Abstract

Abdominal subcutaneous fat deposition (ASFD) is not only related to meat quality in the pig industry but also to human health in medicine. It is of great value to elucidate the potential molecular mechanisms of ASFD. The present study aims to identify obese-specific biomarkers and key pathways correlated with ASFD in pigs. The ASF-related mRNA expression dataset GSE136754 was retrieved from the Gene Expression Omnibus (GEO) database and systematically analyzed using a comprehensive bioinformatics method. A total of 565 differentially expressed genes (DEGs) were identified between three obese and three lean pigs, and these DEGs were mainly involved in the p53 signaling pathway, MAPK signaling pathway and fatty acid metabolism. A protein-protein interaction (PPI) network, consisting of 540 nodes and 1,065 edges, was constructed, and the top ten genes with the highest degree scores—ABL1, HDAC1, CDC42, HDAC2, MRPS5, MRPS10, MDM2, JUP, RPL7L1 and UQCRFS1—were identified as hub genes in the whole PPI network. Especially HDAC1, MDM2, MRPS10 and RPL7L1 were identified as potential robust obese-specific biomarkers due to their significant differences in single gene expression levels and high ROC area; this was further verified by quantitative real-time PCR (qRT-PCR) on abdominal subcutaneous fat samples from obese-type (Saba) and lean-type (Large White) pigs. Additionally, a mRNA-miRNA-lncRNA ceRNA network consisting of four potential biomarkers, 15 miRNAs and 51 lncRNAs was established, and two targeted lncRNAs with more connections, XIST and NEAT1, were identified as potentially important regulatory factors. 24. Improving consolidation of dredged slurry by vacuum preloading using prefabricated vertical drains (PVDs) with varying filter pore sizes

Abstract

Prefabricated vertical drains (PVDs) have been used extensively to accelerate the consolidation rate of dredged slurry. While some fine particles from dredged slurry can easily squeeze through the filter into the drainage channel, many cannot. As such, these soil particles deposit on the filter surface causing partial clogging of the drainage path. Although the pore size of filter is recognized as an important factor that influences PVD clogging, the standards for determining the pore size of the filter are lacking. To this end, the traditional gradient ratio tests with four different filter pore sizes were conducted, and the results show that the permeability of the filter at a given head increases with the increase in the pore size of the filter. To remove the effect of the difference between static hydraulic gradient and vacuum pressure, the vacuum preloading tests with varying pore sizes of filters were further conducted. Through these vacuum preloading tests, the degree of vacuum, settlement, pore-water pressure, water content, vane shear strength, and other parameters of PVDs with various filter pore sizes were obtained, and the optimal pore size of filter was determined. Les drains verticaux prefabriques (DVP) ont ete largement utilises pour accelerer le taux de consolidation des boues de dragage. Certaines particules fines de boues de dragage peuvent facilement passer a travers le filtre dans le canal de drainage, mais beaucoup ne le peuvent pas. En tant que telles, ces particules de sol se deposent sur la surface du filtre, provoquant un colmatage partiel du chemin de drainage. Bien que la taille des pores du filtre soit reconnue comme un facteur important qui influence le colmatage du DVP, les normes permettant de determiner la taille des pores du filtre sont insuffisantes. A cette fin, les tests de rapport de gradient traditionnels avec quatre tailles de pores de filtre differentes ont ete realises, et les resultats montrent que la permeabilite du filtre a une hauteur donnee augmente avec l'augmentation de la taille des pores du filtre. Afin de supprimer l'effet de la difference entre le gradient hydraulique statique et la pression de vide, les tests de prechargement sous vide avec des filtres de differentes tailles de pores ont egalement ete realises. Grace a ces tests de prechargement sous vide, le degre de vide, le tassement, la pression de l'eau interstitielle, la teneur en eau, la resistance au cisaillement de l'essai scissometrique et d'autres parametres des DVP avec differentes tailles de pores du filtre ont ete obtenus et la taille de pores optimale du filtre a ete determinee. Abstract

N6-Methyladenosine-related long noncoding RNAs play an essential role in many cancers’ development. However, the relationship between m6A-related lncRNAs and acute myelogenous leukemia (AML) prognosis remains unclear. We systematically analyzed the association of m6A-related lncRNAs with the prognosis and tumor immune microenvironment (TME) features using the therapeutically applicable research to generate effective treatment (TARGET) database. We screened 315 lncRNAs associated with AML prognosis and identified nine key lncRNAs associated with m6A by the LASSO Cox analysis. A model was established based on these nine lncRNAs and the predictive power was explored in The Cancer Genome Atlas (TCGA) database. The areas under the ROC curve of TARGET and TCGA databases for ROC at 1, 3, and 5 years are 0.701, 0.704, and 0.696, and 0.587, 0.639, and 0.685, respectively. The nomogram and decision curve analysis (DCA) showed that the risk score was more accurate than other clinical indicators in evaluating patients’ prognoses. The clusters with a better prognosis enrich the AML pathways and immune-related pathways. We also found a close correlation between prognostic m6A-related lncRNAs and tumor immune cell infiltration. LAG3 expression at the immune checkpoint was lower in the worse prognostic cluster. In conclusion, m6A-related lncRNAs partly affected AML prognosis by remodeling the TME and affecting the anticarcinogenic ability of immune checkpoints, especially LAG3 inhibitors. The prognostic model constructed with nine key m6A-related lncRNAs can provide a method to assess the prognosis of AML patients in both adults and children.

Abstract

Novel TiO2 nanoparticles-coated CNTs (TiO2@CNTs) fabricated by the alcohol-thermal method were used as the reinforcement to prepare aluminum matrix composites through high-intensity ultrasonic-assisted casting, followed by heat treatment. The results of optical microscope showed that the optimum heat treatment process was solution treatment (495 °C, 2 h) + aging treatment (180 °C, 12 h). Under this condition, the Al2Cu and Al2CuMg phases were uniformly precipitated in the composites, and most of them were in a discrete state. TEM results revealed that TiO2 nanoparticles dispersed on the surface of CNTs played the role of an interlayer structure between CNTs and matrix, locking the CNTs and improving the CNTs–Al interfacial bonding. The ultimate tensile strength (UTS) and yield strength (YS) of 1.0 wt%TiO2@CNTs/2024 composite after T6 heat treatment reach 324 MPa and 219 MPa, respectively, which exhibited enhanced UTS and YS than the monolithic 2024 alloy. This was attributed to the synergistic strengthening of multi-mechanisms, such as grain refinement strengthening, load transfer strengthening, dislocation strengthening and, especially, Orowan strengthening.
