Abstract

The polyphagous pest Spodoptera frugiperda (J. E. Smith) damage a number of important crops. The fertility and the reproductive rate of S. frugiperda can be altered by the CRISPR/Cas9 mediated mutagenesis of target genes, which produces a cascade of frame-shift mutations. The gene mediating reproductive behaviour in adult moths by suppressing the female receptivity in a variety of lepidopteran pests is the sex peptide receptor gene. PCR amplification and cloning of the S. frugiperda sex peptide receptor gene (1039 bp) was carried out. Utilizing CRISPOR, an online bioinformatics tool, SFSPR exons were used as input to create 'On target maximised and off target minimised sgRNA (20 bp)'. The in vitro restriction assay using designed sgRNA resulted in band size of 790 bp which is a release from the 1039 bp CDS which further verified the designed sgRNA's efficiency. Abstract

Pushback is a mechanism for defending against Distributed Denial-of-Service (DDoS) attacks. DDoS attacks are treated as a congestion-control problem, but because most such congestion is caused by malicious hosts not obeying traditional end-to-end congestion control, the problem must be handled by the routers. Functionality is added to each router to detect and preferentially drop packets that probably belong to an attack. Upstream routers are also notified to drop such packets in order that the router's resources be used to route legitimate traffic hence term pushback. Client puzzles have been advocated as a promising countermeasure to DoS attacks in the recent years. In order to identify the attackers, the victim server issues a puzzle to the client that sent the traffic. When the client is able to solve the puzzle, it is assumed to be authentic and the traffic from it is allowed into the server. If the victim suspects that the puzzles are solved by most of the clients, it increases the complexity of the puzzles. This puzzle solving technique allows the traversal of the attack traffic throughout the intermediate routers before reaching the destination. In order to attain the advantages of both pushback and puzzle solving techniques, a hybrid scheme called Router based Pushback technique, which involves both the techniques to solve the problem of DDoS attacks is proposed. In this proposal, the puzzle solving mechanism is pushed back to the core routers rather than having at the victim. The router based client puzzle mechanism checks the host system whether it is legitimate or not by providing a puzzle to be solved by the suspected host.

Abstract

The melon fly (Zeugodacus cucurbitae) is widely distributed throughout the world with more than 81 host species in which fruit losses can range from 30 to 100 per cent. CRISPR-Cas9 mediated gene editing has the potential to bring an area wide management of this devastating pest. The sgRNA employed in a CRISPR/Cas9 experiment has a significant impact on the efficiency of mutagenesis. In this study, scarlet gene of melon fly that encodes ATP-binding cassette (ABC) transporters, which are responsible for the translocation of various eye pigments into the proper eye compartment, was selected as target gene since it can impart a visual phenotype on mutation, which is a shift from wild brick red to mutant yellow eye colour. The scarlet gene coding region of size 2454 bp was amplified, cloned using pTZ57R/T vector followed by sequencing. From the sequences obtained, three sgRNAs (sgRNA1, sgRNA2 and sgRNA3) were designed using CHOP-CHOP and CRISPOR online tools, considering different principles such as the presence of protospacer adjacent motif (PAM) region, NGG; GC content (35-50%) and the secondary structures. The in vitro transcribed sgRNAs were incubated with Cas9 protein to form Ribo nucleo protein (RNP) complex. In vitro restriction assays were done to establish that the RNP complex effectively recognizes and cleaves the PCR amplified scarlet gene by releasing fragments upon gel analysis. The sgRNA2 and sgRNA3 have shown successful in vitro restriction with fragments of 2454bp, 2055bp, 393 bp and 2454 bp, 1958 bp, 496 bp, respectively. The failure of sgRNA1 might be due to the deviation from ideal secondary structure. It is evident that despite the in silico predictions, not all sgRNAs exhibit the same cleavage effectiveness. To discover this contradiction, in vitro technique offers selection of most efficient sgRNAs that may effectively cause double-stranded break at specific targets prior to delivering genome editing agents into live cells. Abstract

Genome editing mediated by programmable CRISPR/Cas system is a new, emerging technology that holds greater potential in developing genetic pest management strategies via precision guided sterile insect technique; genetic improvement of pollinators, natural enemies etc. The seemingly innocuous prokaryotic immunity has now been translated into a mammoth site specific editing technology for its use in medical, agricultural, diagnostic fields etc. Until recently most of the genome editing work was dependent on RNA directed site specific cleavage by Cas9 enzyme followed by error prone, non-homologous end joining (NHEJ) resulting in random mutations. Later development of improved editing systems like base editor and prime editor have enabled obtaining site-specific, pre determined mutations without a double stranded break. Similarly many engineered Cas9 variants and other Cas proteins belonging to different class and types such as Cas3, Cas12a, Cas13a have improved the existing editing tool box. Currently efforts are being made to design and implement suitable gene drive methods for various pests such as spotted wing Drosophila, Drosophila suzukii Matsumura, Diamondback moth, Plutella xylostella (Linnaeus) , many species mosquito, Med fly, Ceratitis capitata Wiedem etc..
