Brown algae have the unique ability to produce phlorotannins, which are phenolics with potent antioxidant capacity. These valuable metabolites have numerous applications in medicine, cosmetics, and food industries. Therefore, it is extremely important to develop strategies to continuously produce these compounds without having to constantly harvest seaweeds from the environment. In this work, the possibility of growing Fucus spiralis under artificial conditions by tissue culture techniques was evaluated and optimized. After growing for four months, callus were excited from the original explants and evaluated for phlorotannin content by the Folin- Ciocalteu method and scavenging potential by the 2,2-diphenyl-2-picrilhidrazil (DPPH) scavenging assay. Phlorotannin content and scavenging potential were compared with wild-collected tissue. Fucus spiralis grown callus exposed to 60 μmol photons.m-2.s-1 presented the highest phlorotannins’ content and the highest scavenging potential with 250.02 ± 7.24 mg phloroglucinol equivalents/ g extract and an IC50 of 125.2 (101.3 - 139.5) μg/mL, respectively. This research represents the first evaluation of the potential of Fucus spiralis to be cultured under artificial conditions for phlorotannins extraction, opening a new window for further optimization and development of photobioreactors for continuous phlorotannins production. info:eu-repo/semantics/publishedVersion

Nowadays, consumers demand high quality food products with an extended shelf-life without chemical additives. Edible coatings (EC) containing natural compounds are a promising preservation technology for raw seafood without compromising fresh-like appeal and nutritional content.The aim of this work was to evaluate the effect of Thunnus obesus skin gelatin-based EC containing Codium spp. or Fucus vesiculosus extracts on raw tuna preservation. Three gelatin-based EC (gelatin (5 %) + glycerol (25 %); gelatin (5 %) + Codium spp. (1 %) + glycerol (25 %); gelatin (5 %) + Fucus vesiculosus (1 %) + glycerol (25 %)) were applied directly on the surface of tuna fillets. Functional properties of gelatin and gelatin-based EC containing seaweed extracts were also studied. The gelatin was extracted by an acid-swelling process in the presence/absence of pepsin, followed by subsequent heating/refrigeration, after a pre-treatment with NaOH. The type of acid, temperature and concentration of NaOH greatly influence the process yield. The higher extraction yield was achieved using acetic acid in the presence of pepsin by subsequent refrigeration, especially when skins were previously treated with NaOH (0.2 M). Tuna quality was assessed over 12 days of storage at 4 ± 1 °C in terms of chemical and microbial indices. Results showed that tuna skin gelatin-based EC avoids tuna deterioration. Microbial growth, assessed by total viable counts, and total volatile basic nitrogen were maintained below the maximum limits recommended, contrarily to the control. Additionally, the use of EC increased the stability of red colour during storage.

Objective: To evaluate the possible association between hyperuricemia and cardiorespiratory fitness levels/nutritional profile, grouped into a single variable, in schoolchildren. Method: Cross-sectional study of 2335 students from Elementary schools, aged 7–17 years of both genders, stratified by conglomerates of a municipality in Southern Brazil. Screening of antioxidant potential of dichloromethane and methanolic extracts of twenty-seven seaweeds from the Peniche coast was performed by: total phenolic contents (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and oxygen radical absorbance capacity (ORAC). Seaweeds revealing the highest antioxidant activity were screened for cytoprotective potential in MCF-7 cells, including the mitochondrial membrane potential analysis and the caspase-9 activity. High correlation was found between TPC of seaweed extracts and their scavenging capacity on DPPH and peroxyl radicals. The highest antioxidant activity was displayed by the methanolic fraction of brown seaweeds belonging to Fucales, however Ulva compressa presented the highest cytoprotective effect by blunting the apoptosis process. These results suggest that high antioxidant activity may not be directly related with high cytoprotective potential. The indiscriminate use of carbapenems in the fight against multidrug resistant Gram-negative bacteria leads to the emergence of resistance to these antimicrobial agents. We examine the in vitro activity of carbapenems and tigecycline against ESBL-producing E. coli and Klebsiella spp. isolated in a single hospital at two different periods eight years apart. Overall resistance to carbapenems ranged from 18.7% in 2007 to 19.1% in 2015/2016. We found no isolates resistant to tigecycline, but two intermediary profiles in the 2015/2016 period. Tigecycline is an important option for treating multidrug resistant Gram-negative infections and helps in the fight against global dissemination of resistance to carbapenems.

Justificativa e objetivos: O estresse ocupacional pode estar relacionado ao surgimento de doenças físicas e psíquicas. Objetiva-se avaliar a associação dos aspectos psicossociais do trabalho, de acordo com o Modelo Demanda-Controle, com a ocorrência de transtornos mentais entre os trabalhadores das Estratégias de Saúde da Família em Santa Cruz do Sul, RS. Métodos: estudo com abordagem epidemiológica, de corte transversal e quantitativo. A população alvo do estudo foi constituída por 89 profissionais, entre médicos, enfermeiros, técnicos/auxiliares de enfermagem (TE) e agentes comunitários de saúde, pertencentes às equipes das 11 Estratégias de Saúde da Família do município de Santa Cruz do Sul. Utilizou-se um questionário contendo questões sociodemográficas, ocupacionais e outras pertencentes à Job Stress Scale e ao Self Reporting Questionnaire. Resultados: Constatou-se que 30,1% dos profissionais apresentavam alta demanda e baixo controle sobre o trabalho, resultando em alta exposição ao desenvolvimento de estresse ocupacional. A prevalência de transtorno mental comum nos profissionais submetidos ao trabalho de alta exigência foi de 38%. Conclusão: Frente a esses resultados preocupantes faz-se necessário que se tomem atitudes visando a prevenção ou redução do estresse laboral e do adoecimento no trabalho. Summary The aim of this work was to study the proximate composition and the bioactive profile of Bifurcaria bifurcata. It contains 73.31 ± 0.69% of moisture, 8.57 ± 0.11 g per 100 g dry weight (d.w.) of protein, 5.81 ± 0.14 g per 100 g d.w. of lipid content and 30.15 ± 0.00 g per 100 g d.w. of ash. The polyunsaturated fatty acids were the most abundant fatty acid (FA), accounting for 2426.56 mg per 100 g which represents 41.77% of the total FA. The methanolic fraction showed high quantity of polyphenols (220.01 ± 0.010 phloroglucinol equivalents g−1 extract), DPPH radical reduction capacity (EC50:58.82 μg mL−1) and oxygen radical absorbent capacity (3151.35 ± 119.33 μmol Trolox equivalents g−1 extract). The highest antimicrobial effect was observed against Pseudomonas aeruginosa (11.3 ± 1.5 mm) and Saccharomyces cerevisiae (IC50:17.07 μg mL−1) induced by methanolic and dichloromethane fractions, respectively. Dichloromethane fraction revealed the highest antitumor activity on Caco-2 and HepG-2 cells. Bifurcaria bifurcata can be a promising source of bioactive compounds and functional ingredients.

Objective: The purpose of this study was to measure the prevalence of common mental health disorders among employees of the Family Health Strategy in Santa Cruz do Sul, RS, and to describe sociodemographic and occupational factors associated with these disorders. Methods: This was a cross-sectional, quantitative study, conducted from June to August 2013. In total, 83 employees, who worked at 11 family health units in the city, participated. Participants included doctors, nurses, technicians/nursing assistants, and community health workers. The survey was conducted using a self-administered questionnaire about sociodemographic and occupational issues as well as the matters related to the Self-Reporting Questionnaire. Results: The average age of participants was 37.1±8.9 years. The majority of community health workers were female workers who were married, had children, had a per capita income of up to 2 minimum wages, and had completed high school. The general prevalence of common mental health disorders was 19.7%. Mental health disorders were present in all occupational categories, but were more frequent among male nurses (25%), although this was not statistically significant. Conclusion: This study, in addition to characterizing the population of employees of the Family Health Strategy in Santa Cruz do Sul, RS, in terms of demographic and occupational characteristics, found that psychological distress was present, to varying degrees, in all professional categories evaluated. Given the high prevalence of mental health disorders among these employees, measures are required to ensure the protection and promotion of mental health in this setting.

Macroalgae‐associated bacteria have already proved to be an interesting source of compounds with therapeutic potential. Accordingly, the main aim of this study was to characterize Asparagopsis armata‐associated bacteria community and evaluate their capacity to produce substances with antitumor and antimicrobial potential. Bacteria were selected according to their phenotype and isolated by the streak plate technique. The identification was carried out by the RNA ribosomal 16s gene amplification through PCR techniques. The antimicrobial activities were evaluated against seven microorganisms (Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Salmonella enteritidis, Staphylococcus aureus, Saccharomyces cerevisiae, Candida albicans) by following their growth through spectrophotometric readings. Antitumor activities were evaluated in vitro on human cell lines derived from hepatocellular (HepG‐2) and breast carcinoma (MCF‐7) using the MTT method. The present work identified a total of 21 bacteria belonging to the genus Vibrio, Staphylococcus, Shewanella, Alteromonadaceae, Bacillus, Cobetia, and Photobacterium, with Vibrio being the most abundant (42.86%). The extract of Shewanella sp. ASP 26 bacterial strain induced the highest antimicrobial activity, namely against Bacillus subtilis and Staphylococcus aureus with an IC 50 of 151.1 and 346.8 μg/mL, respectively. These bacteria (Shewanella sp.) were also the ones with highest antitumor potential, demonstrating antiproliferative activity on HepG‐2 cells. Asparagopsis armata‐associated bacteria revealed to be a potential source of compounds with antitumor and antibacterial activity., Macroalgae‐associated bacteria have showed to be an interesting source of compounds with therapeutic potential. Fucus spiralis is an edible brown seaweed (SW) found in the Portuguese Coast that is still undervalued and not very well studied. However, SW are known to be nutritious and healthy foods, in part, for their potential to supply mineral nutrients to the body. Nevertheless, with the increasing levels of pollution in the coastal areas, SW can also accumulate some contaminants. Therefore, to expand the knowledge about this marine resource, the contents of Mg, Mn, Co, I, Cd, Pb, As and Hg in fresh and freeze-dried Fucus spiralis were determined before and after in vitro human digestion simulation. Magnesium was the major element with 11.86 mg/g dry weigh (DW), but it must be highlighted that I in this SW presented concentration values of approximately 135 μg/g DW. Concerning contaminants, F. spiralis seems to have low levels of Cd, Pb, and Hg. The elements’ bioaccessibility was always high, ranging between 45.8% (I – Fresh SW) and 88.5% (Hg – FD SW). The freeze-drying process seemed to enhance element bioaccessibility. Highlights: F. spiralis is a good dietary source of iodine; This seaweed presented low Cd, Pb, and Hg levels; Essential and contaminant elements displayed high bioaccessibility percentages. Cryptococcus gattii has the ability to escape from the host’s immune system through poorly understood mechanisms and can lead to the death of healthy individuals. The role of mannoproteins in C. gattii pathogenicity is not completely understood. The present work characterized a protein, Kpr1, that is essential for the maintenance of C. gattii main virulence factor, the polysaccharide capsule. Our data contribute to the understanding of the role of Kpr1 in capsule structuring, mainly by modulating the distribution of glucans in C. gattii cell wall., The yeast-like pathogen Cryptococcus gattii is an etiological agent of cryptococcosis. The major cryptococcal virulence factor is the polysaccharide capsule, which is composed of glucuronoxylomannan (GXM), galactoxylomannan (GalXM), and mannoproteins (MPs). The GXM and GalXM polysaccharides have been extensively characterized; however, there is little information about the role of mannoproteins in capsule assembly and their participation in yeast pathogenicity. The present study characterized the function of a predicted mannoprotein from C. gattii, designated Krp1. Loss-of-function and gain-of-function mutants were generated, and phenotypes associated with the capsular architecture were evaluated. The null mutant cells were more sensitive to a cell wall stressor that disrupts beta-glucan synthesis. Also, these cells displayed increased GXM release to the culture supernatant than the wild-type strain did. The loss of Krp1 influenced cell-associated cryptococcal polysaccharide thickness and phagocytosis by J774.A1 macrophages in the early hours of interaction, but no difference in virulence in a murine model of cryptococcosis was observed. In addition, recombinant Krp1 was antigenic and differentially recognized by serum from an individual with cryptococcosis, but not with serum from an individual with candidiasis. Taken together, these results indicate that C. gattii Krp1 is important for the cell wall structure, thereby influencing capsule assembly, but is not essential for virulence in vivo. Methanol, n-hexane and dichloromethane extracts of twelve marine macro-algae (Rhodophyta, Chlorophyta and Heterokontophyta divisions) from Peniche coast (Portugal) were evaluated for their antibacterial and antifungal activity. The antibacterial activity was evaluated by disc diffusion method against Bacillus subtilis (gram positive bacteria) and Escherichia coli (gram negative bacteria). Saccharomyces cerevisiae was used as a model for the antifungal activity by evaluating the growth inhibitory activity of the extracts. The high antibacterial activity was obtained by the Asparagopsis armata methanolic extract (10 mm-0.1 mg/disc), followed by the Sphaerococcus coronopifolius n-hexane extract (8 mm-0.1 mg/disc), and the Asparagopsis armata dichloromethane extract (12 mm-0.3 mg/disc) against Bacillus subtilis. There were no positive results against Escherichia coli. Sphaerococcus coronopifolius revealed high antifungal potential for n-hexane (IC50 = 40.2 µg/ml), dichloromethane (IC50 = 78.9 µg/ml) and methanolic (IC50 = 55.18 µg/ml) extracts against Saccharomyces cerevisiae growth. The antifungal potency of the Sphaerococcus coronopifolius extracts was similar with the standard amphotericin B. Asparagopsis armata and Sphaerococcus coronopifolius reveal to be interesting sources of natural compounds with antimicrobial properties.

Estudo do conceito do conservadorismo político. A dissertação analisa o conceito do conservadorismo, que é um recorte da direita política. Dado à manifesta abrangência semântica que este termo possui, este trabalho visa identificar uma organização e precisão de significados que permita o uso do mesmo como léxico da filosofia política. Para tanto, acompanha-se o contexto de seu surgimento no período revolucionário francês no final do século XVIII, as dissensões entre as ideias da esquerda e direita políticas que estão diretamente relacionadas com o verbete examinado. O estudo aborda a carga emotiva que acompanha essa categoria, as críticas progressistas, parte delas especificamente marxistas e algumas inter-relações com a psicologia política e social. O uso do termo na ciência econômica é apresentado como parte importante da cartografia de utilização léxico conservadorismo, porquanto fecunda para validar aspectos nucleares das demarcações léxicas previamente balizadas. O estudo compreendeu a análise de uma das mais importantes obras para a categoria: Reflexões sobre a Revolução na França, de Edmund Burke, assim como o texto que inaugura um debate que se tornaria recorrente na política, os Direitos do Homem, de Thomas Paine. Os estudos contemporâneos diretamente relacionados à construção semântica albergaram o capítulo 11 do livro A Companion to Contemporary Political Philosophy, de Robert E. Goodin, Philip Pettit e Thomas Pogge, cujo tema é: Conservadorismo (Conservatism), da lavra de Anthony Quinton; o verbete ―Conservadorismo‖, escrito por Tiziano Bonnazi, no Dicionário de Política de Norberto Bobbio, Nicola Matteucci e Gianfranco Pasquino; e a obra O Conservadorismo Clássico, de Laura Escorsim Netto. Estas constituíram as fontes primárias desse estudo. O trabalho se conclui destacando a relevância e o alcance que esta categoria contemporânea pode usufruir na filosofia política, constituindo um esforço para dar conta de facticidades que se apresentam conflituosas e paradoxais na realidade social. Study of the concept of political conservatism. The dissertation analyzes the concept of conservatism, which is a piece of the political right. Given the manifest semantic comprehensiveness that this term has, this work aims to identify an organization and precision of meanings that allows its use as a lexicon of political philosophy. To this aim, we explore the context of its emergence in the French revolutionary period in the late eighteenth century, which is followed by the dissensions between the ideas of the political left and right that are directly related to the concept examined. The study addresses the emotive charge that accompanies this category, the progressive and specific Marxist critiques, and some interrelationships with political and social psychologies. The use of the term in economics is presented as part of the cartography of the lexicon conservatism, whose observation is fruitful to validate inner aspects of the previously marked lexical demarcations. The study comprised the analysis of one of the most important works for the category: Reflections on the Revolution in France, by Edmund Burke, as well as the text that inaugurates the historical antinomy of conservatism, Rights of Man, by Thomas Paine. Contemporary studies directly related to semantic construction have housed chapter 11 of Robert E. Goodin's The Companion to Contemporary Political Philosophy, Philip Pettit and Thomas Pogge, whose theme is: Conservatism - from the work of Anthony Quinton; The entry "Conservatism", written by Tiziano Bonnazi, in the Dictionary of Politics of Norberto Bobbio, Nicola Matteucci and Gianfranco Pasquino; and the book The Classical Conservatism, by Laura Escorsim Netto. These were the primary sources of this study. This work concludes by highlighting the relevance and scope that this contemporary category can have in political philosophy, constituting an effort to interpret events that are conflicted and paradoxical in social reality.

We evaluated the influence of hesperidin and vitamin C (VitC) on glycemic parameters, lipid profile, and DNA damage in male Wistar rats treated with sucrose overload. Rats were divided into six experimental groups: I-water control; II-sucrose control; III-hesperidin control; IV-VitC control; V-co-treatment of sucrose plus hesperidin; VI-co-treatment of sucrose plus VitC. We measured the levels of triglycerides, total cholesterol, HDL-c, LDL-c, fasting glucose, and glycated hemoglobin (A1C). DNA damage was evaluated in blood and brain cells using the comet assay and the micronucleus test was used to evaluate chromosomal damages in the rat bone marrow. Co-treatment with VitC, but not with hesperidin, normalized the serum glucose. No effect of co-treatments was observed on A1C. The co-treatment with VitC or hesperidin did not influence the lipid profile (p>0.05). Rats co-treated with hesperidin had a significantly lower DNA damage level in blood (p0.05). Hesperidin and VitC showed different effects on sucrose and DNA damage levels. While VitC lowered the serum glucose, hesperidin reduced the DNA damage.

Background: Little has been studied on heart rate and its relationship with metabolic disorders. Objective: To identify possible association between heart rate (HR) and metabolic disorders in children and adolescents. Methods: This cross-sectional study evaluated 2.098 subjects, aged between 7 and 17 years. The variables evaluated were: HR, systolic (SBP) and diastolic blood pressure (DBP), pulse pressure (PP), double-product (DP), myocardial oxygen consumption (mVO2), lipids, glucose and uric acid levels, body mass index (BMI) and waist circumference (WC). The values of HR at rest and effort were divided into quartiles. The association between continuous values of HR and cardiometabolic indicators was tested by linear regression. Results: LDL cholesterol presented a significantly higher mean (p = 0.003) in schoolchildren with resting HR greater or equal to 91 bpm, compared to students with less than 75 bpm. Compared with the quartiles of effort HR, SBP, DBP, glucose and uric acid presented high values when HR was greater or equal than 185 bpm. SBP, glucose and HDL cholesterol demonstrated a significant association with resting HR. Uric acid was observed as a predictor of increased effort HR. Conclusion: Schoolchildren with a higher resting HR have higher mean of LDL cholesterol. For effort HR, there was an increase in blood pressure, glucose and uric acid levels. Uric acid has been shown to be a predictor of elevated effort HR. Resumo Fundamento: Pouco se tem estudado sobre frequência cardíaca e suas relações com alterações metabólicas. Objetivo: Verificar se existe associação entre frequência cardíaca e disfunções metabólicas em crianças e adolescentes. Método: Estudo transversal com 2.098 escolares, com idade entre 7 e 17 anos. As variáveis avaliadas foram: frequência cardíaca (FC), pressão arterial sistólica (PAS), diastólica (PAD) e de pulso (PP), duplo-produto (DP), consumo de oxigênio pelo miocárdio (mVO2), perfil lipídico e glicêmico, níveis de ácido úrico, índice de massa corporal (IMC) e circunferência da cintura (CC). Os valores de FC de repouso e esforço foram divididos em quartis. A associação entre os valores contínuos de FC com indicadores cardiometabólicos foi testada por meio da regressão linear. Resultados: O colesterol LDL apresentou média significativamente superior (p = 0,003) nos escolares com FC de repouso maior ou igual a 91 bpm, em comparação aos escolares que apresentaram menos de 75 bpm. Comparados com os quartis da FC de esforço, a PAS, PAD, glicose e ácido úrico apresentaram valores elevados quando a FC foi igual ou superior a 185 bpm. A PAS, a glicose e o colesterol HDL demonstraram associação significativa com a FC de repouso. Observou-se o ácido úrico como um preditor do aumento da FC de esforço. Conclusão: Escolares com FC de repouso mais elevada apresentam médias superiores de colesterol LDL. Para FC de esforço, observou-se elevação na pressão arterial, nos níveis de glicose e de ácido úrico. O ácido úrico demonstrou ser preditor da elevação da FC de esforço.

The aim of this study was the isolation and identification of epiphytic bacteria from the marine brown alga, Bifurcaria bifurcata, and the evaluation of the antioxidant and antimicrobial activity of bacteria extracts. The identification of epiphytic bacteria was determined by 16S rRNA gene sequencing. Bacteria extracts were obtained with methanol and dichloromethane (1:1) extraction. The antioxidant activity of extracts was performed by quantification of total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and oxygen radical absorbance capacity (ORAC). Antimicrobial activities were evaluated against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Salmonella enteritidis, Staphylococcus aureus, Saccharomyces cerevisiae and Candida albicans. A total of 39 Bifurcaria bifurcata-associated bacteria were isolated and 33 were identified as Vibrio sp. (48.72%), Alteromonas sp. (12.82%), Shewanella sp. (12.26%), Serratia sp. (2.56%), Citricoccus sp. (2.56%), Cellulophaga sp. (2.56%), Ruegeria sp. (2.56%) and Staphylococcus sp. (2.56%). Six (15.38%) of the 39 bacteria Bifurcaria bifurcata-associated bacteria presented less than a 90% Basic Local Alignment Search Tool (BLAST) match, and some of those could be new. The highest antioxidant activity and antimicrobial activity (against B. subtilis) was exhibited by strain 16 (Shewanella sp.). Several strains also presented high antimicrobial activity against S. aureus, mainly belonging to Alteromonas sp. and Vibrio sp. There were no positive results against fungi and Gram-negative bacteria. Bifurcaria bifurcata epiphytic bacteria were revealed to be excellent sources of natural antioxidant and antimicrobial compounds.

Chitin was extracted from Polybius henslowii, a swimming crab, captured in large quantities throughout the Portuguese coast by purse seine vessels as bycatch. After standard chitin extraction procedures, water-soluble chitosan products were obtained via two different methods: (1) N-acetylation with the addition of acetic anhydride and (2) a reaction with hydrogen peroxide. The chemical structure and molecular weight of chitosan derivatives, water-soluble chitosan (WSC) and chitooligosaccharides (COS), were confirmed by Fourier Transform Infrared Spectroscopy (FT-IR) and gel permeation chromatography (GPC). Antioxidant and metal chelation activities were evaluated, and the growth inhibition capacity was tested on four phytopatogens. The chitooligosaccharides from pereopods (pCOS) and shell body parts (sCOS) inhibited all fungal species tested, particularly Cryphonectria parasitica with 84.7% and 85.5%, respectively. Both radical scavenging and antifungal activities proved to be dose-dependent. Chitooligosaccharides with a low molecular weight (2.7, 7.4, and 10.4 Kg&middot, mol&minus, 1) showed the highest activity among all properties tested. These results suggested that chitosan derivatives from P. henslowii raw material could potentially be used against phytopathogens or as ingredient in cosmetics and other products related to oxidative stress.

International audience; The marine environment offers both economic and scientific potential which are relatively untapped from a biotechnological point of view. These environments whilst harsh are ironically fragile and dependent on a harmonious life form balance. Exploitation of natural resources by exhaustive wild harvesting has obvious negative environmental consequences. From a European industry perspective marine organisms are a largely underutilised resource. This is not due to lack of interest but due to a lack of choice the industry faces for cost competitive, sustainable and environmentally conscientious product alternatives. Knowledge of the biotechnological potential of marine organisms together with the development of sustainable systems for their cultivation, processing and utilisation are essential. In 2010, the European Commission recognised this need and funded a collaborative RTD/SME project under the Framework 7-Knowledge Based Bio-Economy (KBBE) Theme 2 Programme ‘Sustainable culture of marine microorganisms, algae and/or invertebrates for high value added products’. The scope of that project entitled ‘Sustainable Production of Biologically Active Molecules of Marine Based Origin’ (BAMMBO) is outlined. Although the Union is a global leader in many technologies, it faces increasing competition from traditional rivals and emerging economies alike and must therefore improve its innovation performance. For this reason innovation is placed at the heart of a European Horizon 2020 Strategy wherein the challenge is to connect economic performance to eco performance. This article provides a synopsis of the research activities of the BAMMBO project as they fit within the wider scope of sustainable environmentally conscientious marine resource exploitation for high-value biomolecules.

Prediabetes (intermediate hyperglycemia) is a high-risk state for diabetes that is defined by higher than normal glycemic levels that are below the level required for a diagnosis of diabetes. Prediabetes is characterized by oxidative stress, yet the associated DNA damage and cytotoxicity remain unknown to date. Therefore, we evaluated the relationship between glycemic alterations, DNA damage and cytotoxicity in the lymphocytes of individuals with pre-diabetes. Fasting plasma glucose (FPG) and glycated hemoglobin (A1C) levels were quantified and used as inclusion criteria. Anthropometric parameters were also evaluated. The cytokinesis-block micronucleus cytome assay (CBMN Cyt) was used to evaluate DNA damage and cytotoxicity. FPG correlated with A1C (r=0.562, p=0.002). Background: Cancer represents a serious threat for human health with high social and economic impacts worldwide. Therefore, the development of new anticancer drugs is of most importance. The aim of the present study was to evaluate the antitumor potential of twelve algae from Portugal coast on an in vitro model of human hepatocellular carcinoma (HepG-2 cells).Results: Both extracts of Asparagopsis armata (1000 µg/ml; 24 h) presented high cytotoxicity with 11.22 ± 2.98 and 1.51 ± 0.38 % of HepG-2 live cells, respectively. Sphaerococcus coronopifolius methanolic and dichloromethane extracts (1000 µg/ml) also generated high reduction on HepG-2 viability (14.04 ± 2.62 and 12.84 ± 3.82 % of HepG-2 live cells, respectively). The most potent anti-proliferative activity was induced by dichloromethane extract (1000 µg/ml; 24 h) of Sphaerococcus coronopifolius, Asparagopsis armata and Plocamium cartilagineum with 99.61 ± 0.27, 98.56 ± 0.81 and 85.13 ± 1.04 % of cell’s proliferation reduction, respectively. Sphaerococcus coronopifolius dichloromethane extract exhibited the highest potency both on cytotoxicity and anti-proliferation assays with an IC50 of 14.1 and 32.3 μg/ml, respectively.Conclusions: Sphaerococcus coronopifolius is a promising source of new molecules with possible application on cancer therapeutics.

FUNDAMENTO: Durante a infância e adolescência, o sedentarismo, o excesso de peso e a alimentação inadequada são fatores de risco para doenças crônicas, sobretudo obesidade, hipertensão arterial sistêmica e diabete melito. A intervenção precoce pode prevenir o desenvolvimento dessas complicações. OBJETIVO: Verificar a presença de fatores de risco cardiovasculares (obesidade e hipertensão arterial) e suas possíveis interações com a capacidade cardiorrespiratória. Métodos: Estudo transversal composto de amostra estratificada por conglomerados, de 1.666 escolares, com idades entre 7 e 17 anos, 873 (52,4%) do sexo masculino e 793 (47,6%) do sexo feminino. Avaliaram-se as pressões arteriais sistólica (PAS) e diastólica (PAD), índice de massa corporal (IMC), percentual de gordura (%G) e capacidade cardiorrespiratória. Ainda, PAS e PAD foram correlacionadas com circunferência da cintura (CC), relação cintura-quadril (RCQ), somatório de dobras cutâneas (ΣDC) e capacidade cardiorrespiratória. RESULTADOS: A avaliação do IMC dos escolares evidenciou 26,7% de sobrepeso ou obesidade e 35,9% com o percentual de gordura acima de moderadamente alto. Com relação aos níveis pressóricos, encontraram-se 13,9% e 12,1% de escolares limítrofes e hipertensos, para PAS e PAD, respectivamente. Houve associação entre hipertensão, obesidade e capacidade cardiorrespiratória. Observou-se correlação significativa em relação à PAS e PAD, para todas as variáveis analisadas, apresentando, ainda, uma relação fraca a moderada com as variáveis idade, peso, estatura, IMC e circunferência da cintura. METHODS: This was a cross-sectional study conducted in a stratified cluster sample of 1,666 schoolchildren, aged between 7 and 17 years, 873 (52.4%) of them male and 793 (47.6%) of them female. The following variables were evaluated: systolic blood pressure (SBP), diastolic blood pressure (DBP), body mass index (BMI), body fat percentage (BF %), and cardio-respiratory fitness. SBP and DBP were correlated with waist circumference (WC), waist-hip ratio (WHR), sum of skin folds (ΣSF), and cardio-respiratory fitness. RESULTS: A BMI assessment of the students showed that 26.7% of them were overweight or obese, and 35.9% had body fat percentage over moderately high. As to blood pressure, we found that 13.9% and 12.1% of the students were borderline or hypertensive, for SBP and DBP, respectively. There was an association among hypertension, obesity, and cardio-respiratory fitness. There was a significant correlation of SBP and DBP with all variables, and also a weak to moderate correlation with age, weight, height, BMI, and waist circumference. CONCLUSION: The presence of hypertension associated with obesity and its effects on cardio-respiratory fitness stress the importance of recommending, since childhood, a more active and healthy lifestyle.

Neste estudo estimou-se a distribuição e prevalência de β-lactamases de espectro estendido pertencentes às famílias TEM, SHV e CTX-M entre amostras de Escherichia coli e Klebsiella spp. no Hospital Universitário de Santa Maria, Rio Grande do Sul. Durante 14 meses, 90 microrganismos foram selecionados como prováveis produtores de ESBL. Os isolados foram submetidos a testes fenotípicos confirmatórios para a presença de ESBL. A seguir, os tipos de ESBLs presentes em cada microrganismo foram determinados através da pesquisa dos respectivos genes através da reação em cadeia da polimerase. Empregando-se o método do disco combinado, a presença de ESBLs foi confirmada em 55 (61,1%) amostras; quando o método do duplo disco foi utilizado, 57 (63,3%) amostras foramprodutoras de ESBLs. Com base na PCR, as ESBLs do tipo TEM e SHV foram mais presentes em Klebsiella pneumoniae enquanto que ESBL do tipo CTX-M foram mais presentes em Klebsiella oxytoca.In this study, the distribution and prevalence of extended-spectrum β-lactamases belonging to the TEM, SHV and CTX-M families were estimated among samples of Escherichia coli and Klebsiella spp. at the university hospital of Santa Maria, Rio Grande do Sul. Over a 14-month period, 90 microorganisms were selected as likely ESBL producers. The isolates were subjected to confirmatory phenotype tests for the presence of ESBL. Through investigating the respective genes using the polymerase chain reaction, the ESBL types present in each microorganism were then determined. Fifty-five samples (61.1%) were confirmed as ESBL-positive by means of the combined disc method, and 57 (63.3%) were found to be ESBL producers by means of the double disc method. From the polymerase chain reaction, ESBLs of TEM and SHV types were more frequently present in Klebsiella pneumoniae, while ESBL of CTX-M type was more frequently present in Klebsiella oxytoca.

As marine organisms are well known to provide a wide range of original compounds, the aim of this study was to investigate the bioactivity of the main constituents of the cosmopolitan red alga, Sphaerococcus coronopifolius. The structure of several bromoditerpenes was determined by extensive spectroscopic analysis and comparison with literature data. Five molecules were isolated and characterized which include a new brominated diterpene belonging to the rare dactylomelane family and named sphaerodactylomelol (1), along with four already known sphaerane bromoditerpenes (2–5). Antitumor activity was assessed by cytotoxicity and anti-proliferative assays on an in vitro model of human hepatocellular carcinoma (HepG-2 cells). Antimicrobial activity was evaluated against four pathogenic microorganisms: Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida albicans. Compound 4 exhibited the highest antimicrobial activity against S. aureus (IC50 6.35 μM) and compound 5 the highest anti-proliferative activity on HepG-2 cells (IC50 42.9 μM). Também se discute a vigência de conflito entre preocupação com o equilíbrio orçamentário e políticas de desenvolvimento. Na primeira seção são analisadas as receitas e despesas estaduais, com ênfase na classificação dos comportamentos assumidos por essas variáveis ao longo do período e na determinação das principais rubricas que as compunham. Também é examinada a evolução da dívida fundada, assim como sua correlação com as despesas públicas. A segunda seção analisa especificamente os gastos com a infraestrutura de transportes, nomeadamente ferrovias e estradas de rodagem. O diabetes mellitus (DM) inclui um grupo de doenças metabólicas resultante de disfunção na secreção e/ou ação da insulina. O diagnóstico de DM é definido com base na avaliação da glicemia de jejum(GJ) e hemoglobina glicada (A1C) que também informa a persistência da hiperglicemia. Entretanto novas tecnologias analíticas estão sempre sob constante investigação. A espectroscopia de absorção molecular no infravermelho com Transformada de Fourier (FT-IR) associada à análise multivariada tem surgido como uma alternativa nesse contexto. O presente estudo buscou avaliar a FT-IR, através da técnica de reflectância difusa, para quantificar glicose e A1C em um grupo de indivíduos adultos. O estudo foi composto por 46 pacientes de 32 a 59 anos, com valores GJ de 71,67 a 135 mg/dL e de A1C de 5,3 a 7,2 %. O modelo PLS-DRIFTS para GJ o apresentou RMSECV=1,37 mg/dL e R²=0,995 e para A1C, RMSECV=0,06% R²=0,993 sem exclusão de amostras e usando a faixa de 4000-2401, 2300-600 cm-1. Dessa forma foi possível concluir que FT-IR associada a análise multivariada se mostrou adequado para predizer os parâmetros bioquímicos de glicose e A1C de jejum com um erro mínimo, atendendo os requisitos analíticos da ANVISA para quantificação destes parâmetros.

Hepatic ischemia-reperfusion injury has a significant impact on liver resection and transplantation. Many strategies have been developed to reduce the effects of ischemia-reperfusion injury, including pharmacologic and ischemic preconditioning; however, studies comparing these two methods are lacking.An experimental study was performed in a swine model. Eighteen swine were randomly assigned to three different groups: an ischemic preconditioning (IschPC) group, a pharmacologic preconditioning (PharmPC) group, and a control group. All animals underwent a 40-min liver ischemia, followed by 40 min of reperfusion. The IschPC group received a short period of ischemia (10 min) and a short period of reperfusion (15 min) before prolonged ischemia. The PharmPC group received inhaled sevoflurane for 30 min before prolonged ischemia. The control group did not receive any intervention before prolonged ischemia. Blood samples and liver tissue were obtained after ischemic and reperfusion periods. Injury was evaluated by measure of DNA damage (using COMET assay) and serum biochemical markers (transaminases, alkaline phosphatase, amylase, bilirubin, and C-reactive protein [CRP]).No significant difference was found in serum biochemical markers, except for the C-reactive protein level that was lower in the PharmPC group than in the control group soon after hepatic ischemia. Soon after prolonged ischemia, DNA damage index, both in blood samples and in liver tissue samples, was similar among the groups. However, an increase in DNA damage after reperfusion was higher in the control group than in the PharmPC group (P 0.05). The increase in DNA damage in the IschPC group was half of that observed in the control, but this difference was not statistically significant.Our results suggest an early protective effect of PharmPC (lower levels of C-reactive protein soon after ischemia). The protective effect observed after reperfusion was higher with PharmPC than with ischemic preconditioning. The simultaneous use of both methods could potentiate protection for ischemia-reperfusion.

Antioxidants play an important role as Reactive Oxygen Species (ROS) chelating agents and, therefore, the screening for potent antioxidants from natural sources as potential protective agents is of great relevance. The main aim of this study was to obtain antioxidant-enriched fractions from the common seaweed Fucus spiralis and evaluate their activity and efficiency in protecting human cells (MCF-7 cells) on an oxidative stress condition induced by H₂O₂. Five fractions, F1-F5, were obtained by reversed-phase vacuum liquid chromatography. F3, F4 and F5 revealed the highest phlorotannin content, also showing the strongest antioxidant effects. The cell death induced by H₂O₂ was reduced by all fractions following the potency order F4 > F2 > F3 > F5 > F1. Only fraction F4 completely inhibited the H₂O₂ effect. To understand the possible mechanisms of action of these fractions, the cellular production of H₂O₂, the mitochondrial membrane potential and the caspase 9 activity were studied. Fractions F3 and F4 presented the highest reduction on H₂O₂ cell production. All fractions decreased both caspase-9 activity and cell membrane depolarization (except F1). Taken all together, the edible F. spiralis reveal that they provide protection against oxidative stress induced by H₂O₂ on the human MCF-7 cellular model, probably acting as upstream blockers of apoptosis. info:eu-repo/semantics/publishedVersion

Biogeography of Mexican terrestrial mollusks is poorly known, although many studies have described some of their biological aspects. This study aimed to contribute with new information on the distribution of terrestrial mollusks in this region of high altitudinal, topographic and climatic variations of Southern Tamaulipas State, México. For this, terrestrial gastropods were surveyed from December 1990 to November 2006. A total of 525 samples were obtained from 121 localities, characterized with different vegetation types. The primary zoogeographical feature was the presence of many endemic taxa, with 45 species or subspecies, representing the 43%, the highest value for Northeastern México. In addition, a total of 24 species (23%), were of Neotropical and Neartic distributions, and 15 species or subspecies (14%) exhibited Neotropical distributions. Furthermore, the family Spiraxidae showed the largest number of endemic species, with 17 species and two subspecies, and had the largest number of endemic species (27) for Northeastern México. La biogeografía de los moluscos terrestres mexicanos es pobremente conocida. Entre diciembre 1990 y noviembre 2006 se estudiaron los gasterópodos terrestres de la región sur del estado de Tamaulipas, México. Esta región posee una gran variación altitudinal, topográfica y climática. Los muestreos (525) fueron obtenidos en 121 localidades en diferentes tipos de vegetación. La principal característica zoogeográfica fue el endemismo, con 45 especies o subespecies (43% de la malacofauna, el valor más alto se dio en el noreste de México). Además, un total de 24 especies (23%) tuvo una distribución neotropical y neártica, y 15 especies o subespecies (14%) una distribución exclusivamente neotropical. La familia Spiraxidae fue la que presentó mayor endemismo (17 especies y dos subespecies). Esta familia tiene el mayor número de especies endémicas (27) en el noreste de México.

The risk of developing cardiovascular disease (CVD) is related to lifestyle (e.g. diet, physical activity and smoking) as well as to genetic factors. This study aimed at evaluating the association between CVD risk factors and DNA damage levels in children and adolescents. Anthropometry, diet and serum CVD risk factors were evaluated by standard procedures. DNA damage levels were accessed by the comet assay (Single cell gel electrophoresis; SCGE) and cytokinesis-blocked micronucleus (CBMN) assays in leukocytes. Empregando-se o método do disco combinado, a presença de ESBLs foi confirmada em 55 (61,1%) amostras; quando o método do duplo disco foi utilizado, 57 (63,3%) amostras foramprodutoras de ESBLs. Com base na PCR, as ESBLs do tipo TEM e SHV foram mais presentes em Klebsiella pneumoniae enquanto que ESBL do tipo CTX-M foram mais presentes em Klebsiella oxytoca.

This study evaluated the association between primary DNA damage and chromosomal damage with iron intake and red blood cell parameters of iron status in a sample of healthy children and adolescents from a low-socioeconomic community.The level of primary DNA damage was assessed using an alkaline comet assay and the level of chromosomal damage was assessed using the cytokinesis-block micronucleus assay. A automated complete blood count was used to evaluate red blood cell status. The intake of iron was measured using a food-recall questionnaire.According to hemoglobin levels, only 1 of the 30 subjects evaluated was anemic. Nevertheless, 43% of the sampled subjects showed decreased mean corpuscular volume in addition to an increased amount of primary DNA damage (P0.05). Mean corpuscular volume was negatively correlated with primary DNA damage (r = -0.429, P = 0.020) but not with chromosomal damage. The association between iron and primary DNA damage showed a U-shaped curve, indicating that an intake of approximately 15 mg of iron per day (up to two-fold of the dietary recommended intake) could minimize primary DNA damage in this age group. The frequency of micronuclei and nucleoplasmic bridges, indicators of chromosomal breakage/loss and chromosomal end-fusions, respectively, showed a negative correlation with iron intake. These results indicate that an intake of iron15 mg/d could increase genomic stability in binucleated lymphocytes of the same group.An intake of iron ≥ 15 mg/d can decrease DNA damage in young subjects.
