2. Implications of FCC and HCP cobalt phases on wear performance of weld deposited cobalt-based coating

SS316L possesses comparatively low wear and abrasion resistance properties and thus needs proper surface modification to sustain under severe industrial applications. Cobalt-based Stellite 6, a suitable coating for such applications, contains 4–5 % of tungsten that provides solid solution strengthening to the cobalt-based matrix, due to which it shows good wear resistance in abnormal working conditions. Stellite 6 is dominated by face-centered cubic (FCC) and hexagonal close packed (HCP) cobalt along with different carbide phases such as M7C3, M23C6, MC and M6C where M is Cr, W, Mo, Co and Fe. Weld deposited cobalt-based Stellite 6 coating on SS316L substrate, contains FCC structure as primary phases. Pure cobalt contains a stable HCP phase at temperature below 417 °C and FCC phase at and above 417 °C up to melting point of pure cobalt i.e., 1495 °C. Under the frictional stresses caused during wear test, impact more on FCC cobalt than HCP cobalt due to the high slip system of FCC. Because of this, FCC cobalt underwent through high wear losses compared to HCP cobalt in cobalt-based coating. Present study revealed that, high wear losses are seen in samples where FCC cobalt phases are dominant. The observed mechanism by SEM shows abrasive wear in which material removal takes place by ploughing, micro cutting and indentation. The targeted industrial applications where such wear losses are supposed to happen includes slurry transportation applications such as pipes, sea engineering components, submersible pumps, control valves.

Introduction: Cannabis-based medicinal products (CBMPs) have been identified as a promising novel therapeutic for symptoms and comorbidities related to autism spectrum disorder (ASD). However, there is a paucity of clinical evidence of their efficacy and safety. Objective: This case series aims to assess changes to health-related quality of life and the incidence of adverse events in patients treated with CBMPs for associated symptoms of ASD enrolled on the UK Medical Cannabis Registry (UKMCR). Methods: Patients treated with CBMPs for ASD-related symptoms for a minimum of 1 month were identified from the UKMCR. 8. Outcome of complex tibial plateau fractures with Ilizarov external fixation with or without minimal internal fixation

Purpose: To evaluate the clinico-radiological outcome of complex tibial plateau fractures treated with Ilizarov external fixation with or without minimal internal fixation. Methods: This retrospective review was conducted on all the cases of Schatzker types V and VI tibial plateau fractures treated by Ilizarov external fixation between July 2006 and December 2015 with the minimum follow-up duration of one year. There were 30 patients: 24 males and 6 females, mean age 43.33 years, and mean follow-up 3.6 years. Three of them were open fractures; 15 cases were Schatzkertype V fractures and the other 15 type VI. According to AO/OTA classification, there were 11 type C1, 12 C2 and 7 type C3 fractures. Outcome assessment was made with American Knee Society Score (AKSS) and Rasmussen's Radiological Score (RRS) at final follow-up. Results: Out of the 30 cases, mini-open reduction was performed in 7, bone graft in 4, minimal internal fixation in 10 and knee temporary immobilisation in 11 patients. Mean duration of external fixation was 11.8 weeks. All fractures united. Pin tract infections in 7 and common peroneal neuropathy in 2 patients were self-limiting. Two patients had axial misalignment of less than 10°. At final follow-up, the mean knee range of motion was 114.7, mean AKSS 81.5 and mean RRS 16.7. On statistical analysis, Schatzker type of fractures, use of minimal internal fixation and knee-spanning did not influence the final outcome. Conclusion: Ilizarov external fixator with or without minimal internal fixation provides acceptable outcome for complex tibial plateau fractures. Care must be taken to look for minor loss of alignment, especially in Type VI Schatzker fractures after removal of the fixator. However small sample size precludes firm conclusions. 9. Metallurgical characterisation of multi-track Stellite 6 coating on SS316L substrate

- Abstract
ABSTRACTThis article attempts to investigate the root cause of the possible failure of the cobalt-based Stellite 6 coating deposited on SS316L substrate using plasma-transferred arc welding through an extensive metallurgical and mechanical analysis by considering parameters such as contact angle, dilution and aspect ratio. The chosen sample with optimum input parameters, such as weld current 100–180 A, travel speed 100–180 mm/min, etc., having good mechanical properties but with traces of minor coating irregularities is thoroughly investigated for its metallurgical performance. Studies revealed that the Stellite 6 coating is dominated by the FCC cobalt and HCP cobalt along with carbides of different elements such as M7C3, M6C and M23C6phases where M is tungsten and Chromium. Stellite 6 microstructure shows a hypoeutectic structure of dendrites rich in cobalt and interdendrites rich in chromium increasing microhardness. As arc current increases, percentage dilution increases, leading to a decrease in weld coating contact angle.

10. On the performance of metallurgical behaviour of Stellite 6 cladding deposited on SS316L substrate with PTAW process

- Abstract
ABSTRACTCobalt rich Stellite 6 cladding shows a superior result against wear, corrosion and galling. However, the metallurgical bonding at the cladding interface, if not achieved carefully, may result in failure during service. Hence, the proposed Stellite 6 weld overlay is deposited by plasma transferred arc welding (PTAW) process on SS316L substrate. Mechanical characterisation of this combination has been already attempted earlier with the help of a fine-tuned welding parameters. The same work has been reconsidered in the present research study for its detailed metallurgical investigation. In order to investigate the metallurgical variation at the interface, two samples with different weld parameters (sample A and B) have been considered. Cobalt rich phases have been obtained in both samples such as γ-Co (FCC), ϵ-Co (HCP), CrCo along with different carbides of chromium and tungsten such as Cr7C3, Cr23C6, W6C which are confirmed by EDS and XRD analysis. The formation of different phases shows the W6C, interdendritic eutectic phases Cr7C3, Cr23C6and primary dendritic phases γ-Co, ϵ-Co and CrCo that are chiefly responsible for the higher hardness of sample B which has lower heat input.

- Abstract
Voltage-gated sodium channel NaV1.7 is a genetically validated target for pain. Identification of NaV1.7 inhibitors with all of the desired properties to develop as an oral therapeutic for pain has been a major challenge. Herein, we report systematic structure–activity relationship (SAR) studies carried out to identify novel sulfonamide derivatives as potent, selective, and state-dependent NaV1.7 inhibitors for pain. Scaffold hopping from benzoxazine to chroman and indane bicyclic system followed by thiazole replacement on sulfonamide led to identification of lead molecules with significant improvement in solubility, selectivity over NaV1.5, and CYP2C9 inhibition. The lead molecules 13, 29, 32, 43, and 51showed a favorable pharmacokinetics (PK) profile across different species and robust efficacy in veratridine and formalin-induced inflammatory pain models in mice. Compound 51also showed significant effects on the CCI-induced neuropathic pain model. The profile of 51indicated that it has the potential for further evaluation as a therapeutic for pain.

12. Optimising fracture in automotive tail cap by firefly algorithm

- Abstract
Deep drawing is a manufacturing process in which sheet metal is progressively formed into a three-dimensional shape through the mechanical action of a punch forming the metal inside die. The flow of metal is complex mechanism. Pots, pans for cooking, containers, sinks, automobile body parts such as panels and gas tanks are among a few of the items manufactured by deep drawing. Uniform strain distribution in forming results in quality components. The predominant failure modes in sheet metal parts are springback, wrinkling and fracture. Fracture or necking occurs in a drawn part, which is under excessive tensile loading. The prediction and prevention of fracture depends on the design of tooling and selection of process parameters. Firefly algorithm is one of the nature inspired optimisation algorithms and is inspired by firefly's behaviour in nature. The proposed research work presents novel approach to optimise fracture in automotive component-tail cap. The optimisation problem has been defined to optimise fracture within the constraints of radius on die, radius on punch and blank holding force. Fire fly algorithm has been applied to find optimum process parameters. Numerical experimentation has been conducted to validate the results.

Adverse events were recorded and analysed. Statistical significance was determined by p< 0.050.Results: Seventy-four patients with ASD were included in the analysis. The mean age of participants was 32.7 (±11.6) years. There were significant improvements in general health-related quality of life and sleep as assessed by the EQ-5D-5L, SQS and GAD-7 at 1 and 3 months, with sustained changes in EQ-5D-5L and SQS at 6 months (p< 0.010). There were 180 (243.2%) adverse events reported by 14 (18.9%) participants. If present, adverse events were commonly mild (n= 58; 78.4%) or moderate (n= 81; 109.5%), rather than severe (n= 41; 55.4%).Conclusion: This study demonstrated an associated improvement in general health-related quality of life, and anxiety- and sleep-specific symptoms following initiation of treatment with CBMPs in patients with ASD. These findings, while promising, are limited by the confines of the study which lacks a control arm and is subject to attrition bias. Therefore, further evaluation is required with randomised controlled trials.

- Abstract
Monoclonal antibodies (mAbs) are currently the dominant class of biopharmaceuticals. Due to the high dosage requirements of most mAb therapeutics, high productivity and low aggregation are prevailing criteria during cell line generation and process development. Given that light chains (LCs) play an important role in antibody folding and assembly, and that most mAb producing cell lines also manufacture free LCs, we sought to investigate whether there was a relationship between free LC levels in cell culture media and mAb productivity/quality. To this end, a series of analytical methods were developed in order to quantify free LC content in cell culture media and assess mAb productivity and aggregation levels. Afterwards, conditioned media samples from different cell lines at identical culturing conditions and a single clone under varying culturing conditions were analyzed. Higher LC expression was found to correlate with higher cell viability, higher mAb productivity, and lower aggregation. While LC expression cannot yet be definitively considered the root cause of these phenomena, these results are consistent with the role of LCs in mAb production, suggesting that free LC expression levels may potentially serve as a parameter for cell line generation and cell culture process optimization., (© 2013 American Institute of Chemical Engineers.)
