Kube Energy is a renewable energy services company that has most of its solar power plants planned around a single or multiple off-takers (end-users) connected in a small mini-grid. What is common for these off-takers is that a large proportion of their power consumption is to cover air conditioning and water heating for offices and residences. To date, Kube has designed their systems with larger battery banks or spinning capacity in diesel generators to provide stability in the grid during the day, as well as to provide electricity during the night. This year, Kube Energy has started a project with an aim at optimizing consumption of electricity to better match generation. This mainly entails scheduling loads for when there is high production from the PV (e.g., reheating water in hot water tanks or charging electrical vehicles) or cutting loads if there is an unscheduled decrease in generation (clouds). In this effort, Kube Energy is looking at methods for forecasting consumption and generation. This thesis will explore multiple methodologies for consumption forecasting in the UNHCR offices in Kukuma, Kenya to help Kube build a power management system (PMS) that will improve the profitability of such system by reducing the gap between generation and consumption of electricity. An important feature of the PMS is to ensure adequate balance between power generation and consumption to avoid load shedding, in case power consumption is larger than power production, or other strategies when other type of load variations take place. The thesis looks at parametric and non-parametric models and compares the models’ prediction accuracy to choose the best performing or reliable one. The models used in this thesis are the Auto Regressive Integrated Moving Average (ARIMA), Error Trend and Seasonality (ETS), linear regression (simple and multiple), and Long-Short Term Memory (LSTM). The LSTM outperforms the others significantly and was selected as the most reliable one for forecasting consumption values at the site in question. The decision was based on the performance metrics of the models. The non-parametric model or LSTM had a better performance than the two remaining parametric models due to many reasons. One of the advantages of the LSTM is the ability to tune hyperparameters to obtain a well performing forecasting model, giving flexibility to the user if overparameterization is avoided.

Huntington's Huntington's disease (HD) is a progressive neurodegenerative disease. It is caused by an abnormal expansion of CAG repeats in the gene coding for the Huntingtin protein (HTT). Neurons in the striatum are affected by the toxicity of the mutation. HD is characterized by motor, cognitive and psychiatric symptoms. Studies in the lab have shown that the genes that define striatal identity and the genes involved in the mechanisms of neuronal plasticity are down-regulated in patients and mouse models of HD. The mechanism of transcriptional deregulation could involve the transcription factor Serum Response Factor (SRF), promoting synaptic plasticity and memory processes. The objective of my thesis work was to characterize the regulation of SRF in the context of HD and to evaluate its role in pathogenesis. Indeed, the analysis of SRF at the transcriptional and/or protein level shows that it is down-regulated at an early stage in the striatum of mouse models of HD. To establish a possible causal link between SRF down-regulation in the striatum and Huntington's phenotypes, we used an AAV-based viral approach to over-express SRF in the striatum of R6/1 transgenic mouse, a model of HD. Using different behavioral tests, we showed that overexpression of the full length of SRF in the striatum of transgenic mice leads to an aggravation of motor symptoms. Moreover, histological and molecular analyses (q-RT-PCR, RNAseq and Western-blot) showed no rescue of SRF targets in the striatum of R6/1 mice, despite its high overexpression. Our results suggest that the striatal regulation of SRF and its target genes is altered by mutated HTT. We then overexpressed a construct consisting of the fusion of the SRF binding domain with the VP16 transactivation domain (SRF-VP16) in the striatum of R6/1 mice. Our data indicate that although neuronal plasticity genes are up-regulated by this construct, the behavioral phenotype of R6/1 mice is not improved, probably due to the absence of a major effect of SRF-VP16 on the striatal identity genes. This could suggest that restoring the level of SRF in the HD is not sufficient to significantly rescue HD pathogenesis., La maladie de Huntington (MH) est une maladie neurodégénérative progressive. Elle est due à une expansion anormale des répétitions CAG au niveau du gène codant pour la protéine Huntingtine (HTT). Les neurones du striatum sont affectés par la toxicité de la mutation. La MH est caractérisée par des symptômes moteurs, cognitifs et psychiatriques. Des études du laboratoire ont montré que les gènes qui définissent l’identité striatale et les gènes impliqués dans les mécanismes de plasticité neuronale sont sous-régulés chez les patients et les modèles murins de la MH. Le mécanisme de dérégulation transcriptionnelle pourrait impliquer le facteur de transcription Serum Response Factor (SRF), favorisant la plasticité synaptique et les processus de mémoire. L'objectif de mes travaux de thèse a été de caractériser la régulation de SRF dans le contexte de la MH et d'évaluer son rôle dans la pathogenèse. L’analyse de SRF au niveau transcriptionnel et/ou protéique montre en effet qu’il est sous-régulé de façon précoce dans le striatum de souris modèles de la MH. Pour établir un éventuel lien de causalité entre la sous-regulation de SRF dans le striatum et les phénotypes Huntington, nous avons utilisé une approche virale basée sur un AAV pour sur-exprimer SRF dans le striatum des souris transgéniques R6/1, modèles de la MH. En utilisant différents tests comportementaux, nous avons montré que la surexpression de la forme complète de SRF dans le striatum des souris transgéniques entraîne une aggravation des symptômes moteurs. De plus, les analyses histologiques et moléculaires (q-RT-PCR, RNAseq et Western-blot) ne montrent aucun sauvetage des cibles de SRF dans le striatum des souris R6/1, malgré sa surexpression élevée. Nos résultats suggèrent que la régulation striatale de SRF et de ses gènes cibles est altérée par la HTT mutée. Nous avons alors surexprimé une construction consistant en la fusion du domaine de liaison de SRF avec le domaine de transactivation de VP16 (SRF-VP16) dans le striatum des souris R6/1. Nos données indiquent que bien que les gènes de plasticité sont sur-régulés par cette construction, le phénotype comportemental des souris R6/1 n’est pas amélioré, probablement du fait de l’absence d’effet majeur de SRF-VP16 sur les gènes de l’identité striatale. Cela pourrait suggérer que la restauration du niveau de SRF dans la MH n'est pas suffisante pour sauver de manière significative la pathogenèse de la MH.

Light-induced electron and energy transfer in molecular arrays is being extensively studied in view of artificial photosynthesis. In this context, [Ru(bpy)3]2+ derivatives have been widely used, due to their favorable photophysical properties, i.e. lifetimes in the µs range at room temperature, high emission quantum yields, and strong oxidizing and reducing capability. The electronic structural design of most ruthenium(II) complexes is of great importance by specifying the complexes’ photophysical, photochemical, and electrochemical properties. As a consequence, the main theme of this thesis is to explore the coordination properties of new tridentate ligands on the ruthenium cation. This work describes the synthesis of these new tridentate ligands based on the acridine heterocycle with different moieties at positions 4 and 5. Octahedral ruthenium(II) complexes were then constructed around these acridine-based ligands, with tuned energy levels of the functionally active 3MC and 3MLCT excited states. Very few acridine derivatives are substituted at positions 4 and 5. The synthetic strategy for the target ligands starts from acridone. The 4,5-dibromoacridine was obtained in four synthetic steps and this common precursor was subsequently used for the construction of tridentate (NNN) ligands with pyridine and pyrazole. In addition, a thioether acridine-based tridentate ligand (SNS) analogue was synthesized, and another tridentate thioether (SNS) ligand, without an acridine heterocycle, was prepared to gain some flexibility within the ligand scaffold. The corresponding homoleptic complexes with 6-membered metallacycles only adopted a facial octahedral geometry. Thus, the heteroleptic complexes were prepared in combination with a planar terpyridine ligand in order to force the constrained and rigid ligands to coordinate in a meridional fashion on the metal. The electrochemical, photophysical, and photochemical properties of all complexes were studied and the structure for most of these Ru(II) complexes was deduced by X-ray diffraction crystallography experiments. The bis-tridentate Ru(II) complexes based on the flexible (SNS) thioether ligand were photoactive: they were able to undergo a photosubstitution reaction, by replacing the tridentate thioether ligand with monodentate ligands, upon white light exposure., Le transfert d'électron et d'énergie induit par la lumière dans des dispositifs moléculaires est largement étudié dans le domaine de la photosynthèse artificielle. Dans ce contexte, les dérivés du [Ru(bpy)3]2+ ont été très utilisés, en raison de leurs propriétés photophysiques favorables, à savoir des durées de vie de l’ordre des µs à température ambiante, des rendements quantiques d’émission élevés et un fort pouvoir d’oxydation et de réduction. La conception de la structure électronique des complexes de ruthénium(II) revêt une très grande importance, car celle-ci dictera les propriétés photophysiques, photochimiques et électrochimiques de ces complexes. En conséquence, le sujet principal de cette thèse est d'explorer les propriétés de coordination de nouveaux ligands tridentes sur le cation ruthénium. Ce travail décrit la synthèse de ces nouveaux ligands basés sur l'hétérocycle acridine avec différents fragments en positions 4 et 5. Les complexes de ruthénium(II) octaédriques correspondants ont ensuite été construits autour de ces ligands à base d'acridine, avec des niveaux d'énergie ajustés des états excités 3MC et 3MLCT fonctionnellement actifs. Très peu de dérivés d’acridine sont substitués aux positions 4 et 5. La stratégie de synthèse pour les ligands cibles commence sur l’acridone. La 4,5-dibromoacridine a été obtenue en quatre étapes synthétiques et ce précurseur commun a ensuite été utilisé pour la construction de ligands tridentés (NNN) avec de la pyridine et du pyrazole. De plus, un analogue de ligand tridentate (SNS) à base de thioéther acridine a été synthétisé, et un autre ligand tridentate de thioéther (SNS), sans hétérocycle acridine, a été préparé dans le but d’acquérir une certaine flexibilité dans la structure. Les complexes homoleptiques correspondants avec des métallacycles à 6 chaînons ont tous adopté une géométrie octaédrique faciale. Ainsi, les complexes hétéroleptiques ont été préparés en combinaison avec un ligand terpyridine plan afin de forcer les ligands contraints et rigides à se coordiner de manière méridienne sur le métal. Les propriétés électrochimiques, photophysiques et photochimiques de tous les complexes ont été étudiées et la structure de la plupart de ces complexes de Ru(II) a été déduite par des expériences de cristallographie par diffraction des rayons X. Les complexes bis-tridentés de Ru(II) basés sur le ligand thioéther flexible (SNS), se sont montrés photoactifs: lors d'une exposition à la lumière blanche ils subissent une réaction de photosubstitution, remplaçant le ligand thioéther tridentate par des ligands monodentates.

Spintronik ergänzt die konventionelle Halbleiterelektronik bereits an vielen Stellen der Informationstechnologie, die dadurch neben dem elektronischen Freiheitsgrad der Ladung auch den des Elektronenspins nutzen kann. Die klassische Trennung von elektronischer Datenverarbeitung und magnetischer Speicherung in der Informationstechnologie wurde mit der Entdeckung magnetoresistiver Effekte in magnetischen Schichtsystemen aufgehoben - an dieser Stelle treten die beiden Elektroneneigenschaften Spin und Ladung in Wechselwirkung. Die enge Verknüpfung der magnetischen und elektrischen Eigenschaften erfordert ein genaues Verständnis der elektronischen Vorgänge in dotierten Metalloxid-Halbleitern, um Materialien wie die zwei Verbindungen SnO2 und TiO2 gezielter für spintronische Anforderungen zu modellieren. Zu diesem Zweck wurden Untersuchungen an diesen Materialien unternommen, um die grundlegenden Prozesse der Schichtherstellung, der Nachbehandlungen und die morphologische Charakterisierung bis zu deren Auswirkungen auf die elektrischen Transportmechanismen und die magnetische Phänomenologie zu studieren. In der vorliegenden Arbeit werden die Herstellung von dünnen Zinndioxid- und Titandioxid-Schichten und die Resultate verschiedener physikalischer Charakterisierungsmethoden beschrieben. Die beiden Metalloxide wurden mit einem Sputterverfahren und mit einem thermischen Verdampfungsverfahren als dünne Schichten aufgebracht. Es wurden Mikroskop- und Röntgen-Beugungsuntersuchungen durchgeführt, um die Morphologie der Materialien zu untersuchen. Einige SnO2-Proben wurden einem Annealing unterzogen, die TiO2-Schichten wurden nachträglich mit Fe-Ionen dotiert. Die Untersuchungen konnten in einigen Fällen ferromagnetische Ordnung in TiO2:Fe mit Ordnungstemperaturen über 300 K bestätigen. Die Leitfähigkeitsmessungen der Schichten identifizieren den dominierenden Leitungsmechanismus als Variable Range Hopping zwischen lokalisierten Zuständen tief in der Bandlücke., Spintronics already increase the functionality of conventional semiconductor electronics, by utilizing the degree of freedom of the electrons charge and spin at the same time. The traditional separation of electronical data processing and magnetical data storage has been overturned by the discovery of magneto resistive effects in magnetic layer structures - at this point the spin and the charge of the electron interact. To use compounds like SnO2 and TiO2 for spintronic devices a detailed understanding of the electronical processes in such materials is required. Mainly that is the close interconnection between magnetical and electronical properties in doped metal oxide semiconductors. For this purpose the fundamental processes of thin film deposition, the effects of secondary treatment and the morphological properties of these materials have been investigated, to study the influence on the electrical transport and the magnetical phenomenology. In this work different deposition methods for tin dioxide und titanium dioxide thin films are described and the results of several physical characterizations are shown. Both compounds have been deposited as thin films with sputtering and with thermal evaporation. Investigations of the materials morphology were done by transmission electron and atomic force microscopy and X-Ray diffraction. Some SnO2 samples have been annealed, while the TiO2 thin films have been doped with Fe ions post-preparational. In some cases the results show ferromagnetical behaviour in TiO2:Fe with ordering temperatures above 300 K. The conductivity measurements identify the Variable Range Hopping of localized states in the band gap as the dominating electronic transport process.
