The intra-erythrocytic apicomplexan Babesia microti is the predominant pathogen that causes human babesiosis, an infectious disease that occurs worldwide. B. microti relies on the antioxidant including thioredoxin system to maintain the redox balance during the erythrocytic stage. In the present study, the full-length B. microti thioredoxin 3 (BmTrx3) gene was cloned, expressed in vitro , and its response to antiprotozoal drugs were tested. The full-length BmTrx3 was 663 bp and contained an intact open reading frame of 567 bp. The encoded polypeptide was 188 amino acids and the predicted molecular weight of the protein was 21.7 kDa. A conserved thioredoxin-like family domain was found in BmTrx3. The expression of BmTrx3 was upregulated on both the third and eighth day post-infection in mice, whereas expression was downregulated during the beginning and later stages. Western blot analysis showed that mouse anti-BmTrx3 serum could recognize the native BmTrx3 in parasite lysates and that the mouse anti- B. microti serum could recognize the recombinant BmTrx3 protein. Immunofluorescence microscopy showed that BmTrx3 localized in the cell cytoplasm of B. microti merozoites in B. microti- infected red blood cells. The results of bovine insulin reduction assay indicated the enzyme activity of the purified recombinant BmTrx3 protein. The anti-malaria drug chloroquine significantly inhibited the expression of BmTrx3, however, another anti-malaria drug qunine, and a known anti-babesiosis drug clindamycin, induced significantly higher upregulation of BmTrx3 mRNA. The results of the present study demonstrate that BmTrx3 is a functional enzyme with antioxidant activity and may be involved in the response of B. microti to anti-parasite drugs.

Ethnopharmacological relevance Hua Ju Hong (HJH, peels of Citrus grandis (L.) Osbeck) is a popularly used traditional Chinese medicine recorded by “Compendium of Materia Medica” (Ben Cao Gang Mu) in Ming Dynasty of China (1578 A.D.). With flavonoid components, HJH possesses hypolipidemic effect associated with antioxidation action. However, no report was found regarding the flavonoid profile and antioxidant activity of HJH. Materials and methods Five purified flavonoid extracts (TFCA, TFCB, TFCC, TFCD and TFCE.) were obtained from HJH using Ca(OH)2 and HPD-300 macroporous resins, and their total flavonoids and representative flavonoid components were analyzed. In vitro tests of DPPH free radical scavenging activity, reducing power, and total antioxidant activity of each extract were evaluated. The most effective extract was selected for in vivo antioxidative evaluation using a rat hyperlipemia model. Results The total flavonoid content was varying among each HJH extract and decreased in an order of TFCB>TFCD>TFCC>TFCE>TFCA. TFCB, TFCD, and TFCC contained more than 50% of total flavonoids, the highest content of which was found in TFCB (80.7%). HPLC analysis showed that the contents of three flavonoid components, narirutin, naringin and neohesperidin, displayed a similar trend as that of total flavonoids. In vitro antioxidative tests determined that TFCB at 0.24 to 1.2 mg/ml possessed the most significant antioxidant effects among other extracts and was also superior to BHT. In vivo experiment also revealed the significant antioxidant and antihyperlipidemic activities of TFCB started from 50 to 200 mg/kg after oral administration to hyperlipemia rats. These results indicate that TFCB with the highest flavonoid contents has the strongest antioxidant-associated activities. Conclusion This is the first report regarding antioxidant-associated activities and relevant flavonoid components of HJH extracts, providing a promising candidate of traditional Chinese medicine for antioxidative treatment.

Chronic fatigue accumulation increases the incidence of cardiovascular disease while the treatment of antioxidants could prevent this development. We have previously shown that quercetin-3-O-gentiobiose (QG), a flavonoid isolated from tonic herb Okra, possesses anti-oxidative properties. In the present study, the protective effects of QG were evaluated in a rat model of load-induced endurance swimming. Oral administration of QG at the doses of 25-75mg/kg could significantly improve the endurance capability of rats to fatigue along with decrease serum lactic acid and blood urea nitrogen levels were decreased. Moreover, QG could alleviate vascular impairments, enhance the activities of antioxidant enzymes and attenuate the levels of inflammatory cytokines (MCP-1, IL-6 and TNF-α). The results indicated that QG had anti-fatigue and vasoprotective effects and represented a potential agent for the treatment of aortic pathology involved with fatigue- and related syndrome.
