Balb/c mice wound healing. Dolastatin 10 (Dol-10), a leading marine pentapeptide isolated from the Indian Ocean mollusk Dolabella auricularia, contains three unique amino acid residues. Dol-10 can effectively induce apoptosis of lung cancer cells and other tumor cells at nanomolar concentration, and it has been developed into commercial drugs for treating some specific lymphomas, so it has received wide attention in recent years. In vitro experiments showed that Dol-10 and its derivatives were highly lethal to common tumor cells, such as L1210 leukemia cells (IC50 = 0.03 nM), small cell lung cancer NCI-H69 cells (IC50 = 0.059 nM), and human prostate cancer DU-145 cells (IC50 = 0.5 nM), etc. With the rise of antibody-drug conjugates (ADCs), milestone progress was made in clinical research based on Dol-10. A variety of ADCs constructed by combining MMAE or MMAF (Dol-10 derivatives) with a specific antibody not only ensured the antitumor activity of the drugs themself but also improved their tumor targeting and reduced the systemic toxicity. They are currently undergoing clinical trials or have been approved for marketing, such as Adcetris®, which had been approved for the treatment of anaplastic large T-cell systemic malignant lymphoma and Hodgkin lymphoma. Dol-10, as one of the most medically valuable natural compounds discovered up to now, has brought unprecedented hope for tumor treatment. It is particularly noteworthy that, by modifying the chemical structure of Dol-10 and combining with the application of ADCs technology, Dol-10 as a new drug candidate still has great potential for development. Clarifying the genetic relationship and divergence among Curcuma L. (Zingiberaceae) species around the world is intractable, especially among the species located in China. In this study, Reduced Representation Sequencing (RRS), as one of the next generation sequences, has been applied to infer large scale genotyping of major Chinese Curcuma species which present little differentiation of morphological characteristics and genetic traits. The 1295 high-quality SNPs (reduced-filtered SNPs) were chosen from 997,988 SNPs of which were detected from the cleaned 437,061 loci by RRS to investigate the phylogeny and divergence among eight major Curcuma species locate in the Hengduan Mountains of the Qinghai–Tibet Plateau (QTP) in China. The results showed that all the population individuals were clustered together within species, and species were obviously separated; the clustering results were recovered in PCA (Principal Component Analysis); the phylogeny was (((((C. Phaeocaulis, C. yunnanensis), C. kwangsiensis), (C. amarissima, C. sichuanensis)), C. longa), (C. wenyujin, C. aromatica)); Curcuma in China originated around ~7.45 Mya (Million years ago) in the Miocene, and interspecific divergence appeared at ca. 4–2 Mya, which might be sped up rapidly along with the third intense uplift of QTP. The occurrence of adventitious roots is necessary for the survival of cuttings. In this study, comparative transcriptome analysis between two ramie (Boehmeria nivea L.) varieties with different adventitious root (AR) patterns was performed by mRNA-Seq before rooting (control, CK) and 10 days water-induced adventitious rooting (treatment, T) to reveal the regulatory mechanism of rooting. Characterization of the two ramie cultivars, Zhongzhu No 2 (Z2) and Huazhu No 4 (H4), indicated that Z2 had a high adventitious rooting rate but H4 had a low rooting rate. Twelve cDNA libraries of the two varieties were constructed, and a total of 26,723 genes were expressed. In the non-water culture condition, the number of the distinctive genes in H4 was 2.7 times of that in Z2, while in the water culture condition, the number of the distinctive genes in Z2 was nearly 2 times of that in H4. A total of 4411 and 5195 differentially expressed genes (DEGs) were identified in the comparison of H4CK vs. H4T and Z2CK vs. Z2T, respectively. After the water culture, more DEGs were upregulated in Z2, but more DEGs were downregulated in H4. Gene ontology (GO) functional analysis of the DEGs indicated that the polysaccharide metabolic process, carbohydrate metabolic process, cellular carbohydrate metabolic process, cell wall macromolecule metabolic process, and photosystem GO terms were distinctively significantly enriched in H4. Simultaneously, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that photosynthesis, photosynthesis antenna proteins, and starch and sucrose metabolism pathways were distinctively significantly enriched in H4. Moreover, KEGG analysis showed that jasmonic acid (JA) could interact with ethylene to regulate the occurrence and number of AR in Z2. This study reveals the transcriptomic divergence of two ramie varieties with high and low adventitious rooting rates, and provides insights into the molecular regulatory mechanism of AR formation in ramie. Water and N are the most important factors affecting ramie (Boehmeria nivea (L.) Gaudich) growth. In this study, de novo transcriptome assembly and Tandem Mass Tags (TMT) based quantitative proteome analysis of ramie under nitrogen and water co-limitation conditions were performed, and exposed to treatments, including drought and N-deficit (WdNd), proper water but N-deficit (WNd), proper N but drought (WdN), and proper N and water (CK), respectively. A total of 64,848 unigenes (41.92% of total unigenes) were annotated in at least one database, including NCBI non-redundant protein sequences (Nr), Swiss-Prot, Protein family (Pfam), Gene Ontology (GO) and KEGG Orthology (KO), and 4268 protein groups were identified. Most significant changes in transcript levels happened under water-limited conditions, but most significant changes in protein level happened under water-limited conditions only with proper N. Poor correlation between differentially expressed genes (DEGs) and differentially expressed proteins (DEPs) was observed in ramie responding to the treatments. DEG/DEP regulation patterns related to major metabolic processes responding to water and N deficiency were analyzed, including photosynthesis, ethylene responding, glycolysis, and nitrogen metabolism. Moreover, 41 DEGs and 61 DEPs involved in regulating adaptation of ramie under water and N stresses were provided in the study, including DEGs/DEPs related to UDP—glucuronosyhransferase (UGT), ATP synthase, and carbonate dehydratase. The strong dependency of N-response of ramie on water conditions at the gene and protein levels was highlighted. Advices for simultaneously improving water and N efficiency in ramie were also provided, especially in breeding N efficient varieties with drought resistance. Nitrogen-use efficiency (NUE) has significant impacts on plant growth and development. NUE in plants differs substantially in physiological resilience to nitrogen stress; however, the molecular mechanisms underlying enhanced resilience of high-NUE plants to nitrogen deficiency remains unclear. We compared transcriptome-wide gene expression between high-NUE and low-NUE ramie (Boehmeria nivea (L.) Gaudich) genotypes under nitrogen (N)-deficient and normal conditions to identify the transcriptomic expression patterns that contribute to ramie resilience to nitrogen deficiency. Two ramie genotypes with contrasting NUE were used in the study, including T29 (NUE = 46.01%) and T13 (NUE = 15.81%). Our results showed that high-NUE genotypes had higher gene expression under the control condition across 94 genes, including frontloaded genes such as GDSL esterase and lipase, gibberellin, UDP-glycosyltransferase, and omega-6 fatty acid desaturase. Seventeen stress-tolerance genes showed lower expression levels and varied little in response to N-deficiency stress in high-NUE genotypes. In contrast, 170 genes were upregulated under N deficiency in high-NUE genotypes but downregulated in low-NUE genotypes compared with the controls. Furthermore, we identified the potential key genes that enable ramie to maintain physiological resilience under N-deficiency stress, and categorized these genes into three groups based on the transcriptome and their expression patterns. The transcriptomic and clustering analysis of these nitrogen-utilization-related genes could provide insight to better understand the mechanism of linking among the three gene classes that enhance resilience in high-NUE ramie genotypes. Apocynum plants, especially A. venetum and A. hendersonii, are rich in flavonoids. In the present study, a whole genome survey of the two species was initially carried out to optimize the flavonoid biosynthesis-correlated gene mining. Then, the metabolome and transcriptome analyses were combined to elucidate the flavonoid biosynthesis pathways. Both species have small genome sizes of 232.80 Mb (A. venetum) and 233.74 Mb (A. hendersonii) and showed similar metabolite profiles with flavonols being the main differentiated flavonoids between the two specie. Positive correlation of gene expression levels (flavonone-3 hydroxylase, anthocyanidin reductase, and flavonoid 3-O-glucosyltransferase) and total flavonoid content were observed. The contents of quercitrin, hyperoside, and total anthocyanin in A. venetum were found to be much higher than in A. hendersonii, and such was thought to be the reason for the morphological difference in color of A. venetum and A. hendersonii. This study provides valuable genomic and metabolome information for understanding of A. venetum and A. hendersonii, and lays a foundation for elucidating Apocynum genus plant flavonoid biosynthesis. Human lung cancer is highly invasive and the most malignant among human tumors. Adenocarcinoma as a specific type of non-small cell lung cancer occurs with high frequency and is also highly resistant to radiation therapy. Thus, how to avoid radiation resistance and improve radiotherapy effectiveness is a crucial question. In the present study, human lung cancer A549 and H1299 cells were irradiated using γ-rays from a Co60 irradiator. Protein expression was detected by Western blotting. Cell cycle and apoptosis were measured by flow cytometry. Surviving fraction was determined by colony formation assay. γH2AX and 53BP1 foci formation were examined by fluorescence microscopy. In the results, we show that CHMP4C, a subunit of Endosomal sorting complex-III (ESCRT-III), is involved in radiation-induced cellular response. Radiation-induced Aurora B expression enhances CHMP4C phosphorylation in non-small cell lung cancer (NSCLC) cells, maintaining cell cycle check-point and cellular viability as well as resisting apoptosis. CHMP4C depletion enhances cellular sensitivity to radiation, delays S-phase of cell cycle and reduces ionizing radiation (IR)-induced γH2AX foci formation. We found that Aurora B targets CHMP4C and inhibition of Aurora B exhibits similar effects with silencing of CHMP4C in radioresistance. We also confirm that CHMP4C phosphorylation is elevated after IR both in p53-positive and-negative cells, indicating that the close correlation between CHMP4C and Aurora B signaling pathway in mediating radiation resistance is not p53 dependent. Together, our work establishes a new function of CHMP4C in radiation resistance, which will offer a potential strategy for non-small cell lung cancer by disrupting CHMP4C.
