The reasonable design of hole spacing for buried heat exchangers is the first issue to be solved when developing shallow geothermal energy in the Nanjing erosion and accumulation plain area. In order to obtain the hole spacing for design reference and ensure ground-source heat pump system to operate long and stably, based on the ground-source heat pump test project at No. 102 Shimenkan, Qinhuai District of Nanjing, the hole spacing of ground heat exchanger and time effect of 10-year operation with ground-source heat pump system have been studied with the temperature monitoring system and numerical simulation method, and the hole spacing of ground heat exchanger and the operation mode have been obtained. The results show that : (1) during the complete operation cycle of summer, post summer recovery, winter and post winter recovery working conditions of ground-source heat pump system, formation temperature undergoes periodic change of heating, cooling, slow cooling and basic recovery. (2) The economical and reasonable hole spacing of ground heat exchanger is 4.0 meters suitable for the erosion accumulation plain of Nanjing. (3) In order to eliminate the heat accumulation during the summer working condition and promote the service life of ground-source heat pump system, the operation mode of equal cooling time in the summer and heating time in the winter is suggested, and the post summer recovery working condition is extended as much as possible. This study can provide scientific basis for the design of buried pipes and shallow geothermal energy engineering research in the Nanjing area.

Abstract To optimize and evaluate the accuracy of the vault-predicting formula generated from a very high-frequency digital ultrasound robotic scanner (Artemis Insight 100). The relationship between the achieved lens vault (LVa) at one month after intraocular collamer lens (ICL) implantation surgery and the predicted vault (LVp) was analyzed by a retrospective study, and an optimized formula was built up. Then, the accuracy of the optimized vault-predicting formula was evaluated in a prospective study by comparing the LVa and the predicted vault from the optimized formula (LVop). The retrospective study included 77 patients (133 eyes) while the prospective study enrolled 90 patients (170 eyes). Abstract Background Women with polycystic ovary syndrome (PCOS) are usually selected to undergo an ovulation induction regimen or a programmed regimen for endometrial preparation in the frozen-thawed embryo transfer (FET) during their IVF/ICSI treatment. The programmed regimen permits flexible scheduling of embryo transfer but requires long-term usage of exogenous estrogen and higher dosages of luteal support while the letrozole ovulation regimen needs lower dosages of luteal support only. Recently, multiple studies have shown that the letrozole ovulation regimen can improve pregnancy outcomes of FET in women with PCOS compared with the programmed regimen. However, most of these studies are retrospective, and prospective studies are urgently needed the evidence from the perspective study is insufficient. Methods/design We are undertaking a multicentre, randomized, controlled clinical trial of an endometrial preparation regimen for FET in women with PCOS. The eligible women are randomly assigned to either the letrozole ovulation regimen or the programmed regimen for endometrial preparation. The primary outcome is the clinical pregnancy rate. Discussion The results of this study will provide evidence for whether the letrozole ovulation regimen for endometrial preparation could improve pregnancy outcomes in PCOS women undergoing FET. Abstract Activation of the NF-κB pathway is strictly regulated to prevent excessive inflammatory and immune responses. In a well-known negative feedback model, IκBα-dependent NF-κB termination is a delayed response pattern in the later stage of activation, and the mechanisms mediating the rapid termination of active NF-κB remain unclear. Here, we showed IκBα-independent rapid termination of nuclear NF-κB mediated by CLK2, which negatively regulated active NF-κB by phosphorylating the RelA/p65 subunit of NF-κB at Ser180 in the nucleus to limit its transcriptional activation through degradation and nuclear export. Depletion of CLK2 increased the production of inflammatory cytokines, reduced viral replication and increased the survival of the mice. Mechanistically, CLK2 phosphorylated RelA/p65 at Ser180 in the nucleus, leading to ubiquitin‒proteasome-mediated degradation and cytoplasmic redistribution. Importantly, a CLK2 inhibitor promoted cytokine production, reduced viral replication, and accelerated murine psoriasis. This study revealed an IκBα-independent mechanism of early-stage termination of NF-κB in which phosphorylated Ser180 RelA/p65 turned off posttranslational modifications associated with transcriptional activation, ultimately resulting in the degradation and nuclear export of RelA/p65 to inhibit excessive inflammatory activation. Our findings showed that the phosphorylation of RelA/p65 at Ser180 in the nucleus inhibits early-stage NF-κB activation, thereby mediating the negative regulation of NF-κB.

Abstract Background The precision of anterior segment biometric measurements in eyes has become increasingly important in refractive surgery. The purpose of this study is to assess the repeatability of the automatic measurements provided by a new spectral-domain optical coherence tomograph (SD-OCT)/Placido topographer (MS-39, CSO) and its agreement with a swept-source OCT (SS-OCT) biometer (CASIA SS-1000, Tomey) in patients with myopia. Methods The right eye of 235 subjects was scanned 3 times with both devices. The evaluated parameters included central corneal radius of the steep meridian, central corneal radius of the flat meridian, mean central corneal radius, thinnest corneal thickness, central corneal thickness, anterior chamber depth, corneal volume and diameter. The intraobserver repeatability of the MS-39 measurements was calculated using intraclass correlation coefficient (ICC), within subject standard deviation, coefficient of repeatability, coefficient of variation and repeated-measures analysis of variance of the 3 repeated measurements. The agreement between the two devices was evaluated by 95% limits of agreement (LoA). Results The majority of the parameters acquired from MS-39 showed high repeatability. The repeatability of corneal diameter was slightly lower than the other measurements, although the ICC remained high. Agreement with the CASIA SS-1000 was good, indicated by the Bland-Altman plots with narrow 95% LoA values for all parameters assessed. Conclusions The high repeatability of automatic measurements by the new device supports its clinical application in eyes with myopia, and the good agreement between the two devices indicates they could be used interchangeably for the parameters evaluated.

Abstract The alteration of metabolic processes has been found to have significant impacts on the development of hepatocellular carcinoma (HCC). Nevertheless, the effects of dysfunction of tyrosine metabolism on the development of HCC remains to be discovered. This research demonstrated that tyrosine hydroxylase (TH), which responsible for the initial and limiting step in the bio-generation of the neuro-transmitters dopamine and adrenaline, et al. was shown to be reduced in HCC. Increased expression of TH was found facilitates the survival of HCC patients. In addition, decreased TH indicated larger tumor size, much more numbers of tumor, higher level of AFP, and the presence of cirrhosis. TH effectively impairs the growth and metastasis of HCC cells, a process dependent on the phosphorylation of serine residues (S19/S40). TH directly binds to Smad2 and hinders the cascade activation of TGFβ/Smad signaling with the treatment of TGFβ1. In summary, our study uncovered the non-metabolic functions of TH in the development of HCC and proposes that TH might be a promising biomarker for diagnosis as well as an innovative target for metastatic HCC.

According to a study from World Health Organization's Global Burden of Disease, mental and neurological disorders have accounted for 13% of global diseases in recent years and are on the rise. Neuropsychiatric conditions or neuroinflammatory disorders are linked by the presence of an exaggerated immune response both peripherally and in the central nervous system (CNS). Cognitive dysfunction (CD) encompasses a complex group of diseases and has frequently been described in the field of autoimmune diseases, especially in multiple non-CNS-related autoimmune diseases. Recent studies have provided various hypotheses regarding the occurrence of cognitive impairment in autoimmune diseases, including that abnormally activated immune cells can disrupt the integrity of the blood-brain barrier (BBB) to trigger a central neuroinflammatory response. When the BBB is intact, autoantibodies and pro-inflammatory molecules in peripheral circulation can enter the brain to activate microglia, inducing CNS inflammation and CD. However, the mechanisms explaining the association between the immune system and neural function and their contribution to diseases are uncertain. In this review, we used clinical statistics to illustrate the correlation between CD and autoimmune diseases that do not directly affect the CNS, summarized the clinical features and mechanisms by which autoimmune diseases trigger cognitive impairment, and explored existing knowledge regarding the link between CD and autoimmune diseases from the perspective of the field of neuroimmunology.

Two new termite-pathogenic species, Ophiocordyceps globiperitheciata and O. longistipes, are described from Yunnan Province, China. Six-locus (ITS, nrSSU, nrLSU, tef-1α, rpb1 and rpb2) phylogenetic analyses in combination with morphological observations were employed to characterize these two species. Phylogenetically, O. globiperitheciata is most closely related to Hirsutella cryptosclerotium and O. communis, whereas O. longistipes shares a sister relationship with O. fusiformis. However, O. globiperitheciata differs from H. cryptosclerotium by parasitizing Blattodea and producing clavate, unbifurcated stromata. Ophiocordyceps globiperitheciata is distinguished from O. communis by multiple stromata, shorter asci and ascospores. Ophiocordyceps longistipes differs from O. fusiformis in producing larger stromata, perithecia, asci and ascospores, as well as smaller citriform or oval conidia. Morphological descriptions of the two new species and a dichotomous key to the 19 termite-pathogenic Ophiocordyceps species are presented.

Dual-phase amorphous-nanocrystalline Al-x at.% Mo (x = 20, 26, 30) alloy films prepared in the way of dual-phase co-growth directly through magnetron sputtering were studied in terms of thermally induced microstructural evolution and mechanical properties via annealing at 550 °C. It was observed that, the annealed Al-26 and 30 at.% Mo alloy films maintained stable dual-phase amorphous-nanocrysatlline structure and precipitated Al8Mo3 nano-grains dispersed in amorphous matrix uniformly. After annealing, the surface roughness of Al-26 and 30 at.% Mo alloy films remained unchanged of 7.5 nm and the hardness and elastic modulus of them were as high as 13 and 210 GPa which enhanced about 50 % and 24 % than before, respectively. In contrast, the Al-20 at.% Mo alloy film crystallized completely and mainly contained intermetallic Al12Mo phase along with apparent grain coarsening after annealing, which resulted in significant increase of surface roughness and deterioration of hardness. The excellent thermal stability of dual-phase amorphous-nanocrystalline Al-26 and 30 at.% Mo alloy films stem from the small driving force for precipitated Al8Mo3 nano-grain growth, as well as benefit from effect of diffusion field impingement.

Abstract Point cloud completion is a challenging task that involves predicting missing parts in incomplete 3D shapes. While existing strategies have shown effectiveness on point cloud datasets with regular shapes and continuous surfaces, they struggled to manage the morphologically diverse structures commonly encountered in real-world scenarios. This research proposed a new point cloud completion method, called SegCompletion, to derive complete 3D geometries from a partial shape with different structures and discontinuous surfaces. To achieve this, morphological segmentation was introduced before point cloud completion by deep hierarchical feature learning on point sets, and thus, the complex morphological structure was segmented into regular shapes and continuous surfaces. Additionally, each instance of a point cloud that belonged to the same type of feature could also be effectively identified using HDBSCAN (Hierarchical Density-Based Spatial Clustering of Applications with Noise). Furthermore, the multiscale generative network achieved sophisticated patching of missing point clouds under the same geometric feature based on feature points. To compensate for the variance in the mean distances between the centers of the patches and their closest neighbors, a simple yet effective uniform loss was utilized. A number of experiments on ShapeNet and Pheno4D datasets have shown the performance of SegCompletion on public datasets. Moreover, the contribution of SegCompletion to our dataset (Cotton3D) was discussed. The experimental results demonstrated that SegCompletion performed better than existing methods reported in the literature.

Background: As adequate support, virtual reality (VR) has been increasingly introduced into the classroom to help students learn a new language. However, empirical studies exploring the educational potential of using 360° spherical video-based virtual reality (SVVR) in high-school Chinese language classrooms are still lacking. Objectives: A mind mapping-based SVVR learning system is proposed for international high-school students' Chinese courses and examines its effects on improving Chinese learning achievements and perceptions among Grade 10 students who learn Chinese as a foreign language. Methods: This study adopted a quasi-experiment design. Participants (N = 66) were assigned to one of three conditions: mind mapping-based SVVR (MMBSVVR), conventional SVVR (CSVVR), or traditional lecturing (TL). Our mixed-method approach employed pre-and post-tests to measure Chinese learning achievements, questionnaires to measure Chinese learning motivation, problem-solving ability, and self-efficacy, and semi-structured interviews to further explore students' feelings and attitudes about Chinese courses in corresponding learning environments. Results and Conclusions: The results showed that the MMBSVVR classroom significantly improved high students' Chinese learning achievements in Chinese vocabulary and making sentence, problem-solving ability, and self-efficacy. Moreover, SVVR stimulates students' learning interest and class participation, and mind mapping provides students with a clear guide to arrange the knowledge. These findings help interpret the relationships between MMBSVVR and learning achievements, Chinese learning motivation, problem-solving ability, and self-efficacy in an international high school Chinese courses, thus providing insight on integrating MMBSVVR into existing classrooms.

Many key functionalities of optical frequency combs, such as self-referencing and broad spectral access, rely on coherent supercontinuum generation (SCG). While nanophotonic waveguides have emerged as a compact and power-efficient platform for SCG, their geometric degrees of freedom have not been fully utilized due to the underlying complex nonlinear and noise-dependent stochastic physics. Here, we introduce inverse design to unlock free-form waveguides for coherent SCG. The efficacy of our design is numerically and experimentally demonstrated on Si3N4 waveguides, producing flat and coherent spectra from visible to mid-infrared wavelengths. Our work has direct applications in developing chip-based broadband light sources for spectroscopy, metrology, and sensing across multiple spectral regimes.

BackgroundFor Rheumatoid Arthritis (RA), a long-term chronic illness, it is essential to identify and describe patient subtypes with comparable goal status and molecular biomarkers. This study aims to develop and validate a new subtyping scheme that integrates genome-scale transcriptomic profiles of RA peripheral blood genes, providing a fresh perspective for stratified treatments.MethodsWe utilized independent microarray datasets of RA peripheral blood mononuclear cells (PBMCs). Up-regulated differentially expressed genes (DEGs) were subjected to functional enrichment analysis. Unsupervised cluster analysis was then employed to identify RA peripheral blood gene expression-driven subtypes. We defined three distinct clustering subtypes based on the identified 404 up-regulated DEGs.ResultsSubtype A, named NE-driving, was enriched in pathways related to neutrophil activation and responses to bacteria. Subtype B, termed interferon-driving (IFN-driving), exhibited abundant B cells and showed increased expression of transcripts involved in IFN signaling and defense responses to viruses. In Subtype C, an enrichment of CD8+ T-cells was found, ultimately defining it as CD8+ T-cells-driving. The RA subtyping scheme was validated using the XGBoost machine learning algorithm. We also evaluated the therapeutic outcomes of biological disease-modifying anti-rheumatic drugs. ConclusionsThe findings provide valuable insights for deep stratification, enabling the design of molecular diagnosis and serving as a reference for stratified therapy in RA patients in the future.

The Cambrian Terreneuvian Yuertusi Formation of the Tarim Basin, regarded as a potential hydrocarbon source rock (TOCmax = 29.8 wt%), preserves the record of the Ediacaran to Cambrian Series 2 transition. This study presents a high-resolution multi-proxy investigation of the Sugaitebulake and Yutixi sections of the NW Tarim Basin. Evidence of hydrothermal activity and euxinic conditions in the earliest Cambrian has been delineated by samples from the lower part of the Yuertusi Formation group A, featured by weakly positive Ce anomalies (Ce/Ce*avg. = 0.46), extremely positive Eu anomalies (Eu/Eu*avg. = 26.39), Y/Ho ratios (Y/Hoavg. = 40.48) and several barite layers. Mo and U covariation suggests that the Yuertusi Formation group A and Xiaoerbulake Formation were deposited under sulfidic conditions, whereas the Ediacaran Qigebulake Formation, and Yuertusi Formation groups B and C and D accumulated under suboxic to anoxic conditions. The basin has been weakly restricted and characterized by elevated productivity (Ba-excessavg. = 6,410.42 ppm) during Terreneuvian time. Suboxic conditions in late Ediacaran time (Qigebulake Formation) became euxinic conditions in association with increased sea level and productivity sustained by hydrothermal activity and upwelling, which have been preserved in the organic-rich lower part of the Yuertusi Formation. Suboxic conditions were reestablished during the Cambrian Terreneuvian time as reflected in the geochemistry of the upper part of the Yuertusi Formation. Euxinic conditions were once again established early in Cambrian Series 2 Xiaoerbulake Formation. Euxinic conditions were coincident with transgressions of the South Tianshan Ocean during that interval. The accumulation and preservation of organic matter of the Early Cambrian northern Tarim Platform reflect the interaction of hydrothermal activity, coastal upwelling, and sea level change.

Quantum fluctuations disrupt the cyclic motions of dissipative Kerr solitons (DKSs) in nonlinear optical microresonators and consequently cause timing jitter of the emitted pulse trains. This problem is translated to the performance of several applications that employ DKSs as compact frequency comb sources. Recently, device manufacturing and noise reduction technologies have advanced to unveil the quantum properties of DKSs. Here we investigate the quantum decoherence of DKSs existing in normal-dispersion microresonators known as dark pulses. By virtue of the very large material nonlinearity, we directly observe the quantum decoherence of dark pulses in an AlGaAs-on-insulator microresonator, and the underlying dynamical processes are resolved by injecting stochastic photons into the microresonators. Moreover, phase correlation measurements show that the uniformity of comb spacing of quantum-limited dark pulses is better than 1.2 × 10-16 and 2.5 × 10-13 when normalized to the optical carrier frequencies and repetition frequencies, respectively. Comparing DKSs generated in different material platforms explicitly confirms the advantages of dark pulses over bright solitons in terms of quantum-limited coherence. Our work establishes a critical performance assessment of DKSs, providing guidelines for coherence engineering of chip-scale optical frequency combs.

In recent years, several researchers have introduced spherical video-based virtual reality (SVVR) into classroom teaching to help students learn different subjects. In SVVR learning, learners are typically presented with great autonomy over their learning process. Therefore, learners should engage in self-regulated strategy (SRS) learning in order to successfully study in SVVR. However, learners often struggle to self-regulate their learning. Therefore, we implemented a SRS intervention in SVVR. In this study, a SVVR environment was developed to situate students in art history contexts; moreover, the SRS was employed to guide students to improve their performance and to make reflections on their own performance. In order to evaluate the proposed approach's effectiveness, we carried out an experiment in a university art history class. The experiment results reveal more positive effects of the SRS-based SVVR approach compared with the non-SRS-based SVVR approach in terms of the learners' learning achievements, self-regulation, meta-cognitive awareness, as well as self-efficacy. In addition, this study specified that this approach would not have an effect on students' cognitive load. As a result, the SRS-based SVVR approach proposed in the current study was effective in art history education.

According to previous studies, traditional laboratory safety courses are delivered in a classroom setting where the instructor teaches and the students listen and read the course materials passively. The course content is also uninspiring and dull. Additionally, the teaching period is spread out, which adds to the instructor's workload. As a result, students become less motivated to learn. In contrast, artificially intelligent educational robots (AIERs), help students learn while lessening the workload on instructors by enhancing teaching strategies, using robots to substitute for teachers, giving students access to a variety of instructional content, and improving interaction with students through the use of intelligent voice interactions and Q&A systems to promote student engagement in learning. If the robot is used for a long time for learning, it may lead to a decrease in students' interest in learning. Therefore, this study introduces the GAFCC model (the theory-driven gamification goal, access, feedback, challenge, collaboration design model) as an instructional design model to guide the development of a gamified AIER system, aiming to improve students' motivation and learning effectiveness for laboratory safety courses. To test the effectiveness of the system, this study conducted an experimental study at a university in China in the summer of 2022. 53 participants participated in the research, with a random sample taken from each group. Each participant was able to choose the time of their free time to engage in the experiment. There were 18, 19, and 16 participants in experimental group 1, experimental group 2, and the traditional group, respectively. Students in experimental group 1 learned using the gamified AIER system, students in experimental group 2 learned on a general anthropomorphic robot system and the control group received traditional classroom learning. The experimental results showed that compared to the other two groups, the gamified AIER system guided by the GAFCC model significantly improved students' learning achievement and enhanced their learning motivation, flow experience, and problem-solving tendency. In addition, students who adopted this approach exhibited more positive behaviors and reduced cognitive load in the learning process.

Abstract Traditional palladium‐catalyzed dearomatization of (hetero)arenes takes place via an ionic pathway and usually requires elevated temperatures to overcome the energy barrier of the dearomative insertion step. Herein, a combination of the radical and two‐electron pathways is disclosed, which enables room temperature dearomative 3D transformations of nonactivated phenyl rings with Pd(0) as the catalyst. Experimental results together with density functional theory (DFT) calculations indicate a versatile π‐allyl Pd(II) species, cyclohexadienyl Pd(II), possibly is involved in the dearomatization. This species is generated by combining the cyclohexadienyl radical and Pd(I). The cyclohexadienyl Pd(II) provides chemoselective (carboamination and trieneylation), regioselective (1,2‐carboamination), and diastereoselective (carbonyl‐group directed face selectivity) conversions.

Summary: Glioblastoma is a highly aggressive malignant tumor of the central nervous system, but the interaction between glioblastoma and different types of neurons remains unclear. Here, we established a co-culture model in vitro using 3D printed molds with microchannels, in which glioblastoma organoids (GB), dorsal forebrain organoids (DO, mainly composed of excitatory neurons), and ventral forebrain organoids (VO, mainly composed of inhibitory neurons) were assembled. Our results indicate that DO has a greater impact on altered gene expression profiles of GB, resulting in increased invasive potential. GB cells preferentially invaded DO along axons, whereas this phenomenon was not observed in VO. Furthermore, GB cells selectively inhibited neurite outgrowth in DOs and reduced the expression of the vesicular GABA transporter (VGAT), leading to neuronal hyperexcitability. By revealing how glioblastoma interacts with brain cells, our study provides a more comprehensive understanding of this disease.

Abstract Background Although body mass index (BMI) and eye rubbing are linked to an increased risk of keratoconus (KC), the interactive effect of eye rubbing and BMI on KC is largely unknown. This study aimed to evaluate the independent and interactive effects of BMI and eye rubbing on KC and to further explore the role of environmental factors on the occurrence of KC. Methods A total of 621 individuals (291 KC patients and 330 control individuals) were enrolled in this hospital‑based study on KC patients in Central China after individuals missing BMI data were excluded. BMI was calculated as weight in kilograms divided by the square of height in meters. Data on eye rubbing was recorded through face-to-face interviews. Generalized linear regression models were used to analyze associations among BMI, eye rubbing and KC. Interaction plots were used to describe the interactive effects of BMI and eye rubbing on KC. Abstract Background The catenin beta 1 gene (CTNNB1) plays a crucial role in the malignant progression of various cancers. Recent studies have suggested that CTNNB1 hyperactivation is closely related to the occurrence and development of bladder cancer (BCa). As a member of the deubiquitinating enzyme (DUB) family, ubiquitin C-terminal hydrolase L3 (UCHL3) is abnormally expressed in various cancers. In this study, we discovered that UCHL3 is a novel oncogene in bladder cancer, suggesting it is a promising target against bladder cancer. Methods We utilized CRISPR‒Cas9 technology to construct cell lines with UCHL3 stably overexpressed or knocked out. The successful overexpression or knockout of UCHL3 was determined using Western blotting. Then, we performed CCK-8, colony formation, soft agar and Transwell migration assays to determine the impact of the UCHL3 gene on cell phenotype. RNA-seq was performed with UCHL3-depleted T24 cells (established via CRISPR–Cas9-mediated genomic editing). We analyzed differences in WNT pathway gene expression in wild-type and UCHL3-deficient T24 cell lines using a heatmap and by gene set enrichment analysis (GSEA). Then, we validated the effect of UCHL3 on the Wnt pathway using a dual fluorescence reporter. We then analyzed the underlying mechanisms involved using Western blots, co-IP, and immunofluorescence results. We also conducted nude mouse tumor formation experiments. Moreover, conditional UCHL3-knockout mice and bladder cancer model mice were established for research. Results We found that the overexpression of UCHL3 boosted bladder cancer cell proliferation, invasion and migration, while the depletion of UCHL3 in bladder cancer cells delayed tumor tumorigenesis in vitro and in vivo. UCHL3 was highly associated with the Wnt signaling pathway and triggered the activation of the Wnt signaling pathway, which showed that its functions depend on its deubiquitination activity. Notably, Uchl3-deficient mice were less susceptible to bladder tumorigenesis. Additionally, UCHL3 was highly expressed in bladder cancer cells and associated with indicators of advanced clinicopathology. Conclusion In summary, we found that UCHL3 is amplified in bladder cancer and functions as a tumor promoter that enhances proliferation and migration of tumor cells in vitro and bladder tumorigenesis and progression in vivo. Furthermore, we revealed that UCHL3 stabilizes CTNNB1 expression, resulting in the activation of the oncogenic Wnt signaling pathway. Therefore, our findings strongly suggest that UCHL3 is a promising therapeutic target for bladder cancer.

Dynamic liquid level monitoring and measurement in oil wells is essential in ensuring the safe and efficient operation of oil extraction machinery and formulating rational extraction policies that enhance the productivity of oilfields. This paper presents an intelligent infrasound-based measurement method for oil wells’ dynamic liquid levels; it is designed to address the challenges of conventional measurement methods, including high costs, low precision, low robustness and inadequate real-time performance. Firstly, a novel noise reduction algorithm is introduced to effectively mitigate both periodic and stochastic noise, thereby significantly improving the accuracy of dynamic liquid level detection. Additionally, leveraging the PyQT framework, a software platform for real-time dynamic liquid level monitoring is engineered, capable of generating liquid level profiles, computing the sound velocity and liquid depth and visualizing the monitoring data. To bolster the data storage and analytical capabilities, the system incorporates an around-the-clock unattended monitoring approach, utilizing Internet of Things (IoT) technology to facilitate the transmission of the collected dynamic liquid level data and computed results to the oilfield’s central data repository via LoRa and 4G communication modules. Field trials on dynamic liquid level monitoring and measurement in oil wells demonstrate a measurement range of 600 m to 3000 m, with consistent and reliable results, fulfilling the requirements for oil well dynamic liquid level monitoring and measurement. This innovative system offers a new perspective and methodology for the computation and surveillance of dynamic liquid level depths.

To improve our understanding of the selfing of G. lingzhi basidiospore monokaryons and increase the efficiency of breeding excellent strains, 52 basidiospore monokaryons were isolated from a commercial G. lingzhi strain (laboratory number P). A severe partial segregation was observed using the chi-square test, the growth rate of the monokaryotic strains was normally distributed, and colonies exhibited 5 forms. The genetic diversity of the monokaryotic strains was further demonstrated by intersimple sequence repeat (ISSR) analysis, and the similarity coefficient was in the range of 0.49–1, which was consistent with the genotype classification results. In total, 14 AxBx monokaryotic strains were randomly selected for selfing with the 1 AyBy strain when the similarity coefficient was 0.76, and a total of 14 offspring were obtained via selfing, all of which were incompatible with their parents. The traits of the selfing progenies were diverse. The mycelial growth rate, fruiting body yield, and polysaccharide, triterpene, and sterol contents were the main indices. According to the membership function value, 71.43% of the selfing progeny were super parent, and the A88 strain with the best comprehensive traits was selected. These findings prove that ISSR molecular marker-assisted breeding reduces blindness, greatly reduces workload, and improves work efficiency.
