Many have dedicated their time trying to determine the ideal conditions for a cylinder to have equal probabilities of falling with one of its faces facing upwards or on its side. However, to this day, there is no concrete analysis of what these conditions should be. In order to determine such circumstances, a theoretical analysis was conducted, considering approaches from Rigid Body Dynamics and Statistical Mechanics. An experimental system was also built to improve control over the launches, and a comparative analysis was performed between the results obtained experimentally and the theory. It was concluded that the environment and other launching conditions have a significant influence; nevertheless, it is possible, under controlled conditions, to determine, within certain limits, the expected probabilities.

Malaria remains a global health challenge, necessitating the development of effective vaccines. The RTS,S vaccination prevents Plasmodium falciparum (Pf) malaria but is ineffective against Plasmodium vivax (Pv) disease. Herein, we evaluated the murine immunogenicity of a recombinant PvCSP incorporating prevalent polymorphisms, adjuvanted with Alhydrogel or Poly I:C. Both formulations induced prolonged IgG responses, with IgG1 dominance by the Alhydrogel group and high titers of all IgG isotypes by the Poly I:C counterpart. Poly I:C-adjuvanted vaccination increased splenic plasma cells, terminally-differentiated memory cells (MBCs), and precursors relative to the Alhydrogel-combined immunization. Splenic B-cells from Poly I:C-vaccinated mice revealed an antibody-secreting cell- and MBC-differentiating gene expression profile. Biological processes such as antibody folding and secretion were highlighted by the Poly I:C-adjuvanted vaccination. These findings underscore the potential of Poly I:C to strengthen immune responses against Pv malaria.

Abstract Artificial neural networks are becoming an integral part of digital solutions to complex problems. However, employing neural networks on quantum processors faces challenges related to the implementation of non-linear functions using quantum circuits. In this paper, we use repeat-until-success circuits enabled by real-time control-flow feedback to realize quantum neurons with non-linear activation functions. These neurons constitute elementary building blocks that can be arranged in a variety of layouts to carry out deep learning tasks quantum coherently. As an example, we construct a minimal feedforward quantum neural network capable of learning all 2-to-1-bit Boolean functions by optimization of network activation parameters within the supervised-learning paradigm. This model is shown to perform non-linear classification and effectively learns from multiple copies of a single training state consisting of the maximal superposition of all inputs.

Cationic gemini surfactants have emerged as potential gene delivery agents as they can co-assemble with DNA due to a strong electrostatic association. Commonly, DNA complexation is enhanced by the inclusion of a helper lipid (HL), which also plays a key role in transfection efficiency. The formation of lipoplexes, used as non-viral vectors for transfection, through electrostatic and hydrophobic interactions is affected by various physicochemical parameters, such as cationic surfactant:HL molar ratio, (+/−) charge ratio, and the morphological structure of the lipoplexes. Herein, we investigated the DNA complexation ability of mixtures of serine-based gemini surfactants, (nSer)2N5, and monoolein (MO) as a helper lipid. The micelle-forming serine surfactants contain long lipophilic chains (12 to 18 C atoms) and a five CH2 spacer, both linked to the nitrogen atoms of the serine residues by amine linkages. The (nSer)2N5:MO aggregates are non-cytotoxic up to 35–90 µM, depending on surfactant and surfactant/MO mixing ratio, and in general, higher MO content and longer surfactant chain length tend to promote higher cell viability. All systems efficaciously complex DNA, but the (18Ser)2N5:MO one clearly stands as the best-performing one. Incorporating MO into the serine surfactant system affects the morphology and size distribution of the formed mixed aggregates. In the low concentration regime, gemini–MO systems aggregate in the form of vesicles, while at high concentrations the formation of a lamellar liquid crystalline phase is observed. This suggests that lipoplexes might share a similar bilayer-based structure.

This work seeks to present an investigation about the trajectories followed by rays of light passing through refractive index gradients that was entirely carried by high school students. Such trajectories are curved, therefore contradicting the common sense that light should always travel along straight lines. This fact causes the formation of distorted and striated images, similarly to a type of mirage known as Fata Morgana. Using a rectangular aquarium containing solutions with different gradients of sugar or water with a temperature gradient, we analyzed the conditions for image inversion to occur. Also, we were able to reconstruct a distorted image through our theoretical predictions.

The search for antivirals against SARS-CoV-2 continue due to the emergence of variants of concerns, able to escape the vaccinal humoral response. In this work, authors pre-clinically explore the potential of kinetin against SARS-CoV-2, which could be used alone or in combination with other antivirals.

Chondrosia reniformis is a collagen-rich marine sponge that is considered a sustainable and viable option for producing an alternative to mammalian-origin collagens. However, there is a lack of knowledge regarding the properties of collagen isolated from different sponge parts, namely the outer region, or cortex, (ectosome) and the inner region (choanosome), and how it affects the development of biomaterials. In this study, a brief histological analysis focusing on C. reniformis collagen spatial distribution and a comprehensive comparative analysis between collagen isolated from ectosome and choanosome are presented. The isolated collagen characterization was based on isolation yield, Fourier-transformed infrared spectroscopy (FTIR), circular dichroism (CD), SDS-PAGE, dot blot, and amino acid composition, as well as their cytocompatibility envisaging the development of future biomedical applications. An isolation yield of approximately 20% was similar for both sponge parts, as well as the FTIR, CD, and SDS-PAGE profiles, which demonstrated that both isolated collagens presented a high purity degree and preserved their triple helix and fibrillar conformation. Ectosome collagen had a higher OHpro content and possessed collagen type I and IV, while the choanosome was predominately constituted by collagen type IV. In vitro cytotoxicity assays using the L929 fibroblast cell line displayed a significant cytotoxic effect of choanosome collagen at 2 mg/mL, while ectosome collagen enhanced cell metabolism and proliferation, thus indicating the latter as being more suitable for the development of biomaterials. This research represents a unique comparative study of C. reniformis body parts, serving as a support for further establishing this marine sponge as a promising alternative collagen source for the future development of biomedical applications.

Quantum key distribution (QKD) for information-theoretic secure distribution of cryptographic keys is one of the best known applications that will be enabled by the Quantum Internet. Here, the authors deploy the protocol of measurement-device-independent quantum key distribution into a realistic network environment, testing its field performances and showing the co-existing of this QKD protocol and the classical network infrastructure.

IntroductionScale insects (Hemiptera: Coccomorpha) feed on a wide variety of agricultural crops and forest and ornamental trees worldwide. These pest insects damage plants not only by causing sap loss but also by reducing the plant's photosynthetic activity. This is because the honeydew they produce acts as a substrate for mold, which covers leaf surfaces. In the last decades, several outbreaks of Physokermes spp. (soft-scale insects) have occurred throughout Europe and have partly been attributed to unusual weather conditions or climate change, as some species seem to be expanding their distribution range. However, the small size of these insects and their large intraspecific morphological variation have hindered the identification of the species responsible for outbreaks.MethodsIn this study, mitochondrial DNA (cytochrome c oxidase subunit I, COI), ribosomal RNA (28S), and nuclear (elongation factor 1α, EF1α) DNA markers were used to reconstruct the phylogenetic relationships of four Physokermes species sampled throughout Europe in 2013–2015.Results and discussionThe results allowed us to clearly distinguish P. hellenicus and P. inopinatus from each other, as these appeared in well-supported clades in the phylogenetic trees and from P. piceae and P. hemicryphys. However, P. hemicryphus appeared in a single clade in trees based on 28S and EF1α but among P. piceae in the COI tree. Further investigations are therefore required to determine the taxonomic status of P. piceae and P. hemicryphys, which seem to comprise a species complex.

Amyotrophic lateral sclerosis is the most common adult-onset neurodegenerative disease affecting motor neurons. Its defining feature is progressive loss of motor neuron function in the cortex, brainstem, and spinal cord, leading to paralysis and death. Despite major advances in identifying genes that can cause disease when mutated and model the disease in animals and cellular models, it still remains unclear why motor symptoms suddenly appear after a long pre-symptomatic phase of apparently normal function. One hypothesis is that age-related deregulation of specific proteins within key cell types, especially motor neurons themselves, initiates disease symptom appearance and may also drive progressive degeneration. Genome-wide in vivo cell-type-specific screening tools are enabling identification of candidates for such proteins. In this minireview, we first briefly discuss the methodology used in a recent study that applied a motor neuron-specific RNA-Seq screening approach to a standard model of TAR DNA-binding protein-43 (TDP-43)-driven amyotrophic lateral sclerosis. A key finding of this study is that synaptogyrin-4 and pleckstrin homology domain-containing family B member 1 are also deregulated at the protein level within motor neurons of two unrelated mouse models of mutant TDP-43 driven amyotrophic lateral sclerosis. Guided by what is known about molecular and cellular functions of these proteins and their orthologs, we outline here specific hypotheses for how changes in their levels might potentially alter cellular physiology of motor neurons and detrimentally affect motor neuron function. Where possible, we also discuss how this information could potentially be used in a translational context to develop new therapeutic strategies for this currently incurable, devastating disease.

Summary: We present an optimized protocol set to study the production of drug metabolites in different in vitro systems. We detail the necessary steps to identify the metabolites of xenobiotics produced in different metabolic-competent systems, from purified enzymes to primary cell cultures. It is coupled to a high-resolution mass spectrometry analytical approach and can be adapted to study any xenobiotic. This protocol was optimized using montelukast, an antagonist of the cysteinyl leukotriene receptor 1, widely used for asthma management.For complete details on the use and execution of this protocol, please refer to Marques et al. Summary: We present here an optimized protocol to obtain primary neuron-enriched cultures from embryonic chicken brains with no need for an animal facility. The protocol details the steps to isolate a neuron-enriched cell fraction from chicken embryos, followed by characterization of the chicken neurons with mass spectrometry proteomics and cell staining. Abstract Malaria is a highly prevalent parasitic disease in regions with tropical and subtropical climates worldwide. Among the species of Plasmodium causing human malaria, P. vivax is the second most prevalent and the most geographically widespread species. A major target of a pre-erythrocytic vaccine is the P. vivax circumsporozoite protein (PvCSP). In previous studies, we fused two recombinant proteins representing three allelic variants of PvCSP (VK210, VK247 and P. vivax-like) to the mumps virus nucleocapsid protein to enhance immune responses against PvCSP. The objective of the present study was to evaluate the protective efficacy of these recombinants in mice challenged with transgenic P. berghei parasites expressing PvCSP allelic variants. Formulations containing Poly (I:C) or Montanide ISA720 as adjuvants elicited high and long-lasting IgG antibody titers specific to each PvCSP allelic variant. Immunized mice were challenged with two existing chimeric P. berghei parasite lines expressing PvCSP-VK210 and PvCSP-VK247. We also developed a novel chimeric line expressing the third allelic variant, PvCSP-P. vivax-like, as a new murine immunization-challenge model. Our formulations conferred partial protection (significant delay in the time to reach 1% parasitemia) against challenge with the three chimeric parasites. Our results provide insights into the development of a vaccine targeting multiple strains of P. vivax.

Mg-Zn-Zr (ZK) alloys exhibit notably high mechanical strength amongst all magnesium alloy grades. However, due to the formation of low melting point Mg3Zn7-precipitates, these alloys are susceptible to hot cracking, thus compromising their metallurgical processing. The addition of rare earths to ZK alloys is an alternative to form higher melting point intermetallic compounds, speed up dynamic recrystallization, refine grain size, enhance corrosion resistance and extend the service temperature due to improved creep resistance. This work deals with the effect of Ce-base mischmetal addition on the hot rolling behaviour of as-cast ZK60 alloy. The microstructure investigation conducted using electron microscopy and X-Ray diffraction shows that precipitation of Mg7Zn3 intermetallics occur during hot rolling, whereas no further precipitation is observed for the ZK60-Mm alloys. The fragmentation of the intermetallic compounds occur during hot rolling and finer particles of Mg7Zn3 are observed for the ZK60, whereas Mg7Zn3 and MgZn2Ce intermetallics are formed in the alloy modified with mischmetal addition. A higher fraction of dynamically recrystallized grains is observed for the ZK60-Mm in comparison to the ZK60. Continuous recrystallization takes place in ZK60 with the formation of sub-grains near to the intermetallics and the addition of mischmetal promotes the occurrence of discontinuous recrystallization with the nucleation of new grains close to the precipitates. The mechanical strength and, in particular, the ductility of the hot-rolled alloys are notably improved when compared to the same alloys in the as-cast condition. The mechanical strength is, however, higher for the ZK60 alloy. Less solid solution strengthening, softer MgZn2Ce intermetallics and more extensive recrystallization contribute to reduce the mechanical strength of ZK60-Mm. Failure in both alloys are initiated at coarse intermetallics and propagate through intermetallic-rich regions.

Most polymeric materials are synthetic and derived from petroleum, hence they accumulate in landfills or the ocean, and recent studies have focused on alternatives to replace them with biodegradable materials from renewable sources. Biodegradable wastes from food and agroindustry, such as spent coffee grounds (SCGs), are annually discarded on a large scale and are rich in organic compounds, such as polysaccharides, that could be used as precursors to produce films. Around 6.5 million tons of SCGs are discarded every year, generating an environmental problem around the world. Therefore, it was the aim of this work to develop films from the SCGs polysaccharide fraction, which is comprised of cellulose, galactomannans and arabinogalactans. Two types of crosslinking were performed: the first forming coordination bonds of calcium ions with polysaccharides; and the second through covalent bonds with 1,4-phenylenediboronic acid (PDBA). The films with Ca2+ ions exhibited a greater barrier to water vapor with a reduction of 44% of water permeability vapor and 26% greater tensile strength than the control film (without crosslinkers). Films crosslinked with PDBA presented 55–81% higher moisture contents, 85–125% greater permeability to water vapor and 67–150% larger elongations at break than the films with Ca2+ ions. Film biodegradability was demonstrated to be affected by the crosslinking density, with the higher the crosslinking density, the longer the time for the film to fully biodegrade. The results are promising and suggest that future research should focus on enhancing the properties of these films to expand the range of possible applications.

Untargeted metabolomics approaches require complex samples containing the endogenous metabolites of a biological system. Here, we describe a set of protocols that can be applied to various types of samples, including prokaryotic and eukaryotic cells, as well as animal and human samples. Following a single extraction step, samples are analysed using different chromatographic conditions coupled to high-resolution mass spectrometry. Quantification of metabolite changes between samples is performed without internal standards, using peak areas from extracted ion chromatograms for statistical analysis. Bioinformatics annotation of the results allows a pathway- and process-oriented analysis across biological sample conditions, allowing a complete pathway interrogation.

Cytokines and other growth factors are essential for cell expansion, health, function, and immune stimulation. Stem cells have the additional reliance on these factors to direct differentiation to the appropriate terminal cell type. Successful manufacturing of allogeneic cell therapies from induced pluripotent stem cells (iPSCs) requires close attention to the selection and control of cytokines and factors used throughout the manufacturing process, as well as after administration to the patient. This paper employs iPSC-derived natural killer cell/T cell therapeutics to illustrate the use of cytokines, growth factors, and transcription factors at different stages of the manufacturing process, ranging from the generation of iPSCs to controlling of iPSC differentiation into immune-effector cells through the support of cell therapy after patient administration.

Collagen is the major structural protein in extracellular matrix present in connective tissues, including skin, being considered a promising material for skin regeneration. Marine organisms have been attracting interest amongst the industry as an alternative collagen source. In the present work, Atlantic codfish skin collagen was analyzed, to evaluate its potential for skincare. The collagen was extracted from two different skin batches (food industry by-product) using acetic acid (ASColl), confirming the method reproducibility since no significant yield differences were observed. The extracts characterization confirmed a profile compatible with type I collagen, without significant differences between batches or with bovine skin collagen (a reference material in biomedicine). Thermal analyses suggested ASColl’s native structure loss at 25 °C, and an inferior thermal stability to bovine skin collagen. No cytotoxicity was found for ASColl up to 10 mg/mL in keratinocytes (HaCaT cells). ASColl was used to develop membranes, which revealed smooth surfaces without significative morphological or biodegradability differences between batches. Their water absorption capacity and water contact angle indicated a hydrophilic feature. The metabolic activity and proliferation of HaCaT were improved by the membranes. Hence, ASColl membranes exhibited attractive characteristics to be applied in the biomedical and cosmeceutical field envisaging skincare.

Interactions between polymers (P) and surfactants (S) in aqueous solution lead to interfacial and aggregation phenomena that are not only of great interest in physical chemistry but also important for many industrial applications, such as the development of detergents and fabric softeners. Here, we synthesized two ionic derivatives—sodium carboxymethylcellulose (NaCMC) and quaternized cellulose (QC)—from cellulose recycled from textile wastes and then explored the interactions of these polymers with assorted surfactants—cationic (CTAB, gemini), anionic (SDS, SDBS) and nonionic (TX-100)—commonly used in the textile industry. We obtained surface tension curves of the P/S mixtures by fixing the polymer concentration and then increasing the surfactant concentration. In mixtures where polymer and surfactant are oppositely charged (P−/S+ and P+/S−), a strong association is observed, and from the surface tension curves, we determined the critical aggregation concentration (cac) and critical micelle concentration in the presence of polymer (cmcp). For mixtures of similar charge (P+/S+ and P−/S−), virtually no interactions are observed, with the notable exception of the QC/CTAB system, which is much more surface active than the neat CTAB. We further investigated the effect of oppositely charged P/S mixtures on hydrophilicity by measuring the contact angles of aqueous droplets on a hydrophobic textile substrate. Significantly, both P−/S+ and P+/S− systems greatly enhance the hydrophilicity of the substrate at much lower surfactant concentrations than the surfactant alone (in particular in the QC/SDBS and QC/SDS systems).

Adjuvants are essential for vaccine development, especially subunit-based vaccines such as those containing recombinant proteins. Increasing the knowledge of the immune response mechanisms generated by adjuvants should facilitate the formulation of vaccines in the future. The present work describes the immune phenotypes induced by Poly (I:C) and Montanide ISA 720 in the context of mice immunization with a recombinant protein based on the Plasmodium vivax circumsporozoite protein (PvCSP) sequence. Mice immunized with the recombinant protein plus Montanide ISA 720 showed an overall more robust humoral response, inducing antibodies with greater avidity to the antigen. A general trend for mixed Th1/Th2 inflammatory cytokine profile was increased in Montanide-adjuvanted mice, while a balanced profile was observed in Poly (I:C)-adjuvanted mice. Montanide ISA 720 induced a gene signature in B lymphocytes characteristic of heme biosynthesis, suggesting increased differentiation to Plasma Cells. On the other hand, Poly (I:C) provoked more perturbations in T cell transcriptome. These results extend the understanding of the modulation of specific immune responses induced by different classes of adjuvants, and could support the optimization of subunit-based vaccines.

A free software package for the computation of the three-dimensional normal modes of an hydrostatic atmosphere is presented. This software performs the computations in isobaric coordinates and was developed for two user-friendly languages: MATLAB and Python. The software can be used to expand the global atmospheric circulation onto the 3-D normal modes. This expansion allows the computation of a 3-D energetic scheme, which partitions the energy reservoirs and energy interactions between 3-D spatial scales, barotropic and baroclinic components, and balanced (rotational) and unbalanced (divergent) circulation fields. Moreover, by retaining only a subset of the expansion coefficients, the 3-D normal mode expansion can be used for spatial-scale filtering of atmospheric motion, filtering of balanced motion and mass unbalanced motions, and barotropic and baroclinic components. Fixing the meridional scale, the 3-D normal mode filtering can be used to isolate tropical components of the atmospheric circulation. All of these features are useful both in data analysis and in assessment of general circulation atmospheric models.

Mixtures of cationic and anionic surfactants often originate bilayer structures, such as vesicles and lamellar liquid crystals, that can be explored as model membranes for fundamental studies or as drug and gene nanocarriers. Here, we investigated the aggregation properties of two catanionic mixtures containing biomimetic surfactants derived from serine. The mixtures are designated as 12Ser/8-8Ser and 14Ser/10-10Ser, where mSer is a cationic, single-chained surfactant and n-nSer is an anionic, double-chained one (m and n being the C atoms in the alkyl chains). Our goal was to investigate the effects of total chain length and chain length asymmetry of the catanionic pair on the formation of catanionic vesicles, the vesicle properties and the vesicle/micelle transitions. Ocular observations, surface tension measurements, video-enhanced light microscopy, cryogenic scanning electron microscopy, dynamic and electrophoretic light scattering were used to monitor the self-assembly process and the aggregate properties. Catanionic vesicles were indeed found in both systems for molar fractions of cationic surfactant ≥0.40, always possessing positive zeta potentials (ζ = +35–50 mV), even for equimolar sample compositions. Furthermore, the 14Ser/10-10Ser vesicles were only found as single aggregates (i.e., without coexisting micelles) in a very narrow compositional range and as a bimodal population (average diameters of 80 and 300 nm). In contrast, the 12Ser/8-8Ser vesicles were found for a wider sample compositional range and as unimodal or bimodal populations, depending on the mixing ratio. The aggregate size, pH and zeta potential of the mixtures were further investigated. The unimodal 12Ser/8-8Ser vesicles ( ≈ 250 nm, pH ≈ 7–8, ζ ≈ +32 mV and a cationic/anionic molar ratio of ≈2:1) are particularly promising for application as drug/gene nanocarriers. Both chain length asymmetry and total length play a key role in the aggregation features of the two systems. Molecular insights are provided by the main findings.

Arthropods, the largest animal phylum, including insects, spiders and crustaceans, are characterized by their bodies being covered primarily in chitin. Besides being a source of this biopolymer, crustaceans have also attracted attention from biotechnology given their cuticles’ remarkable and diverse mechanical properties. The goose barnacle, Pollicipes pollicipes, is a sessile crustacean characterized by their body parts covered with calcified plates and a peduncle attached to a substrate covered with a cuticle. In this work, the composition and structure of these plates and cuticle were characterized. The morphology of the tergum plate revealed a compact homogeneous structure of calcium carbonate, a typical composition among marine invertebrate hard structures. The cuticle consisted of an outer zone covered with scales and an inner homogenous zone, predominantly organic, composed of successive layers parallel to the surface. The scales are similar to the tergum plate and are arranged in parallel and oriented semi-vertically. Structural and biochemical characterization confirmed a bulk composition of ɑ-chitin and suggested the presence of elastin-based proteins and collagen. The mechanical properties of the cuticle showed that the stiffness values are within the range of values described in elastomers and soft crustacean cuticles resulting from molting. The removal of calcified components exposed round holes, detailed the structure of the lamina, and changed the protein properties, increasing the rigidity of the material. This flexible cuticle, predominantly inorganic, can provide bioinspiration for developing biocompatible and mechanically suitable biomaterials for diverse applications, including in tissue engineering approaches.

Remineralization of tooth enamel can be achieved by applying a complex of casein phosphopeptides and amorphous calcium phosphate (CPP-ACP). However, the efficacy and optimization of this agent in molar–incisor hypomineralization (MIH) lacks evidence. The purpose of this study is to evaluate the efficacy of CPP-ACP tooth mousse in remineralizing MIH-affected enamel in an optimized 28-day protocol using polarized Raman microscopy and scanning electron microscopy. The protocol was applied to two types of MIH opacities, white and yellow, and compared against sound enamel specimens before and after treatment. Data was analyzed using a one-way ANOVA and LSD post hoc multiple comparisons test (p < 0.05) for the Raman analysis. Hypomineralized enamel showed an improvement of its structure after CPP-ACP supplementation. In addition, Raman spectroscopy results showed a decrease in the depolarization ratio of the symmetric stretching band of phosphate (p < 0.05 for both groups). In conclusion, there was an improvement in mineral density and organization of the hypomineralized enamel after treatment with CPP-ACP tooth mousse.

Remineralization of tooth enamel can be partially achieved by the application of a casein phosphopeptides and amorphous phosphate (CPP-ACP) complex. However, evidence to support its effectiveness in Molar-incisor-hypomineralization (MIH)-affected teeth is scarce. The study’s aim is to evaluate the efficacy of CPP-ACP mousse in remineralizing MIH-affected enamel using a Vickers microhardness test. Two groups of enamel opacities of hypomineralized permanent teeth, white (group A) and yellow (group B) lesions (n = 14), went through a 28-day treatment protocol with GC Tooth Mousse. Before and after treatment, microhardness was measured in three different areas of each tooth (hypomineralized, transition, and outside the hypomineralized area). Data were analyzed using parametric and non-parametric tests with a significance of p < 0.05. The mean microhardness values increased in the hypomineralized and transition areas in both groups after the treatment protocol (in group A, 105.38 ± 11.70 to 158.26 ± 37.34; 123.04 ± 22.84 to 156.33 ± 35.70; in group B, 108.63 ± 14.66 to 143.06 ± 22.81; 132.55 ± 20.66 to 146.00 ± 12.88) and the differences pre/post-treatment were statistically significant within each group (p < 0.001 for both groups). Between groups, there was a statistically significant difference in the same areas (hypomineralized: p = 0.003; transition: p = 0.008) with a higher improvement in enamel hardness in group A. Topical application of CPP-ACP showed an increase in the physical strength of the hypomineralized and transition areas of MIH-affected enamel, likely due to an increase in mineral content.

La presencia de toros con características seminales deficientes ocasiona fallas considerables en la eficiencia reproductiva de las unidades de producción, debido a la disminución del número de vacas preñadas, siendo más marcado en sistemas bajo programas de inseminación artificial (IA). Los rasgos de calidad seminal (CS) pueden constituir un criterio importante para la selección de machos reproductores utilizados en IA. La identificación de marcadores moleculares asociados con CS en el toro, podría facilitar la selección para estos rasgos. Este estudio tuvo como objetivo evaluar la asociación de los polimorfismos del gen Leptina, sobre la CS de toros de la raza Carora. Se evaluaron las variables volumen de eyaculado (VE), motilidad masal (MM), motilidad individual (MI) y concentración espermática (CE) de 43 toros reproductores Carora, organizados en 8 grupos de edad. Los polimorfismos nucleotídicos simples (SNP) del gen Leptina evaluados fueron: rs29004487 (SNP1), rs29004488 (SNP2), rs29004501 (SNP3) y rs29004508 (SNP4). Se utilizó un análisis de varianza mediante un modelo lineal generalizado (GLM). El factor genotipo contó con 10 niveles y el factor edad, 8 niveles. No se observó efecto del SNP1 sobre ninguna de las variables evaluadas, mientras que el SNP3 tuvo un efecto significativo sobre la CE. Los SNP2 y SNP4 presentaron un efecto altamente significativo sobre la MI y CE. Finalmente, las variables VE y MM no fueron afectadas por ninguno de los SNP estudiados. Los resultados del presente estudio sugieren marcadores potenciales para la valoración y selección genética de bovinos reproductores.

Due to their relevance to the environment and economy, coastal areas are considered national strategic segments that should be preserved. Since erosion phenomena occur intensively in those areas, it is capital to monitor them in order to identify risk zones. In addition to national and regional studies, it is also necessary to conduct local monitoring of erosion prone areas, especially those which are often frequented by people, such as beaches limited by high cliffs. Large scale vertical mapping is necessary to model their susceptibility to mass movements, in order to provide adequate prevention, protection and assistance. Recent techniques like laser scanning or aerial photogrammetry using UAVs allow the definition of the status quo of a cliff wall and its situation a few years back. But to assess the susceptibility to rock mass movements in such cliff segments, inventories of past events are of primordial importance. These inventories allow applying several statistic models to better understand susceptibility together with a set of variables of internal and external nature regarding the cliff site. We present a case study focused on the beach of Ribeira d’Ilhas (Mafra, Portugal), where a workflow of terrestrial photogrammetry for present day situation and recovery of old analogue stereoscopic pairs (1999, 2000, 2003) was implemented. A mass movement inventory (1999-2014) was compiled by multitemporal comparison followed by a detailed characterization of the cliff using a GIS software. Finally, the application of the logistic regression method allowed the definition of a susceptibility map of the cliff wall towards the occurrence of mass movements.

Increasing environmental distress is associated with a growing asthma incidence; no treatments are available but montelukast (MTK)—an antagonist of the cysteinyl leukotrienes receptor 1—is widely used in the management of symptoms among adults and children. Recently, new molecular targets have been identified and MTK has been proposed for repurposing in other therapeutic applications, with several ongoing clinical trials. The proposed applications include neuroinflammation control, which could be explored in some neurodegenerative disorders, such as Alzheimer’s and Parkinson’s diseases (AD and PD). However, this drug has been associated with an increasing number of reported neuropsychiatric adverse drug reactions (ADRs). Besides, and despite being on the market since 1998, MTK metabolism is still poorly understood and the mechanisms underlying neuropsychiatric ADRs remain unknown. We review the role of MTK as a modulator of leukotriene pathways and systematize the current knowledge about MTK metabolism. Known toxic effects of MTK are discussed, and repurposing applications are presented comprehensively, with a focus on AD and PD.

Malaria is a serious public health problem that affects mostly the poorest countries in the world, killing more than 400,000 people per year, mainly children under 5 years old. Among the control and prevention strategies, the differential diagnosis of the Plasmodium–infecting species is an important factor for selecting a treatment and, consequently, for preventing the spread of the disease. One of the main difficulties for the detection of a specific Plasmodium sp is that most of the existing methods for malaria diagnosis focus on detecting P. falciparum. Thus, in many cases, the diagnostic methods neglect the other non-falciparum species and underestimate their prevalence and severity. Traditional methods for diagnosing malaria may present low specificity or sensitivity to non-falciparum spp. Therefore, there is high demand for new alternative methods able to differentiate Plasmodium species in a faster, cheaper and easier manner to execute. This review details the classical procedures and new perspectives of diagnostic methods for malaria non-falciparum differential detection and the possibilities of their application in different circumstances.

ABSTRACT: Staphylococcus spp. plays a significant role in the etiology of bovine mastitis. Staphylococcus aureus is considered the most important species due to the high prevalence and the difficulty of in vivo treatment that is related to the expression of virulence factors and biofilm formation. This study aimed to detect the phenotypic expression of the biofilm formation in 20 S. aureus isolated from bovine mastitis and to evaluate the expression and regulation of genes involved in its production. MALDI-TOF and phenogenotypic identification assays were performed to characterize the isolates. The phenotypic biofilm production and the presence of icaA and icaD and bap genes were evaluated. The Agr system was typified (agr I, agr II, agr III and agr IV) and its regulator (agr RNAIII) was detected. Furtherly, Real-time PCR (qPCR) was performed at chosen times to quantify the expression of icaA, icaD and hld genes in three selected isolates. All 20 strains were biofilm producers and most presented icaA and icaD genes. Only one isolate presented the bap gene. The agr gene type II showed a prevalence of 70%. Transcriptional analysis revealed increased expression of ica genes at eight hours of growth. These results confirm that polysaccharides production mediated by the icaADBC operon genes is an essential mechanism to the biofilm formation and contributes to the early stages of bacterial growth.

The purpose of this in vitro study is to evaluate the efficacy of a commercially available tooth mousse in the remineralization of human enamel in cases of Molar-incisor-hypomineralization (MIH).Treatment protocol with GC Tooth Mousse containing Casein phosphopeptide amorphous calcium phosphate (CPP-ACP) was applied to 12 teeth (4 healthy, 4 hypomineralized with white opacities and 4 hypomineralized with yellow opacities) and the samples were evaluated with polarized Raman microscopy. The depolarization ratio of the symmetric stretching band of phosphate band was compared in each sample before and after treatment with the tooth mousse.The mean depolarization ratio values decreased in all three groups, after the treatment protocol, with significant differences (p
