The Mediterranean will be one of the focal points of climate change. The predicted dry and hot summers will lead to water scarcity in agriculture, which may limit crop production and growth. The olive tree serves as a model woody plant for studying drought stress and improving water resource management; thus, it is critical to identify genotypes that are more drought tolerant and perform better under low irrigation or even rainfed conditions. In this study, the metabolomic approach was used to highlight variations in metabolites in stems and leaves of three Italian olive cultivars (previously characterized physiologically) under two and four weeks of drought stress. Phenolic and lipophilic profiles were obtained by gas chromatography-mass spectrometry and ultra-high performance liquid chromatography-mass spectrometry, respectively. The findings identified the leaf as the primary organ in which phenolic variations occurred. The Maurino cultivar exhibited a strong stress response in the form of phenolic compound accumulation, most likely to counteract oxidative stress. The phenolic compound content of ‘Giarraffa’ and ‘Leccino’ plants remained relatively stable whether they were exposed to drought or not. Variations in the lipid profile occurred in leaves and stems of all the cultivars. A high accumulation of compounds related to epicuticular wax components was observed in the leaf of ‘Giarraffa’, while a strong reduction of lipids and long-chain alkanes occurred in ‘Maurino’ when exposed to drought stress conditions. The aim of this work was to evaluate the photosynthetic performance of Pear trees (cv. ‘Rocha’) infected with Erwinia amylovora, three months after suffering a pruning of infected branches (P-trees) compared with asymptomatic trees (C-trees) of the same orchard. Three months after pruning, P-trees looked healthy and were negative for the presence of E. amylovora. In September of 2018, fully expanded leaves of both P- and C- trees were sampled and analysed for photosynthetic parameters related to chlorophyll a fluorescence and gas exchange, alongside with pigments, total soluble sugars, starch, and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) contents. No significant differences were found in chlorophyll and carotenoids levels, but anthocyanins significantly decreased in P-trees. Also, despite the maximum quantum yield (Fv/Fm) significantly decreased in P-trees, the effective quantum yield of the PSII was maintained, paralleled with no changes in gas exchange parameters (PN, gs, Ci, E, iWUE, PN/gs), nor in RuBisCO relative content. Finally, the maintenance of the levels of total soluble sugars and starch also supports that the photosynthetic performance of P-trees, three months after pruning, reached values similar to those of the C-trees, contributing to the normal development and ripening of the fruit. Data support that pruning represents a reliable control measure against this quarantine pathogen. This work is the first evaluation of pruning in fire blight management regarding carbon metabolism in P. communis trees.

Global climate change-induced abiotic stresses (e.g., drought, salinity, extreme temperatures, heavy metals, and UV radiation) have destabilized the fragile agroecosystems and impaired plant performance and thereby reducing crop productivity and quality. Biostimulants, as a promising and eco-friendly approach, are widely used to address environmental concerns and fulfill the need for developing sustainable/modern agriculture. Current knowledge revealed that plant and animal derived stimulants (e.g., seaweeds and phytoextracts, humic substances, and protein hydrolysate) as well as microbial stimulants (e.g., plant beneficial bacteria or fungi) have great potential to elicit plant tolerance to various abiotic stresses and thus enhancing plant growth and performance-related parameters (such as root growth/diameter, flowering, nutrient use efficiency/translocation, soil water holding capacity, and microbial activity). However, to successfully implement biostimulant-based agriculture in the field under changing climate, the understanding of agricultural functions and action mechanism of biostimulants coping with various abiotic stresses at physicochemical, metabolic, and molecular levels is needed. Therefore, this review attempts to unravel the underlying mechanisms of action mediated by diverse biostimulants in relation to abiotic stress alleviation as well as to discuss the current challenges in their commercialization and implementation in agriculture under changing climate conditions. Plant-growth-promoting bacteria (PGPB) are gaining attention as a sustainable alternative to current agrochemicals. This study evaluated the impact of three Bacillus spp. (5PB1, 1PB1, FV46) and one Brevibacillus sp. (C9F) on the important crop tomato (Solanum lycopersicum) using the model cv. 'MicroTom'. The effects of these isolates were assessed on (a) seedlings' growth and vigor, and (b) adult potted plants. In potted plants, several photosynthetic parameters (chlorophylls (a and b), carotenoids and anthocyanins contents, transpiration rate, stomatal conductance, net CO2 photosynthetic rate, and intercellular CO2 concentration, and on chlorophyll fluorescence yields of light- and dark-adapted leaves)), as well as soluble sugars and starch contents, were quantified. Additionally, the effects on redox status were evaluated. While the growth of seedlings was, overall, not influenced by the strains, some effects were observed on adult plants. The Bacillus safensis FV46 stimulated the content of pigments, compared to C9F. Bacillus zhangzhouensis 5PB1 increased starch levels and was positively correlated with some parameters of the photophosphorylation and the gas exchange phases. Interestingly, Bacillus megaterium 1PB1 decreased superoxide (O2-) content, and B. safensis FV46 promoted non-enzymatic antioxidant defenses, increasing total phenol content levels. These results, conducted on a model cultivar, support the theory that these isolates differently act on tomato plant physiology, and that their activity depends on the age of the plant, and may differently influence photosynthesis.
